Abstract:
A pesar de la amplia bibliografía sobre arquitectos del Movimiento Moderno y sobre los debates de las Vanguardias, aún permanecen obras sin estudiar de arquitectos y artistas, cuyos compromisos contribuyeron en el Nacimiento de las Vanguardias, abriendo camino a la eclosión del Movimiento Moderno. Esta tesis se desarrolla desde la implicación de dos artistas, el arquitecto Michael Loewe (1871-1955) y su hija, la artista Margot Loewe (1905-1974), cuyas obras nos permiten reconocer como se entrelazaron dos generaciones en pro de unos mismos ideales, y como el dialogo entre técnica y arte y entre arte e industria propició nuevos espacios y nuevos modos de vivir. La investigación se ubica en Berlín, escenario cultural de confrontación entre tradición y modernidad, de búsqueda de una nueva identidad arquitectónica, y gran laboratorio urbanístico y social que pasa de pequeña capital de Provincia a Metrópoli, entre 1871 y 1933, desde Bismarck hasta la República de Weimar. El estudio desglosa la trayectoria profesional del arquitecto alemán, partiendo de su formación y carrera como Regierungbaumeister en la Königliche Technische Hochschule de Berlin-Charlottenburg, y sus obras construidas en Berlín entre 1907 y 1933. Por orden de aparición, las obras presentadas son los edificios de Bismarckstrasse 82-83 (1907), Olivaerplatz 8 (1909), Manfred-von-Richthofen-Strasse 2 (1912), Pariser Strasse 20 (1913), los grupos de viviendas de Prausestrasse 5-27 (1920), de Kuno-Fischer-Strasse 3-4-5 (1923) y de Pichelswerder (1927). En las obras de Michael Loewe identificamos una primera etapa, previa a la I Guerra Mundial, de reacción contra el exceso ornamental de los estilos históricos y del Jugendstil, moviéndose entre la desnudez y la simplicidad geométrica, con una clara influencia del Biedermeier y del Neoclasicismo Alemán, y en la que colabora con el arquitecto-ingeniero de prestigio Bruno Möhring. En su segunda etapa, de Entre Guerras, evoluciona y se compromete tempranamente hacia un lenguaje próximo a la Neue Sachlichkeit, en que integra su proyecto en el paisaje, explorando nuevas tecnologías y nuevas formas de vivir, que le permiten expresarse con más libertad, situándose en la primera generación de influencia de F. L. Wright en Europa. En ese temprano compromiso por la Modernidad, Michael Loewe despertó reacciones favorables de partidarios del Neues Bauen, pero también fue punto de mira de los adversarios de la Nueva Arquitectura, cuyos artículos y posicionamientos se radicalizaron, convirtiéndose en objeto de lucha política En su obra afloran sus compromisos profesionales a favor de la Nueva Objetividad, que plantea el problema de la posición del arte en la sociedad, de la relación entre arte y artesanía, entre arte y técnica y finalmente el de la forma y función. Sus compromisos personales le llevaron a ejercer de promotor en los momentos de crisis económica, a adherirse a propuestas de Existenzminimum, y a experimentar nuevas formas de vivir. También le llevaron a impulsar a su hija Margot Loewe a acceder al Bauhaus, que estudia entre 1929 y 1933 en Bauhaus-Dessau, bajo la dirección de Hannes Meyer y de Mies van der Rohe. La trayectoria artística de Margot Loewe que se expone aquí, corresponde al breve periodo comprendido entre los años de sus estudios en Bauhaus-Dessau y el exilio, pero entendido como parte del debate entre las Vanguardias utópicas y la Bauhaus, en su contexto de dialogo entre tecnología y arte, en el que la tecnología permitió una nueva concepción del espacio, argumento común de la arquitectura y del arte. Los compromisos técnicos y sociales de Michael Loewe le llevaron a nuevas propuestas formales y a explorar nuevas maneras de vivir, y en sus motivaciones artísticas participó con su hija Margot Loewe en esa nueva mirada subjetiva, provocando impresiones visuales y emociones, que empuja al arquitecto a experimentar con nuevos espacios, y a la artista a la abstracción de su pintura.
Despite the extensive literature concerning architects of the Modern Movement and debates on the Avant-Garde, several works of architects and artists remain unstudied. The commitments of these individuals contributed to the birth of the Avant-Garde Movement, opening the door for the flourishing of the Modern Movement. This thesis was developed and inspired by the works of two artists, architect Michael Loewe (1871 -1955) and his daughter, the artist Margot Loewe (1905-1974). These artists' work allows us to recognize how these two generations were interwined and contributed the same ideals, as well as how the dialogues between technique and art and between art and industry led to new spaces and new ways of living. This research project was conducted in Berlin, a cultural center of confrontation between tradition and modernity and a city that is in search of a new architectural identity. Berlin represents a major urban and social laboratory that changed from a small provincial capital into a metropolis between 1871 and 1933. This study anlyses this German architect's career, starting from is training and career as a Regierungbaumeister in the Königliche Technische Hochschule of Berlin-Charlottenburg to his architectural accomplishments that were built between 1907 and 1933. The works presented include the buildings of Bismarckstrasse 82-83 (1907), Olivaerplatz 8 (1909), Manfred-von-Richthofen-Strasse 2 (1912), Pariserstrasse 20 (1913), the housing projects of Prausestrasse 5-27 (1920) and of Kuno-Fischerstrasse 3-4-5 (1923), and Pichelwerder (1927). Regarding the works of Michael Loewe prior to World War I, we have identified an initial stage of reaction against the ornamental excess of historical styles and Jugendstil that moved between unadorned designs and geometric simplicity with clear influence from German Neoclassicism . Collaborative works with the prestigious architect-engineer Bruno Möhring were also identified in this stage. In the second stage of hi work in the years between the wars , Michael Loewe envlved and undertook early steps towards a !anguage close to the Neue Sachlichkeit, integrating his project ¡nto the landscape and exploring new technologies and new ways of living that allowed him to express himsell more freely. During his early commitment to Modernity, Michael Loewe's work elicited favorable reactions from supporters of the Neues Bauen, but he was also targeted by the opponents of the New Architecture, whose papers and positions were radicalized, and became the subject of political conflict. In Michael Loewe's work, his professional commitments emerge in favor of the New Objectivity,which raises the issue of the position of art in society and the relationships between art and craft, art and technology, and form and function . Michael Loewe's personal commitments led him to join Existenzminimum proposals, and to experience new ways of living . He encourages his daughter Margot Loewe to study between 1929 and 1933 at the Bauhaus-Dessau under the guidance of Hannes Meyer and Mies van der Rohe. The artistic trajectory of Margot Loewe that is presented corresponds to the brief period between the years of her studies at the Bauhaus-Dessau and her exile. Margot Loewe's work is understood as part of the discussion between utopian Avant-Garde and Bauhaus, as well as its context for dialogue between technology and art, where techlology enabled a new conception of space, which is a common theme in both architecture and art. The technical commitments of Michael Loewe led him to create new formal proposals and as a result of his artistic motivations, he also participated alongside his daughter Margot Loewe in new forms of subjective insight, which created visual impressions and emotions that pushed the architect to experiment with new spaces and led the artist to the abstraction of her painting.

Abstract:
This thesis has been written in order to provide, especially for the non-German-reading musician, a fuller picture of Loewe and his ballads than has been available up to now. This picture is developed within the literary background history of the ballad poems, and the literary, mental, and musical climate at the beginning of the Romantic era; further, Loewe's life, as revealed in his many letters, his diaries, and his autobiography, provides the human context from which the ballads emerge as a logical extension of his personality. These earlier parts of the thesis have considerable bearing on the appreciation of Loewe's timely position in musical history, treating as they do with the popularity of the ballad poems, the rapid expansion of the means of musical/emotional expression, and the complete acceptance of that most romantic and versatile of soloinstruments, the piano. Loewe's temperamental affinity with the poetry of the ballads is shown to have affected his choice of subject, and in many cases the ultimate quality of the music is obviously dependent upon the strength or otherwise of his attraction. After observations on Loewe's vocal and piano writing, the thesis treats the ballads primarily with regard to their feeling and emotional content, and investigates the musical means by which this is conveyed. Categories are suggested, and ballads of similar dramatic, pictorial, or emotional type are discussed and compared. Certain formal characteristics are examined, in particular Loewe's use of highly organised motivic work in certain ballads, which foreshadows its later use by Liszt, Wagner and others. Over one hundred of Loewe's 120 ballads are dealt with, some in extensive detail~ and copious musical examples are given. The few comparatively well-known ballads receive due attention, but it was regarded as important to bring to light some of the more neglected or unknown ballads, many of which possess great beauty and originality, amply repaying study and, still more, performance. As a corollary, the approach of the performer is considered, and the Conclusion argues for an informed :esthetic appreciation of Loewe's ballads and their place in teday' s vocal repertoire.

Abstract:
Carl Loewe (1796-1869) was a prolific composer of works for voice and piano with an output exceeding 400 pieces. Just as Schubert pioneered Lieder as a new genre of art music in the nineteenth century, Loewe can be credited for his comparable innovation with the ballad, a narrative song that depicts a story. Though Loewe is often considered a conservative musical figure in the nineteenth century, later romantic composers like Richard Wagner and Hugo Wolf held his ballads in high regard, as they show Loewe's compositional originality in boldly producing drama through the piano-singer format. This document displays how Loewe in his ballad cycle Gregor auf dem Stein, Op. 38 (1834) creates a continuous musical drama to enhance a theological legend. This work is an example of how Loewe foreshadows aspects of later German Romanticism, more fully realized by Wagner and Wolf, through use of musical and dramatic continuity, progressive tonality, motives, and declamatory vocal style.

Abstract:
Now all but forgotten, the songs of Sidney Homer (1864-1953) were at one time well-regarded and often performed. Married to the great American contralto Louise Homer, he was in a unique position to have his songs performed by the great artists of the time. Unlike the cloying "parlor songs" of many of his contemporaries, his works consistently demonstrate a respect for both the great poets as well as the European art-song tradition. One of the most cosmopolitan of the American composers of his day, his involvement with Louise's career brought him into contact with many of the great composers and performers of the day including Massenet, Puccini, Humperdinck, Mahler, Toscanini and Caruso. When viewed in their entirety, his songs reveal not only a tremendous variety, but also the maturation of his compositional style.

Resumo: Música e poesia. Como as duas artes interagem, que significados são criados, reiterados, modificados e como a performance se constrói e se enriquece a partir desta interação é a reflexão que impulsiona o presente trabalho. Propomos uma análise músico-textual visando a interpretação e performance de quatro Lieder do período Romântico construídos sobre o poema Gretchen am Spinnrade de J. W. Goethe. Serão contempladas as canções dos compositores Louis Spohr (Gretchen), Franz Schubert (Gretchen am Spinnrade), Carl Loewe (Meine Ruh ist hin), e Richard Wagner (Meine Ruh ist hin). Para isso, é feita uma introdução sobre o gênero Lied e sobre o poeta. Em seguida, é proposta uma análise literária do poema, que servirá de base para a interpretação das canções. São abordados aspectos conteudísticos e formais do texto, assim como sua contextualização dentro da peça Fausto I, de que foi extraído. Em seguida, é feita uma análise musical das obras, com foco nas relações texto-música presentes e contemplando aspectos tanto da linha vocal quanto do acompanhamento. Ao longo da análise são feitas sugestões interpretativas baseadas nas relações texto-música
Abstract: Music and Poetry. How the two art forms interact, what meanings are created, emphasized, modified and how the performance is built and enriched from this interaction is the reflection that impels the present work. A text-music analysis aiming towards the interpretation and performance of four Lieder - settings from the Romantic period of the poem Gretchen am Spinnrade from J. W. Goethe is presented here. The settings of Louis Spohr (Gretchen), Franz Schubert (Gretchen am Spinnrade), Carl Loewe (Meine Ruh ist hin), e Richard Wagner (Meine Ruh ist hin) will be approached. After introductory considerations about the genre Lied and about the poet are made, an analysis of the poem is presented, which will ground the interpretation of the songs. Content and formal aspects of the text are considered, as well as its contextualization in the play Faust I, from where it was extracted. Afterwards, a musical analysis of the songs is presented, focusing on the existing text-music relations and taking aspects from the vocal line as well as the accompaniment into consideration. During the analysis interpretative suggestions are made, based on the text-music relations

Abstract:
The Karhunen-Loeve Expansion (K-L expansion) is a bi-orthogonal stochastic process expansion. In the field of stochastic process, the Karhunen-Loeve expansion decomposes the process into a series of orthogonal functions with the random coefficients. The essential idea of the expansion is to solve the Fredholm integral equation, associated with the covariance kernel of the process, which defines a Reproducing Kernel Hilbert Space (RKHS). This either has an analytical solution or special numerical methods are needed. This thesis applies the Karhunen-Loeve expansion to some fields of statistics. The first two chapters review the theoretical background of the Karhunen-Loeve expansion and introduce the numerical methods, including the integral method and the expansion method, when the analytical solution to the expansion is unavailable. Chapter 3 applies the theory of the Karhunen-Loeve expansion to the field of the design experiment using a criteria called "maximum entropy sampling". Under such setting, a type of duality is set up between maximum entropy sampling and the D- optimal design of the classical optimal design. Chapter 4 uses the Karhunen-Loeve expansion to calculate the conditional mean and variance for a given set of observations, with application to prediction. Chapter 5 extends the theory of the Karhunen- Loeve expansion from the univariate setting to the multivariate setting: multivariate space, univariate time. Adaptations of numerical methods of Chapter 2 are also provided for the multivariate setting, with a full matrix development. Chapter 6 applies the numerical method developed in Chapter 5 to the emerging area of multivariate functional data analysis with a detailed example on a trivariate autoregressive process.

Abstract:
The Karhunen-Loeve Transform (KLT) is a popular transform used in multiple image processing scenarios. Sometimes, the application of the KLT is not carried out as a single transform over an entire image Rather, the image is divided into smaller spatial regions (segments), each of which is transformed by a smaller dimensional KLT. Such a situation may penalize the transform efficiency. An improvement for the segmented KLT, aiming at mitigating discontinuities arising on the edge of adjacent regions, is proposed in this paper. In the case of moderately varying image regions, discontinuities occur as the consequence of disregarded similarity between transform domains, as the order and sign of eigenvectors in the transform matrices are mismatched. In the proposed method, the KLT is adjusted to guarantee the best achievable similarity via the optimal assignment and sign correspondence for eigenvectors. Experimental results indicate that the proposed transform improves the similarity between transform domains, and reduces RMSE on the edge of adjacent regions. In consequence, images processed by the adjusted KLT present better cohesion and continuity between independently transformed regions.

Abstract:
This thesis proposes a speaker-independent, continuous-speech stop recognition algorithm. The method incorporates knowledge into the system by means of rules whose conditions are linked to acoustic events. Coarticulatory effects present in the speech are taken into account with the analysis of the transient portions of the signal.
The novelty of this approach consists of the use of the Karhunen-Loeve transform to segment the speech and to extract both steady-state and dynamic features. Sequences of stops followed by vowels were segmented with the second Karhunen-Loeve transform coefficient, whereas information about the formant transitions and energy distribution was extracted by means of the basis vectors of the transformed space.
Tests of the proposed rule-based system resulted in 71% correct classification of the stops' place of articulation, and 80% correct classification of the stops' manner of voicing.
More complex systems have previously attained better results. However, the simplicity and computational cost efficiency of this approach would be of considerable benefit to any large ASR system.

Sinais podem ser amostrados, armazenados e reproduzidos por computadores digitais. Adicionalmente, sinais podem ser analisados de modo que formas comportadas de sua representação possam ser extraídas, economizando o espaço de memória necessário para o armazenamento. A transformada de Fourier tem sido tradicionalmente utilizada para este fim. Este trabalho trata da utilização da transformada Karhunen-Loeve como método alternativo de representação de tons musicais, possibilitando grande economia de recursos computacionais comparada com a transformada de Fourier.
Signals can be sampled, stored and played back by digital computers. Furthermore, signals can be analized in such a way that compressed forms of their representation can be extracted, reducing the amount of memory needed to store them. The Fourier transform have been traditionally used in this way. This dissertation deals with the use of the Karhunen-Loeve transform as an alternative method of representing a particular and dynamic signal, the musical tones, allowing bigger savings of computing resources, as compared to the Fourier transform.

Pour contribuer a elucider la base moleculaire du syndrome oculocerebrorenal de lowe (ocrl), nous avons analyse la region genomique du point de cassure d'une translocation t(x; 3) associee de novo a l'affection. Nous montrons que le yac rs88 contient la region cible. Deux fragments de ce yac ont une sequence conservee chez les mammiferes. Ces fragments n'ont permis de detecter aucun clone positif lors du criblage de trois banques d'adnc. Le yac rs41, chevauchant rs88, a ete utilise directement pour cribler une banque adnc de cristallin bovin, permettant l'identification d'un premier gene au locus ocrl. Ce gene n'est probablement pas implique dans le syndrome ocrl: un transcrit identique aux temoins est present chez tous les patients ocrl analyses, et sur 4,4 kb de sequence transcrite, 3,8 kb sont localises proximalement au point de cassure de la translocation. Au sein de la sequence transcrite un cadre ouvert de lecture presente une sequence peptidique deduite ayant une forme similitude avec les proteines snf2/swi2 et sthi chez s. Cerevisiae, brahma chez d. Melanogaster. Aucune activite snf2 n'a cependant pu etre observee lorsque ce gene est exprime chez s. Cerevisiae. Nous avons ensuite crible une banque adnc de rein humain avec le yac rs88, et identifie un autre gene qui est vraisemblablement le gene du syndrome de lowe: i) sa structure est interrompue par la translocation t(x; 3), ii) le transcrit est absent chez deux patientes porteuses de translocations (x; autosome) associees au syndrome ocrl, iii) le transcrit est absent ou anormal chez neuf patients ocrl sans rearrangement genomique detectable. Un cadre ouvert de lecture dont la sequence peptidique deduite presente une forte similitude avec une inositol polyphosphate 5-phosphatase, suggere que le syndrome de lowe serait du a un defaut du metabolisme des inositol phosphates

ENGLISH ABSTRACT: It has been acknowledged in two recent studies that the translation of key biblical terms is an area that needs urgent attention. Many lexicons provide the meaning of a word by describing its etymology, hardly paying any attention to the socio-cultural contexts within which it is used. Thus, lexicons are often of limited value for Bible interpretation and translation. This dissertation argues that the meaning of a word can only be fully determined by taking into consideration the linguistic and socio-cultural contexts within which it functions. A basic assumption is that the biblical source text serves as a frame of reference for the semantic analysis of a particular word. The text provides an integrative semantic and pragmatic framework within which a biblical term must be investigated with reference to its wider socio-cultural setting. In the light of this framework, this study investigates the meaning of dsj in the book of Ruth from a Lomwe perspective. Although the word occurs only three times (Ruth 1:8, 2:20 and 3:10) with reference to Ruth, Boaz and Yahweh as subjects, respectively, the book is a “dsj story”, which represents the essence of the covenant between Yahweh and His people. The essence of this covenant is demonstrated by the main characters of the story, which unveil the theological depth that dsj brings to the understanding of this narrative. Since the aim of the study is to evaluate the suitability of the terms osivela, osivela combined with woororomeleya and ikharari in relation to others that are potentially available in Lomwe to convey the conceptual complexity denoted by dsj, a Cognitive Frames of Reference (CFR) approach was introduced for the translation. To bridge the cognitive gap between the socio-cultural worlds of the biblical audience and the target audience, the study used different dimensions of CFR namely the textual, socio-cultural, communicational and the organizational frames of reference. Using the book of Ruth as a starting point for the translation of the word dsj into Lomwe, it is argued that this approach offers a better understanding of the meaning of dsj in Ruth 1:8, 2:20 and 3:10. Since osivela waya woororomeleya does not do justice to the meaning of dsj in the three passages, the words ikharari (1:8 and 2:20) and oreera murima (3:10) have been proposed as exegetically and socio-culturally more appropriate alternatives.
AFRIKAANSE OPSOMMING: In twee onlangse studies is aangedui dat daar dringend aandag geskenk behoort te word aan die vertaling van sleutel bybelse terme. Baie woordeboeke verskaf die betekenis van woord deur die etimologie daarvan te beskryf, met beperkte fokus op die sosio-kulturele kontekse waarin dit gebruik word. Gevoglik is die waarde van woordeboeke beperk met betrekking tot bybelinterpretasie en -vertaling. Hierdie proefskrif argumenteer dat die betekenis van woord slegs volkome bepaal kan word deur die inagneming van die literêre en sosio-kulturele kontekste waarin dit funksioneer. Basiese aanname is dat die bybelse bronteks as verwysingsraamwerk dien vir die semantiese analise van bepaalde woord. Die teks verskaf geïntegreerde semantiese en pragmatiese raamwerk waarin bybelse term ondersoek moet word met verwysing na sy breër sosio-kulturele milieu. In die lig van hierdie raamwerk ondersoek hierdie studie dus die betekenis van dsj in die Boek van Rut vanuit Lomwe perspektief. Alhoewel die woord slegs driekeer voorkom (Rut 1:8, 2:20 en 3:10) met betrekking tot onderskeidelik Rut, Boaz en Jahwe as onderwerpe, is die boek “dsj storie” wat die essensie van die verbond tussen Jahwe en sy volk verbeeld. Die wese van dié verbond word gedemonstreer deur die storie se hoofkarakters wat die teologiese diepte van dsj tot beter verstaan van die narratief blootlê. Aangesien die studie evaluering van toepaslike terme osivela, osivela, gekombineer met woororomeleya, en ikharari, in verhouding tot andere wat moontlik in Lomwe beskikbaar is, om die konseptuele kompleksiteit weer te gee, ten doel het, is Kognitiewe Verwysingsraamwerk (KWR) benadering vir vertaling voorgestel. Ten einde die kognitiewe gaping tussen die sosiokulturele wêrelde van die bybelse gehoor en die teikengehoor te oorbrug, het hierdie studie verskillende dimensies van KWR, te wete die tekstuele, sosio-kulturele, kommunikatiewe en organisatoriese verwysingsraamwerke aangewend. Deur die Boek Rut as vertrekpunt te neem vir die vertaling van dsj in Lomwe, word geargumeenteer dat dié benadering beter verstaan van dsj se betekenis in Rut 1:8, 2:20, 3:10 tot gevolg het. Aangesien osivela waya woororomeleya nie reg laat geskied aan die betekenis van dsj in hierdie drie perikope nie, is die woorde ikharari (1:8 en 2:20) en oreera murima (3:10) as eksegeties en sosio-kultureel meer toepaslike alternatiewe voorgestel.

Abstract:
La cytocinèse est la dernière étape de la division cellulaire permettant la séparation de deux cellules filles topologiquement distinctes. L’étape finale de la cytocinèse, coupure physique des deux cellules filles ou abscission, est encore à élucider. Les études de ces dernières années ont permis de mettre en évidence que le trafic intracellulaire ainsi que la composition lipidique de la membrane du pont intercellulaire jouent un rôle essentiel pendant la cytocinèse. Néanmoins, la régulation des lipides par le trafic intracellulaire reste une question ouverte et nécessaire à la compréhension de ce processus clé de la division cellulaire. Nous avons montré que la GTPase Rab35 est requise pour la localisation au pont de cytocinèse de son effecteur, OCRL, une PI(4,5)P2 5-phosphatase impliquée dans une maladie récessive liée à l’X, le syndrome de Lowe. L’inhibition de cette voie entraine une accumulation de PI(4,5)P2 et d’actine filamenteuse au pont de cytocinèse ce qui conduit à l’inhibition de l’abscission. Ainsi, nous avons montré que l’hydrolyse du PI(4,5)P2 est nécessaire à la complétion de la cytocinèse et est régulée par Rab35 et OCRL. En parallèle, nous avons montré que l’inhibition de l’abscission observée après déplétion de Rab35 ou d’OCRL ou dans des cellules issues de patients atteints du syndrome de Lowe peut être corrigée par l’addition de faibles doses de latrunculine A, un agent dépolymérisant l’actine-F. L’ensemble de ces travaux a permis de mettre en évidence un lien nouveau entre la régulation locale, par une GTPase associée à une phosphatase, des niveaux de PI(4,5)P2, du cytosquelette d’actine et des mécanismes régulant les étapes finales de la cytocinèse

Le PI(4,5)P2 est un des principaux régulateurs de la dynamique de l'actine. Entre autres, ce lipide active de multiples complexes impliqués directement dans la polymérisation de l'actine. Comprendre comment est régulée la concentration locale de PI(4,5)P2 dans le temps et l'espace est à cet égard un enjeu biologique important. L'hydrolyse du PI(4,5)P2 est cruciale pour le trafic endosomal et dépend essentiellement de la phosphatase OCRL dans les cellules non-neuronales. Cependant, le mécanisme qui permet le recrutement de cette enzyme sur les vésicules recouvertes de clathrine, précisément après leur scission de la membrane plasmique, est encore méconnu. Nous avons montré que la GTPase Rab35 recrute directement la phosphatase OCRL immédiatement après la scission de la vésicule recouverte de clathrine. De plus, la déplétion de Rab35 ou d'OCRL entraîne l'accumulation de récepteurs internalisés, tels que le CI-MPR, sur des endosomes à la périphérie de la cellule. Ces endosomes, alors anormalement associés à la clathrine, présentent également une accumulation de protéines liées au PI(4,5)P2 (telles que AP-2) ou à l'actine (telles que cortactine ou Arp2/3). Nous avons par ailleurs observé que le recrutement de Rab35 sur les vésicules recouvertes de clathrine suit rapidement le recrutement de DENND1A, la protéine GEF de Rab35 et la disparition d'EPI64B, la GAP de Rab35. Nos résultats montrent que l'activation spatiale et temporelle précise de la GTPase Rab35 déclenche le recrutement de la protéine OCRL sur les endosomes nouvellement formés pour contrôler les niveaux de PI(4,5)P2 et d'actine, et permettre un trafic intracellulaire normal à partir de ces endosomes
PI(4,5)P2 is one of the major regulators of actin dynamics. This lipid activates numerous complexes that are directly involved in actin polymerisation. Therefore, it is a major biological issue to understand how the local concentration of PI(4,5)P2 is regulated in time and space. The hydrolysis of PI(4,5)P2 is crucial for endosomal trafficking and principally depends on the phosphatase OCRL in non-neuronal cells. However, this enzyme is recruited on clathrin-coated vesicles right after their separation from the plasma membrane but the underlying mechanism remained unanswered so far. During my thesis, I have shown that the GTPase Rab35 recruits a phosphatase, OCRL, straight after the separation of the clathrin-coated vesicle from the plasma membrane. The depletion of Rab35 or OCRL leads to an accumulation of internalised receptors such as CI-MPR on peripheral endosomes. These endosomes are abnormally associated with clathrin and also present an accumulation of proteins linked to PI(4,5)P2 (such as AP-2) or to actin (such as cortactin or Arp2/3). Moreover, we observed that the recruitment of Rab35 on clathrin-coated vesicles quickly follows the recruitment of DENND1A, the GEF of Rab35, and the disappearance of EPI64B, the GAP of Rab35. Our results reveal that the precise spatial and temporal activation of GTPase Rab35 triggers the recruitment of the protein OCRL on newly formed endosomes to control the levels of PI(4,5)P2 and actin thus enabling normal intracellular trafficking from these endosomes

Oculocerebrorenal or Lowe Syndrome (OMIM #309000) is an X-linked recessive condition characterized by a triad of congenital cataracts, proximal renal tubular dysfunction, and variable central nervous system involvement. Nearly all affected boys will be hemizygous for a pathogenic variant in the OCRL (NM_000276.4 c.2615delC) gene. We present a clinical and molecular characterization of an extended multiplex family of three affected boys with Lowe Syndrome and describe a novel variant, predicted to be pathogenic, in the OCRL gene. This is to the best of our knowledge the first description of its kind in South African patients and future research into more families with Lowe syndrome will be beneficial.

Abstract:
Lowe syndrome is a rare X linked disorder, characterized by renal, ocular and cerebral defects, caused by mutation in the protein OCRL1. OCRL1 has been implicated in a plethora of cellular functions, and loss of its catalytic conversion of PtdIns(4,5)P2 into PtdIns(4)P is proposed to underly many of the cellular phenotypes associated with lack of OCRL1. The interaction with other proteins such as IPIP27A are also required for the correct function of OCRL1. Renal tubular dysfunction similar to that seen in Lowe syndrome patients is seen in zebrafish models of ocrl1 and ipip27a mutation. Using zebrafish as a model of Lowe syndrome, a reduction of increased PtdIns(4,5)P2 levels in ocrl1-/- embryos is shown to alleviate the renal tubular dysfunction. This demonstrates that targeting PtdIns(4,5)P2 is a viable option for therapeutic treatment of Lowe syndrome. Novel transgenic zebrafish lines are also described, that provide megalin specific, fluorescent and luminescent readouts of proximal tubular endocytic function. These will be an important tool to perform high throughput screens for compounds that alleviate the symptoms of Lowe syndrome. The importance of the binding of IPIP27A to its interaction partners OCRL1 and SH3 containing proteins such as PACSIN2 is demonstrated by rescue of the ipip27a-/- mutant with ipip27a with mutated binding sites. The phenotype of ipip27a-/- mutant embryos is further characterised to demonstrate there is no long term growth defect or defect in tubular polarity, however tubular dilation is seen, suggesting possible mild ciliary defects. In the zebrafish proximal tubule in fish with no functional IPIP27A or OCRL1, a more severe defect in 10 kDa dextran endocytosis is seen, as well as hydrocephaly and curved body axis, cilia impairment related phenotypes. This indicates that IPIP27A and OCRL1 are acting in the same pathway, and therefore depletion of both exacerbates phenotypes. Finally, transgenic lines expressing ubiquitous or pronephric tubule specific fluorescently tagged Rab proteins as markers of membrane compartments in zebrafish are described.

Un territoire est indéniablement associé à Napoléon et à sa légende dans l'esprit des populations. Il ne s'agit pas de la Corse, lieu de naissance, mais bien de Sainte-Hélène, lieu d’exil, de mort mais aussi de construction de la légende napoléonienne. C'est sur cette île de l'Atlantique sud qu'entre 1815 et 1821 celui qui, pendant plus de vingt ans, a fait trembler l'Europe, vit les dernières années de son existence. Le captif pendant toutes les années de sa détention sur Sainte-Hélène n'est peut-être à aucun autre moment de sa vie si étroitement observé. Cependant, les Français, entre 1815 et 1821, ignorent ce qui se passe sur cette île perdue de l’Atlantique sud. Les nouvelles sur ce personnage sont rares sinon absentes et l'absence d'information véridique sur les événements de l'île est considérable. Ce constat est accentué par la situation de Sainte-Hélène. Il s'agit d'un lieu éloigné, inconnu et difficilement compréhensible pour la grandes majorités des Français du XIXe siècle. Cette absence d'information entraine les plus folles rumeurs. Effectivement, les fausses nouvelles parsèment l'exil de Napoléon, des Français répandent des bruits, prétendent connaître la vérité et la transmettre généralement oralement, plus rarement par l'écrit, aux autres. Si les bruits sont essentiellement émis entre 1815 et 1821 ils se poursuivent au cours des années 1820 et même au-delà par de multiples relais.Nombre de bruits et d'ouvrages offrent de multiples histoires toutes plus sensationnelles les unes par rapport aux autres sur l'exil de Napoléon. Parmi elles, Napoléon s'est évadé de Sainte‑Hélène. De véritables faits ont inspiré les nombreuses théories évasionistes qui fleurissent à partir de 1815. Ainsi, des projets d’enlèvement ont indéniablement été pensés. Néanmoins, aucun des plans n’est mis à exécution, amenant en conséquence un questionnement sur les raisons de l’absence de leur concrétisation, n'ont-ils pas simplement été de simples échanges verbaux entre Bonapartistes ?Enfin, les rumeurs perdurent après 1821 en partie grâce aux arts. En effet, l'internement de Napoléon a beaucoup été représenté par la littérature, la peinture et plus récemment par le cinéma. Par exemple, de très nombreux auteurs du XIXe siècle se sont inspirés du séjour de l'Empereur à Sainte-Hélène de façon plus ou moins évidente et ont réinvesti les éléments constitutifs de la légende.Toutes les rumeurs et les représentations artistiques de la captivité déforment les dernières années de Napoléon. Or, ces représentations altérées restent dans la mémoire collective car elles sont plus faciles à retenir, elles sont de meilleures histoires que la réalité... Et donc, la mythologie fini par altérer la réalité historique. Par exemple, l'idée d’un Napoléon humilié en permanence par les Anglais est ancrée dans la mentalité populaire comme parmi les élites. D'ailleurs cette distorsion de la réalité est une partie intégrante de la légende napoléonienne. Sans l'idée du calvaire de Napoléon à Sainte-Hélène prisonnier des puissances de la Sainte-Alliance et l’image négative de Hudson Lowe, la figure du captif auprès des libéraux et des nationalistes au XIXe siècle n’aurait pas été la même. La légende de Sainte-Hélène est donc une partie fondamentale de l’histoire napoléonienne
A territory is undeniably associated with Napoleon and his legend in the minds of the peoples. It is not a question of Corsica, place of birth, but of Saint Helena, place of exile, of death but also of construction of the napoleonic legend. It is on this island of the South Atlantic that between 1815 and 1821 he who, during more than twenty years, made tremble Europe, saw the last years of its existence. The captive during all the years of his detention on St. Helena Napoléon is closely observed. However, the French, between 1815 and 1821, do not know what is happening on this lost island of the South Atlantic. News about this character is rare if not absent and the lack of truthful information about the events of the island is considerable. This observation is accentuated by the situation of St. Helena. It is a distant place, unknown and difficult to understand for the great majority of the French of the nineteenth century. This lack of information leads to the wildest rumors. Indeed, the false news strew the exile of Napoleon, French spread noises, claim to know the truth and to transmit it generally orally, more rarely in writing, to others. If the noises are essentially emitted between 1815 and 1821 they continue during the 1820s and even beyond by multiple relays. Many noises and books offer multiple stories all about Napoleon's exile. Among them, Napoleon escaped from Saint Helena. Real facts have inspired the many evasive theories that flourished from 1815. Thus, kidnapping projects have undeniably been thought out. Nevertheless, none of the plans are put into execution, leading consequently a questioning on the reasons of the absence of their concretization. Maybe, they were simple verbal exchanges between Bonapartists? Finally, rumors endure after 1821 partly thanks to the arts. Indeed, the internment of Napoleon was much represented by literature, painting and more recently by cinema. For example, many nineteenth-century writers drew inspiration from the Emperor's stay in Saint Helena in a more or less obvious way and re-invested the constituent elements of the legend. All the rumors and artistic representations of captivity distort the last years of Napoleon. However, these altered representations remain in the collective memory because they are easier to remember, they are better stories than reality ... And so, mythology ended up altering the historical reality. For example, the idea of ​​a Napoleon permanently humiliated by the British is rooted in the popular mentality as among the elites. Moreover this distortion of reality is an integral part of the Napoleonic legend. Without the idea of ​​Napoleon's ordeal on St. Helena prisoner of the Holy Alliance and the negative image of Hudson Lowe, the figure of the captive among liberals and nationalists in the nineteenth century would not have been the same. Abstract:
Oculocerebrorenal syndrome of Lowe (OCRL) is caused by mutations in a phosphatidylinositol 5-phosphatase, OCRL1, and is believed to lead to an elevation of its preferred substrate, PI(4,5)P2. To date, much of the work on OCRL1 has centred on its role at Golgi and endosomal membranes. However, there is also evidence of plasma membrane activity for OCRL1, where its PI(4,5)P2 substrate is known to be highly abundant. PI(4,5)P2 regulates a wide array of downstream cellular functions such as cytoskeletal dynamics, membrane trafficking and signalling. The tight regulation of PI(4,5)P2 levels and localisation, like other phosphoinositides, provides a framework upon which many of these cellular processes work. In this thesis, effects of OCRL1 loss have been tested through siRNA depletion of OCRL1, focussing where possible on multiple PI(4,5)P2-dependent mechanisms, and also focussing on cells forming polarised epithelia. Firstly, we have visualised the localisation of PI(4,5)P2 in living HeLa cells lacking OCRL1 through immunostaining for Annexin A2, which showed a marked translocation to the plasma membrane. This change in distribution of Annexin A2 suggested that OCRL1 depletion may have an effect on intracellular calcium dynamics as well as PI(4,5)P2 localisation. We also used a GFP-chimera of the well characterised PI(4,5)P2-binding pleckstrin homology domain of PLCδ1. This showed no difference in localisation upon OCRL1 depletion. As OCRL1 is highly enriched at the TGN, we fused the pleckstrin homology domain of PLCδ1 to a mutated pleckstrin homology domain of OSBP known to bind ARF1 at the TGN, to act as a coincidence detector for PI(4,5)P2 at the TGN. This construct also showed no reproducible effect of OCRL1 depletion. Secondly we tested the effect of loss of OCRL1 on cytosolic calcium levels. Using two phospholipase C (PLC) agonists, and a SERCA pump inhibitor, we found no consistent differences in calcium handling upon depletion of OCRL1. Thirdly, we have assessed the potential specialised role that OCRL1 has in polarised epithelial cells, which might relate to the clinical picture in Lowe Syndrome. We found that OCRL1 targets the tight junctions of immortalised lines and primary cells. Through co-immunoprecipitation, we found OCRL1 in complexes with the tight junction scaffold protein ZO-1. Most significantly, we found that depletion of OCRL1 in human polarised epithelial cell lines interfered with epithelial differentiation, reducing cell number and altering morphology, to produce large flat cells. We attribute this phenotype, stronger than any other so far described experimentally, to a defect in tight junction maturation.

Le syndrome de Lowe (LS) est une maladie génétique récessive rare liée à l’X associée à des mutations de perte de fonction du gène OCRL. Elle est caractérisée par une cataracte congénitale, une tubulopathie rénale proximale et des signes neurologiques. La physiopathologie des signes neurologiques n’est toujours pas claire. Ma thèse se concentre sur l’étude des bases cellulaires et moléculaires des signes neurologiques du LS. Nous avons mimé la pathologie in vitro à l’aide des neurones de l’hippocampe de souris où nous avons inhibé l’expression d’OCRL. Nous avons recherché les conséquences de cette perte sur des phénotypes neuronaux de déficience intellectuelle. Nous avons exploré la formation du cil primaire au cours de la différenciation in vitro. Nous avons analysé le rôle d’OCRL1 dans la formation du cerveau cortical de la souris. Nous avons identifié différents phénotypes perturbés par la perte d’expression d’OCRL dans le LS. Les neurones inhibés pour OCRL sont en moyenne plus petits avec moins de branches et une arborisation moins complexe. Ce défaut de dendritogenèse laisse supposer qu’OCRL joue un rôle lors de la dendritogenèse. En étudiant précisément les dendrites, on observe une dérégulation de la formation des épines dendritiques accompagnée d’un défaut de synaptogenèse. Lors de l’inhibition d’OCRL, on constate un défaut de ciliogenèse. Ce défaut de cil primaire se traduit également in vivo par un retard de la migration corticale des neurones inhibés pour OCRL. OCRL aurait donc aussi un rôle dans la ciliogénèse. Ces anomalies constitueraient ainsi des bases cellulaires et moléculaires de la physiopathologie des signes neurologiques de la maladie. Les mutations d’OCRL ont été observées non seulement chez les patients atteints du LS classique, mais également chez des patients plus modérément atteints (quasi absence de signes neurologiques et oculaires), classés Dent-2. Cette situation suggère qu’il existe des différences individuelles dans la capacité de compenser la perte d’OCRL. La seconde partie de ma de thèse explore les mécanismes de compensation existants entre les patients Lowe et Dent-2.Des études dans le laboratoire ont montré que les fibroblastes de patients Lowe et Dent-2 avaient des phénotypes différents et identifié un gène, PLCβ1, comme étant exprimé de façon différente chez les patients Lowe et Dent. PLCβ1 étant impliqué dans les mêmes voies métaboliques qu’OCRL.Nous avons donc étudié, des phénotypes perturbés dans les fibroblastes de patients Lowe et ayant un phénotype moins sévère dans les fibroblastes de patients Dent-2. L’inhibition de PLCβ1 dans des fibroblastes de patients Lowe, nous a permis d'observer que les fibroblastes présentent un « phénotype de type Dent-2 ». De façon réciproque, lorsque l’on surexprime PLCβ1 dans des fibroblastes Dent-2, nous avons constaté que les fibroblastes présentent un « phénotype de type Lowe ». Ces travaux suggèrent que PLCβ1 est un gène modificateur du LS, qui aurait un rôle dans l’hétérogénéité des signes neurologiques associés aux mutations d’OCRL. Mon projet a consisté à évaluer PLCβ1 en tant que gène modificateur du LS. Nous avons testé cette hypothèse, par la modulation de PLCβ1 dans les neurones down régulés pour OCRL. Les neurones à la fois inhibés pour OCRL et PLCβ1, ont des phénotypes quasi similaires aux contrôles pour tous les paramètres que nous avons étudiés précédemment. Les neurones modulés à la fois pour OCRL et PLCβ1 sont en moyenne plus grands avec plus de branches et une arborisation plus complexe que les neurones KO pour OCRL Nous observons une restauration de la densité des épines dendritiques et synaptiques, un réseau d’actine plus dense et une amélioration du défaut de ciliogenèse. L’ensemble de ces résultats confirme l’hypothèse de l’implication de PLCβ1 dans la variabilité phénotypique des signes neurologiques entre le LS et la maladie de Dent-2, donc que PLCβ1 est un gène modificateur du LS
Lowe's syndrome is a rare X-linked recessive genetic disorder associated with loss of function mutations in the OCRL gene. It is characterized by congenital cataract, proximal renal tubulopathy and neurological signs. Currently, the pathophysiology of neurological signs is still unclear. My thesis focuses on the study of cellular and molecular bases of neurological signs of Lowe's syndrome.We began by mimicking the pathology in vitro using neurons from the mouse hippocampus where we inhibited OCRL expression. We investigated the consequences of this loss on neuronal phenotypes reported in various models of intellectual disability. We also explored primary cilia formation during in vitro differentiation. We then analyzed the role of OCRL1 in mouse cortical brain formation using in utero electroporation of shRNA in the brain of mouse embryos.We identified different phenotypes disrupted by the loss of OCRL expression in Lowe's syndrome. Neurons inhibited for OCRL are on average smaller with fewer branches and a less complex arborization. This lack of dendritogenesis suggests that OCRL plays a role in dendritogenesis. By studying precisely the dendrites, we observe a dysregulation of the formation of dendritic spines associated with a defect of synaptogenesis.When OCRL is inhibited, there is a defect in ciliogenesis. This primary cilia defect is also reflected in vivo by a delay in the cortical migration of neurons inhibited for OCRL. Thus OCRL also has a role in neurons ciliogenesis.These abnormalities thus constitute cellular and molecular bases of the pathophysiology of the neurological signs of the disease.OCRL mutations were observed not only in patients with classic Lowe's syndrome, but also in patients with more moderate disease (almost no neurological and ocular signs), classified as Dent-2. Other patients present an intermediate symptomatology between the two pathologies showing the existence of a clinical continuum between Dent-2 disease and Lowe's syndrome. This situation suggests that there are individual differences in the ability to compensate for the loss of enzymatic function. Despite numerous studies, the mechanisms leading to restricted clinical manifestations are still poorly understood.The second part of my thesis explores the compensation mechanisms existing between Lowe and Dent-2 patients.Studies in the laboratory have shown that fibroblasts from Lowe and Dent-2 patients have different phenotypes and identified a gene, PLCβ1, as being differently expressed in Lowe and Dent patients. PLCβ1 being involved in the same metabolic pathways as OCRL. We therefore investigated phenotypes of Lowe patient’s fibroblasts that were less severe in Dent-2 patient’s fibroblasts. Inhibition of PLCβ1 in Lowe patient’s fibroblasts allowed us to observe that the fibroblasts have a "Dent-2 type phenotype". Reciprocally, when overexpressing PLCβ1 in Dent-2 fibroblasts, we found that the fibroblasts exhibit a "Lowe-like phenotype".This work suggests that PLCβ1 is a modifier gene of Lowe's syndrome, and that it has a role in the heterogeneity of the neurological signs associated with OCRL mutations. My project was to evaluate PLCβ1 as a modifier gene of Lowe's syndrome. We tested this hypothesis, by modulating PLCβ1 in neurons downregulated for OCRL. Neurons both inhibited for OCRL and PLCβ1, have phenotypes almost similar to controls for all the parameters we studied previously. The modulated neurons for both OCRL and PLCβ1 are on average larger with more branches and a more complex arborization than the neurons KO for OCRL. We observe a restoration of the density of the dendritic spines and synapses, a denser actin network and an improvement of the ciliogenesis defect.All these results confirm the hypothesis of the involvement of PLCβ1 in the phenotypic variability of neurological signs between Lowe syndrome and Dent-2 disease, and that PLCβ1 is a modifier gene of Lowe's syndrome

La modélisation de systèmes complexes conduit à la résolution numérique de problèmes de très grande taille, par exemple dans le cas de discrétisations par des méthodes d'éléments finis. Une alternative consiste à utiliser des données connues a priori pour réduire la taille du système à résoudre. Etant donnée la solution u d'un problème d'évolution, les méthodes de décomposition orthogonale propre (POD) permettent, à partir de valeurs instantanées u(ti) connues a priori, de construire une base de fonctions sur laquelle on projette le problème : on parle alors de méthodes de POD-Galerkin. Cette méthode a été mise en œuvre notamment dans le cadre de problèmes issus de la mécanique des fluides. On présente une synthèse des méthodes de POD. On montre des résultats de convergence et des estimations dans le cadre parabolique et dans le cadre d'équations générales de la mécanique des fluides. La stabilité de la méthode est prouvée. Des simulations numériques montrent que la taille des systèmes à résoudre est très petite en comparaison des méthodes classiques d'éléments finis. On présente une application intéressante pour la résolution de problèmes de contrôle optimal
Modelling complex systems leads one to solve problems of very high size numerically, for example in case of finite elements discretization. An alternative consists in using a priori known data to reduce the size of the system to solve. Given the solution u of an evolution problem, proper orthogonal decomposition (POD) methods allow one to define a basis of functions from a priori known snapshots u(ti) on which the problem is projected : one speaks of POD-Galerkin methods. This method has been especially implemented in case of fluids mechanics problems. We present a synthesis of POD methods. We show convergence results and estimates in the parabolic case and in the case of general equations in fluids mechanics. The stability of the method is proven. Numerical computations show that the size of systems to solve is very small compared to classical finite elements methods. We present an interesting application to compute the solution of optimal control problems

Cette méthode comprend plusieurs parties : 1) un filtrage par ajustement polynomial ; 2) un traitement du signal (analyse en séries de Fourier) ; 3) une reconnaissance de forme (double transformation de Karhunen-Loeve). La nouvelle méthode de filtrage polynomial par segment sans changement de phase est décrite. L'intérêt du coefficient polynomial de courbure des principales composantes du P. E. S. Est étudié. La transformation de Karhunen-Loeve entraîne une importante réduction des données. La méthode d'analyse compare une série de trois P. E. S. à trois fonctions créneaux (un mobile et deux fixes). Ce traitement est un essai de quantification des latences interpics des P. E. S.

Le gène OCRL1, responsable du syndrome de Lowe de transmission récessive liée à l'X, code pour une PIP2 5-phosphatase associée à l'appareil de Golgi. 107 mutations du gène OCRL1 ont été identifiées chez 146 familles de syndrome de Lowe et 6 mutations ont été identifiées chez 23 patients atteints de maladie de Dent sans mutation du gène CLCN5. L'haplotypage de 18 familles a mis en évidence 3 cas de mosaïcisme germinal et somatique. L'activité PIP2 5-phosphatase fibroblastique des patients est très abaissée par rapport à des fibroblastes normaux. L'analyse par Western blot de la protéine OCRL1 montre une diminution variable de la quantité protéique et pas forcément corrélée à l'activité résiduelle de la protéine. L'analyse des transcrits d'OCRL1 montre qu'il existe une variabilité quantitative chez les patients mais également chez les témoins. Les premières études cliniques chez 55 patients atteints de syndrome de Lowe ne montrent pas de corrélation génotype-phénotype évidente.
