Full textAcquisition of oxygenic photosynthesis by eukaryotes occurred through a unique primary endosymbiosis of a cyanobacterium within a heterotrophic ancestor that resulted in the emergence of the three primary lineages known as the Archaeplastida consisting of the Rhodophyceae (red algae), Chloroplastida (green algae) and Glaucophyta (glaucophytes)Recently, a paradigm shift in the acquisition of photosynthesis was proposed: the implication of an intracellular obligate pathogen in plastid establishment. This hypothesis, dubbed the Ménage-à-trois Hypothesis (MATH), specifically addresses the central issue of disconnected supply and demand of carbon at the time of plastid endosymbiosis, suggesting an active and direct role of Chlamydiales in the success of primary endosymbiosis, which would have provided many critical genes to the cyanobiont hosted in a common vesicle known as the chlamydial inclusion. The expression and efficient localization of specific genes, such as key transporters and glucan transferases, would have initiated the biochemical fluxes of symbiosis. MATH is supported by molecular, biochemical and phylogenetic evidence but remains highly controversial. The major criticism concerns both the interpretation of single gene phylogenetic trees and the existence of other contributions assigned to different groups of bacteria thereby questioning a specific role for Chlamydiales in the endosymbiotic process. This work aims to test the Ménage à Trois Hypothesis by first evaluating the chlamydial footprint in the evolution of Archaeplastida and then comparing this signal to analogous contributions from other lineages of the bacterial domain. A bioinformatic pipeline was designed to identify all lateral gene transfer (LGT) events between Chlamydia and Archaeplastida, for which a manual analysis of the trees confirmed the occurrence very early during the endosymbiotic process. We then compared this chlamydial signal in the Archaeplastida to control signals, to ensure the specificity of both the bacterial donors and the eukaryotic acceptors involved in these LGT

Full textIn response to various environmental stimuli, neurons undertake specific cognitive functions, such as learning and memory. Many studies have shown that epigenetics, notably histone acetylation, is crucial for the regulation of gene expression involved in these processes. Moreover, dysregulation of signaling pathways that regulate epigenetics is clearly recognized as a main actor in the pathogenesis of several neurological disorders. Even if the origin of these disorders is sometimes genetic, their etiology remains elusive and environmental factors are also suspected. In particular, infectious agents are considered as serious candidates, notably for viruses that persist in the central nervous system (CNS). The Bornavirus (BDV) constitutes an interesting model to assess the impact of viral infection on neuronal epigenetics. This non-cytolytic RNA virus infects neurons of the CNS and causes various cognitive disorders in animals. It has the remarkable property to replicate in the nucleus, in close association with chromatin, thus leading to our working hypothesis that it may modify the epigenetics of infected neurons. During my Ph.D., we used primary cultures of cortical neurons to demonstrate that BDV infection decreases H2B and H4 acetylation levels on selected lysine residues, through inhibition of histone acetyltransferase (HAT) activities. We also showed that the viral phosphoprotein was responsible for this perturbation, acting on histone acetylation and HAT activities in a protein kinase C dependent manner. Finally, using pharmacological agents, we observed that histone acetylation levels were correlated with replication of BDV. Thus, BDV action on histone acetylation pathway may represent an original mechanism to control viral replication, thereby favoring long-term persistence in neurons. I am currently examining the consequences of this virus-mediated perturbation on H2B/H4 acetylation at the genome scale, using ChIP-sequencing experiments

Full textA key characteristic of the process of economic development is the shrinking spatial mismatch between economic activity and population. This thesis analyzes what happens when this spatial mismatch is reduced, as people’s places of residence and work or the geographical distribution of economic activity is altered. Chapter I deals with the relationship between international migrants and their households of origin. It sheds light on the importance for remittances and investments of migrants' beliefs and the information asymmetry between remittance senders and recipients that distance aggravates. Chapter II explores the transformation of the receiving economy due to an influx of rural-to-urban migrants in China. It first quantifies the effect of immigrants on the labor market at destination, and then investigates their impact on the reallocation of production factors and factor-market constraints faced by urban firms. Chapter III focuses on the long-term impact of a large industrialization plan in China. It reveals a reversal pattern due to the distortions in local labor markets induced by the presence of big plants. It highlights the role of migration in overcoming such imperfections and in bringing about the later stage of structural transformation—from heavy industry to consumption goods and services

Full textIn my doctoral thesis, I investigate the information feedback from stock prices to managers’ decisions. More specifically, I study whether and how managers learn new information from stock prices to guide their corporate decisions. My doctoral thesis includes three essays focusing on this topic. The first essay studies the relationship between stock market informational efficiency and real economy efficiency at firm-level. In the first essay, I find that when stock prices reflect greater amount of information that managers care about, corporate decisions made by managers become more efficient. The second essay studies whether managers seek to learn short sellers’ information from stock prices and use it in corporate decisions. In the second essay, I overcome the empirical difficulties by exploiting a unique institutional feature in Hong Kong stock market that only stocks included in an official list are allowed for short sales. I find that that non-shortable firms’ managers can learn short sellers’ information on external conditions from shortable peers’ stock prices and use it in their corporate decisions. The third essay studies the real effects of long-term option trading. I find that long-term option trading stimulates the production of long-term information, which managers can use to guide their long-term investment decisions
