Full textI peptidi in difesa dell’ospite (HDPs) esercitano molteplici ruoli nell’immunità, agendo sia come molecole antimicrobiche ad azione diretta sia come agenti immuno-moduatori. Il ruolo all’interfaccia tra immunità innata ed adattativa li rende molecole ideali per la futura applicazione nel trattamento di malattie infettive. Lo scopo di questo lavoro è stato quello di valutare le caratteristiche funzionali e strutturali di Catelicidine e Defensine selezionate al fine di correlare queste proprietà con l’ attività selettiva su cellule eucariotiche e procariotiche. Metodi biofisici e biochimici sono stati applicati alla catelicidina umana LL37 ed alcuni analoghi (ortologhi di primate e peptidi artificiali) con lo scopo di studiare la loro struttura e aggregazione in contatto con membrane biologiche e modello (i). Lo stesso approccio è stato anche applicato alle defensine umane hBD2 e 3 ed a loro analoghi. Inoltre, tecniche di microscopia, quali la microscopia a trasmissione elettronica (TEM), la microspettroscopia infrarossa in trasformata di fourier accoppiata ad una sorgente di sicrotrone (µSR-FTIR) e la citofluorimetria, sono state utilizzate in modo complementare al fine di studiare l’interazione a breve termine di hBD2 con cellule presentanti l’antigene, in particolare le cellule dendritiche immature (ii). (i) Lo studio strutturale e l’interazione di membrana di catelicidine ortologhe ci ha permesso di scoprire come l’evoluzione abbia lavorato sulla sequenza dei peptidi inducendo una diversa capacità di strutturare nei diversi ortologhi. Questo ha portato ad un’interazione differenziata e specifica con le membrane e a diversi meccanismi di lisi di membrana cellulare e probabilmente diversi modi di interagire con le cellule dell’ospite. (ii) Inoltre, abbiamo individuato una rapida interazione di hBD2 con le cellule presentanti l’antigene. hBD2 sembra indurre un riarrangiamento generale dei lipidi cellulari che sembra comportare un aumento nella fluidità di membrana e una ri- organizzazione del sistema endomembranoso. Queste variazioni potrebbero essere responsabili di un cambiamento morfologico delle cellule che promuoverebbe la mobilità cellulare in risposta a stimuli esterni. Questo studio dimostra l’esistenza di un posibile meccanismo alternativo di motilità cellulare rispetto alla chemotassi recettore-mediata, indicando un meccanimo di azione di hBD2 sulle iDC più complesso rispetto quanto riportato fino ad oggi.

The interactions of terpenes with membranes of erythrocyte, fibroblasts, stratum corneum and the model membranes of 1,2-dipalmitoylsn -glycero-3-phosphocholine were investigated by using the the electron paramagnetic resonance and fluorescence spectroscopic of lipophilic probes. It has been shown that when added at high concentrations to systems having a high lipid/solvent ratio, terpenes such as 1,8-cineol, α-terpineol, (+)-limonene and nerolidol are able to self-stabilize in molecular aggregates which can extract the bilayers lipids. Studies on the hemolytic and cytotoxic potential of various terpenes showed that cell damage caused by these molecules are concentration dependent and that among the studied terpenes, nerolidol and α-terpineol are the most hemolytic and cytotoxic, while (+)-limonene and 1,8-cineole are the least hemolytic and cytotoxic. However, the low correlation between these two tests indicates that the processes involved in each case are not completely dependent. It was also shown that once embedded in the membrane, terpenes increase the fluidity of lipid bilayers and decrease the temperature of the main phase transition. Differences between increased fluidity promoted by sesquiterpene nerolidol and all monoterpenes studied were observed. Meanwhile, in a comparison of the effect of the monoterpenes studied, no significant differences in their ability to increase membrane fluidity were detected. Furthermore, it was demonstrated by using confocal and atomic force microscopy and fluorescence spectroscopy that the 1,2-distearoylsn -glycero-3-(Aurora nanoparticles) is better incorporated in lipid membranes under fluid phase and that the addition of 0.1% of these conjugated nanoparticles do not produces large variations in membrane fluidity and no causes substantial morphological changes of lipid bilayers.
As intera¸c˜oes de terpenos com membranas de eritr´ocito, fibroblastos, estrato c´orneo e membrana modelo composta de 1,2-dipalmitoil-sn -glicero-3-fosfocolina foram investigadas por meio das espectroscopias de ressonˆancia paramagn´etica eletrˆ onica e de fluorescˆencia por meio do uso de sondas lipof´ılicas. Foi poss´ıvel demonstrar que quando adicionados em altas concentra¸c˜oes `a sistemas que possuem uma alta rela¸c˜ao lip´ıdio/solvente, terpenos como o 1,8-cineol, α-terpineol, (+)-limoneno e nerolidol s˜ao capazes de se estabilizar em agregados moleculares capazes de extrair os lip´ıdios das bicamadas. Estudos sobre o potencial hemol´ıtico e citot´oxico de v´arios terpenos demostraram que os danos celulares causados por estas mol´eculas s˜ao dependentes da concentra¸c˜ao e que dentre os terpenos estudados, nerolidol e terpineol s˜ao os mais hemol´ıticos e citot´oxicos enquanto limoneno e cineol s˜ao os menos hemol´ıticos e citot´oxicos. Entretanto, a baixa correla¸c˜ao entre estes dois testes indica que os processos envolvidos em cada caso n˜ao s˜ao totalmente dependentes. Ficou demonstrado ainda que uma vez incorporados nas membranas, os terpenos aumentam a fluidez das bicamadas lip´ıdicas e diminuem a temperatura de sua transi¸c˜ao de fase principal. Diferen¸cas entre o aumento de fluidez promovido pelo sesquiterpeno nerolidol e por todos os monoterpenos estudados foram verificadas. Contudo, uma compara¸c˜ao entre o efeito dos monoterpenos estudados, n˜ao aponta para diferen¸cas significativas entre suas capacidades de aumento de fluidez. Al´em disso, foi demostrado atrav´es das microscopias confocal e de for¸ca atˆomica e da espectroscopia de fluorescˆencia que a 1,2-distearoil-sn -glicero-3-(Nanopart´ıculas Aurora) ´e melhor incorporada em membranas lip´ıdicas em fase fluida e que a adi¸c˜ao de 0,1% destas nanopart´ıculas conjugadas n˜ao produz grandes varia¸c˜oes na fluidez e n˜ao provoca mudan¸cas morfol´ogicas substanciais das bicamadas lip´ıdicas.

Full textChitosans are polysaccharides used in medicine, pharmacy, dentistry and in the inhibition of microorganisms growth (eg. as bactericidal agent). In these applications their action should depend on the interaction with cell membranes, which is difficult to verify because isolating a membrane is not easy. An alternative is to investigate the interaction with membrane models, such as a Langmuir film of phospholipids, from which information on the molecular level can be obtained. This dissertation evaluates the influence of the average content of N-acetylglucosamine units (GlcNAc) of chitosan and molecular interaction with Langmuir films of dipalmitoyl phosphatidic acid (DMPA). Chitosans with different average degrees of acetylation and low molecular weight were produced with the high-intensity ultrasound procedure. Chitosans affect the surface pressure and surface potential isotherms at large areas per molecule due to electrostatic and hydrophobic interactions with DMPA. In condensed films, they are located in the subsurface with little effect on the isotherms. The chitosan with fewer GLcNAc groups induced larger changes in the isotherms and in the film elasticity, probably due to stronger electrostatic interaction owing to a larger number of amine groups in chitosan interacting with the polar heads of DMPA. The most effective sample to induce changes in the DMPA monolayers was the low molecular weight chitosan, which can be attributed to the ease of adsorption. A minimum size chain seems essential, however, for mixtures of repeating units N-acetylglucosamine (GlcNAc) and glucosamine (GlcN) did not change the surface pressure isotherms and the elasticity of the DMPA films, owing to the absence of hydrophobic interactions. We conclude that the chitosan with better prospects for biological applications relying on the cell membrane interaction should have a low degree of acetylation and low molecular weight.

Full textThe study of amphiphilic aggregates is extremely important due to their cell membrane mimic, which are essential for the life of the cell. It is known that phospholipids do not have molecular structure well defined in membranes, but play an essential role in maintaining of their integrity. Zwitterionic phospholipids are one of the main components of cell membranes, and a simplified model for the membranes are the bilayers they can form in aqueous medium. The main characteristic of lipid bilayers is the self-organization of lipids, making it necessary to study natural and spontaneous process, as their structural and dynamical properties. The fluorescence spectroscopy has been used to study many processes and systems of biological interest, especially by measurement of fluorescence anisotropy, which gives information about the rotational dynamics of the fluorescent probe inserted in the systems of interest, reflecting the combined effects of flexibility, fluidity and static interactions with surrounding molecules. In this work we examined the structural and dynamic properties of phospholipid model membranes formed of 1,2-dipalmitoyl-sn-glycero-3-phosphocoline DPPC by techniques related to fluorescence spectroscopy, mainly by measurements of steady-state and time resolved anisotropy of the probes 1,6-diphenyl-1,3,5-hexatriene (DPH), 7-nitrobenz-2-oxa-1,3-diazol-yl (NBD) attached to different regions of phospholipid molecules and also the lipophilic probe 2-amino-N-hexadecyl-benzamide (Ahba). The measurements were perfomed above and below of the phase transition temperature of the phospholipid bilayers of DPPC, gel and liquid-crystalline phase, due to the difference in the lateral organization of hydrocarbon chains in these two phases. Measures of dynamic light scattering (DLS) was performed to confirm the formation of the unilamellar vesicles by extrusion of lipid suspension containing multilamellar vesicles, and the technique of differential scanning calorimetry (DSC) was used to verify if the low concentration of fluorescent probes in lipid vesicles affect its packing. From the results, we found that the behavior of the three different fluorescent probes differ in both phases of phospholipid bilayers, revealing their structural and dynamic properties, mainly because to specific locations of the fluorophores. We verify that, due to the affinity for the hydrophobic region, the motion of the DPH is restricted to the \"wobbling\" motion, limited by hydrocarbon chains. For the NBD labeled in lipids, the motion of the fluorescent analogues as a whole depends on the location of the fluorophore and on the lipid conformation in both phases of lipid bilayers. Because of the location of the fluorescent group of the probe Ahba in the interface of lipid bilayers, we found that its rotational motion increases as the bilayers becomes more fluid, showing a dependency of the motion with the microviscosity of these bilayers.

Full textSyftet med detta arbete är att undersöka om det är möjligt att bygga en modell över innerörat med dess basilära membran. Om detta skulle visa sig vara möjligt, är nästa frågeställning hur en sådan modell skulle kunna fungera pedagogiskt och uppfattas av elever.
Modellen som arbetet lett fram till är konstruerad som en låda, gjord av plexiglas, vilken skall motsvara en utrullad hörselsnäcka. Som membran har en uppsättning gummiband använts. Vidare aktiveras modellen med hjälp av en högtalare som får motsvara örats trumhinna. Det har visat sig att modellens membran beter sig på ett sätt som liknar det basilära membranet i hörselsnäckan. De vibrationer som bildas på modellens membran förflyttar sig, beroende på inkommande frekvens. Detta förlopp sker även på det basilära membranet.
Modellen har testats på en niondeklass som har fått utföra laborationer med hjälp av modellen. Eleverna har observerats under arbetet och har efter laborationen fått skriva en utvärdering, i form av en laborationsrapport och en enkätundersökning. Observation och utvärdering visar att modellen har ett pedagogiskt värde, men att det krävs förkunskaper inom området för att laborationen med modellen skall bli ett givande medel för inlärning.

Full textPhotodynamic Therapy (PDT) is a cancer treatment that has had great advances over the years and consists in the interaction between light and a photosensitizer compound, transforming the molecular oxygen in singlet oxygen, highly reactive and toxic to the cell. In this context, the use of Chorioallantoic Membrane (CAM) enables direct access to blood vessels, making possible to study the vascular effects involved in this therapy. The development of a tumor in this environment previously vascularized allows the study and the understanding of mechanisms that involves the growing and the destruction of tumor, improving different therapeutic modalities. The tumor cells used to develop the tumor in this model can be from several lines and ways of application, since cell culture to biopsy of some tumor. The main objective was investigating PDT in blood vessels and neoplasic cells in the tumor model in CAM. With this model studied for melanoma cells and Ehrlich, it was chosen to work with Ehrlich tumor because its facility of laboratory manipulation. After the observation the interaction between blood vessels and tumor cells, with confocal microscopy analysis, it was possible to understand and quantify the individualized effect of photosensitizers, including the pharmacodynamic of Photogem® and the Protoporphyrin IX from Aminolevulinic acid, both topic and intravenous application. From this, the tumor was illuminated and the Photdynamic Therapy effects was analyzed both tumor and blood vessels that feeds it. Still using the curcumin, photosensitizer derivated of saffron which has alone a vascular effect, was applied PDT to analysis on the Chorioallantoic Membrane and the blood vessels effect. The understanding of this effect of curcumin enables the extension of its useful as photosensitizer in the cancer treatment and vascular diseases. Therefore, with the model established, it was possible to follow all PS studying both their action in blood vessels and in the tumor region, causing damage.

Full textInteractions of the antimicrobial peptides with biological membrane models have been broadly studied to understand the function and the action mechanism of this class of peptides. Many efforts have been realized to increase the potency and the specificity of these peptides with the purpose of obtain more selective pathogen antimicrobials with decrease of the toxic effects and for a better explanation of the biological process concerned in the peptide action. The action mechanism approached to antimicrobial peptides concern the cellular membrane permeation by pore formation or other type of membrane disruption. A fundamental point in the knowledge of the activity is the distinct lipid composition of pathogen and host cells that is conceived as the principal key point in the selectivity of the antimicrobial peptides. The aim of the present work is to contribute for the knowledge of the action of the antimicrobial peptide indolicidin and some of its mutants by molecular dynamics simulation. The indolicidin is a short 13 amino acid residues antimicrobial peptide that was isolated from bovine neutrofils that have the function of ingest and kill pathogens. The action mechanism of the indolicidin is not yet known despite of numerous experimental studies realized with this peptide. The interaction of the indolicidin with the membrane models is important both for the knowledge of the defense machinery of the live organisms and for the development of new antimicrobials. These questions were approached by the study of the indolicidin and some of its mutants in solution and in interaction with cell membrane models.

Full textA busca por produtos para o tratamento de feridas provocadas principalmente por queimaduras é cada vez maior. Diversos produtos são desenvolvidos e comercializados com finalidade de atuarem acelerando a dinâmica cicatricial, tais como, agentes tópicos, soluções, enxertos, curativos interativos e biofilmes. A gelatina têm sido um dos materiais mais estudados para esse fim, visto que apresenta arquitetura celular adequada para atuar como suporte na construção de novos tecidos, bem como, possibilita a incorporação de substâncias ativas em sua estrutura proteica. O ácido úsnico (AU), um metabólito secundário de líquens, tem apresentado propriedades biológicas promissoras para o tratamento de feridas, como atividade antimicrobiana e anti-inflamatória. Porém é um metabólito insolúvel em água e para sua incorporação às membranas de gelatina se faz necessário o uso de lipossomas, que possibilitam a incorporação de fármacos insolúveis para posterior liberação controlada. Nesta perspectiva, o objetivo desse estudo foi avaliar o efeito de membranas de gelatina contendo AU/lipossomas sobre o reparo cicatricial em queimaduras em suínos. O ensaio biológico foi realizado com nove (09) suínos machos, 03 animais em cada grupo, formando três grupos, que foram sacrificados 8, 18 e 30 dias após a indução da queimadura, para remoção dos espécimes e preparo das lâminas histológicas. Foram realizadas três queimaduras no dorso de cada animal, as quais foram recobertas com três produtos: pomada de sulfadiazina de prata (grupo SDZ); Duoderme® (grupo GDU) e filmes de gelatina contendo lipossoma/ácido úsnico (grupo UAL). Os resultados da macroscopia demonstraram redução progressiva da área lesada ao longo do tempo experimental para os três grupos estudados, além disso, não foi observado sinais clínicos de infecção secundária. Na análise do índice médio da retração das feridas não houve diferença estatisticamente significativa entre os índices de retração obtidos nos animais em nenhum grupo e tempos analisados (p>0,05). Na microscopia, em 8 dias, os três grupos mostraram degeneração hidrópica do epitélio, com intensa infiltração neutrofílica. A reação de granulação observada na região central das feridas do grupo SDZ foi mais imatura em relação aos grupos GDU e UAL. Em 18 dias, a neoformação epidérmica, apesar de parcial nos três grupos, se mostrou bastante incipiente no SDZ quando comparada aos grupos GDU e UAL. A reação inflamatória foi reduzida em todos os grupos, entretanto a reação de granulação mostrou-se mais imatura no grupo SDZ, em relação à reação de granulação exuberante, ricamente celularizada, evidenciada nos grupos GDU e UAL. Em 30 dias, a reação de granulação, demostrou estar mais extensa no grupo SDZ, contrapondo-se a restrita reação de granulação observada nos grupos GDU e UAL, presente apenas na região abaixo do epitélio. No grupo SDZ, ainda observou-se presença de infiltrado inflamatório envolvido na reação de granulação. Na análise de colágeno por picrossírius verificou-se uma gradativa substituição das fibras de colágeno tipo III para tipo I, bem como, melhora da densidade de colagenização em todos os grupos, porém no UAL, quando comparado aos grupos SDZ e GDU, observou a substituição mais rápida das fibras de colágeno, assim como melhora na densidade da colagenização. Conclui-se assim, que o grupo UAL promoveu o desenvolvimento e maturação da reação de granulação, do reparo cicatricial comparáveis ao duoderme e melhores que a pomada de sualfadizina de prata, bem como maior incremento da deposição colagênica em relação aos dois outros grupos.

Full textGeotechnical engineers have been faced with difficult problems due to failure of geological materials caused by stresses exceeding the strength. The strain around a circular tunnel during and after excavation, for example, may lead to failure due to stresses redistribution. Particularly in rock masses with time-dependent deformation properties, the new stress field can lead to large strains in the long term or even to the closure of the tunnel due to creep behavior under constant stress and temperature. This work aimed originally at simulating the interaction of concrete support structures with evaporites identifying the effects of creep phenomenon in underground excavations. During the development of the work, it was decided to extend the scope to include other phenomena related to the interaction between the first and second layers of tunnel support structures. These include partial friction or the mechanical interface of a waterproofing sprayed membrane. For the development of the present study, numerical simulation of the time-dependent interaction between support and rock masses, during and after excavation of a tunnel were carried out with the commercial FEM software Abaqus®, chosen due to its versatility for the solution of non-linear problems.

Full textGRA proteins are secreted by the intracellular parasite Toxoplasma gondii into the parasitophorous vacuole, where most of them interact with two systems of tubular membranes: the Host Organelle Sequestering Tubules (HaSTs) and the Membrane Nanotubules Network (RNM). Although most of the GRA proteins contain potential transmembrane domains, they are secreted as soluble forms and become membrane-associated only when they reach their target membranes. This unusual property led to consider them as attractive models of protein-membrane interactions. 1 developed two experimental approaches to study the interactions of GRA proteins, extracted trom the parasite or trom the vacuole, with model membranes. Firstly, biochemical approaches using Small Unilamellar Vesic1es (SUVs) led to characterize the solubilisation forms of GRA proteins and their association with SUVs membranes. Secondly, 1 developed a Giant Unilamellar Vesic1es (GUVs) model to study the interactions of GRA proteins with membranes by fluorescence spectroscopy methods. The results provide elements 1) which help to decipher the traffic of GRA proteins within the parasite and the PV, and 2) which open the way to set up an in vitro minimal system to study the building up of the parasitophorous vacuole and of its associated tubular membranes

Peptídeos antimicrobianos são moléculas biologicamente ativas que, geralmente, tem as membranas fosfolipídicas como alvo primário. Resultados de diferentes técnicas experimentais têm sugerido que esses peptídeos permeabilizam as membranas pela formação de poros. Parte dos peptídeos caracterizados apresentam especificidade de disrupção para membranas de bactérias, em detrimento das membranas dos hospedeiros. Essa característica tem atraído a atenção da comunidade científica internacional, porque indica que estas moléculas podem ser modelos para o desenvolvimento de novos antibióticos, portanto o entendimento do mecanismo de ação, ou seja, do mecanismo de formação de poro, tem extrema importância. Simulações por Dinâmica Molecular foram produzidas para investigarmos o impacto que peptídeos antimicrobianos da família Mastoparano tem sobre membranas lipídicas modelo. Dois cenários foram explorados: (i) de baixa concentração peptídeo/lipídeo, P/L=1/128, que consistia de simulações fine-grained das interações de um peptídeo com uma bicamada pura de 128 lipídeos aniônicos (POPG) ou zwiteriônicos (POPC); (ii) de alta concentração, P/L=1/21, que abordava as interações de seis peptídeos com uma bicamada mista de 128 lipídeos POPC/POPG (1/1) usando uma modelagem coarse-grained. Tomando o peptídeo MP1 como caso paradigmático, verificamos que em baixo P/L é possível sugerir que sua característica seletiva surge da capacidade de coordenar e perturbar maior número de lipídeos em membrana aniônica comparada à neutra. Essa capacidade fica acentuada nas simulações com membrana mista, onde a atração dos lipídeos aniônicos pelos peptídeos catiônicos guiou a separação local e a formação de domínios de lipídeos aniônicos, o que facilitou o afinamento local da membrana e a formação de poro transmembrânico. Antimicrobial peptides are biologically active molecules that, usually, have the phospholipid membranes as a primary target. Results from different experimental techniques have suggested these peptides permeabilize membranes by the pore formation. Part of the characterized peptides have specificity of disruption for bacterial membranes, instead of host membrane. This feature has attracted the attention of the international scientific community, because it indicates that these molecules can be models for the development of novel antibiotics, so understanding the mechanism of action, ie, the mechanism of pore formation, is extremely important. Molecular dynamics simulations were performed to investigate the impact of antimicrobial peptides from the Mastoparano family have on model lipid membranes. Two scenarios were explored: (i) of low peptide/lipid concentration, P/L=1/128, which consisted of fine-grained simulations of the interactions of a peptide with a pure bilayer of 128 anionic (POPG) or zwitterionic (POPC) lipids; (ii) of high concentration, P/L=1/21, which addressed the interactions of six peptides with a mixed bilayer of 128 POPC/POPG (1/1) lipids, using a coarse-grained modeling. Taking the MP1 peptide as a paradigmatic case, we found that in low P/L is possible to suggest that its selective feature arises of its ability to coordinate and disturb large number of lipids in the anionic membrane compared to neutral one. This ability is accentuated in simulations with mixed membrane, where the attraction of the anionic lipids by the cationic peptides led to the local segregation and formation of POPG lipid domains, which facilitated the local thinning of the membrane and the formation of transmembrane pore. These findings help to explain how short peptides, such as MP1, are able of forming pores in a membrane whose thickness is larger than the length of the peptide

Os ovos embrionados de codorna podem ser utilizados para inoculação de diversos tipos de vírus, por diferentes vias, assim como técnicas já conhecidas e aplicadas em ovos embrionados de galinha (Gallus domesticus) com algumas adequações. Podem ser atribuídas vantagens a esse processo, tais como: custo menor de produção, redução do tempo de incubação, necessidade de menor espaço físico e, talvez a mais importante, ser de uma espécie diferente do Gallus domesticus e desta forma não ser portadora de antígenos e anticorpos de interesse na avicultura comercial moderna, não interferindo no diagnóstico e na replicação viral. Para avaliar o potencial de replicação viral, foram utilizados 351 ovos embrionados de codorna de casca branca (Coturnix coturnix japonica) da linhagem NISSEI e amostras dos vírus que acometem aves (vírus da doença de Newcastle, vírus da varíola aviária e vírus da bronquite infecciosa das galinhas) e também em outras espécies como vírus da influenza equina. Nos ovos inoculados com o vírus de Newcastle foi coletado o liquido alantoíde para a realização do teste de HA, que apresentou melhor resultado no tempo de 72 horas com o volume de 100μl. Na inoculação do vírus da varíola aviária foram coletadas as membranas corioalantoídes para analisar as les es po e an lise histopatol gica, sendo que todos os volumes utilizados (20, 40, 80 e 100μl) apresentaram lesões características desta enfermidade. Nos vírus da bronquite infecciosa das galinhas, de todos os volumes utilizados, somente o de 40μl produziu lesões no embrião, como nanismo e enrolamento. A replicação do vírus influenza equina, assim como aconteceu com o VDN, foi dependente da concentração e o tempo de incubação, sendo o melhor tempo o intervalo de 48 horas, utilizando o volume de 40μl. Os ovos embrionados de codornas de casca branca também foram utilizados para testar a viabilidade de uso em cultivo primário de fibroblastos, sendo que os mesmos permaneceram viáveis após quatro passagens in vitro. Este modelo biológico foi eficiente para replicação do vírus da doença de Newcastle, vírus da varíola aviária, vírus da bronquite infecciosa das galinhas e vírus da influenza equina. Estes embriões se mostraram aptos também para serem utilizados em cultivo primário de fibroblastos.
Quail embryonated eggs may be used for inoculation of several viruses by different ways, like the well-known techniques used in embryonated chicken (Gallus domesticus) eggs with some light adjustments. Some advantages may be assigned to this process like: lower production costs, reduction in incubation time, no need for large spaces and, maybe the most important one, for being a different species of Gallus domesticus and so, not carrying antigens and antibodies of interest for modern commercial poultry, it does not interfere in the diagnose or in the viral replication. To assess the viral replication potential, we used a total of 351 embryonated quail eggs of white eggshell type (Coturnix coturnix japonica) NISSEI lineage and samples of virus that affect birds (Newcastle disease virus, avipoxvirus and infectious bronchitis chickens and also, some other species like the equine influenza virus.From the eggs inoculated with Newcastle virus it was collected the allantois fluid to perform HA test which presented the better result for 72 hours period and a volume of 100μl. From the inoculation with Fowl pox we collected the chorioallantoic membrane o analyze both pox lesions and for histopathological analysis, all considered volumes (20, 40, 80 e 100μl) presented the disease characteristic lesions. For the Chicken Infectious Bronchitis Virus, from all considered volumes used only the 40μl volume produced embryo lesion like dwarfism and winding. The Equine Influenza Virus replication similar to what was observed with NDV was concentration and incubation time dependent, being the best result achieved for a 48 hours interval using a 40μl volume. White Shell quail embryonated eggs were also used to test its viability for primary cultures of fibroblasts and it was verified that they remain viable after four passages in vitro. This biological model demonstrated efficient for virus replication of Newcastle Disease, Fowl Pox, infectious bronchitis chickens and for equine influenza virus. These embryos also proved suitable for use in primary fibroblasts culture.

Full textSulfonamides and their different derivatives are extensively used in Medicine due to their pharmacological properties, such as antibacterial activity. Modified toxicological properties have been observed when those sulfonamides are administered in the form of metal complexes. In this work, we used the molecules sulfadimethoxine (4-p-aminobenzenesulfonamido-2,6-dimethoxypyrimidine) and sulfisoxazole (N (1) - (3,4-dimethyl-5-isoxazolyl) sulfanilamide) complexes with Cu(II) to assess their mechanism of interaction with lipid model membranes. Differential Scanning Calorimetry (DSC) and Electron Paramagnetic Resonance (EPR) experiments were carried out to monitor the influence of the complexes on the lipid thermotropic behavior. Samples consisting of the multilamellar vesicles of dipalmitoylphosphatidylcholine (DDPC) and, in the case of EPR, spin labeled phospholipids were used as models for the membrane. DSC results showed that the presence of the complexes affects both the pre-transition and the main phase transition of the lipids in the membrane. A disordering effect is evidenced by the decrease in Tm and Tp values. ESR results showed that the major alterations are detected in the lipid gel phase and for the labels positioned close to the lipid-water interface. Overall our results indicate that non-specific interactions between the complexes and the membrane might be a mechanism used by the drugs to overcome the physical barriers found in the cell.

Full textThe peptide hormones Angiotensin II (Ang II) and bradykinin (BK) trigger signal transduction by binding to G Protein Coupled Receptors (GPCR). This work proposes the study of conformational properties of Ang II and BK, as well as their analogues containing the spin label 2,2,6,6-tetramethylpiperidine-1-oxyl-4-amino-4-carboxylic acid, TOAC (TOAC1-Ang II, TOAC3-Ang II, TOAC0-BK, TOAC3-BK) making use of fluorescence spectroscopy. Studies were performed in solution (effect of pH) and also of the interaction between the peptides and model membranes - micelles and bilayers - formed by amphiphiles, either zwitterionic or negatively charged. The intrinsic fluorescence of aromatic residues (Tyr4 in Ang II and Phe5 and Phe8 in BK) was monitored. Fluorescence quenching by the TOAC-carrying analogues provided information about the proximity between TOAC and the fluorophores. The fluorescence was pH-dependent and evinced regions corresponding to pKs of ionizable groups. Peptide-model membrane interactions were also examined. Stronger interactions were detected between the peptides and membranes formed by negatively charged amphiphiles, pointing to the importance of electrostatic interactions for binding. However, hydrophobic interactions were also involved, as suggested by the fact that the peptides also bound to zwitterionic membranes. Variable pH studies showed the effect of this parameter on peptide-membrane interaction. The peptide-membrane interactions promoted changes in the pKs of ionizable residues. Titrations with increasing membrane concentrations allowed calculation of binding constants. Binding to membranes is a function of peptide conformation. In particular, TOAC at position 3 seems to decrease the affinity of both Ang II and BK for membranes. Fluorescence quenching studies made use of three different approaches: 1) quenching by water soluble acrylamide, 2) quenching of the fluorescence of phospholipids carrying the fluorescent group NBD in different positions by the spin-labeled TOAC-bearing analogues, 3) quenching of the peptides fluorescence by methyl esters of stearic acid containing the nitroxide moiety at different positions in the acyl chain. These studies indicated that the peptides are located at the water-membrane interface. Measurements of fluorescence anisotropy also evinced binding of the peptides to the membranes and showed that the peptides undergo more restricted motion under these conditions. A peptide containing residues 92-100 (fEL1) of the Ang II AT1 human receptor was also studied. Predictions based on rhodopsin crystalline structure estimate that this sequence is located in the receptor´s first extra-cellular loop. Preliminary results suggest that fEL1 interacts with Ang II and TOAC1-Ang II, but not TOAC3-Ang II. This latter result is probably due to the TOAC-induced bend that restricts the freedom of motion of the peptide backbone. This feature is probably the cause of lack of biological activity of TOAC3-Ang II and TOAC3-BK, while the N-terminally labeled analogues retain partial activity (Nakaie et al., 2002). GPCR-binding peptides have been proposed to bind to the lipid bilayer and reach their receptors by diffusion in the bilayer. In aqueous solution these molecules exist as a dynamic equilibrium between various flexible conformations, among them, the receptor-bound conformation. The present study provides contributions for the understanding, at the molecular level, of the interaction between the peptide hormones and lipid membranes.

Resumo: Este trabalho teve como objetivo o desenvolvimento de uma microbomba de membrana acionada eletromagneticamente, tornando possível sua utilização futura em dispositivos que reúnam de maneira eficiente diversos processos microfluídicos. Para atingir este objetivo foi necessário o desenvolvimento do atuador, composto de uma membrana flexível e uma microbobina, e do modelo, aqui desenvolvido utilizando a técnica de grafos de conexão, que permita o planejamento futuro das aplicações deste dispositivo. A membrana flexível foi fabricada utilizando-se resina fotocurável de poliuretano-acrilato ou silício corroído em banho KOH e recebeu filmes de níquel puro, depositados por processo à vácuo e eletroquímico e filmes depositados eletroquimicamente da liga CoNiMnP. Tais membranas atingiram deslocamentos próximos a 1?m frente a campos magnéticos de aproximadamente 250 G. As microbobinas, produzidas por meio de deposição eletrolítica de ouro sobre alumina, foram capazes de gerar campos magnéticos ao redor de 50 G. A microbomba com câmara de bombeamento de 10 mm, fabricada com corpo em poliuretano-acrilato e com o atuador descrito apresentou um bom fluxo de líquido nos testes com água.
Abstract: This investigation was conducted to develop a new membrane micropump, magnetically actuated, able to be used in lab-on-a-chip devices. To reach this target, a new actuator was eloped, containing a microcoil and a flexible diaphragm covered with metallic film, and a bond-graph model, leading future applications planning. The diaphragm was made using photocurable polyurethane acrylate resin or silicon etched in KOH bath, covered with pure nickel films, deposited by sputtering and electroplating techniques and electroplated CoNiMnP films. Diaphragms reached 1 ?m displacement facing 250G magnetic fields. Microcoils, obtained by electroplating over alumina surfaces, produced magnetic fields close to 50G. Micropump pumping chamber diameter of 10 mm, made using polyurethane acrylate and using the described actuator produced good water flow in the tests

Resumo: Neste trabalho de tese, foi realizado um estudo do potencial do processo de pervaporação como um processo de separação de misturas azeotrópicas. Foram realizados o estudo, a avaliação e a comparação de dois modelos matemáticos selecionados da literatura par o processo de pervaporação, que têm como base o mecanismo solução-difusão. Forma desenvolvidos dois programas computacionais, o PERKAT e o PERBRUN, que pemitiram avaliar, segundo estes modelos, qualquer tipo de mistura binária, aqui exemplificados pelos sistemas etanol/água e acetato de metila/água. Foi proposto um modelo matemático a partir do mecanismo solução-difusão procurando minimizar a necessidade de dados experimentais e assim, possibilitar a avaliação rápida em estágios iniciais de projetos de processos. Foi , ainda, elaborado um algoritmo de calculo para o processo de pervaporação. Afim de obter um modelo mais robusto, minimizando a necessidade de dados experimentais, foram realizados e estudo da predição do coeficiente de difusão na membrana, a determinação do parâmetro de interação binário solvente/polímero, a validação do método de predição com dados experimentais e uma análise de sensibilidade paramétrica. Com isso, foi desenvolvido o simulador ¿PERVAP¿ a partir do modelo matemático proposto neste trabalho. Abstract: In this work, a study of the potential of pervaporation process for the separation of azeotropic mixtures was carries out. Two mathematical models were chosen from the literature for pervaporation process based on solution diffusion mechanism and a study, an evaluation and a comparison between these models were made. Two computational programs were developed, PERKAT and PERBRUN, which allow the evaluation, according to these models, of any type of binary mixture, here exemplified by the system ethanol/water and methyl acetate/water. A mathematical model was proposed based on the solution-diffusion mechanism with the aim of minimizing the necessity of experimental data and, thus, to make possible the fast evaluation in early stages of process design.Furthermore, it was elaborated a calculation algorithm for the pervaporation process. In order to get a more robust model, minimizing the necessity of experimental data, the study of the prediction of the diffusion coefficient in the membrane, the determination of the binary interaction parameter of solvent/polymer, the validation of the prediction method with experimental data and a parametric sensitivity analysis were carried out. With this, the "PERVAP" simulator was developed from the mathematical model proposed in this work. Full textMembrane structures used in roofs are constructive systems formed mainly by structural membrane, which has also the function of sealing. Structural membranes are flexible sheets that resist to external loads due to their form, to their physical characteristics and to their pre-stressing. The pre-stressing of the membrane can be achieved either through its stretching using cables (surface-tensioned membranes) or through gases pressurization (pneumatic structures). These structures are being utilized in roofs over airports, stadiums, industrial facilities, shopping precincts and buildings for industrial or agricultural use, among others. However, membrane structures are not yet well known, studied and used here in Brazil. In this context, this work has as objectives to systematize the scientific knowledge existent on the theme, to introduce it in Brazil and, also, to point out ways for scientific investigation on membrane structures in this country. This research presents the constructive system, the design process and the analysis methods of membrane structures used in roofs. This work systematizes the membrane structural design, with recommendations and study of case. This work proposes an integrated model of research for this technology development in Brazil. This model should assist the coordination and the mutual appreciation of the scientific investigation efforts in the different knowledge areas involved in the membrane structure development

Full textIn this work we showed results from studies about the use of fluorescence spectroscopy techniques as a tool to investigate amphiphilic aggregates, used as a model of the cell membrane. We performed measurements on the spectral properties of light absorption and emission of adequate chromophors, registered the experimental timeresolved decay of fluorescence and time correlated fluorescence emission of the probes and used also adequate methodologies for the analysis and interpretation of experimental data. Several compounds presenting absorption and emission in the UV/visible spectral range were employed: the lipophilic probes 2-Amino-N-hexadecil-benzamida (Ahba), 6-lauryl-2-dimethylaminonaphthalene (Laurdan), N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl) (NBD), in different environment:homogeneous aqueous medium, micelles of surfactants like Sodium Dodecyl Sulfate (SDS), Cetyl-trimethyl-ammonium-bromide (CTAB) and 3- (Dodecyl-Dimethyl-Ammonio)-propane-sulfonate (DPS) and phospholipid vesicles of 1,2-Dimyristoyl-sn-Glycero-3-Phosphocholine (DMPC), 1,2-Dimyristoyl-sn-Glycero-3- [Phospho-rac-(1-glycerol)](Sodium Salt) (DMPG) and 1-Palmitoyl-2-Oleoyl-sn-Glycero-3-Phosphocholine (POPC). Alkyilpyridinium halides with different alkyl chain length were employed fluorescence quenchers of the Ahba probe. Using the Stern-Volmer model to describe the quenching phenomena dependent on fluorophor/quencher collision, we observed that higher quenching rates were obtained in the presence of negatively charged amphiphilic agreggates: SDS micelles and DMPG vesicles; in the presence of zwitterionic vesicles the quenching efficiency was more efficient when the quencher hydrophobicity was high (long alkyl chain). We performed Förster resonance energy transfer (FRET) experiments where the fluorescent moiety of the probe Ahba was the energy donor. As acceptors molecules we used Acridine Orange, Ethylene-diamine-dinitrophenyl (Eddnp) and NBD-labeled phospholipids. The computational package CONTIN was adapted to analyze the experimentally obtained fluorescence decay profiles of the donor in the presence of the acceptor, in order to determine the distance distribution between the Ahba/Eddnp and Ahba/NBD-phospholipids pairs in the presence of lipid vesicles. For the Ahba/NBD pair, the distances were dependent on the emperature of the system (or the phase bilayer behavior), the acceptor concentration and the NBD position in the phospholipid. We observed that the Laurdan probe can be used in studies about DMPC vesicles diffusion using fluorescence correlation spectroscopy techniques. Investigation about the use of the probe Ahba with this technique had shown that its maximum absorption for two photon excitation occurs near to 695 nm, but it is not an appropriated probe to FCS experiments due to its very low brightness. On the other hand, Ahba can be used as a membrane fluorescent label in membrane fluorescence microscopy, as we can see in the fluorescence imaging experiments with giant vesicles labeled with Ahba.

Full textThis study is aimed at understanding the interaction of capsaicinoids with cell membranes using model systems. Among the alkaloids derived from plants of the genus Capsicum, capsaicin and dihydrocapsaicin account for 90% of capsaicinoids, which are used as analgesic and anti-inflammatory due to their interaction with specific receptors. The neuropharmacological mechanism has been well studied, but the non-neural mode of action has not been elucidated. Here, we use crude extracts (EBs) of malagueta and dedo-de-moça chilli peppers, which are surface active, and affected Langmuir monolayers of dipalmitoyl phosphatidylcholine (DPPC) and dipalmitoyl phosphatidyl glycerol (DPPG). Such effects did not depend on the charge, since EB from dedo-de-moça interacted more strongly with DPPC than with DPPG, while the opposite applied for malagueta. In addition, there was no significant difference between the two EBs. For Langmuir monolayers representing the bacteria S. aureus, both EBs affected the surface pressure isotherms and the polarizationmodulated infrared reflection-absorption spectroscopy (PM-IRRAS) data, without significant distinction between dedo-de-moça and malagueta. However, in the leakage measurements with liposomes the EB from dedo-de-moça was more efficient in rupturing the liposome, which is consistent with the bactericidal activity for S. aureus. In fact, the minimum inhibitory concentration (MIC) was 0.13 mg mL-1 for dedo-de-moça and 4.0 mg mL-1 for malagueta. For the Langmuir monolayers mimicking the E. coli membrane, the EBs interacted much in the same way, while the EB from dedo-de-moça caused larger leakage in liposomes. There was no bactericidal effect of the EBs. This is explained by the fact that gram-negative bacteria, such as E. coli, have a protective outer layer of liposaccharides (LPS). In monolayers representing LPS, there was little incorporation of EBs, from which one infers that the EBs cannot cause disruption of the LPS layer. Taking all these results together, it appears that the mechanism of action for S. aureus involves partial solubilization of the membrane. Furthermore, there is no relationship between pungency and bactericidal activity because dedo-de-moça, which is less pungent, had greater effect than malagueta. It seems also that the action of pepper extracts must depend on the interaction with membrane receptors, which explains why the use of such extracts has been essentially in topical applications.

Full textTritrpticin (TRP3) is a 13-residue antimicrobial peptide that contains three sequential Ws. With the aim of contributing to the understanding of its mechanism of action, functional and conformational studies were performed with TRP3 and two of its analogues where one (WLW) or two (LWL) of the W were replaced by L. The peptides were equally active against both Gram positive and Gram negative bacteria. Higher concentrations were required for hemolytic activity which varied in the order: TRP3>WLW>LWL. The peptides permeabilized membranes model membranes mimicking E. coli\'s lipid composition or containing different negatively charged phospholipids. CD spectra suggested the peptides acquired different conformations upon binding to bilayers or micelles. Fluorescence studies showed that membrane binding decreases in the order: TRP3>WLW>LWL and that the peptides are located close to the water-membrane interface. EPR spectra of lipid spin labels indicated that peptide binding alter lipid organization, increasing molecular packing

Resumo: O objetivo do presente estudo foi observar o efeito de uma membrana de nanofibras poliméricas a base de poli-epsilon-caprolactona (PCL), impregnadas ou não com hidroxiapatita (HA), na reparação óssea em defeitos críticos produzidos artificialmente na calvária de ratos. Foram utilizados 36 ratos, divididos aleatoriamente em seis grupos de seis animais. Para a indução do defeito ósseo padronizado foram realizadas duas perfurações, por meio de trefina de 3 mm de diâmetro, na calvária dos animais, uma em cada lado, sendo que o lado direito recebeu os tratamentos (HA ou PCL-HA) e os controles (coágulo ou PCL) foram feitos no lado esquerdo. Decorridos 30, 60 e 90 dias da cirurgia, os animais foram mortos por aprofundamento da anestesia. As cabeças foram submetidas à tomografia, considerando um voxel de 0.12 mm, com campo visual de 06 × 16 cm e tempo de aquisição de 40s. Foram utilizados 120 kV, 8 mA e 36.12 mA/s na aquisição. Os diâmetros dos orifícios foram medidos por meio do software Invesalius 3.0. As cabeças foram fixadas com solução de formol tamponado (pH 7,2) em tampão fosfato de sódio a 0,1 M, durante 72 horas e submetidas à descalcificação em solução de EDTA a 7% e formol a 5% até verificação de completa descalcificação (aproximadamente 30 dias). Cortes semi-seriados de 7 ?m foram corados por Hematoxilina-Eosina e foram observadas as características histológicas do processo de cicatrização óssea nas lâminas nos diferentes tempos do estudo. A comparação quantitativa das medidas dos orifícios obtidas com a tomografia foi feita pelo teste de Kruskal-Wallis (teste de Dunn como post hoc), considerando um nível de significância de 5%. As imagens tomográficas revelaram uma tendência de redução do volume do orifício ao longo do tempo, a qual foi maior após 90 dias para todos os tratamentos. O tratamento com PCL+HA mostrou menor volume de orifício do que os outros tratamentos independentemente do período. As medidas histológicas mostraram maior formação de osso induzida pela membrana de PCL com ou sem HA do que os outros tratamentos independentemente do período. A HA mostrou maior aumento da cicatrização óssea com ou sem o PCL. Concluímos que a HA adicionada às nanofibras de PCL melhorou significativamente a cicatrização óssea em defeitos provocados na calvária de ratos
Abstract: The aim of the present study was to observe the effect of a polymeric-nanofiber membrane of poly-epsilon-caprolactone (PCL) with or without hydroxyapatite (HA) on bone healing of critical defects induced in rat calvaria. 36 animals were randomly divided into six groups. The standardized bone defects were obtained by two perforations with 3.0 mm diameter trephine directly into the animals¿ calvaria, one in each side. The right side received the treatments (HA or PCL-HA) and the left side the controls (blood clot or PCL). After 30, 60 and 90 days of the surgical procedure, all animals were killed; the head was sectioned and submitted to tomography (voxel of 0.12 mm, visual field of 6x16 cm, acquisition time of 40s, 120 kV, 8mA and 36.12 mA/s). The perforation diameters were measured using the software Invesalius 3.0. After tomography, all heads were fixed by buffered formol (pH=7.2) in 0.1M sodium-phosphate buffer during 72 hours and submitted to decalcification in 7% EDTA/5% formol solution until complete decalcification (approximately 30 days). Semi-serial 7 µm cuts were stained with Hematoxicilin-Eosin. Histological characteristics of bone healing were observed according to groups and periods. Quantitative comparisons of perforation measurements from both tomography and histological analysis were performed by Kruskal-Wallis (Dunn post hoc) test with a 5% significance level. Tomography images revealed a tendency to reduce the perforation volume along time, which was higher after 90 days for all treatments. The treatment with PCL+HA showed the lower volumes of perforations than the other treatments irrespectively of the period. Histological measurements showed more osseous formation induced by treatment with PCL with or without HA than the other treatments irrespectively of the period. HA appear to increase bone healing with or without PCL. We concluded that HA added to PCL nanofibers significantly improved the bone healing in bone defects of rat calvaria

Full textLeishmaniasis is a complex of diseases part of the neglected tropical diseases caused by several species of the genus Leishmania. It reaches a large part of the poorest people in the world and its visceral form, which is fatal if left untreated, has been spread around big cities, increasing the number of people at risk of infection. Among the used drugs for the treatment, there is the synthetic lipid analogue hexadecylphosphocholine (miltefosine), orally administrated, which acts in the cell membranes and can induce apoptosis like death, but its mechanism of action is not totally clear. The first interactions site of this drug is the cell membrane, and it is important to know its mechanism of interaction. In this work we explore properties of several membrane models with different compositions, taking into account the existent knowledge about the composition of the Leishmania plasma membrane. Therefore, the model membranes were giant and large unilamellar vesicles (GUVs and LUVs), formed from pure phospholipids, binary mixtures of phospholipids and cholesterol and ternary mixtures with ceramide, a sterol present in the Leishmania membranes. The interaction with miltefosine was studied in different intervals of drug concentration. The main techniques used were the steady-state and time-resolved fluorescence spectroscopy, fluorescence correlation spectroscopy, confocal and fluorescence microscopy and fluorescence lifetime imaging. The interaction depends on i) the molar ratio of drug and lipids; ii) the real concentration of the drug, if it is below or above the critical micelle concentration (CMC); iii) the composition of the model membrane and the lipid phase of the bilayer. In concentration below the CMC, miltefosine has an effect of bilayer fluidization, mainly when it is in a more ordered phase, but this effect is less pronounced in cholesterol presence, because this compound protects the bilayers from the drug effect. In vesicles from ternary mixtures of phospholipid, cholesterol and ceramide in high concentration, there is no phase separation, and the presence of 10 mol% of miltefosine promotes ceramide domains formation; in vesicles in which ceramide is in low concentration, forming domains, the phase separation is less evident with miltefosine addition. In high concentration ratio miltefosine/lipids, but below CMC, it is observed a decrease in vesicles size with drug/lipids aggregates formation from portion of the bilayer. In concentrations above the CMC, drastic effects occur, with solubilization of bigger portions of the membrane bilayer, and the effects occur in lower times for higher drug concentration. Therefore, generally, cholesterol protects bilayer from the effect of miltefosine, but the drug has a pronounced effect in model membranes of ternary mixtures containing ceramide. The effects vary with miltefosine concentration, increasing the bilayer fluidity in lower drug/lipid ratio, solubilization of small portions of the bilayer and decrease of vesicles size in higher ratios, but still below CMC, and above CMC, formation of aggregates of the drug with portions of bilayer lipids, and membrane fragmentation.

Resumo: Peptídeos antimicrobianos são moléculas biologicamente ativas que, geralmente, tem as membranas fosfolipídicas como alvo primário. Resultados de diferentes técnicas experimentais têm sugerido que esses peptídeos permeabilizam as membranas pela formação de poros. Parte dos peptídeos caracterizados apresentam especificidade de disrupção para membranas de bactérias, em detrimento das membranas dos hospedeiros. Essa característica tem atraído a atenção da comunidade científica internacional, porque indica que estas moléculas podem ser modelos para o desenvolvimento de novos antibióticos, portanto o entendimento do mecanismo de ação, ou seja, do mecanismo de formação de poro, tem extrema importância. Simulações por Dinâmica Molecular foram produzidas para investigarmos o impacto que peptídeos antimicrobianos da família Mastoparano tem sobre membranas lipídicas modelo. Dois cenários foram explorados: (i) de baixa concentração peptídeo/lipídeo, P/L=1/128, que consistia de simulações fine-grained das interações de um peptídeo com uma bicamada pura de 128 lipídeos aniônicos (POPG) ou zwiteriônicos (POPC); (ii) de alta concentração, P/L=1/21, que abordava as interações de seis peptídeos com uma bicamada mista de 128 lipídeos POPC/POPG (1/1) usando uma modelagem coarse-grained. Tomando o peptídeo MP1 como caso paradigmático, verificamos que em baixo P/L é possível sugerir que sua característica seletiva surge da capacidade de coordenar e perturbar maior número de lipídeos em membrana aniônica comparada à neutra. Essa capacidade fica acentuada nas simulações com membrana mista, onde a atração dos lipídeos aniônicos pelos peptídeos catiônicos guiou a separação local e a formação de domínios de lipídeos aniônicos, o que facilitou o afinamento local da membrana e a formação de poro transmembrânico. Abstract: Antimicrobial peptides are biologically active molecules that, usually, have the phospholipid membranes as a primary target. Results from different experimental techniques have suggested these peptides permeabilize membranes by the pore formation. Part of the characterized peptides have specificity of disruption for bacterial membranes, instead of host membrane. This feature has attracted the attention of the international scientific community, because it indicates that these molecules can be models for the development of novel antibiotics, so understanding the mechanism of action, ie, the mechanism of pore formation, is extremely important. Molecular dynamics simulations were performed to investigate the impact of antimicrobial peptides from the Mastoparano family have on model lipid membranes. Two scenarios were explored: (i) of low peptide/lipid concentration, P/L=1/128, which consisted of fine-grained simulations of the interactions of a peptide with a pure bilayer of 128 anionic (POPG) or zwitterionic (POPC) lipids; (ii) of high concentration, P/L=1/21, which addressed the interactions of six peptides with a mixed bilayer of 128 POPC/POPG (1/1) lipids, using a coarse-grained modeling. Taking the MP1 peptide as a paradigmatic case, we found that in low P/L is possible to suggest that its selective feature arises of its ability to coordinate and disturb large number of lipids in the anionic membrane compared to neutral one. This ability is accentuated in simulations with mixed membrane, where the attraction of the anionic lipids by the cationic peptides led to the local segregation and formation of POPG lipid domains, which facilitated the local thinning of the membrane and the formation of transmembrane pore. These findings help to explain how short peptides, such as MP1, are able of forming pores in a membrane whose thickness is larger than the length of the peptide

Resumo: A inibição crônica do óxido nítrico (NO) leva ao desenvolvimento de hipertensão arterial, lesão renal isquêmica e glomeruloesderose. A administração concomitante de Uma dieta hipersódica leva ao desenvolvimento de albuminúria maciça. Nós investigamos os mecanismos responsáveis por esse aumento de permeabilidade glomerular a macromoléculas.Ratos Munich Wistar machos e adultos receberam dietas padrão (NS) ou hipersódica (HS) e associado ou não à administração de um inibido r específico da síntese de óxido nítrico, o Nw Nitro-L-Arginina-Metil Ester (L-NAME), via oral a 70 mgjKgj dia. Após 15 e 30 dias medimos: a °albuminúria (UalbV, mgj min), a pressão arterial média (P AM, mmHg), o clearance de inulina (Ci, mIj min), o de dextran (Cd, mIj min) e o clearance fracional de dextran (8=Ci/Cd). Investigamos também a densidade de cargas aniônicas da membrana basal glomerular (MBG), perfundindo o rim com uma solução contendo ferritina catiônica e quantificando a porcentagem de depósitos de ferritina em micrografias eletrônica de glomérulos. N ossos resultados sugerem que a inibiç'ão crônica do NO quando associada a sobrecarga salina aumenta a permeabilidade glomerular através de: (1) um defeito na restrição por tamanho à filtração de macromoléculas; (2) perda da barreira eletrostática glomerular. Observamos também que 30 dias após interrupção dos tratamentos ocorre reversão dessas alterações
Abstract: We have previously described that chronic nitric oxide (NO) inhibition leads to progressive systemic hypertension and renal structural injury. Concomitant dietary salt overload and NO blockade lead to the development of massive albuminuria. Here we investigated the mechanism whereby these treatments increase the glomerular permeability to macromolecules. Adult male Munich Wistar rats were given standard-salt (55, 0.5% Na) or high salt (H5, 3.12% Na) diet and received either no treatment or the NO inhibitor Nco-L-nitroarginine metilester (NAME), 70 mg/kgl day in drinking water. At 15 and 30 days albuminuria (UalbV, mg/min), inulin clearance (Cin, mI/min), mean arterial pressure (MAP, mmHg), neutral dextran clearance (Cd, mIl min) and the fractional neutral dextran clearance (8= Cinl Cd) were measure,d. The density of fixed anionic sites at glomerular basement membrane (GBM) was also estimated by a point-counting technique, after renal perfusion with a solution containing cationic ferritin. The results suggest that chronic NO inhibition combined with H5 enhances the permeability of the glomerulus by : (1) impairing its size selectivity (by direct wall injury); (2) impairing especialIy its charge selectivity. We also observed that 30 days after the interruption of treatments all these alterations were reverted.

Full textO objetivo do presente trabalho é comparar, do ponto de vista elétrico, a membrana do neurônio ganglionar com a da célula de neuroblastoma, analisando os efeitos das cargas fixas sobre o potencial elétrico nas superfícies da bicamada lipídica e também sobre o comportamento do perfil de potencial através da membrana, considerando as condiçõesfísico-químicas do estado de repouso e do estado de potencial de ação. As condições para a ocorrência dos referidos estados foram baseadas em valores numéricos de parâmetros elétricos e químicos, característicos dessas células, obtidos na literatura. O neurônio ganglionar exemplifica um neurônio sadio, e a célula de neuroblastoma, que é uma célula tumoral, exemplifica um neurônio patológico, alterado por esta condição. O neuroblastoma é um tumor que se origina das células da crista neural (neuroblastos), que é uma estrutura embrionária que dá origem a muitas partes do sistema nervoso, podendo surgir em diversos locais do organismo, desde a região do crânio até a área mais inferior da coluna. O modelo adotado para simular a membrana de neurônio inclui: (a) as distribuições espaciais de cargas elétricas fixas no glicocálix e na rede de proteínas citoplasmáticas; (b) as distribuições de cargas na solução eletrolítica dos meios externo e interno; e (c) as cargas superficiais da bicamada lipídica. Os resultados que obtivemos mostraram que, nos estados de repouso e de ação, os potenciais superficiais da bicamada interno (ÁSbc) e externo (ÁSgb) da célula de neuroblastoma não sofrem alteração mensurável, quando a densidade de carga na superfície interna (QSbc) torna-se 50 vezes mais negativa, tanto para uma densidade de carga na superfície externa da bicamada nula (QSgb = 0), como para um valor de QSgb 6= 0. Porém, no estado de repouso, uma leve queda em ÁSbc do neur^onio ganglionar pode ser observada com este nível de variação de carga, sendo que ÁSgb do neurônio ganglionar é mais negativo quando QSgb = 1=1100 e/A2. No estado de ação, para QSgb = 0, o aumento da negatividade de QSbc não provoca alteração detectável de ÁSbc e ÁSgb para os dois neurônios. Quando consideramos QSgb = 1=1100 e/A2, ÁSgb do neurônio ganglionar se torna mais negativo, não se observando variações detectáveis nos potenciais superficiais da célula de neuroblastoma. Tanto no repouso quanto no estado de ação, ÁSgb das duas células não sofre variação sensível com o aumento da negatividade da carga fixa distribuída espacialmente no citoplasma. Já a ÁSbc sofre uma queda gradativa nos dois tipos celulares; porém, no estado de ação, esta queda é mais rápida. Descobrimos diferenças importantes nos perfis de potencial das duas células, especialmente na região do glicocálix.
The aim of our work is to compare, from the electrical point of view, the ganglion neuron membrane with the neuroblastoma cell's membrane, analyzing the effects of fixed charges on the electric potential of the surfaces of the lipidic bilayer and on the behavior of the potential profile across the membrane, considering the physicochemical conditions of the resting state and of the action potential state. The conditions for the occurrence of these states were defined, based on numerical values of electrical and chemical parameters of these cells, obtained in the literature. The ganglion neuron portrays a healthy neuron,and the neuroblastoma cell, which is a tumor cell, represents a pathologic neuron, different from the ganglion cell, due to this condition. A neuroblastoma is a tumor, originated from neural crest cells (neuroblasts), which is an embryonic structure that gives rise to many parts of the nervous system and can arise in various body sites, from the region of the skull all the way to the lower spinal column area.The model used to simulate the neuron membrane includes: (a) the spatial distribution of the fixed electric charges on the glycocalyx and on the network of cytoplasmic proteins; (b) the distribution of the charges in the electrolytic solution of outer and inner resources; and (c) the surface charges of the lipidic bilayer. The results we obtained show that, in the resting and action states, the inner (ÁSbc) and outer (ÁSgb) surface potential of neuroblastoma cells do not change measurably, when the charge density on the inner surface (QSbc) becomes 50 times more negative, for both null charge density on the outer surface (QSgb = 0) and for QSgb 6= 0. However, a slight drop in ÁSbc of a ganglion neuron can be observed with this level of charge variation, but ÁSgb of ganglion neuron is more negative when QSgb = 1=1100 e/A2. At action potential state, for QSgb = 0, the negative increase of QSbc does not measurably change ÁSbc and ÁSgb , for both neurons. When we consider QSgb = 1=1100 e/A2, for the ganglion neuron ÁSgb becomes more negative, with no significant detectable changes in the neuroblastoma cell's surface potentials. At the resting and action states, ÁSgb of both cells does not undergo substantial changes with the negative increasing of fixed charges uniformly distributed in the cytoplasm. However,ÁSbc undergoes a gradual decrease in both cell types, although for the action state, this fall is faster. We discovered important dierences among the potential profile of the two cells, especially in the glicocalyx region.

Nos últimos anos, o interesse por estudar peptídeos com atividade antimicrobiana e anticâncer tem aumentado consideravelmente. Neste trabalho, direcionamos nossos estudos para os peptídeos antimicrobianos da classe dos matoparanos extraídos do veneno da vespa social Polybia paulista: o Polybia-MP III (MP-III), Polybia-MP I (MP-I) e seu sintético análogo, o Asn2-Polybia-MP I (NMP-I). Com o objetivo de estudar a interação desses peptídeos com um modelo de membrana composta por lipídeos do tipo POPC (Palmitoil-Oleil-Fosfatidil-Colina) em solução aquosa, realizamos dois tipos de simulações de Dinâmica Molecular (DM). Primeiramente, foram realizadas simulações no equilíbrio com a finalidade de amostrar o comportamento geral do sistema. Em seguida, para investigarmos especificamente o processo de adsorção do peptídeo à bicamada, realizamos simulações de DM com a utilização do método Adaptive Biasing Force (ABF), o que nos permitiu calcular o perfil de energia livre desse processo. Dentre os três peptídeos estudados, o que mais se destacou com relação a sua interação com o modelo de membrana, na simulação no equilíbrio, foi o peptídeo MP-I, por ser o único a se adsorver na bicamada com 200ns de simulação. Com a utilização do método ABF, verificamos que a posição mais estável para cada peptídeo é a posição na qual estes estão paralelos à face da bicamada, com a face hidrofóbica de cada peptídeo voltada para o interior da membrana, e a face hidrofílica voltada para o meio aquoso. Além disso, os resíduos hidrofílicos estão em contato com o grupo polar dos fosfolipídeos e com a água, e o resíduo de triptofano encontra-se posicionado na interface hidrofóbica/hidrofílica. Os resultados obtidos na simulação no equilíbrio para o peptídeo MP-I, que foi o único a se adsorver na bicamada, estão de acordo com os resultados obtidos pelo método ABF, e ambos concordam com resultados experimentais da literatura.
In recent years, interest in studying peptides with antimicrobial and anti-cancer activity has increased considerably. In this study, we focus our studies on antimicrobial peptides of the mastoparans class extracted from the venom of the social wasp Polybia paulista: the Polybia-MP III (MP III), Polybia-MP I (MP-I) and its synthetic analogue, the Asn2-Polybia-MP I (NMP-I). In order to study the interaction of these peptides with a model of membrane composed of POPC (Palmitoyl-Oleoyl-Phosphatidyl-Choline) lipids type in aqueous solution, we conducted two types of molecular dynamics simulations (MD). At first, a balance system simulation was performed in order to get a sampling of the general behavior of the system. Then, to investigate the adsorption of the peptide to the bilayer, MD simulations using Adaptive Biasing Force (ABF) method was performed, which allowed us to calculate the free energy profile of this process. Among the three studied peptides, the MP-I peptide was the one that stood out related to its interaction with the membrane model, in the simulation on balance, for being the one to adsorb the bilayer with simulation of 200ns. Using the ABF method, we verified that the most stable position for each peptide is the position in which they are parallel to the surface of the bilayer with the hydrophobic side of each peptide facing into the membrane, and the hydrophilic side facing the aqueous solution. In addition, the hydrophilic residues are in contact with the polar group of the phospholipid and water, and the tryptophan residue is positioned on the hydrophobic/hydrophilic interface. The results of the simulation in balance for the MP-I peptide, which was the only adsorbed in the bilayer, are in agreement with the results obtained by the method ABF, and both agree with experimental results in the literature.

Full textModelling the dynamics of neurons is relevant in studies on neurosciences. In this work, a computational model of neuron based on the behavior of the ionic channels found in its membrane is proposed. The model comprises microscopic elements, as the behavior of the individual channels, and macroscopic elements, as the tension along a membrane patch. Simulations were performed with the aim of reproducing biological data and results derived from classical theoretical models of the field. It was possible to reproduce with good agreement the action potential, the phenomenon of adaptation, the curve of the input current versus the spike frequency, and the excitatory postsynaptic potential.

Resumo: As fases lamelares fonnadas em sistemas lipídeo/agua possuem características estruturais que servem como base para estudos de fenômenos de superficie e interface. As multicamadas de fosfolipídeos constituem-se em um eficiente modelo estrutural no estudo das propriedades das membranas biológicas. Em particular, as vesículas multilamelares ou lipossomas são atualmente motivo de intensas pesquisas em áreas tão diversas como o desenvolvimento de cosméticos e as aplicações médicas, como o encapsulamento de drogas e imunogênicos. O presente trabalho teve como objetivo analisar, do ponto de vista estrutural, os parâmetros envolvidos no processo de incorporação de vários tipos de compostos hidrofóbicos na estrutura lamelar de lipossomas e multicamadas do fosfolipídeo zwitteriônico DPPC. Técnicas de difração e espalhamento de raios X usando radiação síncrotron foram usadas para acompanhar mudanças estruturais de diversos sistemas de bicarnadas registrando as alterações nas temperaturas de transição de fase tennotrópicas e na ordem do empacotamento lamelar. Dessa maneira foi possível estudar o comportamento das fases Lb e La em função do conteúdo de água em sistemas de vesículas multilamelares de DPPC com adição do alcaloide Elipticina em várias concentrações. Os resultados mostraram uma diminuição da temperatura de pré transição Lb'-->Pb' assim como da transição de fase principal Pb'-->La. A taxa de decréscimo do espaçamento lamelar da membrana modelo na fase líquido cristalina em função da temperatura nos permitiu descrever o comportamento do coeficiente de compressão ténnica da fase La. Foi possível provar para este sistema, que adicionando o lipídeo aniônico Cardiolipina à membrana mista DPPC/CL. consegue-se otimizar a incorpomção da Elipticina. Os métodos previamente mencionados foram também usados para o estudo das fases lamelares de DPPC com adição de hidrocarbonetos aromáticos policíclicos com vários graus de ação carcinogênica. Foi verificado nestas experiências que a temperatura da transição de fase principal assim como a distância de repetição da estrutura lamelar das vesículas na fase La sofrem mudanças com a incorporação dos HAPs. Mostrou-se assim, que a incorporação destes compostos hidrofóbicos favorece a fonnação da fase líquido cristalina, causando um alargamento da região da transição Pb' --> La. Os resultados obtidos neste trabalho são de importância básica no desenvolvimento de lipossomas termo-sensíveis que podem atuar como carreadores de drogas e no entendimento das perturbações que podem produzir moléculas hidrofóbicas quando interagem com as membranas celulares.
Abstract: The structural features of the lamellar phases of the lipid/water systems are frequently used as a basic model for the study of surface and interface phenomena. Phospholipid bilayers represent an efficient structural model for the study of biological membranes. In particular, multilamellar vesicles or liposomes are the subject of intense research in areas so diverse as de development of cosmetics or medical applications, such as drug and antigenic material encapsulation. The objective of this work has been to study, from the structural point of view, the process of incorporation of various types of hydrophobic compounds into the lamellar phase of liposomes and multilayers of the zwitterionic phospholipid DPPC. X-ray diffraction and scattering techniques using synchrotron radiation, have been used to monitor structural changes of several bilayer systems. Thermotropic phase transitions as well as the order of the lamellar packing were studied in in situ experiments. The behavior of the Lb' e Laphases was followed as a function of the water content in dispersions of DPPC multi lamellar vesicles with the addition of the alkaloid Ellipticine in several concentrations. The results showed a decrease in the temperature of the pre-transition as well as that of the main transition (Pb' --> La). The decrease of the lamellar spacing as a function oftemperature in the liquid crystalline phase leads to the description ofthe thermal compression coefficient in the w phase. It was also proved that addition of the anionic lipid Cardiolipin to the DPPC/water system it is possible to promote the incorporation of Ellipticine. The aforementioned methods were used in another set of experiments to study the lamellar phases of DPPC with the addition of Polycyclic Aromatic Hydrocarbons known to have various degrees of carcinogenic activity. It was verified that the main transition temperature as well as the lamellar spacing of the multilamellar vesicles in the La phase have a change with the incorporation of these compounds. It was possible to show that the hydrophobic PAHs favor the formation of the liquid crystalline phase, causing a broadening of the Pb' --> La phase transition. The results obtained in this work are basically important in the development of thermo sensitive liposomes, which can act as drug carriers. They also contribute to the understanding of the perturbations caused by the interaction of highly hydrophobic molecules with the cellular membranes.

Full textThis present work studied the process of cross flow microfiltration using tubular ceramic applied in the vinasse clarification from sugar cane, analyzing fluid dynamic features of process and the types of pore blocking applied to math models. In this research was used \'alfa\'-alumina (\'alfa\'-\'AL IND.2\'O IND. 3) ceramic membrane with pore size of 1.2 \'mü\'m and 0.8 \'mü\'m, respectively called M12 and M08. Permeate samples (transmembrane flux) and retentate were analyzed measuring their specific mass, pH, electrical conductivity, viscosity, surface tension, total soluble solid (ºBrix), color and turbidity. The virgin membranes were analyzed first with tap water in different fluid dynamics conditions and transmembrane flux was measured in function of the time to quantify its proprieties before to have impregnation in its surface. Vinasse was deluded in water in the proportion of 50% vol and it was microfiltered in varied fluid dynamics using transmembrane pressure of 200 kPa, 300 kPa, 400 kPa and 500 kPa in turbulent flow with Reynolds number 11,500; 22,500 and 33,500. The experimental data obtained were analyzed through to graphics of transmembrane flux versus time and resistances (membrane resistance, total resistance, polarization resistance and fouling resistance) versus pressure, and produced tables with physical-chemical proprieties values. The influence of interaction between vinasse and membrane surface was investigated using math models to pore blockage. The high performance was obtained with M12 membrane with permeate of 31.48 L/h.\'M POT.2\' in transmembrane pressure of 500 kPa (Re = 13,500). For M08 membrane permeate flux (39.47 L/h.\'M POT.2\') was higher for the situation of greater tangential velocity (Re = 33,500) and higher pressure (\'delta\'P = 500 kPa). The analysis for the pore blockage showed that the permeate flux decreases primarily by phenomena related to the cake formation of polarization and partial blockage of pores. Analyses of pH and viscosity of permeate flux showed that Reynolds number influences on values. The study showed that the clarification of vinasse by ceramic membranes of microfiltration has the potential to remove suspension particles and to reduce turbidity and color.

Full textResumo: Os peptídeos sintéticos L1A (IDGLKAIWKKVADLLKNT-NH2, Q = +3e) e seu análogo acetilado (acL1A, Q = +2e) utilizados neste estudo foram projetados para que tenham características estruturais semelhantes ao peptídeo Polybia-MP1 extraído do veneno da vespa Polybia paulista, em que um dos dois resíduos ácidos ocupa a segunda posição na região Nterminal, e resíduos básicos são terceiros e/ou quartos vizinhos dos resíduos ácidos. Esses peptídeos possuem significativa atividade bactericida seletiva para bactérias Gram-negativas, especialmente Escherichia coli, sem serem hemolíticos. Estudos anteriores, em sistemas modelo, demonstraram que a acetilação do N-terminal resultou no aumento da atividade lítica em vesículas aniônicas (8POPC/2POPG) em comparação com o L1A, o que sugeriu perturbação do empacotamento lipídico de modo mais eficaz para o análogo que é menos carregado. Considerando que a membrana plasmática de bactérias Gram-negativas contém majoritariamente fosfatidiletanolamina (PE) e fosfatidilglicerol (PG), o presente trabalho propôs investigar o impacto dos peptídeos L1A e acL1A em membranas modelo compostas por 3POPE/1DOPG utilizando uma variedade de técnicas experimentais. Os resultados demonstraram que ambos os peptídeos induziram segregação lipídica, sendo o análogo acetilado mais eficiente em recrutar PG e segregar PE.
Abstract: The synthetic peptides L1A (IDGLKAIWKKVADLLKNT-NH2, Q = +3e) and its acetylated analog (acL1A, Q = +2e) used in this study were designed to have some structural features similar to the peptide Polybia-MP1 extracted from the venom of the wasp Polybia paulista, in which one of the acidic residues occupies the second position on the N-terminus region and basic residues are third and/or fourth neighbors of the acidic residues. These peptides display significant bactericidal activity against Gram-negative bacteria, especially Escherichia coli, being non-hemolytic. Previous work performed in model membrane systems has shown that the N-terminal acetylation led to an increase on the lytic activity in anionic vesicles (8POPC/2POPG) compared with L1A, suggesting that the less charged peptide has higher ability to perturb the lipid-packing. Considering that the Gram-negative cell membranes contain mainly phosphatidylethanolamine (PE) and phosphatidylglycerol (PG), the present work proposed to investigate the impact of L1A and acL1A on model membranes composed of 3POPE/1DOPG using a variety of experimental techniques. The results suggested that both peptides induced lipid segregation being the acetylated analog more efficient in recruiting PG and segregating PE.

The main predictive models of absorption of potential new drugs in preclinical stage are focused on the gastrointestinal mucosa, given the predominance of this pathway in drug administration. Often, the fraction absorbed (Fa) can be predicted in ex vivo models (p.e. Ussing chambers), in vitro (p.e. Caco-2 cells monolayers), intestinal perfusion studies in situ and in vivo absorption. In the present study, from an adaptation of Snapwell ™ inserts, a new ex vivo model to evaluate the permeability of substances passively absorbed is proposed. High permeable drugs (metoprolol, caffeine and theophylline) and low permeable drugs (atenolol, ranitidine and cimetidine) were maintained in an incubator at 37 ° C under constant stirring (60 rpm) and carbogenic atmosphere (5% CO2). The viability of the jejunal membrane (52 Ω.cm2 ± 8.0) was observed remaining above 20 Ω.cm2 for 120 min incubation, under all conditions evaluated, including the addition of co-solvents (1% DMSO and 1% EtOH). Values of apparent permeability coefficients obtained (Papp) were characteristic of ex vivo permeation studies (3.8 to 12.6 x10-6 cm / s). Strong correlation was observed between the data obtained here versus data intestinal perfusion in vivo (r = 0.89), as well as the fraction absorbed in humans (r = 0.85), reported in the literature. Additionally, the model features high sensitivity and accuracy compared to other commonly used models in classification permeability of substances. In line, we can infer that the MTSSNAPWELL model demonstrates, yet, potential application in studies of screening for selection of low molecular weight, such as potential phytochemicals, as well as their synthetic analogues evaluated with low amount of sample (ca 10 mg).
Os principais modelos preditivos da absorção de potenciais novos fármacos na etapa pré-clinica são focados na mucosa gastrointestinal, haja vista a predominância desta via na administração medicamentosa. Frequentemente, a fração absorvida (Fa) pode ser predita em modelos ex vivo em câmaras de Ussing, in vitro em monocamadas de células Caco-2, perfusão intestinal in situ e estudos de absorção in vivo. No presente estudo, a partir de uma adaptação do aparato Snapwell™, um novo modelo ex vivo de avaliação da permeabilidade para substâncias absorvidas por difusão passiva é proposto. Substâncias de alta (metoprolol, cafeína e teofilina) e baixa (atenolol, ranitidina e cimetidina) permeabilidade, foram mantidos em incubadora à 37OC, sob agitação constante (60 rpm) e atmosfera carbogênica (5% CO2). A viabilidade da membrana jejunal (52 ± 8,0 Ω.cm2) foi observada mantendose acima de 20 Ω.cm2 por até 120 min de incubação, sob todas condições avaliadas incluindo a adição de co-solventes (DMSO 1% e EtOH 1%). Os valores de coeficientes de permeabilidade aparente obtidos (Papp) mostraram-se característicos de estudos ex vivo de permeação (3,8 – 12,6 x10-6 cm/s). Forte correlação foi observada entre os dados aqui obtidos versus dados de perfusão intestinal in vivo (r = 0,89), assim como da fração absorvida em humanos (r = 0,85), relatados na literatura. Adicionalmente, o modelo apresenta elevada sensibilidade e precisão frente aos demais modelos comumente utilizados na classificação da permeabilidade de substâncias. Em consonância, pode-se inferir que o modelo MTSSNAPWELL demonstra, até o momento, potencial aplicação em estudos de screening para seleção de moléculas de baixo peso molecular, tais como potenciais fitofármacos, assim como seus análogos sintéticos avaliados com baixa quantidade de amostra (c.a. 10 mg).

Full textLipids in aqueous solution form a variety of different structures which include monolayers of surfactants at the water-air interface, known as Langmuir monolayers, as well as unilamellar or plurilamellar vesicles within the solution. Under temperature variation, these structures display different phases, observed through calorimetry or isothermal variation of lateral pressure. Among the phases presented by these structures, the two most important differ in the order of the lipid chains. From the point of view of the thermodynamic phases, our understanding is that Langmuir monolayers and lipid bilayers constitute the same physical system under different constraints. In this work, we develop a statistical model for the order - disorder transition of lipid bilayers which adds density fluctuations to Doniach\'s 1980 model, which has been considered the basis for many theoretical studies for lipid systems phase transitions. Density fluctuations are essential in the description of the properties of charged vesicles in solution, which consist of surfactants whose polar head dissociates in water. The study in the laboratory of thermal and structural properties of artificial charged lipid membranes is relatively new, and was developed largely in the IFUSP Laboratory of Biophysics. Such membranes exhibit distinct behavior if compared to neutral membranes, notoriously influenced by the solution salt concentration. The experimental investigations motivated us to develop a second model, in which we describe the polar headgroups through a pair of opposite charges. The lipid layer is attached to the lattice restricted primitive model, which plays the role of the saline solution. The first model was studied both through a mean-field approximation as well as through Monte Carlo simulations, whereas the second model was investigated only through numerical simulations. The study of the charged model was preceded by a thorough investigation of the simulation techniques for Coulomb interaction systems, leading to the development of a methodology suitable for non isotropic boundary conditions and with reduced computational cost. The statistical models proposed by us led to two important results. To our knowledge, our model for neutral lipid layers is the only statistical model which, aside from describing simultaneously both the gel-fluid transition of lipid bilayers and the condensed liquid - expanded liquid transition of Langmuir monolayers, also describes the gas- expanded liquid transition at the air-water interface. The model for lipid layers that dissociate in water reproduces the abrupt change in dissociation, concomitant with the order-disorder transition, a property that allows us to interpret experimental studies related to conductivity of the corresponding lipid solutions.

Full textNeste trabalho estudou-se a estrutura, função e mecanismo de ação do peptídeo antibacteriano Protonectarina-MP (isolado de veneno da vespa social Protonectarina sylveirae) tendo seu resíduo C-terminal nas formas ácida (-OH) e amidada (-NH2). Os peptídeos foram sintetizados, utilizando-se a estratégia Fmoc, purificados por cromatografia líquida de alta performance. O monitoramento do material sintético foi feito por espectrometria de massas ESI-MS e por seqüenciamento através de Química Degradativa de Edman. A estrutura secundária foi investigada pelo uso de espectroscopia de dicroísmo circular e modelagem molecular. Atividade lítica (extravasamento) e interação do resíduo de triptofano em vesículas foram investigadas pelo uso de espectrômetro de fluorescência. Foram realizados ensaios sobre as interações desses peptídeos em meio de vesículas zwitteriônicas e aniônica, formando complexos proteolipossomos que foram submetidos à troca isotópica H/D monitorada por espectrometria de massas ESI-MS e MS/MS. Além disso, foram realizados ensaios biológicos de atividade hemolítica, de desgranulação de mastócito, de liberação da enzima citoplasmática Lactato Desidrogenase e de atividade antimicrobianas. Os dados de CD revelam uma tendência dos peptídeos se estruturarem em hélice-α em ambiente hidrofóbico e em ambiente de membranas. Porém, o mesmo não pode ser observado em meio aquoso. Os modelos obtidos para ambos os peptídeos por modelagem molecular mostram uma estruturação em hélice-α anfipática. Nos ensaios de atividade lítica em vesículas, os peptídeos apresentaram um processo com cooperatividade positiva, com curvas de dose-resposta que mostram uma dependência sigmoidal com a concentração do peptídeo. Os resultados da fluorescência do triptofanos mostram um deslocamento da emissão para a região de onda do azul para o peptídeo...
In the present work was studied the structure, function and mechanism of action of the antibacterial peptide Protonectarina-MP (isolated from venom of social wasp Protonectarina sylveirae) with its carboxyamidation (-NH2) and carboxyl-free (-OH) Cterminal forms. The peptides were manually synthesized on-solid phase by using Fmoc strategy and purified under HPLC. The homogeneity of the synthetic material was analyzed by ESI mass spectrometry and Edman Degradation Chemistry. The secondary structure was investigated through circular dichroism (CD) spectroscopy and molecular modeling. Lytic activity and peptides interaction with the membranes was also investigated through tryptophan emission, by fluorescence spectrometry. The interaction of peptides with zwitterionic and anionic vesicles was investigated through the combination of H/D exchange and ESI-mass spectrometry. Some biological activities, like: mast cell degranulation, release of cytoplasmic enzyme lactate dehydrogenase, hemolysis and antibiosis were investigated for both peptides. The CD spectra revealed that the peptides in hydrophobic environments or in presence of biological membranes have the tendency to form helix conformations; however, organized structures were not observed in aqueous or buffer solutions. The models obtained by molecular modeling show that both peptides form an amphipathic α-helix. The peptides presented a positive cooperative process in the lytic activity of vesicles, with dose-response curves presenting a sigmoidal dependence with the peptide concentration. The results of the fluorescence of tryptophans showed a shift of the emission wavelength to the blue region of the peptide Protonectarina-MP (-NH2), which was not observed for its analogue presenting the C-terminal residue in free acid form. Full text[ES] En la presente Tesis Doctoral se investigó la aplicación de dos técnicas no convencionales (disoluciones salinas y campos eléctricos) para limpiar membranas de ultrafiltración que previamente habían sido ensuciadas con disoluciones modelo de lactosuero. Además, se estudió el ensuciamiento de las membranas causado por las diferentes disoluciones ensayadas, caracterizando el mismo mediante el ajuste de distintos modelos matemáticos semi-empíricos a los resultados experimentales de variación de la densidad de flujo de permeado con el tiempo. Los ensayos se realizaron a escala de laboratorio con cuatro membranas de ultrafiltración de diferente material (cerámicas y orgánicas) y umbral de corte molecular (5, 15, 30 y 50 kDa) y con tres disoluciones modelo de lactosuero consistentes en disoluciones acuosas de seroalbúmina bovina (BSA), BSA con CaCl2 y concentrado de proteínas de lactosuero (WPC). Cada ensayo fue dividido en cuatro etapas: ensuciamiento con la disolución modelo, primer aclarado, limpieza y segundo aclarado. Las condiciones experimentales durante la primera etapa fueron las mismas en todos los casos, para poder evaluar la influencia de las condiciones experimentales durante la etapa de limpieza (tipo de disolución salina, concentración de sal, temperatura de la disolución, velocidad tangencial y potencial de campo eléctrico aplicado) en la eficacia del proceso de limpieza. Los modelos matemáticos considerados fueron los modelos de Hermia adaptados a flujo tangencial, un modelo combinado basado en las ecuaciones de Hermia de bloqueo completo de poros y formación de torta y un modelo de resistencias en serie. Los resultados obtenidos durante la etapa de ensuciamiento demostraron que los modelos matemáticos utilizados son capaces de predecir con una elevada exactitud el descenso de la densidad de flujo de permeado con el tiempo, determinándose el valor de los parámetros característicos de dichos modelos y los mecanismos de ensuciamiento responsables principalmente de dicho descenso. En cuanto a los ensayos de limpieza de las membranas utilizadas, los resultados indicaron que tanto las disoluciones salinas como los campos eléctricos son técnicas efectivas para recuperar las propiedades permselectivas de las membranas, utilizando un intervalo de concentración de sal óptimo.

A més, es va estudiar l'embrutament de les membranes causat per les diferents dissolucions assajades, caracteritzant el mateix mitjançant l'ajust de distints models matemàtics semi-empírics als resultats experimentals de variació de la densitat de flux de permeat amb el temps. Els experiments es realitzaren a escala de laboratori amb quatre membranes d'ultrafiltració de diferent material (ceràmiques i orgàniques) i umbral de tall molecular (5, 15, 30 i 50 kDa) i amb tres dissolucions model de sèrum de llet consistents en dissolucions aquoses de seroalbúmina bovina (BSA), BSA amb CaCl2 i concentrat de proteïnes de sèrum de llet (WPC). Cada experiment fou dividit en quatre etapes: embrutament amb la dissolució model, primer aclarat, neteja i segon aclarat. Resumo: Domínios lipídicos são regiões de composição diferente em uma mesma membrana, quer seja biológica ou sintética. A dinâmica destes domínios pode participar de processos bioquímicos e fisiológicos. Anestésicos locais são fármacos que agem sobre proteínas de membranas por um mecanismo ainda não totalmente conhecido, e cuja potência está fortemente correlacionada com sua hidrofobicidade. Oentre estes, a dibucaína (OSC) é um potente anestésico, com alto coeficiente de partição em favor das membranas, capaz de interferir com diversos processos fisiológicos e de reverter alguns efeitos do íon cálcio, um conhecido indutor de domínios lipídicos. Investigamos a interação da OBC com membranas e o efeito deste AL sobre domínios lipídicos naturais e induzidos por cálcio. As técnicas usadas foram Fluorescência Estática, Ressonância Paramagnética Eletrônica e Microscopia de Fluorescência. Verificamos que a oac possui um pKa de 8,4 e que deve localizar-se na altura do glicerol em membranas compostas por OOPC, o que favorece sua competição com o cálcio pelas cabeças polares de lipídios, e que a forma neutra apresenta maior afinidade pela membrana (_ = 1500) que a protonada (_ = 930). A fluorescência intrínseca da molécula foi analisada em água, em membranas e em soluções de diferentes polaridades; observamos diminuição no rendimento quântico de fluorescência da OSC, concomitante a um deslocamento para o azul nos meios mais apoiares. Experimentos de Ressonância Paramagnética Eletrônica com marcadores de spin do tipo nitróxido em alta concentração (exibindo alargamento dos espectros devido à interação de troca de estado de spin) mostraram que a OBC reverte domínios lipídicos induzidos por cálcio e por magnésio, em sistemas ternários de OOPC: POPA: 5-MeSL. Por microscopia de fluorescência (usando sondas do tipo nitro-benzoxadiol amino, NBO) notamos diferenças evidentes no padrão de formação de domínios lipídicos entre os tratamentos controle, com cálcio, com OBC e com ambos os aditivos, tanto em vesículas unilamelares gigantes quanto em eritrócitos e ghosts de eritrócitos marcados. As diferenças entre os tratamentos foram qualitativamente consistentes para os parâmetros de análise das imagens utilizados. Vimos também aqui o antagonismo entre a OSC e o cálcio e, além disso, uma aparente capacidade da OSC em reverter domínios induzidos por proteínas de membrana. Nossos resultados sugerem que a OSC reverte a separação lateral de fases induzida por cálcio ou proteínas de membrana; esta capacidade da OSC - não'observada em outros anestésicos locais - estaria relacionada à localização preferencial da DSC próximo à cabeça polar dos fosfolipídios. Tal fenômeno pode ser relevante nos processos membranares em que a DSC reconhecidamente interfere, mediados pela alteração do padrão dos domínios lipídicos, dentre os quais a fusão celular e a própria anestesia
Abstract: Lipid domains are compositionally different regions within biological or artificial membranes that may play an important role in physiological and biochemical events as well. Local anesthetics, however, acts upon membrane proteins by an still unknown mechanism and exibit a strong correlation between partition to membranes and anesthetic potency. Among this class of pharmaceuticals, dibucaine (OBC) is a potent one, with a high membrane partition, able to alter several physiological processes and revert some calciuminduced effects on artificial membranes. We have investigated the interaction of neutral and protonated forms of OBC with natural and artificial membranes and its effect upon naturally and calcium-induced lipid domains both in biological and artificial membrane systems. Our approach was based on Static Fluorescence, Electronic Paramagnetic Resonance (EPR) and Fluorescence Microscopy. Our experiments reveal a pKa of 8,4 for OBC and that its aromatic ring may be located close to the glycerol backbone of phospholipid membranes. Surprisingly, OBC emission decreased simultaneously to a classical blue shift when membranes are available or by reducing its solution polarity. EPR experiments strongly suggest that OBC reverts Ca+2- or Mg+2-induced lipid domains. The parameter ais increased in ternary systems composed by OOPC: POPA: 5MeSL when OBC is present, which is antagonic to calcium ion. Fluorescence Microscopy experiments revealed distinct patterns among OBC, Ca+2 or OBC plus Ca+2 treatments, in NBO-PA labeled liposomes, ghosts and erythrocytes. Although Kruskal-Wallis test were not able to tell which treatment differences were statistically significant, those differences were consistent through ali parameters analysed. We could observe antagonism between OBC and Ca+2, besides a further ability of OBC to disrupt naturally occurring lipid domains. These data suggest that OBC is able to revert Ca+2_, Mg+2 or protein-induced domains, for its favoured location close to the phospholipid head. Such phenomena may be relevant in lipid domain mediated processes, which may include anesthesia itself

Full textThe understanding at the molecular level of the interaction of both disease-causing viruses (or part thereof, such as proteins and peptides) and drug-candidate molecules is of paramount importance for the therapy and treatment of such diseases. In this work, we use biophysical techniques to study the interaction between membrane mimetics and two groups of small molecules. The first one is related to a disease, the Dengue fusion peptide, putative sequence of Dengue virus glycoprotein E, which is conserved among other flaviviruses, such as the Zika virus. The second group consists of coordination compounds of copper, Phenantroline and the dipeptides Ala-Gly and Ala-Phe: Cu (L-dipeptide) (phenanthroline), which are potential candidates for antitumor drugs. Our results show that the molecules form both groups are capable of interacting and modifying the physic-chemical properties of the mimetic membrane system. This indicates that, on one hand, one can think in strategies of blocking the interaction to avoid virus infection and, on the other hand, one can use our results to improve membrane interaction in drug delivery mechanisms and/or for facilitating diffusion through the physical barrier represented by the membranes in tumor cells
