Abstract:
ABSTRACTThis research aims to explore and describe the ethnomathematics on Pasuruan society from the concept of geometry for elementary school on the Pasedahan Suropati batik motive, the philosophical meaning of it, and the alternative usage in teaching geometry in primary school. This research is a qualitative research with ethnographic approach, in wich the technique of collecting a literature study, observation, interviews, fields note, and documentation. Based on the data collecting, there are 6 kinds of Pasedahan Suropati batik in Pasuruan, they are Suruh Leaves motive, Podang Bird motive, Harmoni Built motive, Tembikar motive, Phoniex Bird motive, and Mangrove motive. The meaning of Pasedahan Suropati batik philosophically is Untung Suropati is very wise person, so people who wear this Pasedahan Suropati batik is expected has a wise attitude Untung Suropati. The concept of geometry of elementary school on Pasedahan Suropati batik motive is the concept of point, straight lines, curved lines, zig-zag lines, high lines, parallel lines, angels, triangle, rectangle, oval, and fold symmetry. Thus, the geometry concept of Pasedahan Suropati batik motive in elementary school, can be used in learning geometry such as the introduction of line, the introduction of the angle, and the introduction of shape. ABSTRAKPenelitian ini bertujuan untuk menggali dan mendeskripsikan etnomatematika yang ada pada masyarakat Kota Pasuruan yang dilihat dari konsep geometri untuk sekolah dasar yang terdapat pada motif batik Pasedahan Suropati, makna filosofis yang terkandung di dalamnya, serta alternatif penggunaannya dalam pembelajaran geometri di sekolah dasar. Penelitian ini merupakan penelitian kualitatif dengan pendekatan etnografi, yang mana teknik pengumpulan datanya dilakukan dengan cara studi kepustakaan, observasi, wawancara, catatan lapangan, dan dokumentasi. Berdasarkan hasil pengumpulan data penelitian diperoleh 6 jenis motif batik Pasedahan Suropati yang ada di Kota Pasuruan, yaitu motif Daun Suruh, motf Burung Podang, motif Gedung Harmoni, motif Tembikar, motif Burung Phoenix, dan motif Mangrove. Secara filosofis batik Pasedahan Suropati memiliki makna bahwa Untung Suropati adalah orang yang sangat bijaksana, jadinya orang yang memakai batik Pasedahan Suropati diharapkan memiliki sifat bijaksana laksana Untung Suropati. Konsep geometri untuk sekolah dasar yang ada pada motif batik Pasedahan Suropati adalah konsep titik, garis lurus, garis lengkung, garis zig-zag, garis tinggi, garis sejajar, sudut, segitiga, persegipanjang, oval, dan simetri lipat. Dengan demiian, motif batik Pasedahan Suropati yang memiliki konsep geometri untuk sekolah dasar, tentunya dapat digunakan dalam pembelejaran geometri seperti pada pengenalan garis, pengenalan sudut, dan pengenalan bangun datar sederhana. Abstract:
Lupus autoimmunity frequently presents with neuropsychiatric manifestations, but underlying etiology remains poorly understood. Human brain cytoplasmic 200 RNA (BC200 RNA) is a translational regulator in neuronal synapto-dendritic domains. Here, we show that a BC200 guanosine-adenosine dendritic transport motif is recognized by autoantibodies from a subset of neuropsychiatric lupus patients. These autoantibodies impact BC200 functionality by quasi irreversibly displacing two RNA transport factors from the guanosine-adenosine transport motif. Such anti-BC autoantibodies, which can gain access to brains of neuropsychiatric lupus patients, give rise to clinical manifestations including seizures. To establish causality, naive mice with a permeabilized blood–brain barrier were injected with anti-BC autoantibodies from lupus patients with seizures. Animals so injected developed seizure susceptibility with high mortality. Seizure activity was entirely precluded when animals were injected with lupus anti-BC autoantibodies together with BC200 decoy autoantigen. Seizures are a common clinical manifestation in neuropsychiatric lupus, and our work identifies anti-BC autoantibody activity as a mechanistic cause. The results demonstrate potential utility of BC200 decoys for autoantibody-specific therapeutic interventions in neuropsychiatric lupus.

Abstract:
ObjectiveSLE is an autoimmune disease with a complex pathogenesis. T-cell infiltration into organs contributes to inflammation and organ damage in SLE. Recently, G-protein signalling modulator 2 (GPSM2) has been shown to be implicated in T-cell migration.MethodsWe analysed the expression levels of GPSM2 and of a truncated isoform of GPSM2 containing the GoLoco motif region in CD4+ T cells from patients with SLE and from healthy individuals by western blot. In a next step, we studied the role of the truncated GPSM2 isoform using a CD4+ T-cell migration assay.ResultsOur experiments revealed comparable levels of GPSM2 in CD4+ T cells from patients with SLE and healthy controls. In contrast, the truncated 35 kDa isoform of GPSM2 was significantly more highly expressed in CD4+ T cells from patients with SLE as compared with healthy subjects. Antibody-mediated blockade of the 35 kDa GPSM2 isoform reduced the in vitro capacity of CD4+ T cells to migrate towards the chemokine CCL20.ConclusionsA truncated GPSM2 isoform containing the GoLoco motif region is upregulated in CD4+ T cells from patients with SLE and promotes CD4+ T-cell migration. Targeting this isoform with specific antibodies might be a promising approach to reduce CD4+ T-cell infiltration into inflamed tissues and to prevent organ damage in SLE.

Abstract:
Objective.Systemic lupus erythematosus (SLE) is a prototype of systemic autoimmune disease in which cytokines such as B lymphocyte chemoattractant (BLC, or CXC motif ligand 13, CXCL13) may play important roles in pathogenesis. We investigated the implications of CXCL13 in SLE and lupus nephritis.Methods.Serum samples from 425 patients with SLE and 106 healthy control individuals were analyzed for the concentration of CXCL13 by ELISA. Tissue expression of CXCL13 and its corresponding receptor CXCR5 were observed in lupus kidney. The CXCR5-bearing B cells in SLE patients were analyzed by flow cytometry.Results.Serum levels of CXCL13 were higher in SLE patients compared to controls. SLE patients with lupus nephritis or positive anti-dsDNA antibodies had significantly higher serum CXCL13 levels. The peripheral venous blood B cells that bear CXCR5 were more abundant in SLE patients as detected by flow cytometry. CXCR5 and CXCL13 were highly expressed in the renal cortex from patients with lupus nephritis.Conclusions.Our results suggest that BLC/CXCL13 as well as its corresponding receptor, CXCR5, may play important roles in the pathogenesis of SLE and in lupus nephritis.

Tujuan penelitian ini adalah untuk mengidentifikasikan etnomatematika yang ada pada motif kain tenun Sumba Barat. Penelitian ini termasuk jenis penelitian deskriptif kualitatif dengan pendekatan etnografi. Data dalam penelitian ini adalah aktivitas etnomatematika pada kain tenun Sumba Barat yang diperoleh dari data pustaka dan data lisan. Teknik pengumpulan data pada penelitian ini menggunakan wawancara jarak jauh dan dokumentasi serta vidio. Teknik analisis data dilakukan terhadap data kualitatif (motif-motif yang ditemukan pada kain tenun Sumba Barat). Hasil penelitian di analisis dengan mengidentifikasi konsep matematika yang terkandung dalam motif kain tenun Sumba Barat dan kemudian dideskripsikan sesuai dengan sifat-sifatnya. Hasil penelitian menunjukan bahwa Beberapa motif pada kain tenun Sumba Barat memunculkan konsep matematika yang teridentifikasi sebagai konsep geometri bidang datar seperti garis lurus, garis sejajar, simetri lipat, belah ketupat dan segitiga. Konsep-konsep geometri tersebut dapat ditemukan dalam motif mata kerbau, bintang, mamoli, serta pada desain garis dasar pada kain tenun. Hasil penelitian ini diharapkan dapat diterapkan dalam proses pembelajaran berbasis etnomatematika pada budaya Kabupaten Sumba Barat.

Memiliki berbagai macam motif yang berbeda-beda dari setiap daerah di Indonesia, salah satunya motif Madras yang hingga kini masih mendominasi pada kain Sarung di Indonesia. Terpaku dengan identiknya motif Madras saat ini, diperlukan strategi untuk menghadapi trend-trend baru di masa yang akan datang, sehingga kelestarian kain sarung tidak tergores oleh perkembangan zaman. Inovasi perlu dilakukan karena Semakin berkembangnya motif pada kain Sarung akan memberikan banyak pilihan pada para peminat sekaligus memperluas cakrawala pemanfaatannya. Metode yang digunakan dalam perancangan ini yakni metode kualitatif bertujuan untuk mengetahui keadaan sebenarnya. Wawancara dan observasi adalah dua metode pengumpulan data kualitatif sebagai bagian dari penelitian. Hasil wawancara dan observasi menunjukan bahwa motif pada saat ini kurang variatif dikarnakan keterbatasan mesin yang digunakan. Mesin ATBM (alat tenun bukan mesin) hanya bisa menghasilkan motif madras dan garis tegak lurus saja, cukup sulit diterapkan pada banyaknya permintaan konsumen yang menginginkan motif kain Sarung yang lebih modern. Seperti halnya pada komunitas pemuda hijrah yang memiliki harapan motif kain Sarung bisa mewakili kaum muda saat ini. Abstract:
Background Systemic lupus erythematosus (SLE) is characterized by a type I interferon (IFN) signature, and traditional methods for its measurement like gene expression analysis are cumbersome for routine use. Thus, we aimed to study galectin-9 as a biomarker and compared it with a validated marker, C-X-C motifchemokine ligand 10(CXCL-10). Methods Ninety-seven patients with SLE (26 years; 89 females) were included and stratified based on renal involvement and activity into - active (SLEDAI > 4) renal (35), active non-renal (32) and inactive renal subgroups (30) along with 20 healthy controls (HC, 25 years; 15 females). The median disease duration was 24 months (6-48), and SLEDAI 2K was 9 (2–15). Serum and urine galectin-9 and CXCL-10 levels were measured by ELISA. Urine levels were normalized with spot urine creatinine values. Follow-up serum and urine galectin-9 levels were measured for those in the active renal group at 6 months. Results Patients with SLE had higher serum galectin-9 (5.6 vs 1.7 μg/mL, p = .0001) but not urine galectin-9 (0.52 vs 0.32 μg, p = .7) levels as compared to HC. Serum galectin-9 but not urine galectin-9 was higher in patients with active as compared to inactive lupus (12.9 - active renal, 16.7 - active non-renal vs 3.57 μg/mL, p = .04 and .005). Serum CXCL-10 (0.16 vs 0.05, p = .01) and urine CXCL-10 (0 vs 0, p = .01) were both significantly higher in the SLE group as compared with HC. Serum but not urine CXCL-10 was higher in the active as compared to inactive lupus (0.2 - active renal, 0.3 - active non-renal vs 0.08 μg/mL, p = .9 and .02). Serum galectin-9 showed a modest correlation with CXCL-10 0.4 (0.2–0.6), whereas none was found between their urine levels. Serum galectin-9 and CXCL-10 showed a moderate positive correlation with SLEDAI 2K. Serum galectin-9 showed a greater AUC than CXCL-10 (0.77 vs 0.67) in differentiating active from inactive SLE, and both tested together had the best AUC of 0.82. However, urinary levels had no association with SLEDAI 2K or renal SLEDAI. In a subset of patients with active renal disease, serum galectin-9 but not urine levels declined significantly after 6 months. Conclusion Serum galectin-9 is a good marker of lupus activity; however, it does not differentiate between active renal and active non-renal disease. It performs slightly better than CXCL-10. Urinary galectin-9 does not reflect renal activity.

El lupus eritematoso sistémico es una enfermedad autoinmunitaria que puede debutar con un espectro clínico de presentación muy amplio. Al ser una enfermedad potencialmente mortal es necesario un conocimiento exhaustivo del tema. En ocasiones puede debutar con manifestaciones neurológicas, compromiso renal, entre otras, las cuales son predictores de mal pronóstico. Un 15-20% de los pacientes desarrolla la enfermedad durante la edad pediátrica; por este motivo se presenta un caso de un paciente pediátrico con diagnóstico de lupus eritematoso sistémico y factores de mal pronóstico. En este artículo se discute sobre la evolución de la enfermedad y su manejo.

Penelitian ini berfokus pada proses pembuatan desain Batiklontara.com dengan memperhatikan unsur dan prinsip desain dalam proses pembuatannya, dengan memperoleh informasi dari Rachmat Nurfansyah, pemilik Batiklontara.com, dan desainer Batiklontara.com, Mustakim dan Darmadi. Adapun data yang dihasilkan adalah proses pembuatan desain motif Batiklontara.com memiliki 2 proses, yaitu dengan metode manual dan metode desain grafis. Proses pembuatan dengan menggunakan metode manual diawali dengan proses pembuatan sketsa awal, kemudian sketsa di scan menjadi bentuk file gambar, lalu digambar kembali menggunakan pen tablet melalui software Adobe Illustrator CC. setelah file diubah ke dalam bentuk vektor, ekspor file gambar tersebut ke dalam software Coreldraw X8 untuk penyusunan komposisi gambar pada kain batik. Setelah itu, desain yang sudah dikomposisikan kemudian memasuki proses pewarnaan. Desain yang telah jadi kemudian diaplikasikan ke dalam bentuk mockup menggunakan software Adobe Photoshop CC. Adapun proses pembuatan menggunakan metode desain grafis memiliki perbedaan hanya di bagian pembuatan sketsa awal. Selebihnya cara dan prosesnya sama saja. Unsur desain yang diterapkan dalam proses pembuatan desain motif Batiklontara.com yaitu garis, contohnya seperti garis lengkung, garis lurus, garis diagonal, garis gradasi, garis putus-putus, garis diagonal, garis zigzag dan garis vertikal. bidang, dan warna. Sedangkan prinsip desain yang diterapkan dalam proses pembuatan desain yaitu prinsip keseimbangan, tekanan, irama, dan kesatuan.

Abstract:
The maintenance of autoreactive B cells in a quiescent state is crucial for preventing autoimmunity. Here we identify a variant of human immunoglobulin G1 (IgG1) with a Gly396→Arg substitution (hIgG1-G396R), which positively correlates with systemic lupus erythematosus. In induced lupus models, murine homolog Gly390→Arg (G390R) knockin mice generate excessive numbers of plasma cells, leading to a burst of broad-spectrum autoantibodies. This enhanced production of antibodies is also observed in hapten-immunized G390R mice, as well as in influenza-vaccinated human G396R homozygous carriers. This variant potentiates the phosphorylation of the IgG1 immunoglobulin tail tyrosine (ITT) motif. This, in turn, alters the availability of phospho-ITT to trigger longer adaptor protein Grb2 dwell times in immunological synapses, leading to hyper–Grb2–Bruton’s tyrosine kinase (Btk) signaling upon antigen binding. Thus, the hIgG1-G396R variant is important for both lupus pathogenesis and antibody responses after vaccination.

El lupus eritematoso sistémico es una enfermedad autoinmune compleja con numerosas manifestaciones clínicas y de laboratorio. La incidencia aumenta notablemente en la mujer. El motivo del predominio en el género femenino aún está en estudio. Se ha descrito en algunos estudios que el diagnóstico de lupus en población pediátrica masculina puede tener peor pronóstico con afectación más severa. Métodos: En este trabajo se describen las características clínicas y serológicas de una población pediátrica mexicana en un hospital mexicano de tercer nivel. Resultados: El trastorno renal fue más alto que otros informes, incluidos los informes de América Latina. Los síntomas constitucionales también fueron muy frecuentes. Y dentro del perfil inmunológico destacamos la positividad de anticuerpos anti-ADN, el consumo de complemento y el anticoagulante lúpico positivo. Conclusión: Creemos que el mejor conocimiento sobre poblaciones homogéneas podría mejorar las opciones de tratamiento. El lupus masculino en nuestro grupo pediátrico mexicano tuvo una mayor incidencia de afectación renal, en comparación con otros estudios reportados en población latinoamericana.

In this study, we used an imiquimod-induced lupus mouse model focused on the TLR7 pathway and proteomics analysis to determine the specific pathway related to viral infection and the related protein expressions in splenic B cells to obtain insight into B-cell responses to viral infection in the lupus model. Materials and Methods: We treated FVB/N wild-type mice with imiquimod for 8 weeks to induce lupus symptoms and signs, retrieved splenocytes, selected B cells, and conducted the proteomic analysis. The B cells were co-cultured with CD40L+ feeder cells for another week before performing Western blot analysis. Panther pathway analysis was used to disclose the pathways activated and the protein–protein interactome was analyzed by the STRING database in this lupus murine model. Results: The lupus model was well established and well demonstrated with serology evidence and pathology proof of lupus-mimicking organ damage. Proteomics data of splenic B cells revealed that the most important activated pathways (fold enrichment > 100) demonstrated positive regulation of the MDA5 signaling pathway, negative regulation of IP-10 production, negative regulation of chemokine (C-X-C motif) ligand 2 production, and positive regulation of the RIG-I signaling pathway. A unique protein–protein interactome containing 10 genes was discovered, within which ISG15, IFIH1, IFIT1, DDX60, and DHX58 were demonstrated to be downstream effectors of MDA5 signaling. Finally, we found B-cell intracellular cytosolic proteins via Western blot experiment and continued to observe MDA5-related pathway activation. Conclusion: In this experiment, we confirmed that the B cells in the lupus murine model focusing on the TLR7 pathway were activated through the MDA5 signaling pathway, an important RNA sensor implicated in the detection of viral infections and autoimmunity. The MDA5 agonist/antagonist RNAs and the detailed molecular interactions within B cells are worthy of further investigation for lupus therapy.

Only a fraction (12%) of 268 "autoreactive" T cell clones derived from lupus-prone mice can selectively induce the production of pathogenic anti-DNA autoantibodies in vitro and accelerate the development of lupus nephritis when transferred in vivo. The CDR3 loops of T cell receptor beta chains expressed by these pathogenic T helper (Th) clones contain a recurrent motif of anionic residues suggesting that they are selected by autoantigens with cationic residues. Herein, we found that approximately 50% of these pathogenic Th clones were specific for nucleosomal antigens, but none of them responded to cationic idiopeptides shared by variable regions of pathogenic anti-DNA autoantibodies. Nucleosomes did not stimulate the T cells as a nonspecific mitogen or superantigen. Only the pathogenic Th cells of lupus responded to nucleosomal antigens that were processed and presented via the major histocompatibility class II pathway. Although the presentation of purified mononucleosomes to the Th clones could be blocked by inhibitors of endosomal proteases, neither of the two components of the nucleosomes--free DNA or histones by themselves--could stimulate the Th clones. Thus critical peptide epitopes for the Th cells were probably protected during uptake and processing of the nucleosome particle as a whole. The nucleosome-specific Th clones preferentially augmented the production of IgG autoantibodies to histone-DNA complex in vitro. In vivo, nucleosome-specific, CD4+ T cells were not detectable in normal mice, but they were found in the spleens of lupus-prone mice as early as 1 mo of age, long before other autoimmune manifestations. Immunization of young, preautoimmune lupus mice with nucleosomes augmented the production of autoantibodies and markedly accelerated the development of severe glomerulonephritis. Previously, crude preparations containing nucleosomes were shown by others to have polyclonal mitogenic activity for B cells from normal as well as lupus mice. Identification here of pure mononucleosome as a lupus-specific immunogen for the Th cells that selectively help the pathogenic anti-DNA autoantibody producing B cells of lupus could lead to the design of specific therapy against this pathogenic autoimmune response.

Abstract Dysregulated B cell activation plays pivotal roles in systemic lupus erythematosus (SLE), which makes B-cell-depletion a potential strategy for SLE treatment. The clinical success of anti-CD19 chimeric antigen receptor (CAR) T cells in treating B-cell malignancies has attracted the attention of researchers. In this study, we aimed to investigate the feasibility of applying anti-CD19 CAR-T cell therapy into SLE treatment in a mouse disease model. We have constructed murine anti-CD19 CARs with either CD28 or 4-1BB as the intracellular costimulatory motif and evaluated the therapeutic functions of corresponding CAR-T cells by infusing them into MRL-lpr mice. We found that adoptive transfer of anti-CD19 CAR-T cells was able to achieve sustained B-cell-depletion in MRL-lpr mice. More importantly, transferring syngeneic anti-CD19 CAR-T cells can not only prevent disease pathogenesis before the onset of disease symptoms but also have substantial therapeutic benefits in disease treatment at a later stage after disease progression. Comparing to CAR-T cells with CD28 as a costimulatory motif, CAR-T cells with 4-1BB costimulatory motif work better for mouse SLE treatment. In summary, our study claimed that anti-CD19 CAR-T cell therapy worked effectively in treating murine SLE, indicating its potential in treating human patients in clinic.

Aicardi-Goutières syndrome is an early-onset encephalopathy with a presumed immune pathogenesis caused by inherited defects in nucleic acid metabolism. The clinical picture resembles a congenital viral infection despite negative investigations for common viruses. In addition to leukoencephalopathy with calcifications of basal ganglia, patients show increased levels of the antiviral cytokine interferon-α in cerebrospinal fluid. We report on a 12-year-old boy with Aicardi-Goutières syndrome and systemic lupus erythematosus (SLE) due to mutations in the SAMHD1 (sterile alpha motif domain and HD domain-containing protein 1) gene, illustrating an emerging pattern of the natural history of Aicardi-Goutières syndrome characterized by neurological disease followed by symptoms of systemic autoimmunity. Thus, Aicardi-Goutières syndrome constitutes a model disease for systemic autoimmunity triggered by the activation of the innate immune system. Recognition of the etiologic link between Aicardi-Goutières syndrome and systemic lupus erythematosus has direct implications on therapeutic management and suggests that early immune modulatory intervention can improve neurological outcome.

Neutrophil dysregulation is implicated in the pathogenesis of systemic lupus erythematosus (SLE). SLE is characterized by elevated levels of a pathogenic neutrophil subset known as low-density granulocytes (LDGs). The origin and phenotypic, functional, and pathogenic heterogeneity of LDGs remain to be systematically determined. Transcriptomics and epigenetic assessment of lupus LDGs, autologous normal-density neutrophils, and healthy control neutrophils was performed by bulk and single-cell RNA sequencing and assay for transposase-accessible chromatin sequencing. Functional readouts were compared among neutrophil subsets. SLE LDGs display significant transcriptional and epigenetic heterogeneity and comprise 2 subpopulations of intermediate-mature and immature neutrophils, with different degrees of chromatin accessibility and differences in transcription factor motif analysis. Differences in neutrophil extracellular trap (NET) formation, oxidized mitochondrial DNA release, chemotaxis, phagocytosis, degranulation, ability to harm the endothelium, and responses to type I interferon (IFN) stimulation are evident among LDG subsets. Compared with other immune cell subsets, LDGs display the highest expression of IFN-inducible genes. Distinct LDG subsets correlate with specific clinical features of lupus and with the presence and severity of coronary artery disease. Phenotypic, functional, and pathogenic neutrophil heterogeneity are prevalent in SLE and may promote immune dysregulation and prominent vascular damage characteristic of this disease.

Objectives: To determine the clinical significance of Ro52 protein/tripartite motif-containing 21 antibody and specific antinuclear antibody patterns using indirect immunofluorescence technique. Method: The retrospective study was conducted at the clinical laboratory of the First Affiliated Hospital of Chongqing Medical University, China, and comprised data from January 2017 to December 2021 of patients who underwent antinuclear antibody and anti-extractable nuclear antigen antibody detection. Inpatients with Ro52 antibody-positive status were taken as the cases, while anti-Ro52 negative patients with clear clinical diagnosis were taken as the controls. Data was analysed using SPSS 19. Results: There were 1802 cases and 1211 controls. Positive Ro52 showed significantly greater frequency in patients with primary Sjogren’s syndrome, systemic lupus erythematosus, inflammatory myositis, dry eyes and interstitial lung disease (p<0.05). Ro52 antibody showed high positive predictive value for primary Sjogren’s syndrome 25(96.15%), systemic lupus erythematosus 259(91.20%), connective tissue disease-associated interstitial lung disease 45(86.67%) and inflammatory myositis 60(86.67%). Antinuclear antibody indirect immunofluorescence patterns most frequently detected were nuclear speckled 128(40.89%) and cytoplasmic speckled 126(40.26%) (p<0.05). Interstitial lung disease was associated with the presence of cytoplasmic speckled antinuclear antibody indirect immunofluorescence pattern 24(19.2%), while tumours 47(36.5%) and hepatitis B 26(20.3%) seemed to be more frequent with nuclear speckled pattern (p<0.05). The simultaneous reactivity extractable nuclear antigen antibodies most frequently detected were antinuclear antibody+Ro52+anti-Sjogren's syndrome A+ 558(33.96%). Conclusion: Ro52 antibody positivity was found to be associated with Sjogren’s syndrome, systemic lupus erythematosus, inflammatory myositis, dry eye and interstitial lung disease. The antinuclear antibody immunofluorescence pattern of Ro52 positive was single and primarily granular cytoplasm type. Antinuclear antibody negative and Ro52 positive in the serum of patients also had certain significance in auxiliary disease diagnosis. Hanya UMKM yang lulus seleksi dan memiliki kualifikasi skala produksi mapan yang bisa berpartisipasi. Oleh karenanya, mayoritas peserta pameran kerajinan adalah peserta lama yang telah berulang kali bahkan lebih dari 3 kali mengikuti pameran. Dengan mengetahui faktor yang mempengaruhinya, akan dapat disimpulkan benar atau tidaknya motif utama berpromosi diikuti oleh motif lain yang sama pentingnya. Untuk menghasilkan data yang tepat, penelitian lapangan dilakukan melalui observasi, kuesioner, dan wawancara. Data dianalisis menggunakan analisis faktor eksplorasi. Hasil penelitian menunjukkan bahwa ada 2 faktor yang mempengaruhi, faktor ekonomi dan faktor sosial. Faktor ekonomi termasuk mendapatkan pelanggan dan mendapatkan keuntungan tinggi. Faktor sosial termasuk daya saing, keberadaan/eksistensi, cerita sukses peserta lain, dan pengakuan.

Chemokine C–X–C ligand 10 (CXCL10), also known as interferon-γ-inducible protein 10 (IP-10), exerts biological function mainly through binding to its specific receptor, CXCR3. Studies have shown that renal resident mesangial cells, renal tubular epithelial cells, podocytes, endothelial cells, and infiltrating inflammatory cells express CXCL10 and CXCR3 under inflammatory conditions. In the last few years, strong experimental and clinical evidence has indicated that CXCL10 is involved in the development of renal diseases through the chemoattraction of inflammatory cells and facilitation of cell growth and angiostatic effects. In addition, CXCL10 has been shown to be a significant biomarker of disease severity, and it can be used as a prognostic indicator for a variety of renal diseases, such as renal allograft dysfunction and lupus nephritis. In this review, we summarize the structures and biological functions of CXCL10 and CXCR3, focusing on the important role of CXCL10 in the pathogenesis of kidney disease, and provide a theoretical basis for CXCL10 as a potential biomarker and therapeutic target in human kidney disease.

Abstract TRIM21 belongs to the large family of tripartite motif containing proteins, and is often targeted by autoantibodies in lupus and Sjogren’s syndrome. Considering the significant protein domain homology between different TRIM proteins, we hypothesized that additional TRIM proteins are targets of autoimmunity. Based on the literature, in this study we investigated autoantibody responses to TRIM38. While 9% of lupus patients (n=149) had anti-TRIM38 antibodies, the incidence in Sjogren’s syndrome patients (n=150) was 12%, and in controls (n=50) it was 4%. With respect to TRIM21, the incidence was 62%, 68%, and 4% respectively. In Sjogren’s syndrome patients, the presence of anti-TRIM38 antibodies was closely associated with the increased severity of dry eye parameters. Epitope mapping studies showed that anti-TRIM21 antibodies reacted with the RING, Coiled coil and PRY-SPRY domains of TRIM21, whereas anti-TRIM38 antibodies reacted only with the Coiled coil and PRY-SPRY domains of TRIM38. All anti-TRIM38 positive patients also had anti-TRIM21 antibodies. Affinity purified anti-TRIM21 antibodies from lupus patients did not immunoprecipitate TRIM38, indicating lack of cross-reactivity at B cell level. However, we observed T cell cross-reactivity between TRIM21298-312 and TRIM38302-316. Our study suggests that immune responses to TRIM proteins can evolve through epitope spreading and contribute towards exacerbating the pathogenesis in autoimmune disorders.

Abstract The production of pathogenic anti-DNA autoantibodies in mice with lupus nephritis is dependent on special autoimmune Th cells that can also transfer the disease into preautoimmune mice. In previous work, these pathogenic Th cells were cloned and their TCR beta-chains were sequenced to reveal a recurrent motif of anionic residues in their CDR3 loops. Accordingly, approximately half of the Th clones were found to be specific for nucleosomal Ag that contain cationic residues. Herein, we analyzed the TCR alpha-chain repertoire of 15 of these pathogenic Th clones and found them to be heterogeneous, even among the nucleosome-specific Th clones. Most of these autoimmune TCR alpha-chains contained anionic residues in their CDR3 in addition to cationic residues. Therefore, these pathogenic Th clones of lupus probably recognize epitopes with mixed charge runs that are derived from autoantigens, such as histone-DNA complexes. Interestingly, the V alpha gene segments used by 10 of these Th clones derived from the (SWR x NZB)F1 lupus mice differed from previously reported sequences indicating that they were new members or alleles of the respective V alpha gene family. One of the Th clones used a gene from an entirely new murine V alpha gene family, identified here as V alpha 23, which consisted of approximately two members that were conserved among strains with different V alpha haplotypes. Knowledge of the primary structure of the TCR expressed by these pathogenic Th clones of lupus would help in the analysis of their antigenic specificities and also would be essential for studying their regulation in transgenic mice carrying these autoimmune TCR genes.

Despite great efforts, our knowledge of serum protein patterns in severe SLE phenotypes is still limited.ObjectivesThis study investigated the serum proteomic profile of SLE, in order to identify relevant clinical patterns.MethodsProximity extension immunoassay (PEA, Olink) was used to assess the serum levels of one hundred and eighty-four inflammation and organ damage-related proteins in patients with SLE (n = 72) and age-matched healthy donors (HD) (n = 18). In parallel, an extensive clinical and analytical profile of recruited subjects was performed. To evaluate the contribution of molecular profiles to the disease severity, unsupervised clustering analyses were developed. Gene ontology enrichment analysis were also carried out to interrogate the biological meaning associated with the molecular signatures identified.ResultsSeveral circulating proteins related to inflammation and organ damage were coordinately altered in the serum of SLE patients in relation to HD. Unsupervised clustering analyses differentiated 2 patients clusters presenting different proteomic profiles. Clinically, although no differences were found in terms of age or disease duration, patients belonging to cluster 1 were characterized by a significantly higher status of disease activity (SLEDAI > 6) and prevalence of positivity for anti-ENA and anti-dsDNA antibodies than patients belonging to cluster 2, along with preponderance of patients with biopsy-diagnosed lupus nephritis (LN), abnormal creatinine and proteinuria. Twenty-three serum proteins were found deregulated between clusters. In this article, I trace the various ways Ancient Greek artisans used seashell-motifs, and how they interpreted these natural forms in artistic contexts. The reason for the popularity of seashells lies in their visual and haptic variety, their wealth of association, and the device of something secret hidden under the hard outer shell. I examine three possible variants of the motif’s treatment in relief and volume: the shell-container as part of a vessel, the shell as an independent container, the shell as a sculpted motif on a vessel. Of particular interest are the vases where the integration of shell and vessel is especially marked, such as plastic aryballoi, lekythoi and other types (often with the inclusion of mythological subjects, such as the birth of Aphrodite). Terracotta and metal pyxides are also considered. Alongside these, I examine the rare “marble shells”, stone vessels carved in the intricate shape of “pelican’s foot” shells.

Although the presence of autoantibodies against TRIM21 in various autoimmune diseases suggests potential pathogenetic roles, no studies have clarified its exact implications especially in lupus.ObjectivesWe aimed to elucidate the functions of TRIM21 in dysregulation of type I interferon signals in lupus.MethodsTo investigate effects of TRIM21 dysfunction in lupus pathogenesis, two independent lupus animal models, the R848-induced model and the B6/lprmice model were performed using TRIM21 knockout mice and their phenotypes and immunological profiles were determined. In addition, we investigated the degree of TRIM21 dysfunction and therapeutic effects ofin vivodelivery of TRIM21 in MRL/lprmice. To evaluate the E3 ubiquitin ligase activity of TRIM21 for targeted proteins in type I interferon pathways, we performed specialized immunoblot assay.ResultsThe R848-induced model and the B6/lprmodel both presented with more severe lupus-like phenotypes such as nephritis, lymphadenopathies, and inflammatory immune cell profiles in TRIM21 knockout mice than in control mice. TRIM21 deficiency resulted in activation of intracellular factors related to type I interferon pathways such as STING, TBK1, and IRF3 in both models. MRL/lprmice presented with activation of type I interferon pathways including STING, TBK1, and IRF3, and decreased expressions of TRIM21. Overexpression of TRIM21 attenuated the disease phenotypes in MRL/lprmice. The current problem is that the Angkola Batak Ulos weaving is not widely known to the public, because the types of Ulos in North Sumatra are very diverse. This research was conducted to describe the shape of the motifs, the meaning of the motifs and the weaving technique of Angkola Batak Ulos in South Tapanuli Regency. The research method used is descriptive qualitative with data types in the form of primary data and secondary data. Collecting data in this study using observation techniques, interviews and documentation. The results of the research show that the Ulos Batak Angkola weaving motifs in South Tapanuli Regency consist of 17 motifs, namely pusuk robung, distance, cap mumbang, hiok-hiok, sijobang, singap, simata na maraturan, simata namaradopan, Ruang, lus-lus, bungo ros, iran-iran, letters, naginjang signs, sirat, jojak, suri-suri. The shape of the Ulos weaving motif is geometric and decorative. The source of inspiration for the Angkola Batak Ulos weaving motifs is living things and the surrounding environment. The meaning contained in the Ulos Batak Angkola woven motif is in the form of advice, hopes and prayers as well as the family tree of the people of North Sumatra. The Angkola Batak Ulos weaving technique is to carry out the preparatory process, namely mangunggas, manjomur, mangukul, mangasoli, mangani. After that, weaving can only begin, in weaving the position of the two legs must be straight ahead, the soles of the feet are right on the point. The sitting position must be fixed (not shifting) because it will affect the balance of the body when weaving. In making a motif, the first thing to do is move the guyun first, then carry the guyun with both hands, so that you can see the voids in the warp threads, use balobas to make it easier to count the threads, after that the motif making can be done according to the calculation of the motif. Use the tipak to secure the thread. So that the Ulos Batak Angkola weaving in South Tapanuli Regency can develop according to fashion developments but refers to the characteristics of regional motifs so that it becomes attractive to consumers, for this, targeted guidance and support from the government is needed for weaving craftsmen in South Tapanuli Regency.Keywords: weaving, Ulos Batak Angkola. Setelah itu, baru menenun dapat dimulai, dalam menenun posisi kedua kaki harus lurus kedepan, kedua telapak kaki tepat pada tadokan. Posisi tempat duduk harus tetap (tidak bergeser-geser) karena akan mempengaruhi keseimbangan badan saat menenun. Dalam pembuatan motif, hal yang pertama dilakukan adalah menggerakkan guyun terlebih dahulu, setelah itu baru guyun dijinjing dengan kedua tangan, sehingga terlihat rongga pada benang lungsinya, gunakan balobas untuk mempermudah penghitungan benangnya, setelah itu pembuatan motif dapat dilakukan sesuai dengan perhitungan motifnya. Gunakan tipak untuk merapatkan benangnya. Introducción: el lupus eritematoso sistémico (LES) se conoce como una enfermedad autoinmune multisistémica, cuya causa es desconocida, se caracteriza por la presencia de complejos inmunes y autoanticuerpos. En series internacionales se describe una presentación de procesos infecciosos en estos pacientes hasta en un 75 % de los casos, las infecciones ocurren tanto al inicio de la enfermedad como en etapas tardías, y son causa directa de muerte en 30 % a 60 % de los casos y motivo de hospitalización hasta de 30 %. Objetivos: determinar los procesos infecciosos más frecuentes en pacientes con lupus eritematoso sistémico internados en el Servicio de Clínica Médica del Hospital Nacional en el periodo 2022-2023. Metodología: diseño observacional, descriptivo de corte transversal de pacientes con diagnóstico de lupus eritematoso sistémico según criterios de European League Against Rheumatism (EULAR)-2019 que se encuentran internados en el servicio de Clínica Médica del Hospital Nacional (Itaugua-Paraguay) en el periodo de 2022-2023. Resultados: la edad media de los pacientes fue de 34 años, con predominio del sexo femenino en el 88.18 %. Las infecciones del aparato respiratorio fue la más frecuentemente diagnosticada en 50 (45.45 %) pacientes, seguido por la infección de vías urinarias 47 (42.38 %) pacientes, el condicionante de riesgo predominante fue el uso de corticoides en un total 105 (96.40 %) pacientes, se estableció en un total de 97 (88.18 %) pacientes con antibioticoterapia, dentro del aspecto demográfico de la zona urbana 56 (50.90 %) pacientes y rural 54(49.1 %). Conclusión: la infección del aparato respiratorio fue la infección más frecuente, el condicionante de riesgo predominante es el uso de corticoides y recibieron antibioticoterapia la cual en monoterapia fue la más utilizada.

Protein ubiquitination, which is catalyzed by ubiquitin-activating enzymes, ubiquitin-conjugating enzymes, and ubiquitin ligases, is a crucial post-translational modification to regulate numerous cellular functions in a spatio–temporal-specific manner. The human genome encodes ~100 deubiquitinating enzymes (DUBs), which antagonistically regulate the ubiquitin system. OTUD1, an ovarian tumor protease (OTU) family DUB, has an N-terminal-disordered alanine-, proline-, glycine-rich region (APGR), a catalytic OTU domain, and a ubiquitin-interacting motif (UIM). OTUD1 preferentially hydrolyzes lysine-63-linked ubiquitin chains in vitro; however, recent studies indicate that OTUD1 cleaves various ubiquitin linkages, and is involved in the regulation of multiple cellular functions. Thus, OTUD1 predominantly functions as a tumor suppressor by targeting p53, SMAD7, PTEN, AKT, IREB2, YAP, MCL1, and AIF. Furthermore, OTUD1 regulates antiviral signaling, innate and acquired immune responses, and cell death pathways. Similar to Nrf2, OTUD1 contains a KEAP1-binding ETGE motif in its APGR and regulates the reactive oxygen species (ROS)-mediated oxidative stress response and cell death. Importantly, in addition to its association with various cancers, including multiple myeloma, OTUD1 is involved in acute graft-versus-host disease and autoimmune diseases such as systemic lupus erythematosus, rheumatoid arthritis, and ulcerative colitis. Thus, OTUD1 is an important DUB as a therapeutic target for a variety of diseases.

Using SELEX (systematic evolution of ligands by exponential enrichment), we serendipitously discovered a ssDNA aptamer that binds selectively to the anti-FLAG M2 antibody. The aptamer consisted of two motifs (CCTTA and TGTCTWCC) separated by 2-3 bases, and the elimination of one or the other motif abrogated binding. The DNA aptamer and FLAG peptide competed for binding to the antigen-binding pocket of the M2 antibody. In addition, the aptamer eluted FLAG-tagged proteins from the antibody, suggesting a commercial application in protein purification. These findings demonstrate the feasibility of using SELEX to develop ssDNA aptamers that block the function of a specific antibody, a capability that could lead to the development of novel therapeutic modalities for patients with systemic lupus erythematosus, rheumatoid arthritis, and other autoimmune diseases.

Systemic lupus erythematosus (SLE) is a chronic, multifactorial autoimmune disease that predominantly affects young females. Dysregulation of different immune cell populations leads to self-tolerance breakdown and subsequent multiple organ damage as the disease develops. Plasmacytoid dendritic cells (pDCs) are potent producers of type I interferon (IFN), while myeloid dendritic cells (mDCs) are more specialized in antigen presentations. We have previously reported that bone-marrow (BM)-derived pDCs from the murine lupus model New Zealand black/white F1 (BWF1) possess abnormalities. Therefore, this study continues to investigate what aberrant properties peripheral pDCs and mDCs possess in BWF1 and how they mediate SLE progression, by comparing their properties in pre-symptomatic and symptomatic mice. Results showed that CD11chiCD11b+ myeloid DCs expanded during the disease state with down-regulation of co-stimulatory molecules and major histocompatibility complex class II molecules (MHC II), but their capacity to stimulate T cells was not hampered. During the disease state, this subset of mDCs displayed heightened toll-like receptors 7 and 9 (TLR 7/9) responses with increased interleukin 10 (IL-10) and C-X-C motif chemokine ligand 13 (CXCL13) expressions. Moreover, the expressions of myeloid differentiation primary response 88 (Myd88) and nuclear factor kappa B subunit 1 (Nfkb1) were higher in CD11chiCD11b+ DCs at the disease stage, leading to higher nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) p65 phosphorylation activity. In summary, we reported aberrant phenotypic properties with enhanced TLR7/9 responses of CD11chiCD11b+ DCs in SLE mediated by aberrant NF-κB signaling pathway. Our findings add additional and novel information to our current understanding of the role of DCs in lupus immunopathogenesis. Lastly, molecular candidates in the NF-κB pathway should be exploited for developing therapeutic targets for SLE.

Abstract Expression of Fli-1, a member of the Ets family of transcription factors, is implicated in the development of nephritis in murine models of systemic lupus erythematosus (SLE). Expression of monocyte chemotactic protein-1 (MCP-1) and Chemokine (C-C motif) ligand 5 (CCL5, also known as RANTES) plays an important role in the inflammatory cell infiltration. In this study, we examined the role of Fli-1 on chemokine production and inflammatory cell infiltration in conjunction with nephritis development in NZM2410 mice, an animal model of SLE. We generated Fli-1 heterozygous knockout NZM2410 mice (Fli1+/-; Fli-1 homozygous knockout is embryonic lethal) and wild-type (WT) littermate (Fli1+/+) mice as controls. Expression of MCP-1 and CCL5 in the kidneys from 18-week old Fli-1+/- NZM2410 mice was significantly decreased compared to that from WT littermates. The number of macrophages, neutrophil granulocytes, T cells and B cells in the kidneys from 34-week Fli-1+/- NZM2410 mice decreased by 44 - 75% compared to WT littermate controls. The productions of MCP-1 in endothelial cells transfected with specific Fli-1 siRNA were significantly reduced compared to cells transfected with negative control siRNA. Our data indicate that Fli-1 plays an important role in production of inflammatory chemokine MCP-1 and inflammatory cell infiltration in the lupus nephritis development.

Human red blood cells (RBCs), senescent or damaged due to particular stress, can be removed by programmed suicidal death, a process called eryptosis. There are various molecular mechanisms underlying eryptosis. The most frequent is the increase in the cytoplasmic concentration of Ca2+ ions, later exposure of erythrocytes to oxidative stress, hyperosmotic shock, ceramide formation, stimulation of caspases, and energy depletion. Phosphatidylserine (PS) exposed by eryptotic RBCs due to interaction with endothelial CXC-Motiv-Chemokin-16/Scavenger-receptor, causes the RBCs to adhere to vascular wall with consequent damage to the microcirculation. Eryptosis can be triggered by various xenobiotics and endogenous molecules, such as high cholesterol levels. The possible diseases associated with eryptosis are various, including anemia, chronic kidney disease, liver failure, diabetes, hypertension, heart failure, thrombosis, obesity, metabolic syndrome, arthritis, and lupus. This review addresses and collates the existing ex vivo and animal studies on the inhibition of eryptosis by food-derived phytochemicals and natural compounds including phenolic compounds (PC), alkaloids, and other substances that could be a therapeutic and/or co-adjuvant option in eryptotic-driven disorders, especially if they are introduced through the diet.

El Lupus Eritematoso Sistémico (LES) es una enfermedad sistémica autoinmune. La verdadera incidencia de miocarditis es difícil de determinar; se encuentra en 1 a 9% de las autopsias de rutina y de 5 a 12% de las muertes súbitas no explicadas. La miocarditis debe ser diagnosticada por la combinación de criterios clínicos y patológicos, por su presentación heterogénea. La miocarditis es una manifestación rara, pero potencialmente fatal; por ese motivo su reconocimiento temprano y el manejo de soporte e inicio temprano de terapia inmunosupresora pueden lograr resultados favorables. Reportamos el primer caso documentado de miopericarditis lúpica en una mujer joven sin evidencia de infección viral asociada, realizándose el diagnóstico por la evolución y la respuesta terapéutica. Destacamos además la secuencia de los eventos; inicialmente como pericarditis efusiva masiva y que rápidamente progresó a disfunción sistólica severa del ventrículo izquierdo.

Systemic lupus erythematosus (SLE) is a chronic, systemic, and autoimmune disease, whose etiology is still unknown. Although there has been progress in the treatment of SLE through the use of glucocorticoid and immunosuppressive drugs, these drugs have limited efficacy and pose significant risks of toxicity. Moreover, prognosis of patients with SLE has remained difficult to assess. TRIM21/Ro52/SS-A1, a 52-kDa protein, is an autoantigen recognized by antibodies in sera of patients with SLE and Sjögren's syndrome (SS), another systemic autoimmune disease, and anti-TRIM21 antibodies have been used as a diagnostic marker for decades. TRIM21 belongs to the tripartite motif-containing (TRIM) super family, which has been found to play important roles in innate and acquired immunity. Recently, TRIM21 has been shown to be involved in both physiological immune responses and pathological autoimmune processes. For example, TRIM21 ubiquitylates proteins of the interferon-regulatory factor (IRF) family and regulates type I interferon and proinflammatory cytokines. In this paper, we summarize molecular features of TRIM21 revealed so far and discuss its potential as an attractive therapeutic target for SLE.

Dari segi pengumpulan data, penulis melakukan observasi dengan mengamati gejala yang ada dalam objek penelitian serta wawancara dengan Bhikkhu Khanthidaro selaku Kepala Vihara Dhammadipa Arama, Bhikkhu Santacito, Samanera, Atthasilani, dan dokumentasi berupa buku-buku yang terkait dengan penelitian. Sedangkan pengolahan data penulis menggunakan analisis data yaitu dengan metode deskriptif.Data yang dianalisis adalah orientasi agama bagi Samanera dan Atthasilani. Hasil penelitian menunjukkan dua hal. Pertama, motif yang mendorong menjadi Samanera dan Atthasilani yaitu berasal dari kesadaran diri dan didorong dari keluarga, saudara, Guru, teman yang membuat semakin yakin menjalani kehidupan Samanera serta Atthasilani. Kedua, tahapan menjadi Samanera dan Atthasilani melalui tes yang diuji Bhikkhu dengan menghafal beberapa sila, lalu ditahbis menjadi Samanera dan Atthasilani dengan melalui prosesi cukur rambut dan pemakaian jubah. Pada saat menjalani kehidupan Samanera dan Atthasilani diberi penilaian dalam menerapkan 10 sila bagi Samanera ditambah 75 latihan tambahan dan 8 sila bagi Atthasilani untuk konsisten sampai lulus ke jenjang Bhikkhu maupun Atthasilani tetap.Kata kunci: Orientasi, agama, Samanera, Atthasilani.

Metode yang digunakan dalam penelitian ini menggunakan pendekatan kualitatif dengan jenis penelitian studi kasus. Teknik pemilihan informan dalam penelitian ini menggunakan purposive sampling dengan jumlah informan dalam penelitian ini ada 15 mahasiswa, 5 mahasiswa dari kelas 16, 5 mahasiswa dari angkatan 2017, dan 5 mahasiswa dari angkatan 2018 yang sudah menyelesaikan skripsi atau sudah lulus. Teknik pengumpulan data dilakukan dengan cara wawancara mendalam, observasi non partisipan dan studi dokumentasi. Untuk memperoleh keabsahan data dilakukan triangulasi data. Teknik analisis data menggunakan model Milles dan Huberman. Penelitian ini dianalisis dengan teori tindakan sosial Max Weber yang menjelaskan bahwa teori tindakan sosial Max Weber berorientasi pada motif dan tujuan aktor. Dengan menggunakan teori ini kita dapat memahami perilaku setiap individu dan kelompok yang masing-masing memiliki motif dan tujuan yang berbeda dari suatu tindakan yang dilakukan. Hasil penelitian ini menunjukkan bahwa pada masa pandemi Covid-19 terdapat empat strategi penyelesaian skripsi bagi mahasiswa Departemen Sosiologi Fakultas Ilmu Sosial Universitas Negeri Padang pada masa pandemi Covid-19 yaitu pertama Memanfaatkan Teknologi dan Media Online, kedua Komunikasi dan Bimbingan Intens dengan Dosen Pembimbing, dan ketiga Pemilihan Topik Skripsi Terkait Pandemi Covid-19.

Abstract T follicular helper (Tfh) cell-derived signals promote activation and proliferation of antigen-primed B cells. It remains unclear whether epigenetic regulation is involved in the B cell responses to Tfh cell-derived signals. Here, we demonstrate that Tfh cell-mimicking signals induce the expression of histone demethylases KDM4A and KDM4C, and the concomitant global down-regulation of their substrates, H3K9me3/me2, in B cells. Depletion of KDM4A and KDM4C potentiates B cell activation and proliferation in response to Tfh cell-derived signals. ChIP-seq and de novo motif analysis reveals NF-κB p65 as a binding partner of KDM4A and KDM4C. Their co-targeting to Wdr5, a MLL complex member promoting H3K4 methylation, up-regulates cell cycle inhibitors Cdkn2c and Cdkn3. Thus, Tfh cell-derived signals trigger KDM4A/KDM4C - WDR5 - Cdkn2c/Cdkn3 cascade in vitro, an epigenetic mechanism regulating proper proliferation of activated B cells. This pathway is dysregulated in B cells from systemic lupus erythematosus patients and may represent a pathological link.

Interleukin- (IL-) 33 contributes to various inflammatory processes. IL-33/ST2 activation participates in systemic lupus erythematous via binding to the receptor of Suppression of Tumorigenicity 2 protein (ST2). However, whether IL-33/ST2 interferes with the nosogenesis of cutaneous lupus erythematosus (CLE) has not been reported so far. Herein, we proposed to disclose the impacts on IL-33/ST2 activation and Ro60 on CLE and their potential implications in the photosensitization of CLE cells. IL-33, ST2, and Ro60 in CLE patients’ skin lesions were detected. Murine keratinocytes stimulated with or without IL-33 were irradiated by ultraviolet B (UVB), and the levels of Ro60 and inflammation markers were determined. Keratinocytes were cocultured with J774.2 macrophages and stimulated with IL-33 for analysis of chemostasis. The results identified that IL-33, ST2, and downstream inflammation markers were significantly upregulated in CLE lesions with Ro60 overexpression. Additionally, IL-33 treatment promoted the upregulation of Ro60 induced by UVB treatment in murine keratinocytes. Moreover, IL-33 stimulates keratinocytes to induce macrophage migration via enhancing the generation of the chemokine (C–C motif) ligands 17 and 22. Meanwhile, the silencing of ST2 or nuclear factor-kappa B (NF-κB) suppression abolished IL-33-induced upregulation of Ro60 in keratinocytes. Similarly, the inhibition of SOX17 expression was followed by downregulation of Ro60 in keratinocytes following IL-33 stimulation. In addition, UVB irradiation upregulated SOX17 in keratinocytes. Conclusively, the IL-33/ST2 axis interferes with Ro60-regulated photosensitization via activating the NF-κB- and PI3K/Akt- and SOX17-related pathways.

Abstract T follicular helper (TFH) cells are functional subset CD4+ T cells in providing help to B cells and aberrant expansion of TFH cells is observed in systemic lupus erythematosus (SLE), which suggests a global defect in the maintenance of T and B cell tolerance. The mechanisms underlying uncontrolled TFH cells development and differentiation in SLE pathogenesis is fragmentary understand. Casitas B-lineage lymphoma (Cbl) family is an E3 ubiquitin ligase and has been implicated in SLE pathogenesis as a potent regulator in activating the immune cells and maintaining peripheral tolerance. The deficiency of cbl-b mice spontaneously develops autoimmunity and is highly susceptible to develop pathogenic auto-antibodies of autoimmune diseases. Through CRISPR KO system, we identified the deficiency of cbl/cbl-b in human naïve CD4+ T cells induces the TFH expansion in T cell stimulation with Activin A & IL12. Indeed, higher levels of chemokine (C-X-C motif) ligand 13 (CXCL13) which is constitutively produced in germinal center (GC) TFH cells were detected. Functionally, cbl/cbl-b KO TFH cells are more potent B cell activators compared to cbl/cbl-b competent TFH cells. There was increased plasmablast differentiation which secrete IgG. These data suggest that the deficiency of Cbl family in human elicits heightened T-dependent immune activation with augmentation of TFH responses, thus regulating humoral immune responses and the susceptibility to SLE in human. This project will advance our understanding of the mechanism of activation and maintenance of TFH cells in lupus and dissect possible avenues of therapeutic intervention in disease.

La dimension ludique de la fiction tient dans le fait qu’elle repose sur un acte de faire semblant. Inversement, un jeu possède une dimension fictionnelle quand son terrain représente un monde et quand les actions du joueur, au lieu de suivre des règles abstraites et conventionnelles, imitent des actions concrètes dont le but répond à des intérêts authentiquement humains. Caillois pensait que la fiction est incompatible avec les jeux de type ludus, basés sur les règles et la compétition, mais les jeux vidéo, ainsi que les diverses tentatives, dès le XVIIe siècle, d’associer le jeu de l’oie à des thèmes narratifs démentissent son opinion. La réconciliation du jeu et de l’histoire ne se réalisera toutefois pleinement que lorsque deux conditions seront remplies : 1. le joueur sera motivé par l’intérêt pris à l’histoire; 2. le joueur construira l’histoire par ses actions, et chaque fois qu’il jouera, il produira une nouvelle histoire.

One of the most abundant non-canonical bases widely occurring on various RNA molecules is 5-methyluridine (m5U). Recent studies have revealed its influences on the development of breast cancer, systemic lupus erythematosus, and the regulation of stress responses. The accurate identification of m5U sites is crucial for understanding their biological functions. We propose RNADSN, the first transfer learning deep neural network that learns common features between tRNA m5U and mRNA m5U to enhance the prediction of mRNA m5U. Without seeing the experimentally detected mRNA m5U sites, RNADSN has already outperformed the state-of-the-art method, m5UPred. Using mRNA m5U classification as an additional layer of supervision, our model achieved another distinct improvement and presented an average area under the receiver operating characteristic curve (AUC) of 0.9422 and an average precision (AP) of 0.7855. The robust performance of RNADSN was also verified by cross-technical and cross-cellular validation. The interpretation of RNADSN also revealed the sequence motif of common features. Therefore, RNADSN should be a useful tool for studying m5U modification.

La criptococosis meníngea tiene una alta prevalencia en todo el mundo y afecta particularmente a pacientes infectados por VIH con recuentos bajos de CD4 y a personas que reciben terapia inmunosupresora. Paciente de 40 años con antecedentes de Lupus eritematoso sistémico, se evidencia alteración del estado de conciencia acompañado de cefalea de 7 días de evolución, motivo por el cual acude a la casa de salud mas cercana.
