Teng, Y. "Nucleotide excision repair in the Saccharomyces cerevisiae MFA2 gene." Thesis, Swansea University, 2000. http://ethos.bl.uk/OrderDetails.do?uin=uk.bl.ethos.639174.

Full text
Abstract:
An end-labelling strategy for mapping DNA damage in yeast Saccharomyces cerevisiae at the level of the nucleotide is presented. Compared with other high resolution damage mapping protocols, the method described in this thesis is simpler and more efficient with less radioactive manipulation. The MFA2 gene, which is actively transcribed in a-mating type cells whereas repressed in α-mating type cells, provides an ideal model system for studying DNA repair due to its small size and easy control of its expression. The data from nucleotide excision repair competent strains showed that repair of Cyclobutane Pyrimidine Dimers (CPDs) in the upstream region of the MFA2 gene is enhanced in the Mcm1 binding region, and the closer to the transcription start point, the quicker the repair rate observed except for the TATA box region. This enhanced repair rate becomes fastest in the MFA2 transcribed region due to the transcription-coupled repair (TCR). The data from the rad16 mutant strain provided the first evidence that Rad16 is not absolutely required to repair CPDs in some upstream control sequences despite being nontranscribed regions. Experiments with a rad26 mutant showed that Rad26 is essential for TCR of the whole MFA2 transcribed region. Here, although TCR was abolished in the transcribed region, enhanced repair still occurred in the control region of MFA2. Studies with yeast replication protein A mutants indicated that the DNA binding domain, not the protein interaction domain of RPA70, confers a defect in global repair and only partly impairs the process of TCR. The experiments with transcription adaptor mutants, Δgcn5 and Δada2, identified a requirement for these gene products in Nucleotide Excision Repair (NER) of CPDs. Comparing the two adaptors examined, Gcn5 plays a more important role than Ada2 in both MFA2 transcription and CPD repair.

Abstract:
The Saccharomyces cerevisiae checkpoint genes RAD9, RAD17, RAD24, and MEC3 are required for the transcriptional induction of a group of genes including some for DNA repair. In this thesis two of the checkpoint genes, RAD9 and RAD24, and two nucleotide excision repair (NER) genes RAD16 and RAD26, have been investigated for their role in inducible NER at the MFA2 gene at nucleotide resolution. In wild type cells, enhanced NER is detected in cells received a minor UV irradiation one hour prior to a main UV dose compared to cells only treated with the main UV dose. This inducible NER is not detectable in mutant strains rad9, rad24, rad9rad24, rad16 and rad26. These data suggest that inducible NER is dependent on the checkpoint genes RAD9 and RAD24, on the RAD16 gene that is essential for global genome repair (GGR), and on the RAD26 gene that is required for efficient transcription-coupled repair (TCR). A second aspect of the thesis examined the role of histone acetylation in NER. Histone acetyltransferases (HATs) play an important role in remodelling chromatin structure during transcription activation. Here, the role of one HAT, Gen5, in the NER of UV induced CPDs in the upstream region of MFA2 has been examined. In Δgcn5 strains, NER of both strands in the control region of MFA2 is slower than in wild type cells. As Gen5 regulates MFA2 transcription, probably through chromatin modification, its role in chromatin organization in the control region of MFA2 would appear to influence DNA repair in that region.

Tur, Torres Juan. "Role of Mfn2 in Macrophage Inflammatory Responses." Doctoral thesis, Universitat de Barcelona, 2017. http://hdl.handle.net/10803/402898.

Full text
Abstract:
Mitochondria are well known for their role as bioenergetic and biosynthetic organelles. Recently, they also have emerged as one of the main regulators of innate immune responses, mostly for its ability to modulate several signaling pathways through mechanisms such ROS production. Mitofusin-2 (Mfn2) is a GTPase located in the external mitochondrial and ER membranes. It is responsible for the fusion between mitochondria and the ER-mitochondria contacts, both necessary for the correct functioning of both organelles. Even that, the role of Mfn2 in immune responses. Here we demonstrate that Mfn2 is crucial for the pro-inflammatory activation of macrophages. Murine Mfn2-/- macrophages show a disruption in the mitochondrial network morphology that leads to loss in the mitochondrial membrane potential and in mitochondrial respiration. These two defects in the mitochondrial function do not affect their ability to normally generate ATP. However, the production of ROS in the mitochondria from Mfn2-/- macrophages is markedly decreased. This defect in ROS generation leads to a severe dysfunction in macrophage responses, particularly in the inflammatory activation, phagocytosis, and the processing of proteins. First, the decrease in ROS in Mfn2-/- macrophages results in a defective activation of p38, ERK, and NF-kB signaling pathways in response to LPS. The decrease in these signaling cascades leads to a reduction in the production of pro- inflammatory cytokines, severely impairing their ability to undergo pro- inflammatory activation. Second, in addition to show reduced ROS levels, Mfn2-/- also show an accumulation of autophagosomes due to a Mfn2-dependent defect in the autophagosome-lysosome fusion. As a result to both increased autophagy and decreased ROS levels, Mfn2-/- macrophages show decreased expression of type- A scavenger receptors. We demonstrate that these alterations lead to a widespread defect in the phagocytic capabilities of Mfn2-/- macrophages, showing defective phagocytosis of bacteria (both gram positive and gram negative) and apoptotic bodies. Thirdly, Mfn2-/- macrophages also show a defect in the bactericidal activity of phagocyted bacteria, as well as a defective proteolysis, being unable to process antigens to present them to CD4+ cells in a MHC-II context, and therefore, potentially impairing their ability to initiate adaptive immune responses. Finally, we demonstrated that Mfn2 is relevant in in vivo models of inflammation or infection. Myeloid-conditional Mfn2-/- mice were infected with either Listeria monocytogenes or Mycobacterium tuberculosis. In both models, Mfn2-/- mice showed a severe decrease in their survival, when compared to their WT counterparts. Furthermore, the colony counts in selected organs (spleen and liver for listeria, spleen and lung for tuberculosis) was significantly increased in Mfn2-/- mice, indicating that Mfn2 in macrophages is required to effectively control bacterial infections. In addition, we performed a model of sterile inflammation using the irritant DNFB on the mice’s ear. Confirming the in vitro results, Mfn2-/- mice show decreased inflammation in the ear, as confirmed by the decrease in size and weight, and the reduced expression of inflammatory cytokines. All these findings suggest that Mfn2 is a crucial regulator of macrophage pro- inflammatory responses, including production of pro-inflammatory cytokines, phagocytosis, and antigen presentation, through modulation of mitochondrial ROS production, autophagy, and protein processing.

Abstract:
This thesis overviews my experience during graduate school making tangible, object oriented sculpture. I have been working formally to compose space in a way that develops a narrative between parts. The work is also a bridge between the fields of painting and sculpture, in terms of drawing with form and both painted and local, material color. My palette has mostly consisted of bronze casting, steel fabrication, fiberglass and epoxy resin, paint, the found object, woodworking, and mold making. This work is also conceptually based in showing the hand worked qualities of the materials, the transfer of meaning through casting, and my emotional relationship with the various parts of the sculptures. The three major themes of the work are: divine love and the complex of the apocalypse, the complexities and psychology concerning the relationship between a man and a woman, and the intrigue, potential energy, and beauty of the systems mankind hasinvented to harness the atom. The major artistic influences for this body of work have been: Jasper Johns, Marcel Duchamp, Constantine Brancusi, Alberto Giacommetti, Reg Butler, Henry Moore, Lynn Chadwick, Kenneth Armitage, Jeff Koons, Terry Winters, William DeKooning, Richard Diebenkorn, David Smith and Charles Long. I retain a strongrelationship with the movements of Dada, Surrealism, Futurism, and Assemblage, and amalso currently involved in solidifying the Manifesto of Raubeaux with a small group ofesteemed colleagues.

The purpose of this phenomenographical case study is to explore the ways in which a group of MFA students conceive of their learning as they are enmeshed within an MFA community. The research follows along two guiding research questions: 1) What does artistic learning involve for graduate students in an MFA community? 2) How is one's artistic practice shaped by one's active participation in an MFA community? The findings of this study have been presented as lines of artistic learning and help to show the various conceptions that MFA students have of their learning as artists while in an MFA program of study. Ultimately, it is in better understanding one's lines of artistic learning that MFA students can be better supported in their journeying to become professional, practicing artists.

Abstract:
This is the thorough examination of Allyson Malandra's process of becoming a character, and the many different challenges and triumphs she has faced throughout the past three years of developing her craft. It also aims to bring into focus where the work will be headed throughout the many years to come. The goal of this process paper is to examine the many different details and intricacies that go into a role, and also how to troubleshoot problems that may arise throughout the journey. Our process is how we, as actors, attempt to create life on stage, and discover and unfold the underlying truth that connects all human beings. It is understood that each role for an actor will present a different set of chges, and therefore it is important to understand a baseline process that gets at the core of the actor's work. Through examination of this process, previous and current chges in the work will be discussed, as well as ways to move through these issues. Ideas on what is valuable in acting will be discussed, as a way of staying centered and focused, and also retaining quality in the craft. The examination will also explore three main pillars of the work--breathe, listen, and play--and how these anchor Allyson's artistry. The ultimate goals of this examination are to create a support for the process, to chge her work in the future, and to help her continue to grow and expand, both as an artist and as a person.

Schultz, Heath. "Becoming-professional: notes on the university and the production of MFAs." Thesis, University of Iowa, 2011. https://ir.uiowa.edu/etd/2767.

Full text
Abstract:
This paper begins by looking at the MFA as a worker within the context of the contemporary university and from there attempts to situate that position in relationship to capitalism by charting out how the university uses workers for its own ends much like any capitalist business would, which results in the over-producing MFAs. From here, we can look toward the broader consequences of this large production of cultural producers and their becoming-professional. The consequences of this becoming-professional, I argue, are much more problematic than they initially appear, which further destabilizes our ability to act as anti-capitalist and anti-authoritarian cultural producers without further strengthening the forces we seek to oppose. Finally, I'll try and develop Stefano Harney and Fred Moten's concept of the criminal as well as Deleuze & Guattari's thinking on smooth spaces and a socio-political shifting toward that of the control society. Last I look at the various ways of thinking about fleeing or evacuating to help us chart escape routes by moving past traditional artistic notions of institutional critique and other professionalizing discourses learned within the spaces of MFA production.

Abstract:
Mitochondria continuously undergo changes in their morphology by two dynamic processes called mitochondrial fusion and mitochondrial fission. Mitochondrial fusion results in longer mitochondria and is important for the complementation of mtDNA and intermixing of essential mitochondrial proteins and nutrients to maintain healthy mitochondrial population. Mfn1 and Mfn2 are involved in outer mitochondrial membrane fusion while OPA1 mediates inner mitochondrial membrane fusion. On the other hand, mitochondria are broken into smaller units for easy transport and removal of damaged mitochondria by Drp1. Defective mitochondrial dynamics has been linked with various common neuropathies and neurodegenerative diseases. Charcot Marie Tooth Type 2A (CMT2A) and its subtype Hereditary motor and sensory neuropathy type VI (HMSNVI) are caused by mutations in Mfn2 and result in progressive loss of distal motor and sensory neurons of peripheral nervous system. However, the pathomechanism of Mfn2 mutations and specific degeneration of peripheral motor and sensory neurons is still unclear.

Foster, Helen Cecilia. "US apparel imports from China in the context of MFA IV." Diss., Virginia Tech, 1990. http://hdl.handle.net/10919/39908.

Full text
Abstract:
The purpose of this research was to analyze US apparel trade building up to and following the MFA IV renewal in 1986, so as to gain insight into the reasons for US apparel industry support of the new fiber inclusions, and to gauge effects of MFA IV on US apparel imports overall and specifically from the PRC. The study focused on the period 1978 - 1988. Total imports from all suppliers (World) were examined by country of origin: China (PRC), and rest of world (ROW); fiber content: MFA IV-fiber or all fibers and whether knitted or woven construction. The imports were then examined to determine the impact of economic variables such as the exchange rate, and US personal apparel consumption expenditure (PCE). Imports were found to have increased from 1978 to 1987 and then to have declined in all categories except one from 1987 to 1988. The MFA IV, implemented in August 1986 was believed to have caused a reduction of import volumes after an initial lag period (expected in international trade). However, other factors were implicated in the trade reduction primarily the fall of the dollar. The dollar was strong through the early 1980s until its peak in 1985, it then declined for the rest of the study. The one category which continued to increase after the trade restriction was items of MFA IV woven apparel. The reason for this continued increase is not clear but it is believed to be the absence of specific restriction, i.e., pure silk had not been included in bilateral agreements so it is possible that suppliers were switching the product mix to increasing quantities of the less restricted group. Also this would have been the case if the items were coming from new and/or unrestricted suppliers. In conclusion, it is felt that the apparel import rate or increase was the primary concern and the factor that determined action not the actual volume of imports as the MFA IV-fibers were increasingly used in apparel production.

Abstract:

Ciacci, Luca <1984&gt. "Integration of MFA and LCA methodologies: the anthropogenic aluminium cycle in Italy." Doctoral thesis, Alma Mater Studiorum - Università di Bologna, 2013. http://amsdottorato.unibo.it/5524/.

Abstract:
MFA and LCA methodologies were applied to analyse the anthropogenic aluminium cycle in Italy with focus on historical evolution of stocks and flows of the metal, embodied GHG emissions, and potentials from recycling to provide key features to Italy for prioritizing industrial policy toward low-carbon technologies and materials. Historical trend series were collected from 1947 to 2009 and balanced with data from production, manufacturing and waste management of aluminium-containing products, using a ‘top-down’ approach to quantify the contemporary in-use stock of the metal, and helping to identify ‘applications where aluminium is not yet being recycled to its full potential and to identify present and future recycling flows’. The MFA results were used as a basis for the LCA aimed at evaluating the carbon footprint evolution, from primary and electrical energy, the smelting process and the transportation, embodied in the Italian aluminium. A discussion about how the main factors, according to the Kaya Identity equation, they did influence the Italian GHG emissions pattern over time, and which are the levers to mitigate it, it has been also reported. The contemporary anthropogenic reservoirs of aluminium was estimated at about 320 kg per capita, mainly embedded within the transportation and building and construction sectors. Cumulative in-use stock represents approximately 11 years of supply at current usage rates (about 20 Mt versus 1.7 Mt/year), and it would imply a potential of about 160 Mt of CO2eq emissions savings. A discussion of criticality related to aluminium waste recovery from the transportation and the containers and packaging sectors was also included in the study, providing an example for how MFA and LCA may support decision-making at sectorial or regional level. The research constitutes the first attempt of an integrated approach between MFA and LCA applied to the aluminium cycle in Italy.

Abstract:
This post-qualitative inquiry project investigated subjectivity (sense of self) among graduates of creative writing Master of Fine Arts (MFA) programs. The project asked how subjectivity is involved in the creative writing process and how that process fuels further writing after a creative piece (such as the MFA thesis) is completed. A post-qualitative, thinking-with-theory approach was used to explore the role of subjectivity among four anonymous graduates of creative writing MFA programs who provided writing samples describing their creative writing processes. Following the thinking-with-theory approach, the data were analyzed using Deleuze and Guattari’s theory of productive desire. Study findings are presented in two formats. First, a traditional, qualitative presentation of findings describes how unconscious desires develop a beneficial weakening of subjectivity that may encourage creative writers to continue writing after completion of the MFA degree. Next, further findings are presented via a nonlinear, rhizomatic data assemblage. The project concludes with recommendations for the use of Deleuzian productive desire as a pedagogical framework in graduate-level creative writing courses, as well as a call for the consideration of post-qualitative research methods in the field of education.

Lin, Junjun. "Molecular Regulation of a Novel Pro-Survival Bnip3 Spliced Variant NIPLET in Cardiac Myocytes Functionally Couples ER and Mitochondria." Circulation, 2015. http://hdl.handle.net/1993/31215.

Full text
Abstract:
Abstract Alternative splicing provides a versatile mechanism by which cells can generate proteins with different or even antagonistic properties. Herein we describe a novel splice variant of the hypoxia-inducible death gene Bnip3. Sequence analysis of the new Bnip3 protein revealed an N-terminus that was identical to Bnip3 but contained an Endoplasmic reticulum (ER) retention motif within the C-terminus, therefore we designated the new Bnip3 isoform NIPLET for (Nip-Like ER Target). While Bnip3 was predominately localized to mitochondria and promoted mitochondrial perturbations and cell death, NIPLET was preferentially localized to the ER and opposed the cytotoxic actions of Bnip3. Interestingly, NIPLET suppressed mitochondrial injury from Bnip3 activation and mitochondrial permeability transition pore opening by a mechanism dependent upon the dynamin motor protein Mitofusin-2 (MFN2). Notably, mutations of NIPLET within the critical ER retention motif rendered NIPLET defective for interacting with MFN2 and suppressed necrosis induced by Bnip3 or hypoxia. Hence, our findings reveal a novel signaling pathway that functionally couples ER and mitochondria for cell survival to a mechanism that is mutually dependent and obligatorily linked to a novel BNIP3 protein in cardiac myocytes.

Abstract:
In the course of studying painting for the past three years at the University of Iowa, I have found collaborating with other artists to be a great way for me to try on different hats. Two of these collaborations in particular, The Old Man Study Group with Hamlett Dobbins (Memphis, TN) and The Coracle Drawing Club with David Dunlap (Iowa City, IA), have given me the license and opportunity to pretend to be someone else. These collaborative projects have asked me to consider, and at times adopt, even if only for a moment, the interests and concerns of another maker. A few months into these two projects, I noticed that the work I was making on my own, in the isolation of my own studio, was suddenly open to the world's innovations, and not just my own.

Abstract:

Abstract:

Abstract:
North Atlantic Black is a collection of contemplative, lyrical poems that explore issues of coming out, suicide, yearning, and male relationships. Woven together, North Atlantic Black moves through different questions of masculinity encountered by the poet through the process of coming out. Early poems explore themes of masks, of theater, and of dressing and costume as means of escaping the traditional bounds of masculinity North Atlantic Black further braids in concepts of home, how they relate to identity through heritage and expectation, and how they inform the poet’s thoughts on what it means for men to have relationships—how ideas of masculinity have imposed on the poet’s life, and weigh on the relationships he wishes to pursue. Throughout, the moody colors of the Maritimes and the North East, of sealing ports and cold, forested mountains, loom over these confessions and contemplations.

Abstract:
Kayaking is a watersport activity that involves paddling performed within leisure purposes. Although kayaking provides pleasure to the practitioners, there are some adverse environmental issues concerning the site used to perform kayaking due to the equipment. This paper identifies and analyzes the lifecycle stages in which negative environmental impacts are generated. Melker of Sweden is an outdoor company specialized in delivering high-quality kayaks. This study aims to present an overview of the current environmental performance of Melker of Sweden’s kayaks. For this purpose, two environmental assessment tools are applied: material flow analysis and life cycle assessment. The Material Flow Analysis (MFA) results show that the transport of material to the manufacturing unit generates a considerable amount of emissions. Additionally, hull manufacturing and assembling accessories were found to be the least resource-efficient operation among all. The Life Cycle Assessment (LCA) results identify the transport of material and the manufacturing phase as the primary sources responsible for environmental impacts. On the one hand, the use of epoxy resin and gel coat is the root cause of high contribution of the manufacturing phase. On the other hand, the use of flax fiber found to be the least contributing material to adverse environmental impacts. This report also presents a list of recommendations regarding the import of material, the efficacy of the manufacturing operations, the type of raw material, and waste treatment alternatives.

Abstract:
Cellular metabolism is one of the most fundamental systems for any living organisms, involving thousands of metabolites and reactions that forms large interconnected metabolic networks. Proper and comprehensive understanding of the metabolism in human cells has been a field of research for a long time. One of the key parameters in understanding the metabolism are the metabolic fluxes, which are the rates of conversion of metabolic intermediates. Currently, one of the main approaches for determining these fluxes is metabolic flux analysis (MFA), in which isotope-labelled compounds are introduced into the system and measured. Mathematical models are then used to calculate a prediction of the systems flux configuration. However, the current paradigm of MFA lack established methods for validating that a model can accurately predict quantities for which there are no experimental data. In this study, a model for the central human metabolism was created and evaluated with regards to the model’s ability to predict a validation dataset. Further, an uncertainty analysis of these predictions were performed with a prediction profile likelihood analysis. This study has conclusively shown that MFA models can be validated against experimental data that the model has never seen before. Additionally, such model predictions were shown to be observable with a well determined prediction uncertainty. These results shows that a systematic validation of MFA models is possible. This in turn allows for a greater trust to be placed in the models, and in any conclusions that are based on such models.

Abstract:
In the last decades, many tools have been developed to understand and manage the anthropogenic cycles of materials, with different approaches. Each handles the material flows in society in different ways and each possesses its respective databases that fuel their uses. Yet there seems to be no common ground of communication between design activities and cycling activities, as well as their respective stakeholders, which hinders the information exchanges required for a proper management of discarded products (and their materials). This thesis provides two original contributions to circular product design: a tool for the integration of material circularity in product design and a framework to characterize material cycling networks. The tool is composed of a multi-criteria indicator for circular material value that is used in the Design for Material Circularity method. The framework is based on an extensive literature review enhanced by industry experts’ interviews and provides a basis for data collection and knowledge capitalization on cycling activities. The open-loop recycling networks of eight materials, from the three main material classes, are characterized using this framework (steel, aluminium, copper, precious metals, specialty metals, rare earth elements, plastics and glass). Two case studies detail the deployment of these contributions. The first focuses on the optimal choice of material and end-of-life scenario for a 1,5-litre bottle container. The second is aimed at identifying material circularity hotspots and ideal end-of-life scenarios for a vehicular lithium-ion battery pack

In my research into interdisciplinary choreographic processes, I found there to be a lacking representation of recent scholarship dedicated to exploring the relationship between literature and dance. As a dance theatre choreographer who often utilizes textual sources as impetuses for artistic creation, I have employed methods of traditional dramaturgy in my practice and, in seeking scholarship on this subject, have noted a need for clear examples of dance dramaturgy in practice. In this thesis study, I employed methods from literary studies and dramaturgy in the process of adapting a work of short fiction by Kurt Vonnegut for the stage. I documented my process and have structured the information for the benefit of those who may read it. The artistic product of this research was presented to an audience in direct relation to its literary source. At the conclusion of this research, I posit that dance can function as literary criticism.

Abstract:
Mitofusin 1 and 2 are outer-mitochondrial membrane proteins that have been shown to be involved in fusion. Mitofusin 2 has also been associated with apoptosis and development. When Mfn1 and Mfn2 were each conditionally knocked out from the cerebellum, Purkinje cells in Mfn2 deficient cerebellum during development had undergone neurodegeneration. Mutations in Mfn2 have also been associated with the Charcot Marie Tooth Type 2A (CMT2A). We want to asses the effect Mfn2 and Mfn1 might have on the development of other regions of the brain such as the telencephalon. We generated Mfn1 and Mfn2 conditional knockouts in the telencephalon by crossing them with Foxg1 Cre - a cre expressed in the telencephalon. We found that Mfn1 deficient mice have lost their corpus callosum at the midline, but survive over 6 months with a decrease in progenitor cells postnatally. Mfn2 deficient mice die between P9 and P12 with a decrease in progenitor cells postnatally and a decrease in number of neurons in the cortex. Therefore, our results suggest that Mfn1 and Mfn2 play a significant role in the development of the telencephalon.

This paper offers a contextualization of my art practice, within particular trends in contemporary art and its discourse. It traces an expressed interest in networked objects, and the indeterminacy of meaning in art; from researched examples and texts, through specific expressions of these ideas in my thesis art exhibition. The paper then outlines key areas of practical interest to the experimental viability of my art practice. The paper goes on to explore the relationships between indeterminacy, experimentation, and creativity in the arts; within the context of an original thought experiment that draws an analogy between topics in physics and the human mind. Non-rational art ideas are lauded for their uniquely explorative potential within this conjectural model. Optimism, and a will to think experimentally, are offered as the abiding principles of my art practice; as well as being universal tools that all human beings can depend upon.

Abstract:

Full text
Abstract:
The purpose of this research study aimed to critically examine personal and professional issues related to a small group of visual arts MFA graduate students and myself as we participated in a semester-long course entitled, “Issues of Relevance and Character in the Fine Arts,” at a large public university in the United States. In the form of a colloquium series, the course aimed to explore a graduate student’s developing sense of self and its impact on the different roles he or she often embodies while pursuing an MFA in a School of Art. These roles include those of an individual, a student, an artist, a teaching assistant, and a future professional in the field of the visual arts. Employing a participatory action research methodology, I was also a participant in the study. I regularly documented the oral, written, and visual data that emerged from the participants’ considerations of their immediate circumstances both inside and outside the School of Art as well as their interactions with the reflexive pedagogies utilized in the colloquium sessions. This dissertation also reviewed research studies conducted on graduate student development, students in visual arts MFA programs, the historical development of the MFA degree, as well as faculty members in MFA programs and their curricular and pedagogical practices. Findings indicated the following: First, strong convictions seemed to be intrinsic to the participants’ pursuit of their MFA degrees. They appeared eager to learn about becoming a professional in the field, and each participant expressed interest in teaching the visual arts as instructors in higher education. Second, participating in a course such as “Issues of Relevance and Character in the Fine Arts,” seemed to offer the participants a receptive and reflective platform to convey the voices of the “characters” they embodied as MFA students. For example, the participants appeared to clarify intra- and interpersonal priorities, educational goals and artistic aspirations. Their personal and professional development was influenced by the complex relationships they shared with others in their MFA programs. The MFA participants grappled with the connections and disconnections that appeared to exist between their professors, peers, and themselves. A third theme addressed the impact of my participation within the study. With an established background in both teaching and taking courses in the studio arts and Art and Visual Culture Education, the findings suggest that I was able to empathize with the three other participants on several fronts. Fourth, the findings also address the curricular and pedagogical strengths and limitations of the course. The implications of this research study suggest the need for (a) more action research studies of MFA graduate students, as the methodology seemed to enhance the reflexive and exploratory nature of self-inquiry; (b) a series of required courses within MFA curricula that help graduates to systematically reflect upon their roles as graduate students in terms of their immediate goals, as well as their aspirations for the future; (c) research and professional development opportunities for faculty members so that they might become more aware of the ways that their MFA curricula and instructional practices influence a graduate student's sense of self both positively and negatively; and (d) research on how art and visual culture educators can be involved in assisting MFA graduate students and faculty members develop new perspectives related to their instructional and mentoring practices.

As a result of the complexity increase in current industrial processes, requests for treatment of nonlinear systems have been overburdening modern control techniques. The modern control theory is based on a model to represent these processes, however complex models can result in a complicated and difficult controller to maintain. Data-Driven Control techniques are getting featured in areas where the system complexity, or the absence of a model, can be overcame by a lot of available data, which can be used to calculate the control signal directly. Among Data-Driven Control methods, the Model Free Adaptive Control (MFAC) technique stands out for characteristics, such as being on-line, using just input and output data from the plant and reference signal, as well as having formulations for systems with varying degrees of non-linearity. However, the MFAC is still making unanswered questions, such as the choice of controller parameters. The tuning of these parameters can be transformed into an optimization problem, nevertheless, a control project usually involves multiple objectives to be attended. Therefore, this work will define strategies and a multi-objective evolutionary algorithm, based on differential evolution and directed immigrants, to adjust the MFAC controller parameters. Several cases will be evaluated and two adaptive strategies for these parameters will be implemented: An off-line strategy, at which the parameters are optimized in all acting period, and another on-line, where the controller uses the optimized parameters obtained in the previous strategy performing optimizations at smaller intervals when some situations are detected. The results obtained through simulations suggest that the controller with optimized parameters off-line is better than parameters found in the literature. In addition, the proposed on-line strategy has been able to improve or at least maintain the benefits of off-line optimization.

Abstract:
Textiles and apparel trade has been governed by the Multi-Fiber Arrangement (MFA) for three decades. Trade restrictions have generated substantial welfare losses and price wedges in exporting and importing countries through trade distortions. Beginning in 1995, textiles and apparel trade underwent fundamental changes in trade flows and patterns. The World Trade Organization’s Agreement on Textiles and Clothing (ATC) aimed to remove all MFA quotas by January 2005. This study established an equilibrium displacement model to investigate the impact on textile and cotton sectors of different countries and country-groups of removing the MFA quota. The model specifies the basic linkages of textile and cotton markets in the United States, China and four other country-groups. With different assumptions about U.S. textile supply elasticity, foreign cotton exporters’ reaction and changes in the U.S. farm program payments, alternative scenarios are simulated to predict changes in domestic and import demand for textiles and apparel, import demand for U.S. cotton, domestic and import price of textiles and apparel, U.S. cotton price and adjusted world cotton price. Uniform distribution was imposed for selected parameters involved in the model to overcome the deficiency of equilibrium displacement models of assuming certainty of known related parameters. Results indicate increased import demand for U.S. cotton by China, higher U.S. cotton supply, more textile and apparel supply from China, decreased domestic demand for U.S. cotton, and lower U.S. domestic demand for textiles and apparel. However, prices of both textile and cotton markets experience both positive and negative changes under different scenarios. Holding other assumptions unchanged, when farm program payments increase, U.S. cotton price and adjusted world cotton price declined. When farm program payments are held constant, prices rise. The changes expected in U.S. cotton price are, in absolute value, greater than those of the adjusted world price.

Abstract:
My work explores the gray area and the shifting border between "us" and "other." It investigates the helplessness and the submissiveness on both sides of this spectrum and one's passivity that makes the "pain of others" inevitable. Moreover, it examines the individual and collective experiences of guilt and complicity in relation to world events. I am interested in the selective and repressed memories of individuals and nations, the reluctance to look and the ability to forget.
