Abstract:
In prospective studies, increased levels of cardiolipin-biding antibodies and autoantibodies to oxidized low-density lipoprotein (LDL) have been observed in patients with myocardial infarction (MI). These findings suggest that antiphospholipid antibodies may contribute to the development of MI. The ‘oxidative-modification hypothesis’ in the pathogenesis of atherosclerotic heart disease is based on the oxidation of LDL, its accumulation into arterial wall, and the development of chronic inflammation in the atheroma. Evidence of enhanced lipid peroxidation and its association with antiphospholipid antibodies has been recently reported in SLE patients. There is also epidemiological data showing a remarkably increased risk of MI in SLE. In this review, the role of different types of antiphospholipid antibodies in the development of atherosclerotic heart disease is evaluated with particular attention to their potential pathogenic mechanisms and the possibilities in the prevention of MI associated with antiphospholipid antibodies.

Abstract:
Arazyme and extracts of soy leaves (ESLs) are used as ingredients for functional foods; however, their combined administration has not been studied. This study assessed the combined effect of Arazyme and ESLs in high-fat-diet (HFD)-induced obese C57BL/6J mice fed 2 mg/kg Arazyme, 50 mg/kg ESLs, or a combination of 2 mg/kg Arazyme and 50 mg/kg ESLs by oral gavage for 13 weeks. Individually, Arazyme and ESLs had no effect on the HFD-induced phenotypes. The combination of Arazyme and ESLs significantly suppressed body weight gain, improved glucose and insulin tolerance, and suppressed hepatic steatosis by reducing lipid synthesis and enhancing lipid utilization gene expression. Furthermore, the combination significantly reduced HFD-induced plasma bile acid reabsorption by suppressing bile acid transporter expression, including the ATP biding cassette subfamily B member 11 (Abcb11), solute carrier family 10 member 1 (Slc10a1), Slc10a2, Slc51a, and Slc51b in the liver and gut. Moreover, the combination of Arazyme and ESLs significantly prevented HFD-induced islet compensation in the pancreas. These results suggest that the incorporation of Arazyme combined with ESLs reduces HFD-induced body weight, hyperglycemia, and hepatic steatosis by regulating liver–gut bile acid circulation in HFD-fed mice. This combination can markedly reduce treatment doses and enhance their therapeutic effects, thereby reducing therapeutic healthcare costs.

Abstract:
Antigens extracted using ethanol (EtOH) and incorporated in the EtOH vortex ELISA (EVELISA) test have previously shown high specificity and sensitivity for detecting Mycobacterium avium subspecies paratuberculosis (Map) and M. bovis infections in cattle. The objective of this study is to define the components present in the EtOH extract. We show that this extract is composed of lipid, carbohydrate, and proteins on the surface of the bacilli, and that EtOH removes the outer layer structure of Map which comprise these elements. To identify proteins, polyclonal antibodies to the EtOH prep were produced and used to screen a Map genomic expression library. Seven overlapping clones were identified with a single open reading frame, MAP_0585, common to all. MAP_0585, which encodes a hypothetical protein, was recombinantly produced and used to demonstrate strong reactivity in sera from hyperimmunized rabbits, but this protein is not strongly immunogenic in cattle with Johne’s disease. A panel of monoclonal antibodies was used to determine the presence of additional proteins in the EtOH extract. These antibodies demonstrated that a well-known antigen, termed MPB83, is present in M. bovis EtOH extracts and a fatty acid desaturase (MAP_2698c) is present in Map EtOH extracts, while lipoarabinomannan was common to both. The lipid and carbohydrate components of the extract were analyzed using thin layer chromatography and lectin binding, respectively. Lectin biding and protease treatment of the EtOH extract suggest the antigenic component is carbohydrate and not protein. These results give further insight into this important antigen prep for detecting mycobacterial diseases of cattle.

Abstract Objectives To evaluate the effect of selected bioactive compounds from cocoa shell and its aqueous extract on the activation of fibroblast growth factor 21 (FGF21) signaling in hepatocytes and the resulting modulation of lipid and glucose metabolism, oxidative stress, and mitochondrial function. Methods The bioactive compounds from cocoa shells were extracted using water and characterized by UPLC-MS/MS. An in vitro model of non-alcoholic fatty liver disease was used: HepG2 cells were co-treated with either pure theobromine (TH), protocatechuic acid (PCA), procyanidin B2 (PB2), epicatechin (EPI), catechin (CAT) (10–50 µmol L−1), or the aqueous extract from cocoa shells (CAE, 20–100 µg mL−1) in the presence or absence of palmitic acid (PA, 500 µmol L−1). FGF21 receptor interaction with cocoa shell phytochemical was evaluated in silico. Biomarkers of cell metabolism were assessed 24 h after the co-treatment in cell supernatants and lysates using chemical, biochemical, and immunochemical techniques. Results Phytochemicals from cocoa shell were able to interact in silico with FGF21 receptor (biding energies from −6.0 to −11.0 kcal mol−1) denoting a potential interaction causing FGF21 receptor response. FGF21 hepatic signaling was activated (1.1 to 2.3-fold enhance in ERK1/2 phosphorylation). CAE, TH, PCA, PB2, EPI, and CAT diminished PA-elicited lipid accumulation (5–86%, P < 0.05) by reducing fatty acid synthase expression, and stimulated lipolysis (1–87%, P < 0.05) through the up-regulation of lipoprotein lipase expression. CAE and therein-bioactive compounds, mainly TH and PCA, promoted glucose uptake (4–35%, P < 0.05) via the increase in the phosphorylation of the insulin receptor (2.2-fold, P < 0.05), protein kinase B (2.8-fold, P < 0.05), AMPKα (2.0-fold, P < 0.05), and ERK1/2 (2.8-fold, P < 0.05) phosphorylation. Oxidative stress was prevented (50–100%, P < 0.05), and the loss of mitochondrial content (15–100%, P < 0.05) and function, measured as ATP production, alleviated (28–100%, P < 0.05). Conclusions Results demonstrated that the major bioactive compounds, primarily TH and PCA, found in cocoa shell could activate FGF21 signaling and regulated hepatic lipid and glucose metabolism, preventing insulin resistance, oxidative stress, and mitochondrial dysfunction. Abstract Introduction: Childhood obesity is one of the most important public health issues of the 21st century. Epidemiological studies have suggested that obesity during childhood increases the risk of developing comorbidities, such as type 2 diabetes, later in life. Childhood obesity is a complex disease whose molecular mechanisms are not completely elucidated. In this context, a system biology approach could contribute to the scientific knowledge regarding genetic factors related to childhood obesity onset. Aim: To identify molecular mechanisms involved in childhood obesity by implementing a system biology approach. Methods: Experimentally validated and computationally predicted genes related to Pediatric Obesity (C2362324) were downloaded from the DisGeNET v7.0 database. The protein-protein interaction (PPI) network was constructed using the STRING v11.0 database and analyzed using NetworkAnalyst v3.0 and Cytoscape v3.8.1. The relevance of each node for the network structure and functionality was assessed using the degree method to define hub genes. Functional and pathway enrichment analyses were performed based on Gene Ontology (GO) terms and KEGG Pathways. Results: The search on the DisGeNET database retrieved 191 childhood obesity-related genes. The PPI network of these genes showed 19 hub genes (STAT3, SIRT1, BCL2, IRS1, PPARG, SOCS3, TGFB1, HDAC4, DNMT1, ADCY3, PPARA, NEDD4L, ACACB, NR0B2, VEGFA, APOA1, GHR, CALR, and MKKS). These hub genes were involved in biological processes of lipid storage / kinase activity, regulation of fatty-acid metabolic processes, regulation of pri-miRNA transcription by RNA polymerase II, and negative regulation of small molecules and carbohydrate metabolic processes. In terms of molecular functions, repressing of transcription factors biding was found enriched. Regarding KEGG Pathways, the hub genes are involved with adipocytokine signaling, insulin resistance, longevity regulation, and cytokine signaling pathways. Conclusion: Our approach identified 19 hub genes, which are highly connected and probably have a key role in childhood obesity. Moreover, functional enrichment analyses demonstrated they are enriched in several biological processes and pathways related to the underlying molecular mechanisms of obesity. These findings provide a more comprehensive information regarding genetic and molecular factors behind childhood obesity pathogenesis. Further experimental investigation of our findings may shed light on the pathophysiology of this disease and contribute to the identification of new therapeutic targets.

Abstract. Due to low toxicity to nontarget species and rapid degradation after its application, organophosphate (OP) remains a widely used class of pesticide. Suicidal or accidental overdose of OP can result in acute tubular necrosis. Experimental evidence shows little correlation between the renal tubular necrosis and the degree of OP-induced acetylcholinesterase inhibition, the main mechanism of OP's toxicity, suggesting the involvement of alternate mechanisms. Since reactive oxygen species (ROS) are known mediators of many toxin-induced renal injuries, this study was conducted to investigate whether ROS play a role in Bidrin® (BD)-induced renal tubular epithelial cell (LLC-PK1) toxicity. BD is an OP insecticide formulation with dicrotophos as the active ingredient. LLC-PK1 cell death, determined by lactate dehydrogenase (LDH) release (% of total), rose concentration- and time-dependently after exposure of the cells to 1000, 1250, 1500, 1750, and 2000 ppm of BD for 6, 12, 24, and 48 h. Antioxidants 2-methylaminochroman (2-MAC; 0.3 to 2.5 μM) and desferrioxamine (DFO; 0.25 to 2 mM) reduced cell damage induced by 1250 ppm of BD over a 24-h incubation in a concentration-related manner. The greatest reductions in % LDH were produced by DFO 2 mM and 2-MAC 2.5 μM, both significantly lower than BD alone. H2O2 levels (μmol/mg protein per h) were significantly elevated after exposure to 1250 ppm of BD. Significantly increased malondialdehyde formation (nmol/mg protein) compared with control was also found in BD-exposed cells indicating enhanced lipid peroxidation. Malondialdehyde generation was significantly suppressed by 2-MAC and DFO. These results demonstrate that the organophosphate BD can cause direct tubular cytotoxicity, and implicate, at least in part, a role for ROS and accompanying lipid peroxidation in cytotoxicity. Based on these direct in vitro findings, it is hypothesized that, besides hypotension that often accompanies OP intoxication, OP-induced oxidative stress at the tubular level may play a role in the pathogenesis of acute tubular necrosis.

The aim of the research was to know the capability of actinomycetes isolate from Bangka Belitung, which stored at Microbiology-LIPI Culture Collection, in producing chitinase enzyme. This isolate which could produce chitinolitic enzyme, signed by clear zone at medium contain 1% chitine. The chitinase activity of the isolate which incubated for 1-7 days in the room temperature was analyzed by spectrophotometer in λ 584 nm. The result of this experiment was highest chitinase activities with incubated for 3 days, were 1.66 . 10-2 U/ml. Maximum chitinase activities was found at 1% starch soluble substrate 2.83 . 10-2, pH 8.0 and at 50°C condition were 9.3 . 10-2 and 12.98 .10-2 U/ml respectively.Key words : chitinase, clear zone, spectrophotometer

Abstract:
Sorgum (Sorghum bicolor L.) merupakan tanaman serealia yang potensial untuk dibudidayakan dan dikembangkan, khususnya pada daerah-daerah marginal dan kering di Indonesia. Pengetahuan tentang karakterisasi fisiologis pada fase awal pertumbuhan pertumbuhan tanaman sangat penting, terkait dengan pemenuhan air dan hara untuk aktivitas fisiologis dan efisiensi pemanfaatan air pada tanaman sorgum. Penelitian ini bertujuan untuk mengetahui pola pertumbuhan 15 aksesi sorgum, khususnya pada fase vegetatif pertumbuhan awal sebagai dasar pengetahuan pada fase pertumuhan dan perkembangan berikutnya. Penelitian dilaksanakan di rumah kaca Pusat Penelitian Bioteknologi LIPI Cibinong dengan 15 aksesi sorgum yang ditanam Januari hingga Februari 2019. Penanganan hasil panen di Laboratorium Fisiologi Stres dan Konservasi Benih Bidang Botani Pusat Penelitian Biologi LIPI. Analisis kuantitatif, dilakukan berdasarkan Rancangan Acak Kelompok dengan 15 nomor aksesi sorgum dan 3 kali ulangan. Analisis data dilakukan dengan menyajikan perbandingan rata-rata hasil perolehan data ke dalam bentuk tabel dan histogram, terdiri dari pengamatan hasil pengamatan dan pengukuran. Dari keseluruhan data pertumbuhan meliputi data tinggi tanaman, jumlah daun, kadar klorofil,dan potensial daun dari 15 aksesi sorgum selama 3 MST, dapat dibandingkan keseluruhan data yang diperoleh. Dapat disimpulkan dari beberapa data pengukuran, aksesi yang dinilai kurang baik adalah aksesi Kawali dengan nilai yang selalu terendah dari seluruh aksesi pada semua parameter pengukuran. Sedangkan aksesi yang dapat dinilai cukup baik adalah Buleleng Empok (berdasarkan data tinggi tanaman dan jumlah daun), Suri 4 (berdasarkan data tinggi tanaman, jumlah daun dan potensial air daun), dan Jagung Rote (berdasarkan data tinggi tanaman, jumlah daun, dan kadar klorofil daun).
