Abstract:
The hallmark of Gram-negative bacteria is their cell envelope, which is composed of two membranes, the inner or cytoplasmic membrane (IM), and the outer membrane (OM), separated by a compartment (the periplasm) that contains a thin peptidoglycan layer. Lipopolysaccharide (LPS) is the major component of the OM, and it acts as a selective barrier together with the OM proteins (OMPs), preventing the entry of many toxic molecules into the cell. Despite the structure and composition of OM have been elucidated in pivotal studies in the 50s and in the 70s, the factors required for the assembly of this organelle have only recently been identified. LPS, once it is synthesized in the cytoplasm, has to be translocated through out the cell envelope. Seven essential proteins cooperate in a unique fashion to extract the macromolecule from the IM and deliver it in the outer leaflet of the OM. LptBCFG form the IM complex that empowers the translocation process by ATP hydrolysis (Narita and Tokuda, 2009), LptDE constitute a complex embedded in the OM that finally flips LPS across the OM and deliver it to its final destination (Chng et al., 2010a; Freinkman et al., 2011), and LptA is a periplasmic protein that contacts both the IM and OM complexes (Sperandeo et al. 2007; Tran et al., 2008). Notably, LptC is single-pass IM protein with a large periplasm-protruding region. LptC single mutants were obtained in this work by random-mutagenesis, and used in vivo and in vitro experiments to characterize two regions of the protein that distinctly interact with LptA and the IM protein complex LptBFG, respectively. Chimera versions of LptC, either missing the transmembrane (TM) sequence, or with the IM anchor substituted by a heterologous sequence, were additionally constructed to this purpose. Moreover, Both LptA and LptC were previously demonstrated to bind LPS in vitro, here it is presented a rapid bioinformatic tool which has been implemented to discover the molecular determinants of LptA for the interaction with Lipid A, the main component of LPS. Genetic evidences previously obtained in our laboratory together with the presented data strongly support the LPS transport machinery model defined as the trans-envelope complex by Chng and coworkers (Chng et al., 2010a): indeed LptA interacts both with the IM and the OM protein complexes (LptBCFG and LptDE respectively), bridging them together. In support of this model, a phylogeny and structural motif conservation analysis of the Lpt components suggests that the unique structural domain retained in these proteins—despite the low sequence similarity—is the key to make possible the interaction between all the Lpt components.

Abstract:
Bakgrund: Patienters delaktighet i sin vård ses idag som en självklarhet i Sverige och personcentrerad vård (PCV) är ett verktyg för att öka delaktigheten. Genom att lyssna till patientens berättelse, sträva efter ett jämbördigt partnerskap och dokumentera det som framkommer får sjuksköterskan möjlighet att med patienten skapa en hållbar och långsiktig effekt av vården. Inom tvångsvården enligt Lagen om psykiatrisk tvångsvård (LPT) ges sjukvården rätten och skyldigheten att vårda patienten mot deras vilja när det anses vara befogat för att skydda patientens liv och hälsa. Enligt LPT är syftet med tvångsvården att förmå patienten till att ta emot vården som erbjuds frivilligt.   Syfte: Syftet med denna studie är att undersöka erfarenheter hos sjuksköterskor i psykiatrisk vård av att arbeta med PCV med patienter som vårdas enligt LPT.  Metod: Studien har genomförts med en kvalitativ semistrukturerad intervju. Detta har sedan analyserats med en kvalitativ innehållsanalys.  Resultat: Studiens resultat uppvisar erfarenheter av att främja patientberättelsen men även hinder för denna. Det framkommer även erfarenheter av partnerskap som bedrivs under LPT. Partnerskapet främjas till stor del av sjuksköterskans bemötande. Till sist framkommer erfarenheter av dokumentationens betydelse för PCV som bedrivs enligt LPT. Slutsats: Utifrån den fakta som läggs fram rörande PCV och LPT i bakgrunden är dessa två begrepp relativt komplicerade att förena. I resultatet av denna studie kan det konstateras att delaktighet kan främjas PVC i vård som bedrivs enligt LPT, som i sin tur främja delaktighet. Förhållandet mellan begreppen PCV och delaktighet blir i det närmsta cirkulärt där de stödjer varandra och skapar uppbyggande symbios Kliniska implikationer: Baserat på resultatet framkommer en tilltro till att PCV kan vara positiv i bemärkelsen att det skapas ökad delaktighet mellan patient och sjuksköterska i vård som bedrivs enligt LPT.
Background: Patients' participation in their care is today seen as a matter of course in Sweden and person-centred care (PCV) is a tool to increase participation. By listening to the patient's story, striving for an equal partnership and documenting what emerges, the nurse has the opportunity to create a sustainable and long-term effect of care with the patient. In coercive care under the Compulsory Psychiatric Care Act (LPT), healthcare is given the right and obligation to care for the patient against their will when this is considered justified to protect the patient's life and health. According to LPT, the purpose of coercive care is to induce the patient to receive the care offered voluntarily.   Aim: The aim of this study is to investigate the experiences of nurses in psychiatric care of working with PCV with patients being cared for according to LPT. Method: The study has been conducted with a qualitative semi-structured interview. This has then been analyzed with a qualitative content analysis. Results: The results of the study show experiences in promoting the patient's story but also obstacles to it. There is also experience of partnerships conducted under LPT. This partnership is largely promoted by the nurse's treatment. Finally, experience of the importance of documentation for PCVs carried out under LPT emerges. Conclusion: Based on the facts presented regarding PCV and LPT in the background, these two concepts are relatively complex to reconcile. In the results of this study, it can be concluded that participation can be promoted PVC in care conducted according to the LPT, which in turn promotes participation. The relationship between the concepts of PCV and participation becomes almost circular where they support each other and create constructive symbiosis. Clinical implications: Based on the results, there is a belief that PCV can be positive in the sense that increased participation between patient and nurse is created in care conducted according to LPT.

Abstract:
Modern turbomachinery design is characterized by a tendency towards thinner, lighter and highly loaded blades, which in turn gives rise to increased sensitivity to flow induced vibration such as flutter. Flutter is a self-excited and self-sustained instability phenomenon that may lead to structural failure due to High Cycle Fatigue (HCF) or material overload. In order to be able to predict potential flutter situations, it is necessary to accurately assess the unsteady aerodynamics during flutter and to understand the physics behind its driving mechanisms. Current numerical tools used for predicting unsteady aerodynamics of vibrating turbomachinery components are capable of modeling the flow field at high level of detail, but may fail in predicting the correct unsteady aerodynamics under certain conditions. Continuous validation of numerical models against experimental data therefore plays significant role in improving the prediction accuracy and reliability of the models.   In flutter investigations, it is common to consider aerodynamically symmetric (tuned) setups. Due to manufacturing tolerances, assembly inaccuracies as well as in-service wear, the aerodynamic properties in a blade row may become asymmetric. Such asymmetries can be observed both in terms of steady as well as unsteady aerodynamic properties, and it is of great interest to understand the effects this may have on the aeroelastic stability of the system.   Under certain conditions vibratory modes of realistic blade profiles tend to be coupled i.e. the contents of a given mode of vibration include displacements perpendicular and parallel to the chord as well as torsion of the profile. Current design trends for compressor blades that are resulting in low aspect ratio blades potentially reduce the frequency spacing between certain modes (i.e. 2F & 1T). Combined modes are also likely to occur in case of the vibration of a bladed disk with a comparatively soft disk and rigid blades or due to tying blades together in sectors (e.g. in turbines).   The present investigation focuses on two areas that are of importance for improving the understanding of aeroelastic behavior of oscillating blade rows. Firstly, aeroelastic properties of combined mode shapes in an oscillating Low Pressure Turbine (LPT) cascade were studied and validity of the mode superposition principle was assessed. Secondly, the effects of aerodynamic mistuning on the aeroelastic properties of the cascade were addressed. The aerodynamic mistuning considered here is caused by blade-to-blade stagger angle variations   The work has been carried out as compound experimental and numerical investigation, where numerical results are validated against test data. On the experimental side a test facility comprising an annular sector of seven free-standing LPT blades is used. The aeroelastic response phenomena were studied in the influence coefficient domain where one of the blades is made to oscillate in three-dimensional pure or combined modes, while the unsteady blade surface pressure is acquired on the oscillating blade itself and on the non-oscillating neighbor blades. On the numerical side, a series of numerical simulations were carried out using a commercial CFD code on a full-scale time-marching 3D viscous model. In accordance with the experimental part the simulations are performed using the influence coefficient approach, with only one blade oscillating.   The results of combined modes studies suggest the validity of combining the aeroelastic properties of two modes over the investigated range of operating parameters. Quality parameters, indicating differences in mean absolute and imaginary values of the unsteady response between combined mode data and superposed data, feature values that are well below measurement accuracy of the setup.   The findings of aerodynamic mistuning investigations indicate that the effect of de-staggering a single blade on steady aerodynamics in the cascade seem to be predominantly an effect of the change in passage throat. The changes in steady aerodynamics are thereby observed on the unsteady aerodynamics where distinctive effects on flow velocity lead to changes in the local unsteady pressure coefficients. In order to assess the overall aeroelastic stability of a randomly mistuned blade row, a Reduced Order Model (ROM) model is introduced, allowing for probabilistic analyses. From the analyses, an effect of destabilization due to aero-asymmetries was observed. However the observed effect was of moderate magnitude.

På hösten 2018 riktades blickarna mot vården i Sverige. Inte vilken del av vården som helst utan den som berör två av de kanske mest utsatta grupperna i samhället, missbruksvården och psykiatrin. I kölvattnet efter Uppdraggransknings dokumentärer om Sanne beslutade regeringen att det behövdes en (ny) översyn av huvudmannaskapet vid samsjuklighet av missbruk och psykisk sjukdom. (Ny) är denna översyn eftersom Missbruksutredningen 2011 lämnade över sitt slutbetänkande ”Bättre insatser vid missbruk och beroende” som pga. det höga antalet av samsjuklighet yrkade på ett lagförslag där Lag om vård av missbrukare i vissa fall (LVM) och Lag om psykiatrisk tvångsvård (LPT) skulle bli en och samma lag. Regeringen avslog det yrkandet. Hur upplever läkarna, som möter dessa patienter dagligen, att det är att ha två huvudmän för den här patientgruppen? Uppsatsens syfte är att ta reda på hur specialistläkarna inom psykiatri och beroendevården på Vrinnevisjukhuset i Norrköping uppfattar gråzonen mellan LVM och LPT. Detta har gjorts genom intervjuer med läkare som arbetar på Beroendekliniken och på psykiatriska kliniken på Vrinnevisjukhuset i Norrköping. Uppfattningen bland läkarna är att det finns ett behov av ett bättre samarbete mellan de olika instanserna och att strukturerna för hur man ska ha detta samarbete måste vara tydligt. Läkarna anser att lagarna bör fortsatt vara åtskilda men att det kunde vara önskvärt med en och samma huvudman för att minska risken för att patienter hamnar mellan stolarna.

Abstract:
The present work deals with post-processing by means of Proper Orthogonal Decomposition (POD) of data obtained from experimental investigations on Low-Pressure Turbine (LPT) cascades. The aim is to provide a strategy useful to coarsening design space grids and hence, reduce the data acquisition time during experimental investigations or high-fidelity simulations. The experimental work has been conducted in a wind tunnel installed at the Laboratory of Aerodynamics and Turbomachinery of the University of Genova. The experimental procedure consists in surveying the flow upstream and downstream of the cascade, aiming to evaluate the total pressure drop through the cascade, and hence the overall losses. Specifically, effects on losses of design parameters such as freestream turbulence intensity, vane solidity, incoming wake parameters (velocity defect, turbulent content and reduced frequency), flow Reynolds number and incidence flow angle have been evaluated. Then, POD has been applied to data constituted by total pressure loss coefficient distributions in the pitchwise direction under different conditions in order to: • highlight the loss trend vs the design space parameter variation • identify the minimum number of requested conditions in the design space, i.e., the number of independent conditions useful for the education of a predictive model These objectives have been achieved due to the POD capability in providing compact subspaces able to reproduce the same loss trend observed in the physical space. Moreover, in the POD subspace it has been possible to define a smooth kernel with the number of independent conditions in order to estimate the loss distribution of the remaining conditions. Hence, a model has been educated by means of Least Squares Method (LSM) on the POD design space coefficients choosing the predictors as low order polynomials of the independent variables. Results will show good agreement between measured and modelled loss distribution varying each design space parameter, showing that the model is able to predict loss distribution within the uncertainty measurement. Furthermore, keeping the same strategy for the education of the kernel on another dataset characterizing the loss distribution of two cascades with different geometrical parameters, loss distribution and magnitude are still well captured. Thus, analysis show that the rank of the problem is much lower than the tested conditions, and consequently a reduced number of tests are really necessary. This technique could be useful to reduce the number of scale-resolving simulations or detailed experiments without significant loss of accuracy.

Läkemedelshantering såsom administrering av medicin till patienter är en av de dagliga omvårdnadshandlingarna för en sjuksköterska. För en patient som vårdas under Lagen om Psykiatrisk Tvångsvård (LPT) (SFS, 1991:1128) kan upplevelser av att få medicin vara blandade. I sjuksköterskans omvårdnadsarbete är det viktigt att se patientens unika upplevelser av situationen. Syftet med denna studie är att belysa patienters upplevelser av läkemedelsadministrering då de vårdas under LPT. Metoden är av kvalitativ natur där empirin samlats in med hjälp av intervjuer som har analyserats med manifest innehållsanalys. Resultatet visar blandade upplevelser hos patienter då Acceptans och Välbefinnande blandat med Fruktan, Resignation, Utsatthet, Ovilja och Brist på information framkommit. Av resultatet drogs Slutsatsen att kommunikation mellan patient och sjuksköterska är viktig samt att Brist på kommunikation kan leda till lidande för patienten.

Abstract:
This article is about the application of the two coercive legislations, Care of Abusers (Special Provisions) Act (LVM) and  the Compulsory Psychiatric Care Act (LPT) when individuals suffer from comorbidity. The authority who files the application for coercive interventions differs between the legislations. In LVM the social welfare board files the application and in LPT the chief psychiatrist.  Earlier research has shown that the individuals of the comorbidity group do not get their needs met in either of the legislations. It has also occurred that concrete arguments which relate to statutory criteria are missing. Our aim was to gain knowledge about how the court argued for these criteria, especially in cases where comorbidity exist. Another aim was to see which legislation that gave legal impact. Finally, we wanted to examine the roles of the chief psychiatrist and the social welfare board in court negotiations. We decided to study twenty court cases from Administrative court, ten cases from LVM and ten from LPT where the same individual had been committed to coercive care according to both legislations, this to capture the comorbidity. We interpreted the documents using discourse analysis and selected two concepts, inspired by Foucault and Fairclough, that we thought could be fruitful for our analysis. The result shows that there is more often argumentation about if the legal criteria in LVM are fulfilled compared to LPT where the argumentation is less informative. This result may have been influenced by the secrecy of Law. We also found out that the legislation that was given legal impact depended on the authorities assess of which need that was most urgent. The last finding was that the chief psychiatrist`s opinion was never questioned by the court, but the statements made by the social welfare board was.

Abstract:
Il rivestimento dei batteri Gram negativi consiste in una membrana interna (IM) e una membrana esterna (OM) separate da uno spazio periplasmatico contenente un sottile strato di peptidoglicano (PG) ancorato alla OM tramite la lipoproteina di Braun (LPP). Mentre la IM è costituita da un doppio strato di fosfolipidi, la OM è una membrana lipidica asimmetrica, contenente fosfolipidi nel foglietto interno, e un lipide complesso il Lipopolisaccaride (LPS) nel foglietto esterno. Il LPS sintetizzato nel citoplasma, viene traslocato sul lato periplasmatico della IM e preso in carico dal complesso multiproteico Lpt (LPS transport), composto in Escherichia coli da sette proteine essenziali (Lpt ABCDEFG) che si occupano del suo trasporto fino al raggiungimento della sua sede finale, la OM. Analisi biochimiche hanno dimostrato che le sette proteine Lpt formano un complesso “transenvelope” che connette IM e OM e studi di tipo genetico suggeriscono che esse operino in concerto come un singolo macchinario. Infatti, la deplezione di un qualsiasi componente del complesso Lpt causa lo stesso fenotipo, ovvero l’accumulo del LPS nel versante periplasmatico della IM, la decorazione del LPS con acido colanico e la formazione di una IM anomala, con una densità intermedia tra la IM e la OM. Nel nostro laboratorio è stata condotta l’analisi differenziale del proteoma delle membrane totali di E. coli in deplezione di LptC, per studiare la risposta globale al blocco del trasporto del LPS. Tra le proteine il cui livello cambia nel confronto tra il ceppo non depleto e il depleto sono state trovate proteine coinvolte nella biogenesi e nel rimodellamento del PG. Lo scopo di questa tesi è stato lo studio della correlazione tra il blocco del LPS e il rimodellamento del PG. Inizialmente è stata analizzata la struttura del PG in deplezione di LptC. Questa analisi ha evidenziato che in questa condizione la struttura del PG varia sia per composizione che per tipo di legami crociati tra i filamenti glicanici adiacenti. Nei batteri Gram negativi il legame tra i filamenti glicanici è generalmente un legame diretto 3-4, che si forma tra il gruppo aminico del diaminoacido in posizione 3 di un tetrapeptide e il gruppo carbossilico della D-alanina in posizione 4 del tretrapeptide adiacente. Il legame 3-4 avviene ad opera delle D,D transpeptidasi PBP. Un altro tipo di legame crociato presente nel PG è quello tipo 3-3 che si forma tra il gruppo aminico del diaminoacido in posizione 3 ed il gruppo carbossilico del diaminoacido presente nel tetrapeptide del filamento glicanico adiacente ed è catalizzato da L,D-transpeptidasi . In E. coli, sono noti cinque enzimi con attività L,D-transpeptidasica, di cui tre (LdtA, LdtB, LdtC) ancorano la lipoproteina più abbondante della OM (lipoproteina di Braun) al PG e due (LdtD, LdtE) catalizzano il legame crociato 3-3. La delezione di tutti questi geni, singolarmente o in combinazione, non presenta nessun fenotipo, suggerendo che in condizioni normali questo legame è dispensabile. Per studiare la correlazione tra il legame crociato 3-3 e il blocco del trasporto del LPS abbiamo creato mutanti arabinosio dipendenti per alcuni dei componenti del sistema Lpt deleti contemporaneamente per i geni che esprimono le L,D-transpeptidasi LdtD e LdtE. In precedenza, nel nostro laboratorio è stato dimostrato che la deplezione di uno qualsiasi dei geni lpt causa la formazione di cellule filamentose e l’arresto della crescita ma non la lisi cellulare. Invece, nei mutanti ΔldtDΔldtE, in deplezione dei geni lpt, oltre alla formazione di cellule filamentose si osserva la formazione di un setto anomalo e la lisi cellulare. Questi dati suggeriscono che il rimodellamento del peptidoglicano a seguito della formazione di legami 3-3 potrebbe essere una forma di riposta al danno alla membrana esterna.
The cell envelope of Gram-negative bacteria is a complex multi-layered structure consisting of a cytoplasmic and an outer membrane (CM and OM), which delimit the periplasm containing a thin layer of peptidoglycan (PG) called the sacculus. The primary function of the OM is to establish a selective permeability barrier that enables the cell to maintain favourable intracellular conditions even in hash environments and the lipopolysaccharide (LPS) layer greatly contributes to this peculiar property. The integrity of the PG mesh is essential to protect the cell from bursting due to its turgor and maintain the shape of the cell. OM and PG are synthetized and assembled by multiprotein machineries that need to be finely coordinated as imbalanced growth of these layers may compromise structural integrity of the cell. In order to gain more insight in the mechanism by which the cells coordinate the growth of these two layers, we analysed the PG composition when the biogenesis of OM is compromised due to the block of LPS transport. In this work we shown that when OM is impaired, E. coli cells remodel PG architecture by increasing the non-canonical 3- 3 cross-linkage. We can assume that this is a salvage mechanism to prevent cell lysis when OM is damaged.

El presente Proyecto Profesional consiste en un plan estratégico desarrollado para la empresa comercializadora de cierres, Importaciones LPT ZIPPER SAC, para los años 2017- 2022. Para ello, se ha seguido el método propuesto por el reconocido doctor Fernando D´Alessio, director de la escuela de negocios Centrum, de la Pontificia Universidad Católica del Perú, en su libro: ―El proceso estratégico, un enfoque de gerencia‖1. La elaboración de un plan estratégico requiere de un estudio profundo de la industria a la que pertenece a la empresa. En el caso de LPT ZIPPER SAC, el análisis externo de la empresa, ha arrojado información favorable, como por ejemplo, el pronóstico de recuperación y crecimiento del sector para un futuro próximo. Pero también, ha puesto en evidencia las amenazas propias de la industria, como lo son una alta rivalidad de competidores y la elevada presencia de productos sustitutos o de nuevos competidores, además del gran poder de negociación de los clientes y proveedores del rubro. Asimismo, es necesario un análisis interno para identificar las fortalezas y debilidades de LPT ZIPPER. Una fortaleza a destacar es la alta calidad de sus productos; sin embargo, al ser débil para aprovechar el entorno y afrontar las amenazas, no responde correctamente a las exigencias del mercado. Visto ya el contexto de LPT ZIPPER SAC, se definieron la visión, misión, valores y código de ética de la empresa, debido a que no las tenía formalmente establecidas. Se ha trazado como visión para el año 2022, que LPT ZIPPER SAC se posicione como una de las empresas peruanas más reconocidas en la venta de productos textiles y asesoría personalizada. La empresa estará respaldada por la calidad de sus cierres y por su conocimiento de las últimas tendencias de la moda. Para alcanzar esta meta institucional, se han formulado ocho objetivos de largo plazo y siete intereses organizacionales. En base a ellos, se formularon veinte estrategias, de las cuales se han seleccionado dieciséis, y el resto quedaron como estrategias de contingencia. Para lograr los objetivos de largo plazo se han establecido cuarenta y seis objetivos a corto plazo, a los cuales se les ha asignado los recursos necesarios y un presupuesto para el periodo especificado. Finalmente, una adecuada implementación de un plan estratégico, requiere de una herramienta de control que evalúe la consecución de los objetivos: el Balance Scorecard. Cada objetivo a corto plazo cuenta con indicadores variados, así como proyecciones de crecimiento hasta el año 2022. Como conclusión, se reconoce que este plan estratégico plantea nuevos retos para LPT ZIPPER SAC. Uno de los más importantes, y que ya ha empezado a llevarse a cabo, es el de una reestructuración organizacional que cree una subdivisión de la empresa en dos unidades de negocio: mayoristas y minoristas. Esta y otras medidas desarrolladas en este trabajo, contribuirán a que Importaciones LPT ZIPPER logre alcanzar la visión y objetivos trazados, consiguiendo no sólo una supervivencia sostenible, rentable y atractiva para sus dueños, sino también, poseer un potencial sólido que le permita ingresar a nuevos mercados.
The present Professional Project consists of a strategic plan developed for the company marketer of zippers, LPT ZIPPER SAC Imports, for the years 2017-2022. In order to do this, the method proposed by the renowned Dr. Fernando D'Alessio, director of the Centrum business school, of the Pontifical Catholic University of Peru, in his book "The strategic process, a management approach", has been followed. The elaboration of a strategic plan requires an in-depth study of the industry to which the company belongs. In the case of LPT ZIPPER SAC, the external and internal analysis of the company, has provided favorable information, such as the forecast of recovery and growth of the sector in the near future; but it has also exposed the threats of the industry, such as a high rivalry of competitors and the high presence of substitute products or new competitors, in addition to the great bargaining power of customers and suppliers of the item Likewise, an internal analysis is required to identify the strengths and weaknesses of LPT ZIPPER. A strength to be highlighted is the high quality of its products; however, being weak to take advantage of the environment and face the threats, does not respond correctly to the demands of the market. Having already seen the context of LPT ZIPPER SAC, the vision, mission, values and code of ethics of the company were defined, because they were not formally established. It has been outlined as vision for the year 2022, that LPT ZIPPER SAC be positioned within the first 5 companies most recognized in the sale of zippers of great quality and diversity of models. To achieve this institutional goal, eight long-term objectives and seven organizational interests have been formulated. Based on them, twenty strategies were formulated, of which sixteen were chosen, and the rest remained as contingency strategies. To achieve the long-term objectives, forty-six short-term objectives have been established, which have been allocated the necessary resources and a budget for the specified period. Finally, a proper implementation of a strategic plan requires a control tool that evaluates the achievement of the objectives: the Scorecard Balance. Each short-term objective has varied indicators, as well as growth projections up to 2022. As a conclusion, it is recognized that this strategic plan poses new challenges for LPT ZIPPER SAC. One of the most important, and that has already begun to take place, is an organizational restructuring that creates a subdivision of the company into two business units: wholesalers and retailers. This and other measures developed in this work will contribute to LPT ZIPPER Imports achieve the vision objectives outlined, achieving not only a sustainable, profitable and attractive survival for its owners, but also, to possess a solid potential that allows him to enter new markets.

I förarbeten till lagarna LVM och LPT framkommer det att det finns en problematik kring vården av personer som lider av både missbruk och psykisk ohälsa. Det framgår att samarbetet mellan psykiatri och socialtjänst i vissa avseende är att anse som bristfälligt och att brukarna faller mellan stolarna. Syftet med studien har varit att, genom granskning av rättskällorna, ta reda på och tydliggöra vad som ska ses som gällande rätt, för om en person med både missbruk och psykisk ohälsa ska få vård enligt LVM eller LPT. Metoden som har använts är en rättsvetenskaplig metod där fokus legat på granskning av rättskällorna. En rättsfallsstudie har även gjorts för att bredda bilden av hur rekvisiten för lagstiftningarna tillämpas. Resultatet har analyserats och redovisats utifrån den rättsdogmatiska metoden. Rättsfallen har tematiserats och redovisats i diagram. Studien visar att det finns gällande rätt kring ämnet, men att gällande rätt varierar beroende på personens situation. Huvuddraget i resultatet är att LPT vid allvarlig psykisk ohälsa bör gå före LVM. Vidare framkommer det att det inte finns något hinder, för att under en kortare period, vårdas enligt båda lagstiftningarna samtidigt.
In the preparatory works to the laws of LVM and LPT it has been pointed out that there is a problem regarding the care of persons that both suffer from an addiction as well as mental illness. It appears that the cooperation between psychiatry and social services is deficient and that the users are falling through the cracks. The purpose of the study has been, through review of the sources of law, to find out and define what is to be considered a correct, application of the law for a person that suffers from an addiction as well as a mental illness. Is it to be admitted to compulsory care and treatment according LVM or LPT? The method and material used for this purpose is the sources of law. To broaden the picture a study of court cases has also been made, regarding how the legislation is applied by a lower administrative court. The results has been analyzed and reported through the legal method. Court cases has been thematized and presented in different diagrams. The study shows that the outcome of the law varies depending on the person’s situation. The main feature of the result is that if someone is suffering from serious mental illness, LPT should be chosen instead of LVM. Further it is possible, during a shorter time, to be treated according to both LVM and LPT simultaneously.

The outer membrane (OM) of Gram-negative bacteria is an asymmetric bilayer formed by phospholipids in the inner leaflet and lipopolysaccharide (LPS) in the outer leaflet, with a large number of embedded or associated proteins. The primary function of this structure essential for Gram-negative viability is to establish an additional selective permeability barrier that enables the cell to maintain favourable intracellular conditions even in harsh environments and the LPS layer greatly contributes to this peculiar property. The transport of LPS to the cell surface is an essential process for OM biogenesis; the LPS transport (Lpt) system, originally identified in E. coli, is the protein machine responsible for LPS delivery from the periplasmic side of the inner membrane (IM) to the OM. It is composed of seven proteins forming a complex which spans from IM to OM. At the IM the ABC transporter LptBFG, associated to the membrane-bound protein LptC interacts with the periplasmic protein LptA that connects, through structurally conserved domains, the IM ABC transporter with the OM translocon LptDE, responsible for LPS assembly at the cell surface. In order to gain more insight in the mechanism of LPS transport and more in general in OM homeostasis we used both a genetic and a proteomic approach. The former was based on the selection of suppressors of LPS transport defects obtained with two different types of mutants. i) a quadruple non-lethal lptA mutant (lptA41) that displayed increased sensitivity to toxic compounds, and ii) a lethal deletion mutant of lptC. Genome sequencing analysis of spontaneous suppressors of lptA41 phenotype revealed two different mechanism of suppression: one mechanism involves the Mla system, a protein machinery which contributes to maintain OM asymmetry; the second mechanism involves both an intragenic mutation improving LptA41 protein stability and an extragenic mutation affecting osmoregulated periplasmic glucans (OPGs) synthesis. Viable mutants lacking lptC were obtained using a plasmid shuffling technique. Genome sequencing of such mutants revealed single amino acid substitutions at the R212 residue of the IM component LptF (lptFSupmutants). Our results suggest that LptC may serve as a chaperon of the Lpt machine assembly and/or activity rather than an essential structural component and the periplasmic domain of LptF might be implicated in the formation of the Lpt bridge. The latter approach consisted of the analysis of differential envelope proteins content of an E. coli lptC conditional expression mutant upon depletion of LptC and thus impairment of LPS transport. By two-dimensional chromatography coupled to tandem mass spectrometry (Multidimensional Protein Identification Technology, MudPIT) we identified 123 proteins whose level is significantly modulated upon LptC depletion. Most of these proteins belong to pathways that contribute to repair OM and restore its permeability barrier properties, including protein involved in maintaining OM asymmetry, in the synthesis of phospholipids and exopolysaccharides as substrate for lipid A modification enzymes, and in peptidoglycan synthesis/remodelling. Overall these data contribute to our understanding of the multiple strategies that E. coli cells may adopt to respond to perturbations of the OM permeability barrier and to restore OM functionality.

Idag råder synsättet att individens delaktighet är att föredra i så gott som alla behandlings- och insatsformer samt att relationen mellan den professionelle och den som tar emot hjälpen är av stor vikt. Tvångsvård och tvångsåtgärder är något som strider mot den allmänna synen på hur man bäst tar hand om människor. I situationer där någon kan vara till fara för sig själv eller andra och inte inser detta kan dock tvång vara nödvändigt. Hur är det möjligt att göra någon delaktig i en behandling som sker mot dennes vilja? Syftet med denna uppsats var att undersöka hur personal som utövar olika former av tvångsvård går tillväga för att skapa en god behandlingsrelation och för att få klienten delaktig i behandlingen. Sju kvalitativa intervjuer med åtta respondenter genomfördes på en psykiatrisk klinik i Norrland. Resultatet visar att personalen utifrån de förhållanden som råder kan göra patienterna delaktiga genom att samtala med dem och bemöta dem med respekt och ömsesidighet. Om delaktighet definieras som en känsla hos individen snarare än att delta aktivt i saker kan delaktighet under tvång utrycka sig genom att den som blir utsatt för tvånget ändå känner sig sedd, respekterad och likvärdig.

Bakgrund: Cirka 12 000 patienter vårdas årligen enligt Lagen om psykiatrisk tvångsvård (LPT) i Sverige. Könsfördelningen är jämnt fördelad men unga kvinnor är den grupp som står för flest antal fastspänningar. Att vårda patienter under tvång ställer krav på vården både i  kvalité och i speciellt hänsyn till patientens autonomi. Studier visar att patienter som vårdas under tvång ofta är missförstådda av vårdpersonalen och att patienters erfarenhet av denna typ av omvårdnad främst är av negativ aspekt.   Syfte: Patienters upplevelser av att ha vårdats under lagen om psykiatrisk tvångsvård. Metod: Kvalitativ litteraturstudie med bloggar som litteraturkälla och en innehållsanalys med induktiv ansats. Resultat: Resultatet presenteras i fyra kategorier. Upplevelse av att vara inlåst, omfattar patienternas upplevelser av att sitta inlåst på vuxenpsykiatrin och de känslomässiga konsekvenserna av det. Upplevelse att inte bli hörd, sammanfattar patienternas upplevelser av kommunikationen mellan patient och vårdpersonal. Fråntagen beslutsrätten, handlar om hur patienter som vårdats under LPT upplevde att deras rätt till självbestämmande vid flera tillfällen blev fråntagen och Betydelse av omhändertagande som omfattar patienternas förståelse för lagen om psykiatrisk tvångsvård.  Slutsats: Studien kan bidra till en ökad förståelse för patienters upplevelser av att ha vårdats under LPT. Den kan även bidra till att förbättra sjuksköterskors insikt i hur behandling påverkar patienterna för att bättre kunna möta deras behov. Patienter som vårdas under LPT skall kunna uppleva delaktighet i sin egen vård och kunna påverka sin egen behandling. Tidvattenmodellen kan här ge möjligheter att öka vårdpersonalens engagemang till att bättre kunna förstå patienten och att utveckla en individualiserad vård

Abstract:
The Standard Penetration Test (SPT) is the most widely used in-situ soil test in the world. "Large Penetration Test" (LPT) is a term used to describe any scaled up version of the SPT. Several types of LPT have been developed around the world for the purpose of characterizing gravel deposits, as SPT blow counts are less reliable in gravels than in sands. Both tests suffer from the lack of a reliable means of determining transferred energy. Further, the use of LPT blow counts is generally limited to calculation of equivalent SPT blow counts using correlation factors measured in sands. Variation of LPT blow counts with grain size is assumed to be negligible. This research shows that safety hammer energies can be reliably estimated from measurements of hammer impact velocity for both SPT and LPT. This approach to determining transferred energy is relatively simple, and avoids the primary limitation of existing methods, which is the inability to calibrate the instrumentation. Transferred energies and hammer impact velocities are collected from various sources. These data are used to determine the ratio between the hammer kinetic energy and the transferred energy (energy transfer ratio, ETR), which is found to follow a roughly Normal distribution for the various hammers represented. An assessment of uncertainty is used to demonstrate that an ETR based approach could be superior to existing energy measurement methods. SPT grain size effects have primarily been characterized as the variation of an empirical relative density correlation factor, (CD)SPT, with mean grain size. In this thesis, equivalent (CD)LPT data are back-calculated from measured SPT-LPT correlation factors (CS/L). Results of a numerical study suggest that SPT and LPT grain size effects should be similar and related to the ratio of the sample size to the mean grain size. Based on this observation, trend-lines with the same shape as the (CD)SPT trend-line are established for the back-calculated (CD)LPT data. A method for generating the grain size effect trend-line for LPT is then proposed. These trend lines provide a rational approach to direct interpretation of LPT data, or to improved prediction of equivalent SPT blow counts.

Tvångsbältning är ett av de äldsta problemen inom institutionaliserad psykiatri och är än idag en ifrågasatt metod. Sverige är ett av dem länder som använder sig av tvångsbältning i hög utsträckning. Metoden kritiseras för att vara tortyrliknande och patienter upplever den som ett övergrepp. Syftet med denna studie är därför att undersöka hur individer som utsätts för metoden har påverkats ur ett psykosocialt perspektiv. Studiens kunskapsbas vilar på tidigare forskning inom området och kvalitativa intervjuer. I analysen av materialet kan intervjupersonernas olika förståelse och inställning inför tvångsbältning som metod urskiljas. I resultatet visar det sig dock att tvångbältning alltid fick negativa konsekvenser för undersökningspersonerna. Oavsett förståelse för metoden, hade undersökningspersonerna alltid ett behov av att bearbeta tvångsbältningen via samtalsstöd. Resultatet pekar även på bristerna och det stora tolkningsutrymmet i Lagen om psykiatrisk tvångsvård [LPT] och hur det kan ha påverkat undersökningspersonerna. Slutsatser som dras är att undersöknings personernas inställning till tvångsbältning påverkas av personalens bemötande både före, under och efter en tvångsbältning. Bemötandet är också a och o för att tvångsbältningen ska ge så lindriga konsekvenser som möjligt i efterhand för undersökningspersonerna. Det sociala livet påverkades också enligt intervjupersonerna. Främst handlade det om att dem upplevt sig stigmatiserade på grund av deras psykiska ohälsa och att de utsatts för tvångsbältning.

Abstract:
This thesis presents the design method and development of a high performance Low Pressure Turbine (LPT) for turbocompounding applications to be used in a 1.0L "cost-effective, ultra-efficient heavily downsized gasoline engine for a small and large segment passenger car". Under this assumption, the LPT was designed to recover the latent energy of discharged exhaust gases at low pressure ratios (1.05 - 1.3) and to drive a small electric generator with a maximum power output of 1.0 kW. The design speed was fixed at 50,000 rpm with a pressure ratio, PR of 1.1. Commercially available turbines are not suitable for this purpose due to the very low efficiencies experienced when operating in these pressure ratio ranges. A bespoke mean-line model was developed to evaluate the turbine performance and to generate a preliminary LPT design. Prior to the design work, the mean-line model was validated against an existing turbine. A good agreement between the predicted turbine performance and the test result was found and a minimum Relative Standard Deviation value of 1.62% was achieved. By fixing all the LPT requirements, the turbine loss model was combined with the geometrical model to calculate preliminary LPT geometry. The LPT features a mixed-flow turbine with a cone angle of 40˚ and 9 blades, with an inlet blade angle at radius mean square of +20˚. The exit-to-inlet area ratio value is approximately 0.372 which is outside of the conventional range indicating the novelty of the approach. A single passage Computational Fluid Dynamics (CFD) model was applied to optimize the preliminary LPT design by changing the inlet absolute angle. The investigation found the optimal inlet absolute angle was 77˚ and this was used to design the volute. Turbine off-design performance was then predicted from the mean-line model, single passage CFD and full turbine CFD model. The full turbine CFD and a refined single passage CFD were modelled to analyze the turbine flow field in the volute and rotor passage. A rapid prototype of the LPT was manufactured and tested in Imperial College turbocharger testing facility under steady-state and pulsating flow. The steady-state testing was conducted over speed parameter ranges from 1206 rpm/K0.5 to 1809 rpm/K0.5. The test results showed a typical flow capacity trend as a conventional radial turbine but the LPT had higher total-to-static efficiency, nt-s in the lower pressure ratio regions. A maximum total-to-static efficiency, nt-s of 0.758 at pressure ratio, PR≈1.103 was found, no available turbines exist in this range as parameters. A validation of the predicted off-design performance against the LPT test result found a minimum total-to-static efficiency Standard Deviation of ±0.019 points for mean-line model at 1206 rpm/K0.5 and the full turbine CFD model showed a minimum Mass Flow Parameter Standard Deviation of ±0.09 kg/s.K0.5/bar also at 1206 rpm/K0.5. The pulsating flow testing was carried out at LPT power of 1.0 kW for a pulsating frequency range of 20 Hz to 80 Hz over the turbine speed parameter range between 1206 rpm/K0.5 and 1809 rpm/K0.5. A hysteresis turbine performance encapsulated the steady state turbine map due to a ‘filling and emptying’ was shown for all frequencies. The pulse pressure amplitude and the chopper plate pulse frequency are found to have influenced the measured unsteadiness characterization. Strouhal number, St.* and [Lambda criterion symbol appears here] lambda criterion which commonly used in quasi-steady analysis were used to quantified the unsteadiness level. The LPT was implemented into a validated 1-D engine model to investigate the impact on the Brake Specific Fuel Consumption and Brake Mean Effective Pressure. The validated 1-D engine model was run at full load at three LPT locations: waste-gated, pre-catalyst and post catalyst. The study found the optimum location was at post catalyst and a maximum BSFC reduction of 2.6% can be achieved. The part-load 1-D engine model found that an installation of the turbocompounding unit increased the Pumping Mean Effective Pressure hence; it increased the Brake Specific Fuel Consumption up to 0.72%. However, as soon as the LPT power was reused into the engine, a maximum reduction of BSFC approximately 2.6 % can be achieved. Engine testing to analyze the effect of the LPT was carried out at Ricardo UK ltd, Shoreham Technical Centre. The LPT was installed at exhaust post catalyst of a heavily downsized 1.0L gasoline engine. A commercial compressor Garret GT28RS was attached to load the LPT. The results showed a maximum Brake Specific Fuel Consumption reduction of 2.6 % was achieved at an engine speed of 2500 rpm during the part load condition. Finally, the engine model results were compared against the engine testing. A comparison between the engine testing and 1-D engine model showed a good agreement at part load condition with a minimum Brake Specific Fuel Consumption Standard Deviation of 0.0238 at engine speed of 3000 rpm.

Rättsväsendet i västvärlden har sedan antiken gett dessa förbrytare en särställning i lagens ögon. De flesta av oss har en åsikt om frågan. Jag har gjort en djupdykning i problemet för att räta ut eventuella frågetecken som man möter. Intresset har alltid funnits för detta ämne men först nu har jag fått chansen att verkligen ta reda på svaren.   I uppsatsen tas rättsväsendets syn på dessa förbrytare upp, ända från antiken fram till idag. Vilka lagar och regler som gäller presenteras och, ett antal rättsfall med dess utgång redovisas. Uppsatsen kretsar kring tre grundläggande frågor som tidigt i uppsatsen ställs och i slutet besvaras utifrån resultaten som insamlats och deskriptivt presenterats.   Som komplement till redovisningen om rättsväsendet rörande denna grupp av kriminella, presenteras även den biologiska faktorn bakom en psykiskt störd individ. Olika typer av störningar och dess särdrag, samt orsaken till dessa beskrivs för att ge en komplett bild av problemet och dess orsaker. Det är vikigt är att hålla i minnet är att även om de flesta grova brottslingar kan antas ha någon form av störnig, så är det långt ifrån alla psykiskt störda som kan tänkas vara eller bli brottslingar. Man måste hålla sig borta från stigmatisering.

Abstract:
Adverse health effects attributable to both short- and long-term exposure to air pollution have turned the focus on different microenvironments. The interior of vehicles is of relevance as road traffic emissions and re-suspension of road dust are major sources of pollutants associated with adverse health effects. Hence, the air quality inside vehicles deserves attention regarding human health. This thesis presents a new virtual methodology, using CFD, to study the distribution of fine particulate matter, PM2.5, inside a car cabin. In the CFD model, unsteady RANS and Lagrangian particle tracking were used to simulate particles entering from the exterior. In this study, a practical measurement of interior particle concentrations was also carried out as a first attempt to validate the CFD model. The objective was to find positions inside the cabin where elevated concentrations of PM2.5 are present. The results from the CFD simulations showed that significantly higher concentrations are present at head height in the front row. Due to a discrepancy in the investigated positions in the CFD model and the practical measurement, the simulation results could not be validated. Nevertheless, the simulation results in this study have provided guidelines for future measurements of interior particle concentrations.

This study presents the results of an experimental study that investigates the effects of uniform/waveform tip injection along the camberline on the total pressure loss characteristics downstream of a row of Low Pressure Turbine (LPT) blades. The experiments are performed in a low speed cascade facility. This injection technique involves spanwise jets at the tip that are issued from a series of holes along the camber line normal to the freestream flow direction. The injection mass flow rate from each hole is individually controlled using computer driven solenoid valves and therefore the flow injection geometrical pattern at the tip can be adjusted to any desired waveform shape, and can be uniform as well as waveform along the camber. Measurements involve Kiel probe traverses for different injection scenarios 0.5 axial chords downstream of the blades. Results show that, instead of performing uniform mass injection along the camberline, by selecting an appropriate waveform injection pattern one can reduce the total loss levels of the blade, including the tip leakage loss as well as the wake losses.

Bakgrund: All sjukvård i Sverige styrs av Hälso-och sjukvårdslagen [HSL]. Under vissa omständigheter kan denna lag behövas kompletteras med Lagen om psykiatrisk tvångsvård [LPT]. Dessa personer vårdas inom psykiatrisk slutenvård. Omvårdnadens syfte är att patienten samtycker till vården och tvångsvården kan över gå till frivillig vård. Dessa personers problematik är dock ofta komplex och omvårdnaden upplevs innebära många svårigheter för sjuksköterskorna. Syfte: Att beskriva sjuksköterskors erfarenheter och upplevelse av omvårdnad i samband med LPT-vård inom sluten psykiatrisk vård. Metod: Empirisk kvalitativ studie med deskriptiv design. Studien baseras på sju semistrukturerade intervjuer med sjuksköterskor. Dessa analyserades med manifest innehållsanalys. Resultat: Resultatet visade att en förutsättning för att uppnå målet med tvångsvården genom omvårdnad var att skapa en allians med patienten. Kommunikation som innefattade individuellt anpassad information var en nyckelfaktor till alliansskapande. Omvårdnadsarbetet innefattade samverkan och sjuksköterskorna upplevde ett behov av kompetensutveckling. Slutsats: Sjuksköterskor upplever att omvårdnad enligt LPT är en utmaning och sammankopplat med flertalet svårigheter. Erhålls kompetens, tid för reflektion och trygghet i arbetsgruppen ses dock inte omvårdnaden som en omöjlighet utan kan upplevas som positivt för både den som vårdar och för den som vårdas.
Background: All health care in Sweden is governed by the Health Care Act. Under certain circumstances, this law needs to be supplemented with the Compulsory mental care act. These people cared for within psychiatric inpatient care. Nursing aim is that the patient agrees to care and compulsory care may go to voluntary treatment. These people's problems are often complex and the nursing care experienced many difficulties. Aim: To describe nurses’ caring and experiences for patients under compulsory mental care act within psychiatric inpatient care. Method: Empiric qualitative study with descriptive design. The study is based on seven semi-structured interviews with nurses. These were analysed with manifest content analysis. Result: The results showed that a prerequisite for reach the goal of compulsory care through nursing was to establish alliance with the patient. Communication that included individualized information was a key factor to alliance building. Nursing included collaboration and nurses experienced a need for skills development. Conclusion: Nurses experiencing that caring according to compulsory mental care act is a challenge and is linked to a number of difficulties. Obtained competence, time for reflection and perceived safety in the working group does not care felt as an impossibility, but could be experienced as positive for both the caring and the cared for.

I Sverige finns idag en vårdform där patienter som begått brott under grav psykisk störning samvårdas med personer som har ett allvarligt självskadebeteende. Dessa inrättningar måste då förhålla sig till två olika vårdsätt. Vi vill därför undersöka om personalen beskriver och upplever eventuella skillnader i interaktionen med patienter vilka vårdas med stöd av olika tvångsvårdslagar på en rättspsykiatrisk avdelning. Vi har använt oss av en kvalitativt ansats där vi har intervjuat sex yrkesverksamma inom rättspsykiatrin. Med hjälp av symbolisk interaktionism samt vårdkulturteori har vi analyserat den empiri som framkommit från intervjuerna. Resultaten visar att personalen vill se patienterna såsom en homogen grupp men brottet/handlingen påverkar patientsynen. Resultaten visar även att interaktionen villkoras av personalens attityder och värderingar samt att den individuella vården försvåras ytterligare av samvården. Slutsatsen blev att personalen skildrade en diskrepans där de å ena sidan beskrev att interaktionen var likadan oberoende av vilken patientgrupp det var. Samtidigt framkom att de upplevde och gjorde skillnader mellan LRV- och LPT-patienter.

Studiens syfte är att ge ökad kunskap kring professionellas erfarenheter om hur personal motivera patienter till vård och behandling inom den svenska rättspsykiatrin. Studien lyfter svårigheter och möjligheter med att motivera patienter till att delta i vårdprocessen, i en miljö präglad av säkerhet. Patienter som befinner sig inom rättspsykiatrin, vårdas enligt två lagar; Psykiatrisk tvångsvård (LPT) eller rättspsykiatrisk tvångsvård (LRV). Patienter inom rättspsykiatrin vårdas under tvång och syftet med rättspsykiatrisk vård är att ge patienten rätt behandling och öka kunskapen hos patienten kring patientens sjukdomsbild. Rättspsykiatrisk vård är ska också vara en konsekvens av ett brott, som begåtts under en allvarlig psykisk störning. Målet med behandlingen inom rättspsykiatrin är att patienten inte ska återfalla i brott. Denna studien är av intresse ur ett socialpedagogiskt perspektiv då studien lyfter delaktighet, inkludering, exkludering, relation mellan professionell och patient och motivationsskapande och hur dessa kan uppfyllas i en miljö präglad av säkerhet.  Studien bygger på ett kvalitativt ansats, med hjälp av semistrukturerade intervjuer och med hermeneutiken som metodologisk utgångspunkt. De teoretiska utgångspunkter som används i denna studie är; Maslows behovshierarki och utvecklingsekologin. Studiens slutsats, baserat på den insamlade empirin och respondenternas utsagor, är att motivation är en viktig faktor både för patient och personal, samt att det är viktigt som professionell att inkludera patienten i vårdprocessen. Den insamlade empirin pekar även på stora kunskapsluckor och behovet av mer forskning inom den svenska rättspsykiatrin.

Den digitala utvecklingen får konsekvenser för arbetslivet och dess organisering. I studien undersöks genom fem semistrukturerade intervjuer hur fackligt aktiva orderplockare resonerar kring plocksystemet genom vilket de utför sitt jobb och hur de digitala systemen påverkar maktförhållanden på arbetsplatsen.   Respondenterna arbetar alla inom livsmedelsdistribution på arbetsplatser som arbetar med det digitala plocksystemet pick-by-voice (PbV). Med utgångspunkt i Edwards (1979) tre övergripande kontrollmekanismer styrning, utvärdering och disciplinering samt Kelloggs (2020) sex mekanismer genom vilka algoritmisk kontroll utövas, de så kallade 6R:en, undersöks hur den digitala utvecklingen påverkar möjligheten för arbetsmarknadens parter att utöva kontroll och göra motstånd i arbetsprocessen. Studien finner att PbV innebär att flera arbetsledande funktioner flyttas till det tekniska systemet. Detta innebär en starkare styrning av arbetet genom att systemet begränsar de anställdas handlingsutrymme och en mer omfattande utvärdering och disciplinering genom att varje arbetsmoment loggas på individnivå. Genom realtidsutvärdering sker disciplineringen dels genom självdisciplinering, dels genom belöning för de som rankas högt enligt måltalen och bestraffning för de som inte lever upp till förväntningarna. Här fungerar tillgång till helgarbete, fasta anställningar och rotation som motiverande instrument. Studien finner också att de bemanningsanställda disciplineras hårdare då de pressas genom otryggheten i anställningen. Genom det digitala systemet blir kontrollen främst algoritmisk. Samtidigt förefaller den direkta kontrollen samverka med den algoritmiska kontrollen i utdelandet av både belöningar och bestraffningar utifrån den digitala utvärderingen. Detta motverkar institutionaliserandet av makt i det tekniska systemet och skapar konfliktytor kring det digitala systemet.

The emergence of multidrug-resistant strains of Gram-negative pathogens that rapidly spread in the clinic is of great concern, since the range of antibiotics still effective against these organisms is limited and will continue to diminish. Therefore, the identification of novel and unexplored drug targets is an urgent need. Gram-negative bacteria possess an outer membrane (OM) with highly selective permeability properties due to its asymmetric structure with LPS in the outer leaflet and phospholipids in the inner leaflet. Dissecting the biogenesis of the OM and gaining insights into the multiprotein machineries that assemble this structure is vital if we want to succeed in developing novel antibiotic compounds that can target these machineries. This thesis focuses on the machinery that transports lipopolysaccharide (LPS) to the cell surface: the LPS transport (Lpt) machinery. In Escherichia coli, the Lpt system is composed of seven essential proteins spanning the cell envelope: the ABC transporter LptB2FGC powers the LPS extraction from the inner membrane (IM) and its transport along the periplasmic bridge, comprising LptA, to the OM LptDE translocon, which assembles LPS on the cell surface. Due to its vital role in cell physiology, the Lpt system represents a good target for the development of antibiotics with an innovative mechanism of action. Encouragingly, two promising inhibitors of this machinery have been discovered: murepavadin, which is currently in preclinical development, and thanatin. The research project of this thesis focuses on two main topics: elucidating the mechanism behind thanatin’s antibacterial activity, and the characterization of a mutant six-component Lpt machinery that is functional without LptC. Thanatin is a host-defence antimicrobial peptide recently shown to cause defects in membrane assembly and to bind to the N-terminal β-strand of LptA in vitro (Vetterli et al., 2018). Since this region is involved in both LptA dimerization and interaction with LptC, we implemented the Bacterial Adenylate Cyclase Two-Hybrid (BACTH) system to detect these interactions in the periplasm and probe which is the target of thanatin. With this technique, we found that thanatin targets both interactions and has a stronger inhibitory effect on the LptC-LptA interaction (Moura et al., 2020: https://doi.org/10.3389/fmicb.2020.00909). Further demonstrating a direct effect upon the LPS transport, we observed in thanatin-treated cells the degradation of LptA and the accumulation of LPS decorated with colanic acid (Moura et al., 2020), both of which have been previously reported to be indicative of LPS transport defects (Sperandeo et al., 2008, 2011). We further explored how thanatin affects the integrity of the cell envelope and observed that it induces promoters regulated by envelope-specific stress response systems (unpublished data). Although all seven Lpt proteins have been shown to be essential, viable mutants lacking LptC but carrying suppressor mutations at the residue R212 in the periplasmic domain of LptF were isolated by our group (Benedet et al., 2016). Interestingly, LptC was recently proposed to have a regulatory role on the LptB2FGC transporter by modulating its ATPase activity (Owens et al., 2019; Li et al., 2019), thus adding to the mystery of how the suppressor mutants can survive without LptC. In the second part of the project, we elucidated how the cell can bypass the presence of LptC and its regulatory role in the machinery by performing a biochemical characterization of the most representative suppressor mutant (manuscript ready for submission). Moreover, by analysing the interaction networks around the residue R212 of LptF, we also formulated a putative mechanism adopted by the Lpt transporter to regulate LPS transfer from LptB2FGC to LptA.

Les bactéries présentent plusieurs mécanismes qui leur confèrent la capacité de faire face à des situations difficiles.Dans un contexte global de résistance aux antibiotiques, les bactéries à Gram négatif présentent des mécanismes de résistance intrinsèque. L'enveloppe multicouche complexe et dynamique, enrobée de lipopolysaccharides (LPS), confère à ces bactéries une capacité de survie accrue. La biosynthèse de ces glycolipides est initiée dans le cytoplasme et son transport se déroule depuis la membrane interne jusqu'à la membrane externe, avec une voie de biosynthèse/transport dédiée.La machinerie de transport des lipopolysaccharides (Lpt) comprend sept protéines fondamentales (LptA à LptG) qui couvrent toute l'enveloppe. Au niveau de la membrane interne, le transporteur LptB2FG couple l'hydrolyse de l'ATP avec l'extraction du LPS. LptB2 est directement en charge de l’hydrolyse de l’ATP tandis que LptF/G interagit avec le LPS et le transporte vers LptC/LptA dans le périplasme.Cette machinerie utilise une architecture conservée avec des domaines de jellyroll dédiés présents sur LptF/G/C/A qui s'assemblent en un pont permettant au LPS de s'écouler vers la membrane externe.Les molécules qui seraient capables de perturber les interactions entre protéines et les différents domaines jellyroll du système, pourraient devenir de puissants inhibiteurs de la construction de la paroi cellulaire. La thanatine, un peptide antimicrobien naturel, a été décrite comme ciblant les domaines jellyroll de la machinerie. Nous avons examiné son effet dans la perturbation du complexe LptC/A. La thanatine se lie pas à LptC mais uniquement à LptA et inhibe l'assemblage du complexe à faible concentration (de l’ordre du nao molaire), démontrant ainsi le potentiel des interactions entre les jellyrolls du système LptC.Le réseau d'interactions entre LptB2FG et LptC/A n'est pas entièrement compris. Le LptB2FG a été produit dans des micelles de détergents et dans des particules de type nanodisque, pour sonder les interactions avec LptC et LptA à l'échelle atomique, à l'aide de diverses techniques biophysiques.Dans l'assemblage du pont LptB2FGCA, LptC/F interagissent principalement à travers les domaines jellyroll. Une mutation dans le résidu R212 de LptF a rendu la présence de la protéine LptC facultative in vivo.La caractérisation biophysique/biochimique a montré une interaction inchangée du mutant LptB2FG avec LptC et LptA, tandis que l'activité ATPase a montré un manque de régulation par la présence de ses partenaires. Cela nous a conduit à proposer que R212 soit un point de contrôle dans LptF pour que LptB2FG détecte le bon assemblage de la machinerie.Lorsque le complexe LptB2FGCA est assemblé in vitro, LptB2 s'est avérée capable de catalyser le phosphotransfert entre deux molécules d'ADP, générant de l'ATP et de l'AMP, et représentant une nouvelle activité (Adenylate Kinase) jusqu'alors non décrite pour cette protéine. Étant un sujet très récent dans la littérature, le rôle des transporteurs à double fonction n'est pas encore bien compris. Pour caractériser l'équilibre entre ATPase et Adenylate Kinase, nous avons muté LptB2 sur des motifs ABC clés pour sonder l'emplacement de l'activité Adenylate Kinase. L’étude du complexe LptB2FG préparé dans des particules de nanodisques, suggère que l'équilibre entre les activités dépend de l'assemblage dynamique de LptB2FGCA. La caractérisation structurale de LptB2 dans sa forme apo et lié aux nucléotides a été initiée.Ce projet, axé sur le système Lpt essentiel pour la survie bactérienne, met en lumière l'importance des interactions protéine-protéine comme cibles pour la conception de futurs composés antimicrobiens. L’intérêt de cibler des transporteurs à double fonction, à la fois impliqués dans la synthèse de la paroi cellulaire et l'exportation de médicaments, pourrait aussi représenter une piste prometteuse pour la recherche future de nouvelles drogues
Bacteria display several intrinsic mechanisms which confers them the ability to cope with disadvantageous situations, such as nutrient deprivation, environmental inter/intra-species competition, managing adaptation to detrimental conditions, and handling effects of antibacterial compounds.In a global context of antibiotic resistance accelerated by anthropogenic activities, gram negative bacteria display intrinsic resistance mechanisms. The complex and dynamic multilayered envelope, coated with lipopolysaccharides (LPS), confers these bacteria increased survivability. Biosynthesis of these complex glycolipids is initiated in the cytoplasm, and its transport proceeds along the inner membrane, periplasm, until reaching the outer membrane, with a dedicated biosynthetic pathway and transport machinery.The Lipopolysaccharide Transport (Lpt) machinery comprises 7 fundamental proteins (LptA to LptG) that span the entire envelope. More specifically, at the inner membrane, LptB2FG ABC transporter couples ATP hydrolysis with LPS extraction. LptB2 cycles ATP while LptF/G interact with LPS and carry it towards LptC and LptA in the periplasm.This machinery uses a conserved architecture with dedicated jellyroll domains present on LptF, LptG, LptC and LptA that assemble into a bridge that allow LPS flow to the outer membrane.Molecules that would disrupt protein-protein interactions between the different jellyroll domains of the Lpt system could become potent cell wall inhibitors. Thanatin, a natural occurring antimicrobial peptide, has been described as targeting the jellyroll domains of the machinery. We screened its effect in the disruption of LptC-LptA complex. Thanatin binds to LptA but not LptC and inhibits the assembly of the complex at low nM concentrations, showing the potential of targeting Lpt Jellyroll-jellyroll interactions.The network of interactions between the Inner membrane complex, LptB2FG and periplasmic LptC and LptA is not fully understood. LptB2FG was produced in detergent micelles and within nanodisc particles, to probe interactions with LptC and LptA at an atomic scale, using Nuclear Magnetic Resonance (NMR) and biophysical techniques.In the assembly of the LptB2FGCA bridge, LptC and LptF interact mostly through the jellyroll domains. A mutation in the LptF jellyroll (R212 residue) rendered LptC presence facultative in vivo.Biophysical and biochemical characterization showed unaltered interaction of mutant LptB2FG with LptC and LptA, whereas ATPase activity showed lack of regulation by presence of its partners. This led us to propose that R212 is a checkpoint in the LptF jellyroll, acting as a hub for LptB2FG to sense proper assembly of the machinery.When LptB2FGCA complex is assembled in vitro, LptB2 was found capable of catalyzing phosphotransfer between ADP molecules, generating ATP and AMP, a novel activity (Adenylate Kinase) previously undescribed for this protein. Being a topic of very recent interest in the literature, the role of dual-function transporters is not understood. To characterize the balance between ATPase and AK, we mutated LptB2 on key ABC motifs to probe possible location for AK activity. LptB2FG studied in nanodisc particles, suggests that balance between activities depends on the dynamic assembly of LptB2FGCA, with regulatory mechanisms possibly not being shared between both activities. Structural characterization of LptB2 in apo and nucleotide bound-state was initiated .This project, focused on the essential Lpt system, sheds light on the importance of protein-protein interactions as targets for designing future antimicrobial compounds. It could also be worth evaluating if dual-function transporters, involved in cell wall synthesis and drug export, are valid targets for future drug screenings

The publisher of the Degrees of Reading Power test of reading comprehension (DRP) calibrate their test using an item response model called the Rasch or one-parameter logistic model. The relationship between the use of the Rasch model in calibration of the DRP and the use of the DRP as a component of the Virginia Literacy Passport Testing Program (LPT) is addressed. Analyses concentrate on sixth grade students who were administered the DRP in 1991. The question that arises is whether the Rasch model is the appropriate model to use to calibrate the DRP in this high-stakes setting. The majority of research that has been reported by the publisher of the DRP to assess the adequacy of the Rasch model have not included direct checks on model assumptions, model features or model predictions. Instead, they have relied almost exclusively on statistical tests in assessment of model fit. This study will assess the adequacy of fitting DRP test data to the Rasch model through direct examination of the assumptions, features and predictions of the IRT model. This is accomplished by comparing the Rasch model to the less restrictive two- and three-parameter logistic models. Robust IRT-based goodness-of-fit techniques are conducted. When the DRP is used in a high stakes setting, guessing is likely for those in jeopardy of failing. Under these circumstances, we must attend to the possibility that guessing may be a factor and thereby calibrate the DRP with the three-parameter model, as this model takes guessing into account.

The thesis draws on transaction cost economics (TCE), agency, contingency, and labour process theories in the context of appraising factors motivating outsourcing. Two empirical phases have been undertaken in the study. The first phase involved a series of interviews with general managers and financial controllers in large South East Queensland hotels. The second phase involved two distinct questionnaire surveys of large Australian hotels. The first was administered to hotel general managers, and the second was administered to hotel financial controllers. Significant findings arising from the study include: 1. In light of the substantial international literature describing hotel outsourcing, it appears that outsourcing in Australian hotels is relatively limited. This appears to be particularly the case with respect to food and beverage related activities. 2. Mixed support is offered for the TCE model. Both the survey and interview data provide some support for TCE's prescription that frequently conducted activities will not tend to be outsourced. Two specific extensions are offered to this aspect of the model, however. Firstly, where activities are conducted to a minimal extent, it can be uneconomic to outsource. Secondly, where large activities are undertaken by a group of organisations, their enhanced purchasing power can result in inexpensive outsourcing arrangements. With respect to TCE's uncertainty proposition, support is offered for the view that the propensity to outsource will be greater where behavioural uncertainty is lower. No support has been offered with respect to environmental uncertainty. The interview data provides some support for TCE's asset specificity proposition, however, minimal support was found in the survey phase. Despite this, the many dimensions of asset specificity (eg. site specificity, human asset specificity, etc) provided a useful checklist of issues to be considered in relation to the outsourcing decision. 3. Negligible support was found for labour process theory (LPT) in the interview phase of the study. In light of this, and the need to narrow the study’s focus in the survey phase, LPT was not pursued further. LPT is a difficult construct to operationalise, given the social desirability error that may result. This may partially account for the absence of significant LPT findings in the interview phase. 4. The survey data provides some support for the agency theory view that risky activities will tend to be outsourced. 5. Considerable cross-hotel variation exists in management of, and accounting's involvement in, outsourcing decision making and control systems. Abstract:
Estudos epidemiológicos e experimentais mostram que a poluição do ar pode causar diversos efeitos adversos na saúde, dentre eles inflamação sistêmica e pulmonar, doenças cardiovasculares e exacerbação de doenças pré-existentes. A síndrome do desconforto respiratório aguda é caraterizada por intenso infiltrado inflamatório, dano na barreira alvéolo-capilar e hipoxemia, e desde a sua descrição em 1967 ainda apresenta elevados índices de mortalidade. O presente estudo tem como objetivos: 1. Avaliar qual o impacto da poluição atmosférica de uma região urbana sobre a progressão da LPA induzida por LPS; e 2. Avaliar se a lesão induzida por LPS é alterada em um indivíduo previamente exposto à poluição atmosférica. Os principais achados relativos ao objetivo 1 mostram que, de uma forma geral, quando há a interação do material particulado fino (MP2,5) anterior à fase aguda da LPA, esta não se desenvolve de forma tão grave. Quanto aos parâmetros inflamatórios, nota-se que, na maioria dos parâmetros, as células inflamatórias e citocinas pró-inflamatórias aumentam no grupo LPS 24 horas, mas não, ou não tanto quanto, no grupo poluição+LPS. Como base nestes resultados podemos hipotetizar que pode ter ocorrido uma alteração do perfil da resposta inflamatória ou imunotoxicidade. Quando observamos os resultados referente ao objetivo 2, nota-se que, o grupo LPS + poluição permanece em um estado inflamatório persistente com número de leucócitos aumentado no lavado bronco-alveolar e níveis elevados de citocinas pró-inflamatórias (IL-1beta, IL-6 e IL-8) no tecido pulmonar, enquanto o grupo LPS 5 semanas tem estes parâmetros mais próximos do grupo controle. Na morfologia tecidual, o grupo LPS + poluição a diminuição do espaço aéreo alveolar e o espessamento septal. É bastante plausível que a poluição do ar dificulte recuperação e o reparo adequado da lesão causada pelo LPS, uma vez que a poluição do ar, e especialmente o material particulado fino, exerce um papel pró-inflamatório contínuo sobre a lesão. Podemos concluir que: o uso do LPS por nebulização é um modelo viável para a reprodução dos parâmetros característicos da lesão pulmonar aguda, a exposição ao material particulado pode alterar o perfil de resposta imediata (24 horas) na lesão pulmonar aguda e pode dificultar a recuperação da lesão. Estudo adicionais são necessários para entender o possível papel da modulação da resposta imunológica nos mecanismos envolvidos nestes processos
Epidemiological and experimental studies show that the air pollution can cause several harmful outcomes to the health, which include systemic and pulmonary inflammation, cardiovascular diseases, and exacerbation of preexisting diseases. The acute respiratory distress syndrome is characterized by intense inflammatory response, alveolo-capillary barrier damage and hypoxemia and since it was described for the first time in 1967 it still has high mortality rates. This study aims to 1. Evaluate the impact of urban air pollution exposure on the acute lung injury progression and 2. Evaluate if the LPS-induced injury is altered in an individual previously exposed to the air pollution. The main findings regarding objective 1 show that when there is an interaction of the particulate matter on the acute phase of LPS-induced injury, the lesion is not as severe as in the group that received only LPS. The inflammatory parameters show that inflammatory cells and pro inflammatory cytokines are increased in the LPS 24 hour, whereas not, or not as much as, in the air pollution + LPS group. Based on these results, we can hypothesize that may have occurred a shift of the inflammatory profile or immunotoxicity. Results of the objective 2 show that the group LPS + air pollution remains in a persistent inflammatory condition with increased leucocytes in BALF and pro inflammatory cytokines (IL-1beta, IL-6 e IL-8) also increased in the lung tissue, while the LPS 5 weeks group shows these parameters levels closer to the control group. The tissue morphology displays a diminished alveolar air space and septal thickening. It is very likely that the air pollution interferes on the adequate LPS-induced lesion recovery and repair, once that the air pollution, specially the fine particulate matter, has a continuous pro-inflammatory role over the lesion. We can conclude that: the use of nebulized LPS is a feasible acute lung injury model; the exposure to the particulate matter could alter the profile of the immediate response (24 hours) of the acute lung injury and it can impair the lesion recovery. Additional studies are necessary to understand the possible role of the immunological response modulation mechanisms involved in these processes

This paper presents work being done at NASA/GSFC by the Operating Missions as Nodes on the Internet (OMNI) project to demonstrate the application of Internet communication technologies to space communication. The goal is to provide global addressability and standard network protocols and applications for future space missions. It describes the communication architecture and operations concepts that will be deployed and tested on a Space Shuttle flight in July 2002. This is a NASA Hitchhiker mission called Communication and Navigation Demonstration On Shuttle (CANDOS). The mission will be using a small programmable transceiver mounted in the Shuttle bay that can communicate through NASA’s ground tracking stations as well as NASA’s space relay satellite system. The transceiver includes a processor running the Linux operating system and a standard synchronous serial interface that supports the High-level Data Link Control (HDLC) framing protocol. One of the main goals will be to test the operation of the Mobile IP protocol (RFC 2002) for automatic routing of data as the Shuttle passes from one contact to another. Other protocols to be utilized onboard CANDOS include secure login (SSH), UDP-based reliable file transfer (MDP), and blind commanding using UDP. The paper describes how each of these standard protocols available in the Linux operating system can be used to support communication with a space vehicle. It will discuss how each protocol is suited to support the range of special communication needs of space missions.

Abstract:
Lipopolysaccharide (LPS) is the glycolipid that comprises the outer leaflet of the Gram-negative outer membrane (OM). Because it is essential in nearly all Gram-negative species, and because it is responsible for making these bacteria impervious to many types of antibiotics, LPS biogenesis has become an important area of research. While its biosynthesis at the cytoplasmic face of the inner membrane (IM) is well studied, the process by which it is removed from the IM, transported across the aqueous periplasmic compartment, and specifically inserted into the outer leaflet of the OM is only beginning to be understood. This transport process is mediated by the essential seven-protein LPS transport (Lpt) complex, LptA/B/C/D/E/F/G. The OM portion of the exporter, LptD/E, is a unique plug-and-barrel protein complex in which LptE, a lipoprotein, sits inside of LptD, a β-barrel integral membrane protein. LptD is of particular interest, as it is the target of an antibiotic in Pseudomonas aeruginosa. Part I of this thesis investigates how the cell forms the two non-consecutive disulfide bonds that connect LptD's C-terminal β-barrel to its N-terminal soluble domain. These disulfides, one of which is almost universally conserved among Gram-negatives, are essential for cell viability. Here, we show that an intermediate oxidation state with non-native disulfide bonds accumulates in the absence of LptE and in strains defective in either LptE or LptD. We then demonstrate that this observed intermediate is on-pathway and part of the native LptD oxidative folding pathway. Using a defective mutant of DsbA, the protein that introduces disulfide bonds into LptD, we are able to identify additional intermediates in the LptD oxidative folding pathway. We ultimately demonstrate that the disulfide rearrangement that activates the LptD/E complex occurs following an exceptionally slow β-barrel assembly step and is dependent on the presence of LptE. Part II describes work towards obtaining X-ray crystal structures of the LptD N-terminal domain and LptD/E complex. Expression construct and purification optimization enabled the production of stable LptD/E in quantities that make crystallography feasible. Numerous precipitants, detergents, and additives were screened, ultimately resulting in protein crystals that diffract to a resolution of 3.85 Å.

Piezoelectric materials are well known for their applications in surface (SAW) and bulk acoustic wave (BAW) devices such as oscillators, resonators and sensors. Quartz has been the main material used in such applications. Ternary calcium gallium germanate (CGG) structure-type materials, so-called langasites, recently emerged as very promising because of their piezoelectric properties superior to quartz. This thesis discusses the growth of langasite-type La3Ga5.5Ta0.5O14 (LGT) films by liquid phase epitaxy (LPE) technique and their chemical and structural characterization. In addition, the different techniques suitable for the growth of LGT are discussed and compared. To adjust the materials properties for given applications, doping by selected ions can be used. However, the dopants must be homogeneously distributed. In the current study, Al, Ti, Cr and Ca were investigated as dopants. In an earlier study, Al and Ti had been chosen because of their ability to substitute the octahedral site of LGT, normally occupied by Ga (CN=VI) with a segregation coefficient near unity in Czochralski growth. Doping with Ca and Cr has never been reported before, and therefore, the segregation behavior was unknown. In this study, Al, Ti and co-doping with Cr and Ca has been investigated for both X and Y-oriented films. The dopant distribution in the films was quantitatively evaluated by Secondary Ion Mass Spectroscopy (SIMS), using ion-implanted LGT substrates as standards. The drop of dopant concentration, in the SIMS profile, allows for the identification of the film-substrate interface and to accurately measure the thickness of the films. The film thickness is found to be typically of the order 0.5 to 2µm, depending on growth conditions. The solvent was found a reliable choice, as solvent ions were not incorporated in the films above the detection limits of the characterization techniques. A lead oxide solvent system is used as a solvent for the growth of LGT LPE films with different orientations. Extensive structural characterization was performed. The crystallinity of substrates and films grown with different orientations was compared by X-ray diffraction (XRD). The films show a very high structural perfection, with typically FWHM values of 0.035 for the (004) reflection of the XRD rocking curve. The films were also characterized by TEM. The optical transmittance of the films was characterized by Varian optical spectrophotometer, and the value obtained of approximately 80% is comparable with the transmittance value of the Czochralski grown polished substrate.

The purpose of this essay is to try and bring clarity to the question, what is Chick Lit and which factors make it so popular. My thesis  endeavors to explain that it is not only the promise of light entertainment that draws the reader, but also the possibility that in an easy way they can read about existential questions such as self-development  and life -choices.

As well as mapping out Chick Lit´s specific characteristics, followed by previous research on the subject and the litterateur’s history, I have found it interesting to discuss the female characters, their personalities and life choice’s against a backdrop of existentialistic philosophy.

In my research of this form of literateur I have discovered that chick lit often deals with existential universal problems, and that in order to be entertaining these books must contain a serious element.

A significant number of Low Probability of Intercept (LPI) radars are used in various military applications, from guided weapons (such anti-ship missile), to large platforms (aircrafts, ships), to large systems (Integrated Air Defense Systems - IADS). The purpose of this thesis is to evaluate the performance of netted LPI radar systems. To do so, it commences with establishing the theoretical background for the LPI radar techniques and detection methods. Additionally, it presents existing LPI assets along with their operational characteristics to provide the reader with a useful tool for comparative analysis of the LPI radar market. As this work focuses on LPI radar networks, specific emphasis is given to clarifying the notion of a netted system; the conceptual and mathematical background for such are presented in a separate chapter.
