Abstract:
ABSTRACT The regulatory network of the cell envelope stress response in Bacillus subtilis involves both extracytoplasmic function σ-factors and two-component signal transducing systems. One such system, LiaRS, responds to cell wall antibiotics that interfere with the undecaprenol cycle and to perturbation of the cytoplasmic membrane. It is encoded by the last two genes of the liaIHGFSR locus. Here, we analyzed the expression of two LiaR-dependent operons, liaIHGFSR and yhcYZ-yhdA, and characterized a palindromic sequence required for LiaR-dependent activation. Since induction of the strong liaI promoter leads to both liaIH and liaIHGFRS transcripts, LiaR is positively autoregulated. Systematic deletion analysis of the liaI operon revealed that LiaF is a potent negative regulator of LiaR-dependent gene expression: a nonpolar liaF deletion led to constitutive activation of both characterized LiaR-dependent promoters. The liaF gene is conserved in both sequence and genomic context in the Firmicutes group of gram-positive bacteria, located directly upstream of liaSR orthologs. LiaH, a homolog of Escherichia coli phage shock protein A, also plays a more subtle role in negatively modulating the bacitracin-inducible expression from LiaR-dependent promoters. Our results support a model in which the LiaFRS module integrates both positive and negative feedback loops to transduce cell envelope stress signals.

Abstract:
Abstract Background Daptomycin (DAP) is a key first-line agent for the treatment of vancomycin-resistant enterococcal infections. Resistance to DAP in enterococci is regulated by the liaFSR three-component regulatory system that consists of a histidine kinase sensor (LiaS), a response regulator (LiaR) and a transmembrane protein of unknown function (LiaF). Previous studies indicate that deletion of isoleucine in position 177 of LiaF results in DAP tolerance and is sufficient to change membrane architecture. Here, we dissect the role of LiaF in DAP resistance Methods We generated three liaF mutants in OG1RF, a DAP-susceptible laboratory strain of Efs (DAP MIC = 2 µg/mL): (i) a non-polar, C-terminal truncation of liaF (OG1RFliaF∆152), (ii) a null liaF mutant with a premature stop-codon (OG1RFliaF*11), and (iii) an isoleucine deletion at position 177 (OG1RFliaF177). We determined DAP MIC by Etest and characterized the localization of anionic phospholipids microdomains using 10-nonyl-acridine-orange (NAO). The expression of the liaXYZ (the main target of LiaR) and liaFSR clusters were evaluated by qRT-PCR and relative expression ratios (Log2 fold change) were calculated by normalizing to gyrB expression. We assessed activation of LiaFSR by evaluating surface exposure of LiaX by ELISA. We used the bacterial adenylate cyclase two-hybrid system (BACTH) to evaluate the protein-protein interaction between LiaF and LiaS. Results Full deletion of liaF or the C-terminal truncation of LiaF did not have any effect on DAP MICs, membrane architecture or a significant increase in LiaX surface exposure compared with parental strain OG1RF. In contrast, deletion of the codon encoding isoleucine in position 177 of LiaF caused a major increase (8-fold) in LiaX exposure and redistribution of anionic phospholipid microdomains away from the septum without changes in the actual DAP MIC. Transcriptional analyses indicated upregulation (>2 log2-fold) in the liaXYZ gene cluster indicating activation of the stress response. We also observed a positive interaction between LiaF and LiaS. Conclusion LiaF is likely a key activator of the LiaFSR stress response and the critical regulatory domain appears to be located in a stretch of four isoleucines toward the C-terminal of the protein. Background The relationship between intention and behavior has been well researched, but most studies fail to capture dynamic, time-varying contextual factors. Ecological momentary assessment through mobile phone technology is an innovative method for collecting data in real time, including time-use data. However, only a limited number of studies have examined day-level plans to be physically active and subsequent physical activity behavior using real-time time-use data to better understand this relationship. Objective This study aims to examine whether plans to be physically active (recorded in advance on an electronic calendar) were associated with objectively assessed physical activity (accelerometry), to identify activities that replaced planned periods of physical activity by using the mobile app Life in a Day (LIAD), and to test the feasibility and acceptability of LIAD for collecting real-time time-use data. Methods The study included 48 university students who were randomly assigned to 1 of 3 protocols, which were defined by 1, 3, or 5 days of data collection. Participants were asked to record their planned activities on a Google Calendar and were provided with mobile phones with LIAD to complete time-use entries in real time for a set of categories (eg, exercise or sports, eating or cooking, school, or personal care). Participants were instructed to wear an accelerometer on their nondominant wrist during the protocol period. A total of 144 days of protocol data were collected from the 48 participants. Results Protocol data for 123 days were eligible for analysis. A Fisher exact test showed a statistically significant association between plans and physical activity behavior (P=.02). The congruence between plans and behavior was fair (Cohen κ=0.220; 95% CI 0.028-0.411). Most participants did not plan to be active, which occurred on 75.6% (93/123) of days. Of these 93 days, no physical activity occurred on 76 (81.7%) days, whereas some physical activity occurred on 17 (18.3%) days. On the remaining 24.4% (30/123) of days, some physical activity was planned. Of these 30 days, no physical activity occurred on 18 (60%) days, whereas some physical activity occurred on 12 (40%) days. LIAD data indicated that activities related to screen time most often replaced planned physical activity, whereas unplanned physical activity was often related to active transport. Feasibility analyses indicated little difficulty in using LIAD, and there were no significant differences in feasibility by protocol length. Conclusions Consistent with previous literature, physical activity plans and physical activity behaviors were linked, but not strongly linked. LIAD offers insight into the relationship between plans and behavior, highlighting the importance of active transport for physical activity and the influence of screen-related behaviors on insufficient physical activity. LIAD is a feasible and practical method for collecting time-use data in real time.

ABSTRACT The Lia system, a cell envelope stress response module of Bacillus subtilis, is comprised of the LiaRS two-component system and a membrane-anchored inhibitor protein, LiaF. It is highly conserved in the Firmicutes bacteria, and all orthologs investigated so far are activated by cell wall antibiotics. In response to envelope stress, the systems in Firmicutes cocci induce the expression of a number of genes that are involved in conferring resistance against its inducers. In contrast, a complete picture of the LiaR regulon of B. subtilis is still missing and no phenotypes could be associated with mutants lacking LiaRS. Here, we performed genome-wide transcriptomic, proteomic, and in-depth phenotypic profiling of constitutive “Lia ON” and “Lia OFF” mutants to obtain a comprehensive picture of the Lia response of Bacillus subtilis. In addition to the known targets liaIH and yhcYZ-yhdA, we identified ydhE as a novel gene affected by LiaR-dependent regulation. The results of detailed follow-up gene expression studies, together with proteomic analysis, demonstrate that the liaIH operon represents the only relevant LiaR target locus in vivo. It encodes a small membrane protein (LiaI) and a phage shock protein homolog (LiaH). LiaH forms large oligomeric rings reminiscent of those described for Escherichia coli PspA or Arabidopsis thaliana Vipp1. The results of comprehensive phenotype studies demonstrated that the gene products of the liaIH operon are involved in protecting the cell against oxidative stress and some cell wall antibiotics. Our data suggest that the LiaFSR system of B. subtilis and, presumably, other Firmicutes bacilli coordinates a phage shock protein-like response.

In order to increase the efficiency of magneto-optical traps, yielding a greater number of trapped atoms and long lifetimes, the effect of lightinduced desorption atom source emerges as an adjuvant and no thermal atoms. A prior experimental investigation (FRITSCH, 2011) showed that the number of rubidium atoms trapped increases when a light desorption focuses on the inner walls of the chamber imprisonment, whose material is stainless steel. The light desorption is applied from an incandescent lamp with power equal to 60 mW. The experimental data are adjusted and parameters are determined from the trap, in case of absence of light. Thus, the theoretical model proposed by Zhang et al. (2009) is applied, describing the increase of the number of atoms in the magneto-optical trap when the light desorption is triggered. In this setting, the rate of desorption and adsorption coefficient is estimated at 5x1017 atoms per second and 55 s-1. The combination of this model with that proposed by Monroe et al. (1990), in case of absence of light, perfectly describes the experimental curve generated when the mechanism for obtaining the magneto-optical trap and light desorption are actuated simultaneously. Furthermore, a theoretical prediction for the desorption rate, the rate of charging of magneto-optical trap and maximum number of atoms trapped on the basis of light desorption power is shown graphically.
Com o intuito de aumentar a eficiência das armadilhas magnéto-ópticas, obtendo-se um número maior de átomos aprisionados e longos tempos de vida, o efeito de dessorção atômica induzida por luz surge como uma fonte coadjuvante e não térmica de átomos. Uma investigação experimental prévia (FRITSCH, 2011) comprovou que o número de átomos de rubídio armadilhados aumenta quando uma luz de dessorção incide sobre as paredes internas da câmara de aprisionamento, cujo material é de aço inoxidável. A luz de dessorção aplicada é proveniente de uma lâmpada incandescente com potência igual a 60 mW. Os dados experimentais são fitados e os parâmetros da armadilha são determinados, para o caso sem luz. Com isso, o modelo teórico proposto por Zhang et al. (2009) é aplicado, descrevendo muito bem o incremento do número de átomos presentes na armadilha magnéto-óptica quando a luz de dessorção é acionada. Desse ajuste, a taxa de dessorção e o coeficiente de adsorção são estimados em 5x1017 átomos por segundo e 55 s-1. A união desse modelo com o proposto por Monroe et al. (1990), para o caso sem luz, descreve perfeitamente a curva experimental gerada quando o mecanismo para a obtenção da armadilha magnéto-óptica e a luz de dessorção são acionados simultaneamente. Além disso, uma previsão teórica para a taxa de dessorção, a taxa de carregamento da armadilha magnéto-óptica e o número máximo de átomos aprisionados em função da potência da luz de dessorção é mostrada graficamente.

Les possibilités remarquables offertes par les techniques de refroidissement laser et de piégeage ont permis des avancées significatives dans le domaine des standards de fréquence à base d'atomes neutres. Parmi les différents systèmes permettant d'accéder à des valeurs de stabilité et précision élevés, l'atome d'argent est considéré comme l'un des candidats les plus prometteurs comme standard optique de fréquence. Cette thèse traite du développement d'une horloge basée sur des atomes neutres d'Ag. La transition 4d105s2 S1/2 > 4d95s2 2D5/2 dans l'Argent peut fournir une référence de fréquence pour les raisons suivantes : le niveau métastable 2D5/2, qui se relaxe par émission d'un photon quadripolaire électrique à 330. 5 nm, possède une durée de vie estimée à 0. 2 s, correspondant à une largeur de raie naturelle très faible de 0. 8 Hz. Il est par conséquent possible de bénéficier d'un long temps d'interaction pour le dispositif de fontaine atomique. De plus, cet état est accessible par une transition à 2 photons à 661. 2 nm, permettant d'éliminer l'effet Doppler du 1er ordre. Comme aspect technique important, la fréquence nécessaire pour la transition d'horloge et celle pour le refroidissement des atomes d'argent (raie D2 à 328 nm) peut être obtenue avec le même cristal de NdYLF (émission possible à 1322 nm/1312 nm). Ce travailde thèse détaille la réalisation de s sources nécessaires au refroidissement des atomes d'argent et à la transition d'horloge à l'aide d'un cristal de NdYLF pompé par diode et par deux étapes de doublage de fréquence intracavité. Par ailleurs, le phénomène LIAD y est présenté comme un compromis intéressant dans la conception d'une horloge atomique optique
The remarkable capabilities of laser cooling and trapping techniques for neutral atoms have led to great advances in the field of neutral atom-based frequency standards. Among the several atomic systems affording excellent prospects of high stability and accuracy in the optical region, the silver atom is regarded as one of the most promising candidates for an optical frequency standard. This thesis deals with the development of a clock based on neutral silver atoms. The Ag 4d105s2 S1/2 > 4d95s2 2D5/2 transition provides an attractive reference frequency for the following reasons : the 2D5/2 metastable level, which decays by emission of electric quadrupole radiation at 330. 5 nm, has an estimated lifetime of 0. 2 s, corresponding to an ultralow natural linewidth of only 0. 8 Hz. It’s therefore possible therefore to benefit from the long interaction time in an atomic fountain setup. Furthermore, the long-lived state is accessible with a two-photon transition at 661. 2 nm, providing a first-order Doppler-free interaction with atoms of all velocities. As an important technical aspect, the frequency needed to drive the clock transition and to cool the silver atoms (D2 line at 328 nm) can be provided by the same crystal Nd:YLF enable to generate both 1322 nm and 1312 nm. This work details especially the realization of the laser sources for both cooling silver atoms and driving the clock transition by diode-pumped Nd-YLF crystal and intracavity frequency doubling operation in two steps (second and fourth harmonic generation). In addition, the Lignt Induced Atomic Desorption (LIAD) phenomenon is described and observed as an interesting compromise in the case of an optical atomic clock conception

The migration dynamics in Brasilia has shown a decrease over the years, however, data point to a population of 53% of people who come from other states, seeking, for the most part, better living through work. Currently the most attractive areas the one that encompasses business activities and the civil service, offered by different spheres of the government sector. The migrant profile that demand Brasilia as the destination has been changing gradually, with an increase in age of those people who usually move with their families. Weint end this study to know aspects of the relationship of couples and families who migrate to Brasilia because of appro valor transfer of a spouse in an open competition, identifying the changes that migration causes in marital and family relationships, identify the timing of the lifecycle What happens to move to Brasilia, the sources of support before and after migration, the strategies used by the family to deal with the possible difficulties of adaptation, as well as meet the future expectations of the family. Participated in this study three families at different stages of the lifecycle, a family with teenage son, a family with little son and a childless couple. The families residing in Brasilia, at most 5 years. It is a qualitative ere search by means of case study, using the following instruments for data collection: semi-structured interview, genogram and family ecopama. An interview with each family, recorded and transcribed was held. Data were analyzed based on the constructive-interpretative method of Gonzalez-Rey. Eight zones of meaning were built from the axis of analysis and indicators. Responding to the specific objectives of the study, it was observed that: the wives showed greater difficulty in establishing meaningful links in Brasilia situation hampered by lack of own goals in the non-domestic sphere; that they have a social network provided by the work favored men make friends and experience to adapt more effectively; the most frequent contact with family members living in the home cities is more common in the family that created less ties in the new town after the change; the lack of social support networks in Brasilia seems to overload the family system may cause difficulties in the marital relationship; the possibility of the return to origin city presents itself as a strategy to sustain the initial difficulties of migration, however, when the system is more adapted and adjusted to the new reality, no longer uses this feature with the same intensity; families that were more adapted to life in the capital and they have devised the migration process in a satisfactory manner are those who can think of a future in Brasilia, not considering returning to hometowns. This work helped in understanding the current migratory dynamics off families seeking Brasilia as a destination city.

A ciranda - manifestação cultural que compreende canto e dança foi categorizada pelos estudiosos como sendo uma manifestação folclórica e pertencente ao povo , como trabalhadores rurais, pescadores e biscateiros. O ambiente em que se cantava e dançava cirandas restringia-se aos locais populares como as beiras de praia e pontas de ruas. Nos anos 1970, as rodas de cirandas se popularizaram entre a classe média, sendo deslocadas para espaços de lazer da cidade do Recife. A prática cultural metamorfoseou-se, apresentando-se com modificações na sua configuração, letras das músicas, sentidos e significados, transformando-se em um espetáculo no calendário turístico da cidade. A espetacularização das cirandas ocorreu, sobretudo, a partir dos Festivais de Cirandas, concursos nos quais os cirandeiros disputavam o título da melhor ciranda do ano. Esses festivais atendiam não apenas a indústria cultural que criava um mercado de produção, distribuição e consumo de bens culturais, mas também a indústria do turismo que buscava entreter turistas e parte da sociedade pernambucana com shows de cirandas . Na realização deste trabalho, dialogamos com diversas fontes documentais, desde os periódicos, os relatos de memória, a iconografia, passando pela análise da música enquanto fonte histórica, buscando estabelecer um diálogo polifônico entre as diversas vozes que delas ecoaram. Nessas fontes, buscamos compreender os sentidos e os significados da ciranda na vida dos cirandeiros (as), isto é, como esses os sujeitos sociais pensaram, agiram, vivenciaram e significaram os processos que metamorfosearam a prática cultural no período de 1960 a 1980. Através das mudanças operadas na manifestação cultural, buscamos compreender as relações que os diversos cirandeiros estabeleceram com instituições públicas, privadas, intelectuais e demais artistas, ou seja, como estes sujeitos sociais interagiram com as transformações que foram operadas nas rodas de cirandas, constituídas de sistemas de símbolos que articularam significados construídos dentro de um grupo-tempo-espaço

Abstract:
Syftet med den här rapporten är att undersöka hur ett trädbyte utförs på en plats som är skyddad som statligt byggnadsminne. Platserna som beskrivs är barockträdgården i Uppsala Botaniska trädgård och barockträdgården i slottsträdgården på Drottningholm. Båda trädgårdarna ägs av staten och förvaltas av Statens fastighetsverk. Riksantikvarieämbetet har det övergripande ansvaret för att anläggningarna inte förvanskas. Botaniska trädgården i Uppsala tillhör Uppsala universitet och underhålls av universitetet. Anläggningarna har ett högt historiskt värde som gör dem skyddsvärda och på grund av detta finns det restriktioner och föreskrifter som dikterar hur platserna ska vårdas och förvaltas. Uppsatsen bygger på intervjuer med projektledaren för båda trädbytena och trädgårdsdirektören och 1:e trädgårdsmästare vid Botaniska trädgården i Uppsala. De gamla lindarna på Drottningholm planterades under sent 1600-tal och tidigt 1700-tal och var till större delen parklind, Tilia x europaea ’Pallida’, importerade från Holland. De gamla lindarna i Botaniska trädgården planterades på 1970-talet då trädgården restaurerades under ledning av Walter Bauer. Trädbytet på Drottningholm utfördes mellan 1997–2011 i fem etapper och trädbytet i Botaniska trädgården utförs från hösten 2016 och förväntas vara klart i juni 2017. Båda anläggningsarbetena och en femårig garantiskötsel utförs på entreprenad. I rapporten beskrivs vad som påverkar valet av träden vid anläggningarna. Det har utförts studier för att art- och sortbestämma de ursprungliga lindarna i alléerna på Drottningholm. De nya lindarna som planterats där och de som ska planteras under våren 2017 i Botaniska trädgården, är av samma sort som de ursprungliga lindarna på Drottningholm. Ett stort antal åtgärder har utförts vid anläggningen och skötseln av träden för att se till att de etablerar sig ordentligt och utvecklas väl.
The aim of this paper is to examine how a tree replacement is carried out in a place that is protected as a cultural heritage. The places described are the Baroque garden in Uppsala Botanical Gardens and the baroque garden in the palace garden at Drottningholm. Both gardens are owned by the state and managed by the National Property Board. The National Heritage Board has overall responsibility that the facilities are not distorted. The Botanical Garden in Uppsala belongs to Uppsala University and is managed by the University. The gardens have a high historical value that make them worthy of protection. Because of this there are restrictions and regulations that dictate how the locations should be maintained and administered. The thesis is based on interviews with the project manager for both of the tree-replacements and the garden Director and 1st gardener at the Botanical Garden in Uppsala. The old lime trees at Drottningholm were planted in the late 1600s and early 1700s and was largely dutch lime, Tilia x europaea 'Pallida', imported from Holland. The old lime trees in the botanical garden was planted in the 1970s, when the garden was restored under the direction of Walter Bauer. The tree replacement at Drottningholm was conducted between 1997–2011 in five stages and the restoration in the Botanical Garden is carried out in autumn 2016 and is expected to be completed in June 2017. Both the construction work and a five-year warranty maintenance is performed under contract. The paper describes what influences the choice of the trees at the facilities. There have been studies to determine the species and varieties of the original lime trees in the avenues at Drottningholm. The new lime trees planted there and those that will be planted in the spring of 2017 in the Botanical Garden, are of the same type as the original lime trees at Drottningholm. A large number of measures have been carried out at the installation and maintenance of the trees to ensure that they establish themselves properly and develop well.

This study aims to verify the impact of the Bolsa Fam?lia Program (BFP) in income and school attendance of poor Brazilian families. It is intended to also check the existence of a possible negative effect of the program on the labor market, titled as sloth effect. For such, microdata from the IBGE Census sample in 2010 were used. Seeking to purge possible selection biases, methodology of Quantilic Treatment Effect (QTE) was applied, in particular the estimator proposed by Firpo (2007), which assumes an exogenous and non-conditional treatment. Moreover, Foster- Greer-Thorbecke (FGT) index was calculated to check if there are fewer households below the poverty line, as well as if the inequality among the poor decreases. Human Opportunity Index (HOI) was also calculated to measure the access of young people / children education. Results showed that BFP has positively influenced the family per capita income and education (number of children aged 5-17 years old attending school). As for the labor market (worked hours and labor income), the program showed a negative effect. Thus, when compared with not benefiting families, those families who receive the BFP have: a) a higher family income (due to the shock of the transfer budget money) b) more children attending school (due to the conditionality imposed by the program); c) less worked hours (due to sloth effect in certain family groups) and d) a lower income from work. All these effects were potentiated separating the sample in the five Brazilian regions, being observed that the BFP strongly influenced the Northeast, showing a greater decrease in income inequality and poverty, and at the same time, achieved a greater negative impact on the labor market
Este estudo tem como objetivo verificar o impacto do Programa Bolsa Fam?lia (PBF) na renda e na frequ?ncia escolar das fam?lias pobres brasileiras. Pretende-se, ainda, verificar a exist?ncia de um poss?vel efeito negativo do programa no mercado de trabalho, intitulado como efeito-pregui?a. Para tanto, foi utilizado os microdados do Censo amostral do IBGE em 2010. Visando expurgar poss?veis vieses de sele??o, aplicou-se a metodologia do Efeito Quant?lico do Tratamento (EQT), em especial o estimador proposto por Firpo (2007), no qual assume um tratamento ex?geno e n?o condicional. Ademais, foi calculado o ?ndice Foster-Greer-Thorbecke (FGT) para verificar se h? menos domic?lios abaixo da linha de pobreza, bem como se a desigualdade entre os pobres diminui. Foi calculado, ainda, o ?ndice de Oportunidades Humanas (IOH) para medir o acesso de jovens/crian?as educa??o. Os resultados mostraram que o PBF influenciou positivamente a renda familiar per capita e a educa??o (n?mero de filhos de 5 a 17 anos que frequentam a escola). Quanto ao mercado de trabalho (horas trabalhadas e renda do trabalho), o programa mostrou um efeito negativo. Assim, quando comparada com as fam?lias n?o beneficiadas, aquelas fam?lias que recebem o PBF possuem: a) uma maior renda familiar (devido ao choque or?ament?rio do repasse monet?rio); b) mais filhos frequentando a escola (devido ? condicionalidade imposta pelo programa); c) menos horas trabalhadas (devido ao efeito-pregui?a em alguns grupos familiares) e; d) uma menor renda oriunda do trabalho. Todos esses efeitos foram diferenciados ao separar a amostra nas cinco regi?es brasileiras, sendo observado que o PBF influenciou mais fortemente o Nordeste e a zona rural, que mostraram um maior impacto negativo no mercado de trabalho e, ao mesmo tempo, obteve uma maior diminui??o da desigualdade de renda e pobreza

Clara Schumann’s 1850 tour of northern Germany with her husband officially ended with a successful concert in Altona where Jenny Lind made a surprise appearance. Immediately thereafter, one more concert featuring the pianist, singer, and Robert’s music was added at the last minute to take place in Hamburg. This too was a success. But a detail that made it especially memorable was Lind’s position behind the piano lid so that, as Clara recounted in her diary, many audience members could hardly catch a glimpse of her. This paper explores the rationales and implications of this singular and fleeting moment, and teases out aspects of the two star performers’ relationship both on and off the stage. In the process, the paper draws attention to hitherto neglected variables in the performance practice of Lieder and seeks to expand our lines of inquiry with regards to the 19th-century Lied as cultural practice.
