Abstract:
Penelitian ini bertujuan untuk mengetahui Pemerataan Bantuan Program Keluarga Harapan (PKH) di Negeri Leahari, Kecamatan Leitimur Selatan. Tipe penelitian yang digunakan adalah penelitian deskriptif kualitatif, Teknik pengumpulan data dalam penelitian ini adalah observasi, wawancara dan dokumentasi. Hasil penelitian yang ditemukan bahwa Proses penyaluran PKH di Negeri Leahari merupakan program pemerintah yang disalurkan kepada masyarakat miskin di Indonesia termasuk masyarakat yang berada di Negeri Leahari, dan sudah menyentu masyarakat secara langsung.. Penyaluran PKH belum tepat sasaran karena masih ada masyarakat miskin yang belum mendapatkan bantuan tersebut. Faktor Yang Mempengaruhi Penyaluran PKH di Negeri Leahari bahwa masyarakat penerima bantuan PHK tertib dan mengikuti prosedur yang sudah ditetapkan oleh Pemerintah. Namun ada beberapa faktor yang menghambat yaitu proses pendataan yang tidak ditindak lanjut oleh pemerintah sehingga daftar yang dikeluarkan berbedah dengan daftar yang dikirim. Kesalahan yang terjadi pada data-data peenrima sehingga perlu di lakukan aktivasi Kembali.

Abstract:
Tujuan penelitian adalah (1). Mengidentifikasi jenis-jenis usaha dan sumber-sumber pendapatan baik dari usaha pertanian maupun luar pertanian di Negeri Leahari Kecamatan Leitimur Selatan. (2). Merumuskan strategi pengembangan usaha ternak ayam broiler terintegrasi dengan dusung di Negeri Leahari Kecamatan Leitimur Selatan. Unit pengamatan terkecil yang diamati pada penelitian ini adalah jenis-jenis usaha dan pemilik usaha yang mengintegrasikan usaha ternak ayam broiler dengan dusung. Hasil penelitian menunjukan : (1). Sumber-sumber pendapatan usaha di Negeri Leahari terbagi menjadi tiga jenis yaitu: Ternak, dusung, dan Ikan. Jenis usaha 1 Memiliki pendapatan dari ternak Rp. 40,139,643/tahun. dan dusung Rp. 50.543.050/tahun. Jenis usaha 2 memiliki pendapatan dari ternak Rp. 98.932.780/tahun, dan dusung sebesar Rp. 8.638.000 /Tahun., Jenis usaha 3 memiliki pendapatan, dari ternak sebesar Rp. 154,337,842/Tahun, dusung sebesar Rp. 17.977.000/tahun, dan ikan sebesar Rp. 58.990. 667/tahun. Strategi untuk mengembangkan usaha peternakan ayam ras di Negeri Leahari adalah dengan SO Strategi, WO Strategi, ST Strategi dan WT Strategi Kombinasi usaha yang baik digunakan untuk meningkatkan pendapatan pemilik usaha adalah kombinasi antara ternak-dusung-ikan.

Abstract:
Agroforestry is a land use activity that aims to maintain or increase total yields in a sustainable manner by combining woody plants with food crops or forage crops on the same plot of land, either simultaneously or alternately, using management practices. according to local ecological, economic, social, and cultural conditions So far, Negeri Leahari has only depended on the season and only on a small scale to meet the economic needs of the community, so farmers do not know how much income they get from managing the hamlet. This study aims to determine the cropping patterns and income levels of agroforestry farmers in Dusung Negeri Leahari,This study used direct observation methods and interviews with hamlet owners. The results showed that there were two hamlet management patterns in Leahari Country, namely Trees Along Border and Mixture Random Of the 20 respondents, 12 people, or 60%, used the Trees Along Border pattern, while those who used the Mixture Random pattern were as many as 8 people, or 40%. The net income obtained using the Trees Along Border pattern is Rp. 41,425,000/year, or 60%, while the net income obtained using Mixture Random, is Rp. 23,176,000/year, or 40%. In conclusion, there are 2 agroforestry management patterns: the Trees Along Border Pattern and the Random Mixture Pattern while the level of farmer opinion with the Trees Along Border Pattern is Rp. 3,452,089, and with the Random Mixture Pattern, the average is Rp. 2,897,000.

Abstract:
Rupiah is the official currency of Indonesia and is widely accepted in economic transactions within the country's borders. The rupiah can be weakened by the use of foreign currencies, the circulation of counterfeit money, and poor handling of money. To promote the importance of the rupiah, the Student Community Service (KKN) from Pattimura University in Ambon organized a program called Love Proud Understanding Rupiah. The socialization event took place at the Leahari State Office and involved preparing educational materials about the value of love, pride, and understanding of the rupiah. On May 23, 2023, the outreach program was carried out in Negeri Leahari to educate people about the significance of preserving and caring for the rupiah currency. By raising awareness about the importance of the rupiah, the socialization activities aimed to maintain the economic stability of Indonesia.

Abstract:
Research has been carried out to determine the cell structure of the epidermis and stomata in some mangrove plants in the species Aegiceras corniculatum and Rhizophora apiculata. Descriptive method is used to describe the cell structure of the epidermis and stomata of Aegiceras corniculatum and Rhizophora apiculata and quantitative leaves to calculate the number of stomata, number of epidermis and stomata index based on nail polish on the cross section of epidermal cells on the lower underside of the leaf using a light microscope, while the incision longitudinal to determine leaf thickness between the upper epidermis and the lower epidermis. The results showed that the two mangrove species that grow in the mouth of the Poka and Leahari villages namely Aegiceras corniculatum and Rhizophora apiculata were found to have the same anatomical structure and leaf anatomical characteristics in terms of the shape of epidermal cells, rectangular, octagonal, elongated, and irregular. Aegiceras corniculatum and Rhizophora apiculata have anomositic stomata type because neighboring cells surround the stomata and have the same shape as epidermal cells. Mangrove species in the river estuary of Poka Village have higher number of stomata and smaller epidermal size and lower stomata index than mangrove species in Leahari Village due to the influence of the shade. This study aimed to analyze the maximum capacity of roads in overcoming bottlenecks using the Maximum Network Flow Model and analyze the interaction of the distribution of the movement with the city located around the city of Ambon by using Gravity Model. Abstract:
Abstract Previous work in our lab found that increased expression of the E6* variant of the HPV E6 viral protein was significantly correlated with worse response to standard-of-care chemoradiation (CRT) and progression-free survival, independently of HPV genotype. In vitro validation showed that overexpression of E6* induces senescence in an HPV 16-positive cervical cancer cell line. Given these findings, we hypothesized that increased E6* expression mediates resistance to CRT by inducing a reversible state of tumor cell senescence. We created a senescence score using ten genes that have been consistently reported in the literature as associated with senescence. Using an institutional cohort of 68 patients with cervical cancer with prospectively collected clinical outcome data, we performed bulk RNA sequencing to assess senescence scores before CRT and determine whether there is an association between senescence scores and E6* expression. Using a second institutional cohort of 35 patients with cervical cancer treated with CRT with pre and posttreatment tumor biopsies, we performed bulk RNA sequencing to evaluate the same relationship and determine whether expression of senescence-related genes was affected by CRT. Nine cervical cancer cell lines stratified into low- and high-E6* expression groups will be assessed regarding their senescence score, beta-galactosidase staining, and SASP expression before and after CRT. In our first cohort, there was no correlation between E6*:E6 expression ratio and senescence score, regardless of HPV genotype. In our second cohort, CRT increased senescence score in CRT-treated samples (p < 0.01), but p16INK4a expression significantly decreased compared to pre-treatment samples (p < 0.01). Increased senescence score was not associated with cancer recurrence. There was no significant difference in senescence scores between different HPV genotypes before therapy. Tumors that were positive for HPV 16 or HPV 18 had significantly lower senescence scores compared to HPV negative tumors (p = 0.037 and p = 0.038, respectively) after CRT. Among HPV 16-positive tumors (n = 12), there was no correlation between E6*:E6 expression ratio and senescence score before or after CRT. We found no association between senescence score and E6* expression, and between senescence score and cancer recurrence after CRT in our dataset. Additionally, our results suggest that p16INK4a might not be an appropriate senescence marker in tumors that have intrinsically high p16INK4a expression, such as in cervical cancer, reinforcing the need for a cervical cancer-specific senescence signature. We are currently testing beta-galactosidase and SASP expression in vitro and in vivo before and after CRT. Abstract:
Abstract Background Breast cancer (BC) is the most frequent cause of leptomeningeal metastases (LM). LM diagnosis is confirmed by the detection of tumor cells in the cerebrospinal fluid (CSF) using conventional cytology (gold standard). However, even with optimal CSF sample volume and time to the analysis, the sensitivity of this technique is low, demanding repeated samples. Here, we aimed to evaluate the value of circulating tumor cell (CTC) detection in CSF using the CellSearch® system for LM diagnosis. Materials and Methods This prospective, monocentric study included adult BC patients with suspected LM (clinical and/or radiological signs). CSF samples from 1-3 lumbar puncture(s) were analyzed: protein level, conventional cytology (60 drops), and CTC detection with the CellSearch® system (60 drops, first lumbar puncture only). Sensitivity (Se) and specificity (Sp) were calculated, using the results of the conventional cytology as the gold-standard. Results Forty-nine eligible patients were included (Jan 2017-Jan 2020): median age 51.8, 95.9% women, 20.4% HER2+ BC, 93.8% previously diagnosed with metastatic BC, 89.8% with clinical symptoms. Among them, 40 were evaluable (CTC detection failure: n=8, eligibility criteria failure: n=1). Median sample volume was 3.0 mL for conventional cytology samples (median time to analysis: 22min) and 3.3 mL for CTC samples. Of the 40 evaluable patients, 18 had a positive cytology (on CSF sample n=°1/n°2: n=16/n=2) and were therefore diagnosed with LM using the gold-standard method. Protein level was elevated in 88.2% of these patients, compared with 45.1% of patients with negative CSF cytology (p=0.005). CTCs were detected in these 18 patients (median 5824 CTCs, range 93-45052). CTCs were also detected in 5/22 patients with a negative cytology (median 2 CTCs, range 1-44). Among them, one patient (44 CTCs) was diagnosed with a cytologically-proven LM 9 months later, while there was no further argument for LM in the other 4 patients’ history (1-3 CTC), who died of the extra-cerebral disease after a median time of 5.2 months (range 0.9-25.9). The detection of at least one CTC in CSF was associated with a Se of 100.0% (IC95% 82.4-100) and a Spe of 77.3% (IC95% 64.3-90.3) for the diagnosis of LM. Considering the number of CTC as a quantitative value, we determined the cut-off maximizing the Youden index using the ROC analysis (93 CTC). The detection of at least 93 CTC in CSF was associated with a Se and a Spe of 100.0% for the diagnosis of LM (area under the curve [AUC]: 1.0). Conclusion CTCs were detected with the CellSearch® system in all patients diagnosed with a cytologically-proven LM, as well as in a few patients without a cytological confirmation of LM. The prognosis of these patients with CSF cytology-/CTCs+ needs to be further investigated in a larger cohort. Garbage is matter which is serious enought to society especially in the main city. Increasing number of society will affect to increasing garbage either as resulted. Volume of garbage which al ways increase without measuring last process of garbage will appear many problems for example, leachate seepage. Leachate is a liquid waste resulted of biology decomposition. Leachate soaks to bottom groundwater will affect decreasing level of field water and allergy on the skin. The aim of this research is to know leahate accumulation and direct seepage arround landfill Sekoto. The method will be conducted by field research by geoelectric configuration diple-dipole. Firstly, the writter survey the location to know the condition of research. Then collected data with 5 line arround landfill Sekoto use resistivitymeter. The data will be process using software Res2dinv and Voxler. Software Res2dinv to show the leachate seepage base on resistivity rock value. Software Voxler is used to show direct seepage as 3 dimension. The research finding shows that lye had been permeated in the south of garbage-can with the leachate seepage reaching 6.79 meters. While, in the north of garbage-can, there was no leachate seepage found. In the west road of Garbage Dump and near to leachate waste pipe, it had been found the leachate seepage reaching 11.8 meters. In summary, the direction of leachate seepage in Garbage Dump is congruence with river flow and the distribution of leachate horizontally.

Abstract:
Coastal communities in Ambon City have local wisdom and traditions in use of natural resources which have continued to this day and have an impact on household food security, which involves women and men. This study aims to analyze gender equality and food security as well as the factors that influence decision making in the household. The research location is in South Leitimur District, Ambon City, especially in Leahari, Hukurila, Kilang and Naku State. The research method is descriptive qualitative where data is collected from key informants in each country. Data collection techniques through in-depth interviews, making field documentation and field observations. The research data were analyzed using Miles and Huberman qualitative analysis. The results show that gender equality and food security in coastal areas are seen in reproductive and productive activities, access to employment opportunities and forms of social control. The factors that influence household decision making on food security are the division of labor based on the appropriateness of work space, technological innovation in the use of natural resources and social and economic institutions that support decision making in the household. The conclusion of the study shows that gender equality and food security in the coastal areas of Ambon City have been implemented but need to be improved. The research recommendation is that it is necessary to increase the community's capacity for gender equality in development so that it has an impact on improving the welfare of life
