Full textThe RTN4R gene is located in the 22q11 region and it encodes an axonal receptor (Nogo-R), which is involved in neuronal growth. Previous studies have also suggested association of RTN4R with schizophrenia related phenotypes. Here, we investigated potential relevance of genetic variation in RTN4R for molecular, brain imaging and behavioral correlates including prefrontal Nogo-R mRNA expression, prefrontal white matter volume and activity during working memory, as well as behavioral performance during cognition. In a post-mortem sample of 268 healthy subjects, we found that a single nucleotide polymorphism within RTN4R, rs696884, was associated with prefrontal Nogo-R mRNA expression, with AA individuals having lower mRNA levels relative to GG subjects. Furthermore, in vivo investigation in 282 healthy individuals indicated that the AA genotype was also associated with lower prefrontal white matter volume, greater prefrontal working memory activity during fMRI, and poorer cognitive abilities during executive functions. Overall these results suggest that genetic variation in RTN4R modulates prefrontal function and call for further investigation of the relationship between this gene and schizophrenia.
