Full textDespite the extensive literature concerning architects of the Modern Movement and debates on the Avant-Garde, several works of architects and artists remain unstudied. The commitments of these individuals contributed to the birth of the Avant-Garde Movement, opening the door for the flourishing of the Modern Movement. This thesis was developed and inspired by the works of two artists, architect Michael Loewe (1871 -1955) and his daughter, the artist Margot Loewe (1905-1974). These artists' work allows us to recognize how these two generations were interwined and contributed the same ideals, as well as how the dialogues between technique and art and between art and industry led to new spaces and new ways of living. This research project was conducted in Berlin, a cultural center of confrontation between tradition and modernity and a city that is in search of a new architectural identity. Berlin represents a major urban and social laboratory that changed from a small provincial capital into a metropolis between 1871 and 1933. This study anlyses this German architect's career, starting from is training and career as a Regierungbaumeister in the Königliche Technische Hochschule of Berlin-Charlottenburg to his architectural accomplishments that were built between 1907 and 1933. The works presented include the buildings of Bismarckstrasse 82-83 (1907), Olivaerplatz 8 (1909), Manfred-von-Richthofen-Strasse 2 (1912), Pariserstrasse 20 (1913), the housing projects of Prausestrasse 5-27 (1920) and of Kuno-Fischerstrasse 3-4-5 (1923), and Pichelwerder (1927). Regarding the works of Michael Loewe prior to World War I, we have identified an initial stage of reaction against the ornamental excess of historical styles and Jugendstil that moved between unadorned designs and geometric simplicity with clear influence from German Neoclassicism . Collaborative works with the prestigious architect-engineer Bruno Möhring were also identified in this stage. In the second stage of hi work in the years between the wars , Michael Loewe envlved and undertook early steps towards a !anguage close to the Neue Sachlichkeit, integrating his project ¡nto the landscape and exploring new technologies and new ways of living that allowed him to express himsell more freely. During his early commitment to Modernity, Michael Loewe's work elicited favorable reactions from supporters of the Neues Bauen, but he was also targeted by the opponents of the New Architecture, whose papers and positions were radicalized, and became the subject of political conflict. In Michael Loewe's work, his professional commitments emerge in favor of the New Objectivity,which raises the issue of the position of art in society and the relationships between art and craft, art and technology, and form and function . Michael Loewe's personal commitments led him to join Existenzminimum proposals, and to experience new ways of living . He encourages his daughter Margot Loewe to study between 1929 and 1933 at the Bauhaus-Dessau under the guidance of Hannes Meyer and Mies van der Rohe. The artistic trajectory of Margot Loewe that is presented corresponds to the brief period between the years of her studies at the Bauhaus-Dessau and her exile. Margot Loewe's work is understood as part of the discussion between utopian Avant-Garde and Bauhaus, as well as its context for dialogue between technology and art, where techlology enabled a new conception of space, which is a common theme in both architecture and art. The technical commitments of Michael Loewe led him to create new formal proposals and as a result of his artistic motivations, he also participated alongside his daughter Margot Loewe in new forms of subjective insight, which created visual impressions and emotions that pushed the architect to experiment with new spaces and led the artist to the abstraction of her painting.

Resumo: Música e poesia. Como as duas artes interagem, que significados são criados, reiterados, modificados e como a performance se constrói e se enriquece a partir desta interação é a reflexão que impulsiona o presente trabalho. Propomos uma análise músico-textual visando a interpretação e performance de quatro Lieder do período Romântico construídos sobre o poema Gretchen am Spinnrade de J. W. Goethe. Serão contempladas as canções dos compositores Louis Spohr (Gretchen), Franz Schubert (Gretchen am Spinnrade), Carl Loewe (Meine Ruh ist hin), e Richard Wagner (Meine Ruh ist hin). Para isso, é feita uma introdução sobre o gênero Lied e sobre o poeta. Em seguida, é proposta uma análise literária do poema, que servirá de base para a interpretação das canções. São abordados aspectos conteudísticos e formais do texto, assim como sua contextualização dentro da peça Fausto I, de que foi extraído. Em seguida, é feita uma análise musical das obras, com foco nas relações texto-música presentes e contemplando aspectos tanto da linha vocal quanto do acompanhamento. Ao longo da análise são feitas sugestões interpretativas baseadas nas relações texto-música
Abstract: Music and Poetry. How the two art forms interact, what meanings are created, emphasized, modified and how the performance is built and enriched from this interaction is the reflection that impels the present work. A text-music analysis aiming towards the interpretation and performance of four Lieder - settings from the Romantic period of the poem Gretchen am Spinnrade from J. W. Goethe is presented here. The settings of Louis Spohr (Gretchen), Franz Schubert (Gretchen am Spinnrade), Carl Loewe (Meine Ruh ist hin), e Richard Wagner (Meine Ruh ist hin) will be approached. After introductory considerations about the genre Lied and about the poet are made, an analysis of the poem is presented, which will ground the interpretation of the songs. Content and formal aspects of the text are considered, as well as its contextualization in the play Faust I, from where it was extracted. Afterwards, a musical analysis of the songs is presented, focusing on the existing text-music relations and taking aspects from the vocal line as well as the accompaniment into consideration. During the analysis interpretative suggestions are made, based on the text-music relations

Full textThe novelty of this approach consists of the use of the Karhunen-Loeve transform to segment the speech and to extract both steady-state and dynamic features. Sequences of stops followed by vowels were segmented with the second Karhunen-Loeve transform coefficient, whereas information about the formant transitions and energy distribution was extracted by means of the basis vectors of the transformed space.
Tests of the proposed rule-based system resulted in 71% correct classification of the stops' place of articulation, and 80% correct classification of the stops' manner of voicing.
More complex systems have previously attained better results. However, the simplicity and computational cost efficiency of this approach would be of considerable benefit to any large ASR system.

Furthermore, signals can be analized in such a way that compressed forms of their representation can be extracted, reducing the amount of memory needed to store them. The Fourier transform have been traditionally used in this way. This dissertation deals with the use of the Karhunen-Loeve transform as an alternative method of representing a particular and dynamic signal, the musical tones, allowing bigger savings of computing resources, as compared to the Fourier transform.

Full textENGLISH ABSTRACT: It has been acknowledged in two recent studies that the translation of key biblical terms is an area that needs urgent attention. Many lexicons provide the meaning of a word by describing its etymology, hardly paying any attention to the socio-cultural contexts within which it is used. Thus, lexicons are often of limited value for Bible interpretation and translation. This dissertation argues that the meaning of a word can only be fully determined by taking into consideration the linguistic and socio-cultural contexts within which it functions. A basic assumption is that the biblical source text serves as a frame of reference for the semantic analysis of a particular word. The text provides an integrative semantic and pragmatic framework within which a biblical term must be investigated with reference to its wider socio-cultural setting. In the light of this framework, this study investigates the meaning of dsj in the book of Ruth from a Lomwe perspective. Although the word occurs only three times (Ruth 1:8, 2:20 and 3:10) with reference to Ruth, Boaz and Yahweh as subjects, respectively, the book is a “dsj story”, which represents the essence of the covenant between Yahweh and His people. The essence of this covenant is demonstrated by the main characters of the story, which unveil the theological depth that dsj brings to the understanding of this narrative. Since the aim of the study is to evaluate the suitability of the terms osivela, osivela combined with woororomeleya and ikharari in relation to others that are potentially available in Lomwe to convey the conceptual complexity denoted by dsj, a Cognitive Frames of Reference (CFR) approach was introduced for the translation. To bridge the cognitive gap between the socio-cultural worlds of the biblical audience and the target audience, the study used different dimensions of CFR namely the textual, socio-cultural, communicational and the organizational frames of reference. Using the book of Ruth as a starting point for the translation of the word dsj into Lomwe, it is argued that this approach offers a better understanding of the meaning of dsj in Ruth 1:8, 2:20 and 3:10. Since osivela waya woororomeleya does not do justice to the meaning of dsj in the three passages, the words ikharari (1:8 and 2:20) and oreera murima (3:10) have been proposed as exegetically and socio-culturally more appropriate alternatives.
AFRIKAANSE OPSOMMING: In twee onlangse studies is aangedui dat daar dringend aandag geskenk behoort te word aan die vertaling van sleutel bybelse terme. Baie woordeboeke verskaf die betekenis van woord deur die etimologie daarvan te beskryf, met beperkte fokus op die sosio-kulturele kontekse waarin dit gebruik word. Gevoglik is die waarde van woordeboeke beperk met betrekking tot bybelinterpretasie en -vertaling. Hierdie proefskrif argumenteer dat die betekenis van woord slegs volkome bepaal kan word deur die inagneming van die literêre en sosio-kulturele kontekste waarin dit funksioneer. Basiese aanname is dat die bybelse bronteks as verwysingsraamwerk dien vir die semantiese analise van bepaalde woord. Die teks verskaf geïntegreerde semantiese en pragmatiese raamwerk waarin bybelse term ondersoek moet word met verwysing na sy breër sosio-kulturele milieu. In die lig van hierdie raamwerk ondersoek hierdie studie dus die betekenis van dsj in die Boek van Rut vanuit Lomwe perspektief. Alhoewel die woord slegs driekeer voorkom (Rut 1:8, 2:20 en 3:10) met betrekking tot onderskeidelik Rut, Boaz en Jahwe as onderwerpe, is die boek “dsj storie” wat die essensie van die verbond tussen Jahwe en sy volk verbeeld. Die wese van dié verbond word gedemonstreer deur die storie se hoofkarakters wat die teologiese diepte van dsj tot beter verstaan van die narratief blootlê. Aangesien die studie evaluering van toepaslike terme osivela, osivela, gekombineer met woororomeleya, en ikharari, in verhouding tot andere wat moontlik in Lomwe beskikbaar is, om die konseptuele kompleksiteit weer te gee, ten doel het, is Kognitiewe Verwysingsraamwerk (KWR) benadering vir vertaling voorgestel. Ten einde die kognitiewe gaping tussen die sosiokulturele wêrelde van die bybelse gehoor en die teikengehoor te oorbrug, het hierdie studie verskillende dimensies van KWR, te wete die tekstuele, sosio-kulturele, kommunikatiewe en organisatoriese verwysingsraamwerke aangewend. Deur die Boek Rut as vertrekpunt te neem vir die vertaling van dsj in Lomwe, word geargumeenteer dat dié benadering beter verstaan van dsj se betekenis in Rut 1:8, 2:20, 3:10 tot gevolg het. Aangesien osivela waya woororomeleya nie reg laat geskied aan die betekenis van dsj in hierdie drie perikope nie, is die woorde ikharari (1:8 en 2:20) en oreera murima (3:10) as eksegeties en sosio-kultureel meer toepaslike alternatiewe voorgestel.

Full textPI(4,5)P2 is one of the major regulators of actin dynamics. This lipid activates numerous complexes that are directly involved in actin polymerisation. Therefore, it is a major biological issue to understand how the local concentration of PI(4,5)P2 is regulated in time and space. The hydrolysis of PI(4,5)P2 is crucial for endosomal trafficking and principally depends on the phosphatase OCRL in non-neuronal cells. However, this enzyme is recruited on clathrin-coated vesicles right after their separation from the plasma membrane but the underlying mechanism remained unanswered so far. During my thesis, I have shown that the GTPase Rab35 recruits a phosphatase, OCRL, straight after the separation of the clathrin-coated vesicle from the plasma membrane. The depletion of Rab35 or OCRL leads to an accumulation of internalised receptors such as CI-MPR on peripheral endosomes. These endosomes are abnormally associated with clathrin and also present an accumulation of proteins linked to PI(4,5)P2 (such as AP-2) or to actin (such as cortactin or Arp2/3). Moreover, we observed that the recruitment of Rab35 on clathrin-coated vesicles quickly follows the recruitment of DENND1A, the GEF of Rab35, and the disappearance of EPI64B, the GAP of Rab35. Our results reveal that the precise spatial and temporal activation of GTPase Rab35 triggers the recruitment of the protein OCRL on newly formed endosomes to control the levels of PI(4,5)P2 and actin thus enabling normal intracellular trafficking from these endosomes

Full textA territory is undeniably associated with Napoleon and his legend in the minds of the peoples. It is not a question of Corsica, place of birth, but of Saint Helena, place of exile, of death but also of construction of the napoleonic legend. It is on this island of the South Atlantic that between 1815 and 1821 he who, during more than twenty years, made tremble Europe, saw the last years of its existence. The captive during all the years of his detention on St. Helena Napoléon is closely observed. However, the French, between 1815 and 1821, do not know what is happening on this lost island of the South Atlantic. News about this character is rare if not absent and the lack of truthful information about the events of the island is considerable. This observation is accentuated by the situation of St. Helena. It is a distant place, unknown and difficult to understand for the great majority of the French of the nineteenth century. This lack of information leads to the wildest rumors. Indeed, the false news strew the exile of Napoleon, French spread noises, claim to know the truth and to transmit it generally orally, more rarely in writing, to others. If the noises are essentially emitted between 1815 and 1821 they continue during the 1820s and even beyond by multiple relays. Many noises and books offer multiple stories all about Napoleon's exile. Among them, Napoleon escaped from Saint Helena. Real facts have inspired the many evasive theories that flourished from 1815. Thus, kidnapping projects have undeniably been thought out. Nevertheless, none of the plans are put into execution, leading consequently a questioning on the reasons of the absence of their concretization. Maybe, they were simple verbal exchanges between Bonapartists? Finally, rumors endure after 1821 partly thanks to the arts. Indeed, the internment of Napoleon was much represented by literature, painting and more recently by cinema. For example, many nineteenth-century writers drew inspiration from the Emperor's stay in Saint Helena in a more or less obvious way and re-invested the constituent elements of the legend. All the rumors and artistic representations of captivity distort the last years of Napoleon. However, these altered representations remain in the collective memory because they are easier to remember, they are better stories than reality ... And so, mythology ended up altering the historical reality. For example, the idea of a Napoleon permanently humiliated by the British is rooted in the popular mentality as among the elites. Moreover this distortion of reality is an integral part of the Napoleonic legend. Without the idea of Napoleon's ordeal on St. Helena prisoner of the Holy Alliance and the negative image of Hudson Lowe, the figure of the captive among liberals and nationalists in the nineteenth century would not have been the same. The legend of St. Helena is therefore a fundamental part of Napoleonic history

Full textLowe's syndrome is a rare X-linked recessive genetic disorder associated with loss of function mutations in the OCRL gene. It is characterized by congenital cataract, proximal renal tubulopathy and neurological signs. Currently, the pathophysiology of neurological signs is still unclear. My thesis focuses on the study of cellular and molecular bases of neurological signs of Lowe's syndrome.We began by mimicking the pathology in vitro using neurons from the mouse hippocampus where we inhibited OCRL expression. We investigated the consequences of this loss on neuronal phenotypes reported in various models of intellectual disability. We also explored primary cilia formation during in vitro differentiation. We then analyzed the role of OCRL1 in mouse cortical brain formation using in utero electroporation of shRNA in the brain of mouse embryos.We identified different phenotypes disrupted by the loss of OCRL expression in Lowe's syndrome. Neurons inhibited for OCRL are on average smaller with fewer branches and a less complex arborization. This lack of dendritogenesis suggests that OCRL plays a role in dendritogenesis. By studying precisely the dendrites, we observe a dysregulation of the formation of dendritic spines associated with a defect of synaptogenesis.When OCRL is inhibited, there is a defect in ciliogenesis. This primary cilia defect is also reflected in vivo by a delay in the cortical migration of neurons inhibited for OCRL. Thus OCRL also has a role in neurons ciliogenesis.These abnormalities thus constitute cellular and molecular bases of the pathophysiology of the neurological signs of the disease.OCRL mutations were observed not only in patients with classic Lowe's syndrome, but also in patients with more moderate disease (almost no neurological and ocular signs), classified as Dent-2. Other patients present an intermediate symptomatology between the two pathologies showing the existence of a clinical continuum between Dent-2 disease and Lowe's syndrome. This situation suggests that there are individual differences in the ability to compensate for the loss of enzymatic function. Despite numerous studies, the mechanisms leading to restricted clinical manifestations are still poorly understood.The second part of my thesis explores the compensation mechanisms existing between Lowe and Dent-2 patients.Studies in the laboratory have shown that fibroblasts from Lowe and Dent-2 patients have different phenotypes and identified a gene, PLCβ1, as being differently expressed in Lowe and Dent patients. PLCβ1 being involved in the same metabolic pathways as OCRL. We therefore investigated phenotypes of Lowe patient’s fibroblasts that were less severe in Dent-2 patient’s fibroblasts. Inhibition of PLCβ1 in Lowe patient’s fibroblasts allowed us to observe that the fibroblasts have a "Dent-2 type phenotype". Reciprocally, when overexpressing PLCβ1 in Dent-2 fibroblasts, we found that the fibroblasts exhibit a "Lowe-like phenotype".This work suggests that PLCβ1 is a modifier gene of Lowe's syndrome, and that it has a role in the heterogeneity of the neurological signs associated with OCRL mutations. My project was to evaluate PLCβ1 as a modifier gene of Lowe's syndrome. We tested this hypothesis, by modulating PLCβ1 in neurons downregulated for OCRL. Neurons both inhibited for OCRL and PLCβ1, have phenotypes almost similar to controls for all the parameters we studied previously. The modulated neurons for both OCRL and PLCβ1 are on average larger with more branches and a more complex arborization than the neurons KO for OCRL. We observe a restoration of the density of the dendritic spines and synapses, a denser actin network and an improvement of the ciliogenesis defect.All these results confirm the hypothesis of the involvement of PLCβ1 in the phenotypic variability of neurological signs between Lowe syndrome and Dent-2 disease, and that PLCβ1 is a modifier gene of Lowe's syndrome

Full textModelling complex systems leads one to solve problems of very high size numerically, for example in case of finite elements discretization. An alternative consists in using a priori known data to reduce the size of the system to solve. Given the solution u of an evolution problem, proper orthogonal decomposition (POD) methods allow one to define a basis of functions from a priori known snapshots u(ti) on which the problem is projected : one speaks of POD-Galerkin methods. This method has been especially implemented in case of fluids mechanics problems. We present a synthesis of POD methods. We show convergence results and estimates in the parabolic case and in the case of general equations in fluids mechanics. The stability of the method is proven. Numerical computations show that the size of systems to solve is very small compared to classical finite elements methods. We present an interesting application to compute the solution of optimal control problems
