In the Phaedo, to explain why the philosopher lives in the unusually ascetic way he does, Socrates explains what someone realizes when philosophy takes possession of his soul and how he changes his behavior on the basis of this information. This paper considers the conception of belief the character uses in this explanation and whether it is the same as the conception Michael Frede thinks the historical Socrates is likely to have held and that the Stoics much later incorporated into their doctrine of practice.

Résumé La treizième réunion du Symposium Aristotelicum, en 1993, a eu une très étrange et très triste destinée. Certes, elle s’est tenue dans le cadre enchanteur de la Chartreuse de Pontignano, près de Sienne ; elle a donné lieu, comme ses devancières, à des communications et à des discussions d’un vif intérêt. Mais l’édition de ses Actes, pour une fois, s’est heurtée à d’insurmontables obstacles. La charge en avait été initialement confiée à Mario Mignucci et à Michael Frede, deux des plus fidèles et stimulants participants du Symposium. Ils ont été tragiquement enlevés à notre admiration et à notre affection, le premier en 2004, sous les coups d’une longue et impitoyable maladie, le second en 2007, en conséquence d’un accident imprévisible et brutal. Le retard causé à la publication du XIIIe Symposium par cette double et douloureuse disparition n’a pu être comblé jusqu’à présent ; les membres du comité organisateur m’ont assuré qu’à leur avis, il risquait de ne l’être jamais. Par une coïncidence émouvante (en tout cas pour moi), trois semaines seulement avant la mort de Michael Frede, mon collègue et ami Thomas De Koninck me demanda si j’accepterais de publier dans le Laval théologique et philosophique l’étude que j’avais présentée, plus de dix ans auparavant, au XIIIe Symposium. Je passe sur les divers scrupules qui me firent hésiter quelque temps. L’insistance du Professeur De Koninck et celle de ses collaborateurs, Paul Asselin et Martin Achard, en eurent finalement raison, ce dont je leur suis très profondément reconnaissant. Quant à ce texte, le lecteur voudra bien se souvenir de la longue histoire dont il est l’ultime fruit. Il serait bien difficile de le résumer : il est, il tente d’être cela même pour quoi il se donne, à savoir pour une lecture détaillée du commentaire par Ammonius du célèbre premier chapitre du De Interpretatione, lecture focalisée non pas tellement sur la lumière que le commentaire ancien peut (ou peut ne pas) jeter sur la lettre et sur l’interprétation du texte aristotélicien que sur ce que ce commentaire peut nous apprendre sur les méthodes, les choix, les comportements intellectuels de son auteur lui-même, et sur ses propres motivations philosophiques et pédagogiques face à un texte comme celui qu’il entreprend de commenter.

Sexto Empírico, seu principal expoente. Recorremos à interpretação de Myles F. Burnyeat, Michael Frede e Jonathan Barnes para elucidar a questão da viabilidade prática da vida cética, problema que tem sido a principal réplica ao ceticismo desde o ataque cético à filosofia da <em>Stoa</em>. Levando então em consideração o acirrado debate ocorrido entre as principais filosofias do período Helenístico, propusemos uma investigação sobre como se articulam o problema do conhecimento e de sua aquisição, da filosofia prática e da atitude discursiva do cético para compreender a réplica de Sexto ao argumento de que o cético se auto-refuta.</p><div class="page" title="Page 1"><div class="layoutArea"><div class="column"><p><strong>Sextus Empiricus: ethics, language and epistemology of the second century a.C. in Alexandria </strong></p><p><strong>Abstract</strong></p><p><span>This paper deals with the following topics of the sceptical philosophy of the Pyrrhonic kind: ethics, language and epistemology, as thought by the Alexandrian medic of the second century A.C. Sextus Empiricus, which was the main exponent of the Pyrrhonism. We use the interpretations of Myles F. Burnyeat, Michael Frede and Jonathan Barnes to elucidate the problem of the practical viability of the sceptical life, which has been the main reply to the scepticism since the sceptical attack to the Stoic philosophy. Considering then the strained debate happened between the main philosophies of the Hellenistic period, we propose an investigation about how the problem of the knowledge and his acquisition articulates himself with the problems of the practical philosophy and of the sceptical’s discursive attitude, this all is necessary to comprehend the Sextus’ reply to the argument of the sceptical self-refutation. </span></p><p><strong>Keywords: </strong><span>practical philosophy; knowledge; Sextus Empiricus; Pyrrhonism </span></p></div></div></div>

El objeto de estudio de este trabajo es la creencia. Nuestro objetivo es mostrar cómo a través de ese concepto el escepticismo moderno de Hume y el antiguo de Pirrón pueden llegar a converger en una posición bastante similar. Ambas actitudes teóricas y a la vez prácticas consideran que es lícito dar “asentimiento a aquello que se presenta al hombre de modo inevitable e independientemente de nuestra voluntad. Sabemos que tanto para Sexto como para Hume hay algo, independientemente del nombre que le demos (“phainómenon”, “perceptions”), que nos “induce” o nos “exige” asentimiento, término fundamental que parece indicar un concepto común al escepticismo antiguo y al pensamiento de Hume. ¿Será entonces legítimo o justificado seguir apoyando el juicio de la historia que calificó en un principio a Hume de “empirista” o posteriormente de “naturalista”? ¿No será más exacto rastrear cierto pirronismo, aunque sea inconsciente, en las ideas del escocés? Nuestra respuesta parte de la interpretación no tradicional de Michael Frede que fundamentó con cierta solidez la idea de que el escéptico no renuncia a tener creencias o, al menos, puede tenerlas sin infringir con ello la epoché. La cuestión central será entonces determinar qué significado habría que atribuir al término “creencia”.

Eine belangvolle Transformation ist das zwölfte Buch (Lambda) der „Metaphysik“ des Aristoteles durch verschiedene Interpretationen im Laufe der Jahrhunderte widerfahren, sodass es als eine theologische Schrift bis heute überliefert ist (1. Kapitel). Dabei waren sowohl der ursprüngliche Sinn des Autors als auch die Struktur des Textes kaum erkennbar geworden. Die vorliegende Arbeit ist eine historische Rekonstruktion der verschiedenen Interpretationen des Textes seit den ersten peripatetischen Denkern bis zur Gegenwart, insbesondere die von Michael Frede (2. Kapitel), Lindsay Judson (3. Kapitel) und Stephen Menn (4. Kapitel). Frede behauptet, das Buch Lambda sei eine Untersuchung der Substanz (Ousiologie); Judson denkt, dass es die erste Philosophie vorlegt; Menn argumentiert, es sei eher eine Forschung der ersten Prinzipien (Archäologie). Hier wird eine weitere Meinung vertreten, nämlich, dass Lambda die Untersuchung der Prinzipien der Substanz ist und somit verkörpert es Aristoteles’ Verständnis der Sophia (5. Kapitel). Überdies löst diese neuartige Lesart des Textes das Problem der Stelle des zwölften Buches innerhalb der „Metaphysik“ dank der Abfassung einer Karte, die seine Struktur verdeutlicht (Anhänge).
The twelfth book (Lambda) of the “Metaphysics” of Aristotle has undergone a crucial transformation through the diverse interpretations over the centuries. Thus it has been transmitted up to today as a theological writing (chapter 1). But this made hardly recognizable both the original intention of the author and the structure of the text. The present work is a historical reconstruction of the various interpretations of the text since the first Peripatetic thinkers until today, especially those by Michael Frede (chapter 2), Lindsay Judson (chapter 3) and Stephen Menn (chapter 4). Frede states that the book Lambda is an inquiry into the substance (ousiology); Judson thinks that it presents the first philosophy; Menn argues that it is rather an investigation about the first principles (archaeology). A different opinion is set forth here, namely, that the book Lambda explores the principles of the substance, and in doing so it embodies Aristotle’s understanding of the sophia (chapter 5). Moreover, this new reading of the text solves the problem of the position of the twelfth book in the “Metaphysics” thanks to the composition of a map, which clarifies its structure (appendix).

The study analysed the way that tourists, the majority being regulars, used to live nude under cedar trees scattered on the beaches, the so-called kavatzas. The study remarks that the profiles of Gavdos travellers are varied but most of them share a certain rejection of modern society, and the spaces they occupy correspond to what the French philosopher Michel Foucault called heterotopias, i.e. spaces adopting behaviours which are at odds with social rules, and everyone can feel free and in harmony with nature. It is suggested that this profile goes beyond degrowth inspired travelling tourism; it is about the meaning of life.

This detailed account of James Marshall Hendrix’s life before he transformed into “Jimi” Hendrix covers his hardscrabble childhood in Seattle, his early musical inspirations, first instruments and bands, and stint in the U.S. Army. Following his discharge, Hendrix embarks on his professional career, playing the chitlin circuit and making his first recordings as a studio musician. He then lands in New York City, where he lives in abject poverty until his “discovery” by Linda Keith and Bryan “Chas” Chandler. Chandler sees him perform “Hey Joe” and Bob Dylan’s “Like a Rolling Stone” in a Greenwich Village club and arranges his passage to London. In a move that will come back to haunt him, Hendrix agrees to let Chandler and Michael Jeffery manage his career.

Abstract:
A cDNA encoding full length single chain urokinase-type plasminogen activator (scu-PA) was cloned and sequenced. The recombinant scu-PA (rscu-PA) cDNA as well as the cDNA of two mutants constructed by deoxyoligonucleotide directed mutagenesis of Lys158 in rscu-PA to Gly158 (rscu-PA-Gly158 ) or to Glu158 (rscu-PA-Glu158 ) were inserted into SV40 early promoter/enhancer based expression vectors, which were used to transfect Chinese Hamster Ovary (CHO) cells. The expression products were purified from serum-free conditioned media by immunoadsorption on an insolubilized monoclonal antibody raised against natural scu-PA (nscu-PA), followed by gel filtration.The amidolytic activity of the three rscu-PAs was low (< 500 IU/mg). The mutant rscu-PAs, in contrast to the rscu-PA and nscu-PA, could not be converted into an amidolytically active two-chain form (tcu-PA) by plasmin. The mutant scu-PAs had a very low specific activity (< 1,000 IU/mg) on fibrin plates, whereas wild type rscu-PA had a specific activity < 1000 IU/mg. The mutant scu-PAs did not cause lysis of a I-fibrin labeled plasma clot immersed in citrated human plasma. Serum-free medium from a control transfected CHO cell line showed no significant plasminogen activating activity.In a purified system, both rscu-PA-Gly and rscu-PA-Glu activate plasminogen following Michaelis-Menten kinetics, with a much lower affinity (K = 60-80 yM) but with a higher catalytic rate constant (k2 = B.01 s-1) as compared to the wild type rscu-PA (K =1.0 yM, k = 0.002 s-1).It is concluded thaz conversion of scu-PA to tcu-PA is prerequisite for the activation of plasminogen. However, Lys158 seems to be important for the stability of the Michaelis complex between scu-PA and plasminogen.

Abstract:
Physiological tissue-on-a-chip technology is enabled by adapting microfluidics to create micro scale drug screening platforms that replicate the complex drug transport and reaction processes in the human liver. The ability to incorporate three-dimensional (3d) tissue models using layered fabrication approaches into devices that can be perfused with drugs offer an optimal analog of the in vivo scenario. The dynamic nature of such in vitro metabolism models demands reliable numerical tools to determine the optimum tissue fabrication process, flow, material, and geometric parameters for the most effective metabolic conversion of the perfused drug into the liver microenvironment. Thus, in this modeling-based study, the authors focus on modeling of in vitro 3d microfluidic microanalytical microorgan devices (3MD), where the human liver analog is replicated by 3d cell encapsulated alginate hydrogel based tissue-engineered constructs. These biopolymer constructs are hosted in the chamber of the 3MD device serving as walls of the microfluidic array of channels through which a fluorescent drug substrate is perfused into the microfluidic printed channel walls at a specified volumetric flow rate assuring Stokes flow conditions (Re<<1). Due to the porous nature of the hydrogel walls, a metabolized drug product is collected as an effluent stream at the outlet port. A rigorous modeling approached aimed to capture both the macro and micro scale transport phenomena is presented. Initially, the Stokes Flow Equations (free flow regime) are solved in combination with the Brinkman Equations (porous flow regime) for the laminar velocity profile and wall shear stresses in the whole shear mediated flow regime. These equations are then coupled with the Convection-Diffusion Equation to yield the drug concentration profile by incorporating a reaction term described by the Michael-Menten Kinetics model. This effectively yields a convection-diffusion–cell kinetics model (steady state and transient), where for the prescribed process and material parameters, the drug concentration profile throughout the flow channels can be predicted. A key consideration that is addressed in this paper is the effect of cell mechanotransduction, where shear stresses imposed on the encapsulated cells alter the functional ability of the liver cell enzymes to metabolize the drug. Different cases are presented, where cells are incorporated into the geometric model either as voids that experience wall shear stress (WSS) around their membrane boundaries or as solid materials, with linear elastic properties. As a last step, transient simulations are implemented showing that there exists a tradeoff with respect the drug metabolized effluent product between the shear stresses required and the residence time needed for drug diffusion.

This paper reports a theoretical study of the power performance and environmental footprint of high-speed vessels in calm deep water, paying particular attention to a high-speed catamaran cargo vessel Faltinsen [1]. The power analysis uses a modified version of the method of Doctors and Day [2]. One modification to their approach is that the wave resistance and free surface wave profiles, have been numerically predicted using two different linear potential flow field theories; Michell thin-ship theory (Yeung and Wan [3]) and the 3D Rankine panel method (Hess and Smith [4]), respectively. We have based the latter method on the combination of the Dawson [5] upstream finite difference operator and the ‘staggered grid’ technique (Jensen et al. [6]). In addition, this paper shows how the viscosity effects modeled on the free surface layer influence the behavior of the wave resistance curves as predicted by Michell’s thin ship theory. This was done by adopting the approaches described by Tuck [7] and Lazauskas [8]. The wave resistance models were verified and validated using the examples of Wigley monohull (Lazauskas [8], Tarafder and Suzuki [9]) and catamaran (Yeung and Wan [3], Tarafder and Suzuki [10]) analytical forms, as well as on the Tuck parabolic strut (Tuck [7]). Furthermore, the resistance models were extended using free surface profile estimates at selected Froude numbers and applied to the case of a high-speed catamaran cargo vessel. The numerical results obtained were compared with published results and their accuracy and application feasibility is discussed from the perspective of preliminary high-speed vessel design. Based on the discussion above, suitable wave resistance models were selected and combined with the modified Doctors and Day [2] method giving the total resistance of the high-speed catamaran cargo vessel in the interval of Froude numbers. Having the estimates of total resistance, enables the effective power to be found. The results were compared with known results and were found to be in good agreement. Through combination of the effective power and propulsion factors, the brake power of the generic propulsion system is predicted. Taking into account the type of vessel being analyzed, the most common type of the propulsion diesel engine was selected. Finally, knowing the indicative environmental footprint of the selected engine, emission levels of Green House Gases (GHG) and solid particles were obtained. The emission results are further discussed in an ecologically friendly high-speed vessel design perspective (IMO [11, 19]).

Abstract:
The knowledge of the pH and ionic strength dependence of kcat and Km for the hydrolysis of S2238 (HD-phe-pip-arg-pNA.2HC1) by alpha-thrombin is essential in determining optimal reaction conditions of residual enzyme in systems where also protease inhibitors and glycosaminoglycan catalysts play a role.We studied the kinetic behavior of S2238 in piperazine/glycylglycine/NaOH buffers from pH 6 to 11 and with a calculated ionic strength up to 0.7 M taking into account the pH-dependent concentration of the buffer species. The kinetic parameters of 60 Michaelis-Menten substrate functions were used for the setup of ionic strength and pH profiles. The kcat values are dependent upon the ionic strength, increasing steeply up to about 0.3 M and decreasing again at high ionic strength.The Km however,reflecting the affinity between enzyme and substrate,is nearly unaffected.The Km values at very alkaline pH are markedly elevated,indicating a conformational form which does not readily bind substrate.The pH profiles for kcat and kcat/Km are displaced towards the low pH side with increasing ionic strength.The ascending limb corresponds to the pK of the Asp-His charge relay system,decreasing with increasing ionic strength from 7.2 to 6.6 in the ES complex and from 6.8 to 6.6 in the free enzyme.Apparently substrate binding provokes a pK increase of the active His residue.The descending limb in the kcat profile could be described by a hypothetical pK varying from 11.5 to 10.7 but the activity decrease is probably due to enzyme inactivation.The alkaline limb of the kcat/Km profile is governed by a pK of 9.4 which is rather independent of the ionic strength and could be attributed to the B-chain terminal isoleucine ,forming a salt bridge with Asp 194 and stabilizing the active site conformation as proven for other serine proteases.Data analysis via a modified Debije function with appropriate estimates for the dielectric constant and the radius of the macro-ion can provide information on the charge density of the enzyme.
