Abstract:
Background:Mercury is a toxic metal that is present in small amounts in the environment, but its level is rising steadily, due to different human activities, such as industrialization. It can reach humans through the food chain, amalgam fillings, and other sources, causing different neurological disorders, memory loss, vision impairment, and may even lead to death; making its detoxification an urgent task. Methods:Various physical and chemical mercury remediation techniques are available, which generally aim at: (i) reducing its mobility or solubility; (ii) causing its vaporization or condensation; (iii) its separation from contaminated soils. Biological remediation techniques, commonly known as bioremediation, are also another possible alternative, which is considered as cheaper than the conventional means and can be accomplished using either (i) organisms harboring themeroperon genes (merB,merA,merR,merP,merT,merD,merF,merC,merE,merHandmerG), or (ii) plants expressing metal-binding proteins. Recently, differentmerdeterminants have been genetically engineered into several organisms, including bacteria and plants, to aid in detoxification of both ionic and organic forms of mercury. Results:Bacteria that are resistant to mercury compounds have at least a mercuric reductase enzyme (MerA) that reduces Hg+2to volatile Hg0, a membrane-bound protein (MerT) for Hg+2uptake and an additional enzyme, MerB, that degrades organomercurials by protonolysis. Presence of bothmerA andmerB genes confer broad-spectrum mercury resistance. However, merA alone confers narrow spectrum inorganic mercury resistance. Conclusion:To conclude, this review discusses the importance of mercury-resistance genes in mercury bioremediation. Functional analysis ofmeroperon genes and the recent advances in genetic engineering techniques could provide the most environmental friendly, safe, effective and fantastic solution to overcome mercuric toxicity.

Abstract:
Amyotrophic lateral sclerosis (ALS) is a devastating neurodegenerative disease for which there is currently no cure. Progress in the characterization of other neurodegenerative mechanisms has shifted the spotlight onto an intracellular structure called mitochondria-endoplasmic reticulum (ER) contacts (MERCs) whose ER portion can be biochemically isolated as mitochondria-associated membranes (MAMs). Within the central nervous system (CNS), these structures control the metabolic output of mitochondria and keep sources of oxidative stress in check via autophagy. The most relevant MERC controllers in the ALS pathogenesis are vesicle-associated membrane protein-associated protein B (VAPB), a mitochondria-ER tether, and the ubiquitin-specific chaperone valosin containing protein (VCP). These two systems cooperate to maintain mitochondrial energy output and prevent oxidative stress. In ALS, mutant VAPB and VCP take a central position in the pathology through MERC dysfunction that ultimately alters or compromises mitochondrial bioenergetics. Intriguingly, both proteins are targets themselves of other ALS mutant proteins, including C9orf72, FUS, or TDP-43. Thus, a new picture emerges, where different triggers cause MERC dysfunction in ALS, subsequently leading to well-known pathological changes including endoplasmic reticulum (ER) stress, inflammation, and motor neuron death.

Abstract:
Objective: To compare blood pressure readings obtained with two commonly used oscillometric monitors: Omron HEM 711 AC (OM) and Welch-Allyn 52000 series NIBP/oximeter (WA) with mercury sphygmomanometers (Merc) in subjects with atrial fibrillation. Methods: We recruited 51 hemodynamically stable subjects with atrial fibrillation. Fifty four subjects in normal sinus rhythm served as controls. Supine blood pressure readings in each arm were recorded simultaneously using one monitor and Merc. The second monitor then replaced the first and readings were repeated. Merc was then switched to the opposite arm, and both monitors retested. Apical heart rates were ascertained with a stethoscope. We used the averaged, same arm Merc readings as “gold standard”. Results: Automated blood pressure readings were obtained in all control subjects and in all but three of those with atrial fibrillation. Both monitors, and operators, noted a difference between apical and radial/brachial pulse rates: apical-recorded: Merc 6.1±15.0; OM 5.5±13.7; WA 10.0±21.2 beats per minute. Both monitors were accurate in controls: over 90% of readings were within 10 mmHg of averaged Merc, and both achieved European Hypertension Society standards. In subjects with atrial fibrillation, about one quarter of all oscillometric readings differed from Merc by more than 10 mmHg. Both falsely high and falsely low readings occurred, some up to 30 mmHg. There was no relation between accuracy and heart rate. Conclusions: Single blood pressure readings, taken with oscillometric monitors in subjects with atrial fibrillation differ, often markedly, from those taken manually. Health care professionals should record multiple readings manually, using validated instruments when making therapeutic decisions.

Abstract:
Abstract Background A bioassay with severely mercury-stressed duckweed (Lemna gibba L.) had revealed growth-inhibiting effects of homeopathically potentised mercury(II) chloride (Mercurius corrosivus, Merc-c.). We hypothesised that effects of potentised preparations are dependent on the stress level of the organisms used in the bioassay. The aim of the present investigation was to examine the response of duckweed to potentised Merc-c. at a lower stress level. Methods Duckweed was moderately stressed with 2.5 mg/L mercury(II) chloride for 48 hours. Afterwards plants grew in either Merc-c. (seven different potency levels, 24x–30x) or water controls (unsuccussed or succussed water) for 7 days. Growth rates of the frond (leaf) area were determined using a computerised image-analysis system for day 0–3 and 3–7. Three independent experiments with potentised Merc-c. and three systematic negative control experiments were performed. All experiments were randomised and blinded. Results Unsuccussed and succussed water did not significantly differ in their effects on duckweed growth rate. The systematic negative control experiments did not yield any significant effects, thus providing evidence for the stability of the experimental system. Data from the two control groups and the seven treatment groups (Merc-c. 24x–30x) were each pooled to increase statistical power. Duckweed growth rates for day 3–7 were enhanced (p < 0.05) after application of Merc-c. compared with the controls. Growth rates for day 0–3 were not influenced by the homeopathic preparations. Conclusions Moderately mercury-stressed Lemna gibba L. yielded evidence of growth-enhancing specific effects of Merc-c. 24x–30x in the second observation period (day 3–7). This observation is complementary to previous experiments with severely mercury-stressed duckweed, in which a decrease in growth was observed in the first observation period (day 0–3). We hypothesise that the differing results are associated with the level of stress intensity (moderate vs. severe).

Abstract:
Background: Mercuric chloride is known to inhibit the activity of enzymes. It is used in homeopathy at ultra low concentration (ULC) and is known as Mercurius corrosivus (Merc cor). ULCs of Merc cor are reported to promote enzyme activity. Objective: To see whether the mother tincture (?) of Merc cor and its ULCs interact with an enzyme invertase at its binding sites and influence enzyme’s action on its substrate sucrose. Methods: Merc cor ? (0.15 M HgCl2) was diluted with deionized and distilled (DD) water 1:100 and succussed 10 times to prepare Merc cor 1 cH or 1st potency. This potency was further diluted and succussed in 200 and 1000 steps to prepare 200cH and 1000cH potencies, respectively. Merc cor 200 cH and 1000cH were prepared in 90% ethanol. The two potencies and blank 90% EtOH were diluted with DD water 1:1000 to minimize ethanol content to a negligible amount 0.09%. The control was DD water (55mM). The drugs, EtOH and water control were mixed separately with 0.037 mM invertase in DD water. Using an isothermal calorimetry (ITC) instrument the substrate sucrose (65mM) was injected at 2 µl every 2 min into 300 µl invertase solution 20 times at 25 0C. Molecular modeling study was done to predict possible binding sites and nature of binding between the enzyme and HgCl2 and between the enzyme and water. Potencies after dilution are virtually water. Fluorescence spectra of invertase (4µM) mixed with drug/control solutions were also obtained to see the effect of drugs on protein folding. Results: Thermodynamic parameters like binding constant, change in enthalpy, entropy and Gibbs free energy showed marked variation in treatment effects on the enzyme. Molecular modeling study also shows variation in binding between invertase and HgCl2 and between invertase and water. Fluorescence spectra show variation in quenching related to different treatments. Conclusion: Merc cor mother tincture and its potencies interact at different binding sites of invertase and modify the enzyme’s action on sucrose. Drug solutions induce conformational changes in the enzyme.

Abstract:
Heterozygosity for the mouse lethal yellow (Ay) mutation leads to obesity, increased tumor susceptibility and increased activity of the agouti coat color gene; homozygosity for Ay results in embryonic death around the time of implantation. Although these pleiotropic effects have not been separated by recombination, previous studies have suggested that the dominant and recessive effects result from distinct genetic lesions. Here we use a combination of genomic and cDNA cloning experiments to demonstrate that the Ay mutation is caused by a 120 kb deletion which lies centromere-proximal to the agouti coat color gene. The deletion removes coding but not 5′ untranslated sequences for a ubiquitously expressed gene predicted to encode a protein similar in sequence to an RNA-binding protein, which we named Merc, for maternally expressed hnRNP C-related gene, but have renamed Raly, since the gene is nearly identical to one reported recently by Michaud et al. (Gene Dev. 7, 1203–1213, 1993). The Ay deletion results in the splicing of Merc/Raly 5′ untranslated sequences to agouti protein-coding sequences, which suggests that ectopic expression of the normal agouti protein by the Ay fusion RNA is responsible for the pleiotropic effects associated with heterozygosity for Ay. We find that Merc/Raly RNA is present in the unfertilized egg and is also transcribed in preimplantation embryos. Using a PCR-based assay to determine the genotype of individual embryos from an Ay/a × Ay/a intercross, we show that, in the absence of zygotic Merc/Raly expression, Ay/Ay embryos develop to the blastocyst stage, but do not hatch from the zona pellucida or form trophoblastic outgrowths. Injection of a Merc/Raly antisense oligonucleotide into non-mutant embryos blocks development prior to the blastocyst stage, and can be rescued by coinjection of a Merc/Raly transgene. These results suggest that maternal expression of Merc/Raly plays an important role in preimplantation development and that its deletion of is sufficient to explain Ay-associated embryonic lethality.

People have rampantly consumed batteries, thereby obtaining mobile, low cost and highly durable energy. However, many consumers do not recognize that these batteries may contain high level of heavy metals in their composition and so discard them inappropriately with their household trash daily. In order to minimize the problems caused by batteries to the environment, legislations have been created throughout the world. In Brazil, Resolution CONAMA 401 of 2008 established the maximum levels of toxic metals in batteries. This thesis was developed with the objective of verifying the levels of cadmium, lead and mercury in order to validate an analytical method for the determination of theses toxic metals in alkaline batteries. 193 samples of alkaline batteries collected from the Brazilian market between 2010 and 2011 were evaluated; none of the evaluated samples had cadmium levels above the allowed maximum (0.002%). However, 10 samples were found to have levels of mercury that exceeded the allowed maximum of 0.0005% and 8 of the analyzed batteries had levels of lead that were higher than the allowed (0.1%). The results obtained show a high percentage of failure (9.3%) of the samples analyzed, which indicates the necessity of controlling the use of toxic metals.

Abstract:
Rhus coriaria L. (R. coriaria), Arctium lappa L. (A. lappa) and Echium amoenum Fisch. (E. amoenum) are medicinal herbs with extensive traditional uses, covering anti-inflammatory skin therapy. The present research aims to investigate the anti-inflammatory effects of different extracts of R. coriaria fruit, A. lappa roots and the E. amoenum flowers in human keratinocytes (HaCaT cells), evaluating extracts prepared using different techniques. Firstly, the herbal extracts were prepared using different methods including cold extraction, maceration, and decoction. The extraction solvents used were water, ethanol, ethanol-water (50:50), acetone, ethyl acetate, and dichloromethane. In the first phase of this study the extracts of E. amoenum, A. lappa and the polar (water (WRC), ethanol-water (EWRC), ethanol (ERC) and macerated ethanol (mERC)) extracts of R. coriaria were tested. The cytotoxicity and IL-8 release using, respectively, MTT and ELISA assays, were performed after 6 hours of TNF-α (10 ng/mL) treatment. Based on these results, the mERC and the EWRC were selected as the active extracts and assessed for NF-κB driven transcription, following identical treatment conditions. A non-toxic extract of A. lappa (EWAL) was also tested for its effect on NF-κB signaling. According to the preliminary data obtained, R. coriaria showed the most consistent and promising effects in inhibition of lL-8 and NF-κB signaling. Therefore, starting from this phase of the research the main focus of the study was on this herb and selectively mERC and the EWRC as the active extracts. The challenged cells by TNF-α, under treatment conditions with mERC and EWRC, were analysed for ICAM-1, VEGF, and MMP-9 releases, as well as NF-κB translocation, by ELISA assays. In addition, both active extracts were investigated for the inflammation induced by UVB (40 mJ/cm2) and were chemically profiled through HPLC-UV-DAD analysis. In the second phase of this research, the lipophilic extracts of R. coriaria (ARC, EARC and DCMRC) were evaluated using the cytotoxicity and IL-8 release assays in TNF-α or PMA-treated cells. Based on these results, active extracts were measured for their effects on NF-κB driven transcription. Among the evaluated herbs, only R. coriaria extracts achieved anti-IL-8 activity. Although all the polar extracts of this plant inhibited the TNF-α-induced IL-8 release, just mERC and EWRC suppressed NF-κB activation, ICAM-1, and MMP-9 secretion. EWRC showed higher inhibition on ICAM-1 and MMP-9 with IC50s of 1.76 ± 0.24 and 1.24 ± 0.33 µg/mL, respectively (mean ± s.d.). On the contrary, mERC significantly decreased VEGF levels whereas EWRC did not show any effect. The HPLC-UV profile of the extracts revealed higher amount of anthocyanins in EWRC in comparison with mERC. mERC, which showed lower amount of anthocyanins than EWRC, blocked both UVB-induced IL-8 release (with IC50 of 7.61 μg/mL) and translocation of NF-κB (at 10 μg/mL), measured by ELISA assay. The R. coriaria acetone (ARC) and ethyl acetate (EARC) extracts significantly inhibited the IL-8 release (at 25 μg/mL), as well as NF-κB driven transcription (at 50 μg/mL), when induced by TNF-α. The dichloromethane extract of this herb failed to show IL-8- inhibitory activity against TNF-α. ARC (with IC50s of 25.77 μg/mL ) and mERC (at 50 μg/mL) showed a significant effect in inhibiting the PMA induced IL-8 release. ARC modulated PMA-induced NF-κB signaling with IC50 of 27.82 μg/mL. Our results suggest the potential positive effect of R. coriaria fruit extracts, mostly mERC, as preventive agent in the treatment of keratinocyte inflammation through their inhibitory effect on the production of skin pro-inflammatory mediators. Our findings on the promising inhibitory effects of R. coriria on NF-κB signaling seem to confirm the traditional use of this herb as a remedy to treat skin inflammatory conditions. Its use as skin anti-inflammatory agent deserves further investigation.

Abstract:
The sea was first a real geographical space in Chateaubriand’s life, the boundless playing field of his childhood. By unfurling the world at large under his gaze, the sea provided him with a singular mode of appropriating it. The author of the Mémoires d’outre-tombe claimed for himself the status of seaman, discoverer, and traveller. The maritime lexicon comes to him spontaneously. While he only spent seven years of his childhood by the sea, then subsequently stopped but briefly in foreign port of calls, he keeps on referring time and again to the sea, and incessantly reinstates it in his texts, thus elaborating a rich netwwork of echoes. His very style evokes the rhythm of the sea and its ever-changing harmonics. The reader always seems to hear resounding waves and backwash in the distance. It is as if the writer’s sensibility and imagination, bearing the stamp of a foundational landscape, had him perceive the world through the shades and against the backdrops peculiar to the shores he left. Chateaubriand anxiously wondered whether the Mémoires would remain readable to his posterity. But writing and the sea conjure up the same idea of eternity : they write in labile script everlasting songs

Abstract:
Land-based marine pollution is the most important and surely the most complex source of marine pollution. Its effects are particularly dangerous within the scope of closed or semi-closed seas. Parallel to a whole non restricting rules which set the guidelines of marine protection and the fight against land-based pollution, a few conventions determine the legal regulations and the means to abate land-based pollution. The consideration of the main international clauses displays the weaks of the worldwide reglementation applicable to land-based pollution. Necessary complement of universal law, regional law implement the general guidelines elaborated at the universal level into a geographicaly limited scope. Regional conventions supply mesures suited to the problems and the specificities of each sea. The formal and the substance similarities and differences which caracterize the methodes of prevention, reduction and abatement of land-based pollution are brought in fore through a comparative analyse of the different regional or subregional conventional systems specifs to each european closed or semi-closed sea. In order to complet the study, the existing interactions between european community law and regional law are emphasized through the analyse of rules related to water and air protection against pollution, which contribute to the prevention of european seas land-based pollution

Abstract:
Energy systems technology development has been born a new so-called “thermo-hydraulic”. This present thesis manuscript studies the new Thermo-hydraulic process CAPILI and its respective applications especially the production of electricity for houses and for Ocean Thermal Energy Conversion (OTEC). The performance of the new process CAPILI have been evaluated firstly on steady state; and secondly by a dynamic study based on equivalent Gibbs system which was performed to evaluate more precisely the performance of such engine and optimize its different components. Finally after the implementation of a simplified experimental CAPILI engine and the validation of the different results, the dynamic behavior and the performances of a small CAPILI-OTEC plant, of about 10kWe, have been simulated

The way in which indigenous people are represented in documentaries has radically changed within the last century. But "If there (still) is one overriding ethical/political / ideological! question to documentary filmmaking it may be, What to do with the people" (Nichols qtd. in Barbash and Taylor, 1997: p. 12). How can people and issues be represented appropriately? How can one make a documentary about somebody or something with a totally different cultural background to one's own without being unethical? The so-called expository documentary was the first prevailing documentary mode and tries to answer these questions with an authoritative voice-over commentary combined with a series of images that aim to be descriptive and informative. The voice-over approaches the spectator directly and offers facts or arguments that are illustrated by the images. It provides abstract information that the image cannot carry or comments on those actions and events that are unfamiliar to the target audience. This is exactly what some filmmakers reacted against - "to explain what the images mean, as if they don't explain themselves, or as if viewers can't be trusted to work the meaning out on their own. Indeed, the voice-over often seems to attribute a reduced meaning to the visuals; that is it denies them a density they might have by themselves" (Barbash and Taylor, 1997: p. 19). It is typical for the expository documentary style that the narrator speaks about or for other people. Some filmmakers see these voice-overs as "colonial, an enemy of the film, the voice of God" or even as "the (non-existent) view from somewhere" (Barbash and Taylor, 1997: p. 47).

Abstract:
Mercer Engineering Research Center (MERC) developed a neural network-based regression method for predicting maximum stress per flight values at four structural tracking locations on the United States Air Force HH-60G helicopter airframe using Individual Vehicle Health and Usage Monitoring (IVHMS) data. Maximum stress per flight is utilized when evaluating a failure criterion within the HH-60G service life analysis, so an accurate, fleet-wide estimation of maximum stress magnitude and likelihood is critical for accurate service life determinations. The model was trained using parametric flight data time histories (from IVHMS) and stress time histories from a strain survey aircraft. The stress time histories were developed from the strain signals using two different methods depending on the location of strain gauges in the vicinity of the tracking locations. For two of the tracking locations, they were derived from a global finite element model using a collection of strain gauge signals throughout the strain survey aircraft. At the other two tracking locations, the strain time histories were derived from single strain gauges installed in close proximity to the tracking locations. Multiple regression methods and input data configurations were evaluated in order to identify an appropriate regression method that predicts a maximum stress per flight accurately without over-fitting the training data. MERC identified that the relationship between parametric flight data and aircraft component strain can be exploited to a high level of accuracy using machine learning regression tools. Achieving a high level of accuracy required an extensive review of independent and dependent variable data quality and thoughtful consideration of model inputs.

Abstract:
Abstract The Metal Effectiveness Rotor Cooling (MERC) facility at the University of Oxford is a new blow-down linear cascade tunnel developed for aerothermal research of turbine rotor blade and rotor platform cooling systems. A high level of engine similarity is achieved with matched Mach numbers, Reynolds numbers, coolant-to-mainstream pressure ratios, and Biot number (using DMLS components of appropriate conductivity). The modular working section includes five blade passages, including platform and fir tree root geometries, and engine representative hub seals. The facility is designed to allow high accuracy IR camera measurement of the blade and platform surfaces (for metal effectiveness measurements) and downstream area traverse measurements. Detailed traverse measurements are possible in a single run, because of the long run time of the facility (up to 5 minutes). The facility is being used for development and optimisation research of novel blade and platform cooling systems, with an emphasis on overall thermal performance of parts (metal effectiveness). Modular cassettes allow the blade components to be rapidly interchanged, and for variations to the designs of the front and rear hub seals. The engine representative seals and coolant feed paths allow for all engine leakage flows to be replicated in the experiment. This is important, because they are influential in determining the platform cooling flow structure (character and extent of secondary flow) and overall metal effectiveness result. Coolant supplied to each of the hub seals, and blade shank pocket can be independently varied to achieve required mass flows and pressure ratios. Rotor blades are typically manufactured using DMLS, allowing fast development and optimisation of fully-featured cooling systems at significantly reduced cost compared to traditional casting techniques. To demonstrate the capability of the new facility, we present full-surface metal-effectiveness measurements of the rotor blade platforms (post-processed using high-accuracy infrared thermography techniques separately developed at the University of Oxford.) The purpose of this paper is to outline the capabilities of the facility, describe the prior work and research context which led to its development, and to demonstrate the accuracy of the measurement techniques employed by presenting typical measured data. An argument is made for the importance of including realistic hub seal leakage paths in experiments investigating platform cooling flows by illustrating their significant impact on overall cooling performance. The MERC facility is a response to the need to develop more advanced rotor platform cooling schemes for future engines.

Abstract:
The use of multiple external representations (MER) in the empowerment of executive functions (EF) was identified and characterized through a systematic mapping. EF are part of the human learning triad, contributing to the development of complex executive skills. Learners may have difficulty developing EF, and can benefit from MER. Data from 18 articles were extracted. Most MER systems used two external representations, verbal-textual and visual-graphical, through desktop applications. In relation to EF, the most considered component was working memory. We identified the need for further research that considers the effects of MERs on cognitive flexibility and inhibition, and the impact of mobile devices to the exhibition of MERs.
