Caenorhabditis elegans est un organisme modèle couramment utilisé pour étudier les processus de développement tels que la formation de la vulve. Ce processus repose sur l'induction précise des cellules qui donnent naissance à la vulve. Les destins cellulaires résultants sont organisés en un motif caractéristique, qui est conservé tout au long du Caenorhabditis et des nématodes rhabditides. En contraste avec cette conservation des cellules vulvaires, ma thèse présente deux axes d'étude qui ont révélé des signatures d'évolution des voies de développement contrôlant l'induction vulvaire. Dans le premier, je présente un changement des motifs cis-régulateurs nécessaires à l'expression cellulaire spécifique de lin-3 (EGF), un gène codant pour la molécule de signalisation qui induit le destin des cellules de la vulve. Les trois sites de liaison aux facteurs de transcription requis pour l'expression de lin-3 chez C. elegans ne sont observés que chez les espèces du groupe Elegans. En utilisant des techniques de FISH à molécule unique et d'édition de génome, je montre que le remplacement des sites endogènes de C. elegans par un fragment de 58 pb contenant un seul site (provenant de C. angaria) produit des niveaux d'ARNm de type sauvage de type lin-3. Je conclus que l'évolution compensatoire pour l'exigence de sites de liaison de C. elegans s'est produite en cis. Dans la deuxième partie de mon étude, j'ai utilisé une approche de cartographie par séquençage et une collection de mutants de la vulve chez Oscheius tipulae, voisin de C. elegans dans le clade rhabditid, afin d'identifier les mutations dans les gènes agissant sur la spécification du modèle de destin des cellules de la vulve. La seule mutation collectée pour rendre les animaux sans vulve correspond à une délétion de la région cis-régulatrice contrôlant l'expression de O. tipulae lin-3, ce que j'ai confirmé par l'édition du génome de CRISPR / Cas9. Les mutants présentant un excès d'induction de la vulve portent des lésions moléculaires dans la voie plexin / semaphorin, un système de signalisation cellulaire qui, selon moi, agit dans le positionnement antéropostérieur des cellules donnant lieu à la vulve par analyse microscopique. Enfin, d’autres animaux d’O. tipulae dont le destin de cellules vulvaires est perturbé présentent des mutations dans des composants de la voie Wnt; pour cette raison, j'ai étudié les profils d'expression des ligands Wnt d'O. tipulae et révélé leur conservation avec C. elegans. Ces deux études mettent en évidence le processus de dérèglement du système de développement (DSD) en tant que processus évolutif qui se produit lors de la formation de la vulve sur des nématodes rhabditides<br>Caenorhabditis elegans is a model organism commonly used to study developmental processes such as vulva formation. This process relies on the precise fate induction of the cells that give rise to the vulva. The resultant cellular fates are arranged into a characteristic pattern, which is conserved across the vulva development of Caenorhabditis and rhabditid nematodes. In contrast to this extensive conservation of the vulval cell fate pattern, my thesis presents two lines of study that revealed signatures of evolution in developmental pathways controlling vulval induction. In the first one, I present a turnover of cis-regulatory motifs required for the cell-specific expression of lin- 3 (EGF), a gene coding for the signalling molecule which induces vulva cell fates. The three transcription factor binding sites required for lin-3 expression in C. elegans are observed only in species of the Elegans group. Using single-molecule FISH and genome editing techniques, I show that the replacement of the endogenous C. elegans binding sites by a 58 bp fragment containing a single site (coming from C. angaria), produces wild-type mRNA levels of lin-3. I conclude that compensatory evolution for the requirement of C. elegans binding sites occurred in cis. In the second line of study, I used a mapping-bysequencing approach and a collection of vulva mutants in Oscheius tipulae, a neighbour of C. elegans in the rhabditid clade, to identify mutations in genes acting on vulva cell fate pattern specification. The only collected mutation to render animals vulvaless corresponds to a deletion of the cis-regulatory region controlling the expression of O. tipulae lin-3, which I confirmed by CRISPR/Cas9 genome editing. Mutants with an excess of vulva induction carry molecular lesions in the plexin/semaphorin pathway, a cell signalling system which I show to act in the antero-posterior positioning of the cells giving rise to the vulva by microscopic analysis. Finally, other O. tipulae animals with disrupted vulval cell fate pattern hold mutations in components of the Wnt pathway; for this reason, I investigated the expression profiles of Wnt ligands of O. tipulae and revealed their conservation with C. elegans. Connues pour leur forte teneur en huile, oméga 3, antioxydants et fibres, les graines de lin sont de plus en plus demandées par les industries. D’où l’importance de contrôler leurs productions. Le but de ce travail est d’étudier l’effet des paramètres environnementaux et de la variété sur la composition de la graine de lin et sur le procédé ultérieur de pressage. Pour cela dix variétés de lin oléagineux ont été cultivées sur deux sites différents (Picardie et centre de la France) au cours des années 2011, 2012 et 2013. L’étude a révélé un fort effet variétal sur les teneurs en huile, C18:3, amidon et le PMG tandis que le lieu de culture n’a affecté que la teneur en lipides et amidon. L’effet année a quant à lui été marqué pour le PMG, les teneurs en protéines, lipides et C18:3. Le pressage des graines précédemment caractérisées a montré une forte corrélation entre teneur protéines et rendement. Afin d’améliorer le processus d'expression, le dépelliculage des graines de lin avant le pressage a été envisagé. Ainsi, l’étude du pressage des graines de lin sur presse hydraulique et presses continue montre la possibilité du pressage des graines dépelliculées. Une concordance entre les performances de pressage à l’échelle laboratoire et à l’échelle pilote a été obtenue pour les graines entières et dépelliculées ce qui laisse présager une transposition possible vers l’échelle industrielle<br>Flaxseed is known for their high oil content, omega 3, antioxidants and fiber. To meet growing demand, this requires controlling their production. The aim of this work is to study the effect of environmental factors and cultivar on linseed composition and subsequent pressing process. Ten linseed cultivars were grown in two different locations (north and south of France) during years 2011, 2012 and 2013. Every year have been characterized in terms of pluviometry, sunshine hours and degree days, each climatic factor has been cumulated from flowering to harvest. A strong impact of cultivar on oil, C18:3 and starch content has been noticed whereas the growing location has only affected the starch and lipid contents. Year effect has been recorded on thousand kernel weight, proteins, oil, C18:3 and starch content. Oil expression using the seeds previously characterized has exhibited a strong correlation between protein content and oil yield. To improve the pressing process, dehulling flax seeds before pressing was considered. The study of the pressing flax seeds on hydraulic and continuous press shows the possibility of pressing the dehulling flaxseeds. The concordance between the pressing performance at the laboratory scale and pilot scale was highlighted for the whole and dehulled seeds. This could be suggests a possible transposition to the industrial scale

The class B synMuv genes encode many proteins known to remodel chromatin and repress transcription. The class A synMuv genes likely also function in transcription, although their specific molecular functions are unknown. We have identified a class A synMuv mutation in the promoter of lin-3 EGF, revealing that lin-3 is the key biological target of the class A synMuv genes in vulval development. Using FISH with single mRNA molecule resolution, we found that class AB synMuv double mutants exhibit widespread ectopic lin-3 expression. Our results show that lin-3 EGF is normally expressed in the germline, and many class B synMuv genes have previously been implicated in inhibiting germline fates in somatic cells. We propose that the class A synMuv genes specifically repress ectopic lin-3 EGF expression through a site in the lin-3 promoter and the class B synMuv genes either directly or indirectly repress lin-3 as a consequence of their role in regulating the germline/soma distinction. The class A and B synMuv genes had previously been thought of as two parallel pathways, but we have found that each of those pathways is actually composed of multiple parallel pathways. While class AB synMuv double mutants have a strong Muv phenotype, most class AA synMuv double mutants exhibit a weak Muv phenotype, and most pairs of class B synMuv mutants can enhance each other in sensitized backgrounds, indicating that most genes within a class can function in parallel. We also found that some pairs of synMuv genes cannot act in parallel, indicating that they function together to repress ectopic lin-3 expression. We also report the molecular characterization of the class A synMuv gene lin-38 and the identification of mcd-1 as a class A synMuv gene. lin-38 and mcd-1 encode paralogous zinc-finger proteins. Unlike previously studied class A synMuv genes that function specifically in vulval development by repressing lin-3, both lin-38 and mcd-1 control multiple aspects of development by regulating target genes other than lin-3.<br>by Adam M. L'huile de lin est considérée comme une des plus riches en acide linolénique (oméga- 3), avec certaines variétés de lin oléagineux produisant jusqu'à 65% d'acide a-linolénique. Cette huile utilisée depuis longtemps en industrie chimique pour la confection de peintures, vernis, agents tensioactifs, présente aussi des bénéfices pour la santé humaine avec un apport journalier d'environ 2 g.f 1 (diminution de la pression artérielle, prévention de thrombose...). Il parait donc raisonnable aujourd'hui d'augmenter la production de l'huile de lin riche en oméga-3 pour répondre à ces différents besoins. La sélection de cultivars producteurs d'huile de la qualité demandée devient impérative. Pour cela, des connaissances sur les mécanismes de synthèse et d'accumulation AG polyinsaturés (PUFA) dans les graines de lin sont à acquérir. Dans ce contexte, nous avons évalué le rôle des enzymes de type LPAAT dans l'accumulation des PUFA dans le but de sélectionner des isoformes compétitives dans leur accumulation au niveau des TAG. Les gènes Lpaat ont été isolés à partir des graines de lin âgées de 20 JAF, phase active de remplissage en huile. La fonctionnalité des protéines correspondantes a été testée par complémentation dans une souche JC201 d’E. coli déficiente en activité LPAAT. Une caractérisation de ces enzymes a été effectuée par des tests in vitro et in vivo. Les résultats ont montré la présence d'une isoforme, nommée LPAAT2A, dont l'activité LPAAT présente une spécificité et sélectivité élevées vis-à-vis des PUFA. Dans un 2ème temps, nous avons mesuré in vitro et in vivo le potentiel des enzymes LPAAT provenant de litchi et de lin vis-à-vis des AG de type cyclopropanique. Ces AG possèdent des propriétés physico-chimiques intéressantes pour l'industrie des lubrifiants et des cosmétiques et sont produits par une enzyme spécifique « la CFA synthase ». Nous avons généré des lignées d'Arabidopsis exprimant la CFA synthase d’E.coli puis co-exprimé le gène Lpaat de litchi. Une augmentation de la teneur en AGC (AG cycliques) dans les lignées transgéniques obtenues reflète la spécificité de la LPAAT de litchi vis-à-vis des AGC. Nous nous sommes aussi intéressés aux enzymes de types sPLA2a (phospholipase A2). L'isolement du gène correspondant a été effectué à partir des graines de lin pendant la phase active de remplissage en huile. La caractérisation de son activité a été effectuée par des tests in vitro et in vivo. Les résultats ont montré que l'expression du gène sP/a2a de lin dans les graines d' Arabidopsis provoque une augmentation du poids de la graine ainsi qu'une augmentation du contenu en acide linolénique dans les TAG. L'ensemble de ces résultats montre l'existence d'un système enzymatique endogène chez le lin efficace vis-àvis des PUFA et qui semble stimuler le métabolisme lipidique une fois exprimé chez Arabidopsis<br>Flaxseed oil contain high amount of oméga-3 and present different industrials applications and human health benefits. ln this work, we aimed to identify enzymes allowing the accumulation of high level of omega-3 in linseed plants. ln this context, we evaluated the role of Lysophophatidic acid acyltransferases (LPAAT) and Phospholipases A2 (PLA2) enzymes in the accumulation of omega-3 by in vitro and in vivo approaches. Results showed the presence of a LPAAT2A isoform into flax genome having a high specificity and selectivity toward omega-3. Expression of Lpaat2A and sPia2a gene in Arabidopsis seeds increase seed weight, oil production and omega-3 content (up to 10% and 11%) in transformant seeds respectively. These results showed the presence in linseed plant of an efficient enzymatic system toward omega-3 accumulation. Furthermore, we have evaluated in vitro and in vivo the potential of Litchi and flax LPAAT enzymes toward the production of cyclopropane fatty acids (CFA). These fatty acids are naturally produced by a specific enzyme «CFA synthase» with physico-chemical properties interesting to lubricant and cosmetics industries. ln this context, we have generated Arabidopsis lines expressing E. coli Cfa synthase with or not co-expression of litchi Lpaat gene. Result showed an increase of 25% in the content of CFA in transgenic line co-expressing Cfa synthase and litchi Lpaat comparing to the transgenic lines expressing the CFA synthase. This increase in the level of CFA in transgenic seeds reflects the specificity of Litchi LPAAT toward CFA

Reading Brexit through BrexLit is a dissertation centred on the 2016 referendum on British exit from the European Union and on Brexit Literature (‘BrexLit’). The thesis focuses on the relation between Englishness and the Leave vote, analysing themes and images at the core of the Brexit campaign in parallel with selected literary works published both before and after the political outcome, and on the impact of the referendum on literature itself.
After an introduction that serves as a framework, the work is divided into three different sections. The first one is dedicated to an analysis of Brexit from a political point of view, outlining global tendencies and British peculiarities that influenced the vote and explaining Brexit as an English (more than British) phenomenon. The second section moves from the political aspects of the study to the literary ones, showing that Brexit themes are to be found in contemporary fictions published before the referendum and focusing on two novels that are particularly concerned both with the structures of feeling at the core of the campaign and with English identity: Speak for England (2005) by James Hawes and The White Family (2002) by Maggie Gee. The third and last section introduces the reader to Brexit Literature (‘BrexLit), which is a new sub-genre that has flourished from the referendum onwards. The section looks at the main characteristics of the sub-genre, distinguishing between two main categories of BrexLit fictions: political and intimate novels. This sections ends with the close reading of two BrexLit novels published in 2018, Perfidious Albion by Sam Byers and Middle England by Jonathan Coe, which are the most representative in relation to the two categories mentioned above and in the way they engage, as the novels of the previous section, with the structures of feeling exploited in Brexit discourse.

Lo studio affronta il problematico argomento della ciclicità in relazione a due grandi lirici tedeschi, Stefan George e Friedrich Hoelderlin, misurandosi con una vasta tradizione critica e con non agevoli dettati ermeneutici. Il lavoro è scandito in sette capitoli, dei quali il primo analizza il termine 'ciclo', partendo dal suo significato etimologico di cerchio, circolo e ruota, per poi soffermarsi sul significato di successione di eventi memorabili all'interno di un'epoca. Il ciclo è inteso quale categoria estetico-semantica che, incardinata nella dimensione riflessiva del linguaggio, permette di leggere le liriche sia come singoli testi autonomi sia come testi poetici inseriti in un insieme più vasto. Nel secondo capitolo si analizza la nozione di rituale ciclico in Stefan George nel contesto della Germania guglielmina, lacerata dalla frattura fra Bourgeoise e Bildungsbuergertum. Da questo prospettiva il rituale ciclico georgeano è concepito come mito che mira a creare una tradizione culturale funzionale ai bisogni della Germania, legittimandone i caratteri di specificità, eccellenza ed esclusività rispetto alle altre nazioni, nonché esorcizzando lo stereotipo tedesco di "verspaetete Nation", condannata a restare in uno stato di frammentazione territoriale fino al 1871. Il mito carente, capace di tradurre in realtà una volontà di potenza di stampo nietzscheano, viene incarnato dal poeta come esponente di una realtà potenziata che si colloca in un punto di scissura tra finito e infinito. Fondandosi sulla ripetizione di un'azione, il rituale estetico garantisce stabilità e continuità e offre un efficace antidoto contro la fugacità del tempo, mentre l'espressione poetica diviene una totalità in sé conchiusa. In Stefan George il culto della bellezza genera un cerchio magico, nel quale egli si trasforma da esteta in vate che preconizza la palingenesi dell'umanità. Oggetto di indagine sono state, a titolo di esempio, le strutture cicliche di Der Teppich des Lebens und die Lieder von Traum und Tod mit einem Vorspiel. Infine il lavoro volge a contemplare il rapporto tra Friedrich Hoelderlin e Stefan George, a partire dall'analisi del contributo di George e dei suoi sodali per la riscoperta dell'opera tarda di Hoelderlin per poi estendersi alle fonti utilizzate nel processo ermeneutico della poesia georgeana, nonché alle analogie tematiche rintracciabili tra i due poeti tedeschi che mantengono inalterata la propria singolarità sul piano stilistico e linguistico. Ciò avviene attraverso l'analisi di testi esemplari come gli hoelderliniani Fragment philosophischer Briefe e Verfahrungsweise des poetischen Geistes oltre agli inni Patmos, Friedensfeier, Brot und Wein e Der Zeitgeist e le loro ricadute entro l'opera georgeana, a fronte dello specifico paradigma della ciclicità.

La presente tesi si occuperà di indagare il bilinguismo, precisamente nella fascia d’età comprensiva 0-3 anni, e di dimostrarne i vantaggi, eliminando in questo modo l’ombra dei vari pregiudizi che hanno spesso circondato questo tema. In particolare, grazie ad un’analisi degli aspetti neurologici e biologici riguardanti sia il cervello infantile ed il suo sviluppo, sia i processi d’acquisizione (monolingue/bilingue) delle lingue del bambino, verranno evidenziati quei fattori fondamentali e le corrispettive variabili che rendono possibile, e spesso in modo più spontaneo e naturale, un’acquisizione bilingue precoce. Poste le premesse a livello neurologico e biologico, di conseguenza a quanto analizzato, successivamente la tesi si sposterà su un piano prettamente pratico, riguardante la dimensione educativa che sottostà al successo di un’acquisizione bilingue. Verrà posta l’attenzione principalmente sulla realtà dell’asilo nido, una struttura che svolge il compito fondamentale di offrire al bambino un adeguato programma di educazione bilingue, come di garantire una stimolazione ed un supporto sufficiente a raggiunge questo fine. Alla luce di ciò, il presente elaborato indagherà ed analizzerà le migliori caratteristiche e modalità che rendono tale un ottimo programma bilingue, soffermandosi singolarmente sul ruolo dell’educatore/insegnante, l’ambiente in cui il bambino è immerso, e le varie attività a cui egli viene sottoposto.

The goal of this dissertation is to study the infrared absorption spectrum of the C 3 (carbon three) molecule. In particular, the aim is to investigate its ground electronic state up to 12:500 cm-1, as this affects the atmospheres of cool C-rich stars. The linear C 3 molecule shows very unusual properties for a linear molecule: a high degree of floppiness, no permanent dipole moment and a strong bentstretch interaction. Consequently, the C 3 spectrum presents particular features such as overtones, hot bands, and, as has been recently detected in Carbon stars and molecular clouds, a quite low fundamental bending frequency (63 cm-1) when in the ground electronic state. This dissertation aims to address each of these features. The first section discusses the context for this work: the stars. It provides a brief introduction about the Astrophysics related to this research project. A review at the recent literature is provided and the experimental results which provide the goal for the results of the theoretical work in the rest of the dissertation are set out. The second section introduces the C 3 molecule and outlines its properties. Previous C 3 studies are discussed and the theoretical approach used to study ro-vibrational spectra of triatomic molecules is set out. Preparatory tests and calculations are carried out to allow a theoretical reproduction of C 3 roto vibrational spectrum in the infrared region to be produced. The third part of the thesis expands on the nuclear motion calculations of section 2 and presents the results of the large scale calculations performed using the DVR3D suit programs written by Tennyson et al. [1]. This program allows the calculation of energy levels, wave-functions, expectation values and Einstein Coefficients. It takes as input the Potential Energy Surface (PES) and a Dipole Moment Surface (DMS) constructed a priori (in section 2) by solving the electronic problem within the Born-Oppenheimer's approximation. Because the quality of the PES sets the accuracy of the ro-vibrational calculations tests on different C 3 PESs and DMSs are performed. To reproduce accurate spectra of cool stars atmosphere in the temperature range of 2000 - 4000 K it was necessary perform calculations with high rotational quantum number. For this reason, tests with J>>0 were necessary to optimize the DVR3DRJZ parameters in order to guarantee a certain degree of accuracy and energy levels convergence. The results of these calculations and associated C 3 line-lists should be very useful to support the observations and model atmospheric studies. Les acides linoléique (LA) et α-linolénique (ALA) sont deux acides gras polyinsaturés. Ils sont indispensables à l’Homme car nous sommes incapables de les synthétiser in situ. Ces deux acides gras sont les représentants des oméga-6 (LA) et oméga-3 (ALA). Bien qu’ils soient indispensables et donc nécessaires, l’enjeu du ratio oméga-6/oméga-3 entre ces deux familles est également important à prendre en compte dans l’alimentation. Les habitudes de consommation des Français, ces trente dernières années, ont fait évoluer la balance en défaveur des oméga-3 par rapport aux oméga-6. Or un rééquilibre de la balance oméga-6/oméga-3 tend à diminuer les risques cardiovasculaires. Les huiles de lin et de cameline sont des cultures endémiques en France extrêmement riches en ALA. Elles sont les plus adaptées à une complémentation en oméga-3 d’origine végétale. Cependant, les hautes teneurs en acides gras polyinsaturés de ces huiles sont responsables de leurs faibles stabilités oxydatives. La consommation d’huiles ou de graisses oxydées n’est pas recommandée car elle peut apporter des molécules toxiques. Ainsi l’enjeu de ce projet était d’améliorer la durée de conservation d’huiles riches en acides gras polyinsaturés en valorisant les antioxydants extraits des tourteaux obtenus après trituration des graines. Parmi les molécules étudiées, l’acide gallique, l’acide caféique, le gallate de propyle ainsi que les pinorésinol et laricirésinol purifiées du lin ont montré un réel intérêt pour ralentir l’oxydation, tandis que le sécoisolaricirésinol s’est montré inefficace<br>Linoleic (LA) and α-linolenic (ALA) acids are two polyunsaturated fatty acids. They are essential for humans because we are unable to synthesize them in situ. These two fatty acids are the representatives of omega-6 (LA) and omega-3 (ALA). Although they are essential and therefore necessary, the issue of the omega-6 / omega-3 ratio between these two families is also important to take into account in the diet. The consumption habits of the French over the past thirty years have shifted the balance against omega-3s compared to omega-6s. Or a rebalancing of the omega-6 / omega-3 balance tends to reduce cardiovascular risks. Linseed and camelina oils are endemic crops in France extremely rich in ALA. They are the most suitable for supplementation with omega-3 of plant origin. However, the high contents of polyunsaturated fatty acids in these oils are responsible for their low oxidative stabilities. The consumption of oxidized oils or fats is not recommended because they can contribute toxic molecules. The aim of this project was to improve the shelf life of oils rich in polyunsaturated fatty acids by enhancing the antioxidant extracts of the cakes obtained after crushing the seeds. Among the molecules studied, gallic acid, caffeic acid, propyl gallate as well as pinoresinol and lariciresinol purified from flax showed a real interest in slowing down oxidation, while secoisolariciresinol was ineffective

Nós conjecturamos que a única 2-álgebra de Lie simples de este tipo é W(1, 3). Assim, nosso principal objetivo é verificar a veracidade desta conjectura para estas álgebras de pequenas dimensões. Como resultados, provamos que esta conjectura é certa para todas estes álgebras de dimensão menor ou igual a 16, e também em alguns casos especiais quando a dimensão é 17.<br>In this work we study the simple Lie 2-algebras of finite dimension, and toral rank 3 over an algebraically closed field characteristic 2. We surmise that the only simple Lie 2-algebra of this type is W(1, 3). So, our main objective is to study the truthful of this conjecture for these algebras of small dimensions. As a result, we prove that this conjecture is true for all these algebras less than or equal to 16 dimension, and also in some special cases when the dimension is 17.

Paul Auster’s latest novel, 4 3 2 1, is one among a number of recent works of fiction presenting multiple alternate versions of a character’s life. It can, therefore, be placed in productive dialogue with two similar, though different in many respects, contemporary novels exploring plural counterfactual realities, namely Philip Roth’s The Counterlife (1986) and Kate Atkinson’s Life After Life (2013). By projecting complex ontologies in which the distinction between actual and non-actual events is often blurred, these texts investigate the inextricable link between truth and imagination, and factuality and possibility, characterizing a life story. Thus, a great part of their appeal lies in their evocation, by “unnatural” means, of a most “natural” and deeply human experience, namely the impression that life cannot be reduced to a single, linear, and consistent course of events. At the same time, all three novels inevitably represent cognitive challenges for readers striving to navigate their impossible textual universes, as their juxtaposition of incompatible scenarios jars both with our real-world experience and with general readerly expectations for a coherent narrative development. By combining a cognitive approach with concepts drawn from possible worlds theory and unnatural narratology, this thesis aims to highlight the complexities of a reader’s journey through the multiple worlds of Auster’s 4 3 2 1, Roth’s The Counterlife, and Atkinson’s Life After Life.

Ett förslag är att lina 3 ska flyttas så den kommer efter lina 1 och före lina 2.</p><p> </p><p>Syftet med arbetet var att ta fram en flödesanalys över vilka montage som krockar flödesmässigt vid en förflyttning av lina 3. En undersökning av olika möjligheter att tilta/vrida hytten gjordes för att hitta en bra arbetsergonomi åt montören. En flödesanalys över vilka montage som krockar vid en förflyttning av lina 3 togs fram med hjälp av processflödesanalys. I slutet av arbetet gjordes ett förslag över vilka montage som bör flyttas till de olika monteringslinorna. Med de förslag som gjorts angående hur man kan vrida hytten, samt vilka montage som ska flyttas till diverse monteringslinor kan man genomföra en omorganisation av lina 3.</p>

The MBTA Green Line, a Boston light rail system serving nearly 230,000 average weekday passengers, will be undergoing major changes in the next two decades, including nearly doubling of ridership and system expansion. In order to prepare for these changes, measures need to be taken to increase capacity and plan for operations on the new segment. Starting in Fall 2010, the MBTA added to a 2-car train operation, and subsequently increased, a number of 3-car trains on three of the four Green Line branches, in order to begin to address the capacity issue. This thesis analyzes service performance before and after implementation of 3-car trains to find that although scheduled capacity increased slightly, the actual capacity of the system remained constant during the morning peak period and decreased during the evening peak period. Furthermore, there were some negative impacts with respect to passenger waiting time and running times, thus worsening the overall passenger experience. However, since 3-car trains will be required for increasing capacity on the Green Line, it is recommended that trials of 3-car trains continue, with the restriction that only 2- or 3-car trains operate on a branch. Furthermore, field observations at terminal stations on two of the branches show differences in operations management practices, which help explain some variability in service along the route, and point to strategies to improve service reliability.<br>by Alexandra A. The double branched cover is a construction which provides a link between problems in knot theory and other questions in low-dimensional topology. Given a knot in a 3-manifold, the double branched cover gives a natural way of associating a 3-manifold to the knot. Similarly, the double branched cover of a properly embedded surface in a 4-manifold is a 4-manifold whose boundary is the double branched cover of the boundary link of the surface. Consequently, whenever a link in S^3 bounds certain types of surfaces, its double branched cover will bound a 4-manifold of an appropriate type. The most familiar situation in which this connection is used is the application to slice knots as the double branched cover of a smoothly slice knot is the boundary of a smooth rational ball. Examples of 3-manifolds which bound rational balls can therefore easily be constructed by taking the double branched covers of slice knots while obstructions to a 3-manifold bounding a rational ball can be interpreted as slicing obstructions. This thesis is primarily concerned with two different extensions of this idea. Given a closed, orientable 3-manifold, it is natural to ask whether it admits a smooth embedding in the four-sphere $S^4$. Examples can be obtained by taking the double branched covers of doubly slice links. These are links which are cross-sections of an unknotted embedding of a two-sphere in S^4. Certain links can be shown to be doubly slice via ribbon diagrams with appropriate properties. Other embeddings can be obtained via Kirby calculus. On the other hand, many obstructions to a 3-manifold bounding a rational ball can be adapted to give stronger obstructions to embedding smoothly in S^4. Using an obstruction based on Donaldson's theorem on the intersection forms of definite 4-manifolds, we determine precisely which connected sums of lens spaces smoothly embed. This method also gives strong constraints on the Seifert invariants of Seifert manifolds which embed when either the base orbifold is non-orientable or the first Betti number is odd. Other applicable methods, also based on obstructions to bounding a rational ball, include the d invariant from Ozsvath and Szabo's Heegaard-Floer homology and the Neumann-Siebenmann mu-bar invariant. These are used, in conjunction with some embedding results derived from doubly slice links, to examine the question of when the double branched cover of a 3 or 4 strand pretzel link embeds. The fact that the double branched cover of a slice knot bounds a rational ball has a second interpretation in terms of knot concordance. In this viewpoint, the double branched cover gives a homomorphism from the concordance group of knots to the rational cobordism group of rational homology 3-spheres. This can be extended to a concordance group of links using a notion of concordance based on Euler characteristic. This yields link concordance groups which contain the knot concordance group as a direct summand with an infinitely generated complement. The double branched cover homomorphism extends to large subgroups containing the knot concordance group.

Previous issue date: 2013<br>Resumo: A invasão do endométrio pelo trofoblasto extraviloso é fundamental no desenvolvimento do feto e da placenta, processo este controlado por fatores ligados à atividade imunológica e hormonal que, quando alterada, pode resultar em interrupção da gestação e/ou geração das chamadas doenças trofoblásticas gestacionais. Em algumas situações, pode haver evolução para o coriocarcinoma, neoplasia maligna do trofoblasto, em que há evidências da atuação das moléculas ligadas ao processo de fusão celular e inflamação. Porém, os estudos neste tema são incipientes e inconclusivos. Considerando essas informações, o objetivo deste trabalho é estudar de forma comparativa a expressão das citocinas LIF, IL-6 e do ativador de transcrição STAT-3, além da telomerase, em material de aborto, de placenta normal a termo e de coriocarcinoma. Métodos: a expressão destas moléculas foi avaliada pelos métodos: imunoistoquímica (IHQ), imunofluorescência (IF), Western Blotting (WB) e Real-Time PCR (RT-PCR), em amostras de material de aborto, placenta normal a termo e coriocarcinoma (N=12 cada um). Os ensaios de WB e Real-Time PCR empregaram material a fresco de placenta normal a termo e seu cultivo celular e cultura da linhagem BeWo. Resultados: no material de aborto, as reações de IHQs evidenciaram expressão moderada de IL-6 em 58,4% dos casos e intensa de STAT-3 em 33,3%. Na placenta normal, observou-se intensa marcação de IL-6 em 50% e de STAT- 3 em 16,7% dos casos, enquanto que, no coriocarcinoma, houve expressão intensa de IL-6 em 50% e de STAT-3 em 75% dos casos. Por outro lado, as reações para LIF tiveram expressão nula em todos os três grupos. Pelo WB houve expressão proteica de IL-6 apenas no material fresco de placenta normal e ausência de expressão na sua cultura primária e na linhagem BeWo; LIF não foi expresso em todos os grupos estudados. STAT-3 foi detectado no citoplasma em todos os grupos, entretanto, a expressão nuclear da STAT-3 fosforilada (pSTAT-3) não foi observada na IF e nem pelo WB. Na análise gênica pelo RTPCR houve forte expressão de IL-6 e STAT-3 no material fresco de placenta normal e expressão muito fraca na cultura primária de placenta normal e na linhagem BeWo; a expressão de LIF foi muito fraca em todos os grupos. Apenas a linhagem BeWo demonstrou forte expressão gênica da telomerase, contrastando com a completa falta de expressão no material fresco de placenta normal e em sua cultura primária. Conclusão: A intensa expressão IHQ de IL-6 e STAT-3 no coriocarcinoma indica a atuação de ambas na carcinogênese. A expressão proteica de IL-6 no material fresco de placenta normal e sua ausência no material de cultura primária e na linhagem BeWo pode ser ocasionado pelo contato célula-a-célula nas culturas aderentes, inibindo o crescimento celular e, consequentemente, as vias de sinalização. A falta de expressão da pSTAT-3 tanto na IF como por WB demonstra que a via JAK-STAT está sendo desativada. A ausência de expressão de LIF, em todos os métodos estudados, sugere que esta citocina poderia estar sendo inibida por meio de proteínas SOCS3 ou, atuando, de modo indireto, na proliferação celular do coriocarcinoma. O aumento da atividade da telomerase nas células BeWo reforça sua relação com o fenótipo maligno e a aponta como um bom marcador para progressão da doença<br>Abstract: The invasion of the endometrium by extravillous trophoblast is a fundamental process in the growth of the fetus and placenta. The process is controlled by factors related to the immune and hormonal activity that, when changed, may result in termination of pregnancy and development of so-called gestational trophoblastic diseases. In some cases, changes can result in malignancy, in which some molecules play a role in cell fusion process and inflammation, although studies in this area are inconclusive. Considering this information, the study had the aim of investigating the expression of cytokines LIF, IL-6, STAT- 3 and the function of telomerase to understand their participation in abortion, in normal at term placenta and choriocarcinoma. Methods: The expression of the molecules was assessed by immunohistochemical assay (IHC), immunofluorescence (IF), Western Blotting (WB) and Real-Time PCR (RT - PCR) using fixed material from biopsies of abortions, normal at term placentas and choriocarcinoma along with fresh tissue of normal at term placenta and their primary culture and BeWo cell line. Paraffin embedded material used in IHC and IF assays were obtained from the Department of Pathology files. Tests of WB and Real-Time PCR employed fresh material, obtained from cell cultures of normal at term placenta and the BeWo line. Results: IHC reactions to abortion biopsies showed moderate staining for IL-6 in 58.4% of cases and intense for STAT-3 in 33.3 % of cases. In biopsies of normal placenta, there was intense reaction for IL-6 in 50% of cases, intense for STAT-3 in 16.7%; choriocarcinoma showed intense staining for IL- 6 in 50% of cases and also for STAT-3 in 75% of cases. On the other hand, LIF expression was missing in all three groups. WB analyses showed IL-6 protein in fresh material from normal placentas, but no expression in placenta primary cultures and BeWo line. LIF was absent in all groups. Cytoplasmic STAT-3 was observed in all groups, while the nuclear expression of phosphorylated STAT-3 was absent. On gene analyses a strong expression of IL-6 and STAT- 3 was observed from fresh normal placenta, but very weak expression in primary cultures of normal placenta and BeWo cell line. LIF expression was very weak in all groups. In regard to the gene expression of telomerase, it was strong in the BeWo line which contrasted with its complete lack of expression in fresh normal placenta and its primary culture. Conclusion: The high expression of IL-6 and STAT-3 in biopsies of choriocarcinoma indicates the role of both in tumor progression. Regarding protein expression, the presence of IL-6 in the material from fresh normal placenta, and its absence in primary culture and BeWo line may be caused by the cell-to-cell contact cultures by inhibiting cell growth and thus signaling pathways. However, the lack of expression of phosphorylated STAT-3 whether through IF or WB shows that its JAK-STAT pathway is inhibited. Lack of expression of the LIF suggests that it might be involved indirectly in choriocarcinoma cell proliferation or be inhibited by SOCS3 protein. Dans la première partie nous présentons l'algèbre de Weyl et nos résultats en ce qui concerne ses sous-algèbres de Lie de dimension finie. La deuxième partie est consacrée à une structure algébrique plus exotique, l'algèbre de Lie d'ordre 3. Nous posons les bases d'une théorie des déformations et contractions de ces structures algébriques. Nous choisissons après une telle algèbre de Lie d'ordre 3 qui étend d'une manière non-triviale, différente de la supersymétrie, l'algébre de Poincaré. Nous nous intéressons à la construction d'un modèle de théories des champs (la supersymétrie cubique ou la 3SUSY) basée sur cette algèbre. Dans ce but nous obtenons des multiplets bosoniques avec lesquels nous construisons des Lagrangiens invariants. Nous étudions la compatibilité entre cette nouvelle symétrie et la symétrie de jauge abelienne. L'analyse des termes d'interactions possible montre que ceux-ci ne sont pas permis par la 3SUSY. Finalement nous établirons les résultats de l'extension en dimension arbitraire de notre modèle<br>In the first part we present the Weyl algebra and our results concerning its finite-dimensional Lie subalgebras. The second part is devoted to a more exotic algebraic structure, the Lie algebra of order 3. We set the basis of a theory of deformations and contractions of these algebraic structures. We then concentrate on a particular such Lie algebra of order 3 which extends in a non-trivial way the Poincaré algebra, this extension being different of the supersymmetric extension. We then focus on the construction of a field theoretical model based on this algebra, the cubic supersymetry (3SUSY). For this purpose we obtain bosonic multiplets with whom we construct invariant Lagrangians. We then study the compatibility between this new symmetry and the abelian gauge symmetry. Furthermore, the analyse of possible interactions shows that interactions terms are not allowed by the 3SUSY invariance. Finally we establish results regarding the extension in arbitrary dimensions of our model

CLIC is an international study of a future multi-TeV electron-positron linear collider, where the energy of a high-intensity drive beam is extracted and transferred to the main beam via Power Extraction and Transfer Structures (PETS) in the form of rf power. The study of power production is therefore essential for the feasibility of CLIC. Power production in PETS has been studied, and experiments have been performed in the decelerator Test Beam Line in the CLIC Test Facility 3.In particular, the correlation of the power production and the beam position inside the structure has been studied. It is shown that the total produced power is constant when the beam has a position offset through the PETS. In addition, the difference between the measured phases from each side is independent of the beam position, which allows for efficient combination of the fields. However, the ratio of the power on each side of the PETS unexpectedly shows a linear dependence on the horizontal offset, with a correlation value of 0.87. This can potentially affect the power transferred to the main linac, and should be taken into account in the design of the high power rf system.A graphical user interface was developed for the Test Beam Line, and the functionality is described in detail. The program is used in the operation of the Test Beam Line for monitoring, matching, steering and power production experiments, and has interfaces to MAD-X and PLACET. A documentation is also given of the Test Beam Line and the methods of the power measurements.

For instance, cellular phones with high technology incorporating Bluetooth, Infrared, Digital Camera, Games, GPS, Internet Browsing, etc., need high performance rechargeable batteries. Lithium ion-conducting glass-ceramics with NASICONtype structure, based on the Li1+xMxM'2-x(PO4)3 system, where M = Al, Cr, Fe, Ga and M' = Ge, Ti, Hf, are considered good candidates as solid electrolytes in lithium ion batteries due to their chemical stability, easy fabrication and high ionic conductivity. In this work, the Li1+xAlxTi2-x(PO4)3 glass composition was synthesized by the traditional method known as Splat Cooling. Glass-ceramics with the same composition as the precursor glass but with different microstructures were obtained by controlled crystallization, using single and double heat treatments. The parent glass was characterized by chemical and thermal analysis and the latter enabled tailoring of the crystallization heat treatments. X-ray diffraction (XRD) results indicated that the resulting glassceramics exhibit the desired NASICON-type structure as primary phase, as well as the segregation of insulating phases, i.e., AlPO4 and/or TiO2, which do not affect the electrical properties. An examination of the microstructure by scanning electron microscopy (SEM) and measurements of electrical conductivity by impedance spectroscopy (IS), combined with the XRD analysis, indicated that the electrical conductivity of single heat-treated samples increases with the heat treatment temperature and hence with the increase in crystallinity. Samples synthesized by double heat treatment showed increasing electrical conductivity with decreasing nucleation treatment time and consequently with increasing average grain size.<br>Os avanços tecnológicos em dispositivos portáteis trazem outro aspecto que chama a atenção, tanto ou mais importante do que os próprios dispositivos desenvolvidos, i.e., a necessidade de fontes de alimentação mais poderosas. Por exemplo, telefones celulares com tecnologia de ponta que incorporam Bluetooth, Infravermelho, Câmara fotográfica, Jogos, GPS, Navegação na Internet, etc., requerem baterias recarregáveis de alto desempenho. Vitrocerâmicas condutoras por íon lítio com estrutura tipo NASICON, concretamente baseadas no sistema Li1+xMxM 2-x(PO4)3, onde M = Al, Cr, Fe, Ga e M = Ge, Ti, Hf, são consideradas boas candidatas como eletrólitos sólidos em baterias de íon lítio devido à sua estabilidade química, facilidade de fabricação e alta condutividade iônica. Neste trabalho foi obtido o vidro de composição Li1.2Al0.6Ti1.6(PO4)2.9 usando o método tradicional de resfriamento rápido conhecido como Splat Cooling. Vitrocerâmicas de composição igual à do vidro matriz, com diferentes microestruturas, foram obtidas por cristalização controlada mediante tratamentos térmicos simples e duplos. O vidro precursor foi caracterizado por análise química e térmica. Esta última permitiu projetar os tratamentos térmicos de cristalização. Os resultados de difração de raios X (DRX) mostraram a obtenção da estrutura tipo NASICON como fase principal, com presença das fases isolantes AlPO4 e/ou TiO2, que não afetam as propriedades elétricas. O estudo da microestrutura e da condutividade elétrica, usando microscopia eletrônica de varredura (MEV) e espectroscopia de impedância (EI), respectivamente, evidenciaram o aumento da condutividade com o crescimento do tamanho médio do grão, atribuído ao maior contato entre grãos e, provavelmente, a um melhor alinhamento dos canais na estrutura do LiTi2(PO4)3. A máxima condutividade de 1,3 x 10-3 S/cm foi atingida na amostra obtida mediante tratamento térmico simples em 1000 °C durante 20 minutos a qual apresentou um tamanho médio de grão de 350 nm.

In this study the goal was to determine if there was a shift in the extent to which children’s attitudes toward deception change as they age. Participants (N=278) enrolled in grades 3-12 completed a survey assessing their lie acceptability and other factors as potential variables associated with a prodeception attitude. Results indicated that greater lie acceptability was correlated with male children who had self-reported acts of bad behavior. Results also suggest that nontraditional family environments may increase one’s perception of the acceptability of lying. These findings provide potential predictors of the acceptability of lying in children and adolescents that offer insight into the development of antisocial attitudes, which may have practical implications regarding the timing of crucial interventions as to prevent the continuance and escalation of such behaviors in the future.

Until recently, obtaining rest-frame optical spectra of galaxies at z > 1 was a time consuming and challenging observation due to the difficult nature of near-infrared (near-IR) spectroscopy. However, with the advent of second generation ground-based near-IR spectrographs (e.g. KMOS, MOSFIRE), and the new low resolution near-IR grisms on the Hubble Space Telescope (HST), we have entered a new era in the study of high redshift galaxies. This thesis explores the physical properties of star-forming galaxies in the redshift range 1 < z < 3 by utilising a custom reduction of the 3D-HST near-IR grism spectroscopic survey. One of the most important observational constraints on the evolution of galaxies is the mass-metallicity relation (MZR), which is sensitive to both the star-formation history and various inflow/outflow processes. I use the 3D-HST spectra to provide a new constraint on the MZR at 2:0 < z < 2:3, and moreover measure the O/H abundance directly from the oxygen and hydrogen emission lines ([OII], [OIII] and Hβ) as opposed to the more common method at high redshift of inferring O/H from the N/H ratio (via [NII] and Hα). I show that the traditional form of the MZR is recovered from the 3D-HST data, with metallicity increasing with the stellar mass of a galaxy. However, the absolute metallicity values I derive are inconsistent with previous N/H-based measurements of metallicity at these redshifts. Moreover, I show that the 3D-HST data is inconsistent with the `fundamental metallicity relation' (FMR), and that, contrary to previous claims, this local Universe relation may not hold out to z & 2. To investigate this metallicity discrepancy further, I measure the evolution of the [OIII]/Hβ nebular emission line ratio in the 3D-HST spectra over the redshift range 1:3 < z < 2:3. I compare this observed line ratio evolution with state-of-the- art theoretical models which take into account the independent evolution of the ionization parameter, electron density and metallicity of star-forming regions with redshift. The homogeneous 3D-HST dataset allows me to perform a consistent analysis of this evolution which takes into account line luminosity selection effects. I show that, according to models, the observed [OIII]/Hβ evolution cannot be accounted for by pure metallicity evolution. Instead I am able to infer that the line ratio evolution is more consistent with, at the very least, an evolution to stronger ionizing conditions at high redshift, and perhaps even denser star-forming regions. I explore how this result can also explain the observed discrepancy between high redshift metallicity measurements. In light of this finding, I revisit the MZR at z >~ 2 and employ a purely theoretical approach to inferring metallicities from nebular lines, which is able to account for an evolution in ionization conditions. I then use a selection of galaxies from the local Universe, which mimic the properties of high redshift galaxies, to derive a more robust ionization sensitive, conversion, between N/H and O/H. With this new conversion which I am able to bring the previous inconsistent metallicity measurements at z >~ 2 back into agreement. Finally, I am able to show that, in this new formalism, the metallicity evolution between z = 2 and z = 3 is perhaps not as large as previously reported. To conclude I discuss ongoing work as part of the KMOS Deep Survey (KDS) being undertaken with the near near-IR Multi-Object Spectrograph KMOS on the VLT. I describe the observations and data reduction that has been completed to date and describe how this instrument will allow me to extend the work presented in this thesis to z > 3. I also introduce FIGS, a new HST near-IR grism survey seeking to spectroscopically identify galaxies at 5:5 < z < 8:5 and work I have begun in exploring this dataset.

Enbridge has proposed the Line 3 “Replacement” Project, a new pipeline project taking a new route strait through Anishinaabe treaty territory in what is known as northern Minnesota. In the middle of the regulation process, the future remains unclear of how the State of Minnesota will move forward with the permitting process, but Anishinaabe communities, a range of non-profit organizations, and local landowners remain firmly against the line. Rooted in varied frameworks of Native sovereignty, the land, and Indigenous feminism, Anishinaabe communities lead the resistance against a product of ongoing settler colonialism, racial capitalism, and environmental racism. This thesis contextualizes the multi-tactical repertoires of those defending the land in the existing work of Indigenous scholars who write on the necessity for land-based resistance towards the unsettling process of decolonization. Ultimately, the resistance against Line 3 is representative of a long-term battle for Native sovereignty and self-determination in defense of the land and future generations.

We observed the [C II] line in 15 lensed galaxies at redshifts 1 < z <. 3 using HIFI on the Herschel Space Observatory and detected 14/15 galaxies at 3 sigma or better. High magnifications enable even modestly luminous galaxies to be detected in [C II] with Herschel. The [C II] luminosity in this sample ranges from 8x10(7) L-circle dot to 3.7x10(9) L-circle dot (after correcting for magnification), confirming that [C II] is a strong tracer of the ISM at high redshifts. The ratio of the [C II] line to the total far-infrared (FIR) luminosity serves as a measure of the ratio of gas to dust cooling and thus the efficiency of the grain photoelectric heating process. It varies between 3.3% and 0.09%. We compare the [C II]/FIR ratio to that of galaxies at z = 0 and at high redshifts and find that they follow similar trends. The [C II]/FIR ratio is lower for galaxies with higher dust temperatures. This is best explained if increased UV intensity leads to higher FIR luminosity and dust temperatures, but gas heating does not rise due to lower photoelectric heating efficiency. The [C II]/FIR ratio shows weaker correlation with FIR luminosity. At low redshifts highly luminous galaxies tend to have warm dust, so the effects of dust temperature and luminosity are degenerate. Luminous galaxies at high redshifts show a range of dust temperatures, showing that [C II]/FIR correlates most strongly with dust temperature. The [C II] to mid-IR ratio for the HELLO sample is similar to the values seen for low-redshift galaxies, indicating that small grains and PAHs dominate the heating in the neutral ISM, although some of the high [CII]/FIR ratios may be due to turbulent heating.

The medical and bioindustrial applications of pluripotent stem cells rely on our understanding of their biology. Pluripotent stem cell lines derived from embryos in different stages depend on distinct signalling pathways. Embryonic stem cells (ESCs), derived from the inner cell mass (ICM) of preimplantation embryos, are dependent on LIF/STAT3 signalling, while epiblast stem cells (EpiSCs), established from the postimplantation embryos, require Activin A/Smad2/3 signalling. Recent studies revealed the presence of intermediate pluripotent stem cell populations. Their growth factor responsiveness, gene expression pattern and associated chromatic signatures, are compatible with the state intermediate between ESCs and EpiSCs. However, it remains unknown whether such cell populations represent a stable clonally intermediate cell state. In this thesis, I describe the discovery and characterization of novel stem cell lines displaying gene expression pattern intermediate between ESCs and EpiSCs. These cells respond to LIF/STAT3 as well as Activin/Smad2/3 signalling at single cell level. They can integrate into the ICM and generate chimeric embryos. In keeping with a more advanced differentiation stage than that of ESCs, the LIF/Activin dual responsive stem cells showed accelerated temporal gene expression kinetics during in vitro differentiation in embryo bodies. I found that these properties are shared by some induced pluripotent stem cell (iPSC) lines. The notion of an intermediate state was consolidated by a genome-wide microarray profiling. The hierarchical clustering analysis grouped LIF/Activin dual responsive stem cells together into a cluster intermediate between ESCs and EpiSCs. These findings advanced our understanding of the regulation of pluripotency. A better understanding of distinct differentiation state of pluripotent stem cells and their signalling responsiveness is crucial for developing tailored strategies for lineage/cell type differentiation.

n the first chapter, I present an experimental analysis of weak-link coordination games in which subjects face specific time constraints in a volatile environment. In a basic 2×2 design, one factor involves situations in which all subjects face decision time (seconds to select a strategy) constraints. Another factor varies feedback time (seconds to review the outcome of the round) constraints. Overall, I find that stringent decision constraints lead to lower minimum effort levels, but increased feedback time seems to improve decision-making in anticipating others’ behaviors and aligning decisions with firm outcomes. Feedback constraints impact coordination outcomes more when decisions are made under time pressure. This finding suggests that if decisions are made under time duress, adequate recovery time can mean the difference between efficient and inefficient coordination. In the second chapter, I study the effect of heterogeneous groups on the efficiency of coordination in repeated weak-link coordination games. I develop an experimental environment to test compositional effects on organizational performance using peer effects to maximize productivity. Our experimental setup models a task environment with two different types of workers. In standard settings, subjects are given five effort choices: Ei ∈{0, 10, 20, 30, 40}. I introduce a second type of worker restricted to the highest effort choices: Ei ∈{30, 40}. I find that heterogeneous groups induce change to higher, more efficient equilibria. Despite increases in group size, firms with restricted workers report higher overall output. Several firms managed to coordinate at much higher levels, with two firms reaching the most efficient coordination level. Employees exert more effort when another employee(s) can be harmed in the firm. This paper provides a mechanism to alleviate coordination failure among large groups, demonstrating how social spillovers can overpower group-size effects. In the third chapter, we test whether participants exert more considerable effort when payoff-equivalent incentives are framed as losses rather than gains. I also intersect loss aversion and social preferences to model real-world situations when employees’ actions can cause another employee to bear a loss. I develop an experimental environment to test stake-size effects on organizational performance. Subjects are given an initial endowment at the beginning of the experiment, which allows us to assess decisions at different reference points and test the degree of loss aversion in each setting. We find significant treatment effects, such that employees in high-stake frames contribute significantly more effort than employees in low-stake frames. Loss aversion is efficiency-enhancing only in high-stake conditions. In other words, framing effects work if the stakes are high enough. We find that social preferences are not as influential in the loss domain compared to our previous findings in the gain domain. It appears that social concerns are influenced by the degree of loss framing. This paper contributes to the coordination literature on framing effects with different stakes and coordination outcomes when loss aversion intersects with social preferences. Indeed, the current COVID-19 pandemic underlines the importance of understanding how exposure to loss can shape decisions and whether this differs with social considerations when employees’ decisions can cause another to suffer the consequences.

In the end, the diagramless homology of a link is found to be equal to some number of copies of the Khovanov homology of that link. There is also a discussion of how one would generalize the diagramless homology theory (hence the theory of Khovanov homology) to links in arbitrary closed oriented 3-manifolds.

This project had a focus on the effect of digitoxin on inflammation in cancer Panc-1 cell line compared to BxPC-3 cell line. Pancreatic cancer, especially the form pancreatic adenocarcinoma is the fourth cause for cancer deaths, seen in the Western world. The progression and the initiation of the disease is a consequence of an interplay between genetic and inflammation events that are linked. Digitoxin is a cardiac glycoside with effect on various cancer types. Panc-1 cells and BxPC-3 cells were treated with different concentrations of digitoxin for 48 hours. Cell viability was measured, ELISA to measure expression of pro-IL-1β and IL-1β, since they are involved in the pathway and qPCR to measure gene expression. Gene expression was done to see if digitoxin affected the genes in the inflammasome pathway. The cell viability decreased with increasing concentration of digitoxin for both cell lines. TNFα was mostly downregulated in both Panc-1 and BxPC-3. NF-κB was upregulated in both cell lines. IL-1β was upregulated in BxPC-3 while gene expression was not seen in Panc-1. Lastly, IL-18 was downregulated in Panc-1, but upregulated in BxPC-3. A downregulation of IL-1β protein expression was seen in Panc-1 and in BxPC-3, a downregulation of pro IL-1β in Panc-1 and a downregulation of pro IL-1β in BxPC-3 was seen in ELISA. This study might provide an insight that is valuable for the researchers in the future to treat cancer from the perspective of an immune disorder.

La théorie d'invariants de type fini des 3-variétés et leurs entrelacs de Goussarov-Habiro repose sur le calcul de claspers, un ensemble d'outils de calcul topologique. Dans cette thèse, on calcule explicitement les invariants en bas degré pour certaines classes d'objets, par une méthode dite graphique. Nous étudions ainsi les cylindres d'homologie sur une surface à 0 ou 1 composante de bord et les string-links framés des boules d'homologie. Leurs invariants de degré 1 sont caractérisés en termes d'invariants classiques, et une correspondance est établie entre les deux cas. On regarde aussi les invariants de Vassiliev des string-links, du point de vue des claspers. Le calcul des invariants de degré 2 implique la construction d'un certain invariant des string-links à 2 cordes. Le lien entre invariants de Vassiliev et de Goussarov-Habiro est étudié pour les string-links.

Si/Ti and Si/Al), via hydroxide and fluoride medium using the cation 1-butyl-3-
methylimidazolium as structure directing agent. Initially, the cation was synthesized in
chloride form and used for the synthesis in hydroxide medium. An anion-exchange (Cl- for
OH-) was required for the synthesis in fluoride medium. Different reactants were used for the
formation of gels synthesis, resulting in the crystallization of MFI and TON phases, the latter
predominant in many compositions. The cation and synthesized zeolites obtained were
characterized by different techniques such as NMR, TG/DTG, XRD, SEM, N2 adsorption and
desorption, DRS and EPMA. Besides characterizing the cation and zeolites, the mother liquor
of hydroxide synthesis was characterized and it was possible to observe a modification of the
cation in the synthesis conditions employed. The materials synthesized in this work can be

Rare sont les études empiriques qui, palliant limites langagières de l’enfant, ont investigué la symptomatologie psychologique et comportementale de l’abus sexuel auprès des enfants en examinant les séquelles de ce trauma au travers le jeu libre. Conséquemment, l’objectif de la présente étude est de relever le profil socioaffectif de deux groupes d’enfants abusés sexuellement dont l’un démontre une capacité à jouer préservée et l’autre se caractérise par une activité ludique limitée. En outre, cette recherche vise à identifier les variables relatives aux profils dégagés qui discriminent de façon plus spécifique chacun des deux groupes d’enfants à l’étude. L’échantillon est constitué de 50 enfants abusés sexuellement âgés entre 3 et 6 ans. Tel que prédit, les résultats démontrent que les enfants ayant un potentiel ludique limité, comparativement aux enfants où le potentiel est préservé, sont caractérisés par un surcroît de comportements de nature anxio-dépressive. De plus, ils démontrent un nombre significativement plus élevé de comportements dissociatifs et leur profil symptomatologique est davantage « Internalisé ». Ces résultats supportent l'idée que l'abus sexuel, agissant comme traumatisme, perturbe la capacité à jouer de deux manières différentes : soit il bloque l'accès au jeu chez certains enfants, soit il trouble l’activité ludique en elle-même chez un autre groupe.

The purpose of this thesis is to investigate a class of four left-invariant optimal control problems on the special orthogonal group SO(3). The set of all control-affine left-invariant control systems on SO(3) can, without loss, be reduced to a class of four typical controllable left-invariant control systems on SO(3) . The left-invariant optimal control problem on SO(3) involves finding a trajectory-control pair on SO (3), which minimizes a cost functional, and satisfies the given dynamical constraints and boundary conditions in a fixed time. The problem is lifted to the cotangent bundle T*SO(3) = SO(3) x so (3)* using the optimal Hamiltonian on so(3)*, where the maximum principle yields the optimal control. In a contribution to the study of this class of optimal control problems on SO(3), the extremal equations on so(3)* (ident ified with JR3) are integrated via elliptic functions to obtain explicit expressions for the solution curves in each typical case.
