Abstract Abstract 4863 THE SECONDARY TRANSFORMATION OF MDS INTO MYELOFIBROSIS IS INDEPENDENT OF THE JAK2 MUTATION Introduction The exact significance of the JAK2 V617F mutation in MDS is unclear. It has previously been suggested that MF could be associated with MDS as one phenotype of myeloproliferative disorders. Ohyashiki et al in 2005 showed that two of six patients with MDS terminating in MF had the mutation at the time of MF, while no MDS patient without MF had the JAK2 V617F mutation, suggesting that MDS patients with the JAK2 V617F mutation may be responsible for secondary MF in MDS patients. We conducted the following experiments to further probe the role of JAK2 mutation in the pathology of MDS with myelofibrosis at our institution. Methods DNA samples of 8 patients in our institution who had MDS with subsequent myelofibrosis were isolated. Genomic DNA isolated using DN easy tissue kit(Qiagen) was amplified by PCR. After confirmation of DNA presence on 1 % agarose gel and purification using QIA quick PCR purification kit (Qiagen), gene sequencing was done. This sequencing was then analyzed for G to T mutation V617F in JAK2. Results We found no evidence of the JAK2 mutation in any of the eight samples. Discussion Given the fact that nearly half the patients with myelofibrosis carry the JAK2 V617F mutation, the possibility of an association between those patients who have myelodysplasia and subsequent myelofibrosis needs to researched. Our cohort which consisted of 8 patients showed no evidence for any such correlation. This evidence confirms the presence of different pathophysiological mechanisms for phenotypic expression of MDS and primary MF and also leads us to suggest that the expression of JAK2 is not the driving mutation in the pathogenesis of MDS progression to myelofibrosis. Clinical decision support is an emerging type of healthcare information technology that aims to actively guide doctors' decision-making processes. In its various forms, it can help physicians design treatment regimens, regulate dosage, avoid potentially harmful drug interactions and allergies, order preventative screenings, and even establish a diagnosis. This project is a journalistic investigation of this new technology, its applications, and its effect on the medical profession. It examines two decision support systems, Partners Healthcare's clinical reminder system, and Logical Images' product VisualDx, in order to explore the potential for these technologies and how their use may change the practice of medicine. Through extensive interviews of experts in medicine, healthcare IT, and healthcare policy, it considers the major problems in implementing decision support, with emphasis on how the technology may affect doctors' autonomy, and how physicians' financial and professional incentives may influence how it is used.

Le curve B-spline comunemente usate in modellazione geometrica sono costituite da segmenti polinomiali aventi tutti lo stesso grado. Nella pratica, è spesso necessario modellare curve composte da segmenti polinomiali di grado differente. In questi casi l’utilizzo delle curve B-spline classiche complica inutilmente la modellazione ed necessita di informazioni superflue alla descrizione matematica della curva. Le curve MD-Spline (Multi Degree Spline) sono una generalizzazione delle curve B-spline costituite da segmenti polinomiali di gradi differenti. Con le MD-Spline un progettista CAD ha la possibilità di gestire il grado di ogni segmento in maniera indipendente dagli altri e può rappresentare una determinata forma specificando solo le informazioni strettamente necessarie a definirla. La conseguenza è che modellare con curve MD-Spline si rivela più semplice e rapido rispetto a quanto possibile con curve B-spline. La tesi tratta le MD-Spline descrivendone gli aspetti teorici alla base ed i principali algoritmi di modellazione. In particolare il lavoro di tesi ha prodotto il primo software di modellazione con curve MD-Spline.

The diploma thesis handles economic analysis of the company MD logistika, a.s. The work is divided into theoretical and practical part. The theoretical part describes the indicators of profitability, liquidity, activity, debt, costs and market indicators. DuPont analysis breaking down ROE was used and was followed by a more detailed decomposition of ROA. Within the value indicatiors was used economic value added, which was calculated by two methods. The credibility, bankruptcy and credibility - bankruptcy models were applied to evaluate the company. The operational leverage, financial leverage and more detailed analysis of stocks analysis were performed. In the practical part each procedure, which was elaborated in the theoretical part of company, was applied to the MD logistika, a.s. In the conclusion the company was compared with the closest competitors and the industry averages.

Abstract:
Knowledge about the structure, conformation and dynamics of carbohydrates is important in our understanding of the way carbohydrates function in biological systems, for example in intermolecular signaling and recognition. This thesis is a summary of five papers studying these properties in carbohydrate-containing molecules with NMR spectroscopy and molecular dynamics simulations. In paper I, the ring-conformations of the six-membered rings of two carbaiduronic analogs were investigated. These carbasugars could potentially be used as hydrolytically stable mimics of iduronic acid in drugs. The study showed that the equilibrium is entirely shifted towards the 4C1 conformation. Paper II is an investigation of the conformational flexibility and dynamics of two (1→6)-linked disaccharides related to an oligosaccharide epitope expressed on malignant tumor cells. In paper III, the conformational space of the glycosidic linkage of an alfa-(1→2) linked mannose disaccharide present in N- and O-linked glycoproteins, was studied. A maximum entropy analysis using different priors as background information was used and four new Karplus equations for 3JC,C and 3JC,H coupling constants, related to the glycosidic linkage, were presented. Paper IV describes a structural elucidation of the exopolysaccharide (EPS) produced by Streptococcus thermophilus ST1, a major dairy starter used in yoghurt and cheese production. The EPS contains a hexasaccharide repeating unit of d-galactose and d-glucose residues, which is a new EPS structure of the S. thermophilus species. In paper V, the dynamics of three generations of glycodendrimers were investigated by NMR diffusion and 13C NMR relaxation studies. Three different correlations times were identified, one global correlation time describing the rotation of the dendrimer as a whole, one local correlation time describing the reorientation of the C-H vectors, and one correlation time describing the pulsation of a dendrimer branch.

t The pink bollworm, Pectinophoragossypiellais a key pest of cotton world-wide. In Israel mating disruption sex pheromone is used in all cotton fields and recent repeated outbreaks of the pest populations has suggested a change in the population sex pheromone characteristics. The research goals were to (1) determine the change in pheromone characteristic of PBW females after long experience to Mating Disruption (MD), (2) to test the male’s antennae response (EAG) to pheromone characteristics of laboratory, naive females, and of field collected, MD experienced females, (3) to analyse the biosynthetic pathway for possible enzyme variations, (4) to determine the male behavioural response to the pheromone blend involved in the resistance to MD. The experiments revealed that (1) MD experienced females produced pheromone blend with higher ZZ ratio than lab reared (MD naive females) that typically produced ZZ:EE ratio of 1:1. (2) Male’s origin did not affect its response to pheromone characteristics of lab or field females. (3) A transcriptome study demonstrated many gene-encode enzymes in the biosynthetic pathway, but some of the transcripts were produced in differing levels in the MD resistant populations. (4) Male origin (field or lab) influenced males’ choice of mate with strong preference to females sharing the same origin. However, when MD was applied, males of both populations were more attracted to females originated form failed MD treated fields. We conclude that in MD failed fields a change in the population mean of the ratio of the pheromone components had occurred. Males in these fields had changed their search “image” accordingly while keeping the wide range of response to all pheromone characteristics. The change in the pheromone blend is due to different level of pheromone related enzyme production.

Abstract:
Because where potentially serious damage is found it is dug and dealt with, models that assess damage severity have no relevance in that context. But, as in-line inspection tools become better adept in identifying and characterizing potential damage, models will be needed to screen the ILI results, to determine whether a dig is warranted, and when that must happen. While criteria like B31G might be used to assess metal loss due to a gouge, the usual complexity of mechanical damage cannot be assessed simply in terms of length and depth, so validated models that can account for the complexity are required. Such models could then be evaluated parametrically to develop prioritization guidelines useful with modern inspection technologies. PRCI Projects MD-4-3 and MD-4-4 were initiated to develop such models, with the results of full-scale testing generated in Project MD-4-1 serving as the basis to assess their validity. This report presents the work done as part of PRCI Project MD-4-4, which targeted an improved model for predicting time and/or cycle-dependent failure of dent and gouge damage. In addition to meeting that objective, this project also developed the basis to predict immediate failure consistent with the scope of Project MD-4-1, which as of this reporting had not emerged from the work of Project MD-4-3. Technology framed in the context of PR3-9305 was used to predict the immediate failure due to burst testing of gouged pipe done as part of MD-4-1, and found to accurately predict those collapse-controlled failures. Technology framed in that same context also was used to predict the delayed failure due to fatigue that developed in cyclic pressure testing of gouged pipes that also was done as part of MD-4-1, and found to reasonably predict those outcomes. However, there were some results that indicated the need to better quantify the crack driving force in terms of re-rounding, and the population of defects that can develop when the damaged pipe re-rounds in the wake of damage.
