Full textMycobacterium marinum (Mm) shares at least 80% amino acid sequence identity with over 3,000 orthologous genes of Mycobacterium tuberculosis (Mtb) and is thus used as a surrogate pathogen for Mtb research. Our laboratory investigates mycobacteriosis using Japanese medaka ( Oryzias latipes) as an aquatic animal model. Mm disease presentation in medaka is similar to Mtb disease presentation in humans, including growth in macrophages, granulomatous lesions, and lifelong chronic disease. We have previously shown that a major route of infection in fish is through an oral route and have thus developed methods to infect medaka with Mm utilizing mosquito larvae as vectors. Recently, our lab was able to show that Mm is able to cross the gut epithelia of medaka in a relatively short-time frame and travel to the underlying submucosa. Therefore, Mm must have the ability to attach to the gut mucosal layer and evade killing by GALT immune cells. Mm is apparently able to exploit macrophages of the mucosal immune system to transport the bacteria to target organs like the head kidney, liver, and spleen for a systemic infection. Utilizing an Mm strain engineered to carry a deletion in the RD-1 region, known to include a number of virulence genes, our lab has shown that mucosal immunity against Mm can be induced in medaka. We have shown that exposure to the mutant RD-1 strain offers some protection against a chronic wild-type oral challenge. Since we know that mutant RD-1 can elicit a mucosal immune response, I tested to see if sensitizing mucosal immunity would also induce systemic immunity by first priming fish with mutant RD-1 and then subsequently challenging them with wild type Mm via an IP route. This thesis demonstrates that mucosal immunity is limited to the gut and thus does not appear to provide broad systemic immunity. Additionally, I tested to see if systemic vaccination would protect against a systemic virulent wild-type challenge by vaccinating and challenging fish via an IP route of infection. Results showed that systemic vaccination does not induce systemic immunity and thus does not protect against an IP injected virulent challenge. Collectively, results from this thesis have shown mutant RD-1 to only be effective as a vaccine against mycobacteriosis if given orally since it was shown to only induce a mucosal immune response and only be protective against an oral virulent wild type challenge.

Full textEl objetivo del presente trabajo fue reunir, analizar y presentar la información más relevante acerca de la infección causada por micobacteriosis en peces, su impacto en la producción y posibles efectos en salud pública. A nivel mundial las enfermedades de peces constituyen una amenaza para el desarrollo y sostenimiento de la acuicultura, ya que si aparecen de forma esporádica, periódica o sostenida limitan la producción. La micobacteriosis (tuberculosis húmeda) en peces o “micobacteriosis atípica” es una enfermedad crónica progresiva; afecta a un amplio rango de especies de agua dulce y salina, particularmente observada en peces de acuario, generando enfermedad crónica sistémica que tiende a formar granulomas externos e internos diseminados; sin embargo, ninguno de estos agentes produce una lesión o signo distintivo, por lo que se requiere hacer un diagnóstico confirmatorio en laboratorios especializados. En las micobacteriosis en peces provocada por micobacterias no tuberculosas (MNT): Mycobacterium marinum, M. chelonae y M. fortuitum, bacterias presentes en el ambiente acuático, la transmisión de la infección se da a través de lesiones en la piel, branquias e intestino, por consumo o contacto con animales o por protozoarios infectados; adicional a ello en peces vivíparos se ha demostrado una transmisión vertical. Sin embargo, el estrés de la manipulación, confinamiento y transporte se consideran como factores predisponentes de la enfermedad. En el hombre, los principales pacientes suelen ser personas mayores o debilitados y que típicamente están relacionados con la manipulación de peces y actividades recreacionales, y se conoce como “granuloma de las piscinas o de los acuarios”, ya que suelen producirse tras pequeños traumatismos cutáneos provocados en piscinas, albercas, estanques o acuarios, además de que de forma característica también existe el antecedente de lesiones con espinas de pescado o al manipular crustáceos. El proceso cutáneo es crónico y con un período de incubación de 2 a 8 semanas. En general suelen ser lesiones únicas, aunque en ocasiones se propagan y ascienden con una distribución lineal esporotricoide. En un tercio de los casos pueden invadir los tejidos profundos llegando a los tendones y los huesos. El diagnóstico de la micobacteriosis atípica se fundamenta en la sospecha clínica ante un historial clínico que sugiere exposición a fuentes de contagio o exposición, en donde pudieran estar presentes MNT. El diagnóstico de confirmación se realiza a partir de biopsias obtenidas de los órganos o tejidos con lesión nodular a los cuales se les practican estudios inmunohistoquímicos, histológicos y microbiológicos. Aunado a los aportes locales e internacionales que la piscicultura ofrece, en la actualidad esta actividad se enfrenta a una gran variedad de factores de riesgo que pueden afectar la productividad, entre los que se incluyen la incidencia y dispersión de enfermedades entre las poblaciones de peces, algunas de las cuales pueden tener importancia zoonótica como el caso de las MNT.

Full textIn the book “Lietuvių literatūra svetur 1945-1967” K. Keblys says, that the novel “Miškais ateina ruduo” (1957) by M. Katiliškis together with the works of such Lithuanian writers as “Mėnesiena” (1957) by A. Baronas and “Balta drobulė” (1958) by Škėma stood out from other works written in 1956-1959. The novels of the first two authors were breath fresh impetus in Lithuanian literature. The biggest contribution was of the novel “Balta drobulė” by A. Škėma and the period of 1956-1959 is associated with the novel “Balta drobulė” by A. Škėma. But M. Katiliškis with this novel stands out from other writers in Lithuanian literature because of its bold spread of eroticism. The topic of the thesis is the spread of eroticism in the novel “Miškais ateina ruduo” by M. Katiliškis. The selection of the topic was determined by the wish to reveal the attitude of the author and his contemporaries towards erotika, to see how each of them perceived and portrayed rather relevant subjects which seem unacceptable (taboo) subjects to Lithuanian nation. The aim of this paper is to investigate the spread of the eroticism in the novel “Miškais ateina ruduo” by M. Katiliškis. Full textMålet med denna uppsats är att ta reda på hur solceller kan förbättra energieffektiviteten ombord M/S Sydfart. Uppsatsen baseras på ett antal energimätningar ombord M/S Sydfart. Solcellernas yta beräknas med hänsyn taget till tillgänglig yta för att installera solpaneler. Globalstrålningsdata kommer ifrån STRÅNG`s databas, den datan används sedan för att beräkna en teoretisk producerad effekt utav solcellerna. Energieffektiviteten analyseras på två sätt. Den första analysen ser på hur mycket av den dagliga energiförbrukningen som kan tillgodogöras med solceller. Den andra analysen baseras på hur stor för- ändring av EEOI som kan uppnås med installerade solceller. Elenergibehovet ombord M/S Sydfart, under sommarhalvåret, kan nästan helt täckas med solceller. Uppsatsen visar dock en del nackdelar med solceller. Under vinterhalvåret är den producerade elenergin från solcellerna låg och en orimligt stor yta solcellsmoduler behövs för att täcka elenergibehovet. Två scenarion utvärderas. I scenario ett utnyttjas all tillgänglig yta för installation av solcellsmoduler. Detta ger ett stort energiöverskott på sommarhalvåret. I scenario två halveras ytan för solcellsmoduler och energiöverskottet blir då mindre. Med hänsyn till att M/S Sydfart har begränsad möjlighet att ta till vara på energiöverskottet, rekommenderas scenario två. Resultatet visar att energieffektiviteten kommer att förbättras. Beroende på årstid, kan EEOI förbättras mellan 0.5% och 12 %. Något krav på förbättring av EEOI finns inte från IMO. Det är rederierna själva som sätter upp interna mål för förbättring.

Synthetic genes from the red marine algae Hypnea musciformis (HML) and Bryothamnion triquetrum (rBTL) were cloned into differents vectors and transformed into several bacterial expression strains. The recombinant lectins were obtained from the soluble fraction of bacterial cultures using Escherichia coli Rosetta-gami 2 (DE3) strain for rHML and E. coli BL21 (DE3) strain for rBTL. Haemagglutination tests showed that rHML and rBTL are able to agglutinate rabbit erythrocytes with strong haemagglutination activity only after treatment with papain and trysine indicating that their ligands are not directly accessible at the cell surface. The haemagglutinating properties of rHML and rBTL confirm the correct folding and functional state of the proteins. A study of the specificity of these lectins by glycan array was conducted. HML, BTL and rBTL showed a restricted specificity for complex N-glycans with core (α1-6) fucose. A more detailed analysis of the specificity of these lectins showed a preference for non bisecting N-glycans, bi- and tri-antennary branching sugars with short chains. Addition of Sialic acid at the non-reducing end of N-glycans favors their recognition by the lectins. This is the first characterization of lectins from red algae by glycan array. An interaction between BTL and a core (α1-6) fucosylated octasaccharides was also observed by STD-NMR. The toxic activity of wild and recombinant lectins were evaluated against Artemia sp. and the human lung adenocarcinoma cell line (A549). In cytotoxicity assays, HML, rHML, BTL and rBTL showed no toxicity against Artemia sp. Only HML and rHML showed a low cytotoxic activity against cell line (A549). The first crystal of rBTL was obtained in micro-scale level using a robot and diffracted at 15 Å.
Os genes sintéticos das lectinas das algas marinhas vermelhas Hypnea musciformis (HML) e Bryothamnion triquetrum (BTL) foram clonados em diferentes vetores e transformados em diferentes células bacterianas de expressão. As lectinas recombinantes foram obtidas a partir da fração solúvel das culturas bacterianas de Escherichia coli Rosetta-gami 2 (DE3) para rHML e BL21 (DE3) para rBTL. Os testes de hemaglutinação mostraram que rHML e rBTL são capazes de aglutinar eritrócitos de coelho tratados com diferentes enzimas proteolíticas. As propriedades hemaglutinantes de rHML e de rBTL confirmam o enovelamento correto e o estado funcional das proteínas. A caracterização da especificidade de ligação a carboidratos da HML, BTL e da rBTL por glycan array mostrou uma especificidade restrita por oligossacarídeos complexos contendo o núcleo de fucosilação (α1-6), com uma preferência particular por N-glicanos não bisectados, bi e tri-antenados de cadeia curta. A presença de ácido siálico na extreminada não-redutora dos glicanos favorece o reconhecimento. Essa foi a primeira caracterização de lectinas de algas vermelhas por glycan array. Experimentos de STD-RMN com a BTL mostraram uma interação com um octassacarídeo contendo o núcleo de fucosilação (α1-6). A atividade tóxica das lectinas selvagens e recombinantes foi avaliada contra Artemia sp. e contra células de adenocarcinoma de pulmão (A549). Nos ensaios de citotoxicidade, HML, rHML, BTL e rBTL não mostraram nenhuma toxicidade contra Artemia sp. e somente HML e rHML mostraram uma baixa toxicidade contra células de adenocarcinoma de pulmão (A549). O primeiro cristal de rBTL foi obtido a um nível de microescala com a ajuda de um robôt de cristalização e difratou a 15 Å de resolução.

Full textThe object of master‘s thesis is the Journalism studies at Vilnius University from its beginning in 1949 to 1991 when the Departments of Printed Journalism, Radio and Television were converted to the Journalism Institute. The main objective of this thesis is to analyze the journalism education in Vilnius University from 1949 to 1991. Primary purpose is to review the main tendencies of soviet high education system and to research the processes of reorganizing the academic departments of Journalism studies. In addition to analyze the changes that took place in Journalism study programme, to represent the academic staff, to introduce the statistics revealing how many school-leavers wanted to enter studies of journalism and how many of them succeeded. The researcher used theoretical and empirical methods: comparative historical, data analysis, interview research methods. The majority of the information and data sources were found in the Archives of Vilnius University (legislations of the rectors, History and Philology Faculty’s protocols (it was reorganized to History Faculty in 1968) council and Journalism Institute’s sessions, personnel department files): it was the only valuable source providing the accurate information about Journalism studies and their development in 1949-1991. Resumo: No capitulo um, apresentamos o planejamento e a síntese de nucleases artificiais baseadas na estrutura da (+/-)-tripargina (19), que poderia intercalar no ADN e apresenta um grupo guanidínico que pode se ligar a grupos fosfatos. Esta foi preparada usando uma estratégia desenvolvida no nosso laboratório em escala multigramas. Dois novos análogos contendo um resíduo guanidínico adicional foram preparados, visto que estes podem aumentar a atividade catalítica desses compostos. O derivado 1,2-bisguanilado 20 foi preparado em 8 etapas com 37% de rendimento global. O análogo 1,9-bisguanilado 21 foi sintetizado com 13% de rendimento para 10 etapas. Também foram preparados três análogos 22-24 contendo uma cadeia hidroxílica lateral em bons rendimentos totais (55, 52 e 31%, respectivamente) a partir do ácido 4-aminoburitírico, bem como três intermediários avançados 70d-72d com duas cadeias guanidínicas e uma cadeia hidroxílica. Estes últimos foram preparados em 12 etapas a partir da triptamina em rendimentos globais variando entre 9-14%. Apesar dos esforços, não encontramos uma condição em que observássemos a atividade catalítica para a (+/-)-tripargina (19) e os derivados bisguanilados 20 e 21, mesmo tendo sido observado que se tenha visto por titulação usando-se RMN-31P uma interação supramolecular entre 19 e o p-nitrofenilfosfato de sódio, com predominância do complexo 1:1 em solução. No capítulo dois, descrevemos a síntese convergente da lingbiabelina M (95) com a finalidade de elucidar sua estrutura tridimensional. Pela estratégia inicial, esta foi dividida em três fragmentos principais: dois deles continham anéis tiazólicos 101 e 106 e foram preparados usando-se uma química clássica para a formação desses heterociclos. A parte policetídica foi sintetizada aplicando-se a metodologia de Masamune para se obter o ácido 107 em 19% de rendimento para 6 etapas. Para finalizar a síntese, os fragmentos 101, 106 e 107 foram acoplados em 49% de rendimento para 6 etapas. Pode-se, assim, confirmar que o produto natural 95 apresenta a esterioquímica (2S, 3S, 14R, 20S) proposta por Gerwick e colaboradores quando de seu isolamento
Abstract: In chapter one, the design and synthesis of artificial nucleases based on the structure of (+/-)-trypargine (19) are introduced. These compounds which contain a guanidine group known to be involved in molecular recognition in biological systems could present the propensity to insert into DNA. Two new analogues containing an additional guanidinic group were prepared, since these may enhance the catalytic activity of these compounds. 1,2-Bisguanylated compound 20 was prepared in 8 steps in 37% overall yield. The analogous 1,9-bisguanylated 21 was synthesized in 13% global yield over 10 steps. Three more analogs 22-24 containing a hydroxylic side chain were prepared in good overall yields (55, 52 and 31%, respectively) from 4-aminoburitiric acid. The synthesis of three advanced intermediates 70d-72d with two guanidinic groups and one hydroxylic chain in 13 steps from tryptamine (31) in overall yields ranging from 9-14% is also disclosed. Despite all efforts, we were not able to find a condition to observe the catalytic activity for (+/-)-trypargine (19) and bisguanylated derivatives 20 and 21, although some supramolecular interaction was observed by 31P-NMR titration between 19 and the p-nitrophenylphosphate sodium salt, predominantly a 1:1 complex in solution. In chapter two, we have described the convergent synthesis of lyngbyabellin M (95) in order to elucidate its stereochemical nature. By retrosynthetic analysis, our target was divided into three main portions: two of them contained thiazole rings 101 and 106, which were prepared using traditional hetericyclic chemistry. The polyketide core was synthesized through the Masamune anti-aldol reaction, giving acid 107 in 19% overall yield over 6 steps. To complete the synthesis, the fragments 101, 106 and 107 were coupled in 49% yield over 6 steps. Thus, we confirmed that the natural product 95 has the stereochemistry (2S, 3S, 14R, 20S) proposed by Gerwick et al, as described in their work of isolation

Full textSynthetic genes from the red marine algae Hypnea musciformis (HML) and Bryothamnion triquetrum (rBTL) were cloned into different vectors and transformed into several bacterial expression strains. The recombinant lectins were obtained from the soluble fraction of bacterial cultures using Escherichia coli Rosettagami 2 (DE3) strain for rHML and E. coli BL21 (DE3) strain for rBTL. Haemagglutination tests showed that rHML and rBTL are able to agglutinate rabbit erythrocytes with strong haemagglutination activity only after treatment with papain and trysine indicating that their ligands are not directly accessible at the cell surface. The haemagglutinating properties of rHML and rBTL confirm the correct folding and functional state of the proteins. A study of the specificity of these lectins by glycan array was conducted. HML, BTL and rBTL showed a restricted specificity for complex N-glycans with core (α1-6) fucose. A more detailed analysis of the specificity of these lectins showed a preference for non bisecting N-glycans, bi- and tri-antennary branching sugars with short chains. Addition of Sialic acid at the non-reducing end of N-glycans favors their recognition by the lectins. This is the first characterization of lectins from red algae by glycan array. An interaction between BTL and a core (α1-6) fucosylated octasaccharides was also observed by STD-NMR. The toxic activity of wild and recombinant lectins were evaluated against Artemia sp. and the human lung adenocarcinoma cell line (A549). In cytotoxicity assays, HML, rHML, BTL and rBTL showed no toxicity against Artemia sp. Only HML and rHML showed a low cytotoxic activity against cell line (A549). Full textThe present study was carried out along a coastline including two bays in Ubatuba, SP, Brazil. The main objectives were (1) examine, at six different rocky shores, the time series of decapod larval settlement in order to understand the physical forcing associated to onshore transport, (2) locate neustonic plankton larval aggregations over the diel cycle to infer on the nature of patchiness and to verify the occurrence of vertical migration in the decapod species examined, and (3) test the relationship between larval supply and settlement, at different spatial scales. The role of larval competency was discussed based on evidence obtained by sampling the plankton at distinct within-bay areas and comparing the results both within and between taxa. From May to August 2005, samples were carried out every other day to estimate the larval settlement rate of two different decapod assemblages in artificial collectors. During two settlement pulses recorded for the megalopae of Pachygrapsus transversus, the nycthemeral neustonic occurrence of that and other decapod stages was assessed. Subsamples of brachyuran megalopae were separated to measure their competency level, by means of both larval rearing, thus obtaining time-to-moult estimates, and microscopic moult-staging of readily fixed individuals. Log-linear models were used to test the effect of wind and tide range on settlement rate, and results were confronted to possible scenarios of surface currents provided by hydrodynamic numerical modeling. Planktonic samples revealed that patches of larvae are coincident to slick fringes, probably caused by the formation of internal waves. A suite of shrimp species undertakes a typical diel vertical migration, while a reversed pattern was found for grapsids, which may assist the final onshore transport to settlement grounds due to marine breezes. Swimming crabs did not show any vertical xi migration trend. Competency is suggested to be better estimated using the larval rearing method. In within-bay areas, portunid megalopae appeared to be more competent to settle than those of grapsids, probably because the former are closer to settlement substrates than the latter, which should be first advected to the nearshore. Within-taxon comparisons revealed that neustonic megalopae of P. transversus collected in within-bay areas are less competent than those obtained from benthic collectors. A transitional stage was missing, suggesting that the transition to the benthos and the development into competent stages are more complex than usually assumed. Significant correlations between megalopae supply and settlement rate were not found. The settlement rate of decapods inhabiting the intertidal region of the rocky coast sampled is apparently dependent on the interaction of wind and tide forcing coupled with larval behaviour. Settlement rate of both grapsids and palaemonids follows a semilunar rhythm, with maxima occurring near neap tides, corroborating the hypothesis that tidally-driven internal motions are also responsible for onshore larval transport coastal decapods. Onshore wind-forcing was found to be the key agent, although dependent on the tide regime, while winds blowing from northeast are suggested to bring larvae from source sites. Coastal circulation patterns and spatially-consistent settlement contrasts indicate that Flamengo Bay is an important area of larval deposition.

Full textThe survey aims to increase the knowledge about circuits, methods and times of dissemination of news in the European and Mediterranean space in early Sixteenth century. The Venetian case allows a wide view over the entire spatial extent because of the eminent position that the city occupies at this time as a political and commercial hub, as a 'bridge' between Europe and the Levant. The restricted periodization (1490-1520) focuses on what appears to be a transition phase for the development of structural forms and languages of diplomatic communication, the growth of postal services and the use of the press in political communication and in wide circulation of news. Recovering and extending the working hypothesis of Sardella (Nouvelles et spéculations, 1948), subsequently reused in Braudel's Méditerranée (1969), Danudo's Diari were used to reconstruct, through the monitoring of correspondence's flows, the pattern and functioning of the Venetian information network.

This study provides analysis, conclusions and recommendations to assist the Deputy Commandant (DC), Manpower and Reserve Affairs Department (M&RA) and DC, Programs and Resources Department (P&R) in structure and process decisions concerning Marine Corps Manpower budget execution. DC, M&RA is the owner of the Marine Human Resource Development Process (HRDP) and the Military Personnel Marine Corps (MPMC) appropriation sponsor, while the DC, P&R has budgetary (1517) authority for MPMC budget execution. In contrast, the Navy has both sponsorship and 1517 authority within one cell at N1. By comparing these two services' organizational factors and Planning, Programming, Budgeting, and Execution Systems (PPBES), relevant differences surface, conclusions are drawn, and recommendations offered for improvements. Recommendations include realignment of 1517 authority within MPMC execution, and the melding of the Programs and Budget Branch of Manpower Plans Division, M&RA with the Military Personnel Branch, Fiscal Division, P&R (RFM). This new office will be responsible for all facets of MPMC programming, budgeting, and execution.
