Abstract:
In this competitive manufacturing environment, material transportation is an important topic for researchers and practical applications. This study presents a real life example of an automotive equipment manufacturer problem. The manufacturer has several depots to serve 54 milk-run suppliers and complained about the fluctuations of milk-run vehicles between work days and depots. The aim of this study is to develop a systematic approach to make milk-run system more stable for manufacturer. Therefore the systematic approach which consists of, the milk-run route optimization with mixed integer linear programming model, container loading heuristic to increase the utilization of milk-run vehicles and time interval assignment heuristic to avoid coincidence of milk-run vehicles at the depots, is suggested to get over existing problems of a manufacturer.

With the rapid development of automobile industry, auto parts logistics emerged as an important part of promoting industrial development. Milk-run is used more and more frequent in auto parts supply. Based on the characteristics of auto parts requirement and milk-run, in order to satisfy the requirement of OEMs, the distribution center takes milk-run supply logistics model to a variety of vendors, using the idea of third party. The design of automobile parts milk-run vehicle routing is the problem of designing the non-full set of goods vehicle routing with time window. This thesis established a vehicle routing model with time window, studied milk-run vehicle routing of automobile parts, used the particle swarm optimization to solve the problem of the example, and obtained a satisfied results in acceptable time. It also verified the validity of the model and algorithm.

Os custos associados aos bens e serviços adquiridos representam um papel importante na tomada de decisão das empresas. É com base nesta importante área da gestão de uma organização, as Compras, que será desenvolvido este trabalho. O presente trabalho analisa as diferentes etapas do processo de abastecimento, pretendendo melhorar e facilitar a gestão das cargas, o abastecimento e receção dos materiais adquiridos e consequentemente reduzir os custos associados com transporte e a contratação de serviços de transporte. É feita uma identificação dos principais problemas encontrados bem como são identificadas oportunidades de melhoria que serão aplicadas à realidade encontrada, tendo sido implementado um método de entregas segundo o modelo Milk Run. Esta alteração trouxe elevadas vantagens ao processo de entregas e redução de custos com transportes tornando o processo mais Lean.
The costs associated with purchased goods and services play an important role in decision making of companies. It is based on this important area of management of an organization, Purchasing, that this work will be developed. This project analyses the different stages of supply and intends to improve and facilitate the management of loads, the supply of purchased goods and consequently reduce the costs associated with transport and hiring transport services. Main problems encountered are described and improvement opportunities identified leading to the implementation of a delivery method according to the Milk Run model. This change brought major advantages to the analysed process of delivery and reduced transport costs making the process leaner.

In today's world the challenge of logistics becomes more and more important for all companies, which consequently contributes to the need of an integrated system of in-house logistics. Goods must flow easily between stations and departments in order to achieve the best utilization of transportation as well as maintaining a good distribution structure to handle these processes in an efficient and effective way to enhance business performance and competitiveness. This report aims to provide research on how to strategically organize, plan and structure the flow of internal logistics within an organization. The research in this study originates from 3 main methods, a theoretical review, a case study and a benchmark. The main idea of the thesis was to contribute with scientific knowledge of improvements that can be achieved within internal logistics targeting companies operating within the service sector. In order to understand the scope of the problem the current state at the case Tropical Islands was analysed and served to identify areas where possible improvements could be achieved. Further on, in combination with the theory and empirical findings the main conclusions of the thesis was the need to strategically organize the flow of internal logistics to improve efficiency and enhance competitiveness. It was made evident that planning is essential in order to successfully implement the milk runs. Other tools could be utilized to further eliminate drawbacks that might occur.

L'attenzione sempre maggiore verso una corretta gestione della propria supply chain spinge sempre più aziende a rivedere le proprie attività logistiche inbound e outbound. All'interno della seguente tesi, maturata durante l'esperienza di tirocinio svolta presso Biesse S.p.A, verrà quindi dettagliato lo studio condotto circa la convenienza economica dell'introduzione di una politica di ritiro della merce in ottica Milk run. L'analisi muove i passi da un primo studio della letteratura matematica legata all'argomento, per poi procedere alla valutazione della situazione attuale dei ricevimenti e termina con la valutazione tecnica e economica della nuova proposta. Particolare enfasi viene posta infine sulla necessità di poter gestire la creazione delle rotte in maniera dinamica, motivazione che ha portato alla realizzazione di un apposito software tramite linguaggio Python.

Questo elaborato, frutto dell'esperienza maturata dal candidato durante il suo percorso di tirocinio, svoltosi presso la sede principale di ACMA S.p.A., ha come obiettivo la razionalizzazione di un campione di fornitori in ottica milk-run, in modo da ridurre i tempi di approvvigionamento di materiale. L'azienda in questione, tra i leader mondiali nella produzione di macchine automatiche per il confezionamento e l'impacchettamento di beni di consumo, ha la necessità di ridurre i tempi di consegna dei propri prodotti, soddisfando le aspettative del mercato, che sono sempre più stringenti, senza dover aumentare la quantità di stock e di conseguenza i relativi costi. Per fare questo si è deciso di progettare una strategia di pianificazione dei prelievi dai fornitori in base alla teoria del milk-run, adattando la metodologia alla particolare situazione aziendale. A tale scopo è stato selezionato, sulla base di analisi sia numeriche sia strategiche, un campione di fornitori su cui progettare la soluzione, in modo da poterla implementare e successivamente, in caso di esito positivo, espanderla a un campione di fornitori più numeroso. I risultati, basati su simulazioni della situazione to-be rispetto a quella attuale, mostrano una notevole riduzione dei Lead Time dei fornitori, con tempi di consegna ridotti fino a due settimane e con una media vicina ai cinque giorni. Mostra, inoltre, un netto miglioramento delle prestazioni logistiche dei fornitori, aumentando la percentuale di OTD e diminuendo i giorni medi di ritardo. I costi totali non subiscono sostanziali variazioni rispetto alla situazione precedente. Il progetto è un punto di partenza per un'estensione di questo strumento all'intera Supply Chain, in modo da avere non solo un aumento del livello di servizio dell'intera Supply Chain, ma anche di ridurre, o comunque di mantenere inalterati, i relativi costi logistici.

Com o processo de globalização das economias em curso, as empresas, em geral, vêm sofrendo crescente concorrência na busca e manutenção de clientes. Qualquer oportunidade que represente vantagem competitiva tem sido analisada e frequentemente adotada pelas mais diversas empresas. Nesse contexto, a cadeia de suprimentos de uma organização pode se constituir em um diferencial competitivo, na medida em que favorece a redução de custos e a melhoria da qualidade dos recursos de produção adquiridos. Por outro lado, atrasos e/ou faltas de insumos na cadeia produtiva, geram perdas de recursos e pode prejudicar a imagem da empresa perante o mercado. Dentro dessa perspectiva foi estudada a cadeia de suprimentos de uma empresa multinacional, fabricante de máquinas e equipamentos, situada no Vale do Paraíba, no Estado de São Paulo. O desenvolvimento do trabalho teve como objetivo identificar os atrasos e faltas de insumos provenientes da cadeia de suprimentos, bem como do processo de compras e da coleta de peças para o sistema de produção até então vigente na empresa. Para reverter esse quadro, optou-se por um novo sistema de compras e de coleta de peças que pudesse contribuir para a eliminação ou a redução de perdas. Para isso, em um processo decisório compartilhado, adotaram os fundamentos do Comakership e da Gestão por Indicadores, visando alcançar os objetivos propostos. Os resultados obtidos evidenciaram a relevância estratégica da integração da cadeia de suprimentos, utilizando-se fornecedores parceiros. A identificação das perdas e a adoção de um novo sistema de suprimentos e de coleta de peças, aplicando-se o processo Milk Run, foram responsáveis pela redução dos custos provenientes de atrasos e faltas de peças que impactavam a produção da empresa, entre outros benefícios. As soluções encontradas geraram uma relação mais estável e duradoura com os fornecedores, além de ampliar a competitividade e as condições de sobrevivência da própria organização.
The current process of economic globalization has brought a growing competitiveness for the maintenance and persuit of customers among the companies. Each opportunity that represents competitive advantage has been analyzed and often adopted by several companies. In this context, the suppliers network of the companies may imply a competitive differential as it favors the reduction of costs and the improvement of quality in the acquired production resources. On the other side, delays and/or missing resources in the production chain generate losses of resources and may damage the image of the company in the market. With that aim a study was carried out in the supply chain of a multinational company which makes equipments and machinery in Vale do Paraíba in the state of São Paulo. The development of this work began with the identification of delays and lacks of the resources that come from the supplying chain, as well as, from the process of purchasing and collecting equipment pieces for the system of production then present in the company. In order to reverse the situation, a new purchasing and collecting system that could contribute for the elimination or reduction of those losses was chosen. For that a shared decisive process in the principles of Comakership aligned to the indicative management aiming were adopted. The obtained results have shown the strategic relevance of the integration of the supplying chain. The partnership with suppliers, identification of losses and adoption of a new supplying and collecting system using the Milk Run process, were responsible for the reduction of costs caused by the delays and lack of equipment pieces that used to get the company production stuck among other benefits. The solutions found generated a more stable and lasting relationship with suppliers besides amplifying the competitiveness and survival conditions of the company itself.

Abstract:
The purposes of this project were: 1) to introduce the FecB gene to the Awassi and Assaf breeds in Israel and the Rambouillet breed in the U.S.A. aiming in the long run to establish Awassi, Assaf and Rambouillet nucclei breeding flocks homozygous for the F gene in which the contribution of the Booroola Merino genetic background will be less than 10%; (In the U.S., Booroola crosses with Suffolk and Targhee were also studied. 2) to evaluate the effect of the FecB gene and different proportions of Booroola Merino genetic background on lamb survival, growth, milk production and wool production in Booroola crosses with the native breeds; 3) to reveal the specific effect of the FecB gene on ovarian development, follicle stimulating hormone (FSH) and inhibin secretion in prepubertal ewe lambs and in adult ewes in order to define physiological criteria for distinguishing carriers of the FecB allele from non-carriers and 4) to identify genetic markers linked to the FecB gene to assist in selection of genotypes within the Booroola crosses. Introgression of the Booroola gene reached the stage of the third backcross in the Awassi, Assaf and the Rambouillet crosses. In all cases the Booroola crosses were superior in prolificacy. However, they were inferior in comparison to the local breeds in production due to Booroola Merino genes other than the FecB. It is expected that the beneficial economic contribution of the Booroola gene will increase along with the upgrading to the local breeds. FSH plasma levels and induced ovulation rate of 5 month old FecB carriers among the crossbreeds. The OarAE101 marker can assist in detecting FecB carriers among Booroola-Awassi crosses. However, this marker is informative only in some of the families.

Despite growing interest in low-speed automated shuttles, pilot deployments have only just begun in a few places in the U.S., and there is a lack of studies that estimate the impacts of a widespread deployment of automated shuttles designed to supplement existing transit networks. This project estimated the potential impacts of automated shuttles based on a deployment scenario generated for a sample geographic area: Santa Clara County, California. The project identified sample deployment markets within Santa Clara County using a GIS screening exercise; tested the mode share changes of an automated shuttle deployment scenario using BEAM, an open-source beta software developed at the Lawrence Berkeley National Laboratory to run traffic simulations with MATSim; elaborated the model outputs within the R environment; and then estimated the related impacts. The main findings have been that the BEAM software, despite still being in its beta version, was able to model a scenario with the automated shuttle service: this report illustrates the potential of the software and the lessons learned. Regarding transportation aspects, the model estimated automated shuttle use throughout the county, with a higher rate of use in the downtown San José area. The shuttles would be preferred mainly by people who had been using gasoline-powered ride hail vehicles for A-to-B trips or going to the bus stop, as well as walking trips and a few car trips directed to public transport stops. As a result, the shuttles contributed to a small decrease in emissions of air pollutants, provided a competitive solution for short trips, and increased the overall use of the public transport system. The shuttles also presented a solution for short night trips—mainly between midnight and 2 am—when there are not many options for moving between points A and B. The conclusion is that the automated shuttle service is a good solution in certain contexts and can increase public transit ridership overall.

Abstract:
Mycobacterium avium subsp. paratuberculosis (MAP) is the etiological agent of a severe inflammatory bowel disease (IBD) in ruminants, known as Johne’s disease or paratuberculosis. Johne’s disease is considered to be one of the most serious diseases affecting dairy cattle both in Israel and worldwide. Heavy economic losses are incurred by dairy farmers due to the severe effect of subclinical infection on milk production, fertility, lower disease resistance and early culling. Its influence in the United States alone is staggering, causing an estimated loss of $1.5 billion to the agriculture industry every year. Isolation of MAP from intestinal tissue and blood of Crohn's patients has lead to concern that it plays a potential pathogenic role in promoting human IDB including Crohn’s disease. There is great concern following the identification of the organism in animal products and shedding of the organism to the environment by subclinically infected animals. Little is known about the molecular basis for MAP virulence. The goal of the original proposed research was to identify MAP genes that are required for the critical stage of initial infection and colonization of ruminants’ intestine by MAP. We proposed to develop and use signature tag mutagenesis (STM) screen to find MAP genes that are specifically required for survival in ruminants upon experimental infection. This research projected was approved as one-year feasibility study to prove the ability of the research team to establish the animal model for mutant screening and alternative in-vitro cell systems. In Israel, neonatal goat kids were repeatedly inoculated with either one of the following organisms; MAP K-10 strain and three transposon mutants of K-10 which were produced and screened by the US PI. Six months after the commencement of inoculation we have necropsied the goats and taken multiple tissue samples from the jejunum, ileum and mesenteric lymph nodes. Both PCR and histopathology analysis indicated on efficient MAP colonization of all the inoculated animals. We have established several systems in the Israeli PI’s laboratory; these include using IS900 PCR for the identification of MAP and using HSP65-based PCR for the differentiation between MAV and MAP. We used Southern blot analysis for the differentiation among transposon mutants of K-10. In addition the Israeli PI has set up a panel of in-vitro screening systems for MAP mutants. These include assays to test adhesion, phagocytosis and survival of MAP to/within macrophages, assays that determine the rate of MAPinduced apoptosis of macrophages and MAP-induced NO production by macrophages, and assays testing the interference with T cell ã Interferon production and T cell proliferation by MAP infected macrophages (macrophage studies were done in BoMac and RAW cell lines, mouse peritoneal macrophages and bovine peripheral blood monocytes derived macrophages, respectively). All partners involved in this project feel that we are currently on track with this novel, highly challenging and ambitious research project. We have managed to establish the above described research systems that will clearly enable us to achieve the original proposed scientific objectives. We have proven ourselves as excellent collaborative groups with very high levels of complementary expertise. The Israeli groups were very fortunate to work with the US group and in a very short time period to master numerous techniques in the field of Mycobacterium research. The Israeli group has proven its ability to run this complicated animal model. This research, if continued, may elucidate new and basic aspects related to the pathogenesis MAP. In addition the work may identify new targets for vaccine and drug development. Considering the possibility that MAP might be a cause of human Crohn’s disease, better understanding of virulence mechanisms of this organism might also be of public health interest as well.
