ABSTRACT Burkholderia mallei is a highly infectious gram-negative pathogen and is the causative agent of human and animal glanders. By generating polar mutations (disruption of bsaQ and bsaZ) in the B. mallei ATCC 23344 animal pathogen-like type III secretion system (TTS), we demonstrate that this bacterial protein delivery system is required for intracellular growth of B. mallei in J774.2 cells, formation of macrophage membrane protrusions, actin polymerization, and phagosomal escape. These findings suggest that TTS plays a role in the intracellular trafficking of B. mallei and may facilitate cell-to-cell spread via actin-based motility.

Abstract:
Burkholderia mallei causes the highly contagious and debilitating zoonosis glanders, which infects via inhalation or percutaneous inoculation and often culminates in life-threatening pneumonia and sepsis. In humans, glanders is difficult to diagnose and requires prolonged antibiotic therapy with low success rates. No vaccine exists to protect against B. mallei, and there is concern regarding its use as a bioweapon. The authors previously identified the protein BpaB as a potential target for devising therapies due to its role in adherence to host cells and the formation of biofilms in vitro and its contribution to pathogenicity in a mouse model of glanders. In the present study, the authors developed an immunostaining approach to probe tissues of experimentally infected animals and demonstrated that BpaB is produced exclusively in vivo by wild-type B. mallei in target organs from mice and marmosets. They detected the expression of BpaB by B. mallei both extracellularly and within macrophages, neutrophils, and epithelial cells in respiratory tissues (7/10 marmoset; 2/2 mouse). The authors also noted the intracellular expression of BpaB by B. mallei in macrophages in the regional lymph nodes of mice (2/2 tissues) and MALT of marmosets (4/5 tissues). It is interesting that B. mallei bacteria infecting distal organs did not express BpaB (2/2 mice; 3/3 marmosets), suggesting that the protein is not necessary for bacterial fitness in these anatomic locations. These findings underscore the value of BpaB as a target for developing medical countermeasures and provide insight into its role in pathogenesis.

ABSTRACT Burkholderia pseudomallei and its host-adapted deletion clone Burkholderia mallei cause the potentially fatal human diseases melioidosis and glanders, respectively. The antibiotic resistance profile and ability to infect via aerosol of these organisms and the absence of protective vaccines have led to their classification as major biothreats and select agents. Although documented infections by these bacteria date back over 100 years, relatively little is known about their virulence and pathogenicity mechanisms. We used in silico genomic subtraction to generate their virulome, a set of 650 putative virulence-related genes shared by B. pseudomallei and B. mallei but not present in five closely related nonpathogenic Burkholderia species. Although most of these genes are clustered in putative operons, the number of targets for mutant construction and verification of reduced virulence in animal models is formidable. Therefore, Galleria mellonella (wax moth) larvae were evaluated as a surrogate host; we found that B. pseudomallei and B. mallei, but not other phylogenetically related bacteria, were highly pathogenic for this insect. More importantly, four previously characterized B. mallei mutants with reduced virulence in hamsters or mice had similarly reduced virulence in G. mellonella larvae. Site-specific inactivation of selected genes in the computationally derived virulome identified three new potential virulence genes, each of which was required for rapid and efficient killing of larvae. Thus, this approach may provide a means to quickly identify high-probability virulence genes in B. pseudomallei, B. mallei, and other pathogens.

Whereas the ME pathway enablesL. caseito grow onl-malate, MLE does not support growth. Themlegene cluster consists of three genes encoding MLE (mleS), the putativel-malate transporter MleT, and the putative regulator MleR. Themaegene cluster consists of four genes encoding ME (maeE), the putative transporter MaeP, and the two-component system MaeKR. Since both pathways compete for the same substrate, we sought to determine whether they are coordinately regulated and their role inl-malate utilization as a carbon source. Transcriptional analyses revealed that themleandmaegenes are independently regulated and showed that MleR acts as an activator and requires internalization ofl-malate to induce the expression ofmlegenes. Notwithstanding, bothl-malate transporters were required for maximall-malate uptake, although only anmleTmutation caused a growth defect onl-malate, indicating its crucial role inl-malate metabolism. However, inactivation of MLE resulted in higher growth rates and higher final optical densities onl-malate. The limited growth onl-malate of the wild-type strain was correlated to a rapid degradation of the availablel-malate tol-lactate, which cannot be further metabolized. Taken together, our results indicate thatL. caseil-malate metabolism is not optimized for utilization ofl-malate as a carbon source but for deacidification of the medium by conversion ofl-malate intol-lactate via MLE.

Aims Cementless acetabular components rely on press-fit fixation for initial stability. In certain cases, initial stability is more difficult to obtain (such as during revision). No current study evaluates how a surgeon’s impaction technique (mallet mass, mallet velocity, and number of strikes) may affect component fixation. This study seeks to answer the following research questions: 1) how does impaction technique affect a) bone strain generation and deterioration (and hence implant stability) and b) seating in different density bones?; and 2) can an impaction technique be recommended to minimize risk of implant loosening while ensuring seating of the acetabular component? Methods A custom drop tower was used to simulate surgical strikes seating acetabular components into synthetic bone. Strike velocity and drop mass were varied. Synthetic bone strain was measured using strain gauges and stability was assessed via push-out tests. Polar gap was measured using optical trackers. Results A phenomenon of strain deterioration was identified if an excessive number of strikes was used to seat a component. This effect was most pronounced in low-density bone at high strike velocities. Polar gap was reduced with increasing strike mass and velocity. Conclusion A high mallet mass with low strike velocity resulted in satisfactory implant stability and polar gap, while minimizing the risk of losing stability due to over-striking. Extreme caution not to over-strike must be exercised when using high velocity strikes in low-density bone for any mallet mass. Cite this article: Bone Joint Res 2020;9(7):386–393.

ABSTRACT Burkholderia mallei is a facultative intracellular pathogen that causes severe disease in animals and humans. Recent studies have shown that the cluster 1 type VI secretion system (T6SS-1) expressed by this organism is essential for survival in a hamster model of glanders. To better understand the role of T6SS-1 in the pathogenesis of disease, studies were initiated to examine the interactions of B. mallei tssE mutants with RAW 264.7 murine macrophages. Results obtained by utilizing modified gentamicin protection assays indicated that although the tssE mutants were able to survive within RAW 264.7 cells, significant growth defects were observed in comparison to controls. In addition, analysis of infected monolayers by differential interference contrast and fluorescence microscopy demonstrated that the tssE mutants lacked the ability to induce multinucleated giant cell formation. Via the use of fluorescence microscopy, tssE mutants were shown to undergo escape from lysosome-associated membrane protein 1-positive vacuoles. Curiously, however, following entry into the cytosol, the mutants exhibited actin polymerization defects resulting in inefficient intra- and intercellular spread characteristics. Importantly, all mutant phenotypes observed in this study could be restored by complementation. Based upon these findings, it appears that T6SS-1 plays a critical role in growth and actin-based motility following uptake of B. mallei by RAW 264.7 cells.

Cela peut signifier au minimum qu’il doit avoir le droit de refuser à l’avance les traitements qui le prolongeraient artificiellement quand tout espoir a disparu. Et cela peut vouloir dire à la limite qu’il doit pouvoir demander qu’on l’aide à quitter sans souffrances ce monde si son existence lui devient intolérable. Les adversaires de l’euthanasie font valoir qu’elle est contraire à la vocation traditionnelle de la médecine et aux principes de base de l’éthique car il existe des cas où l’aide au suicide et l’euthanasie sont des actes de courage et de charité vu que le respect mécanique de la vie peut être un signe d’indifférence. L’avenir verra les sociétés reconnaître de plus en plus le droit de l’individu à échapper à la souffrance et à la dégradation quand il est au bout de sa vie. Une telle démarche n’aura cependant de sens que si, en même temps qu’elle aide à mourir ceux qui sont au bout de leur chemin, tente d’aider à vivre ceux qui n’y arrivent que difficilement.

La vie privée du majeur souffrant d'une maladie mentale ou d'une déficience intellectuelle présente des spécificités par rapport à celle de la personne considérée comme saine d'esprit. Cette différence provient d'abord de la nécessité, dans certains cas, pour l'individu dont les facultés mentales sont altérées d'être hospitalisé dans un hôpital psychiatrique ou de vivre dans une institution sociale ou médico-sociale. Sa liberté d'aller et venir et celle d'aménager librement sa vie quotidienne se trouvent, de ce fait, entravées partiellement voire totalement. Des tiers peuvent, en outre, être amenés à s'immiscer dans certains domaines de la vie privée des personnes mentalement troublées tels que la vie sentimentale, la liberté de procréer et la sante. Cet état de fait pose de délicats problèmes. En ce qui concerne les malades ou déficients mentaux juridiquement capables, les tiers, sauf cas particuliers, n'ont aucun pouvoir et leur intervention peut être qualifiée d'abusive. Lorsque ceux-ci se trouvent soumis à un régime d'incapacité, la loi s'avère lacunaire. Les juridictions, confrontées aux nombreux problèmes qui se posent en pratique, ont cependant reconnu aux organes légaux une mission de protection de la personne de l'incapable majeur. Il s'agit toutefois de savoir s'il est possible de "gérer" la vie privée de l'incapable de la même manière qu'un patrimoine, à l'aide des mêmes techniques et avec la même froideur
The private life of an adult suffering from a mental illness or a mental retardation has a number of distinctive features vis-a-vis the private life of a person who can be considered in full mental health. The difference stems firstly from the need, in certain cases, for an individual whose mental faculties are impaired, to be placed in a psychiatric hospital or to live in a social or medico-social institution. His freedom to come and go and to freely plan his every day life is thus partially or even totally restricted. What is more, outsiders may be led to interfere in some aspects of the private life of the mentally impaired such as emotional life, reproductive freedom and health. This state of facts poses some delicate problems. With regard to the mentally ill or handicapped that retain legal capacity, outsiders, apart form certain specific cases, have no power and their interference may be considered improper. When they are classified in one of the categories of legally incapable persons, the law turns out to be deficient. However, the courts who are confronted with numerous practical problems have recognised that the legal representatives have a role in the protection of the incapable adult. However, we must ask ourselves if it is possible to "manage" the private life of an incapable person in the same way in which one manages a person's pecuniary affairs, using the same techniques and with the same coldness

Projektet handlar om att utveckla mallar för olika lösningar av ljusberäkningar med valda armaturer för ett klassrum i Fässbergsskolan i Mölndal. Projektet utgår från olika befintliga armaturer som består av infällda samt pendlade armaturer. Detta har genomförts med hjälp av lämpliga arbetsverktyg och beräkningar och är utfört på ett modernt och branschkunnigt sätt. Dataprogrammet DIALux har använts för att placera ut belysningsarmaturer och göra samtliga ljusberäkningar för klassrummet. För att utföra en kontroll av ljusberäkningar enligt Svensk standard har man fått justera beräkningsresultaten från DIALux för att minska på produktåtgången och samtidigt uppnå de ställda kraven. Projektet har även utgått från ljus & rum riktlinjer och referenser som är ett dokument med ställda krav och allmänna riktlinjer för hur en belysningsprojektering skall utföras. Mallarna har till grunden bestått av ljusberäkningsmallar över klassrummet framtaget ur dataprogrammet DIALux. Varje utvecklad mall består av en specifik ljusberäkningsmall och CAD-ritning med motsvarande armaturspecifikation, materialkostnader och lönsamhetskalkyl. Resultatet av projektet över de olika lösningarna har innefattat olika armaturer som bland annat bestått av infällda och pendlade armaturer. Resultatet av arbetet visade att infälld i LED-belysning var det bästa alternativet vad gäller energiförbrukning, kostnad och lönsamhet. Totala kostnaden för denna lösning uppkom till 35 000 kr med en återbetalningstid på 3.4 år.

Ce mémoire porte sur l’évolution des conditions de travail des infirmières selon le concept de la qualité de vie. Sur un intervalle d’un demi-siècle, nous avons étudié le discours des élites infirmières à travers deux revues professionnelles francophones, La Garde-Malade Canadienne-Française de 1928 à 1938 et L’infirmière du Québec de 1993 à 2003. À partir des articles et rubriques publiés, le discours des élites témoigne du peu d’importance qu’elles accordent à la qualité de vie des infirmières tout en montrant leur volonté de faire valoir les acquis de la profession notamment la reconnaissance professionnelle des infirmières. Par ailleurs, la récurrence de certains thèmes de publication, l’absence d’articles sur la qualité de vie et la persistance de l’image vocationnelle de l’infirmière indique une lenteur dans l’évolution des mentalités quant à la perception sociale des femmes au tournant du XXIe siècle.

El uso de portainjertos en la vid se ha difundido por su resistencia a filoxera y nemátodos, pero también por su tolerancia a condiciones adversas del suelo. Por otro lado, los portainjertos modifican las relaciones fuente-destino, influyendo en el comportamiento vegetativo y reproductivo de las plantas y en la composición de la uva, lo cuál puede ser utilizado como una herramienta de manejo agronómico. A fin de evaluar si existe un comportamiento diferencial de los portainjertos en cuanto a expresión vegetativa, vigor, rendimiento y composición de la uva, y explicar dichas diferencias en términos de exploración radical, relaciones hídricas, asimilación de carbono, eficiencia en el uso del agua y partición de asimilados se realizó un ensa-yo a campo de cv. Malbec sobre seis portainjertos (3309 C, 1103 P, 140 Ru, SO4, Harmony y Cereza) y a pie franco. Los portainjertos 140 Ru, 1103 P y SO4 tuvieron una mayor tendencia a la producción de uva (mayor Índice de Ravaz), y Franco, Cereza y 3309 C a vegetar, mostrando Harmony una situación intermedia. Las ba-yas sobre el pie Cereza tuvieron un mayor peso (1,96 g) que sobre Harmony (1,75 g). No se encontraron diferencias en los polifenoles de las bayas entre portainjertos. La fotosíntesis de la planta entera (Amax) de Franco, 1103 P y SO4 fue mayor que la de Harmony. La conductancia hidráulica foliar específica (kL) de Harmony fue me-nor que la de Cereza, y su conductancia hidráulica (kH) fue menor que la de Franco, Cereza y SO4. El número de raíces totales de 140 Ru fue mayor que el de 1103 P, SO4 y Harmony. El portainjerto 140 Ru se destacó por privilegiar el desarrollo radi-cal y reproductivo sobre el vegetativo, y por su mayor eficiencia en el uso del agua (EUA). Las diferencias entre portainjertos pueden ser explicadas en parte por dife-rencias en la kL que a su vez incide en el estado hídrico de las plantas (ΨL). De ma-nera que cuando la kL es más baja, el ΨL es menor (i.e., Harmony), y cuando la kL es más alta, el ΨL es mayor (i.e., Franco y Cereza). Mayores ΨL se asocian con mayores superficies foliares.
The use of rootstocks in grapevine has been diffused not only for its resis-tance against phylloxera and nematodes, but also for its tolerance to overcome site difficulties. On the other hand, rootstocks modify the relation between source and sink, having influence on the vegetative and reproductive behaviour of the plants and the grapes quality. Therefore they can be used as an agronomic tool. A trial was performed on field growth plants of cv. Malbec on six rootstocks (3309 C, 1103 P, 140 Ru, S04, Harmony and Cereza) and on own roots (Franco) in order to evaluate if there is a differential behaviour of the rootstocks with regard to vegetative expres-sion, vigour, yield and grape quality and to explain such differences in terms of roots exploration, water relations, CO2 assimilation, water use efficiency and carbohy-drates partitions. The rootstocks 140 Ru, 1103 P and SO4 had a higher tendency to the grape production (higher index of Ravaz), meanwhile Franco, Cereza and 3309 C seemed to vegetate more, and Harmony showed a middle condition. The berries of Cereza had a higher weight (1.96 g) than the one of Harmony (1.75 g). No differ-ences in the poliphenolics of the berries among the rootstocks have been found. The whole plant photosynthesis (Amax) of Franco, 1103 P and SO4 was higher than the Amax of Harmony. Leaf specific hydraulic conductance (kL) of Harmony was lower than the kL of Cereza and its hydraulic conductance (kH) was lower than the kH of Franco, Cereza and SO4. Total roots number of 140 Ru was higher than those of 1103 P, SO4 and Harmony. The rootstock 140 Ru showed off for its favorable car-bohydrates partitions and its higher water use efficiency (EUA). The differences be-tween rootstocks could be explained partly to differences in the kL that affected the water status of the plants (ΨL). So that when the kL is lower, the ΨL is lower (e.g., Harmony) and when the kL is higher, the ΨL is greater (e.g., Franco and Cereza). Higher ΨL are associated with greater leaf areas.

The electrophoretic phenotypes with two codominant alleles were observed at APH-3loci. In MDH and XDH only one zone of activity was observed.

However, minimal research has addressed the impact of individual differences on perceptions of robot trustworthiness. These conclusions can provide guidance to optimize adaptive robotic systems in education, healthcare, and industry settings. This study examined the relationship between personality and human robot interaction in two contexts: (1) error-free and (2) errors. Assessment of individual differences were achieved via the Interpersonal Reactivity Index (IRI) (Davis, 1980) and robot trust assessed using the Multidimensional Measure of Trust (MDMT) (Ullman & Malle, 2018). This project provided a novel contribution in the field of human-robot interaction, highlighting the influence of technological failure on trust impressions of a social robot. Additionally, we sought to understand the degree to which empathy levels mediate these changes in trust.

Background. Research consistently finds that male cannabis users report greater problematic cannabis use (i.e., negative outcomes related to one’s cannabis use), compared to female cannabis users. Further, emotion dysregulation is a transdiagnostic risk factor for substance use and addiction and is associated with greater problematic cannabis use. Therefore, the current study examined whether sex moderates associations between emotion dysregulation (overall and dimensions of emotion dysregulation) and problematic cannabis use. Methods. 741 adults reporting past-month cannabis use (31.44% female) completed an online anonymous survey, including questions on demographics, substance use, problematic cannabis use (via the Marijuana Problem Scale) and emotion dysregulation (via the Difficulties in Emotion Regulation Scale). Independent samples t-tests and hierarchical multiple linear regressions were performed. Results. Male cannabis users reported significantly higher scores on overall emotion dysregulation and five of six dimensions (nonacceptance, impulse, awareness, strategies, and clarity) compared to female cannabis users. Further, sex moderated associations between emotion dysregulation (overall, nonacceptance, goals, impulse, awareness, and strategies) and problematic cannabis use. Overall, nonacceptance, goals, impulse, and strategies were positively associated with problematic cannabis use, and these relationships were stronger in male cannabis users. Lack of emotional awareness was negatively associated with problematic cannabis use in male cannabis users and no relationship was found for female cannabis users. Conclusions. Overall, these results denote the importance of examining individual differences in emotion dysregulation as they relate to problematic cannabis use. Interventions and treatments may need to be tailored for male cannabis users with a focus on specific emotion dysregulation dimensions.

In the process of development and ripening of berries, depending on fluctuations in climatic fac-tors, an aroma characteristic of the genotype of grapevine is formed, and as a result of processing the bunches, a bouquet of young wine is formed. Grapevine berries contain, for the most part, the same aro-matic chemical compounds, however, the specific aroma is due not only to their different mass concentra-tion, but also to their ratio in the aromatic complex of each genotype. The specific shade of aromas of a particular genotype and the accent of aromas of a particular genotype depends to a greater extent on the transmission of hereditary traits from parental pairs of crossing, the degree of ripening of berries, the phy-tosanitary level of plantings and on the influence of factors of the growing environment. BackgroundBystanders often hesitate when rendering first aid to females, particularly it requires disrobing the individual (Leary et al, 2018). In addition to the delayed application of first aid, the lifesaver’s actual task performance may also be less effective than when treating injured males. This can occur, for example, when the lifesaver does not fully expose the wound (Bell et al., 2020). The Army has invested heavily in the acquisition of realistic patient manikins for training combat medicine skills. However, given logistical constraints, it will be difficult to acquire an equal number of female patient manikins. Therefore, the purpose of this study was to develop and test a low-cost manikin “Gender Retrofit Kit” (GRK). The GRK included a breast “vest” that is affixed to the torso, a realistic vagina that is affixed to the groin, a wig, facial makeup, and instructions for “feminizing” the manikin’s appearance. MethodWe recruited a convenience sample of 36 Combat Lifesavers and Combat Medics who were completing their recurrent annual training. At the end of their scheduled training, the participants were invited to practice three medical procedures (treatment of penetrating trauma to the leg via tourniquet, treatment of gunshot wound via application of a chest seal, and treatment of tension pneumothorax via needle chest decompression). Of the three medical procedures, only the last two required disrobing the patient. Therefore, we hypothesized that if performance issues were to occur, they would be localized here. The participants practiced the three procedures using two different manikin types: a standard male manikin and the GRK-outfitted manikin. The order of manikin presentation was counterbalanced. Measures of task quality, task completion times, and usability questionnaires were collected. Results and ConclusionsThe sample was primarily male (78%), and included nearly equal numbers of Combat Lifesavers (42%) and Combat Medics (44%). A post-simulation questionnaire suggested no significant mean differences between the standard vs. GRK manikins with regard to the simulators’ perceived realism, anatomical correctness, or ability to provide meaningful skills practice. However, statistically higher mean scores were reported for questionnaire items that focused on the female manikin’s realistic breast tissue, realistic skin texture, and feminine facial appearance. Linear mixed models were used to separately test the effects of participant gender (or job title), manikin gender, and their interaction on both task performance speed and quality. There were no statistically significant differences in task completion order or speed of task completion. All participants performed the three tasks in accordance with the Army’s MARCH-E algorithm, and all had similar completion times. With regard to the quality of task performance, the analyses revealed only one statistically significant main effect of manikin gender: the GRK manikin had a lower mean task performance score for the treatment of gunshot wounds, which required disrobing the patient to apply the chest seal. Based on the results of this exploratory study, we are prioritizing potential improvements to the GRK, and are planning a more rigorously-controlled validation study with the revised prototype. Additional implications and lessons learned will be discussed.

The main goal of the proposed research was to continue the mutual investigations into the molecular basis of CMS and male fertility restoration [MRF], with the ultimate goal of understanding these phenomena in higher plants. The experiments focused on: (1) dissecting apart the complex CMS - specific mitochondrial S-Pcf locus, in order to distinguish its essential parts which cause sterility from other parts and study its molecular evolution. (2) Studying the expression of the various regions of the S-Pcf locus in fertile and sterile lines and comparing the structure and ultrastructure of sterile and fertile tissues. (3) Determine whether alteration in respiration is genetically associated with CMS. Our mutual investigations further substantiated the association between the S-Pcf locus and CMS by the findings that the fertile phenotype of a population of unstable petunia somatic hybrids which contain the S-Pcf locus, is due to the presence of multiple muclear fertility restoration genes in this group of progenies. The information obtained by our studies indicate that homologous recombination played a major role in the molecular evolution of the S-Pcf locus and the CMS trait and in the generation of mitochondrial mutations in general. Our data suggest that the CMS cytoplasm evolved by introduction of a urs-s containing sublimon into the main mitochondrial genome via homologous recombination. We have also found that the first mutation detected so far in S-Pcf is a consequence of a homologous recombination mechanism involving part of the cox2 coding sequence. In all the cases studied by us, at the molecular level, we found that fusion of two different cells caused mitochondrial DNA recombination followed by sorting out of a specific mtDNA population or sequences. This sequence of events suggested as a mechanism for the generation of novel mitochondrial genomes and the creation of new traits. The present research also provides data concerning the expression of the recombined and complex CMS-specific S-Pcf locus as compared with the expression of additional mitochondrial proteins as well as comparative histological and ultrastructural studies of CMS and fertile Petunia. Evidence is provided for differential localization of mitochondrially encoded proteins in situ at the tissue level. The similar localization patterns of Pcf and atpA may indicate that Pcf product could interfere with the functioning of the mitochondrial ATPase in a tissue undergoing meiosis and microsporogenesis. Studies of respiration in CMS and fertile Petunia lines indicate that they differe in the partitioning of electron transport through the cytochrome oxidase and alternative oxidase pathways. The data indicate that the electron flux through the two oxidase pathways differs between mitochondria from fertile and sterile Petunia lines at certain redox states of the ubiquinone pool. In summary, extensive data concerning the CMS-specific S-Pcf locus of Petunia at the DNA and protein levels as well as information concerning different biochemical activity in CMS as compared to male fertile lines have been accumulated during the three years of this project. In addition, the involvement of the homologous recombination mechanism in the evolution of mt encoded traits is emphasized.

Willingness to pay was uncorrelated with actual recent high pollution. Recipients of SMS alerts indicated having received air pollution information via SMS, along with reporting a high-pollution day in the past week and having stayed indoors on the most recent day they perceived pollution to be high. However, alert recipients were not more accurate in identifying which specific days had high pollution than other respondents. Households that received a free N95 mask were more likely to report utilizing a mask with a filter during the past two weeks but not more likely to report using a mask with a filter on the specific days with high particulate matter.

Drawing on an extensive literature review and four case studies from leading examples of post-conflict local peacebuilding in sub-Saharan Africa, this report argues that the “local” in local peacebuilding is best defined as local knowledge of conflict drivers and dynamics and locally defined, contextually specific definitions of peace. This does not necessarily mean working through or empowering “traditional” actors and institutions (a highly contested category, in any case). Nor should it mean a narrow focus on subnational conflict drivers and peace actors to the detriment of assessing how national and international dynamics shape local peace challenges (and vice versa). International donors and peace actors are most successful when they operate with a keen awareness that all potential peacebuilding actors (national and local actors, but also external donors, "experts," and implementers) have their own agendas and that peacebuilding efforts that work at the sub-national level and engage local actors are not automatically endowed with legitimacy and community buy-in just because of their "localness." International actors must also be flexible and open to partnering with a wide range of local actors, including those that don’t meet preconceived international expectations about what an effective local partner looks like (often, old, male, and "traditional").

This technical assistance project supports the goal of the Asia Near East Operations Research and Technical Assistance (ANE OR/TA) project of helping strengthen family planning (FP)/reproductive health (RH) operations research (OR) capabilities in the Philippines. During the extension period of the ANE OR/TA project, four OR studies were implemented in the Philippines dealing with cross-cutting issues, including a study to assess the feasibility of integrating RTI management within FP/MCH programs; an intervention study dealing with improving quality of care via expanded FP counseling; an intervention to address high unmet need in Pangasinan Province; and an intervention study to increase male involvement in RH in Bukidnon Province. These efforts, along with a study of factors affecting use and discontinuation of the IUD, have generated demand for technical assistance, along with corresponding personnel needed to provide such assistance, thereby necessitating this initiative. Activities include providing advice on technical questions, serving as resource persons in training on research design and methodology, assisting in data analysis, disseminating project findings, traveling to project sites, and monitoring and coordinating activities. The report specifies activities undertaken to fulfill the objectives of capacity building and institutionalization of OR in the country covered under this contract.
