Abstract:
Improving large-seeded waxy corn for food and industrial purposes is one way to increase the value and utilization of new corn cultivars in Thailand. This study aimed to evaluate the agronomic traits, yield and yield components, and response to downy mildew (DM) disease caused by Peronosclerospora sorghi of the developed breeding lines of large-seeded waxy corn. A total of 33 waxy corn breeding lines with different seed sizes, row numbers, and kernel colors were selected from 2015 to 2018. The yield experiment was conducted at the Khon Kaen University, Khon Kaen, Thailand in the 2019/20 dry season and the 2020 rainy season. The breeding lines were also screened for DM resistance through artificial inoculation with spore suspension at farmers’ fields in two locations, namely, Sok Muang and Nong Bua, in the 2020 rainy season. Genotype had a significant effect on yields, yield components, and agronomic traits. However, the interactions between genotype and season were not significant for 1000-kernel dry weight and the number of rows per ear. ‘DRP-4’, RLW-4’, ‘RLW-6’, ‘RLW-7’, and ‘RLW-10’ showed the highest 1000-kernel dry weight and the optimal number of rows. Corn grown at Sok Muang Village had higher disease severity than that grown at Nong Bua Village due to the high rainfall, low temperature, and high humidity at the area. The resistant check cultivar ‘Nei9008’ exhibited moderate resistance to DM at both locations. Although the inbred lines were susceptible to DM disease, they appeared to be a good source of large seeds. Therefore, the backcross breeding method is still needed to obtain DM-resistant germplasm for the further development of waxy corn cultivars.

Abstract:
Ethidium bromide (EB), at 10-5 to 10-4 M, progressively inhibits NAA-induced rooting of mung bean cuttings. Cycloheximide (CH), 6-methylpurine (6-MP) and kinetin (KIN) also inhibited rooting at the same concentrations, although CH and 6-MP were more effective. At 70 and up to 130 hours of incubation, after cuttings received a 1-ml pulse of NAA (10-4 M), they exhibited a progressive increase in the number of observed adventitious roots. The addition of one of the inhibitors, 6-MP, EB or KIN to cuttings, pulsed 48 hours earlier with NAA, showed an initial slight inhibition with increased inhibition over time. CH, however, inhibited rooting immediately after addition. From these and other similar kinetic studies, it appears that 6-MP, EB and KIN operate at the transcriptional level and that CH inhibits translation. Lineweaver-Burk plot analysis of NAA-induced rooting inhibition showed that EB may act as a competitive inhibitor of NAA. Since EB is a known intercalating agent and competitively inhibits NAA-induced rooting, NAA may influence gene expression by ultimately binding to DNA. Studies with space-filling and computer-generated models show that both NAA and EB can bind to certain dinucleotides by an intercalation mechanism.

Abstract:
Khon Kaen District is in the central, north-east part of Thailand and is being developed to handle the country’s growth. Khon Kaen District is undertaking the project of building a light rail as a facility for the people. Consequently, one of the problems is ensuring adequate parking for people using the light rail service. In general, the symmetry concept naturally used in decision making to finding an optimal solution for decision and optimization problems. In this paper, multi-criteria decision analysis (MCDA) and multi-objective decision making (MODM) were used to solve the parking site selection problem, which made the decision easier. This paper proposed an analytic hierarchy process (AHP) technique, combined with the geographical information system (GIS), to evaluate the weight of the criteria used in the analysis and find potential parking solutions. Furthermore, this paper proposed the application of a linguistic technique with fuzzy TOPSIS methods to analyze the appropriateness of parking site selections from potential candidates to support use of the light rail. The results of the MCDA show that the most suitable parking lot location is along the light rail and closest to the business area. The results of the fuzzy TOPSIS method, both positive and negative ideal decisions, can help inform decision makers in selecting which candidate site is optimal for parking.

Abstract:
Aim: This study aims to determine the prevalence of Cryptosporidium spp. infection and to identify the species of Cryptosporidium spp. in newborn dairy calves between December 2016 and March 2017 in Muang District, Khon Kaen Province, Thailand. Materials and Methods: A total of 200 fecal samples from newborn dairy calves of the ages 1 day up to 28 days were collected and the presence of Cryptosporidium oocysts was examined microscopically using the modified Kinyoun's acid-fast staining technique. Then, Cryptosporidium species were identified using nested polymerase chain reaction amplification of 18S rRNA gene and sequencing. Results: The modified Kinyoun's acid-fast staining revealed the presence of Cryptosporidium oocysts in 51% (102/200). Sequence analysis of the 18S rRNA gene identified two species, namely, Cryptosporidium bovis (n=11) and Cryptosporidium ryanae (n=11) and one isolated strain could not be identified. Conclusion: This study indicated that newborn dairy calves aging up to 4 weeks were highly infected with Cryptosporidium spp., and the infection mostly occurred in diarrheic dairy calves. This is the first report of Cryptosporidium in dairy calves in Khon Kaen Province and the results provide baseline information for further studies and control of Cryptosporidium infection in dairy calves in the study area.

Abstract:
Intestinal parasitic infections are still a crucial problem among communities in Northeast Thailand. Misuse of antiparasitic drugs and unhealthy food behaviors are known. This study aimed to explore the prevalence, behavioral health factors, and motivation for self-treatment of anti-parasitic drugs in this area. A community-based cross-sectional study was conducted in Phra Lap sub-district, Mu Khon Kaen district, Khon Kaen province, Northeast Thailand, in 2016. A total of 419 participants were recruited to complete a self-administered questionnaire and stool examination. Binary logistic regression was used to assess the association between the risk factor and parasitic infection. Forty-two participants (10%; 95%CI 7.5–13.3) were positive for at least one parasite species. In this community, the most detected intestinal parasite was Opisthorchis viverrini (5.3%), followed by Strongyloides stercoralis (3.1%). A total of 67.5% of the participants had the experience of anti-parasitic drug treatment within previous 1 year, and “Often eat raw food” was the most common reason for the use of anti-parasitic drugs. On multivariate analysis, parasitic infections were significantly associated with male gender (ORadj. 2.42; 95%CI 1.00–5.85), age ≥ 60 years (ORadj. 7.55; 95%CI 1.60–35.76), and often consuming raw food of at least one type (ORadj. 2.37; 95%CI 1.03–5.44). Given these findings, correction of the dietary habit of eating raw fish/meat, which is the most important measure, and limitation of the use of anthelmintic treatment for individuals with stools positive for ova as well as emphasis on sanitary toilets will be implemented for the prevention and control of parasitic infection in endemic communities.

Abstract:
Coronavirus disease (COVID-19) is widely spread across the world. The COVID-19 lockdown policy has forced dine-in restaurants and cafes to close. Consequently, there is a plunge in the global economy into severe recession due to less economic activities. On the other hand, the demand for online food delivery services has increased during this period. As a result, the changes in consumer behaviours during the COVID-19 crisis are taken into consideration. Are the COVID-19 crisis and the changes in consumer behaviours related? Due to the new model of consumption, our study aims to observe and discern the relationship between COVID-19 crisis and changes in consumer behaviours. Data were collected by a questionnaire asking about participants’ general information, the impacts of COVID-19 on other relevant aspects, and the changes in consumer behaviours during the COVID-19 pandemic. Survey samples, including the population in Mueang Khon Kaen District, Thailand, were randomly selected through a random sampling method with a total of 634 respondents (241 males, 380 females, and 13 others). Our results revealed that the respondents have been highly impacted by the COVID-19 pandemic and their consumer behaviours have been changed significantly compared to the pre-pandemic period. Additionally, statistical analyses showed that there is a significant correlation between the COVID-19 pandemic and the changes in consumer behaviours. This study could serve as a guideline of further studies about the COVID-19 pandemic and consumer behaviours, which can be appropriately adopted in the era for the new normal.

Abstract:
The present study was conducted to find out the impact of inorganic nutrients (Nitrogen @ 20 kgha-1 and Phosphorus 40 kgha-1) blended with and without biofertilizers (Rhizobium and PSB) on growth and yield of mung bean. Significant increments in seed yield of 31.07% followed by 23.34% were recorded from the plots exposed to 20 kgN/ha + 40 kgP2O5/ha along with dual inoculation by Rizobium and PSB (T9) and 20 kgN/ha + 40 kgP2O5/ha + Rizobium (T7). Highest seed yield (13.23 q/ha) realized under T9 supported by increased final aboveground dry matter (29.33% & 26.21%), pod percentage (26.33% & 17.45%) and test weight (17.50% & 11.13%) compared to chemical fertilizers (T4 & T5 respectively). Highest number of nodules (68.17) & nitrogen use efficiency (18.32 kg/kg) were recorded at flowering (50 DAS) stage of the crop under treatment-9. Functional relation between nodulation, NUE, AAR, pods plant-1 and yield were positive and linear (R2=0.987, 0.9987, 0.9501, 0.9115, respectively). To ascertain the higher productivity in mung bean it is recommended that the application of phosphorus @ 40 kgha-1 with starter dosage of N @ 20 kgha-1 along with dual inoculation by Rizobium and PSB should be applied for higher seed yield under temperate agro-ecological conditions.

Abstract:
Background and Aim: Toxoplasmosis is a zoonosis caused by Toxoplasma gondii. Cats are known to be the definitive hosts that can excrete these environmentally resistant oocysts. Other mammals, avians, and even humans can serve as the intermediate host. T. gondii infection is often asymptomatic in healthy individuals; however, it could result in serious health problems in immunocompromised and pregnant individuals. This study investigated the occurrence of T. gondii infection in cats in Khao Suan Kwang and Mueang Khon Kaen. Materials and Methods: In total, 100 serum samples from cats, that is, 62 owned cats (31 males and 31 females) and 38 adopted stray cats (21 males and 17 females), were examined for antibodies against T. gondii through rapid immunochromatographic tests (ICT). Owners were asked to sign a consent form and answer the questionnaires before sample collection. Demographic information about the cats and their owners was also recorded. Results: The overall seroprevalence of cats positive for T. gondii antibodies was found to be 5%. Notably, the Toxoplasma antibody prevalence was significantly higher in the adopted stray cats (10.53% [4/38]) that roamed the zoo than in the owned cats (1.61% [1/62]) (p > 0.05). No significant difference was observed between male (8.33%) and female (1.92%) cats. The cat owners' questionnaire revealed that more than half had never heard of toxoplasmosis before (67.7%), whereas 30.6% knew nothing about the disease transmission routes. Conclusion: This study presented a low seroprevalence of antibodies to T. gondii in owned cats from the Mueang Khon Kaen District, whereas high seroprevalence was detected in the adopted stray cats from Khao Suan Kwang. Adopted stray cats can have a higher potential for T. gondii infection; thus, they could be a source of toxoplasmosis transmission to humans. Therefore, it is essential to control the number of stray cats, and a screening test for antitoxoplasmosis could be recommended before adoption. Although the total seroprevalence was noted to be low, the zoonotic disease was present. Therefore, raising the community's awareness and knowledge might reduce the disease transmission from animals to humans.

Abstract:
In this study, we aimed to explore bioactive volatile compounds in leaves of black rice which emphasized on phenolic compounds. Leaves of four black rice varieties including IR1552 and three Thai black glutinous rice varieties, Kam Huai Hong Khrai (KHHK), Kam Chiang Mai (KCM), and Kam Muang Nan (KMN) were extracted by acidic and basic aqueous solution. Gas chromatography–mass spectrometry analysis of the volatile components in these black rice extracts revealed twelve simple phenols among fifty-five volatile compounds identified. 2-Methoxy-4-vinylphenol was detected as the most abundant volatile, especially, in the booting stage for KHHK followed by the tillering stage of the same variety. Its highest content was approximately 10-folds greater than that of vanillin at the booting stage. To the best of our knowledge, seven simple phenols were detected in black rice leaves for the first time, which included 4-vinylphenol, 2,6-dimethoxyphenol, 4-hydroxybenzaldehyde, 2-methoxy-4-(1-propenyl)phenol, 2,4-bis(1,1-dimethylethyl)phenol, 4-hydroxy-3,5- dimethoxy-benzaldehyde and 4-((1E)-3-hydroxy-1-propenyl)-2-methoxyphenol. 3-Hexen-1-ol was detected at the highest amount and was only identified in the dough-grain stage for KHHK, KCM and KMN. Two major aldehydes (2E)-2-hexenal and nonanal were detected in high quantities in KCM black rice variety in the dough-grain and mature-grain stage. Among the twelve fatty acids identified, (Z,Z,Z)-9,12,15-octadecatrienoic acid was detected at the highest amount, on average, followed by hexadecadienoic acid and octadecanoic acid. Therefore, it could be suggested that the leaves of black rice are rich in phenolic compounds and could be used to promote natural sources of food ingredients that are of great interest to the food industry.
