Abstract:
Lysocardiolipin acyltransferase (LYCAT), a cardiolipin (CL)-remodeling enzyme, is crucial for maintaining normal mitochondrial function and vascular development. Despite the well-characterized role for LYCAT in the regulation of mitochondrial dynamics, its involvement in lung cancer, if any, remains incompletely understood. In this study, in silico analysis of TCGA lung cancer data sets revealed a significant increase in LYCAT expression, which was later corroborated in human lung cancer tissues and immortalized lung cancer cell lines via indirect immunofluorescence and immunoblotting, respectively. Stable knockdown of LYCAT in NSCLC cell lines not only reduced CL and increased monolyso-CL levels but also reduced in vivo tumor growth, as determined by xenograft studies in athymic nude mice. Furthermore, blocking LYCAT activity using a LYCAT mimetic peptide attenuated cell migration, suggesting a novel role for LYCAT activity in promoting NSCLC. Mechanistically, the pro-proliferative effects of LYCAT were mediated by an increase in mitochondrial fusion and a G1/S cell cycle transition, both of which are linked to increased cell proliferation. Taken together, these results demonstrate a novel role for LYCAT in promoting NSCLC and suggest that targeting LYCAT expression or activity in NSCLC may provide new avenues for the therapeutic treatment of lung cancer.

Abstract:
Lung cancer is the leading cause of deaths in United States and non-small cell lung cancer (NSCLC) accounts for ∼85% of all lung cancers with a 5-year survival rate of approximately ∼16%. Therefore there is an immediate need to develop new strategies for early detection and more effective treatments options. Mitochondrial dysfunction including but not limited to defects in mitochondrial genomics and dynamics has long been implicated to play a role in human health and disease particularly in cancer initiation, progression and treatment options since it plays a pivotal role in cell death and survival. Lysocardiolipin acyltransferase (LYCAT), a Cardiolipin remodeling enzyme regulating the 18:2 linoleic acid pattern of mammalian mitochondrial cardiolipin, plays a crucial role in maintaining normal mitochondrial function and vascular development. LYCAT was shown to be up-regulated in cancers; however, the role of LYCAT in lung cancer is yet unclear. Probing the protein expression of LYCAT in lung cancer specimens, non-transformed bronchial epithelial cell lines and 5 lung cancer cell lines revealed increased LYCAT expression and activity in all the lung cancer samples and cell lines tested in comparison to the control lung tissues and non-transformed epithelial cell lines. To determine the role of LYCAT in lung cancer, NSCLC cell lines H2122 and H23 were transfected with either scrambled or LYCAT shRNA and differences in serum-induced cell proliferation, migration, clonogenecity and mitochondrial dynamics were determined. Our results demonstrated that down-regulation of LYCAT by shRNA significantly attenuated cell migration, proliferation, and invasion in NSCLC cell lines compared to control cell lines. Furthermore knockdown of LYCAT expression in NSCLC cell lines inhibited mitochondrial fragmentation and enhanced mitochondrial fusion. Taken together, these data demonstrate a strong association between increased LYCAT expression and cell proliferation, motility, invasion and mitochondrial dynamics in NSCLC cells. Thus, development of targeted therapies to reduce LYCAT expression in NSCLC should be beneficial. This work in part was supported by funds from the College of Medicine, UIC and NIH HL98050 to VN.

Abstract The blast colony-forming cell (BL-CFC) was identified as an equivalent to the hemangioblast during in vitro embryonic stem (ES) cell differentiation. However, the molecular mechanisms underlying the generation of the BL-CFC remain largely unknown. Here we report the isolation of mouse lysocardiolipin acyltransferase (Lycat) based on homology to zebrafish lycat, a candidate gene for the cloche locus. Mouse Lycat is expressed in hematopoietic organs and is enriched in the Lin−C-Kit+Sca-1+ hematopoietic stem cells in bone marrow and in the Flk1+/hCD4+(Scl+) hemangioblast population in embryoid bodies. The forced Lycat transgene leads to increased messenger RNA expression of hematopoietic and endothelial genes as well as increased blast colonies and their progenies, endothelial and hematopoietic lineages. The Lycat small interfering RNA transgene leads to a decrease expression of hematopoietic and endothelial genes. An unbiased genomewide microarray analysis further substantiates that the forced Lycat transgene specifically up-regulates a set of genes related to hemangioblasts and hematopoietic and endothelial lineages. Therefore, mouse Lycat plays an important role in the early specification of hematopoietic and endothelial cells, probably acting at the level of the hemangioblast.

Phosphoinositides (PIPs) are key regulators of membrane traffic and signaling. The interconversion of PIPs by lipid kinases and phosphatases regulates their functionality. Phosphatidylinositol (PI) and PIPs have a unique enrichment of 1-stearoyl-2-arachidonyl acyl species; however, the regulation and function of this specific acyl profile remains poorly understood. We examined the role of the PI acyltransferase LYCAT in control of PIPs and PIP-dependent membrane traffic. LYCAT silencing selectively perturbed the levels and localization of phosphatidylinositol-4,5-bisphosphate [PI(4,5)P2] and phosphatidylinositol-3-phosphate and the membrane traffic dependent on these specific PIPs but was without effect on phosphatidylinositol-4-phosphate or biosynthetic membrane traffic. The acyl profile of PI(4,5)P2 was selectively altered in LYCAT-deficient cells, whereas LYCAT localized with phosphatidylinositol synthase. We propose that LYCAT remodels the acyl chains of PI, which is then channeled into PI(4,5)P2. Our observations suggest that the PIP acyl chain profile may exert broad control of cell physiology.

Abstract. The present study revealed the effects of Lycopene enriched tomato extract (LycT) on chemically induced skin cancer in mice. Skin tumors were induced by topical application of 7,12-Dimethylbenz(a)anthracene (DMBA) [500 nmol/100 ul of acetone, twice a week for two weeks] and 12-O-tetradecanoyl phorbol-13-acetate (TPA) [1.7 nmol/100 ul of acetone, twice a week for eighteen weeks] and LycT (5 mg/kg b.w.) was administered orally. Male Balb/c mice were divided into four groups (n = 15 per group): control, DMBA/TPA, LycT and LycT + DMBA/TPA. The chemopreventive response of LycT to skin tumorigenesis was evident by inhibition in tumor incidence, number, size, burden and volume in LycT + DMBA/TPA group when compared to DMBA/TPA group. This was associated with inhibition of cell proliferation in LycT + DMBA/TPA group as observed by the decrease in epidermal morphometric parameters and mRNA and protein expression of proliferating cell nuclear antigen when compared to DMBA/TPA group (p ≤ 0.05). LycT decreased (p ≤ 0.05) the mRNA and protein expression of angiogenic genes (vascular endothelial growth factor, angiopoietin-2, basic fibroblast growth factor) in LycT + DMBA/TPA group, suggesting its anti-angiogenic effects. The increase (p ≤ 0.05) in protein expression of connexin-32 and 43 in LycT + DMBA/TPA group suggests improved inter cellular communication when compared to DMBA/TPA group. Histochemical studies demonstrated that the components of extracellular matrix (fibrous proteins and mucopolysaccharides) were also modulated during skin carcinogenesis and its chemoprevention by LycT. The decrease in cell proliferation parameters and expression of angiogenesis associated genes, modulation of ECM components and increase in expression of connexins suggest that LycT improved multiple dysregulated processes during chemoprevention of skin cancer.

Usaha menurunkan kolonisasi bakteri dilakukan untuk mengurangi kejadian eksaserbasi salah satuanya adalah dengan vaksinasi. Tujuan dari penelitian ini adalah untuk melihat efek pemberian vaksinasi Lyophilized bacterial lysat pada pasien yang telah diketahui jenis potentialy pathogenic microorganism (PPM).Metode: Desain penelitian uji klinis terbuka (open ckinical trial) pada pemberian Lyophilized bacterial lysate dibandingkan dengan Plasebo terhadap pasien PPOK dengan koloni PPM di poliklinik Paru RSUP Dr.M.Djamil Padang periode Januari sampai Juni 2017.Sampel diambil dengan teknik konsekutif (accidental sampling). Hasil: Penelitian pada 33 pasien PPOK stabil dengan PPM yang diberikan Lyophilized bacterial lysat dan 22 pasien PPOK stabil dengan PPM yang diberika plasebo. Setelah perlakuan selama 3 bulan pada kelompok Lyophilized bacterial lysat tidak ditemukan lagi koloni bakteri ganda (10 menjadi 10) (p=0,002) sedangkan pada bakteri tunggal masih ditemukan koloni, sebelum 23 menjadi 26. Kelompok plasebo tidak didapatkan perubahan baik bakteri ganda (13 menjadi 13) ataupun tunggal (9 menjadi 9). Pada kelompok Lyophilized bacterial lysat terjadi perubahan pola koloni yaitu klebsiella pneumonia berkurang 6, Pseudomonas Aureginosa berkurang 2, Stapilococcus Areus berkurang 6, secara statistik mengalami penurunan (p=0,001) sedangkan pada plasebo tidak terjadi perubahan pola koloni Simpulan: Lyophilized bacterial lysat menurunkan proporsi koloni PPM oleh bakteri ganda dibandingkan dengan plasebo. Lyophilized bacterial lysat juga mengurani koloni terutama Pseudomonas Aureginosa dan stapylococcus Aureus dan klebsiella pneumonia.

Abstract:
In his famous Wager, Blaise Pascal attempted to adduce prudential grounds on which to base a belief in God. His argument founders, however, on the notorious ‘Many Gods Problem’ the problem of selecting among the many equi-probable gods on offer. Lycan and Schlesinger try to treat the Many Gods Problem as a problem of empirical over-determination, attempting to overcome it using methodologies familiar from empirical science. I argue that their strategy fails, but that the Many Gods Problem can be solved (or dissolved) nevertheless. The solution I offer both avoids the problem faced by Lycan and Schlesinger, and does so while respecting the original Pascalian intuitions to a greater extent than any solutions thus far proffered.

Benjamin Percy's novel Red Moon (2013) navigates the problem of the ‘monster’ in the context of post-9/11 representations of Islamist terrorism. Structured around a series of terrorist atrocities carried out by lycan extremists, Percy's novel employs the werewolf as a figure of monstrous otherness in order to deconstruct the very processes of othering by which the monster is produced culturally and politically. Focusing on the distorted ethical justifications of the terrorists and on the roles of political opportunism and media manipulation in shaping US responses, the narrative allows both lycan terrorists and their political antagonists to emerge as more clown than monster. This article draws upon Hannah Arendt's account of the banality of evil, and its development by more recent privation theorists, to situate Red Moon within contemporary popular and theoretical discourses of evil and to read the novel as an interrogation of the processes by which our modern political ‘monsters’ are created.

Brentano (1874) famously claimed that two features demarcate the mental: consciousness and intentionality. Although he claimed that these features are intimately related, subsequent generations of philosophers rarely treated them together. Recently, however, the tide has turned. Many philosophers now accept that consciousness is intentional, where to be intentional is to have representational content, is to represent ‘things as being thus and so — where, for all that, things need not be that way’ (Travis, 2004, 58). In fact, weak representationalism, which holds that perceptual experiences have representational content, is ‘now fairly uncontroversial’ (Lycan, 2004).

The explanatory gap refers to the lack of concepts for understanding “how it is that . . . a state of consciousness comes about as a result of irritating nervous tissue.” By assuming that there are colours in the outside world, Block needlessly widens this gap and Lycan and Kitcher simply fail to see the gap. When such assumptions are abandoned, an unnecessary and incomprehensible constraint disappears. It then becomes clear that the brain can use its own neural language for representing aspects of the outside world. While this may not close the gap, it becomes clearer where we need new concepts.

Body mobil KMHE yang dilombakan oleh kemendikbud tahunan, kebanyakan menggunakan rangka aluminium. Penelitian ini mengajukan bahan carbon fiber-honeycomb sebagai pengganti rangka aluminium. Dalam penelitian metode yang digunakan adalah melapisi honeycomb dengan lapisan carbon fiber-resin. Cara pelapisan bagian dari 1 sampai 3 lapisan pembuatan material menggunakan hand lay-up. Standar specimen yang digunakan adalah ASTM D 638-02 dan ASTM D 790 – 02. Hasil penelitian ini menunjukan bahwa berturut-turut kekuatan tarik dan bending tertinggi di dapat pada 3 lapisan atas komposit karbon fiber-honeycomb 3449,49 kg/mm2 dan untuk pengunjian bending 203,65 kg/mm2. Jika dibandingkan dengan aluminium maka bahan ini dapat digunakan sebagai body mobil KMHE

This research was done to evaluate flexural properties (flexural strength, flexural modulus, and failure mode) of Bambu Tali (Gigantochloa Apus) composite. Lycal 1101 was used as matrix. Two types of laminate were tested in four point bending, unidirectional (00/00) s and symmetric cross ply (00/900) s. Results were then compared with numerical simulation using MSC PATRAN-NASTRAN to determine flexural strength and failure mode prediction for both specimens. All specimens were manufactured using cold press manufacturing method. Results show there was around 28% reduction in flexural strength from unidirectional specimen to symmetric cross ply. Numerical simulation results on failure mode prediction show good agreements with actual four point bending test.

Background: The purpose of this study is to determine the interrater reliability of dynamic muscle tests in the early rehabilitation phase in women after breast cancer surgery with axillary lymph node dissection (ALND) based on the “preventive intervention against lymphedema after breast cancer” (LYCA) randomised controlled trial. Methods: Fifteen women treated with breast cancer surgery including ALND were recruited from participants in the LYCA trial. In this interrater reliability study, women were tested in 4 dynamic muscle tests by 2 physiotherapists at a Capital Hospital in Denmark. Intraclass correlation coefficients (ICCs) with 95% confidence intervals (CIs) was used to assess the relative reliability between raters. A Bland-Altman plot and limits of agreement were calculated to describe the absolute reliability. Results: All 6 subtests displayed high interrater reliability. ICC values were: leg press 0.96 (95% CI = 0.87-0.99), elbow flexion (contralateral) 0.94 (0.83-0.98), elbow flexion (affected arm) 0.93 (0.80-0.98), elbow extension 0.80 (0.41-0.93), shoulder abduction (contralateral) 0.89 (0.68-0.96), and shoulder abduction (affected arm) 0.91 (0.74-0.97). Cumulated interrater reliability for the test battery was very high (ICC = 0.99, 95% CI = 0.990-1.0). The absolute reliability of this study was considered high, and the absence of large shifts between mean and the line of 0 difference suggest no systematic bias that could influence clinical interpretation. Conclusions: The dynamic muscle tests evaluated in this study had high interrater reliability and can be used reliably in women in the postoperative phase after breast cancer surgery with ALND.

Im Zuge der Erarbeitung der Richtlinie VDI 4254 Blatt 2, die die Emissionsmessung von Endotoxinen an Betriebsanlagen beschreibt, wurden Vergleichsmessungen mit Limulus-Amöbozyten-Lysat(LAL)- und rekombinanter-Faktor-C(rFC)-Tests durchgeführt und Verfahrenskenngrößen für das vollständige Messverfahren nach der Richtlinie VDI 4219 bestimmt. Hierfür fanden an einer zwangsbelüfteten Hähnchenmastanlage zum Zeitpunkt hoher bakterieller Emissionen 25 parallele Probenahmen (Doppelbestimmungen) mit dem Impingement-Verfahren statt. Die Endo- toxinaktivitäten wurden von fünf Laboren mithilfe von LAL-und rFC-Tests verschiedener Hersteller gemessen. Beide Methoden erwiesen sich als grundsätzlich für die Analytik von Emissionsproben geeignet, lieferten jedoch Konzentrationsangaben in unterschiedlicher Höhe. Da der rFC-Test keine Pfeilschwanzkrebse benötigt und die Messunsicherheit niedriger ist als beim LAL-Test, wurde für die Aktivitätsbestimmung von Endotoxinen in Emissionsproben nach der Richtlinie VDI 4254 Blatt 2 ausschließlich die Analytik mit dem rFC-Test beschrieben und standardisiert.

Exponents of an ‘explanatory gap’ between physical, functional and intentional facts, on the one hand, and the facts of phenomenal consciousness, on the other, argue that there are reasons of principle why phenomenal consciousness cannot be reductively explained: Jackson (1982), (1986); Levine (1983), (1993), (2001); McGinn (1991); Sturgeon (1994), (2000); Chalmers (1996), (1999). Some of these writers claim that the existence of such a gap would warrant a belief in some form of ontological dualism (Jackson, 1982; Chalmers, 1996), whereas others argue that no such entailment holds (Levine, 1983; McGinn, 1991; Sturgeon, 1994). Das bisher in vielen Laboratorien etablierte Verfahren zur Quantifizierung von Endotoxin ist der Limulus-Amöbozyten-Lysat-(LAL)-Test. Endotoxin wird dabei anhand seiner Fähigkeit quantifiziert, Enzyme aus der Hämolymphe des Pfeilschwanzkrebses zu aktivieren. Um einerseits auf tierisches Material zu verzichten und andererseits eine Verbesserung der Standardisierung zu erreichen, haben verschiedene Hersteller auf der Basis eines rekombinant hergestellten Enzyms Alternativen entwickelt (rFC-Test). Die Umstellung auf ein neues Verfahren, von einem chromogen-kinetischen LAL-Test auf einen einstufigen Fluoreszenzassay, erfordert eine Validierung des neuen Testsystems und Prüfung der zu erwartenden Veränderungen. Ziel der vorliegenden Arbeit war es deshalb, den rFC-Test zusätzlich zum LAL-Test zu etablieren und anhand von Staubfilterextrakten die beiden Methoden zu vergleichen. Dabei erwies sich der rFC-Test als geeignet, den LAL-Test zu ersetzen. Messergebnisse aus beiden Verfahren korrelierten hoch signifikant. Um allerdings Messwerte, z. B. für Arbeitsplatzüberwachungen, über einen längeren Zeitraum miteinander zu vergleichen, ist es erforderlich, immer dieselbe Methode einzusetzen.

Periodic transcription was affected in the mutants as well, since the HTA1 gene was transcribed during the G1 and G2 phases of the cell cycle, periods in the cell cycle when this gene is normally not expressed. A similar loss of cell cycle-dependent transcription was noted for two of the three remaining histone loci, while the HO and CDC9 genes continued to be expressed periodically. Using isolated promoter elements inserted into a heterologous cycl-lacZ fusion gene, we demonstrated that the mutations fell in genes which acted through a negative site in the TRT1 H2A-H2B promoter.

How do you know your own beliefs? And how well do you know them? The two questions are related. I'll recommend a pluralist answer to the first question. The answer to the second question, I'll suggest, varies depending on features of the case.Self-scanning. Shaun Nichols and Stephen Stich (2003) say this: You have in your mind a functionally defined “belief box.” To believe some propositionP is just to have a representation with the content “P” in the belief box. You also have a monitoring mechanism that can scan the contents of the belief box. Normally, you come to know what you believe by deploying that scanner, creating a new belief in the belief box, a belief with the content “I believe thatP.” Self-scanning accounts admit of many possible variations and complications (e.g., Armstrong 1968, 1981, 1999; Lycan 1996; Goldman 2006), but the basic idea is that people have one or more interior monitors or scanners that detect the presence of beliefs and produce, as output, beliefs about those beliefs (or judgments about those beliefs, or representations of those beliefs).

Pada saat ini buah pinang sudah digunakan dalam bidang medis dan kosmetik. Namun akibat dari pengolahan tersebut masih menyisakkan limbah berupa serat buah pinang. Potensi tersebut dapat digunakan sebagai serat dalam pembuatan komposit. Pembuatan komposit kali ini menggunakan resin lycal sebagai matrik dan menggunakan metode vaccum bagging. Penelitian ini bertujuan untuk mengetahui Analisa teknis serta sifat mekanis dari pembuatan komposit serat pinang untuk mendapatkan hasil kekuatan tarik dan bending. Pada pengujian tarik mengacu pada ASTM D-638 dan pengujian bending mengacu pada ASTM D 790. Pada variasi perendaman NaCl didapat nilai tensile tertinggi yaitu 21,78 MPa dan pada variasi perendaman NaOH didapat nilai tensile tertinggi yaitu 30,21 MPa. Pada pengujian bending variasi perendaman NaCl mendapat nilai kekuatan bending tertinggi dengan nilai 509,58 MPa dan untuk variasi perendaman NaOH mendapat nilai kekuatan bending tertinggi dengan nilai 555,82 MPa.

Marbles are secondary decomposition products formed by metamorphism of limestone. Effective classification of marble quarries in terms of quality enables the selection of a sustainable production method and safety application. This evaluation is based on physico-mechanical properties of the samples. Obtained results of physico-mechanical properties of the marbles were statistically analyzed using Stata 14 and SPSS 21 software. The marbles indicated mostly normal physical and mechanical properties. A strong inverse relationship exists between Abrasion Value and Knoop Hardness Determination that indicates a significant nonlinear relationship. Samples were distinguished into 3 groups of close similarity and related properties. The estimated value of the parameters is in the 95 % confidence interval. The equation obtained by regression analysis was used for the determination of resistance to abrasion.

The Secret of Chloris. On the Greek Anthroponyms in Horace’s Odes The trend of poesis docta in Horace’s Odes manifests itself, inter alia, in the sphere of anthroponomy, as many of the names introduced by the poet into his works are of Greek origin. These are pseudonyms, the meaning of which refers to topography (e.g. Lydia, Lycidas, Lyca), to Greek literary tradition (e.g. Neobule, Telephus) and pseudonyms in which there are allusions essential for reading the sense of the work or its part (e.g. Glycera, Lalage, Chloe, Foloe, Chloris, Myrtale, Pyrrus). In poetic translations into Polish, these meaningful nomina propria are not translated, which means that the recipient of the translation is not conscious of the allusion hidden behind the name, and thus is deprived of the possibility of decoding a vastrange of subtexts. The article discusses possible translation solutions that can meet the expectations of various target groups of Polish readers, especially those who do not have linguistic preparation in the field of ancient languages.

Abstract:
The objective was to improve fluvastatin prescribed pulsatile release formulation to get the disintegrative and ruptured lag-time mechanism with a fixed time delay which matches the chronotherapeutics (hypercholesteroidal disorder). Pre formulation studies UV, FTIR (Drug excipient compatibility), solubility studies and flow properties were evaluated for blend and drug. All the values were within the limit. 12 core tablets were prepared with two novel disintegrants, i.e. ludiflash, lycoat in different concentrations after doing the post-compression parameters & drug release F8 was optimized & then coated with PH sensitive polymers HPMC K200M & Ethylcellulose in different concentrations. An evaluation was carried out for all six formulations, and all the values were within the limit. Based on In-vitro dissolution studies, swelling index and rupture test C5F8 is optimized and compared with the marketed product for 10 hours. As per the ICH guidelines optimized formulation (C5F8), stability tests were conducted for three months and was found to be stable. Optimized formulation (C5F8) contains 3:2 polymers (HPM K200M: Ethylcellulose) demonstrates an outstanding pulsatile drug delivery relative to the branded version (Lexcol XL) compared to all other formulations.
