Abstract:
The goal of this diploma thesis is to assess the economic efficiency of the investment consisting in the opening of the new franchise retail network stores, specially Tesco Franchise Stores CR s.r.o. The thesis consists of two main parts - the theoretical part and the analytical part. The theoretical part describes the basic concepts which are related to the investment activities and the methods used to evaluate the investment plans. The ending part of the thesis describes the retail environment and the principles of franchising. The beginning of the analytical parts deals with the strategic analysis and description of the franchise model of the company Tesco Franchise Stores CR s.r.o. In addition, the analytical part focus on a specific investment plan, which is further specified and predicts the expected revenues and costs based and the information obtained from strategic analysis and the financial statements of the company. The end of the analytical part deals with the evaluation of the investment plan using dynamic methods.

The thesis is focused on the statistical analysis of the continuity of power distribution in a specified part of the distribution network. Its aim is to explain the basic concepts and calculations in reliability distribution of electrical energy, which is an essential element for improving the quality of electricity supplied to end customers. The work is divided into two parts (theoretical part and a practical part), which are further divided into chapters and subchapters according to individual areas of focus. In the introduction the explains the aim and content of the of chapters work. You are also given a specific issue and its explanation. The conclusion summarizes the results of the practical part. Abstract:
This master thesis deals with systems for detecting rhythmic structures of music recordings. The field of Music Information Retrieval (MIR) allows us to examine the harmonic and tonal properties of music, rhythm, tempo, etc., and has uses in academic and commercial sphere. Various algorithms are used in the detection of rhythmic structures. However, today, most new methods use neural networks. This work aims to summarize the current research results of systems for detecting beats and tempo, to describe methods of calculating and evaluating the parameters of music recordings, and to implement a program that allows comparison of available detection systems. The result of the work is a script in the Python language, which uses six different systems to detect the rhythmic structure of test recordings. It then checks the outputs of the algorithms according to the given reference and compares the given systems with each other using several evaluation values. It uses two datasets as a reference—one of them is publicly available and the other was created by the author of this thesis (including annotations, i.e., reference beat times, for the sample recordings). The program allows user to see the results in graphs and play any of the sample recordings with detected beat times.

Abstract:
The aim of this work was the analysis of air current from the right outlet, using the smoke method and the subsequent determination of a method for application. A further task was to establish a method for determining the upper and lower vertical routing, yaw angle (ß) and angle of inclination (?) of the air current. The experiment aims to approximate conditions as close to those experienced in the actual vehicle. After assembling a track with a soot blower (outlet) in the laboratory and defining the coordinates of the vehicle, the air current was subjected to visualization within the range of five positions of the vanes. Visualization was achieved by the use of technology for the smoke method. We used a light plane with a customized system of coordinates, a smoke generator for visualizing the air current being emitted and a camera for recording images. During our research we established the upper and lower cut off at the specific closures of the horizontal vanes of the soot blower. We determined a common analogy between the experimental track used in the experiment and the circumstances in the actual vehicle. We acquired the yaw angles (ß) and the angles of inclination (?) in the light plane and, after conversion, in the system of coordinates of the vehicle for all five settings of the vanes. The assessment of the given soot blower was undertaken according to the values achieved.

The aim is to produce parts of demanded shapes, dimensions and surface quality at affordable costs. Ceramics guarantee long terming durability and reliability. The evaluation of ceramics grinding tests from the point of view of cutting forces and surface quality of the machined faces are presented in the last part of the diploma thesis.

Abstract:
The master thesis consists of a theoretical part and a practical part. In the theoretical part jet-grouting technology is described briefly. Mechanical properties of jet-grouted piles were characterized. Various estimation methods of ultimate pile bearing capacity are described in this thesis – analytical calculation of ultimate bearing capacity for bored piles and estimation of ultimate pile bearing capacity by analysis of load-displacement curve defined by CHIN (1970; 1972). Load transfer method for piles and hyperbolic load-transfer curve are introduced. As a part of the load-trasfer method analysis, a method of estimating ultimate pile shaft friction called beta method is defined. In the practical part were evaluated several static load tests of jet-grouted piles and were constructed their load-displacemnt curves. Then reverse analysis of the results from static load tests was performed using load-transfer method. Then ultimate pile bearing capacity was estimated using analytical calculation of pile bearing capacity for bored piles and using analysis of load-displacement curve with method by CHIN (1970; 1972). By evaluation of load transfer method and beta method ultimate shaft resistence for each pile was estimated. Results and load-displacement curves were compared.

The thesis is focused on analysis and comparison of the company's internal project methodology and official project methodology PRINCE2. This thesis aims to evaluate which of the methodologies is more suitable for management of projects in Ahold. A comparative qualitative analysis is used for the comparison of the project methodologies. The analysis is mainly focused on comparison of project phases, processes and documents. Furthermore concrete project is used to substantiate how each of these project methodologies would influence its realization. Comparing internal methodology and PRINCE2 methodology it was found that the internal project methodology that respects the specifics of Ahold is better for project management than official methodology PRINCE2.

Abstract:
This thesis deals with the possibility of using impedance cardiography for calculating cardiac output. Kubicek’s, Sramek‘s and Sramek-Bernstein‘s methods are discussed here. These methods were applied to a data set, obtained by measuring on subjects with implanted cardiostimulators. The subjects’ heart rate was being changed by the programing of cardiostimulators. Thanks to this procedure the measured data were not affected by artifacts, connected with the heart rate change caused by a body stress, or other influences. An influence of heart rate on a cardiac output value based on the statistical processing of the data set was studied.

This thesis describes project management methodology implementation in a consulting company and examines the impact of this methodology on key success factors of the company. The theoretical part deals with issues related to project management implementation in a company and describes well-known methods and standards in this area while focusing on PRINCE2 methodology, which has been chosen for the implementation. The theoretical part continues by describing the process of creating system dynamics models, which allowed the key success factor models to be created as tools for evaluation of the methodology implementation impacts. The practical part of this thesis describes the background for the methodology implementation project and then the implementation itself in detail. The practical part continues by showing and describing the created key success factor models. In the end, the methodology impacts evaluation is carried out, analyzing changes in separate elements of the model breakdown.

Trichinellosis is a worldwide serious disease transmittable from animals to humans. The disease agent is Trichinella spp., which belongs to the phylum of parasitic nematoda called Nematodes. There are 12 genotypes of Trichinella currently known but the most important for the Czech Republic and neighboring countries are T. spiralis, T. britovi and T. pseudospiralis. Man can be infected by ingestion of raw or undercooked meat of infected animals. Treatment is very expensive and can be fatal, therefore a great emphasis is placed on prevention, especially control of animal carcasses which are intended for human consumption. The most famous method, also approved by law, is the digestive method, however there are many other methods for testing the presence of a parasite or for specification of genotype. This paper mainly deals with comparison of known and available methods of detection of trichinosis originators. Furthermore this paper gives character description of majorgenotypes of Trichinella.

The doctoral thesis describes the possibilities of numerical modelling of hollow core concrete panels, SPIROLL type. The commercial finite element program ANSYS 13.0 is used for the modelling. The focus is on the specific flexural support of the panel and on the distribution of the shear stresses in the panel ribs near the support. The goal is to valid actually used method for homogenization of the panel’s foreheads and its influence on the shear resistance by using four numerical models. The numerical models are compared and verified with two experiments. The first experiment is vacuum loading on the hollow core slab. Second experiment is about shear loading by applying the loading blocks. Four methods for design of hollow core panels are evaluated. It is the method based on main stresses, design according EN 1992 and EN 1168 and Yang’s method of prestress effects transfer. Based on the results, a method of considering flexural support for the evaluation of hollow core panels is proposed.

The first part of the thesis is a short description of induction motors, the evaluation method and the inventory of the observed motors related to own consumption of Nuclear power plant Dukovany. The second part of the thesis concerns the design of limits and criteria for evaluating the current state of these important motors. Finally, the thesis shows a short description of the electro monitoring system together with the description of the database environment created. In the supplementary materials of the thesis, there is a design of a protocol template for evaluation of the current state of the motor and a technical report template for regular evaluation in monitored areas MCSA (Motor Current Signature Analysis) of the state of the induction motors.

This master thesis deals with the analysis of the training program, which is implemented in a multinational outsourcing company. During the analysis of training program I am using knowledge of Kirkpatrick model and Success Case Method study, which are described in theoretical part. The aim of this thesis is to describe the processs of the training program (focusing on the description of leadership techniques) and evaluate this course. The basic premise of the thesis is that the training program has positive feedback in the company and it has also good reputation. I'm trying to confirm those claims through an evaluation of questionnaire survey and interviews with graduates of the training program. Evaluation will give me information about employee satisfaction with the organization and content of the course, with the knowledge and skills of the teacher, etc. The questionnaires are focused on identifying what the graduates have learned, how much of it they have used in practice and what they have accomplished thanks to those knowledge. To extend this information I used personal interviews, which provide me with specific examples of the use of leadership techniques in various situations in workplace. Finally, I summarize all the results and make recommendations that would increase the effectiveness of the course.

The pressure-time method is one of two methods of measuring the flow rate on large hydraulic structures applicable to IEC 60041, which is based on the temporal integration of the measured pressure difference and the formation of a water hammer in a closed pipe. The aim of this master thesis is to perform a literature review of this method and to evaluate the flow rate of the measured data. Furthermore, the thesis deals with determination of the sensitivity of the evaluated flow rate to the weights of individual pressure sensors and to numerical modifying of the measured pressures. The first part is made using MS Excel. The flow rate is always evaluates with only one pressure sensor and it is compared with the original flow rate. There is research, how absence of the sensor has an impact on the evaluated flow rate. In the second part of the determination of the sensitivity of the evaluated flow rate, various encroachment (signal smoothing, noise, time delay, frequency band removal) are performed of measured pressure signal in Matlab. Various surrounding influences or sensors failures are simulated.

Abstract:
The task of the master's thesis is introduction with the problems of traffic noise, focusing on noise from road traffic. There is a description what is the noise, how it is formed, its resources and what are the methods of measuring. The following are the various noise reduction measures such as noise barriers and low noise pavements. The main attention is devoted to the noise generated at the tire / road that is reduced by these pavements. The practical part describes the implemented measurements on individual sections at various locations in our country. The measurement was carried by slightly modified method CPX with reference tire directly at the vehicle. The main aim was to evaluate the measurement data, make comparison of different low-noise surfaces with each other and with the commonly used surfaces. Then quantification of the rate reduction of the noise emission for a given section and verification of input data for noise modeling. In conclusion, the obtained results are summarized and based on them were confirmed very good acoustic properties with the recommendation to continue with measurements in the future.

In this master’s thesis there are summarized fundamental findings about air flow velocity measurements and afterwards introduced the measuring track for automobile car vents, measured object and track arrangement and all used components. Velocity field measurement method is how wire anemometry, and apart from 4 main cases measurements of free air jet, there are velocity profiles in piping measured as an input for CFD simulation. Air vent evaluation criteria are described and their assessment is done for specified cases. Further, there is a CFD simulation presented with comparison to experimental data. Uncertainty of flow rate measured and flow rate setting is determined. Improvement points and encountered issues are presented. Visualization and evaluation program coded in MATLAB environment enables easy data display of CTA results and will serve further vent quality assessment.

Introductory part of this thesis concludes the development of air traffic, division of airports in the Czech Republic according to the Ministry of Transport, according to the Law on Civil Aviation and according to the Coding of the airports (Requirement L 14). The aim was to realize theoretical definition and division of output and economic indicators. After that comes choice of appropriate airports from the European Union. The choice has been made on the basis of the number of check-in passengers at the airport in Prague-Ruzyně. Eight most suitable airports have been chosen from all twenty-seven member countries of the EU. The following step is comparative analysis of these chosen airports on the basis of specified output and economic indicators for the years 2005-2009. The year 2010 has not been included, because the chosen airports did not complete their annual reports, balances and statements of profit and loss. From these sources the values for calculation of individual indicators have been chosen. These indicators have been shown in charts, evaluated by means of graphs in Excel Programme and further they have been characterized in words. The aim of the thesis was also the evaluation and comparison of output and economic indicators of effectiveness and after evaluation, proposal of methods of evaluations has followed.

Management projektů a jejich rizik je v současnosti rozvíjející se disciplína, která si získává stále větší pozornost a uplatnění v praxi. Tato práce popisuje úvod do problematiky řízení rizik, zkoumání metod jejich identifikace, vyhodnocení a managementu, předcházení jejich následkům a jejich zvládání. V další části práce byla provedena analýza vzorků rizik z reálných projektů, byly popsány metody pro identifikaci a vyhodnocení následků rizik v úvodních fázích softwarového projektu, taktéž byly popsány atributy rizik a navržen způsob jejich dokumentace. V závěrečné části zadání byl navržen a implementován prototyp modelové aplikace pro podporu managementu rizik softwarových projektů.

Kvítková L.: Evaluation of in vitro activity of potentially antifungal compounds by microdilution broth method - VIII. Over 250 new synthetized compounds divided into 16 chemical groups were tested by microdilution broth method for the establisment of in vitro antifungal activity. This method is based on the standard M27-A (Clinical Laboratory Standard Institute). RPMI 1640 medium buffered to pH 7.0 with MOPS was used and standardized inoculum was prepared from fresh fungal isolates grown on Sabouraud agar. All compounds tested were dissolved and diluted in DMSO. The MICs were determinated visually after 24 or 48 hours of incubation at 35C. The compounds were tested against eight fungal strains. The structure-activity relationships were studied and compared with the calculated log P values. Hydrophobicity of compounds (log P) was computed using the program ACD/LogP (Advanced Chemistry Development Inc., Ontario, Canada). The most active compounds were the derivatives of (E)-1-pyrazine-2- ylethane-1-one-thiosemicarbazone; the majority of the compounds showed moderate or high efficacy against all fungal strains tested. Research of antibacterial substances is one of the most important factor in pharmaceutic industry. The main reason is being the never stopping growth of bacterial resistence. Methods The microdilution broth metod was used to test the substances. This metod was used because of low difficulty and low cost. All the steps including the final analising was done by hand. Results The substances were divided in groups depending their chemical struction. The most effective was the salicylanilide derivates group. The other groups inhibitated growth of bacteria very little or were non-functional. In case of every bacterial stems were analised all substances which were effective. Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Pharmacy The aim of this thesis was to assess the antifungal activity of substances prepared at the Department of pharmaceutical chemistry and drug control, Faculty of Pharmacy of Charles University in Hradec Králové. Substances were tested through use of microdilution broth method at eight strains of pathogenic fungi: Candida albicans, C. tropicalis, C. krusei, C. glabrata, Trichosporon beigelii, Aspergillus fumigatus and Trichophyton mentagrophytes. Substances were divided into nine groups according to their common structural characteristics. The most effective were the derivates of 2-[(1-heteroarylethyliden) hydrazon]-1,3-thiazolidin-4-one with the lowest values of MIC. Derivates of 5,6- diazine-2,3-dicarbonitrile presented no activity. The rate of susceptibility for each tested strain was evaluated. The most susceptible were the Candida albicans and Trichophyton mentagrophytes strains. Objective: With the growing number of systemic fungal diseases and occurrence of resistant strains of fungi it is still necessary to conduct research of new potential drugs. The aim of the thesis was to determine antifungal activity in vitro with 70 substances, which were divided into six groups according to chemical structure. Methods: Antifungal effect was tested on 8 strains of pathogenic fungi, including 5 strains of yeasts and 3 strains of moulds. The microdilution broth method was used for testing. Results: The most expressive antifungal activity showed by a group of substances derived from salicylanilide. SAL-5-Cl-4-Br was the most effective substance, which had a good effect on Trichosporon asahii strain and it reached the lowest value MIC (0,98 µmol/l) at Trichophyton mentagrophytes from all tested substances. Total of 52 substances were tested. Substances were synthetisized at Department of Pharmaceutical Chemistry and Drug Control (Mgr. Jandourek, Mgr. Semelkova). All substances were pyrazine derivates, the substances were divided into six group. Sensitivity of the 8 strains was tested - Candida albicans, Candida tropicalis, Candida krusei, Candida glabrata, Trichosporon asahii, Aspergillus fumigatus, Absidia corymbifera, Trichophyton mentagrophytes. Methods: Microdilution broth method was used. Results: Total of 24 substances displayed antifungal effect, the lowest MIC was 62,5 µmol.l-1 . For testing of the antimicrobial activity was used microdilution broth method. This method is suitable for the quantitative determination of antimicrobial susceptibility. The substances were tested at eight bacterial strains, which consisted of Gram pozitive and Gram negative bacteria including resistant agents of serious nosocomial infections. 52 tested substances were dividend into 6 groups based on a common structure. We tested a totally 10 cathegories of substances: anilides of pyrazincarboxyl acid, arylaminopyrazines, pyrazin-2.3-dinitriles, pyrazincarboxyl acid esters, derivatives of (Z)-5-arylmethyliden-2-thioxothiazolidin-4-on, cholesterol and alkane acids esters, derivatives of thiosalicylamid , diamides, styrylbenzoxazoles, benzoxazepin-diones. Methods: The assesment was carried out using the microdilution broth method. Results: The most striking antifungal activity exhibit derivatives of thiosalicylamid for which MIC values were very low in almost all scale of concentration for all tested fungi. Conclusions: We found that the greatest antimycotic activity is manifested with the substitution of a halogen, mostly by chlorine.

For testing of substances was used the microdilution broth method. This method allows a routine assesment of MIC of a larger amount of antimycotics and also complete strains. The compounds for the strains Candida albicans, C. tropicalis, C. glabrata, C. krusei, Trichosporon beigelii, Aspergillus fumigatus, Absidia corymbifera a Trichophyton mentagrophytes were tested. Substances were tested by using microdilution broth method. Testing was carried out on eight strains of the bacteria: Staphylococcus aureus, Staphylococcus aureus methicilin resistant, Staphylococcus epidermidis, Enterococcus sp., Escherichia coli, Klebsiella pneumoniae and Pseudoonas aeruginosa. We have tested 42 substances, all of them were synthesized at the Department of Inorganic And Organic Chemistry of Faculty of Pharmacy in Hradec Králové. We have used the microdilution broth method for the evaluation. It is a standardize procedure for finding the minimal inhibitive concentration (MIC) of antibiotics (ATB) and thereby the quantitative sensitivity of bacteria to ATB. The principle of the method is the visual readout and evaluation of bacterial growth in wells of microtiter plate. The wells were filled with a live broth, an antibiotic substance and a microbe. The antibiotic effect of the substance was evaluated positive if it came to the visible inhibition of microbial growth in the well. We have tested derivative of sulphonamides and salicylanilides. During the experiment were tested two groups of compounds, derivatives pyrazin-2-carboxamide and salicylanilide. Using microdilution broth method were measured the activities of these substances against eight strains of filamentous fungi and yeasts, which included C. albicans, C. tropicalis, C. krusei, C. glabrata, T. beigelii, A. fumigatus, A. corymbifera and T. mentagrophytes. For determining the sensitivity of these strains of fungi against tested substances was prepared eleven different concentrations (500 mol∙l-1 , 250 mol∙l-1 , 125 mol∙l-1 , 62,5 mol∙l-1 , 31, 25 mol∙l-1 , 15,625 mol∙l-1 , 7,813 mol∙l-1 , 3,906 mol∙l-1 , 1,953 mol∙l-1 , 0,977 mol∙l-1 and 0,488 mol∙l-1 ), which let incubate with the fungi. Methods: The substances were tested by the microdilution broth method at eight strains of yeasts and filamentous fungi. The tested substances were divided into five groups according to their chemical structure: benzylamino substituted derivates of pyrazine-2- carboxamide, aliphatic aminoderivates of pyrazine-2,3-dicarbonitrile, cyclic aminoderivates of pyrazine-2,3-dicarbonitrile, aliphatic aminoderivates of pyrazine -2-carboxamide, cyclic aminoderivates of pyrazine-2-carboxamide. The thesis deals with evaluation of the projection of complex agrarian arrangement in a selected test set represented by the region of South Bohemia The theoretical part focuses on goals and financial measures of the land consolidation in coherence with the financial support programs. Furthermore, it mentiones some of the primary issues, for example the plan of joint facilities and field routes. The practical part determines the appropriate methods and sets leading to obtaining results both in written and in graphic form. Based on data from the districts of South Bohemia we will a) assess and compare the current state of projection with the percentage of both started and completed complex land arrangements in individual districts, b) asses the main reasons for the implementing these arrangements, c) asses the fragmentation of land in relation to land consolidation and d) asses the costs of land consolidation in individual districts. The goal of this thesis is to provide a comprehensive analysis of projected land arrangements and possible suggestions to improve the current state.
