Dr. Walter McAfee surmounted the racism endemic to twentieth-century America and made unique and enduring contributions to the scientific community during his 42 years working as a government scientist in Monmouth County, New Jersey, to include critical contributions to Project Diana, which allowed man’s first “contact” with the moon in 1946. He also made time to quietly battle injustice, and teach and mentor a new generation of innovators and leaders as a professor at Monmouth College (now University) in West Long Branch, a trustee at Brookdale Community College in Lincroft, and an organizer of enrichment programs for high school students. While the communities to which he contributed so much have memorialized him in different ways, the general public knows too little about his accomplishments. We must ensure that the stories of diverse pioneers like Walter McAfee are not excluded from our understanding of the history of the Garden State.

Abstract:
Oral submucous fibrosis (OSF) is a premalignant disorder and persistent activation of myofibroblasts is implicated in this pathological progression. Increasing attention has been addressed towards non-coding RNA-regulated myofibroblasts activities and the effects of phytochemicals on non-coding RNA modulation are of great importance. In the present study, we examined the anti-fibrosis property of α-mangostin, a xanthone isolated from the pericarp of mangosteen. We found that α-mangostin exhibited inhibitory potency in myofibroblast activities and expression of fibrosis markers at the concentrations that caused neglectable damage to normal cells. Apart from the downregulation of TGF-β1/Smad2 signaling, we found that α-mangostin attenuated the expression of long non-coding RNA LincROR as well. Our results demonstrated that the effects of α-mangostin on myofibroblast activation were reverted when LincROR was overexpressed. Additionally, we showed the expression of LincROR in OSF specimens was elevated and silencing of LincROR successfully attenuated myofibroblast characteristics and TGF-β1/Smad2 activation. Taken together, these findings indicated that the anti-fibrosis effects of α-mangostin merit consideration and may be due to the attenuation of LincROR.

ABSTRACT Glioblastoma multiforme (GBM) is the most aggressive malignant brain tumor. One of the reasons for the resistance of MGB to treatment is the extreme heterogeneity of the tumor and, in particular, the presence of cancer stem cells (CSCs) in the population of glioblastoma cells. In this work, we investigated the effect of conditions that reduce the proportion of CSCs in the GBM cell population on the levels of long noncoding RNAs (lincROR and MALAT1) involved in the formation of the phenotype of glioblastoma cancer stem cells. We have shown that culturing under conditions that cause a decrease in cell stemness (when fetal calf serum is added to the culture medium) affected the content of these transcripts: in the cells of most of the analyzed lines, a decrease in the level of the positive stemness regulator lincROR and an increase in the content of MALAT1 were noted.

Abstract:
Abstract Emerging evidence have suggest that patients with metastatic prostate cancer (PCa) will become resistant after receiving docetaxel chemotherapy, but the specific regulatory mechanism is still unclear. LincROR is an important oncogenic lncRNA which plays an important role in regulating tumor carcinogenesis and metastasis; however, the underlying mechanism of lincROR functioning in the docetaxel resistance process of PCa remains largely unknown. In the present study, we found that lincROR is highly expressed in docetaxel-resistant PCa cell lines and was associated with poor docetaxel response in metastatic PCa patients. By using loss- and gain-of-function experiments revealed that lincROR promotes PCa cells growth and docetaxel resistance in vitro and in vivo. Mechanistic studies demonstrated that lincROR specifically interacts with and stabilizes MYH9 protein, which enhances β-catenin/hypoxia-inducible factor 1-alpha (HIF-1α) pathways. Besides, HIF-1α could bind with the promoter region of lincROR to activate its transcription, thus forming the lincROR/MYH9/HIF-1α positive feedback loop. Moreover, lincROR could be packaged into exosomes in an hnRNPA1-dependent manner and then disseminated chemoresistance phenotype to receipt cells. Overall, our study provides evidence supporting exosome-mediated lincROR activates the β-catenin/HIF-1α positive feedback loop by targeting MYH9 protein, which may be exploited for anticancer therapy. Implications: Our findings suggest that targeting hypoxia stress and chemoresistance for therapeutic purposes, and lincROR could promote the improvement of treatment responses in DTX-resistant PCa patients.
