Abstract:
Urbanism conception is inspirated by fragmented structure and courtyards of Staré Brno. The house is dividend into five blocks, which corresponds with scale of surrounding built-up area. First floor (shops, restaurant, multicultural hall, entrances to the apartments and the office) and second floor (Office), they are separated from higher floors with apartments. First and second floors have large-formated glass. Living part (from third to seventh floor) has white facade which gives sunlight to areas between blocks. Bay windows is significant element on facade. It stands out above the surface 35 centimeters. It makes structure and shadows on facade. The multicultural hal lis placed in courtyard.

Abstract:
One of the main objectives is to organize the space into inner courtyard, to define a relationship between the house and surrouding areas, like street and inner courtyard. Current courtyards of the bylcony houses represent a kind of second layer of the city – private zone. Space, which is owned by someone, which has its own scheme and charm in city lifestyle context. Unclear space runs deep into the inner courtyard is defined by three new residential wings and becomes the eleventh courtyard which, will offer comfort gardens and recreational areas for residents. Another important factor is also a fact that we design not only the house but also the whole one side of the street, and there is no corner of the street by now. The new design of both hips when we eject the whole mass before today's bare boards into the street is fair to the satisfaction of cars that disappear from the street - leaving only the tram, a narrow profile so never mind. The opposite principle of a closed block of flats is linear, to the square oriented vertical mass of the townhouse. In place of intersection of two objects there is a quiet green bay, which is the added value of the house in the public space. Vertical brings the last missing facade and at the same time created a full quality corner of the southern mouth of the Mendel Square.

Abstract:
Local DNA structures are alternative DNA conformations which can be formed aside from typical B-DNA conformation. These structures often play pivotal roles in regulation of basic biological processes, such as DNA replication, transcription or binding of specific ligands. This biological significance makes alternative DNA secondary structures a potential drug target. In this diploma thesis, local structures in genomes of viruses from Flaviviridae and Retroviridae families are analysed using bioinformatics tools. Furthermore, these structures are visualised using atomic force microscopy.

Abstract:
Inverted repeats (IR) are common part of DNA of all living prokaryotic and eukaryotic organisms. Inverted repeats plays an important role in the regulation of basics cells processes. They are responsible for formation of cruciform structures. Inverted repeats also cause genomic instability and can be a source of numerous mutations. Cruciform structures can be recognized by DNA-binding proteins and can also act as a transcriptional regulators. Using the Palindrome Analyser tool, the frequency of IR and localization of inverted repeats in bacterial genomes was analyzed. The frequency of IR across the bacterial genome is variable. The frequency of short inverted repeats shows an approximately quadratic dependence on the %GC content in the genome with a minimum of about 50% of GC content. The localization of inverted repeats with respect to “annotated features” show a non-random distribution. The frequency of IR for most features is higher “outside” than “inside”.

Abstract:
This thesis was focused on DNA analysis. The fluorescently labelled oligonucleotides were at first hybridised and subsequently analysed by HRM analysis to determine the melting temperatures of the oligonucleotides depending on the environment. This thesis describes the change of melting temperature of oligonucleotides in environments containing mono and bivalent ions and the influence of protein binding on the stability of these DNA structures. From determined melting points, it was specified whether the ion/protein stabilised or destabilised the oligonucleotide. Furthermore, plasmids were isolated and analyzed by atomic force microscopy.
