Abstract:
Abstract Toxicity of the extracellular products (ECP) and the lethal attribute of serine protease secreted by five pathogenic Vibrio alginolyticus strains from various sources in kuruma prawn Penaeus japonicus were studied. The ECPs of organisms originally isolated from diseased kuruma prawn or small abalone Haliotis diversicolor supertexta were more lethal (LD50 value of 0.48 or 0.41 μg protein/g prawn) than those from diseased tiger prawn P.monodon, yellowfin porgy Acanthopagrus latus or horse mackerel (LD50 value of 0.98 -1.17 μg protein/g prawn). All the ECPs manifested strong, weak and no activities against gelatin, sheep erythrocytes and chitin, respectively. In immunodiffusion tests using rabbit antiserum to a purified 33 kDa serine protease of strain Swy against ECP of each tested strain produced one single precipitation band in each treatment. Furtherm ore, the serine protease was suggested to be the dominant protease secreted by V. alginolyticus strains tested since the majority of enzymatic activity of the respective ECP was inhibited by phenylmethanesulfonyl fluoride (PMSF). A higher inhibition of serine protease activity by PMSF resulted in lower mortality rate of the ECPs injected into the prawns suggesting that the protease is one of the major lethal factor(s) secreted by Valginolyticus.

Although apoferritin has been widely utilized as a new class of natural protein nanovehicles for encapsulation and delivery of nutraceuticals, its ability to remove metal heavy ions has yet to be explored. In this study, for the first time, we demonstrated that the ferritin from kuruma prawns (Marsupenaeus japonicus), named MjF, has a pronouncedly larger ability to resist denaturation induced by Cd2+ and Hg2+ as compared to its analogue, human H-chain ferritin (HuHF), despite the fact that these two proteins share a high similarity in protein structure. Treatment of HuHF with Cd2+ or Hg2+ at a metal ion/protein shell ratio of 100/1 resulted in marked protein aggregation, while the MjF solution was kept constantly clear upon treatment with Cd2+ and Hg2+ at different protein shell/metal ion ratios (50/1, 100/1, 250/1, 500/1, 1000/1, and 2500/1). Structural comparison analyses in conjunction with the newly solved crystal structure of the complex of MjF plus Cd2+ or Hg2+ revealed that cysteine (Cys) is a major residue responsible for such binding, and that the large difference in the ability to resist denaturation induced by these two heavy metal ions between MjF and HuHF is mainly derived from the different positions of Cys residues in these two proteins; namely, Cys residues in HuHF are located on the outer surface, while Cys residues from MjF are buried within the protein shell. All of these findings raise the high possibility that prawn ferritin, as a food-derived protein, could be developed into a novel bio-template to remove heavy metal ions from contaminated food systems.

Abstract Sequence analyses on the complete mitochondrial DNA (mtDNA) control region (992 bp) were conducted to elucidate the population structure of kuruma prawns (Penaeus japonicus) in East Asia. Five populations including 95 individuals were collected. They are separated into the Japan Sea (JS), the north and south of the East China Sea (NECS and SECS), the Taiwan Strait (TS), and the north of the South China Sea (NSCS) populations. There are 292 variable sites without any insertions and deletions. Nucleotide diversity in the total populations is 2.51 ± 0.07%, and the variations within populations ranged from 2.61 ± 0.93% (SECS) to 2.29 ± 0.16% (JS). FST values between the JS and the rest of the populations, between the NECS and NSCS populations, and between the SECS and NSCS populations show significant differences. The UPGMA tree of these five populations shows three distinct clusters; one includes the JS population; another includes the NECS population; the third includes populations from the rest of the areas. The analysis of molecular variance (AMOVA) shows clear genetic difference between the JS and the rest of the populations. Additional AMOVA analysis excluding the JS population indicates significant variation between the NECS population and the other three populations. We, therefore, conclude that three distinct populations exist in East Asia; one is in the JS; another is in the NECS; and the third is distributed in SECS, TS and NSCS.

Abstract:
Many brackish water aquaculture ventures in Australia and overseas have established ponds in coastal regions with acid sulfate soils (ASS). Acid sulphate soils are known to leach relatively high concentrations of metals, acid (metal and H+ ion) and sulfur, however very little is known about how these leached elements affect the water quality of aquaculture ponds. The main objective of this thesis was to describe the hydrochemical processes controlling the water chemistry in the water column and sediment pore water in the studied aquaculture ponds over time and space. Water samples providing the spatio-temporal data were collected from the ponds with the use of adapted sampling methods commonly used in the groundwater environment. A transect of five nested piesometers was installed in two prawn ponds at Pimpama, south east Queensland, Australia. Each piesometer nest contained a multilevel with eight outtakes, a mini ?? horizontal, and a slotted piesometer. Water samples were collected from each nested piesometer on a bi-monthly basis over the prawn-growing season. The unstable elements and water quality variables (pH, Eh, DO, EC, water temperature) were measured in the field. Stable elements were analysed in the laboratory using ICP-OES and ICP-MS. Soil samples were collected at the end of the season for elemental analysis. A number of key sediment/water interactions and processes such as precipitation/dissolution reactions, oxidation-reduction reactions, photosynthesis, adsorption and seawater buffering were identified as important controls on pond water conditions. This is the first study to provide detailed hydrochemcial analysis of the pond water over time and space and aided in identifying that even shallow water bodies can be chemically heterogeneous. Analysis of the water and sediment highlighted the selection of metals that can be associated with ASS and that are mobilised from pond sediments under certain chemical conditions. In Pond 7 Al, As, Ni and Zn concentrations were generally higher at the beginning of the grow-out season. Variability of the metal concentration was observed between the water column (0 ?? 1500 mm) and the pore-water (0 - -1000 mm). The highest concentration of Al (1044 ??g/L) and Zn (104 ??g/L) were sampled in the water column (approximately 400 mm from the surface of the pond). The highest concentration of As (130 ??g/L) and Ni (73 ??g/L) were sampled in the pore water sediment (associated with ASS). Elevated Mn and Fe2+ concentrations were also associated with the sediment pore water. The highest concentrations of Mn and Fe2+ were 4717 ??g/L and 5100 ??g/L respectively. In Pond 10, Ni concentrations (167 ??g/L) were the highest at the beginning of the grow-out season. However, As (97 ??g/L), Al (234 ??g/L) and Zn (308 ??g/L) were most concentrated during the middle of the cycle. The highest mean concentrations of these elements are As (63 ??g/L), Al (91 ??g/L) and Zn (69 ??g/L) which are each associated with the sediment-water interface. These metals are integral in degrading the pond water quality and lead to a loss of beneficial algal blooms, a reduction in pond water pH, poor growth rates and high mortality in shrimp. It is also possible that the dissolved ions and precipitated compounds that are leached from the ASS are discharged into the adjacent coastal estuary of Moreton Bay. With knowledge obtained from this PhD study, effective management and treatment systems can be developed and implemented to minimise the impact of these soils on the pond system and the water discharging into natural coastal ecosystem.

This thesis investigated the characteristics of protease and pathogenesis of Vibrio alginolyticus strain Swy in kuruma prawn. An extracellular lethal toxin produced by Vibrio alginolyticus strain Swy was purified using the AKTA purifier system with hydrophobic interaction chromatography, anion exchange and gel filtration columns. The toxin is an alkaline serine protease, inhibited by phenyl methylsulphonyl fluoride (PMSF), antipain and shows maximal activity at pH 8 to 11(HPA used), having a pI of 4.3 and a molecular weight of 33 kDa. The purified protease was lethal for kuruma prawn at an LD50 of 0.29 g protein/g body weight. The addition of PMSF completely inhibited the lethal toxicity of the purified protease, indicating that this serine protease was a lethal toxin produced by the bacterium. Four chromogenic substrates for characterizing serine protease of strain Swy were evaluated. The protease activity of bacterial extracellular products (ECP), or the 33 kDa purified protease, was completely inhibited by EDTA, EGTA using water-soluble substrates (azoalbumin and azocasein). Since chelating agents and 1,10-phenanthroline are commonly employed as inhibitors to identify metalloprotease, the two water-soluble substrates may not be appropriate for this purpose, except for using 1,10-phenanthroline as an inhibitor. Virulence of ECPs and the lethality attributes of serine proteases secreted by five pathogenic V. alginolyticus strains from various sources in kuruma prawn were studied. The ECPs of organisms originally isolated from diseased kuruma prawn or small abalone Haliotis diversicolor supertexta was more virulent (LD50 value of 0.48 or 0.41 g protein/g body weight) than those from diseased tiger prawn P. monodon, yellowfin porgy Acanthopagrus latus or horse mackerel (LD50 value of 0.98-1.17 g protein/g body weight). All the ECPs manifested strong, weak and no activities against gelatin, sheep erythrocytes and chitin, respectively. The higher inhibition of serine protease activity resulted in lower mortality rate of the ECP injected into the prawns suggesting that the protease is the lethal factor secreted by V. alginolyticus. The toxicity of the respective ECPs to kuruma prawns was only partially neutralized by rabbit antiserum to the formalinized Swy ECP and/or rabbit antiserum to the formalinized Swy serine protease. The haemolymph withdrawn from the moribund prawns injected with the ECP or toxic protease was unable to clot. The coagulogen was fast migrated in CIE following in vitro toxic protease and ECP treatment, but the amount of all components were reduced only in ECP treatment except coagulogen. After these treatment the plasma colour change from blue to pink was recorded. The amount of coagulogen was reduced in CIE following in vivo bacteria and ECP treatment, however that of toxic protease treatment was not remarkable.
