Using a spatially-resolving x-ray spectrometer system, toroidal impurity rotation in Alcator C-Mod plasmas was measured. The propagation of the rotational velocity from the edge to the core of the plasma column was measured during the L- to H-mode transition. Momentum transport was measured in both Ohmic and ICRF-heated discharges, which produced EDA and ELM-free H-modes. The momentum transport was modeled by a simplified diffusion model, in which momentum diffusivity was substantially higher than neoclassically predicted values.

Empirical energy confinement scalings play a crucial role in the design of tokamak fusion reactors, measuring how quickly energy is transported by turbulence from the fusion-producing core to conduction loss at the edge. Unfortunately, experiments often exhibit discontinuous changes in scaling behavior as the plasma parameters are varied, termed confinement transitions. Navigating these transitions requires an understanding of the physical origin and limits of confinement scalings, and is crucial for retiring the physics risk of extrapolating empirical results to future reactors. This thesis explores the connection between two universally observed transitions in tokamak transport: the Linear to Saturated Ohmic Confinement (LOC/SOC) transition and the concomitant intrinsic rotation reversal. Analysis and modeling of rotation reversal hysteresis experiments show that a single turbulent bifurcation underlies both transitions on Alcator C-Mod.
Plasmas on either side of the reversal exhibit different toroidal rotation profiles and therefore different turbulence characteristics despite profiles of density and temperature which are indistinguishable within measurement uncertainty. Elements of this bifurcation are also shown to persist for auxiliary heated L-modes. Within a reduced quasilinear transport model, the deactivation of subdominant (in linear growth rate and contribution to heat transport) ion temperature gradient (ITG) and trapped electron mode (TEM) instabilities is identified as the only possible change in turbulence across the reversal which is consistent with the measured profiles and inferred heat and particle fluxes. Experimental constraints on a possible change from strong to weak turbulence, outside the description of the quasilinear model, are also discussed.
These results indicate an explanation for the LOC/SOC transition that provides a mechanism for the hysteresis through the dynamics of subdominant modes and changes in their relative populations, and does not involve a change in the most linearly unstable ion-scale drift-wave instability. This work highlights the importance of considering the dynamics of the entire mode spectrum, and not just the dominant modes, in making predictions about transport and confinement regimes.

Des protocoles couvrant l'ensemble des étapes de la micropropagation depuis le conditionnement des pieds-mères, et l'introduction in vitro des explants primaires jusqu'à l'enracinement et le sevrage des plantules, ont été mis au point, en attachant une attention toute particulière à l'obtention d'un clonage efficace à long terme à partir de matériel juvénile et adulte. A cette occasion, des variations du mode de croissance des pousses développées in vitro correspondant à une disparition progressive de la croissance rythmique endogène caractéristique de l'espèce, ont été observées. L'étude des conditions permettant leur apparition a montré le rôle primordial de la BA, en présence de BA, l'importance de l'alimentation azotée en tant que facteur de contrôle du modèle de croissance. La réalisation de bilans azotes (n total, n insoluble, n soluble, n minéral, acides aminés et amides libres) au sein des vitroplants, a permis d'associer l'apparition des différents modes de croissance décrits à une accumulation d' n soluble (no#3#, nh#4#+, asparagine, arginine, proline) dans les tiges feuillées, indépendamment des effets induits par l'introduction in vitro elle-même. Ces dosages ont également montré l'importance du cal basal qui assure l'interface entre le milieu de culture et la tige feuillée. Diverses hypothèses permettant d'expliquer le rôle de l'azote dans les mécanismes de contrôle de la croissance rythmique du chêne, ont été proposées

We perform extensive analysis on the physics of L-mode tokamak fusion reactors to identify (1) a favorable parameter space for a large scale steady-state reactor and (2) an operating point for a minimum scale steady-state reactor. The identification of the large scale parameter space is part of the 2008 MIT Nuclear Systems Design Project, which also includes sustainability and economic optimizations to identify a plausible operating point for a large scale (a 14 m major radius) hydrogen production reactor dubbed HYPERION. Due to the potentially prohibitive capital cost (a $50 billion) and exorbitant thermal power (a 35 GWth) of HYPERION, we identify a conservative estimate for the minimum scale of a similar steady-state L-mode reactor of approximately 7.5 meters, half the size of HYPERION and only 20% larger than ITER. This minimum scale reactor would require an on-coil magnetic field of a 16 T and a blanket power density of ~ 5 MW/m 2 . It would produce 7 GWth of power with a power gain of 30, and it would operate far from all stability and confinement limits. To confirm the viability of this operating point, we perform various 1-D calculations. The crucial advantage of a steady-state (or fully non-inductive) reactor is that it is not limited by flux swing and can operate continuously, recharging its solenoid during operation. The crucial advantages of L-mode are that it avoids instabilities associated with edge localized modes (ELMs) and that it allows volumetric heating in the mantle due to the absence of a pedestal. Steady-state L-mode tokamak reactors could be the future of controlled fusion research and even play an important role in meeting the world's clean energy needs.

Le laboratoire qui m'a accueilli en thèse a identifié le régulateur de l'homéostasie du fer, l'hepcidine. La production d'hepcidine est modulée en fonction des demandes en fer de l'organisme : en présence de fer, la synthèse d'hepcidine est augmentée pour éviter l'excès du métal qui est toxique, et, à l'inverse, l'hepcidine est diminuée pour permettre une mobilisation efficace du métal. J'ai participé à la caractérisation du phénotype de souris KO Hepcl qui présentent une surcharge en fer multiviscérale avec une atteinte hépatique et pancréatique particulièrement importante. Dans ce modèle murin, j'ai cherché à établir le lien entre métabolisme du fer et métabolisme du glucose. J'ai étudié les mécanismes de régulation physiopathologique de l'hepcidine par le fer dans un système d'hépatocyte en culture primaire. Ce système de culture d'hépatocytes m'a permis d'étudier la ferroportine, l'exporteur de fer et les mécanismes de mobilisation du fer de l'hépatocyte
The laboratory which welcomed me in thesis identified the regulator of the iron homeostasis, hepcidin. The production of hepcidin is modulated according to the iron demands of the body : in the presence of iron, the synthesis of hepcidin is increased to avoid the excess of the metal which is toxic, and, on the contrary, hepcidin is decreased to allow an effective mobilization of the metal. I participated in the characterization of the phenotype of mouse KO HEPC1 which present a multivisceral iron overload with a particularly important hepatic and pancreatic accumulation of iron. In this murin model, I tried to establish the link between the iron metabolism and the glucose metabolism. I studied the mechanisms of physiopathological regulation of the hepcidin by the iron in a system of primary culture of hepatocytes. This system of culture of hepatocytes allowed me to study ferroportin, the iron excarrier and the mechanisms of mobilization of the iron of the hepatocyte

Cette thèse a pour objectif de dégager les principaux enjeux liés à la réalisation d'un travail en équipe pluridisciplinaire en milieu hospitalier pédiatrique. Pour cela, dans la première partie, l'auteur passe en revue les notions de pluridisciplinarité, d'interdisciplinarité et de transdisciplinarité. Deux études sur des équipes de recherche multidisciplinaires sont examinées. Une analyse de l'évolution du concept de sante est suivie d'un recensement de la littérature concernant la nécessite du travail en équipe de sante pluriprofessionnelle et de sa pratique dans les milieux hospitaliers psychiatrique, général et pédiatrique. Enfin, les travaux traitant directement des enjeux de l'équipe de sante pluriprofessionnelle sont présentés. La deuxième partie est consacrée a la recherche sur le terrain. Des observations ont été menées dans quatre services hospitaliers parisiens -unité d'hémodialyse pédiatrique, unité de greffe de moelle osseuse, service de pédiatrie générale et service de cardiologie infantile- pendant les réunions de synthèse. Les échanges ver- baux entre les divers professionnels sont analyses sous les angles quantitatifs et qualitatif. This thesis aims to underline the main stakes connected to the realization of a work in a pluridisciplinary team in pediatric wards. To achieve this, in the first part, the author has a review of the notions of pluridisciplinary, interdisciplinary and transdisciplinary. Two studies on multidisciplinary research teams are examined. An analysis of the evolution of the health concept will be followed by a census on litterature concerning the necessity of a health pluriprofessional group work and its practise in psychiatric, general and pediatric wards. To finish this subject, the works considering directly the stakes of a pluriprofessional health team, will be presented. The second part of this thesis is devoted to the practical research. Observations were led during the synthesis meetings in four parisian wards : -unity of pediatric hemodialisis, unity of bone marrow transplant, ward of general pediatric and ward of infantile cardiology. What was said between all the present professionnals, is analysed with a quantitative and qualitative approach. The author concludes to the sporadic aspect of the work in a pluridisciplinary team in a pediatric ward in the sample hereby studied. (431 ref. )

As swedes are keen on outdoor recreation and like to spend on travels and gear for forest recreation we want to show if there is a willingness to pay for forest recreation close to home. Starting in hedonic regression we create a model using publicly available data to show how consumer preferences effect house prices. Results show that distance to forest have a small but significant impact on house prices. To conclude we state that a logarithmic model using open source data can be useful in city planning and that there is a positive effect of nearby forests on house prices.

The most prevalent criticism of fission-fusion hybrids is simply that they are too exotic - that they would exacerbate the challenges of both fission and fusion. This is not really true. Intriguingly, hybrids could actually be more viable than stand-alone fusion reactors while mitigating many challenges of fission. This work develops a conceptual design for a fission-fusion hybrid reactor in steady-state L-mode tokamak configuration with a subcritical natural or depleted uranium pebble bed blanket. A liquid lithiumlead alloy breeds enough tritium to replenish that consumed by the D-T fusion reaction. Subcritical operation could obviate the most challenging fuel cycle aspects of fission. The fission blanket augments the fusion power such that the fusion core itself need not have a high power gain, thus allowing for fully non-inductive (steady-state) low confinement mode (L-mode) operation at relatively small physical dimensions. A neutron transport Monte Carlo code models the natural uranium fission blanket. Maximizing the fission power while breeding sufficient tritium allows for the selection of an optimal set of blanket parameters, which yields a maximum prudent fission power gain of 7.7. A 0-D tokamak model suffices to analyze approximate tokamak operating conditions. If the definition of a "reactor" is a device with a total power gain of 40, then this fission blanket would allow the fusion component of a hybrid reactor with the same dimensions as ITER to operate in steady-state L-mode very comfortably with a fusion power gain of 6.7 and a thermal fusion power of 2.1 GW. Taking this further can determine the approximate minimum scale for a steady-state L-mode tokamak hybrid reactor, which is a major radius of 5.2 in and an aspect ratio of 2.8. This minimum scale device operates barely within the steady-state L-mode realm with a thermal fusion power of 1.7 GW. This hybrid, with its very fast neutron spectrum, could be superior to pure fission reactors in terms of breeding fissile fuel and transmuting deleterious fission products. It could operate either as a breeder, producing fuel for pure fission reactors from natural or depleted uranium, or as a deep burner, fissioning heavy metal and transmuting waste with a cycle time of decades. Despite a plethora of potential functions, its primary mission is deemed to be that of a deep burner producing baseload commercial power with a once-through fuel cycle. Although hybrids are often purported a priori to pose an elevated proliferation risk, this reactor breeds plutonium that could actually be more proliferation-resistant than that bred by fast reactors. Furthermore, a novel method (the "variable fixed source method") can maintain constant total hybrid power output as burnup proceeds by varying the neutron source strength. As for engineering feasibility, basic thermal hydraulic analysis demonstrates that pressurized helium could cool the pebble bed fission blanket with a flow rate below 10 m/s. The Brayton cycle thermal efficiency is 41%. This device is dubbed the Steady-State L-Mode Non-Enriched Uranium Tokamak Hybrid (SLEUTH). The purpose of this work is not any sort of elaborate design, but rather the exploration of an idea coupled with corroborating numerical analysis. At this point in the hybrid debate, viable conceptual designs are persuasive while intricate build-ready designs are superfluous. This work conceives such a conceptual design, demonstrates its viability, and will perhaps, incidentally, spur a profusion of pro-fusion sentiment!

Recognizing the current and future requirements of extending Telemetry, TT&C, Earth Resources operations into the C-Band (4400-5200 MHZ) arena, Orbit Communication Systems and Orbit Communication Ltd. are currently designing and testing a Tri-Band auto-tracking / receive / transmit feed technology combining dual polarization with multiple frequency band coverage (L/S/C) into a single feed that can be fitted onto new or existing antenna systems. This technology reduces footprint and minimizes life cycle cost, both of which are important considerations for both our commercial and military customers. This technology has been applied to a number of systems, including flyaway, fixed (or teleport) and shipboard, and is readily applicable to ground and mobile applications. The multiband feed design consists of coaxial, concentric waveguide cavities operating in TE11 and TE21 modes (Patent Pending). The large outermost cavities of the feed operate in the L/S frequency band while the innermost cavities operate at C-Band. The antenna is fed with orthogonally polarized inputs/outputs enabling polarization diversity in all bands. The coaxial cavity feed is ideal for this application because the feed produces high-efficiency, near-optimum illumination patterns and coincident phase centers in all three bands simultaneously. Because it is a waveguide design, the input power levels used in Telemetry and Satellite systems do not present breakdown problems. This type of feed has proved to be an ideal multiband illuminator for parabolic reflectors, and therefore could be used in many military and commercial applications that require multiband operation.

La présente recherche pluridisciplinaire a consacré beaucoup d'efforts au développement d'une analyse spécifique de quantification du comportement de "délestage" des automobilistes, liée à la diffusion d'une information routière en temps réel par panneau à message variable. La gestion des encombrements récurrents journaliers représente une part très importante de l'activité des exploitants autoroutiers en zone urbaine et périurbaine. Il nous a donc semblé opportun de s'intérésser à l'évaluation socio-économique du comportement de l'usager face à l'information routière dynamique. Cette analyse a aboutit à la détermination d'un critère d'évaluation "le taux de délestage". Celui-ci permet de calculer le gain de temps passé en circulation pour la collectivité et indique l'efficacité de l'action d'information des usagers. Nous montrons que les résultats de l'analyse quantitative du comportement des usagers en terme de gains de temps s'intègrent bien dans le cadre de l'évaluation des infrastructures en Ile-de-France. Cette recherche nous a amené à réfléchir sur les améliorations qu'il serait opportun d'apporter à la stratégie d'affichage des messages sur les panneaux.

This paper specifically addresses the manufacture, integration, acceptance testing, installation and use of two 10-Foot / 3.0-Meter Tri-band Telemetry Tracking Systems installed and operated at Patuxent River NAS, Maryland under the auspices of NAVAIR Atlantic Test Range, Telemetry Systems Branch. What is described herein is the final configuration as delivered, antenna test range results, on-site results, and lessons-learned during the development and implementation phases. Charts and tables are presented to assist with clarification of relevant data for the reader.

Abstract:
Virtually all methods of learning dynamic models from data start from the same basic assumption: that the learning algorithm will be provided with a single or multiple sequences of data generated from the dynamic model. However, in quite a few modern time series modeling tasks, the collection of reliable time series data turns out to be a major challenge, due to either slow progression of the dynamic process of interest, or inaccessibility of repetitive measurements of the same dynamic process over time. In most of those situations, however, we observe that it is easier to collect a large amount of non-sequence samples, or random snapshots of the dynamic process of interest without time information. This thesis aims to exploit such non-sequence data in learning a few widely used dynamic models, including fully observable, linear and nonlinear models as well as Hidden Markov Models (HMMs). For fully observable models, we point out several issues on model identifiability when learning from non-sequence data, and develop EM-type learning algorithms based on maximizing approximate likelihood. We also consider the setting where a small amount of sequence data are available in addition to non-sequence data, and propose a novel penalized least square approach that uses non-sequence data to regularize the model. For HMMs, we draw inspiration from recent advances in spectral learning of latent variable models and propose spectral algorithms that provably recover the model parameters, under reasonable assumptions on the generative process of non-sequence data and the true model. To the best of our knowledge, this is the first formal guarantee on learning dynamic models from non-sequence data. We also consider the case where little sequence data are available, and propose learning algorithms that, as in the fully observable case, use non-sequence data to provide regularization, but does so in combination with spectral methods. Experiments on synthetic data and several real data sets, including gene expression and cell image time series, demonstrate the effectiveness of our proposed methods. In the last part of the thesis we return to the usual setting of learning from sequence data, and consider learning bi-clustered vector auto-regressive models, whose transition matrix is both sparse, revealing significant interactions among variables, and bi-clustered, identifying groups of variables that have similar interactions with other variables. Such structures may aid other learning tasks in the same domain that have abundant non-sequence data by providing better regularization in our proposed non-sequence methods.

Cependant, à travers des procédures de commande et de contrôle, le contexte de mise en œuvre a totalement changé. Les directeurs de commande numérique sont de plus en plus complexes. La programmation CN doit être optimisée et contrôlée. Cette évolution nécessite le développement d'interface homme - machine interactive et conviviale, cela induit des contraintes en formation et en utilisation. A travers l'élaboration d'un didacticiel destiné à acquérir la maîtrise de la programmation des machines-outils à commande numérique en langage ISO, on a abordé la dimension pédagogique d'un des domaines de la mécanique. Par ailleurs, une vérification de la justesse des programmes CN est réalisée. Le contexte opérationnel (spécificité du directeur de commande, choix des outils) a été traité afin de mettre l'opérateur en situation. Une interface conviviale, secondée par différents modes d'aides, autorise l'emploi de ce module par des usagers débutants ou confirmés. Une analyse syntaxique, lexicale et grammaticale des informations saisies est effectuée en temps réel de manière à adresser à l'utilisateur des messages pertinents adaptés à son niveau. Le module d'analyse utilise un moteur d'inférence spécifique et des structures algorithmiques en langage C++. Développé en liaison avec des partenaires industriels, le didacticiel réalisé montre la faisabilité de produits fortement interactifs intégrant les contraintes réelles du monde industriel
Current manufacturing process of mechanical pieces have shortly evolved in relation to the used physical process. However, through procedures of control, the context of implementation has totally changed. CNC are increasingly complex. The NC programming has to be optimised and controlled. This evolution necessitates the development of convivial and interactive Human/machine interfaces that induced constraints in training and in utilisation. Through the elaboration of a teaching software destined for acquire the ability to programme CNC machine tool with ISO language, we approached the pedagogical dimension of mechanic. Furthermore, a verification of the precision of CNC programs is realised. The operational context (Characteristic of CNC, choice of tools) has been processed so as to put the operator in the context. A convivial interface, seconded by different types of help, authorises the use of this module by beginners or by experts. Syntactic, lexical and grammatical analysis of typed information's is undertaken in real manner time in order to address to the user relevant messages adapted at its level. The analysis module uses an inference method and algorithmic one, they are developed in C++ language. Developed with collaboration of industrial partners, the software realised shows the feasibility of strongly interactive products integrating real constraints of the industrial world

Abstract:
La regression des herbiers de posidonia oceanica, phanerogames marines de mediterranee, nous a amene a etudier les parametres qui agiraient sur la survie et le developpement de boutures transplantees. La meilleure saison de transplantation pour la survie et le developpement des boutures plagiotropes est le printemps. Pour les boutures orthotropes, les transplantations d'automne donnent les meilleurs taux de survie et celles du printemps le meilleur developpement. Les taux de survie sont superieurs a 74% pour les boutures portant au moins deux faisceaux foliaires au moment de la transplantation. La ramification s'observe des les premiers mois pour les boutures plagiotropes alors qu'elle n'apparait qu'apres une annee pour les boutures orthotropes. Les boutures prelevees en profondeur (30 m) survivent bien lorsqu'elles sont transplantees a plus faibles profondeurs. Inversement, lorsqu'elles sont prelevees pres de la surface (3 m), elles survivent et se developpent d'autant plus mal que leur profondeur de transplantation est elevee. Sur du sable a forte bioturbation et sur la matte morte, les boutures survivent mieux lorsque l'espacement entre elles est inferieur a 10 cm. Sur un substrat sableux stable, l'espacement entre les boutures a peu d'incidence. Un effet de la transplantation sur l'induction de la floraison a ete mis en evidence. L'analyse architecturale des boutures transplantees depuis trois ans et des rhizomes poussant dans les herbiers a differentes profondeurs, a conduit a une modelisation mathematique du fonctionnement de la plante. Les unites de croissance plagiotropes des axes d'ordre 1 presentent un nombre d'entre-nuds formes en un an, variant de 5 a 25 et distribues selon une loi de poisson. Elles peuvent porter une ou deux series independantes d'axes d'ordre 2. Les unites de croissance plagiotropes des axes d'ordre 2 presentent un nombre d'entre-nuds formes en un an, variant de 5 a 15 et generalement distribues selon une loi binomiale, presentant aussi deux zones preferentielles de ramification. Pour les unites de croissance orthotropes, la frequence d'apparition des series de rameaux est beaucoup plus faible et le nombre de feuilles formees en un an varie de 5 a 12. La viabilite des meristemes est plus faible sur les boutures que sur les rhizomes preleves dans les differents herbiers. La croissance de posidonia oceanica, est controlee par une horloge interne virtuelle, qui fournit trois tops sur une annee

The common observation that the onset of a core MARFE (edge localized, poloidally asymmetric, highly radiating region) is followed immediately by a High-to-Low confinement mode transition in DIII-D was investigated by comparing a theoretical prediction of the threshold non-radiative power across the separatrix needed to maintain H-mode with an experimental determination of the non-radiative power flowing across the separatrix. It was found that in three shots with continuous gas fueling that the increased neutral influx associated with the MARFE formation caused a sharp increase in the predicted threshold non-radiative power crossing the separatrix that was required for the plasma to remain in H-mode to a value comparable to the experimental power crossing the separatrix, indicating a theoretical prediction of a H-L transition in agreement with experimental observation.

Dans cette thèse, nous nous intéressons au problème de la prévision spatiale en considérant des modèles non paramétriques conditionnels dont la variable explicative est fonctionnelle. Plus précisément, les points étudiés pour décrire la co-variation spatiale entre une variable réponse réelle et une variable fonctionnelle sont le mode conditionnel et les quantiles conditionnels.En ce qui concerne le mode conditionnel, nous établissons la convergence presque complète, la convergence en norme Lp et la normalité asymptotique d'un estimateur à noyau. Ces propriétés asymptotiques sont obtenues sous des conditions assez générales telles, l'hypothèse de mélange forte et l'hypothèse de concentration de la mesure de probabilité de la variable explicative fonctionnelle. L'implémentation de l'estimateur construit en pratique est illustrée par une application sur des données météorologiques.Le modèle des quantiles conditionnels est abordé dans la deuxième partie de la thèse. Il est traité comme fonction inverse de la fonction de répartition conditionnelle qui est estimée par un estimateur à double noyaux. Sous les mêmes conditions que celles du modèle précédent, nous donnons l'expression de la vitesse de convergence en norme Lp et nous démontrons la normalité asymptotique de l'estimateur construit.Notre étude généralise au cas spatial de nombreux résultats déjà existant en série chronologique fonctionnelle. De plus, l'estimation de nos modèles repose sur une estimation préalable de la densité et de la fonction de répartition conditionnelles et permet de construire des régions prédictives, montrant ainsi l'apport de ce genre de modèles par rapport à la régression classique
The main purpose of this thesis concerns the problem of spatial prediction using some nonparametric conditional models where the covariate variable is a functional one. More precisely, we treat the nonparametric estimation of the conditional mode and that of the conditional quantiles as spatial prediction tools alternative to the classical spatial regression of real response variable given a functional variable.Concerning the first model, that is the conditional mode, it is estimated by maximizing the spatial version of the kernel estimate of the conditional density. Under a general mixing condition and the concentration properties of the probability measure of the functional variable, we establish the almost complete convergence (with rate), the Lp consistency (with rate) and the asymptotic normality of the considered estimator. The usefulness of this estimation is illustrated by an application on real meteorological data.The model of the conditional quantiles is considered in the second part of this thesis and is treated as the inverse function of the conditional cumulative distribution function which is estimated by a double kernel estimator. Under the same general conditions as in the first model, we give the convergence rate in the Lp- norm and we show the asymptotic normality of the constructed estimator. These asymptotic results are closely related to the concentration properties on small balls of the probability measure of the underlying explanatory variable and the regularity of the conditional cumulative distribution function.Our study generalizes to spatial case some existing results in functional times series case. Finally, we highlight what our models brings compared to classical regression, discussing the use of our results as preliminary works to construct predictive regions

De par sa position phylogénétique idéale à la base des Vertébrés, la petite roussette est un modèle d’étude émergent pour l’étude de processus biologiques fondamentaux tel la spermatogenèse, et la recherche de peptides bioactifs. Durant ce travail, nous avons tout d’abord montré que GFRα1et POU2/POU5F1 sont des marqueurs de spermatogonies indifférenciées conservés entre Mammifères et Chondrichtyens alors que PLZF et c-KIT présentent un profil moins spécifique et que HMGB2, HMGB3 et MCM6 seraient de nouveaux marqueurs potentiels. Par la suite, le développement d’un système de culture et d’étude des spermatogonies sur un temps long nous a permis de mettre en évidence la conservation de la fonction du GDNF dans le maintien des spermatogonies souches chez la roussette. Cependant, les études de séquences suggèrent que l’ARTN remplacerait le GDNF dans le maintien de l’auto-renouvèlement des spermatogonies souches via le récepteur GFRα1 chez la roussette. En parallèle, l’étude du mode d’action de trois peptides bioactifs issus du testicule de roussette et de deux autres issus du tissu épigonal a montré des capacités à altérer la croissance de lignées cancéreuses en affectant divers processus cellulaires. Ainsi, bien que deux peptides n’aient pas été davantage caractérisés, les trois autres semblent affecter directement le cytosquelette des cellules et provoquer différents effets secondaires, en fonction des couples lignée/peptide, comme le décalage du cycle cellulaire, l’inhibition de l’autophagie, la nécrose ou encore la déstabilisation membranaire. Ces résultats confirment l’intérêt du modèle roussette pour ces études
Due to its phylogenetic position at the base of the phylogenetic tree of Vertebrates, the dogfish is an emergent model to study fundamental biological process such as spermatogenesis and to identify bioactive peptides. During this work, we have shown that GFRα1and POU2/POU5F1 are conserved markers of undifferentiated spermatogonia from Chondrychtyes to Mammals whereas PLZF and c-KIT presented a less specific expression pattern and that HMGB2, HMGB3 and MCM6 may be new markers. Then, the development of a culture system of dogfish spermatogonia allowed us to show that GDNF function on self-renewal maintenance was also conserved. However, sequences study showed that the ARTN may replace the GDNF to induce self-renewal through GFRα1 in the dogfish. In parallel, the study of the action mode of three peptides isolated from dogfish testis and of two from the epigonal tissue showed antineoplastic effects by affecting various cellular processes. Thus, even if two peptides were further characterized, the three others seem to directly affect the cytoskeleton, which was followed by various cellular perturbations depending on the cell line and the peptide, such as cell-cycle slowing down, autophagy inhibition, necrosis or membrane destabilization. Complementary studies must be done to confirm the first target and identify which cytoskeleton element is involved. Those results suggest finally that the dogfish is a suitable model to study spermatogenesis evolution as well as to search bioactive peptides

Notre travail a suivi deux objectifs principaux, l'une sur la variabilite et le mode d'infection de phoma macdonaldii et l'autre sur l'etude genetique du tournesol vis-a-vis de cette maladie. Les resultats suivants ressortent de cette etude : 1) les souches de l'agent pathogene presentent une forte variabilite phenotypique au niveau de la croissance, de la production et de la taille des pycnides ainsi que la germination des pycniospores. Une methode de contamination artificielle a ete developpee pour apprecier le niveau d'agressivite des souches ainsi que le comportement du tournesol vis-a-vis de cette maladie. Les souches du pathogene montrent une agressivite tres variable. Aucune correlation entre l'agressivite des souches et les caracteres phenotypiques etudiees n'est observee. 2) l'etude du mode d'infection montre que, les spores sont entourees par une couche mucilagineuse de nature polysaccharidique qui joue un role important dans l'adhesion de l'agent pathogene a la surface de l'hote. Ce champignon developpe un coin d'infection qui perfore la cuticule, et penetre directement dans la plante par la cuticule et/ou par les stomates. Apres avoir traverse la cuticule il forme des hyphes subcuticulaires. 3) l'etude genetique montre une grande variabilite au niveau de la resistance entre les genotypes. Nous avons observe le phenomene d'heterosis ainsi qu'un effet nucleo-cytoplasmique sur la resistance du tournesol a cette maladie. Notre etude sur l'aptitude generale a la combinaison des differents genotypes du tournesol, montre que ces valeurs sont significative (positive ou negative). Ce qui prouve l'importance des effets additifs des genes dans le controle de la resistance. L'aptitude specifique a la combinaison de certains hybrides f1 est significative. Ce qui prouve l'existence de l'effet dominant et ou dominant incomplet de genes pour le controle de la resistance a cette maladie.

In campo medico ha un ruolo fondamentale il monitoraggio dei parametri vitali, importanti indicatori dello stato di salute del paziente, essenziali nella diagnosi e nella cura dellle patologie. La frequenza cardiaca occupa un ruolo fondamentale nella clinica, primo indice dell'attività cardio circolatoria del paziente. Il trattato analizza una metodica di monitoraggio del battito cardiaco per mezzo delle videocamere digitali, la ripresa diretta in video del volto del paziente e la presenza di un'adeguata illuminazione ambientale: la pulsazione cardiaca esercita variazioni di pressione a livello periferico generando un cambiamento del colore della pelle sulle zone più esposte, quali il viso. Analizzando l'andamento temporale del segnale RGB registra, per mezzo di algoritmi di elaborazione delle immagini, è possibile stimare accuratamente il battito cardiaco dell'utente. Nell'elaborato viene affrontata la metodica degl algoritmi di image-prcessing presentando un applicativo scritto in linguaggio Python, funzionante e testato su di un sistema Linux (Ubuntu 14.04). Tale metodo si basa sui concetti della fotopletismografia applicata in remoto denotando il non-contact-mode.

Alors que la sociologie et l'histoire se sont intéressées de longue date à l'étude de l'influence du facteur religieux sur les pratiques économiques, les sciences du management en ont découvert l'intérêt depuis peu. C'est dans le champ de l'éthique des affaires, principalement aux Etats-Unis, que cette question suscite, depuis le milieu des années 1990, des recherches en nombre croissant. Au même moment, les méthodologies dominantes de cette disicpline, à caractère quantitatif, sont contestées. Dans ces conditions, l'examen de l'influence des valeurs catholiques du dirigeant de PME sur son mode de management constitue une double tentative d'explorer, d'une part, un thème nouveau dans les sciences du management et de l'éthique des affaires françaises et, d'autre part, de nouvelles voies méthodologiques pour y parvenir. La stratégie de recherche qualitative adoptée, qui s'inspire notamment des approches de Glaser, Strauss et Corbin, permet d'obtenir trois grands types de résultats : la construction d'un idéal du management selon la doctrine sociale de l'Eglise Catholique, l'élaboration d'un modèle encore exploratoire d'analyse de l'influence des valeurs catholiques du dirigeant de PME lorsqu'elles sont confrontées aux exigences et aux contraintes du management des personnes en entreprise, et enfin la mise au point d'un "idéal-type" du dirigeant de PME catholique. Le caractère pluridisciplinaire et biculturel du travail accompli (recherche francophone et anglophone) suscite, en conclusion, des questions épistémologiques sur le paradigme le plus apte à embrasser la nature à la fois plurielle et synthétique du modèle proposé.

Resumo: Introdução: A candidíase oral é uma infecção fúngica oportunista da cavidade oral, cujas taxas de prevalência e incidência vêm aumentando significativamente em todo o mundo. Assim, novas estratégias orientadas para gerir esta doença têm sido propostas, dentre as quais está o uso de óleos essenciais (OE) com propriedades antifúngicas. Evidências indicam que o OE de Coriandrum sativum L. (coentro) é um forte agente antifúngico contra Candida e, portanto, investigações devem dar continuidade ao conhecimento gerado. Objetivo: Este estudo buscou avaliar a atividade antifúngica e modo de ação do OE de C. sativum sobre Candida spp., e determinar os alvos moleculares afetados na expressão global do genoma humano. Material e Métodos: C. sativum foi obtido a partir do Banco de Germoplasmas do Centro Pluridisciplinar de Pesquisas Químicas, Biológicas e Agrícolas (Universidade Estadual de Campinas, SP, Brasil) cujo OE e fração ativa tiveram o perfil fitoquímico determinado por cromatografia gasosa acoplada a espectrometria de massa. Posteriormente, foram realizados testes com cinco cepas de referência de Candida: Determinação da Concentração Inibitória e Fungicida Mínima (CIM/CFM); modo de ação antifúngica (ensaio do sorbitol e ergosterol); Microscopia Eletrônica de Varredura (MEV) de biofilmes de Candida e testes de inibição de aderência em biofilme. Utilizou-se nistatina, anfotericina B ou caspofungina como controles positivos, além de controles negativos. Também, foi testado o efeito do OE sobre a atividade proteolítica de C. albicans. Por fim, um ensaio de farmacogenômica identificou quais alvos moleculares no genoma humano foram afetados pelo OE e fração ativa de C. sativum. Os testes foram realizados em triplicata de experimentos independentes e os dados foram tratados estatisticamente (ANOVA, pós-teste de Tukey, ?=0,05). Os dados da análise farmacogenômica foram processados nas plataformas GeneGo MetaCore® e David Bioinformatics Resources. Resultados: O perfil fitoquímico EO indicou monoterpenos (37,9%) e sesquiterpenos (62,1%) como compostos principais. Os valores de CIM/CFM para o OE variaram de 15,6 a 62,5 µg/mL. Quanto ao modo de ação, o OE de C. sativum parece se ligar ao ergosterol da membrana celular fúngica, aumentando a permeabilidade iônica e causando morte celular; entretanto, o OE não atua sobre vias de biossíntese da parede celular. Estes achados confirmam as alterações na integridade da morfologia do biofilme verificadas nas análises por MEV. Além disso, o OE apresentou atividade antiaderente em biofilme em baixas concentrações (15,6-62,5 µg/mL) contra as cepas testadas, bem como atividade contra proteases produzidas por C. albicans, sendo estatisticamente significante na CIM (p<0,05). Finalmente, o OE e sua fração ativa apresentaram baixa citotoxicidade em células humanas com CI30 de 359,8 e 366,7 µg/mL, respectivamente. As principais vias afetadas estão relacionadas com quimiocinas e MAP-quinase (apoptose, proliferação) bem como proteínas de adesão. Conclusões: O OE das folhas de C. sativum tem forte atividade antifúngica e antiaderente sobre Candida spp. e atividade anti-proteolítica sobre C. albicans, e atua aumentando a permeabilidade iônica da membrana celular, provavelmente devido ao efeito sinérgico de mono e sesquiterpenos. Análise farmacogenômica indicou baixa citotoxicidade do OE e sua fração ativa com alvos moleculares específicos afetados no genoma humano, o que incentiva o desenvolvimento de novas pesquisas pré-clínicas toxicológicas e clínicas nesta área
Abstract: Introduction: Oral candidiasis is a common opportunistic fungal infection of the oral cavity with increasingly significant worldwide prevalence and incidence rates. Accordingly, novel specific-targeted strategies to manage this ailment have been proposed, among which is the use of essential oils (EO) with antifungal properties. Coriandrum sativum L. (coriander) EO has proven antifungal activity against Candida species and thus deserves further investigation. Objective: This study aimed to evaluate the antifungal activity and mode of action of the EO from Coriandrum sativum L. leaves on Candida spp., and to determine the molecular targets affected in the whole-genome expression in human cells. Material and Methods: C. sativum was obtained from the Germoplasm Bank of the Research Center for Chemistry, Biology and Agriculture (University of Campinas, SP, Brazil) whose EO and active fraction had their phytochemical profile determined by Gas Chromatography coupled to Mass Spectrometry. Then, we carried out the following tests with five reference strains of Candida spp. (CBS): Minimum Inhibitory and Fungicidal Concentration (MIC/MFC); antifungal mode of action (sorbitol and ergosterol assays); Scanning Electron Microscopy (SEM) analysis of Candida biofilm and tests of inhibition of biofilm adherence. We used nystatin, amphotericin B or caspofungin as positive controls, in addition to negative controls. Also, we tested the effect of C. sativum EO on the proteolytic activity of C. albicans. Next, a pharmacogenomic assay identified which molecular targets in the human genome were affected by C. sativum EO and its active fraction. Tests were performed in triplicate of independent experiments and data were statistically treated (ANOVA, Tukey¿s post-test, ?=0.05). Pharmacogenomic data were processed on GeneGo MetaCore® and DAVID Bioinformatics Resources. Results: The EO phytochemical profile indicated monoterpenes (37.9 %) and sesquiterpenes (62.1 %) as major compounds. The MIC/MFC values for the EO ranged from 15.6 to 62.5 µg/ml. With regard to the mode of action, C. sativum EO may bind to membrane ergosterol, increasing ionic permeability and causing membrane damage to cell death, but it does not act on cell wall biosynthesis-related pathways. This mode of action confirms the changes in the integrity of the biofilm morphology as verified in the analyses by SEM. The EO showed anti-adherent activity at low concentrations (31.2 ¿ 62.5 µg/ml) against all strains tested, as well as activity against proteases produced by C. albicans, with statistical significance at MIC (P < 0.05). Finally, the EO and its active fraction had low cytotoxicity on human cells with IC30 of 359.8 and 366.7 µg/ml, respectively, affecting the pathways of chemokines and MAP-kinase (apoptosis, proliferation), as well as adhesion proteins. Conclusions: The EO from C. sativum leaves has strong antifungal and anti-adherent activity against Candida spp., as well as anti-proteolytic activity against C. albicans, and acts by increasing cell membrane ionic permeability, probably due to the synergistic effect of mono- and sesquiterpenes. Pharmacogenomic analyses revealed low cytotoxicity with specific targets affected in the human genome, which encourages further pre-clinical toxicological screening and clinical research in this field

La S-adénosyl-L-homocystéine hydrolase (AdoHcy hydrolase) est considérée aujourd'hui comme une bonne cible pharmacologique pour la conception de nouveaux antiviraux à spectre large. Tout au long de cette étude, nous avons cherché à exploiter le mécanisme d'action de l'enzyme et les données structurales récemment publiées pour provoquer son inhibition. La famille des esters homologues de l'adénosine s'est révélée être une famille d'inhibiteurs de type I affectant le cofacteur NAD+. Les thionucléosides ont atteint leur cible : la Cystéine 195. L'étude de la protéine inactivée en ESI-MS puis l'étude des peptides obtenus après digestion par l'EndoLys C de l'enzyme inhibée nous permet de confirmer le mécanisme d'action de ces composés. L'évaluation des acyclonucléosides cible est toujours en cours mais les résultats préliminaires demeurent encourageants et ouvrent la voie vers de nouvelles structures

The family of esters homologous of adenosine was revealed to be a family of the type I inhibitors assigning cofactor NAD+. The thionucleosides reached their target : Cysteine 195. Studies of inactivated protein in ESI-MS then studies of the peptides obtained after digestion by EndoLys C of inhibited enzyme enable us to confirm the mechanism of action of these compounds. Les résultats montrent que la teneur en cendres, protéines brutes, sucres disponibles, oxalates totaux, amidons et phosphore augmente significativement (p<0,05) avec la maturité. En revanche la teneur en amylose baisse avec la maturité. La capacité d'absorption d'eau, l'indice de solubilité, la température et l'enthalpie de gélatinisation (delta H) des farines et amidons augmentent significativement (p<0,05) en fonction du degré de maturité. Quel que soit le degré de maturité, les granules d'amidon ont présenté un diffractogramme caractéristique de type "A". Les précurseurs de brunissement augmentent significativement (p<0,05) avec le degré de maturité. Le paramètre L* des farines et des pâtes augmente alors que a* diminue en fonction du degré de maturité. Le paramètre L* est plus élevé et a* moins élevé dans le cas du séchage électrique que le séchage solaire. Les caractéristiques sensorielles des pâtes de taro Sosso sont affectées positivement par le degré de maturité et le séchage électrique. L'acceptabilité générale des pâtes de taro Sosso est corrélée à la couleur de celles-ci. Cette étude suggère que 10 mois de maturité de taro Sosso et le séchage électrique (40°C) sont des conditions favorables à l'obtention des pâtes de qualité acceptable
Results showed that the ash content, crude proteins, available sugars, total oxalate, starch and phosphorus content increased significantly with the maturity stage. On the other hand the amylose content decreased as function of the maturity. The water adsorption capacity, the water solubility index, the temperature and the enthalpy of gelatinization of the flour and starch increased significantly with the maturity stage. Irrespective of the degree of maturity the starch granules diffractogramm were exhibited an "A" type cristallinity. The browning indicators increased significantly (p<0.05) with the maturity stage of the tuber. The color coordinate L* of the flours and that of its dough increased while a* decreased with the maturity stage. These two coordinates lend themselves better with electric drying than solar drying. The sensory characteristics of the taro Sosso dough were positively affected by the maturity stage and electric drying. The overall acceptability of taro Sosso dough was correlated with its color. These results suggest that 10 months of the taro Sosso maturity and the electric drying (40°C) are the best conditions to obtain the acceptable pastes. These optimum conditions might be used to produce the taro flour at industrial scale

Il sistema è perciò composto da due parti distinte con compiti ben precisi e facilmente ampliabili (o modificabili) in futuro: un'applicazione Android in grado di riconoscere il Mode of Transport dell'utente e di tracciare i suoi spostamenti e una parte Server, sviluppata per ricevere i dati rilevati dall'applicazione Android, memorizzarli e infine mostrarli graficamente agli utenti che desiderano visualizzarli. Questa tesi ha studiato a fondo le modalità di funzionamento del convertitore ZETA. Si è visto che la presenza dei due magnetici determina una condizione di funzionamento non convenzionale (lo stesso accade nel SEPIC) poco studiata in letteratura. Questa condizione, corrispondente al modo discontinuo nei più elementari convertitori, in cui la corrente si annulla sia nel transistor che nel diodo, dà invece luogo ad un ricircolo di corrente pressochè costante in una maglia che comprende entrambe le induttanze. Questa corrente testimonia un intrappolamento di energia magnetica con relativa perdita per dissipazione che presumibilmente degrada l’efficienza del convertitore. Questo è potuto avvenire perchè non vi è nulla che impedisca il flusso di una corrente negativa sui singoli induttori quando la somma algebrica dei due risulti comunque positiva o nulla (diodo in conduzione). Questo problema si può riscontrare sia nel funzionamento in continua (sempre almeno uno fra transistor e diodo in conduzione) che in discontinua (con un intervallo di tempo in cui non conducono nessuno dei due). Per ovviare a questo problema le soluzioni proposte in questa tesi sono quelle di aggiungere un ulteriore diodo rettificatore in serie agli avvolgimenti e/o di gestire il rapporto di induttanze dei due avvolgimenti in modo che nella condizione nominale di funzionamento raggiungano contemporaneamente la condizione di inversione della corrente. Queste possibilità sono state esplorate con successo nell’utilizzo del convertitore ZETA per applicazioni di correzione del fattore di potenza PFC in cui si è proposto un insieme di equazioni di dimensionamento che portano al progetto del convertitore al fine di ottenere le forme d’onda desiderate.

Abstract:
Lo scopo della tesi era quello di analizzare vari parametri delle principali modulazioni PWM per inverter trifase in modo da distinguere quali fossero le più efficienti o le più affidabili. A tale scopo, è stata dapprima effettuata una panoramica dell’inverter trifase e delle relative modulazioni. Sono stati quindi scritti vari codici in Matlab per simulare le diverse tecniche di modulazione. Le modulazioni presentate in questo testo (con i rispettivi codici) sono: Sinusoidal PWM, THIPWM, SVPWM, DSVPWM (0,1,2,3,+,-), ASDSVPWM, Six-step (implementata solo per effettuare un confronto). Al termine della creazione dei vari codici, le modulazioni sono state studiate attraverso uno schema di inverter trifase implementato su Simulink. Il circuito, oltre a permettere la visualizzazione delle varie tensioni e correnti in uscita, ha consentito di studiare varie figure di merito utili per l’obbiettivo. Le figure di merito studiate sono state: THD, ripple di corrente, tensione di modo comune, numero di switching e rendimento di conversione. I risultati ottenuti hanno permesso di capire che le modulazioni meno efficienti sono la six-step (basso rendimento di conversione e THD troppo elevato) e la ASDSVPWM (distorsione troppo elevata nonostante i buoni parametri in potenza e ripple). Al contrario le modulazioni più efficienti sono la DSVPWM0 e la DSVPWM2 grazie alla loro bassa distorsione armonica e ai buoni parametri ottenuti nelle altre figure di merito. Tutte le altre modulazioni in termini di efficienza sono inferiori alle ultime due.

Dans une 1ere partie, nous avons teste l'efficacite d'un nouvel inhibiteur de croissance, le x, insecticide de la famille des benzoilphenylurees, contre 2 especes de termites, r. Santonensis et r. (l. ) grassei. Apres avoir determine quelle etait la formulation du produit la plus efficace et la concentration a utiliser, et apres avoir montre que le x se propageait rapidement a l'ensemble d'un groupe grace aux echanges trophallactiques entre les individus, nous avons utilise cet insecticide pour un traitement de terrain en technique d'appats. Les premiers resultats d'un traitement de ce type se sont averes tout a fait encourageants. Dans une 2eme partie, nous avons suivi l'evolution de la signature chimique des 2 especes de termites, r. Santonensis et r. (l. ) grassei maintenues artificiellement en colonie mixte experimentale. Ceci nous a permis de mieux comprendre les mecanismes impliques dans les phenomenes de regulation des produits cuticulaires, a l'origine de la cohesion d'une societe d'individus: transfert par contact des hydrocarbures allospecifiques, regulation selective des hydrocarbures, relargage des hydrocarbures allospecifiques a partir d'une structure inconnue, ou retour sur la cuticule de ces produits internalises mais non catabolises ou biosynthese de ces produits par les chaines enzymatiques utilisees pour les hydrocarbures homospecifiques, derive temporelle de la signature chimique

Cette thèse porte sur l'étude des propriétés asymptotiques d'un estimateur non paramétrique de la densité de type Parzen-Rosenblatt, sous un modèle de données censurées à droite, vérifiant une structure de dépendance de type associé. Dans ce cadre, nous rappelons d'abord les résultats existants, avec détails, dans les cas i.i.d. et fortement mélangeant (α-mélange). Sous des conditions de régularité classiques, il est établi que la vitesse de coonvergence uniforme presque sûre de l'estimateur étudié, est optimale. Dans la partie dédiée aux résultats de cette thèse, deux résultats principaux et originaux sont présentés : le premier résultat concerne la convergence uniforme presque sûre de l'estimateur étudié sous l'hypothèse d'association. L'outil principal ayant permis l'obtention de la vitesse optimale est l'adaptation du Théorème de Doukhan et Neumann (2007), dans l'étude du terme des fluctuations (partie aléatoire) de l'écart entre l'estimateur considéré et le paramètre étudié (densité). Comme application, la convergence presque sûre de l'estimateur non paramétrique du mode est établie. Les résultats obtenus ont fait l'objet d'un article accepté pour publication dans Communications in Statistics-Theory and Methods ; Le deuxième résultat établit la normalité asymptotique de l'estimateur étudié sous le même modèle et constitute ainsi une extension au cas censuré, du résultat obtenu par Roussas (2000). This thesis deals with the study of asymptotic properties of e kernel (Parzen-Rosenblatt) density estimate under associated and censored model. In this setting, we first recall with details the existing results, studied in both i.i.d. and strong mixing condition (α-mixing) cases. Under mild standard conditions, it is established that the strong uniform almost sure convergence rate, is optimal. In the part dedicated to the results of this thesis, two main and original stated results are presented : the first result concerns the strong uniform consistency rate of the studied estimator under association hypothesis. The main tool having permitted to achieve the optimal speed, is the adaptation of the Theorem due to Doukhan and Neumann (2007), in studying the term of fluctuations (random part) of the gap between the considered estimator and the studied parameter (density). As an application, the almost sure convergence of the kernel mode estimator is established. The stated results have been accepted for publication in Communications in Statistics-Theory & Methods ; The second result establishes the asymptotic normality of the estimator studied under the same model and then, constitute an extension to the censored case, the result stated by Roussas (2000). This result is submitted for publication

Spatial representation of modeling area in travel demand models has changed little over the course of last several decades. In this regard, the state-of-the-art still widely relies on the same centroid-connector system that has been used in classic models. In this approach continuum bidimensional space is lumped on centroids. It is an aggregate approach which ignores the physical variability linked to the scatteredness of disaggregate residence- and activity-places over the local space. Consequently the modeling performance in explaining route and mode choice behavior degrades at local scales: In route choice, disaggregate location influences the propensity between a distant interchange to a highway, or a nearby road. In mode choice, feeder service to public transportations influences the auto vs. transit modal share. We propose a disaggregate approach for spatial representation. Based on a zoning system, a stochastic disaggregate representation is used to characterize the space within a traffic analysis zone. For each zone, anchor-points are defined as the network nodes that are used for accessing to the network from within the local space. An itinerary between a pair of zones is then considered as a chain of legs composed of two terminal legs, corresponding to the intrazonal route sections, and one main leg between two anchor points. The route choice problem is transformed to a joint choice of a pair of anchor points. The vector of random terminal travel times is Multivariate Normal resulting in a Multinomial Probit model of choice of a pair of anchor points. To extend to the multimodal context, a transit composite mode is defined as a chain of access, main, and egress modal legs, and transit platforms are considered as anchor points connecting the feeder legs to the main line-haul leg. A Multinomial Logit mode choice model is estimated based on the 2001 Paris Household Travel Survey for the auto mode and the composite transit modes. It is joined with the two Multinomial Probit models corresponding to the choice of anchor points. The result is a joint model of mode and station choice with a disaggregate representation of the local space

Abstract:
Human papillomavirus (HPV) related cervical cancer is still the most common gynaecological malignancy in developing countries and, as yet, there is no alternative to surgery for the treatment of HPV-associated pre-malignant lesions. HPV 'hijacks' the host-cell ubiquitin-proteasome system to degrade the p53 and Rb tumour suppressor proteins which in turn, leads to the development of cancer. Previous studies have shown that the HIV protease inhibitor lopinavir selectively inhibits the chymotryptic-like activity of the 26S proteasome which stabilises p53 and induces the apoptosis of HPV positive cervical carcinoma cells. Based on this it was hypothesised that lopinavir treatment of HPV positive cervical carcinoma cells would produce changes in the levels of a wide range of cellular proteins that are dis-regulated by HPV-related activation of the proteasome. In order to address this, antibody microarray screening was carried out on lopinavir treated and control untreated HPV positive SiHa cervical carcinoma cells. This showed lopinavir induced alterations in 51 proteins including the cellular antiviral defence protein RNase L. Lopinavir induced both a dose and time dependent increase in RNase L which was subsequently confirmed by western blotting. Transient siRNA silencing of RNase L expression reduced the lopinavir-dependent toxicity in SiHa cells, suggesting an important role for this protein in the toxicity of lopinavir in HPV infected cells. SiHa cells were much more sensitive to lopinavir than CaSKi cervical carcinoma cells which had much higher levels of the E6 protein and did not up regulate RNase L. Furthermore, lopinavir treated HPV16 E6/E7 immortalised keratinocytes were also shown to up regulate RNase L protein expression and these cells were much more sensitive to lopinavir induced apoptosis than mortal control keratinocytes. In addition, transient expression of RNase L in RNase L-deficient C33A cells and the same cells stably transfected with HPV16 E6 (C33AE6) demonstrated that E6 protected these cells from RNaseL-induced cell death. Surprisingly, analysis of RNase L protein levels in these cells demonstrated that E6 did not induce the degradation of the RNase L protein. Instead it was found that E6 stabilised the interaction between RNase L and its endogenous inhibitor protein, ABCE1, and that lopinavir de-stabilised this interaction. Given that C33A tumour cells, E6/E7 immortalised keratinocytes and hTert immortalised keratinocytes are all sensitive to lopinavir, this implies that this compound does not specifically target HPV immortalised cells but rather targets immortalised cells in general, regardless of how this was achieved. The optimum concentration of lopinavir for all these effects was 25 μM, which is 15-fold higher than is observed in cervico-vaginal secretions following oral dosing with the drug Kaletra. In conclusion these results have confirmed the potential of lopinavir to treat HPV related pre-cancerous cervical lesions and provided at least part of the mode-of-action. Indeed they strongly support the use of lopinavir as a low-cost, self-applied topical alternative to surgery for this disease which will be of particular benefit in low-resource countries. Finally, the ability of lopinavir to induce apoptosis of non-HPV related immortalised cells merits further investigation since this indicates this drug may be useful for the treatment of other non HPV related pre-malignant conditions.
