Resumo: As alterações no padrão de sangramento são praticamente universais em usuárias de métodos contraceptivos contendo somente progestógenos, sendo uma importante causa de descontinuação destes métodos. Este estudo foi desenvolvido com o intuito de se avaliar as quimiocinas Interleucina 8 e 6 CKine no endométrio de usuárias do sistema intra-uterino liberador de levonorgestrel e correlacioná-las com as populações de leucócitos, células natural killer uterinas e mastócitos. Amostras de endométrio foram obtidas de dois grupos de usuárias do sistema intra-uterino liberador de levonorgestrel: 11 mulheres em amenorréia e 15 mulheres com algum tipo de sangramento. Os leucócitos foram determinados através de anticorpos específicos para neutrófilos, células natural killer uterinas e mastócitos. Foi realizada imuno-histoquimica para identificação das quimiocinas. Os neutrófilos encontravam-se em pequeno número, não havendo diferença entre os dois grupos. As células natural killer uterinas encontravam-se em número maior no grupo com sangramento (p<0,0001). Não houve diferença entre o número total de mastócitos e mastócitos ativados, porém houve um aumento da área extracelular positiva para triptase (p<0,05) nas mulheres com sangramento. As quimiocinas Interleucina-8 e 6CKine mostravam-se abundantes no estroma e no epitélio, não havendo diferença entre os dois grupos. Houve maior número de células natural killer uterinas e maior área extracelular positiva para triptase em pacientes que apresentavam sangramento. Apesar de não haver diferença entre o número total de mastócitos e mastócitos ativados e das quimiocinas Interleucina-8 e 6CKine, as células natural killer uterinas e os produtos dos mastócitos podem ter exercido papel importante na perda da integridade endometrial em usuárias do sistema intra-uterino de levonorgestrel
Abstract: Changes in the bleeding pattern are almost universal in women using progestin-only methods of contraception, and are a frequent cause of discontinuation of these methods. The present study was designed to assess endometrial chemokines in users of the Levonorgestrel intrauterine system and correlate them with leukocyte populations, uterine natural killer cells, and mast cells. Endometrium was obtained from two groups of Levonorgestrel intrauterine users: 11 amenorrhoeic women and 15 women with some form of bleeding pattern. Leukocytes were assessed using specific antibodies for neutrophils, uterine natural killer cells and mast cells. Immunohistochemistry was performed to locate chemokines 6Ckine and Interleukin-8. Neutrophils were few and without differences between the two groups. Uterine natural killer cells were significantly higher in the bleeding group (p< 0.0001). There was no difference between the total number of mast cells and activated mast cells but there was a greater extracellular area stained for mast cell tryptase (p<0.05) in women with bleeding. Chemokines 6CKine and Interleukin-8 were abundant in the stroma and in the epithelium and there was no difference between the groups. We observed more uterine natural killer cells in users with bleeding, and a greater extracellular area stained for mast cell tryptase, although there were no differences between the number of mast cells and activated mast cells or the chemokines 6CKine and Interleukin-8. Uterine natural killer cells and mast cells products may play a role in provoking break-through bleeding in long term users of the levonorgestrel intrauterine system

Full textMast cell tumor (MCT) is one of the most frequent tumors in dogs, accounting for 16 to 21% of all cutaneous tumors in this species. Histological grading is considered the standard evaluation method to access the prognostic of MCTs. However, its varied biological behavior highlights the need of complementary methods for an accurate evaluation. Currently, a number of studies proposing new molecular markers as prognostic factors in canine MCTs is identified in the indexed literature, but few are consolidated in the clinical and/or diagnostic routine. The aim of this study was to investigate the prognostic value of immunohistochemical markers published in scientific articles through a systematic review and meta-analysis, as well as to evaluate the prognostic value of vascular endothelial growth factor (VEGF) in canine MCTs. The systematic review process has started with 124 studies identified in 5 databases and, after applying the inclusion and exclusion criteria, 7 articles were selected for the meta-analysis. The results demonstrated that the cell proliferation marker Ki67 can be considered a good prognostic factor in canine MCTs. However, poor quality of the articles and insufficient published data made it impossible to evaluate other markers, including the tyrosine kinase receptor KIT. In the retrospective study, immunoexpression of KIT, Ki67 and VEGF markers was evaluated in cutaneous MCTs from 43 dogs. The Ki67 marker showed a significant correlation with the mortality and survival rates, and the KIT marker showed a significant difference in survival between the membranous and diffuse cytoplasmic patterns groups, but no differences were observed in VEGF. In conclusion, these studies validated the cell proliferation marker Ki67 and the KIT tyrosine kinase receptor as major prognostic factors in canine MCTs, whereas VEGF was not indicated. Finally, the systematic review allowed a critical analysis of the quality of the published studies in this area, emphasizing the importance of the standardization of analytical methodologies and the complete documentation of clinical, pathological and statistical data used in controlled studies in veterinary oncology.

Full textThe Mast cells are cells that are found predominantly in the perivascular spaces of almost all tissues and that are easily recognizable for the cytoplasmic contents into metachromatic granules when they are stained by Giemsa or with Toluidine Blue. These granules contain numerous vasoactive and pro-inflammatory mediators that are released after mast cell activation by binding with the receptor for IgE by allergens or other factors. Mast cells derived from multipotent haematopoietic precursors bone marrow, which through the peripheral blood, migrate into the tissues where they differentiate into mature mast cells, under the influence of IL-3 and in particular of the Stem cell factor, which represents the KIT ligand (CD117) , a tyrosine kinase receptor constitutively expressed on mast cells and their precursors. The Mastocytosis is a clonal disorder characterized by an accumulation of mast cells in various organs (skin, bone, gastrointestinal tract, lymph nodes and spleen) and is related in most cases to a point mutation of the gene that encodes the receptor KIT. It differs fundamentally a form of cutaneous mastocytosis (MC), the most frequent and typical of childhood, and systemic mastocytosis (SM), involving at least one organ other than skin. There are no epidemiological data on the prevalence of mastocytosis in the general population: in a study reported 0.66 new cases of maculopapular cutaneous mastocytosis (urticaria pigmentosa-OP) / anno/100.000 inhabitants, while there are no data on the incidence of MS, and for the diagnostic difficulty is probably underdiagnosed disease. The mastocytosis can affect any age and male / female ratio reported varies from 0.8 to 1.5%. This is mostly a sporadic but rare forms are also described family.
