Abstract:
As anfetaminas, grupo de moléculas derivadas da anfetamina, são fármacos estimulantes do sistema nervoso central, e possuem, entre outras, atividade antidepressiva sendo sua ação baseada na inativação da enzima monoamina oxidase (MAO) a qual catalisa a desaminação oxidativa de neurotransmissores, como por exemplo, a serotonina e a noradrenalina. Esta enzima pode ser encontrada em duas diferentes isoformas, a MAO A e MAO B. Este trabalho teve como objetivo o estudo de uma série de derivados anfetamínicos os quais apresentam diferente seletividade e diferentes valores de IC50, variando desde moléculas muito potentes, pouco potentes, até inativas, através da aplicação de técnicas de química-quântica no cálculo de descritores moleculares, bem como a aplicação da mecânica clássica na descrição das interações ligante-receptor a partir de estudos de docking e simulações de dinâmica molecular. Inicialmente, foram aplicados os métodos de química-quântica AM1, HF, DFT (com os funcionais B3LYP e BP86) e MP2 para a determinação do nível de teoria mais apropriado para a otimização da geometria desta série de compostos a partir da comparação entre os resultados teóricos e de raios-X obtidos para a molécula MDMA. O método HF/6-31G(d,p) mostrou os melhores resultados. Desta forma, este método foi utilizado na obtenção das geometrias de mínimo das demais moléculas em estudo. Posteriormente, estes métodos foram utilizados no cálculo das propriedades estruturais, eletrônicas e físico-químicas, distribuição de cargas derivadas do potencial eletrostático, momento dipolar, energia total, energia dos orbitais de fronteira, GAP de energia entre o orbital ocupado de mais alta energia (HOMO) e o orbital desocupado de mais baixa energia (LUMO), dureza, potencial químico, eletronegatividade, eletronegatividade absoluta e eletrofilicidade. Assim, foram verificadas possíveis relações entre os descritores calculados e a atividade biológica determinada por Scorza et al, Hurtado-Guzmán et al and Sterling et al.esta para esta série de compostos. A análise destes resultados foi feita através da aplicação da técnica quimiométrica de análise de componentes principais para, desta forma, verificar-se o agrupamento dos compostos segundo a presença ou ausência de atividade biológica na série estudada. Entretanto, não foi possível se identificar um padrão de agrupamento para as moléculas ativas e inativas, sugerindo assim, que estes descritores não são os mais adequados para a descrição da atividade antidepressiva destes compostos. Em uma segunda etapa, foram feitos estudos de docking para seis diferentes estruturas da MAO B (1OJA, 1OJ9, 2BK3, 2V5Z, 2V60 e 2V61) e cinco da MAO A (2BXR, 2BXS, 2Z5X e 2Z5Y) todas disponíveis no banco de dados de proteína, PDB, no qual se avaliou o modo de interação do ligante no sitio ativo da proteína. Os resultados de docking para a MAO B mostraram que o tamanho do inibidor é importante para uma correta interação no sítio ativo da enzima, tendo-se em vista que o tamanho da cavidade catalítica é dependente da conformação do aminoácido isoleucina 199 e, que a conformação deste aminoácido está relacionada com o tamanho do ligante. Desta forma, a escolha correta da estrutura da proteína torna-se importante para uma correta descrição do sistema. Os resultados sugerem ainda que o átomo do ligante, no qual ocorre a reação com o receptor, deve estar a uma distância média de 3,5 Å do sitio reativo. Os resultados de MD mostraram que este aminoácido é flexível na ausência de um inibidor, onde sua conformação varia entre as chamadas formas aberta e fechada ao longo do tempo, porém, quando o estudo é feito na presença de um ligante, sua conformação perrnanece, de modo geral, constante.
The amphetamine family of drugs is central nervous system stimulant drugs. Amphetamine inhibits the monoamine oxidase enzyme (MAO, isoforms A and B) which catalyzes the oxidative deamination of the neurotransmitter, e. g., serotonin and noradrenalin. In the present study, the aim was to understand the main features of a series of amphetamines derivatives, which have different substrate selectivity and a good range of activity varying from very potent to low potent compounds, even inactive molecules by the application of quantum chemistry techniques to calculate the molecular descriptors, as well as the application of a classical mechanics to describe the ligand-receptor interactions from docking studies and molecular dynamics (MD) simulation. First of all, was applied the AM1, HF, DFT (B3LYP and BP86 functionals) and MP2 quantum chemical methods to analyze which theoretical level is more appropriated for the molecular geometry optimization of this series of compounds from a comparison between the theoretical results for MDMA molecule and it\'s X-ray data. The HF/6-3lG** calculations produced results in close agreement with X-ray crystallography. Thus, was employed the same method for the molecular optimization of the other compounds in the series under investigation. Furthermore, the same method was applied to calculate quantum-chemical parameters (atomic charges, total energy, highest occupied molecular orbital -HOMO- lowest unoccupied molecular orbital -LUMO- dipole moments, hardness, electronegativity, chemical potential, absolute electronegativity and electrophilicity). It was examined possible correlations between the theoretical parameters as calculated by us with the biological activity results as reported by Scorza et al, Hurtado-Guzman et al and Sterling et al. The chemometric technique of Principal Component Analysis for the quantum chemical parameters of these compounds was employed in order to identify some pattern of grouping between the active and inactive molecules. However, it was not possible to identify a pattern between active and inactive compounds suggesting that these above-mentioned parameters are not the best descriptors to evaluate the antidepressant activity for this group of molecules. Later, a docking study was performed for six different PDB structure of MAO B (1OJA, 1OJ9, 2BK3, 2V5Z, 2V60 e 2V61) and five different structure of MAO A (2BXR, 2BXS, 2Z5X e 2Z5Y). It was possible to analyze the ligand-receptor interaction and, according to the bind site size, the activity molecules showed a 3.5A distance between the reactive atoms of the inhibitor and of the protein. Because the conformation of the isoleucine 199 (Ile 199) amino acid can change, the chosen of the correct PDB structure is important for a write description of that interaction. The MD results for the 1OJA structure showed that the Ile199 is flexible in the absence of an inhibitor, where its conformation varies between so-called closed and open forms over the simulation time. However, when the study was done in the presence of a ligand, its conformation remains by generally constant.

Abstract:
The breakup of the Union of Soviet Socialist Republics (USSR) in 1991 has had a variety of significant repercussions on Chinese politics, foreign policy, and other aspects. This doctoral project examines the evolution of Chinese intellectual perceptions of the Soviet Union in the 1980s and 1990s, before and after the collapse. Relying on a larger body of updated Chinese sources, this thesis will offer re-evaluations of many key issues in post-Mao Chinese Sovietology. The following topics will be explored or re-examined: Chinese views of Soviet policies in the early 1980s prior to Mikhail Gorbachev’s assumption of power; Chinese perceptions of Gorbachev’s political reform from the mid-1980s onward, before the outbreak of the Tiananmen Incident in 1989; Chinese scholars’ evolving views on Gorbachev from the 1980s to 1990s; the Chinese use of Vladimir Lenin and his policies in the early 1980s and early 1990s for bolstering and legitimizing the CCP regime after the Cultural Revolution and the Tiananmen Incident, respectively; and the re-evaluations of Leonid Brezhnev and Joseph Stalin since the mid-1990s. First, the thesis argues that the changing Chinese views on the USSR were not only shaped by the ups-and-downs of Sino-Soviet (and later Sino-Russian) relations, China’s domestic political climate, and the political developments in Moscow. Even more importantly, views changed in response to the earth-shaking event of the rise and fall of world communism in the last two decades of the 20th century. Second, by researching the country of the Soviet Union, Chinese Soviet-watchers did not focus on the USSR alone, but mostly attempted to confirm and legitimize the Chinese state policies of reform and open door in both decades. By examining the Soviet past, Chinese scholars not only demonstrated concern for the survival of the CCP regime, but also attempted to envision the future direction and position of China in the post-communist world. This included analysis of how China could rise to be a powerful nation under the authoritarian one-party rule, without succumbing to Western democracy and the sort of collapse that doomed the USSR. In short, Chinese research on Soviet socialism has primarily served to trace the current problems of Chinese socialism, in order to legitimize their solutions – rather than a truth-seeking process devoted to knowledge of the Soviet Union.

Abstract:
Northern Mao is an endangered Afroasiatic-Omotic language of western Ethiopia with fewer than 5,000 speakers. This study is a comprehensive grammar of the language, written from a functional/typological perspective which embraces historical change as an explanation for synchronic structure. The grammar introduces the Northern Mao people, aspects of their culture and history, and the major aspects of the language: contrastive phonology, tone phenomena, nouns, pronouns, demonstratives, numerals, noun phrases, verbs and verbal morphology, single verb constructions, non-final/medial clauses, subordinate clauses and alignment. The tone system has three contrastive levels, where the Mid tones subdivide into two classes which historically derive from two different sources. Nouns each exhibit two tonal melodies: one melody in citation form or other unmodified environments and another melody when syntactically modified. Extensive coverage is given to developments in the pronominal and subject-marking systems as well as the verbal system. In the pronominal and subject marking systems, innovations include the development of a dual opposition, the fusion of an affirmative verbal prefix to subject prefixes, and the development of these subject prefixes into new pronouns. In the verbal system, innovations include the development of new verbal wordforms from subordinate + final verb periphrastic constructions and a set of new subject markers from an old subordinator morpheme. The verbal system is oriented around two oppositional relations: realis vs. irrealis and finite vs. infinitive verb forms. Realis and irrealis verbs have distinct item-arrangement patterns: realis verbs take subject prefixes while irrealis verbs take subject suffixes. Realis is associated with affirmative polarity and non-future tense and may be used with many aspectual distinctions. Irrealis is associated with negative polarity, future tense, and counterfactual constructions; irrealis verbs do not express many aspectual distinctions. Finite versus infinitive verb stems are differentiated by tone. Finite verb stems are used in affirmative declarative and interrogative utterances, non-final/medial constructions and the more finite subordinate clause structures. Infinitive verb stems are used in negative declarative and interrogative utterances, non-final/medial constructions and the less finite subordinate clause structures. The work concludes with a summary of cross-constructional alignment patterns and evaluates the efficacy of a marked-nominative analysis.

Monoamine oxidase (MAO) is a flavin adenine dinucleotide (FAD)-containing enzyme attached to the mitochondrial outer membrane of neurons, glia, and other cells. Its roles include regulation of the levels of biogenic and xenobiotic amines in the brain and peripheral tissues by catalyzing their oxidative deamination. On the basis of their substrate and inhibitor specificities, two isoforms of MAO have been described (A and B). Due to their role in the metabolism of catecholamines neurotransmitters, MAO-A and MAO-B have long been of pharmacological interest. Accordingly, and reversible and irreversible inhibitors of MAO-A and MAO-B have been used in the clinics to treat neurological disorders including depression and Parkinson´s disease. Since the demonstration that I2- imidazoline sites are associated with mitochondrial membranes 15 years ago, several studies have provided evidence that these sites represent regions on MAOs. In line with this view, it has been demonstrated that imidazoline derivatives inhibit MAO activity. This effect has been attributed to a high affinity I2 binding site on MAO-B (I2B) and to a similar lower affinity site on MAO-A (I2A). This study investigated the effect of 4,5-dihydro-1H-imidazole-2-substituted compounds on MAO activity in vitro by spectrophotometric and fluorimetric methods using kynuramine as substrate. Among the compounds that inhibited MAO-A (3c-e, 3j), compound 3d was 73-fold more selective towards MAO-A than MAO-B. Among the compounds that selectively inhibited MAO-B (3g-I, 3k, 3o), imidazoline 3g was shown to be potent with Ki value of 5,3 μM. Some of compounds that selectively bind to I2-sites, such as 3l (benazoline), 3n (2-BFI), and 3p (BU224) showed good inhibitory activity especially against MAO-B. Imidazolines inhibited MAO-A and MAO-B activities in liver with less selectively than in rat brain. The compounds 3d and 3g reversibly inhibited MAO, and kinetics studies showed that compound 3d and 3g inhibited MAO in a mixed manner (decreased Vmax and increased Km values). These results confirm that imidazolines inhibit MAO activity and suggest a relationship between I2 binding site and modulation of central MAO
A monoamina oxidase (MAO) é uma enzima que contém o dinucleotídeo adenina-flavina (FAD) e que está presente na membrana externa da mitocôndria de células neuronais, glia e outras células. Seu papel inclui a regulação dos níveis de aminas biogênicas e xenobióticas no cérebro e em tecidos periféricos pela desaminação oxidativa. Com base na especificidade a substrato e inibidores, são descritas duas isoformas da MAO (A e B). Devido aos seus papéis no metabolismo das catecolaminas neurotransmissoras, a MAO-A e a MAO-B são consideradas farmacologicamente interessantes, e inibidores reversíveis e irreversíveis destas isoformas são usados clinicamente para tratar doenças neurológicas incluindo depressão e doença de Parkinson. Nos últimos 15 anos, desde a demonstração que sítios I2 estão associados com frações da membrana mitocondrial, muitos estudos provem evidências de que estes sítios representam regiões da MAO. Além disso, alguns estudos têm demonstrado que derivados imidazolínicos são capazes de inibir a atividade da MAO. Este efeito tem sido atribuído a sítios I2 de alta afinidade na MAO-B (I2B) e a um sítio similar de baixa afinidade na MAO-A (I2A). Assim, este estudo teve como objetivo investigar o efeito in vitro de compostos 4,5-diidro-1H-imidazol-2-substituídos sobre a atividade da enzima monoamina oxidase através de métodos espectrofotométricos e fluorimétricos usando quinuramina como substrato. Entre os compostos estudados que inibiram preferencialmente a MAO-A (3c-e, 3j) apenas o composto 3d foi seletivo, apresentando um Ki para a MAO-A de aproximadamente 73 vezes menor do que seu Ki para MAO-B. Entre os compostos obtidos que seletivamente inibiram MAO-B (3g-l, 3K, 3o), apenas a imidazolina 3g mostrou ser potente, com valores de Ki de 5,3 μM. Alguns compostos que exercem ligação potente e seletiva à sítios I2, como o 3l (benazolina), 3n (2-BFI) e 3p (BU224) mostraram boa atividade inibitória especialmente contra MAO-B. Em fígado de ratos, as imidazolinas inibiram com menos seletividade a MAO-A e MAO-B quando comparado com cérebro de ratos. Os compostos 3d e 3g inibiram a MAO de maneira reversível e apresentaram inibição de natureza mista (diminuindo o valor de Vmáx e aumentando o valor de Km) sobre a enzima MAO. Estes resultados confirmam que drogas imidazolinas podem inibir a atividade da MAO e sugerem uma relação entre sítios I2 e a modulação da atividade da enzima.

Abstract:
In 1979, the Chinese Communist Party restored its policy of freedom of religion. The intention of this policy was to bring religious movements which had spread underground under the suppression of the Maoist era back under state control. This was to be accomplished through the "patriotic religious organizations," which would be coopted institution accepting party leadership over religion. In the case of Protestants, however, this policy did not succeed in accomplishing the state's objectives, as unsupervised Protestant activities and unwelcome attempts by foreign Protestant groups to play a role in the evangelization of China persisted over the decade. These and more general factors led to a growing desire on the part of the state to strengthen control over Protestant religious activities, but the state's efforts to strengthen control served to undermine the credibility of the TSPM/CCC with Protestant believers, contributing to the failure of the strategy of cooption to unite Protestants under a single institutional structure. At the same time, Protestant leaders were becoming increasingly frustrated with the state's intrusions into religious life, and were seeking to have a sphere of autonomy for the church recognized by the state. By 1990, Protestant leaders were pressing for the state to encode the separation of church and state in law, while the state was more anxious than ever in the post-Tiananmen domestic and international climate to tighten its control over the church, and over society in general.

Abstract:
Le présent travail de thèse a pour but le développement du procédé de traitement de surface d’anodisation micro-arc (MAO), appliqué au titane et alliage de titane. L’objectif est de déterminer l’influence des paramètres tels que la nature du substrat (éléments d’alliage), la chimie de la solution électrolytique et les paramètres électriques, sur le comportement électrochimique in-situ des couches en formation, ainsi que sur les caractéristiques microstructurales et chimiques des revêtements. Des méthodes de caractérisations notamment morphologiques (imagerie MEB et MET), chimiques (EDS, DRX, diffraction des électrons, EELS) et électrochimiques ex-situ (potentiel libre, courbes de polarisation, SIE) sont utilisées afin d’étudier les revêtements formés. L’étude systématique réalisée par voltampérométrie et chronopotentiométrie a permis de différencier trois stades ou régimes d’anodisation (I/ classique, II/ micro-arc, et III/ d’arcs), caractérisés par une réponse électrochimique particulière de l’interface métal/électrolyte, et qui impacte les propriétés de revêtement finalement obtenues. Des modèles phénoménologiques sont proposés aux différents stades d’anodisation et en lien avec les paramètres du traitement MAO. Le titane commercialement pur de Grade 2 et l’alliage de titane Ti-6Al-4V (ou TA6V) ont été étudiés comparativement ; l’influence des éléments d’alliage (aluminium et vanadium) a été discutée sur la conduite du procédé. L’élaboration d’une solution électrolytique a été étudiée dans le but d’obtenir un revêtement épais, compact et composé majoritairement de titanate d’aluminium. L’incorporation dans les revêtements des éléments provenant de l’électrolyte a été discutée en lien avec la réponse électrochimique ; cette étude conduit à une proposition de mécanisme de croissance de couche où interviennent les éléments du substrat et les éléments d’alliage aux différents stades d’anodisation. L’étude des régimes pulsés unipolaires et bipolaires a permis de discuter l’effet des temps de pause et des pulsations cathodiques sur la réponse électrochimique du matériau et sur les propriétés des revêtements. L’étude du ratio des charges anodiques / cathodiques a montré qu’il s’agissait d’un paramètre essentiel pour garantir la croissance d’un revêtement à la fois épais, homogène et compact
This thesis manuscript relates to the study of process set up of an electrochemical surface treatment, called micro-arc oxidation (MAO), and applied to titanium and its alloys. The aim is to determine the influence of parameters such as nature of the substrate (alloying elements), chemistry of the electrolytic solution and electrical parameters, on the process. In-situ electrochemical behaviour of forming oxide layers is studied, as well as microstructural and chemical characteristics of formed coatings. Many methods mostly to characterize morphology (SEM, TEM imagery), chemistry (EDS, XRD, electron diffraction, EELS) and ex- situ electrochemical behaviour (OCP, polarizing, EIS) are used. Systematic study realised by voltamperometry and chronopotentiometry allowed to differentiate three anodizing stages (I/ conventional, II/ micro-arc, III/ of arcs), characterized by a particular electrochemical response of the metal/electrolyte interface, and which impacts obtained coating properties. Phenomenological models are proposed for each stage of anodizing and linked to MAO process parameters. Grade 2 commercially pure titanium and alloy Ti-6Al-4V (or TA6V) are comparatively studied; the influence of alloying elements (aluminium and vanadium) was discussed in relation with running of the process. Development of an electrolytic solution was carried out to obtain a thick and compact coating, mostly composed of aluminium titanate. Incorporation into the coating of elements from the electrolyte was discussed, and linked to in-situ electrochemical response; this study leaded to a proposed coating growth mechanism which involves elements from the substrate and from the electrolyte. Study of unipolar and bipolar pulsed regimes allowed discussing the effect of pause time and cathodic pulses on electrochemical response of the material and on coating properties. Study of the anodic / cathodic charge ratio showed it was an essential parameter to ensure growth of a thick, homogeneous and compact coating

Two interrelated hypotheses are suggested at the outset: (1) Mao had built up a long-time international sense and perspective since he was very young, (2) Mao had shaped his own world view during the defined period at both concrete and abstract levels. They have gained factual and logical validation. A chronological, periodized, and overall outlook portrays Mao as a lively world observer, an enthusiastic commentator, and a bold predictor. Mao's judgments, inferences, and perceptions, whether accurate or erroneous, were the reflection of the connection between the real (objective) world and Mao's mental (subjective) world, either concordant or discrepant. Mao's subjective world was composed of various elements, mainly, nationalist mentality, revolutionary interest and values, historical and cultural heritages, and personal experiences and characteristics. Mao's perceptions of the world as a whole and his images of the major powers, especially Mao's relations with Soviet Union, provide a concrete framework of his world view at empirical basis. A macro analysis of world view-related three groups of concepts (foreign affairs related concepts, famous revolutionary concepts and philosophical concepts) offers abstract principles of Mao's world view at conceptual level. His belief of "understanding the world and changing the world" shows distinctive philosophical ground. Mao's preference of change, struggle, unevenness, and flexibility had particular significance for featuring his world view. Four intellectual sources contributed to Mao's world view, such as: older Chinese traditions, the newly emerged tradition in modern China, western thought and learning, and Marxism-Leninism. Each of them functioned to influence Mao's world view in one way or another. Mao proved to be an eclectic with the label of sinicized Marxism.

Rationale: The serotonergic system is involved in the modulation of emotion and plays an important role for personality and vulnerability for psychiatric disorders. In the papers included in this thesis, we investigate three biological factors that have been studied in relation to psychiatric symptoms: Platelet monoamine oxidase B (MAO-B) activity, and variations in the MAO-A and the serotonin transporter (5HTT) genes. We also study intensity dependent auditory evoked potentials (IAEP) as an intermediate phenotype for serotonergic capacity. Platelet MAO-B has been shown to be a biological marker for the properties of monoamine systems, with low activity being associated with vulnerability for high scores of sensation seeking, monotony avoidance, and impulsiveness, as well as for susceptibility for alcoholism. Functional polymorphisms in the promoter of the genes encoding MAO-A and the serotonin transporter result in high- or low- activity alleles that have been associated with numerous psychiatric symptoms. One hypothesis for the shaping of personality is that these genotype variants have prenatal effects on the wiring of the brain. Thus, exploring how the development of the brain is affected by different prenatal serotonin levels is relevant in this context. Observations: (i) Platelet MAOB activity was associated with monoamine metabolites in cerebrospinal fluid from cisterna magna in monkeys, as well as with voluntary alcohol intake, alcohol-induced aggression, and alcohol sensitivity. (ii) The long 5HTTLPR allele was associated with increased IAEP. (iii) The functional MAOA and 5HTT polymorphisms were associated with symptoms of ADHD-related traits in a population based sample of Swedish adolescents. Associations of these candidate genes with ADHD scores were strenghtened when the platelet MAOB activity was combined with genotype. (iv) Our pilot data showed that treatment of pregnant mice with 5HTT blocking antidepressives resulted in more serotonergic cellbodies in lateral wings of dorsal raphe in the offspring, when compared to saline treatment. Conclusions: Our studies support the notion that platelet MAOB activity and IAEP are endophenotypes for monoaminergic capacity and related behaviours. The functional candidate polymorphisms in MAOA and 5HTT were linked to behaviour, however, the cause-relationship is unclear and the explanation for the associations need to be further investigated, possibly with focus on prenatal effects of the polymorphisms.

Abstract:
The Chinese mass campaign was originally formed in accordance with the Maoist vision of political participation. The "mass line" dictum enunciated by Mao Zedong required a relationship of reciprocity between the masses and the CCP in terms of evolving ideas, improving and amending them through discussion, and implementing decisions. In this participatory process it was vitally important that the masses freely and voluntarily express their own views. Originally, the mass campaign was a positive, pragmatic, and commitment-filled way of completing constructive tasks, one in which the masses themselves played an important role. The vision of participation prevailing at any given time has been a major factor in shaping the nature of the mass campaign. Central to a consideration of the character of participation is the degree to which the masses are permitted to play their independent, active, and integral role. Stemming from the period following the failure of the Great Leap Forward, this original conception of the mass campaign was increasingly corrupted by developing "bureaucratism" and "commandism" and actual mass involvement declined. With the basic tasks of socialist construction seen as completed, the mass campaign began to be used for "reforming the superstructure." Moralizing, lecturing, and behavioural modification were some of the tasks to which the mass campaign was put, as the achievement of social stability and public cooperation became important state goals. Leadership disunity resulted in the manipulation of the mass campaign for factional ends. Since the commencement of the post-Mao reform process, Chinese society has experienced increases in social disorder and in cynical, self-centred and apathetic public behaviour. In responding to these problems, the state has altered the use of the mass campaign to the minimalist one of a tool for social control. Leadership infighting and disagreements over policy direction and party "line" have become especially prominent since the end of the Cultural Revolution. Mass campaigns have been commonly manipulated by individuals and factions within the leadership in recent years; consequently, their administration and structure has become shoddy and their ideological direction haphazard. Campaigns of the 1980s have continued to display these and other trends. Limited to use as lecturing and moralizing tools, subordinated to economic concerns, manipulated by the leadership, lacking constructive or pragmatic goals, and featuring meaningless content, they are increasingly irrelevant to the masses.

Oxidative stress is associated with development and progression of cardiovascular disease. Angiotensin II produces oxidative stress and endothelial dysfunction, and its actions may be attenuated by the activity of angiotensin converting enzyme type 2 (ACE2) which converts angiotensin II to the vasoprotective peptide angiotensin (1-7). Similarly, increased oxidative stress is associated with aortic valve stenosis in humans and mice. In my thesis studies, I explore mechanisms of modulation and generation of oxidative stress in cerebral arteries and heart valves. First, I tested the hypothesis that ACE2 deficiency increases oxidative stress and vasomotor dysfunction in cerebral arteries, and examined the role of ACE2 in vascular aging. Vasomotor function was assessed in the basilar artery ex vivo of adult and old ACE2 deficient (ACE2-/y) and wild type (WT or ACE2+/y) mice. ACE2 was present, but at relatively low levels in cerebral arteries. Systolic blood pressure was similar in adult and old ACE2-/y and ACE2+/y mice. Maximal dilatation to acetylcholine was impaired in the basilar artery from adult ACE2-/y mice compared to adult ACE2+/y. In old mice, maximal vasodilatation to acetylcholine was impaired in ACE2+/y mice and severely impaired in ACE2-/y mice. The antioxidant tempol improved responses to acetylcholine in adult and old ACE2-/y and ACE2+/y mice. Nitrotyrosine staining in the basilar artery was increased in adult ACE2-/y mice and in old ACE2-/y and ACE2+/y mice relative to adult ACE2+/y, which indicates that oxidative stress was higher in cerebral arteries from ACE2 deficient mice and old mice. Expression of NADPH oxidase subunits Nox2 and p47phox, and of pro-inflammatory molecules Rcan1 and TNF alpha; was increased in cerebral arteries from old ACE2-/y and ACE2+/y mice. Additionally, I tested the hypothesis that serotonin induces oxidative stress in human heart valves, and examined mechanisms by which serotonin may increase reactive oxygen species (ROS). Superoxide (O2.-) was measured in heart valves from explanted human hearts that were not used for transplantation. Superoxide levels (lucigenin-enhanced chemiluminescence) were increased in homogenates of cardiac valves and pulmonary artery after incubation with serotonin. A non-specific inhibitor of flavin-oxidases (DPI), or inhibitors of monoamine oxidase-MAO (tranylcypromine and clorgyline), prevented serotonin-induced increase in O2.-. Dopamine, another MAO substrate which is increased in patients with carcinoid syndrome, also increased superoxide levels in heart valves, and this effect was attenuated by clorgyline. Apocynin did not prevent increases in O2.- during serotonin treatment. Addition of serotonin to recombinant human MAO-A generated superoxide, and this effect was prevented by an MAO inhibitor. In conclusion, I have demonstrated that ACE2 deficiency impairs vasomotor function in cerebral arteries from adult mice and augments endothelial dysfunction during aging. Oxidative stress plays a critical role in cerebrovascular dysfunction induced by ACE2 deficiency and aging. I have also identified a novel mechanism whereby MAO-A can contribute to increased oxidative stress in human heart valves and pulmonary artery exposed to serotonin and dopamine.

The meeting between Richard Nixon and Mao Zedong in 1972 The aim of this dissertation is to provide analysis of the meeting that took place in Beijing in February 1972 between Richard Nixon, as President of the United State of America (USA), and Mao Tse-Tung, the President of the People’s Republic of China (PRC).
 First, the analysis will present a brief historical introduction about the first part of the twenty century. In order to highlight the diplomatic issues which caused the postponement of this revolutionary meeting in 1972, this first section will focus on the conclusion of the Second World War and the beginning of the Cold War. Second, the examination of the political and cultural backgrounds of both USA and PRC will clarify the reasons that led to this event.
 Third, the study will underline and describe the main feature of the meeting considering both the so- called public opinion point of view and the behind-the- scenes activities. Finally, the study will stress the importance of the meeting as a symbolic event, that is the beginning of a new era in the relationship between USA and PRC.

Abstract:
The development of cost-effective and sustainable catalytic methods for the production of enantiomerically pure chiral amines is a key challenge facing the pharmaceutical and fine chemical industries. There is an increasing demand for broadly applicable synthetic methods which deliver the desired amine product in high yield and enantiomeric excess (e.e.). Previously we have described the development of variants of monoamine oxidase from Aspergillus niger (MAO-N) which are able to mediate the complete conversion of racemic amines to the corresponding enantiomerically pure products in a single step. In this thesis we report a panel of MAO-N variants (D5, D9 and D11) developed in our laboratory, which are able to mediate the deracemisation of primary, secondary and tertiary amines with broad structural features. In particular, we have synthesized and subjected to deracemisation a broad range of tetrahydroisoquinolines and tetrahydro-β-carbolines checking enantioselectivity and enantiopreference of our biocatalysts. A relation between lipophilicity of the substituents and enantiopreference of the enzyme has been identified. We have also engineered a new MAO-N variant (D11) with a greatly increased substrate scope and enhanced tolerance for bulky substrates. Application of this engineered biocatalyst is highlighted by the asymmetric synthesis of the generic drugs Solifenacin and Levocetirizine as well as a number of important classes of biologically active alkaloid natural products. We also report a novel MAO-N mediated asymmetric oxidative Pictet-Spengler approach to the synthesis of (R)-harmicine.Another challenge facing the chemist in the new millennium is the development of cleaner and more efficient chemical processes. To this aim the combination of two or more catalytic systems to complete a series of cascade reactions is considered particularly appealing. We have reported a concurrent redox cascade for the deracemisation of pyrrolidines and tetrahydroisoquinolines using our monoamine oxidase-N with a biotinylated Ir-complex within streptavidin (SAV). To achieve the final goal it is necessary to shield the metal inside a host to avoid the mutual inactivation of the two catalysts. We have also described the combination of MAO-N with berberine bridge enzyme (BBE) for the synthesis of berbines (tetrahydroprotoberberines), which represent a sub-class of tetrahydro-isoquinoline alkaloids found in various plants. This bi-enzymatic cascade allows the synthesis of these structures achieving a theoretical 100% yield instead of the 50% given by the kinetic resolution using BBE itself.

The present dissertation attempts to examine fictional works written by and about contemporary Chinese youth in the post-Mao era (1976-1984). Among the several thousand stories that flooded the Chinese literary scene since the death of Mao Zedong and the fall of the "Gang of Four", Zhiqing Fiction, or fiction about urban educated youth, has played an important role in terms of its large quantity, its daring social criticism, and its comprehensive revelation of the physical and spiritual journey of contemporary Chinese urban youth since 1966. Zhiqing Fiction enables us to trace that journey from the Red Guard Movement to the rustication program and life from after returning to their city homes. In this thesis, a thematic study of Zhiqing Fiction, the existence of a cyclical pattern is demonstrated. An examination of this thematically circular pattern shows how under the influence of socio-political, literary and personal factors, an important part of Chinese literature was transformed from the affirmation of official (revolutionary) values to the negation of them and finally back to a reaffirmation of similar values in a new historical context. It further shows that, despite the relaxation of literary policy, Chinese literature has not yet escaped political influence. A political exploration of this fact is also seen to be the Chinese writers' inherent desire to influence society through literature. In this thesis portraits of post-Mao youth are discussed under five thematic categories: the Red Guard Movement, rusticated life, rediscovery of love, re-assertion of the self, and the search for new outlets. Besides supporting the cyclical model, these portraits are seen to also provide a window for understanding the loves and hates, aspirations and frustrations, ideals and disillusionments, and above all, the spirit, of contemporary Chinese youth.

Studies of platelet MAO-B activity have revealed a link between low platelet activity and psychiatric syndromes characterized by an inability to control impulses and to anticipate future consequences of behavior (Oreland, 1980; Gottfries, von Knorring, & Oreland, 1980). These characteristics are fundamental to the construct of psychopathy, and we might therefore expect that psychopathy is associated with low MAO activity. Indeed, some investigators have suggested that low platelet MAO-B activity is a potential marker for vulnerability to psychopathy (Schalling, Asberg, Edman, & Oreland, 1987). However, no study to date has directly examined the association between platelet MAO activity and psychometrically-sound indices of psychopathy. The present study measured platelet MAO-B activity in a sample of 54 male offenders, assessed with the Psychopathy Checklist-Revised (PCL-R; Hare, 1991). PCL-R scores were not significantly related to level of platelet MAO activity. The results are discussed in terms of methodological issues involved in conducting biochemical research.

Dans cette thèse qui comprend cinq chapitres (situation historique des poèmes, genres et langue, macrostructures et microstructures, images, intertextualité) nous nous proposons de faire une étude sémiotique de la textualité et de l'intertextualité dans la poésie de Mao. Dans le chapitre "situation historique des poèmes", nous donnons une discussion globale sur la poésie de Mao en soulignant les événements historiques et personnels dont il s'inspire pour écrire ses poèmes. Dans le chapitre "genres et langue", nous présentons les genres poétiques que Mao utilise dans sa poésie, la spécificité des idéogrammes, et les contraintes formelles de la poésie chinoise. Nous y soulignons l'importance du parallélisme comme principe d'organisation dans la poésie chinoise et nous vérifions au passage les théories de Jakobson sur la fonction de l'équivalence et du parallélisme dans le discours poétique. Dans le chapitre "macrostructures et microstructures", notre projet est d'étudier, au niveau des macrostructures, les grandes dominantes structurales qui organisent la poésie de Mao, et au niveau des microstructures, les procédés stylistiques qui se manifestent dans sa poésie. Dans le chapitre "images", nous faisons une étude des images chez Mao. Nous y montrons que la figure, grâce au principe constituant du parallélisme, n'est pas seulement une chose décorative et statique, mais un procédé d'engendrement dynamique du texte. Enfin, dans le dernier chapitre intitule "intertextualité", nous discutons de la pratique de la citation et des rapports poésie – politique dans la tradition poétique chinoise, et nous faisons aussi une étude des opérations d'intertextualité. Dans nos analyses des structures, des images, et du travail d'intertextualité, nous tachons de mettre en relief les systèmes dominants, le fonctionnement des images et des intertextes, afin de mettre en lumière l'idéologie de Mao. Nous donnons en appendice trois lettres de Mao sur la poésie, et une traduction littéraire des quarante-sept poèmes du révolutionnaire
In this thesis which comprises five chapters (historical situation of the poems, genres and language, macrostructures and microstructures, imagery, and intertext), we propose to do a semiotic study on the texture and the intertexture of Mao’s poetry. In the chapter "historical situation of the poems", we give a global discussion of Mao’s poetry while pointing out the historical and personal situations which inspire moa to write poetry. In the chapter "genres and language", we present the genres that Mao uses in his poems, the special characteristics of the Chinese characters, and the rules of versification of Chinese poetry. We point out the importance of parallelism as an organisation principle in Chinese poetry and we verify the theories of Jacobson on the function of equivalence and of parallelism in the poetic discourse. In the chapter "macrostructures and microstructures", our aim is to study the major structures and the stylistic touches in Mao’s poetry. In the chapter "imagery", we study the imagery in Mao’s poetry, and we show that, thanks to the concept of parallelism, imagery is not only decorative and static, but also a dynamic element which generates text. Finally, in the last chapter "intertext", we discuss the practice of intertext and the relationship between poetry and politics in the Chinese poetic tradition, and we also study the operations of intertext in Mao’s poetry. In our analyses of structures, imagery, and the "working" of the intertext, we try to elucidate the dominant structural systems, the functioning of imagery and of intertexts, in order to bring out the ideology of Mao. We also provide in the appendix three letters written by Mao the subject of poetry, and a literary translation of the forty-seven poems of Mao

Alzheimer s disease (AD) is biochemically characterized by the occurrence of extracellular deposits of amyloid beta peptide (Aβ) and intracellular deposits of the hyperphosphorylated tau protein, which are causally related to the pathological hallmarks senile plaques and neurofibrillary tangles. Monoamine oxidase B (MAO-B) activity, an enzyme involved in the oxidation of biogenic monoamines, is particularly high around the senile plaques and increased in AD patients in middle to late clinical stages of the disease. Selegiline, a selective and irreversible MAO-B inhibitor, improves learning and memory in AD patients. Notwithstanding, its mechanism of action is still not completely known. The current study aimed to investigate whether selegiline improves the Aβ25-35 induced cognitive deficit in the object recognition task in mice. In addition, we investigated whether selegiline alters MAO-B and MAO-A activities in the hippocampus, perirhinal and remaining cerebral cortices of Aβ25-35-injected mice. Acute (1 and 10 mg/kg, p.o., immediately post-training) and subchronic (10 mg/kg, p.o., seven days after Aβ25-35 injection and immediately post-training) administration of selegiline reversed the cognitive impairment induced by Aβ25-35 (3 nmol, i.c.v.). Acute administration of selegiline (1 mg/kg, p.o.) in combination with Aβ25-35 (3 nmol) decreased MAO-B activity in the perirhinal and remaining cerebral cortices. Acute administration of selegiline (10 mg/kg, p.o.) decreased MAO-B activity in hippocampus, perirhinal and remaining cerebral cortices, regardless of Aβ25-35 or Aβ35-25 treatment. MAO-A activity was not altered by selegiline or Aβ25-35. In summary, the current findings further support a role for MAO-B in the cognitive deficits observed in AD.
A doença de Alzheimer (DA) é bioquimicamente caracterizada por depósitos extracelulares de peptídeo beta amiloide (Aβ) e de proteína tau hiperfosforilada, que são causalmente relacionadas com as características patológicas, placas neuríticas e emaranhados neurofibrilares. A atividade da monoamina oxidase B (MAO-B), uma enzima envolvida na oxidação de monoaminas biogênicas, é particularmente elevada ao redor das placas senis e aumenta nos pacientes com DA em estágios clínicos de moderado a grave. A selegilina, um inibidor seletivo e irreversível da MAO-B, é relatada por melhorar a memória e o aprendizado em pacientes com DA. Porém, seu mecanismo de ação ainda não é completamente conhecido. O presente estudo teve como objetivo investigar se a selegilina melhora o déficit cognitivo induzido por Aβ25-35. na tarefa de reconhecimento de objetos em camundongos. Além disso, investigou-se a atividade da MAO-A e da MAO-B no hipocampo, no córtex cerebral e no córtex perirrinal de camundongos injetados com Aβ25-35 e com selegilina. Administração aguda (1 e 10 mg/kg, p.o., imediatamente pós-treino) e subcrônica (10 mg/kg, p.o., por sete dias depois da injeção do Aβ25-35 e imediatamente pós-treino) de selegilina preveniu o prejuízo da memória induzido pelo Aβ25-35 (3 nmol, icv). A administração aguda de selegilina (1 mg/kg, p.o.) em combinação com Aβ25-35 (3 nmol) diminuiu a atividade da MAO-B no córtex perirrinal e córtex cerebral. A administração aguda de selegilina (10 mg/kg, p.o.) diminuiu a atividade da MAO-B no hipocampo, no córtex cerebral e no córtex perirrinal independentemente da presença de Aβ25-35. A atividade da MAO-A não foi alterada pelo tratamento com selegilina ou Aβ25-35 em nenhuma das estruturas estudadas. Abstract:
The mitochondrial membrane bound flavoenzymes monoamine oxidase A and B (MAO-A and MAO-B) catalyze the a-carbon oxidation of a variety of amines including neurotransmitters such as dopamine and serotonin. Although the primary structures of these enzymes have been established from the corresponding gene sequences, relatively little is known regarding the structural features of the active sites which lead to the selectivities observed with various substrates and inhibitors. In spite of many efforts, these enzymes have not been crystallized. In the absence of X-ray structures, the design, synthesis, and evaluation of biological activity remain the only way to assess a view of the active sites, through SAR and QSAR studies. The excellent MAO-A and/or B substrate and inhibitor properties of various 1,4-disubstituted-1,2,3,6-tetrahydropyridine derivatives offer an interesting opportunity to probe the active sites of MAO-A and MAO-B. In an effort to explore the spatial features of the active sites, we have synthesized series of substituted tetrahydropyridines, evaluated their biological activity with purified MAO-A and MAO-B, and carried out a topological analysis of the MAO active sites using molecular modeling. In addition, the results described in this thesis provide evidence that the MAO-A and MAO-B active sites differ in shape, regions of activity, and areas that tolerate polar interactions. The role of MAO in neurodegenerative processes such as Parkinson's Disease has been recognized for some time. The structurally unique parkinsonian inducing substrate 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is bioactivated to neurotoxic metabolites. The mechanism of neurotoxicity has been studied extensively and it is known that MAO-B catalyzes the conversion of MPTP to the 2,3-dihydro-1-methyl-4-phenylpyridinium species (MPDP+) which undergoes further oxidation to the neurotoxic metabolite 1-methyl-4-phenyl pyridnium (MPP+). However, the role of MAO-A in mediating a neurotoxic response, has not been fully defined due to the lack of selective MAO-A substrates. In this thesis, we have investigated the neurotoxic potential of several tetrahydropyridines in C57Bl/6 mice and the ability of selective inhibitors to protect against the expression of MAO mediated neurotoxicity.

This thesis studies the prevailing suzhi discourse in post-Mao China. It addresses two main research questions: (1) What are the processes through which suzhi has developed as a state discourse? (2) What is the underlying cultural, philosophical and psychological foundation that propels suzhi discourse? Through an examination of the process of the developments of suzhi and suzhi jiaoyu, this thesis seeks to understand patterns and rationalities underpinning suzhi discourse in politics and education, and how political discourse and the ideology behind that discourse infiltrates the psyche of citizens and those who govern citizens in post-Mao China. This thesis argues that as a prevalent political and cultural idea, suzhi provides a window into the psychological logic and rationale of current day political thinking in China about how a government enlists a civilising program in their quest for national development, modernity and prosperity. This thesis introduces the concept of transformational citizenship as a means of understanding how a political rationale prioritises transforming citizens through education and civilising projects. Transformational citizenship is defined as a type of citizenship that presumes an inadequate citizen and emphasises the necessity and possibility of transforming  through practice and learning  a citizen’s embodied qualities and thought patterns into the essential qualities and thought patterns required of a responsible and eligible citizen.

Abstract:

The subjects were 130 male prisoners in Swedish jails were examined by SCID and the diagnoses given in terms of DSM-IV. The most common mental disorder was drug abuse. On Axis II several personality disorders were diagnosed. Personality assessments were made by KSP. High scores were mainly found for e.g. impulsiveness, sensation seeking, aggression and low scores in socialisation.

MAO assays were performed in 99 male criminal offenders and in 60 non-criminal volunteers. Offenders had lower MAO activity than controls also with the confounding factor smoking under control. It is proposed that platelet MAO is linked to personality traits, which can predispose for criminality.

For testing the existence of combinations of vulnerability factors, a configuration frequency analysis (CFA) was used. The criteria which formed the basis for the subgrouping were; MAO activity below or above –0.5 SD of the mean (L and H), the presence or absence of an Axis I disorder (= drug abuse) (Y/N), the presence or absence of an Axis II disorder (Y/N), or the presence or absence of an Axis I and II disorder (Y/N). In this way eight subgroups were formed. Two significant "types" were found among the criminals: One was characterised by low platelet MAO activity, Cluster B personality diagnosis as well as Drug Abuse Disorder diagnosis (LYY); and the other by a pattern of normal platelet MAO activity, no Cluster B personality disorder, and no Drug Disorder diagnosis (HNN). Also two "antitypes", occurring less frequent than expected, were identified; LYN and LNY. Thus, the aggregation of certain risk factors in the same individual has been shown to contribute to the development of criminal behaviour.

The subgroups HNN, LYN, LNY and LYY were then analysed for a variety of criminological factors. There was a difference in mean age between the subgroups, the HNN being lowest. Economical crimes were more common at an early criminal debut and crimes involving violence at an adult debut. The HNN subgroup had a lower number of crimes and times spent in jail than the other subgroups. More than 50% of the clients in all groups had previously been sentenced to Reformatory.

Abstract:
It is the prevailing critical assessment of Mao Dun's early creative writing that he displays a singular insight in his portrayal of women. This thesis seeks not only to challenge this assessment by a predominantly male body of criticism but also the assumptions on which it is based, namely that an intellectual sympathy for the women’s cause necessarily implies a transcendence of the patriarchal attitudes with which society is imbued. The major short stories and novellas written between 1927 and 1932 are analysed systematically to identify Mao Dun's underlying attitudes towards women. His portrayal of women is assessed from the following perspectives:~ his autobiographical accounts of his encounters with women in his political and personal life and his deliberate association of his female comrades with his creative inspiration;- traditional Chinese perceptions of women and gender roles as these are manifested in the classical tradition;-- Mao Dun's numerous articles and essays on the women's question written during the nineteen twenties and his work in the women's section of the Party in Shanghai;- Mao Dun's attempt to reconcile his conflicting sympathies for feminism and socialism. This thesis relies for its methodology on Western feminist criticism. While the approach is maintained, in its application to the context of early twentieth century China, its eurocentrism in terms of cultural assumptions and perceptions of gender has been replaced by a definition of Chinese values. Since a fundamental prerequisite, of feminist criticism is the assessment of the writer in his/her own cultural context, a historical survey of the portrayal of women in traditional literature is provided to serve as a standard against which to measure Mao Dun’s portrayal.
