Abstract:
Purpose – This paper aims to explore knowledge management (KM) literature to understand further the inter-relationship among KM infrastructure (KMI), KM strategy (KMS), KM process (KMP) and individual competence (IC), which are considered very much essential for achieving a heightened competence at an individual level. In the present scenario, knowledge intensive organizations are striving highly to manage competitiveness for their survival in the market. The role of knowledge capability is very much pivotal in ensuring the success of knowledge process implementation. Design/methodology/approach – A methodical review has been carried out of empirical and conceptual articles to justify the linkages among KMI, KMS, KMP and IC, particularly accessed through sources such as Pro Quest, EBSCO host, Google Scholar and other e-databases. This review integrates a conceptual framework to explore the relationship among the aforesaid constructs. Findings – This study provides valuable insight into the role of KMI and KMS in enabling various KM processes. It also contributes significantly to organizations by helping them to integrate KM processes so as to enhance IC. Research limitations/implications – This paper provides helpful suggestions to drive the KMPs by facilitating suitable congruence of infrastructure capabilities and KMSs within organizations. Further, this study would also be helpful in enhancing IC along with organizational competence. Study limitations are also discussed. Originality/value – This paper contributes to knowledge and competence management literature by adding the fact that KMI and KMS both precede KMP for better outcomes at an individual level.

Abstract:
Reikšminiai žodžiai: hemodializė, lėtinė inkstų liga (LIL), akių pakitimai, tinklainės kraujagyslių skersmuo, optinė koherentinė tomografija (OKT). Darbo tikslas. Įvertinti hemodializuojamų pacientų, sergančių V stadijos lėtine inkstų liga (LIL), inkstų funkcijos rodiklius ir nustatyti, kokius akių pakitimus jie atspindi. Darbo metodika. 2014/11–2015/03 VUL SK atliktas pilotinis tyrimas, kuriame dalyvavo 11 pacientų, sergančių LIL V stadija. Neįtraukti pacientai, sergantys cukriniu diabetu. Nefrologijos centre vertinti epidemiologiniai, laboratoriniai, klinikiniai duomenys. Akių ligų centre – regėjimo aštrumas (RA), akispūdis, akių dugno kraujagyslės. Optinės koherentinės tomografijos (OKT) aparatu įvertintas tinklainės centrinės dalies storis. Glomerulų filtracijos greitis (GFG) apskaičiuotas CKD-EPI skaičiuokle. Statistinė duomenų analizė atlikta SPSS 22.0 programa. Duomenys laikyti statistiškai patikimi, kai p < 0,05. Rezultatai. Ištirta 11 ligonių: 4 vyrai (36,4 proc.), 7 moterys (63,6 proc.). Amžiaus vidurkis 62,5 ± 10,7 metų. LIL V stadijos vidutinė ligos trukmė 10,5 ± 8,2 metų. KMI vidurkis 24,3 ± 2,7. Lygindami tyrimų rezultatus tarp lyties nustatėme, kad vyrų arterijos ir venos santykis yra didesnis, palyginti su moterimis. Rasta statistiškai patikima koreliacija tarp KMI ir tinklainės centrinės dalies storio. Esant nekontroliuojamam AKS, centrinės tinklainės išorinė apatinė dalis yra plonesnė nei esant kontroliuojam AKS. Esant nekontroliuojamam AKS, gauta statistiškai patikima koreliacija tarp KMI ir centrinės tinklainės plonėjimo. Hipertenzine kardiomiopatija (KMP) sirgo 9 (81,8 proc.) pacientai. Jų akių dugno arterijos ir venos santykis yra didesnis, palyginti su tais, kurie KMP neserga. Nustatėme, jog, didėjant amžiui, labiau pažeidžiama premakulinė temporalinė, išorinė temporalinė, premakulinė apatinė ir išorinė apatinė sritys. Didėjant kreatinino koncentracijai, premaculinė temporalinė sritis plonėja labiau. Neradome statistiškai patikimos koreliacijos lygindami amžių, hemodializės ir arterinės hipertenzijos trukmę, GFG su akispūdžiu, OCT tyrimo rezultatais, akių dugno kraujagyslių diametru (p > 0,05). Išvados. Didesnis KMI – rizikos veiksnys tinklainės centrinės dalies plonėjimui. Nors negavome, jog arterinės hipertenzijos trukmė sąlygotų greitesnį tinklainės centrinės dalies plonėjimą, vis dėlto tų pacientų, kurių AKS yra kontroliuojamas, tinklainės centrinė dalis yra storesnė. Arterinės hipertenzijos sukelta KMP sąlygoja akių dugno kraujagyslių diametro pokyčius. Didėjant amžiui ir kreatinino koncentracijai, premakulinė temporalinė tinklainės sritis plonėja labiau. Taip pat lytis galimai įtakoja akių dugno arterijų ir venų kraujagyslių diametrų pokyčius. Tačiau reikia atsižvelgti, jog maža tiriamųjų imtis gali turėti įtakos statistiniams apskaičiavimams ir patikimumo nustatymui.

Abstract:
Osteoarthritis is a degenerative arthropathy that is mainly characterized by dysregulation of inflammatory responses. KMUP-1, a derived chemical synthetic of xanthine, has been shown to have anti-inflammatory and antioxidant properties. Here, we aimed to investigate the in vitro anti-inflammatory and in vivo anti-osteoarthritis effects of KMUP-1. Protein and gene expressions of inflammation markers were determined by ELISA, Western blotting and microarray, respectively. RAW264.7 mouse macrophages were cultured and pretreated with KMUP-1 (1, 5, 10 μM). The productions of TNF-α, IL-6, MMP-2 and MMP- 9 were reduced by KMUP-1 pretreatment in LPS-induced inflammation of RAW264.7 cells. The expressions of iNOS, TNF-α, COX-2, MMP-2 and MMP-9 were also inhibited by KMUP-1 pretreatment. The gene expression levels of TNF and COX families were also downregulated. In addition, KMUP-1 suppressed the activations of ERK, JNK and p38 as well as phosphorylation of IκBα/NF-κB signaling pathways. Furthermore, SIRT1 inhibitor attenuated the inhibitory effect of KMUP-1 in LPS-induced NF-κB activation. In vivo study showed that KMUP-1 reduced mechanical hyperalgesia in monoiodoacetic acid (MIA)-induced rats OA. Additionally, KMUP-1 pretreatment reduced the serum levels of TNF-α and IL-6 in MIA-injected rats. Moreover, macroscopic and histological observation showed that KMUP-1 reduced articular cartilage erosion in rats. Our results demonstrated that KMUP-1 inhibited the inflammatory responses and restored SIRT1 in vitro, alleviated joint-related pain and cartilage destruction in vivo. Taken together, KMUP-1 has the potential to improve MIA-induced articular cartilage degradation by inhibiting the levels and expression of inflammatory mediators suggesting that KMUP-1 might be a potential therapeutic agent for OA.

Abstract:
This study explored whether KMUP-1 improved chronic constriction injury (CCI)-induced BKCa current inhibition in dorsal root ganglion (DRG) neurons. Rats were randomly assigned to four groups: sham, sham + KMUP-1, CCI, and CCI + KMUP-1 (5 mg/kg/day, i.p.). DRG neuronal cells (L4–L6) were isolated on day 7 after CCI surgery. Perforated patch-clamp and inside-out recordings were used to monitor BKCa currents and channel activities, respectively, in the DRG neurons. Additionally, DRG neurons were immunostained with anti-NeuN, anti-NF200 and anti-BKCa. Real-time PCR was used to measure BKCa mRNA levels. In perforated patch-clamp recordings, CCI-mediated nerve injury inhibited BKCa currents in DRG neurons compared with the sham group, whereas KMUP-1 prevented this effect. CCI also decreased BKCa channel activity, which was recovered by KMUP-1 administration. Immunofluorescent staining further demonstrated that CCI reduced BKCa-channel proteins, and KMUP-1 reversed this. KMUP-1 also changed CCI-reduced BKCa mRNA levels. KMUP-1 prevented CCI-induced neuropathic pain and BKCa current inhibition in a peripheral nerve injury model, suggesting that KMUP-1 could be a potential agent for controlling neuropathic pain.

Methanol is one of the important intermediate chemicals which is used widely in many processes to produce final products such as formaldehyde, dimethyl ether (DME), dimethyl terephthalate (DMT), and methyl tertiary butyl ether (MTBE). As a pioneer of methanol producer in Indonesia, PT. Kaltim Methanol Industri (KMI) has a strong commitment for sustainability and saving natural resources including enhancement of energy efficiency during production activities. Since mid-2018, to fulfill the electricity needs, PT. KMI has two main sources of electricity namely internal source using steam turbine generator and tie-in from PT. Kaltim Daya Mandiri (KDM). The modification of electricity sources promotes PT. KMI to evaluate the efficiency of internal electricity production. This has been conducted by performing optimization of blow down rate or cycle steam in boiler operation. Based on the data logbook, this work aims to evaluate the effect of blowdown rate on the energy saving and natural gas consumption. When the number of cycle steam boilers is altered from around 10 to 24, the company could get the potential energy saving around 300 MMBTU/day or 7.14% of total based energy consumption. In the boiler operation, decreasing blowdown rate would raise the cycle steam boiler and give final consequences to reduce energy losses from the release of BFW. The optimization of this cycle steam also cut the boiler specific energy consumption into around 3.0 MMBTU/ton steam. Based on the average heating value, this innovation could decline the natural gas consumption of PT. KMI around 122.91 MMSCF in the period of July 2018 – July 2019. Based on the result above, the modification of the cycle steam boiler would enhance energy efficiency, saving the natural resources and promote the application of sustainable development concept in the chemical industry.Keywords: blowdown rate; boiler energy; cycle steam; energy efficiency; PT. KMIA B S T R A KMetanol merupakan bahan kimia antara yang dipergunakan secara luas sebagai bahan baku untuk menghasilkan produk akhir berupa formaldehyde, dimetil eter (DME), asam dimetil tereftalat (DMT), dan methyl tertiary butyl ether (MTBE). Sebagai satu-satunya produsen metanol di Indonesia, PT. Kaltim Methanol Industri (KMI) senantiasa berkomitmen untuk selalu melakukan penghematan sumber daya alam, salah satunya meningkatkan efisiensi energi. Untuk memenuhi kebutuhan listriknya, perusahaan ini memiliki dua sumber yaitu internal perusahaan menggunakan steam turbine generator dan sambungan (tie-in) dari PT. Kaltim Daya Mandiri (KDM) sejak pertengahan 2018. Adanya perubahan konfigurasi tersebut mendorong PT. KMI melakukan inovasi berupa optimasi laju blowdown atau cycle steam pada boiler untuk meningkatkan efisiensi energi. Berdasarkan data logbook operasi pabrik akan dievaluasi besarnya penghematan energi yang didapatkan dari optimasi tersebut. Peningkatan efisiensi energi dapat diperoleh dengan mengurangi laju blowdown atau peningkatan cycle steam boiler. Perubahan cycle steam pada boiler dari kisaran 10 menjadi 24 akan memberikan penghematan energi sebesar 300 MMBTU/hari atau sekitar 7,14% dari kebutuhan energi mula-mula. Penurunan laju blowdown akan mengakibatkan energi buangan melalui BFW yang dikeluarkan dari boiler berkurang, sehingga dapat menghemat energi yang dibutuhkan oleh boiler. Optimasi cycle steam boiler ini juga membuat konsumsi energi yang dinyatakan dalam energy specific boiler berhasil diturunkan pada kisaran 3,0 MMBTU/ton steam. Berdasarkan tabulasi data, inovasi ini dapat menghemat sumber daya gas alam sebesar 122,91 MMSCF pada kurun waktu Juli 2018 hingga Juli 2019. Dengan demikian, perubahan cycle steam melalui penurunan laju blowdown pada boiler terkonfirmasi dapat meningkatkan efisiensi energi dan menghemat pemakaian sumber daya alam, yang pada akhirnya mendorong aplikasi nyata sustainable development concept di dunia industri. Abstract:
Xanthine-based KMUP-1 was shown to inhibit phosphodiesterases (PDEs) and modulate G-protein coupled receptors (GPCRs) to lower hyperlipidemia and body weight. This study further investigated whether KMUP-1 affects adipogenesis and lipolysis in 3T3-L1 preadipocytes. KMUP-1 (1–40 µM) concentration-dependently attenuated Oil Red O (ORO) staining and decreased triglyceride (TG) accumulation, indicating adipogenesis inhibition in 3T3-L1 cells. In contrast, the β-agonist ractopamine increased ORO staining and TG accumulation and adipogenesis. KMUP-1 (1–40 µM) also reduced MAPKs/Akt/PPARγ expression, PPARγ1/PPARγ2 mRNA, and p-ERK immunoreactivity at the adipogenesis stage, but enhanced hormone sensitive lipase (HSL) immunoreactivity at the lipolysis stage. Addition of protein kinase A (PKA) or protein kinase G (PKG) antagonist (KT5720 or KT5728) to adipocytes did not affect HSL immunoreactivity. However, KMUP-1 did increase HSL immunoreactivity and the effect was reduced by PKA or PKG antagonist. Simvastatin, theophylline, caffeine, and sildenafil, like KMUP-1, also enhanced HSL immunoreactivity. Phosphorylated HSL (p-HSL) was enhanced by KMUP-1, indicating increased lipolysis in mature 3T3-L1 adipocytes. Decreases of MAPKs/Akt/PPARγ during adipogenesis contributed to inhibition of adipocyte differentiation, and increases of PKA/PKG at lipolysis contributed to HSL activation and TG hydrolysis. Taken together, the data suggest that KMUP-1 can inhibit hyperadiposity in 3T3-L1 adipocytes.

Abstract:
To test whether KMUP-1 (7-[2-[4-(2-chlorophenyl) piperazinyl]ethyl]-1,3-dimethylxanthine) prevents myocardial ischemia-induced apoptosis, we examined KMUP-1-treated H9c2 cells culture. Recent attention has focused on the activation of nitric oxide (NO)-guanosine 3', 5'cyclic monophosphate (cGMP)-protein kinase G (PKG) signaling pathway triggered by mitogen-activated protein kinase (MAPK) family, including extracellular-signal regulated kinase 1/2 (ERK1/2), c-Jun N-terminal kinase (JNK), and p38 in the mechanism of cardiac protection during ischemia-induced cell-death. We propose that KMUP-1 inhibits ischemia-induced apoptosis in H9c2 cells culture through these pathways. Cell viability was assessed using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay and apoptotic evaluation was conducted using DNA ladder assay and Hoechst 33342 staining. The level of intracellular calcium was detected using - Fura2-acetoxymethyl (Fura2-AM) staining, and mitochondrial calcium with Rhod 2-acetoxymethyl (Rhod 2-AM) staining under fluorescence microscopic observation. The expression of endothelium NO synthase (eNOS), inducible NO synthase (iNOS), soluble guanylate cyclase α1 (sGCα1), PKG, Bcl-2/Bax ratio, ERK1/2, p38, and JNK proteins were measured by Western blotting assay. KMUP-1 pretreatment improved cell viability and inhibited ischemia-induced apoptosis of H9c2 cells. Calcium overload both in the intracellular and mitochondrial sites was attenuated by KMUP-1 pretreatment. Moreover, KMUP-1 reduced intracellular reactive oxygen species (ROS), increased plasma NOx (nitrite and nitrate) level, and the expression of eNOS. Otherwise, the iNOS expression was downregulated. KMUP-1 pretreatment upregulated the expression of sGCα1 and PKG protein. The ratio of Bcl-2/Bax expression was increased by the elevated level of Bcl2 and decreased level of Bax. In comparison with the ischemia group, KMUP-1 pretreatment groups reduced the expression of phosphorylated extracellular signal-regulated kinases ERK1/2, p-p38, and p-JNK as well. Therefore, KMUP-1 inhibits myocardial ischemia-induced apoptosis by restoration of cellular calcium influx through the mechanism of NO-cGMP-MAPK pathways.

Abstract:
B lymphocyte precursor cells in mouse bone marrow develop in close association with stromal cells which provide essential growth signals. To identify molecules that may normally play a role in this interaction we have examined the in vivo binding of a new monoclonal antibody (mAb) (KMI6) that recognizes a determinant on a bone marrow stromal cell line (BMS2) in vitro. Flow cytometric and radioautographic evaluations revealed that the antigen recognized by KMI6 is represented on the surface of an extremely small number of cells in bone marrow cell suspensions from adult mice. An apparent molecular mass of 110 kD was obtained by surface labeling of a stromal cell clone and immunoprecipitation. Purified mAb KMI6 labeled with 125I was then given intravenously to young C3H/HeJ mice. Unbound mAb was washed out by cardiac perfusion and femoral bone marrow was examined by light and electron microscope radioautography. KMI6 labeling was heavy on the plasma membrane of many stromal cells, especially those located towards the outer subosteal region. The KMI6-labeled stromal cells were usually associated with cells of lymphoid morphology which they often completely surrounded. The labeling was restricted to areas of stromal cell plasma membranes in contact with lymphoid cells. The lymphoid cells themselves, as well as macrophages and other hemopoietic cells, failed to bind mAb KMI6 significantly. Stromal cells in bone marrow depleted of hemopoietic cells by gamma-irradiation (9,5 Gy) bound mAb KMI6 at reduced intensity. The results demonstrate that the KMI6 determinant, a 110-kD protein, is expressed on bone marrow stromal cells in vivo. Its restriction to areas of interaction with lymphoid cells suggests a role in forming microenvironmental niches of B lymphopoiesis. The surface membrane of individual stromal cells may thus be functionally polarized towards interacting B cell precursors and other hemopoietic cells.

Abstract:
The purpose of this work is to create a functional Key Management Server (KMS) with basic functionality for the vSphere 7.0 platform. It should communicate with vCenter and together provide the functionality to encrypt individual virtual machines. Commercial solutions in this area are very expensive and therefore the question arose whether the entire server can be implemented using freely available tools. Since vCenter uses the publicly available KMIP protocol to communicate with KMS, it turns out to be possible. The resulting implementation is based on the Ubuntu 20.04 operating system. The PyKMIP library for python 3.9 was used for the application logic and ETCD as storage. To connect the application and storage, a custom module was created. Bash scripts were created for whole installation and all of the necessary configuration. The overall result is fully functional and no flaws were found during testing. This work was done in cooperation with Master Internet, s.r.o.

Gyventojų mitybos tyrimai turi būti atliekami, siekiant gerinti visuomenės mitybą ir sveikatą bei įgyvendinti nacionalinių, Europos Sąjungos ir Pasaulio sveikatos organizacijos dokumentų nuostatas. Neabejotina, kad netinkama mityba yra daugelio lėtinių neinfekcinių ligų priežastis. Tyrimo tikslas – įvertinti 18-65 metų amžiaus Lietuvos gyventojų mitybos įpročius ir jų pokyčių tendencijas. Tyrimo uždaviniai: 1. Ištirti ir įvertinti Lietuvos gyventojų mitybos įpročius. 2. Nustatyti Lietuvos gyventojų mitybos įpročių pokyčių tendencijas. 3. Įvertinti gyventojų kūno masės indeksą ir KMI pokyčių tendencijas. Tyrimo metodai: Tyrimui buvo atrinkta 3500 Lietuvos suaugusių gyventojų. Tyrimo atsako dažnis 69,1 proc. Tyrimo metu kiekvieno respondento buvo klausta apie mitybos bei gyvensenos ypatumus, požiūrį į tam tikrus mitybos aspektus ir kt. Buvo apskaičiuotas respondentų KMI. Rezultatai: Nustatyta, kad Lietuvos gyventojai nepakankamai vartoja šviežių daržovių: mažiau nei pusė respondentų atsakė, kad šviežias daržoves valgo 3-5 kartus per savaitę, o apie trečdalį nurodė, jog valgo tik 1-2 kartus per savaitę. Moterims pagrindinis maisto pasirinkimo kriterijus yra kaina, vyrams – skonis, tačiau nuo 1997 m. padidėjo gyventojų, kurie renkasi produktus atsižvelgdami į ligų profilaktiką – 1997 m. 8,3 proc. gyventojų pagrindinis maisto pasirinkimo kriterijus buvo ligų profilaktika, o 2007 m. – 17,7 proc. In order to implement provisions of national, EU and WHO documents as well as improve public nutrition and health, nutritional research of population must be done. There is no doubt, that inappropriate nutrition can course many chronic diseases. The aim of research – to evaluate nutrition habits and their tendency of variation of 18-65 years old Lithuanian population. Tasks of research: 1. To examine and evaluate nutrition habits of Lithuanian population. 2. To determine tendency of variation of nutrition habits of Lithuanian population. 3. To evaluate body mass index of population and its tendency of variation. Methods of research: 3500 Lithuania adults were selected to participate in this research. The rate of research reply is 69,1 percent. During the research, each respondent was asked about nutrition and lifestyle peculiarities, certain nutrition aspects as well as how do they evaluate the state of factual nutrition. Body mass index was calculated of all respondents. Results: It was determined that Lithuanian population do not consuming enough fresh vegetables: less than hall of respondents answered, that they eat fresh vegetables 3-5 times per week, and about the third of respondents indicated, that eat fresh vegetables 1-2 times per week. Nustatyti 5-8 klasių mergaičių lankstumo duomenis. 3. Nustatyti 5-8 klasių mergaičių KMI ir lankstumo sąsajas. Hipotezė: Mergaitės su didesniu KMI yra lankstesnės. Svarbiausi rezultatai: Daugiausiai mergaičių yra su nepakankamu KMI – 51.52%, daugiausiai mergaičių yra su vidutiniu lankstumo lygiu – 47.48% ir daugiausiai mergaičių su vidutiniu KMI ir aukščiau vidutinio lankstumo lygio buvo – 30.77%. Išvados: 1. Tarp 5-8 klasių mergaičių daugiausiai buvo su nepakankamu KMI t.y. < 18.5 (kg/cm) Tokių mergaičių yra 51.52%. 2. Daugiausiai mergaičių buvo vidutinio lankstumo lygio, kurių testo rezultatai buvo nuo 20-25cm. Geriausi lankstumo rezultatai pasiekti mergaičių su vidutiniu KMI t.y, nuo 18.5 – 24.9 (kg/cm) , kurių rezultatų – 28.13cm. Procentaliai, tokių mergaičių, pasiekusių aukščiau vidutinio lankstumo lygį t.y. nuo 25 – 30cm., buvo 30.77%, iš visų vidutinį KMI turinčių moksleivių, o blogiausi rezultatai mergaičių su nepakankamu KMI, kurių rezultatų - 26.75cm. Procentaliai, aukščiau vidutinio lankstumo lygio pasiekusių mergaičių yra tik 3.92% iš visų tirtų mergaičių su nepakankamu KMI.
Research object: BMI and flexibility. Research goal: to determine BMI and flexibility in interfaces. Final work tasks. 1. Detection of 5-8 classes for girls BMI data. 2. Detection of 5-8 classes for girls flexibility data. 3. Detection of 5-8 classes for girls BMI and flexibility interface. Hypothesis: the girls with a higher BMI are more flexible. The most important results: Most of the girls are with the lack of BMI - 51.52%, most of the girls are with the average level of flexibility - 47.48% and most of the girls with the average BMI and above average level of flexibility has been – 30.77%. Conclusions: 1. Between 5-8 girls class was largely with a lack of BMI, this is < 18.5 (kg/cm). Such girls are 51.52%. 2. Most of the girls had a moderate level of flexibility, whose test results had been 20-25 cm. Such girls are 47.48%, and the minimum was above the average level of flexibility, the results amounted to between 25-30cm, percent to - 20.20%, girls. 3. The best results of the flexibility is for girls with the average BMI, this is from 18.5 – 24.9 (kg/cm) which results in the - 28.13cm. Such girls, above the, average level of flexibility, this is from 25 – 30cm., was 30.77%, of all students with the average BMI and having the worst results for girls with the lack of BMI, which results in the - 26.75cm. The girls above the average level of flexibility in performing girls there are only 3.92% from the total number of girls with the insufficiency of the BMI.

O intuito desta dissertação é estudar o modelo KMP. Este é um clássico modelo de interação constituído por uma cadeia de osciladores harmônicos unidimensionais desacoplados que trocam energia por meio de um processo estocástico. Cada elo tem um relógio de Poisson. Sempre que o relógio toca, dois osciladores vizinhos redistribuem energia de maneira uniforme. Além disso, o sistema está em contato com reservatórios nas extremidades, à diferentes temperaturas. Neste trabalho, apresentamos o estudo deste modelo e mostramos a validade da Lei de Fourier.

This thesis presents the development of a Web Service for the Hotline module of the Knowledge Management Information Tool (KMIT), a tool that is custom built for the decontamination & decommissionin (D&D) community of the Department Of Energy (DOE). The Hotline module allows interested users to post problems to specific areas of interest in the field of D&D. Various clients working with DOE and KMIT want to display the latest published problems of KMIT Hotline search in their own applications on a regular basis. Considering one of the major benefits of Web Services is the ease of integration of one piece of software with another, the Hotline Service is successfully developed and can be plugged into client’s applications by adding a reference to it. In such a distributed environment, messages can flow from node to node, through firewalls, onto the internet and through various intermediaries. This introduces a variety of message security threats. The research for this thesis included a study of the various security risks and scenarios. Appropriate security model is designed and is successfully implemented. A leishmaniose é uma doença de escala global, que afeta 12 milhões de pessoas e pode causar um espectro de doenças que vai desde a forma cutânea localizada, que tende para a cura espontânea, até a forma visceral que é fatal. Apesar da gravidade da doença, até o momento não existe uma vacina efetiva para prevenir a leishmaniose. Dentre os antígenos promissores para o desenvolvimento de uma vacina, destacam-se as proteínas ribossomais (S4, S6, L3 e L5) e a KMP-11, uma proteína de superfície presente nos membros da família tripanosomatidae. Nosso estudo consistiu em avaliar os efeitos da imunização com estes antígenos frente ao desafio com L. major e com L. braziliensis, empregando modelos experimentais de infecção. Primeiramente, avaliamos a capacidade protetora dos antígenos ribossomais frente à infecção por L. major. Dos quatro antígenos avaliados, apenas L3 ou L5 foram capazes de prevenir o desenvolvimento da lesão e de diminuir a carga parasitária. A vacinação de camundongos com estes antígenos, na presença de CpG, induziu um perfil de resposta Th1, com elevada produção de IFN-γ, baixa produção de IL-10 e presença de anticorpos IgG2a. Em seguida, avaliamos a capacidade protetora dos antígenos L3 e L5 contra o desafio por L. braziliensis, na presença da saliva do vetor. A imunização com os antígenos L3 e/ou L5 também induziu uma elevada produção de IFN-γ, resultando em significativa redução na espessura da lesão e menor carga parasitária. Com relação ao antígeno KMP-11, investigamos a sua capacidade protetora utilizando duas estratégias vacinais: a estratégia homóloga que consistiu na imunização de camundongos com um plasmídeo de DNA que codifica KMP11 (DNA KMP-11) e a estratégia heteróloga que consistiu na imunização com nanopartículas de PLGA contendo DNA KMP-11, seguido de um reforço com nanopartículas contendo a proteína KMP-11 sob forma recombinante, na presença de CpG. As nanopartículas protegem o antígeno da degradação enzimática e promovem a liberação controlada deste, além de atuar como um adjuvante. Ambas as estratégias não impediram o desenvolvimento da lesão, após o desafio com L. braziliensis e na presença de saliva do vetor. Entretanto, os animais imunizados com a estratégia heteróloga apresentaram uma maior redução da carga parasitária comparado com o grupo imunizado pela estratégia homóloga. Este efeito foi associado com uma maior produção de IFN-γ e de TNF-α no sítio da infecção. Por fim, avaliamos a indução da resposta imune inata em macrófagos estimulados com KMP-11 encapsulados em nanopartículas. Observamos que a estimulação de macrófagos murinos com KMP-11, encapsulada em nanopartículas de PLGA, reduziu a carga parasitária intracelular e aumentou a produção de oxido nítrico, superóxido, TNF-α, IFN-γ, IL-6, IL-1β, IL-18, CCL2/MCP-1, CXCL-1/KC sugerindo a indução de uma potente resposta imune inata. Assim, concluímos que os antígenos L3 e/ou L5 mostraram ser promissores para o desenvolvimento de uma vacina que proteja contra as principais espécies de Leishmania e que o encapsulamento de antígenos em nanopartículas é capaz de induzir uma forte resposta imune. Essa estratégia deve ser considerada quanto ao desenvolvimento de vacinas para a leishmaniose.
Leishmaniasis is a global disease affecting 12 million people and can cause diseases that range from self-healing localized cutaneous leishmaniasis to fatal visceral leishmaniasis. Despite the severity of the disease, there is no effective vaccine to prevent leishmaniasis. Among the promising antigens for the development of a vaccine, stand out the ribosomal proteins (S4, S6, L3, and L5) and KMP-11, a surface protein, widely found in the members of family Trypanosomatidae. Our study evaluated the effects of immunization with these antigens upon challenge with L. major and L. braziliensis, employing the experimental models of infection. First, we evaluated the protective ability of ribosomal antigens to infection by L. major. Among the four antigens examined only L3 or L5 were able to prevent lesion development and decrease the parasite load. Mice vaccinated with these antigens, plus CpG, developed a Th1-type response with high production of IFN-γ, low production of IL-10 and presence of IgG2a antibodies. Next, we evaluated the protective capacity of L3 and L5 antigens against challenge by L. braziliensis, in the presence of sand fly saliva. Vaccination with L3 or L5 also induced a high production of IFN-γ, resulting in significant inhibition of lesion development and lower parasite load. Regarding KMP-11, we investigated its protective capacity using two immunization strategies: the homologous strategy, which consisted in immunizing mice with a plasmid DNA encoding KMP-11(DNA KMP-11) while the heterologous immunization strategy consisted of inoculation of PLGA nanoparticles (NPs) containing DNA KMP-11 followed by a booster inoculation with nanoparticles containing recombinant KMP-11, in the presence of CpG. Nanoparticles protect the antigen from enzymatic degradation and promote controlled release, in addition to acting as an adjuvant. Lesion development was not inhibited following either immunization strategy, after challenge with L. braziliensis in the presence of sand fly saliva. However, animals immunized with the heterologous strategy showed a greater reduction in parasite load compared with the group immunized by the homologous strategy. This effect was associated with increased production of IFN-γ e TNF-α at the infection site. Finally, we evaluated the induction of the innate response in macrophages stimulated with KMP-11 encapsulated in NPs. We observed that stimulation of murine macrophages with KMP-11 encapsulated in NPs reduced the parasitic load and increased production of nitric oxide, superoxide, TNF-α, IFN-γ, IL-6, IL-1β, IL-18, CCL2/MCP-1, CXCL-1/KC suggesting the induction of a potent innate immune response. We conclude that the L3 and/or L5 are promising antigens for the development of a vaccine that protects against the main species of Leishmania and that encapsulation of antigens into nanoparticles induces strong immune response. This strategy should be considered for the development of vaccines against leishmaniasis.

Plus spécifiquement, nous reconnaissons dans cette recherche, à l'instar de nombreux auteurs, que c'est moins la dimension technique que les modalités de mise en œuvre de la solution qui importent [Benghozi, 2001]. Dans le cas où les pratiques sont incertaines et complexes parce que mêlant des acteurs différents, une réflexion sur la notion d'usage nous conduira ainsi à mettre en évidence la nécessité d'une co-conception des solutions TIC et à proposer la mise en œuvre de cette démarche à travers une recherche intervention dans le cadre du projet KMP au sein de Telecom Valley. Cette expérimentation nous permettra alors de mettre en exergue le rôle de la construction d'objets frontières dans la co-évolution de la conception et des usages.

As leishmanioses formam um grupo de doenças que afetam 350 milhões de pessoas atualmente, atingindo 88 países em todo o mundo com uma estimativa de 1-2 milhões de novos casos por ano. O desfecho da infecção causada por Leishmania depende tanto de fatores do patógeno como do hospedeiro, embora a virulência de Leishmania possa ser modulada por fatores ambientais e genéticos relacionados aos hospedeiros mamíferos e vetores, os determinantes moleculares são elementos-chave no estabelecimento da infecção, ou seja, são os fatores que determinam a virulência. A KMP-11 é uma glicoproteína que está presente em todos os cinetoplastideos. O presente estudo avaliou a expressão do gene de KMP-11 de L. amazonensis ao nível de RNA e ao nível de proteína, durante passagens sucessivas de promastigotas de fase estacionária através de cultivo in vitro, investigando se há associação entre a sua expressão e a virulência dos parasitos. A avaliação da virulência dos parasitos mantidos em cultura foi realizada através do acompanhamento da evolução da infecção experimental murina (modelo in vivo) durante um período de dez semanas, juntamente com a observação do surgimento de ulceração. A quantificação da carga parasitária foi realizada nos linfonodos drenantes das lesões, através da através da técnica de diluição limitante A avaliação também foi realizada pela infecção de macrófagos murinos (modelo in vitro). Os resultados de medição de pata foram analisados pelo teste não paramétrico ANOVA 2 fatores, seguido do pós-teste de Bonferroni. Além disso, foi realizada a determinação da proporção de metacíclicas nos promastigotas de fase estacionária mantidos em cultura, através da técnica de lise pelo complemento. Nossos resultados mostraram que há um decréscimo da virulência dos promastigotas de fase estacionária ao longo do número de passagens, pois os camundongos infectados com as passagens iniciais desenvolveram lesões maiores do que aqueles com os promastigotas mantidos em cultura por mais tempo. Quanto ao surgimento de ulcerações, na 10a semana pós-infecção, todos os animais infectados com promastigotas de 1a passagem apresentavam lesões ulceradas, enquanto que nenhum dos camundongos infectados com promastigotas de 20a passagem apresentava lesão ulcerada. Na infecção in vitro, a carga parasitária nos macrófagos testados diminuiu em função do número das subculturas, o que foi demonstrado através do decréscimo da porcentagem média de macrófagos infectados e pela quantidade média de amastigotas a cada 100 macrófagos infectados. A quantificação da carga parasitária foi realizada nos linfonodos drenantes da lesão dos camundongos infectados, confirmando a diminuição da virulência dos promastigotas. A quantificação da proporção de promastigotas metacíclicas demonstrou que a porcentagem diminui ao longo do tempo de subcultivo. A avaliação da expressão de KMP-11 na superfície de promastigotas por citometria de fluxo demonstrou um decréscimo na expressão da proteína proporcional ao número de subculturas. Verificou-se, portanto, uma associação entre a expressão da proteína KMP-11 e a virulência de promastigotas de L. amazonensis. Os resultados dos ensaios de PCR em tempo real demonstraram que não há diferença estatisticamente significativa na quantidade de transcritos do gene da proteína KMP-11 entre as passagens analisadas. Entretanto, a perda da virulência associada com a diminuição da expressão da proteína KMP-11 indica que esta molécula possua uma função na infectividade dos promastigotas de Leishmania amazonensis, atuando possivelmente como um fator de virulência.
The leishmaniases are a group of diseases that currently, affects 350 million people reaching 88 countries throughout world with an estimated incidence of 1-2 million new cases per year. The outcome of the infection caused by Leishmania depends on factors from the pathogen and from the host. Although Leishmania virulence can be modulated by environmental and genetic factors related to mammalian hosts and vectors, molecular determinants are key elements in the establishment of infection and for the determination, of virulence. KMP-11 is a glycoprotein which is present in all kinetoplastids. This study evaluated the gene expression of KMP-11 in L. amazonensis at RNA level and at protein level, during successive passages of in vitro culture, investigating a possible correlation between KMP-11 expression and virulence of parasites. The evaluation of the virulence of cultured parasites was performed by monitoring of the progression of the lesions in experimental murine infection (in vivo model) during ten weeks, along with the emergence of cutaneous ulcers. The quantification of parasite load was performed on draining lymph nodes using the limiting dilution analysis. The assessment of parasite virulence was also performed by the infection of murine macrophages (in vitro model). The paw measurement results were analyzed by nonparametric test ANOVA 2 way, followed by Bonferroni post-test. Furthermore, the metacyclic promastigotes proportion in stationary growth phase from cultures with different numbers of passages was evaluated by complement lysis. Our results showed a decrease in promastigotes of stationary phase virulence that correlated with the increase of the number of passages, as mice infected with the early passages developed larger lesions than those infected with promastigotes cultured for longer periods and higher numbers of passages. Concerning the development of ulcers, at 10th week post-infection, all animals infected with promastigotes of first passage presented ulcerated lesions, whereas none of the mice infected mice with promastigotes of the 20th passage showed an ulcerated lesion. Analyzing the in vitro infection, the parasite burden in macrophages decreased with the number of subcultures, as demonstrated by the decrease in the percentage of infected macrophages and in the number of amastigotes per 100 infected macrophages. The quantification of parasites in draining lymph nodes of the infected mice confirmed the decrease in the virulence of promastigotes from cultures with more passages. The estimation of the metacyclic promastigote proportions showed that the percentages decline through the time of subculture. The evaluation of KMP-11 expression on the surface of promastigotes by flow cytometry showed a decrease in protein expression proportional to the number of subcultures. Therefore, there was an association between the expression of KMP-11 protein and the virulence of L. amazonensis promastigotes. The results of real-time PCR assays showed that there is no statistically significant difference in the amount of gene transcripts of KMP-11 protein in the analyzed passages. However, the loss of virulence associated with decreased protein expression of KMP-11 indicates that this molecule may have a role in promastigotes infectivity of Leishmania amazonensis, possibly acting as a virulence factor.

Abstract:
Darbo objektas – Lietuvos sporto universiteto Socialinės pedagogikos studijų krypties studentų KMI ir fizinis aktyvumas. Darbo tikslas – ištirti Lietuvos sporto universiteto Socialinės pedagogikos studijų krypties studentų fizinį aktyvumą, KMI ir sąsajas tarp jų. Uždaviniai: 1. Nustatyti ir įvertinti Lietuvos sporto universiteto Socialinės pedagogikos studijų krypties studentų kūno masės indeksą; 2. Ištirti Lietuvos sporto universiteto Socialinės pedagogikos studijų krypties studentų fizinio aktyvumo lygį; 3. Nustatyti ir įvertinti Lietuvos sporto universiteto Socialinės pedagogikos studijų krypties studentų fizinio aktyvumo bei kūno masės indekso sąsajas. Hipotezė: socialinės pedagogikos studentų fizinis aktyvumas yra per mažas, KMI nėra normalus (18,5 arba daugiau, arba mažiau kaip 25,0). Rezultatai: 1. Didesnės daugumos (80 proc.) apklaustų Lietuvos sporto universiteto Socialinės pedagogikos studijų krypties studentų kūno masės indeksas yra normalus. Vos keletas respondentų (3 proc.) turi per mažą kūno masės indeksą, o likusieji turėjo antsvorio. 2. Lietuvos sporto universiteto Socialinės pedagogikos studijų krypties studentų fizinis aktyvumas nėra aukštas. Daugiau negu pusė (60 proc.) neužsiėminėjo aukšto intensyvumo fiziniu aktyvumu, o kurie užsiėminėjo tai darė 1-2 dienas per savaitę (16,7 proc.) ar 3-4 dienas per savaitę (21,7 proc.). The object: Lithuanian Sports University Social Pedagogy students’ BMI and physical fitness. The aim: to investigate Lithuanian Sports University Social Pedagogy students’ physical fitness, BMI and interconnections between them. The Objectives: 1. To calculate and evaluate BMI (Body Mass Index) of Lithuanian Sports University Social pedagogues. 2. To investigate the level of their physical fitness. 3. To establish and evaluate connections between Social Pedagogy students’ physical fitness and their BMI. Hypothesis: the physical activity of social pedagogues is inadequate, BMI is not normal (18.5 or less, or more than 25.0). The Results: 1. The majority of Lithuanian Sports University Social pedagogues (80%) had an optimal BMI, 3% of them had low BMI and the rest had overweight. 2. The level of physical fitness of the aforementioned students was not very high. More than a half (60%) did not exercise. Those who did took physical activity 1-2 days a week (16.7 %) or 3-4 days a week (21.7 %). The average level of physical fitness was not peculiar for 33.3% of respondents. Moreover, almost three quarters of them (30%) had an average physical activity 1-2 days a week, and 26.7% exercised 3-4 days a week. However, the majority of students (80%) said they go somewhere by foot 5-7 days per week. 3. The physical fitness interrelates with BMI. Abstract Fatigue crack growth thresholds ΔKth are provided by several fitness-for-service (FFS) codes. When evaluating cracked components subjected to cyclic loading, maximum stress intensity factor Kmax and/or minimum stress intensity factor Kmin are required. However, the definitions of the thresholds ΔKth under negative stress ratio R are not clearly written, except for BS (British Standards) 7910. In addition, the ΔKth are given by constant values under negative R. Fatigue crack growth rates under negative stress ratio is recommended to use maximum stress intensity factor Kmax by ASTM (American Society of Testing and Materials) E 647, because of the Kmax being close to crack driving force. Therefore, it deems that the ΔKth under negative R seems to be Kmax. This paper shows that the Kmax converted by the ΔKth are not constant values under negative R based on the survey of experimental data. The Kmax decreases with decreasing the stress ratio R. Therefore, the ΔKth for the FFS codes are less conservative. As experimental data under negative stress ratio R were taken by Kmax – Kmin, the definition of the threshold ΔKth is benefit to use Kmax – Kmin, instead of Kmax.

The conducted studies allow us to conclude that the complexes KMP-1 (Diazophyte, Phosphoenterin, and Biopolycid) and KMP-2 (Phosphoenterin, Biopolycid, Aurill, and Azotobacterin) in water dilution 1:10 can be recommended for varieties with increased resistance to phytopathogens during open stratification to protect grafted grape cuttings from phytopathogens, while applying partial (apical) bacterization. The effectiveness of these microbial complexes is of the same standard as the action of the traditional chemical antiseptic Chinosolum at a concentration of 0.5 %.
