New planktonic stable isotope and ice-rafted debris records from three core sites in the mid-latitude North Atlantic (IODP Site U1313, MD01-2446, MD03-2699) are combined with records of ODP Sites 1056/1058 and 980 to reconstruct hydrographic conditions during the middle Pleistocene spanning Marine Isotope Stages (MIS) 9–14 (300–540 ka). Together the study sites reflect western and eastern basin boundary currents as well as north to south transect sampling of subpolar and transitional water masses. Planktonic δ18O records indicate that during peak interglacial MIS 9 and 11 hydrographic conditions were similar among all the sites with relative stable conditions and confirm prolonged warmth during MIS 11c also for the mid-latitudes. Sea surface temperature (SST) reconstructions further reveal that in the mid-latitude North Atlantic MIS 11c is associated with two plateaus, the younger one of which is slightly warmer. Enhanced subsurface northward heat flux in the eastern boundary current system, especially during early MIS 11c, is denoted by the presence of tropical planktonic foraminifer species. MIS 13 was generally colder and more variable than the younger interglacials. The greatest differences between the sites existed during the glacial inceptions and glacials. Then a north-south trending hydrographic front separated the nearshore and offshore waters off Portugal. While offshore waters originated from the North Atlantic Drift as indicated by the similarities between the records of IODP Site U1313, ODP Site 980 and MD01-2446, nearshore waters as recorded in core MD03-2699 derived from the Azores Current and thus the subtropical gyre. A strong Azores Current influence is seen especially during MIS 12, when SST dropped significantly only during the Heinrich-type ice-rafting event at the onset of Termination V. Given the subtropical overprint on Portuguese nearshore sites such as MD03-2699 and MD01-2443 caution needs to be taken to interpret their records as basin-wide climate signals.

While offshore waters originated from the North Atlantic Current as indicated by the similarities between the records of IODP Site U1313, ODP Site 980 and MD01-2446, nearshore waters as recorded in core MD03-2699 derived from the Azores Current and thus the subtropical gyre. Except for MIS 12, Azores Current influence seems to be related to eastern boundary system dynamics and not to changes in the Atlantic overturning circulation.

Abstract Several studies have shown antitumor activities of the melanoma differentiation–associated gene 7 (mda-7) and the nonsteroidal anti-inflammatory drug sulindac when used as a monotherapies against a wide variety of human cancers. However, the combined effects of mda-7 and sulindac have not previously been tested. Therefore, we tested the antitumor activity of an adenoviral vector expressing mda-7 (Ad-mda7) in combination with sulindac against non–small cell lung cancer cells in vitro and in vivo. When treated with Ad-mda7 in combination with sulindac, human lung cancer cells (A549 and H1299) underwent growth suppression resulting in apoptosis. The growth inhibition induced by Ad-mda7 in combination with sulindac was significantly greater than that observed with Ad-mda7 or sulindac alone. Furthermore, the degree of growth inhibition induced using this combination was dose-dependent for sulindac. Treatment with Ad-mda7 in combination with sulindac had no growth inhibitory effects on human normal lung (CCD-16) fibroblasts. We then investigated the mechanism by which sulindac enhances Ad-mda7-mediated apoptosis. Sulindac increased expression of ectopic MDA-7 protein in tumor cells, thereby increasing the expression of downstream effectors RNA-dependent protein kinase, p38MAPK, caspase-9, and caspase-3 and enhancing apoptosis of non–small cell lung cancer cells. Pulse-chase experiments showed that the increased expression of MDA-7 protein in sulindac-treated cells was due to increased half-life of the MDA-7 protein. Finally, treatment of human lung tumor xenografts in nude mice with Ad-mda7 plus sulindac significantly suppressed growth (P = 0.001) compared with Ad-mda7 or sulindac alone. Our results show for the first time that combined treatment with Ad-mda7 plus sulindac enhances growth inhibition and apoptosis of human lung cancer cells. The increased antitumor activity observed with the combination treatment is a result of increased half-life of MDA-7 protein. Regulation of protein turnover is a heretofore-unrecognized mechanism of this nonsteroidal anti-inflammatory drug.

Abstract Abstract 1474 Melanoma differentiation-associated gene-7/interleukin-24 (MDA7/IL-24) selectively induces apoptosis in cancer cells without harming normal cells. It also exerts immunomodulatory and anti-angiogenic effects, as well as potent antitumor bystander effects, making it an ideal candidate for use in a new anticancer gene therapy. To examine the feasibility of adeno-associated virus (AAV) vectors expressing MDA7/IL-24 in systemic cancer gene therapy for lymphoma, we generated an AAV type 8 vector expressing MDA7/IL-24 (AAV-IL24). In vitro studies showed that medium conditioned by AAV-IL24-transduced C2C12 cells induces tumor cell-specific apoptosis against murine lymphoma cell line (A20 cell). To assess the in vivo effects of muscle targeted AAV-mediated systemic delivery of MDA7/IL-24 we established a lymphoma murine model in which the A20 cells expressing luciferase gene was inoculated into the caudal vein of BALB/C. Using this lymphoma murine model, we can detect the tumor growth and metastases by a real-time in vivo imaging analyze system (IVIS). After single injection of AAV-IL24 (1.5×1011 vg/body) into the right quadriceps muscle of the lymphoma model mice, tumor cell growth was monitored by IVIS. ELISA analysis showed high level of IL-24 was detected in plasma of treated mice (263±16 ng/ml). Suppression of tumor growth was observed in AAV-IL24 injected mice compared to control GFP expressing AAV injected mice (1.3×108vs.2.6×108 photon/sec; p<0.05). Survival effect was also detected in AAV-IL24 mice (66±6 vs. 47±5 days; p<0.03). In addition, TUNEL analyses showed significant induction of tumor cell-specific apoptosis within the tumors and suppression of angiogenesis was also detected in AAV-IL24 treated mice. Finally, immune modulating activity of induction of Th1 cytokines (IL-6, TNFα, IFNγ) secretion was observed after AAV-IL24 injection. These results clearly demonstrate that continuous systemic delivery of MDA7/IL-24 can serve as an effective treatment for lymphoma. Thus, AAV type 8 vector-mediated systemic deliverY of MDA7/IL-24 represents a potentially promising new approach to treat lymphoma. Disclosures: No relevant conflicts of interest to declare.

Abstract. Thanks to its optimal location on the northern Brazilian margin, core MD09-3257 records both ocean circulation and atmospheric changes. The latter occur locally in the form of increased rainfall on the adjacent continent during the cold intervals recorded in Greenland ice and northern North Atlantic sediment cores (i.e., Greenland stadials). These rainfall events are recorded in MD09-3257 as peaks in ln(Ti ∕ Ca). New sedimentary Pa ∕ Th data indicate that mid-depth western equatorial water mass transport decreased during all of the Greenland stadials of the last 40 kyr. Using cross-wavelet transforms and spectrogram analysis, we assess the relative phase between the MD09-3257 sedimentary Pa ∕ Th and ln(Ti ∕ Ca) signals. We show that decreased water mass transport between a depth of ∼1300 and 2300 m in the western equatorial Atlantic preceded increased rainfall over the adjacent continent by 120 to 400 yr at Dansgaard–Oeschger (D–O) frequencies, and by 280 to 980 yr at Heinrich-like frequencies. We suggest that the large lead of ocean circulation changes with respect to changes in tropical South American precipitation at Heinrich-like frequencies is related to the effect of a positive feedback involving iceberg discharges in the North Atlantic. In contrast, the absence of widespread ice rafted detrital layers in North Atlantic cores during D–O stadials supports the hypothesis that a feedback such as this was not triggered in the case of D–O stadials, with circulation slowdowns and subsequent changes remaining more limited during D–O stadials than Heinrich stadials.

The experiment was conducted to evaluate the effect of eggshell powder and phosphorus on growth and yield of MD7 groundnut variety in the spring season in Phu Xuyen, Ha Noi. The two factor experiment was laid out in a split-plot design with three replications. The main plot consisted of three rates of eggshell powder (0, 300, 500 kg/ha). The sub-plot included three rates of phosphorus (30, 60, 90 kg/ha). The results showed that the growth and yield of plants fertilized with eggshell powder were higher than those with unfertilized eggshell powder. In addition, the growth traits and yield of MD7 groundnut variety increased with increasing phosphorus fertilizer doses from 30 kg P2O5/ha to 90 kg P 2O5/ha under the same fertilized or unfertilized eggshell powder conditions. Briefly, the results indicated that the rate of 500 kg eggshell powder/ha combined with 90 kg P2O5/ha was suitable for MD7 groundnut variety to grow, develop and produce the highest yield in the spring season in Phu Xuyen, Ha Noi.

A differentially displayed cDNA clone (MD17) was isolated from tobacco roots (Nicotiana tabacum cv. Xanthi-nc) infected with the arbuscular mycorrhizal (AM) fungus Glomus intraradices. The isolated DNA fragment exhibited a reduced level of expression in response to AM establishment and 90% identity with the 3′ noncoding sequence of two basic chitinases (EC 3.2.1.14) from N. tabacum. Northern (RNA) blots and Western blots (immunoblots), probed with tobacco basic chitinase gene-specific probe and polyclonal antibodies raised against the chitinase enzyme, yielded hybridization patterns similar to those of MD17. Moreover, the up-regulation of the 32-kDa basic chitinase gene expression in tobacco roots by (1,2,3)-thiadiazole-7-carbothioic acid S-methyl ester (BTH) was less effective in mycorrhizal roots than in nonmycorrhizal controls. Suppression of endogenous basic chitinase (32-kDa) expression was also observed in transgenic mycorrhizal plants that constitutively express the 34-kDa basic chitinase A isoform. When plants were grown with an increased phosphate supply, no suppression of the 32-kDa basic chitinase was obtained. These findings indicate that during the colonization and establishment of G. intraradices in tobacco roots, expression of the basic chitinase gene is down-regulated at the mRNA level.

Abstract. Sea surface temperatures (SST) in the Guiana basin over the last 140 ka were obtained by measuring the C37 alkenone unsaturation index U37'k in sediment core MD03-2616 (7° N, 53° W). The resulting dataset is unique for this period in the western tropical Atlantic region. SSTs range from 25.1 to 28.9 °C, i.e. glacial-to-interglacial amplitude of 3.8 °C, which is common in tropical areas. During the last two interglacials (MIS1 and MIS5e) and warm long interstadials (MIS5d-a), the sediments studied trace rapid transmission of the climate variability from arctic-to-tropical latitudes and vice-versa. During these periods, MD03-2616 SSTs showed a remarkable parallelism with temperature changes observed in Greenland and SST records of North Atlantic cores. The last deglaciation in Guiana is particularly revealing. MIS2 stands out as the coldest period of the interval analysed, with SSTs reaching as low as 25.1 °C. It contains reminders of northern latitude events such as the Bølling-Allerød warming and the Younger Dryas cooling which ensued. These oscillations were previously documented in the δ18O of the Sajama tropical ice core and are present in Guiana with rates of ca. 6 °C ka−1 and changes of over 2 °C. During the glacial interval, significant abrupt variability is observed; e.g. oscillations of 0.5–1.2 °C during MIS3, i.e. about 30% of the maximum glacial–interglacial SST change. Nevertheless, in the MD03-2616 record it is hard to identify unambiguously either the Dansgaard–Oeschger type of oscillations described in northern latitudes or the SST drops associated with the Heinrich events characterising North Atlantic records. Although these specific events form the background of the climate variability observed, what truly shapes SSTs in Guiana is a long-term tropical response to precessional changes, which is modulated in the opposite way to polar variability. This lack of synchrony is consistent with other tropical records in locations to the north or south of Guiana and evidences an arctic-to-tropical decoupling when a substantial reduction in the Atlantic meridional overturning circulation (AMOC) takes place.

High-resolution pollen analysis of core MD01-2430 from the Sea of Marmara (40°47.81′ N, 27°43.51′ E) allows us to reconstruct the vegetation response to climatic changes during the past 23 cal ka in the Northeastern Mediterranean. Variation in mesic/temperate forest cover indicates major climatic shifts connected to Heinrich Stadial 1, Bölling-Allerød, Younger Dryas and to the onset of the Holocene. Pollen-anthropogenic indicator approach was used to recognize human-induced landscape changes in the Sea of Marmara. The pollen-inferred onset of the Holocene occurs at ca. 11.5 cal ka, indicating that the Northeastern Mediterranean region represents a transitional zone where higher moisture availability supported an earlier forest expansion than the borderlands of the Aegean Sea and Black Sea. Three major forest retreats occurred during the Holocene at ca. 9.2, 5.5, and 2.1 cal ka while a weaker forest setback was detected at 7.9 cal ka. The Holocene forest setbacks are in phase with previously published alkenone-inferred sea-surface temperature decreases in the Sea of Marmara (core MD01-2430) reconstructed from the same archive. Our new pollen record testifies the sensitivity of Mediterranean forests to changes in moisture availability which is driven by changes in high-latitude climate drivers (North Atlantic Oscillations and/or Siberian High).

Abstract:
This study presents a stratigraphic investigation of the marine sediment core MD04-2822 from the Rockall Trough (56° 50.54' N, 11° 22.96' W; 2344 m water depth). This core is currently the only available high resolution record for the calibration of Late Quaternary sedimentary sequences of the British (Hebridean) margin. It therefore offers an unprecedented archive of changing sedimentological and climatological conditions for the last 175,000 years. The high resolution, multi-proxy records have enabled surface and deep water conditions within the Rockall Trough to be reconstructed. In addition, the fluctuating nature of ice-rafted debris (IRD) inputs to the MD04-2822 site allows a first order attempt of BIS dynamics for the entirety of the last glacial period (i.e. from the demise of the last interglacial to the decay of the Devensian/Weichselian ice sheet) as well as the majority of the penultimate (Saalian/MIS 6) glaciation. Sediment core MD04-2822 is ideally located to capture the dynamics of the British Ice Sheet (BIS) via a continuous record of IRD and fine-grained terrigenous inputs. Fundamental to this is the construction of a robust chronology. This was achieved via: the correlation of the benthic δ¹⁸O record to a global δ¹⁸O stack (SPECMAP); the correlation of the surface proxies (% N. pachyderma (sinistral) and XRF Ca) to the Greenland δ¹⁸O and Antarctic methane ice core records; and radiocarbon dating. This chronology was validated using both radiocarbon dating and tephra horizons. An evaluation of the event stratigraphy approach used in the construction of the MD04-2822 chronology is presented. The marine record provides a valuable archive of past ice sheet dynamics as much terrestrial evidence is removed or obscured by subsequent ice sheet oscillations MD04-2822 provides the first evidence for the expansion of the BIS onto the Hebridean Margin during MIS6 (thereby confirming previous long-range seismic correlations). The continuous sedimentation at MD04-2822 enabled the first insights into the early dynamics of the last BIS. Increases in IRD and fine grained terrigenous material delivered to the MD04-2822 at ca. 72 kyr represent the first significant delivery of material from the BIS across the continental shelf to the core site. The BIS would therefore have attained a marine calving margin by this time. A multi-proxy investigation of provenance was undertaken, however unequivocal provenance determinations remain problematic. The location of the core suggest the proximal BIS as the most likely source of terrigenous inputs. The expanded nature of the MD04-2822 sediments during the penultimate deglacial (Termination II) provides the first details of BIS dynamics for this period: the interplay of large inputs of freshwater from the decay of the Saalian (MIS 6) ice sheets (including the BIS) upon the surface and deep water circulation of the North Atlantic is investigated. In addition, sub-orbital climatic variability is documented at this location throughout the last interglacial (MIS 5e) and appears to be an intrinsic feature of both the N.E. Atlantic surface and deep water circulation of the last 175 kyr.

Abstract:
In this thesis we prove the linear stability to gravitational and electromagnetic perturbations of the Reissner-Nordström family of charged black holes with small charge. Solutions to the linearized Einstein-Maxwell equations around a Reissner-Nordström solution arising from regular initial data remain globally bounded on the black hole exterior and in fact decay to a linearized Kerr-Newman metric. We express the perturbations in geodesic outgoing null foliations, also known as Bondi gauge. To obtain decay of the solution, one must add a residual pure gauge solution which is proved to be itself controlled from initial data. Our results rely on decay statements for the Teukolsky system of spin +/-2 and spin +/-1 satisfied by gauge-invariant null-decomposed curvature components, obtained in earlier works. These decays are then exploited to obtain polynomial decay for all the remaining components of curvature, electromagnetic tensor and Ricci coefficients. In particular, the obtained decay is optimal in the sense that it is the one which is expected to hold in the non-linear problem.

This study employs the western Philippine Sea MD06-3049 core (16.35 ° N, 124.39° E), collected by the 2006 Marco-Polo II cruise, to measure multi-magnetic parameters and to conduct grain size analysis for reconstruction of the paleoenvironment records. The water depth for the core is about 2498 meters and the total recovered length is 32.28 meters. We determine the Brunhes/Matuyama boundary, the Jaramillo subchron, the Olduvai subchron and the Reunion subchron from the characteristics declination and inclination curves in the nature remnant magnetization of the core. In addition, with supplement of the paleointensity low value for comparison, we can differentiate excursions as age control points to construct age model, suggesting that the age span of the core is about 2.4 Ma. The magnetic parameters analysis of the magnetic mineral composition in the core MD06-3049 indicates that the main magnetic mineral is the pseudosingle-domain titanomagnetite. The results of grain size analysis show that the mean grain size positively correlates with the magnetite grain size --- both are bigger during glacial and smaller during interglacial. By using the mean grain size of 10~63μm sediment as an indication of bottom flow speed, we also show that the bottom flow strengthen during glacial and decline during interglacial. The grain size of core MD06-3049 is affected by the changing velocity of the glacial-interglacial bottom flow. The characteristics of continuous records and stable sedimentary rates of marine cores are good for studying whether the paleointensity has orbital cycle or not. Using the anhysteresis remanent magnetization and paleointensity of the MD06-3049 core for spectrum analysis, we find that the anhysteresis remnant magnetization has a main cycle 100kyrs close to that of the Earth’s orbital eccentricity while the paleointensity has not. Although the ratio of nature remanent magnetization and anhysteresis remanent magnetization is used as relative variation of the paleointensity, the signal of anhysteresis remnant magnetization signal still controlled by the grain size of the magnetic mineral. It shows that the cyclical changes of paleointensity may result from changes of grain size, capturing the environmental messages.

A presente dissertação vem no seguimento de algumas questões que foram levantadas numa anterior dissertação de Mestrado, nomeadamente a referência à possível existência de variações morfometricas significativas nos cocólitos de Calcidiscus leptoporus. Desta forma, o principal objectivo do presente estudo foi a identificação das variações morfométricas dos cocólitos da espécie Calcidiscus leptoporus s.l. ao longo de uma série temporal de alta resolução, recorrendo para tal à Analise Multivariada de Morphons (AMM). O estudo foi efectuado com base num troço da sondagem MD01-2443, recolhida na margem ibérica e representativa do estádio isotópico marinho 11. Este apresenta diversas semelhanças com o Holocénico, nomeadamente a geometria orbital e a concentração de gases com efeito de estufa (período pré – industrial) o que permite fazer algumas comparações ao nivel das variações climatéricas e das condições oceanográficas vigentes, com o presente período interglaciar. Um segundo objectivo foi a identitificação e contabilização de uma espécie característica da zona fótica inferior, a Florisphaera profunda. Com base na sua variação ao longo da série temporal seria então possível determinar o índice de produtividade N, que correlaciona a variações de abundância entre espécies da zona fortica superior e inferior. A Analise Multivariada de Morphons permitiu a identificação de 3 morfótipos da espécie Calcidiscus leptoporus presentes na série temporal analisada. Apesar de ligeiramente diferentes, as classes dimensionais encontradas são semelhantes às anteriormente descritas por vários autores. Desta forma comprovou-se a existência de um morfótipo pequeno comdimensões que variam entre os 4,0 μm e os 6,9 μm; um segundo morfótipo intermédio cujo diâmetro mede entre 8,0 μm e 9,9 μm e finalmente um morfótipo grande cujo diâmetro apresenta dimensões superiores a 11,0 μm. Em seguida procurou atribuir-se um significado paleoeoceanografico e/ou paleocologico aos morfótipos identificados. Pelo que foi elaborada uma Analise Factorial da comunidade cenotica presente na sondagem à qual se incluíram os 3 morfótipos de C. leptoporus. Esta permitiu a interpretação paleoceanográfica para a margem ibérica durante o MIS 11.
This work follows on some issues that were raised at a previous MSc in particular possible presence of morfometric variation among Calcidiscus leptoporus coccoliths. Thus, the main goal of this study was to identify the morphometric variations of coccoliths of the species Calcidiscus leptoporus s. l. along a series of high resolution, using the multivariate Morphons analysis (MMA). The study was based on a section of core MD01- 2443, taken in the Iberian margin and representative of the marine isotope stage 11. Similar orbital geometry and greenhouse gas concentrations (pre – industrial period) during marine isotope stage 11 (MIS 11) and allow us to make some comparisons between climatic changes weather and oceanographic conditions prevailing in the present interglacial stage. A second intention was to identity and quantify the lower photic zone, the Florisphaera profunda species. Based on its variation over the time series would then be possible to determine the productivity index N, which correlates to variations in abundance between deep fotic zone species and lower fotic zone species. Multivariate Morphons alalysis permitted the identification of 3 morphotypes of the species Calcidiscus leptoporus present in the time series analyzed. Although slightly different, sizeclasses found are similar to those previously described by several authors. Thus proved the existence of a small morphotype with size dimentions ranging between 4.0 μm and 6.9 μm; a second intermediate morphotype diameter ranges between 8.0 μm and 9.9 μm and finally a large morphotype which has diameter larger than 11.0 μm. Then sought to give paleoeoceanografic and / or paleocologic meaning to the morphotypes identified. Which was drafted by a Factorial Analysis of the kenotic community present in the survey that included the 3 morphotypes of C. leptoporus. This allowed a Paleoceanographic interpretation to the Iberian margin during the MIS 11 based on the nanofóssil community.

We have generated both benthic and planktonic foraminiferal oxygen and carbon isotope records of IMAGES (International Marine Past Global Changes Studies) core MD05-2922 from Ontong Java Plateau (4°59.28 S, 156°08.80 E, 263cm long, water depth 1460m). Based on the 18O records of benthic foraminifera Cibicidoides wuellerstorfi, an age model is established for the past 180 ka. The circulation of Antarctic Intermediate Water is thought to make an important contribution to the global ocean–climate system, but the details of this interaction are not fully understood especially in western Pacific. Difference in 13C values between core MD05-2922 and core MD97-2120 (near the source region) during glacial times were less than those during interglacial times indicating that the influence of AAIW increased during glacial times. However, it may not be due to the strengthened of AAIW but due to the weakened of other water masses in this region. The carbon isotope minimum event at the beginning of glacial terminations is a common feature record in planktonic foraminiferal carbon isotopic records. The 13C minimum records in eastern equatorial pacific were thought due to the enhancement of formation of Antarctic Intermediate Water. Our carbon isotopic data of G. sacculifer and P. obliquiloculata appeared the same patterns during deglacial period. However, we inferred the events in western Pacific were cursed by abruptly increased nutrient or organic matters from continental shelf transported to ocean when sea level rises.

Stable isotope data from Solomon Sea core MD05-2925 (9°20.61'S, 151°27.61'E, 28.43m in length, 1642m water depth) are presented from planktonic foraminifera Globigerinoides ruber, Globigerina sacculifer and Pulleniatina obliquiloculata (top 10m) . The age model for core MD05-2925 is based on correlation of the G. sacculifer oxygen isotope record to the Low Latitude Stake chronology and five radiocarbon dating. In order to reconstruct the hydrology during the last two glacial terminations in the south margin of the Western Equatorial Pacific Warm Pool, we use multiple species approach of planktonic foraminifera as the proxy of water column structure. We compare the oxygon and carbon isotope records of the surface-dwelling foraminifers G. ruber and G. sacculifer and thermocline dwelling P. obliquiloculata with the cores in the eastern equatorial Pacific upwelling region, central Western Equatorial Pacific Warm Pool, and Antarctica. We demonstrate that during the last two glacial terminations, there are obvious 13C minimum events related to the South Ocean warming and CO2 concentration increment. With the support like the oxygen isotope records , we demonstrate that thermocline in the south region of the western Pacific Warm Pool was deepen and was warmer during the last two terminations.
