Abstract:
Cholangiocarcinoma is a rare type of cancer which is an increasingly discernible health threat. The disease is usually very difficult in diagnosis and various treatment modalities are typically not effective. Cholangiocarcinoma is a complex and very heterogeneous malignancy characterized by tumor location, different risk factors, molecular profiling, and prognosis. Cancer cell lines represent an important tool for investigation in various aspects of tumor biology and molecular therapeutics. We established two cell lines, KKU-452 and KKU-023, which were derived from patients residing in the endemic area of liver fluke infection in Thailand. Both of tumor tissues have gross pathology of perihilar and intrahepatic mass-forming cholangiocarcinoma. Two cell lines were characterized for their biological, molecular and genetic properties. KKU-452 and KKU-023 cells are both adherent cells with epithelium morphology, but have some differences in their growth pattern (a doubling time of 17.9 vs 34.8 h, respectively) and the expression of epithelial bile duct markers, CK7 and CK19. Cytogenetic analysis of KKU-452 and KKU-023 cells revealed their highly complex karyotypes; hypertriploid and hypotetraploid, respectively, with multiple chromosomal aberrations. Both cell lines showed mutations in p53 but not in KRAS. KKU-452 showed a very rapid migration and invasion properties in concert with low expression of E-cadherin and high expression of N-cadherin, whereas KKU-023 showed opposite characters. KKU-023, but not KKU-452, showed in vivo tumorigenicity in xenografted nude mice. Those two established cholangiocarcinoma cell lines with unique characters may be valuable for better understanding the process of carcinogenesis and developing new therapeutics for the patients

Abstract:
Cholangiocarcinoma (CCA) is a bile duct cancer, commonly found in Asia including Thailand and especially in the northeastern region of Thailand. To identify the proteins involved in carcinogenesis and metastasis of CCA, protein expression profiles of high-invasive KKU-M213 and low-invasive KKU-100 cell lines were compared using a comparative GeLC-MS/MS proteomics analysis. A total of 651 differentially expressed proteins were detected of which 27 protein candidates were identified as having functions involved in cell motility. A total of 22 proteins were significantly upregulated in KKU-M213, whereas 5 proteins were downregulated in KKU-M213. S100A2, a calcium-binding protein in S100 protein family, is upregulated in KKU-M213. S100A2 is implicated in metastasis development in several cancers. The protein expression level of S100A2 was verified by Western blot analysis. Intriguingly, high-invasive KKU-M213 cells showed higher expression of S100A2 than KKU-100 cells, consistent with proteomic data, suggesting that S100A2 may be a key protein involved in the progression of CCA. However, the biological function of S100A2 in cholangiocarcinoma remains to be elucidated. S100A2 might be a potential biomarker as well as a novel therapeutic target in CCA metastasis.

Coconut crab (Birgus latro) is a fishery resource with high economic value. Coconut Crab is only found in eastern Indonesia, covering Sulawesi, Maluku and North Maluku. One of the habitat of Coconut Crab in North Maluku is Moor Island, Central Halmahera. The coastal typology of this island is predominantly rocky with many gaps and small caves, as well as several steep slopes with dominant vegetation conditions of coconut, beach pandanus and other coastal plants. Walnut Crab is often used as a consumption ingredient because it has delicious taste and high nutritions. However, the use of crustaceans for consumption can also be dangerous. This is because crustaceans also contain several pathogenic bacteria. This study aimed to identify pathogenic bacteria isolated from Coconut Crab which is the largest crustacean in the world. 5 samples of Coconut Crab were taken from Moor Island. Isolated samples marked KK1, KK2, KK3, KK4 and KK5. Pure bacterial isolates were obtained from Coconut Crab samples, evaluated for colony type and identified based on biochemical characterization. The types of pathogenic bacteria identified in samples KK1, KK2, KK3, KK4 and KK5 were Escherichia coli, Pseudomonas sp. and Staphylococcus sp.

Abstract:
Cholangiocarcinoma (CCA) is a highly metastatic tumor, and the majority of patients with CCA have a short survival time because there are no available effective treatments. Hence, a better understanding regarding CCA metastasis may provide an opportunity to improve the strategies for treatment. A comparison study between the highly metastatic cells and their parental cells is an approach to uncover the molecular mechanisms underlying the metastatic process. In the present study, a lung metastatic CCA cell line, KKU-214L5, was established by the in vivo selection of the tail vein-injected mouse model. KKU-214L5 cells possessed mesenchymal spindle-like morphology with higher migration and invasion abilities in vitro than the parental cells (KKU-214). KKU-214L5 also exhibited extremely aggressive lung colonization in the tail vein-injected metastatic model. Epithelial‐mesenchymal transition (EMT) was clearly observed in KKU-214L5 cells. Significant downregulation of epithelial markers (ZO-1 and claudin-1), with unique upregulation of E-cadherin and mesenchymal markers (vimentin, β-catenin, and slug), was observed in KKU-214L5. Increasing MMP-2 and MMP-9 activities and CD147 expression reflected the high invasion activity in KKU-214L5 cells. Suppression of vimentin using siRNA significantly decreased the migration and invasion capabilities of KKU-214L5 to almost the basal levels of the parental cells without any change on the expression levels of other EMT markers and the activities of MMPs. These results suggest that vimentin activation is essential to potentiate the metastatic characters of CCA cells, and suppression of vimentin expression could be a potential strategy to improve the treatment of CCA, a highly metastatic cancer.

Abstract:
An actinomycete strain (KKU-A3) with algicidal activity against Microcystis aeruginosa was isolated from soil in Khon Kaen Province, Thailand. Based on its phenotypic characteristics and 16S rDNA sequence, strain KKU-A3 was identified as Streptomyces rameus. Strain KKU-A3 also exhibited algicidal activity against the cyanobacteria Synechococcus elongatus, Cylindrospermum sp. and Oscillatoria sp. A mathematical and statistical technique was used to optimize the culture conditions and maximize its anti-Microcystis activity. The single factor experiments indicated that glucose and casein were the most effective carbon and nitrogen sources, respectively, and produced the highest anti-Microcystis activity. Response surface methodology indicated that the optimum culture conditions were 19.81 g/L glucose and 2.0 g/L casein at an initial pH of 7.8 and an incubation temperature of 30 °C. The anti-Microcystis activity increased from 82% to 95% under optimum conditions. In an internal airlift loop bioreactor, the removal of M. aeruginosa KKU-13 by the bacterium was investigated in batch and continuous flow experiments. In the batch experiment, KKU-A3 displayed maximum anti-Microcystis activity of 95% at day 7, whereas in the continuous flow experiment, KKU-A3 displayed maximum anti-Microcystis activity of 95% at day 10.

Abstract:
This research aims to investigate the impact of employing the learning management system (Blackboard) on attitudes of King Khalid University (KKU) students. The research examines the students' knowledge and skills in employing the Blackboard in their learning process, and investigates the elements, which influence the students' attitude and performance in using Blackboard in classroom instruction from their perspectives. To this end, the author has selected a sample of 34 of KKU students, where they have been requested to complete a 14-item questionnaire. Findings of the current study revealed that KKU students faced several difficulties in learning English language vocabulary. Consequently, the students have developed negative attitudes towards English vocabulary. The study found that Blackboard had a little influence on the attitude and performance of the KKU students in vocabulary learning. The study also showed that there were some limitations in the use of Blended Learning. It needs an action plan in order to make supportive learning opportunities for the university students.

In this paper we included more reference models such as experiences of Quality instruments affected by demographic criteria. In Higher education systems E-Learning faces two major challenges, first to ensure the interoperability of E-Learning (IEL) and secondly, developing quality learning through e-Learning. This research studies the concept, scope and dimensions of interoperability of E-Learning in education with special reference to King Khalid University then the connection and interdependence between with quality development. To impart learning and teaching through E-learning, KKU has adopted Learning Management Services (LMS) through Blackboard. The university has three types of learning and teaching methods; full online, Blended and Supportive. In this paper we have described the dimensions of quality and the standards of E- Learning for the objectives of IEL and quality development (QD) in KKU. The research is based principally on secondary data observed from KKU E-Learning deanship. Also sample of 20 E-Learning experts at KKU were given closed ended as well as semi closed questionnaires for evaluating the assurance of IEL and QD. These experts are mainly certified online facilitators and admin staff. Results provide the verification of application and presence of IEL and assured the QD process in KKU in imparting knowledge.

Abstract:
Drought remains the most important factor that affects rice productivity, especially in rainfed areas, worldwide. Upland rice is one of the crop choices of farmers in the rainfed environment. Although upland rice varieties require less water than lowland rice varieties, yields often remain limited by drought, particularly in the period of early growth. The aims of this study were to identify the traits related to early drought tolerance in upland rice varieties, and to identify the potential sources of germplasm for early drought tolerance. A total of sixty upland rice varieties were planted in a factorial experiment with a randomized complete block design with 3 replications in the rainy seasons of 2011 and 2012, under greenhouse conditions. Based on the drought tolerance index (DTI), the test germplasm sources were classified into three groups: (i) susceptible; (ii) moderately tolerant; (iii) tolerant to drought stress. Grain yield (GY) showed significant negative correlations with the leaf rolling score (r= − 0.623, P< 0.01), the leaf death score (LDS) (r= − 0.673, P< 0.01) and the recovery score (r= − 0.746, P< 0.01), while leaf dry matter (r= 0.698, P< 0.01) and leaf water potential (r= 0.618, P< 0.01) had significant positive correlations with GY. These findings indicate the suitability of the DTI as the selection criteria for early drought tolerance in a breeding programme. In addition, the upland rice germplasm accessions KKU-ULR011, KKU-ULR012, KKU-ULR125, KKU-ULR199 and KKU-ULR292 were identified as having high levels of stability for drought tolerance in both the 2011 and 2012 experiments, suggesting their potential for further use for rice variety improvement for drought tolerance.

Anemia is a global public health problem. The prevalence of anemia among different ages, genders or ethnic groups must be clarified in order to solve problems. This study proposed to determine the prevalence and factors related to anemia among the Muslim school-age population in Nakhon Si Thammarat, Thailand. Socio-demographic and anthropometric data were collected by a structured questionnaire. Blood samples were collected from 200 school-age subjects. The thalassemia screening was performed with KKU-OF and KKU-DCIP reagents. The prevalence of anemia in this study was 36.5%, divided into males and females, 33.3% and 39.1%, respectively. The means of Hb, Hct, MCV, MCH, and MCHC in the anemic group were significantly lower. The positive results for KKU-OF or KKU-DCIP or both were 15.0%, 2.5%, and 1.0%, respectively. The result of positive OF test was a significantly independent factor for anemia. The number of family members was 5 to 7 and more than 7 persons are related factors for anemia in this study. In summary, the contribution of thalassemia and socio-economic factor are associated factors to anemia in this population. These findings should be addressed in public health strategies for the control of anemia of school-aged Muslims in the region.

Abstract:
The need to understand which factors most strongly affect performance in first-year mathematics programs at Khon Kaen University (KKU), in North Eastern Thailand, provided the main focus of the study which is described. First-year mathematics students in the 1990-1991 academic year, from four KKU faculty groups (Medicine and Nursing, Agriculture, Science and Education, and Engineering) were involved in this study. Research literatures addressing variables which were likely to influence performance in early tertiary mathematical study, and variables associated with difficulties in learning mathematics at the transition from upper secondary school to tertiary studies, were reviewed. The first major aim of the study was to identify the variables which were good predictors of first-year mathematics performance at KKU. Results from stepwise multiple regression analyses indicated that the following predictor variables were statistically significant and entered the regression equations for most Faculty groups: School Mathematics Achievement, Self-Esteem, Study Habits in Mathematics, and Faculty of Study. Other predictor variables that sometimes entered regression equations (depending on the Faculty group) were Socio-Economic-Status, Mathematics Language Competence, Mathematics Confidence, Attitude Towards Mathematics, and Gender. Depending on Faculty group, the statistically significant variables accounted for between 11% and 74% of scores on fist-year KKU mathematics examinations. The predictor variables contributed much more to the variance of scores on first-semester mathematics examinations than to the variance of scores on second-semester mathematics examinations. It was also found that scores on the Direct Entry Examination Mathematics test (administered by KKU) and the School Mathematics Achievement test (developed and administered by the author) had stronger correlations with first-year KKU mathematics performance than did scores on the National Entry Examination Mathematics tests (administered by the Thai Ministry of University Affairs). Scores on the three pre-university mathematics achievement test instruments were better predictors of first-semester mathematics performance than of second-semester mathematics performance. It was found that the mean Mathematics Confidence of male students was statistically significantly higher than that of female students, but there were no statistically significant gender differences in Mathematics Misplaced Confidence. Only about 30% of the main sample ( 30% of the male and 30% of the female sample groups) had appropriate confidence in mathematics, that is, they thought their answers were correct when they were, in fact, correct, and they thought they were wrong when they were, in fact, incorrect. So far as Faculty performance differences were concerned, Engineering students had the highest Mathematics Confidence scores, followed by the Medicine and Nursing group of students and the Science and Education group students. Agriculture students had the lowest mean Mathematics Confidence score. No statistically significant differences occurred in Mathematics Misplaced Confidence between different Faculty groups. The second main aim of the study was to investigate why many first-year students experienced difficulties in coping with their mathematics units. A small group of senior secondary mathematics teachers, university mathematics lecturers, and first-year mathematics students were interviewed during the first semester of the 1990-1991 academic year. Interviews were conducted by the author according to a questionnaire format, and were aimed at identifying factors causing difficulty in the transition from senior secondary to university mathematical study. The analysis of the quantitative data together with the interview data indicated that the major sources of difficulty were associated with: (a) students' mathematical abilities; (b) curriculum content; (c) course organisation; (d) students' study habits; (e) instructional styles; and (f) assessment procedures. The results of the investigation are discussed in the light of the relevant literature and related research. The study concludes with recommendations which are addressed to mathematics teachers and education administrators in senior secondary schools in Thailand, to the Thai Ministry of Education, and to the KKU Department of Mathematics.

Abstract:

This research presents new preconditioners for linear systems. We proceed fromthe most general case to the very specific problem area of sparse optimal control.In the first most general approach, we assume only that the coefficient matrix isnonsingular. We target highly indefinite, nonsymmetric problems that cause difficultiesfor preconditioned iterative solvers, and where standard preconditioners, likeincomplete factorizations, often fail. We experiment with nonsymmetric permutationsand scalings aimed at placing large entries on the diagonal in the context of preconditioningfor general sparse matrices. Our numerical experiments indicate that thereliability and performance of preconditioned iterative solvers are greatly enhancedby such preprocessing.Secondly, we present two new preconditioners for KKT systems. KKT systemsarise in areas such as quadratic programming, sparse optimal control, and mixedfinite element formulations. Our preconditioners approximate a constraint preconditionerwith incomplete factorizations for the normal equations. Numerical experimentscompare these two preconditioners with exact constraint preconditioning andthe approach described above of permuting large entries to the diagonal.Finally, we turn to a specific problem area: sparse optimal control. Many optimalcontrol problems are broken into several phases, and within a phase, mostvariables and constraints depend only on nearby variables and constraints. However,free initial and final times and time-independent parameters impact variables andconstraints throughout a phase, resulting in dense factored blocks in the KKT matrix.We drop fill due to these variables to reduce density within each phase. Theresulting preconditioner is tightly banded and nearly block tri-diagonal. Numericalexperiments demonstrate that the preconditioners are effective, with very little fill inthe factorization.

KSU Course Reader is an android application developed for Kansas State University students to access their course materials, listen to the audio lectures, watch the video lectures and read the lecture slides or other materials from all their enrolled courses with a single application. A student can add RSS xml link associated with the courses enrolled in the current semester in the application and can receive the course materials for each course automatically after every lecture. The aim of this project is to develop a one stop android application for students to access the course materials of all their courses from their android smart phones and tablets. The project also provides a jsp form for course instructors to update the course materials for every lecture which automatically updates the xml file associated with the course. The application also allows students to add other RSS xml like K-State News, K-state Events etc. The same application can be used for adding public rss xml sites and can also be used as a podcast player. It is very essential to provide the course materials readily available to the students all the time wherever they are. Since most students have smart phones or tablets and use them for accessing emails, surf internet etc , it becomes easier for them to use this KSU course reader application to access, read or view all their course materials at one place which is also customized for mobiles and tablets. As android is one of the leading and fastest growing smart phone platforms the project becomes more appropriate to develop in android.

Einerseits wird die Rechtsnorm gem. § 84 Abs. 2 SGB IX aus arbeits- und sozialpolitischer Sicht in Form und Funktion aufgearbeitet. The research paper concentrates on Corporate Integration Management (CIM) in small and medium companies (SME). On the one hand the task consists in reviewing the form and function of the legal norm under Section 84 Subsection 2 of Book IX of the German Social Code (SGB) in terms of labour and social policies. On the other hand a manual-based strategy for implementing CIM shall be developed. Following the integration of the legally valid CIM concept with the appropriately adapted framework the manual is subjected to an empirical review. By accompanying 42 CIM cases in 10 SMEs it has been possible to show on the basis of partly standardized and open guided interviews with company experts that the introduction and implementation of CIM is markedly enhanced by means of this manual. The discussion of the theory-guided findings and the empirical results is followed by an outlook. Suggestions for further research are made while the political establishment is called upon to resolve the challenges existing within the scope of the prevention norm and revealed in the research paper thus facilitating an effective transfer of the CIM theory into the practice field.

Abstract:
The recently initiated German project CARISMA (Crack Initiation and Arrest of Irradiated Steel Materials) will create a data base on pre-irradiated original materials of the four generations of German nuclear pressurized water reactors, which allows the examination of the consequences if the Master Curve instead of the RTNDT concept is applied. Several original materials of the four generations of German nuclear power plants (typical for KWO, KKS - Biblis A, Biblis B, KKU - KKG, KWG, KKP2, KBR - KKE, KK12, GKN2) will be investigated. They have been irradiated in six large scale irradiation capsules in a German research reactor (the VAK plant) at corresponding plant conditions. The capsules contain regular tensile and Charpy impact specimens as well as Pellini and fracture toughness wedge opening load specimens up to a specimen thickness of 100 mm. The first fracture toughness tests have been performed on a weld metal NiCrMo1 UP(mod.)/LW320, LW340 (1. generation, lower bound of the weld materials)—with a fluence Φ = 2,12E19 cm−2 (E &gt; 1 MeV). This weld has a Cu-content of 0.22 wgt.% and it was therefore supposed to show a large transition temperature shift. Some fracture toughness tests in the irradiated material condition were already available and during this project four 100 mm thick wedge opening load (WOL) specimens were tested. At one of the specimens brittle failure could not be achieved during the test, because the load capacity of the test machine was exceeded. Therefore the specimen was loaded by a load-unload-cool-fracture load path to demonstrate the warm pre-stress effect of this highly irradiated specimen. At the final fracture of the specimen at a lower temperature, the failure load was significantly higher than the original one (factor 3 higher), which clearly indicates that the benefit of warm pre-stressing will not be eroded with irradiation.

The dose and time dependence of endotoxin induced activation of the plasma contact system was studied in citrated plasma incubated at 37,4 °C. During the first six hours no significant changes were seen. At 12 hours markedly elevated plasma kalli-krein (KK) activity was found in plasma tested with the highest doses of endotoxin (2000 and 200 γg/ml). Corresponding to the elevated KK activity reductions of both plasma prekallekrein (PKK) and functional kallekrein inhibition (KKI) were seen. Hageman factor (FXII) values determined immunologically were also reduced parallel to the decreases of PKK and KKI. Changes in high molecular weight kininogen (HMwK) values were seen as wellIn plasma tested with doses of endotoxin lower than 200γg/ml no sign of activation was seen at 12 hours. At 24 hours, however, increased KK activity and decreased KKI and PKK values were seen in plasma incubated with 20 γg/ml of endotoxin. In test plasma with endotoxin concentrations of 2 and 0,6 γg/ml we found . sign of activation in 7 of 8 test tubes using 2 γg/ml and in 2 of 6 test tubes using 0,6γg/ml. In test plasma with the lowest dose of endotoxin (2,0 γg/ml) and in control plasma no changes in either KK activity, KKI, PKK, FXII or HMwK values were found at 24 hoursThe study shows thaf- endotoxin induced activation of the contact system in citrated plasma is a slow process which is both time and dose

The results obtained in our randomised study in 3 groups of patients performed in 1982/83 suggested the administration of protease inhibitors to be effective in counteracting the activation of the kallikrein, coagulation and fibrinolytic systems during extracorporeal circulation (ECC) thus lowering the rate of postoperative bleeding complications. Since Cl-esterase-inhibitor (Cl-INH) proved to be the main inhibitor of the kallikrein (KK) system and aprotinin that of plasmin further studies to investigate the efficacy of a combined treatment and to develop an improved dosage regimen were initiated in 1985.Pilot:1: 3 patients/aprotinin; pilot 2: 3 patients/Cl-INH Concentrate; pilot 3: 5 patients/Cl-INH Concentrate + aprotinin. Test parameters included: protein/Biuret; Cl-INH/rocket immune electrophoresis and chromogenic substrate method; kaolin induced kallikrein generation; plasminogen activation using streptokinase; plasmin inhibiton capacity; coagulation factors I, II, V, VII, VIII, and X; reptilase clotting time, thrombin time; platelets and the thrombelastogram.15 blood samples were taken at regular intervals over approx. 16 hours before, during and after surgery, as well as on the first and second postoperative day.Dosage in pilot 1 and 2 was 500 × 103 KIU aprotinin and 1500 PU Cl-INH, respectively, followed by the continous i.v. infusion of 125 × 103 KIU aprotinin/h and 125 PU CI-INH/h, respectively over approx. 16 hours. This regimen still appeared inadequate to prevent the activation of the KK and fibrinolytic system. In pilot 3 therefore, the continous infusion of both concentrates was increased by 100 % (250 × 103 KIU aprotinin/h + 250 PU Cl-INH/h), while the bolus-injection remained the same as in pilot 1 and 2. Using this optimised combined regimen parameters of the kallikrein, coagulation and fibrinolytic system remained almost unchanged compared to values obtained prior to ECC.

The kinematic Kalman filter (KKF) is a sensor-based state estimator which is immune to the external disturbances and the parameter uncertainties of mechanical plants. This paper extends the original idea of the KKF to a more general form as a means to enhance a real time vision sensor for the end-effector control of a robot manipulator, the performance of which is often limited by its slow sampling rate. The original one-dimensional KKF is reformulated in a higher dimensional form by incorporating the measurements from the vision sensor, accelerometers and gyroscopes. A nonlinear state space model of the kinematics of the end-effector is derived including the time delay associated with vision sensing. Then, the new KKF is formulated as a state estimator combining the inter-sample predictions with an extended Kalman filter (EKF). The paper discusses practical issues such as the real time computation to implement the EKF and the vision sensor to measure the absolute position. Experimental results are presented to confirm the benefits of the new KKF using a two-link direct drive manipulator equipped with a dual axis MEMS accelerometer, a single axis MEMS gyroscope and an end-effector mounted vision camera. The accurate estimation of the position and velocity of the end-effector from the new KKF will be useful for the real time visual-servo and the task space control of robot manipulators.

The complex loading of the knee is believed to play an extensive role in the development and advancement of joint disease and injury. Computational models provide a powerful tool in knee biomechanics, but validation of these models is difficult. The objective of this study was to develop a dynamic 3-D subject-specific computational knee model and to validate the model using experimental data from a dynamic knee simulator. The developed knee model was placed in a previously validated model of the Kansas Knee Simulator (KKS) dynamic testing machine [1]. The kinematics from walking and squat profiles simulated in the KKS model and performed on the knee specimen during KKS experimental testing were compared for model validation.

Dynamic knee simulators aim to reproduce prescribed physiological loading and motions of the knee. The natural knee achieves stability through a complex interaction of the neuro-musculoskeletal system; thus, a knee simulator also requires a sophisticated control system to replicate human motion. Guess and Maletsky [1] developed a computational model to predict the required simulator inputs to produce the desired knee loading for dynamic activities on the Kansas Knee Simulator (KKS). The model built demonstrated conceptually that multi-body dynamics models could be used to simulate the KKS. However, as desired loading profiles became more complex, key limitations were discovered in the model; such as the model controller limited to a single axis under feedback control, no out-of-plane loading, not accounting for dynamic joint friction or damping of the actuators, and an inability of the model to flex pass 80° degrees of knee flexion. Thus, there was a need for a new computational model to overcome the limitations and to provide a more robust and complete comparison to the KKS. The new computational model will allow better utilization of the KKS capabilities for future cadaveric and prosthetic testing. This work outlines the sagittal-plane validation of the new computational model.

The major benefit of the kinematic Kalman filter (KKF), i.e., the state estimation based on kinematic model is that it is immune to parameter variations and unknown disturbances regardless of the operating conditions. In carrying out complex motion tasks such as the coordinated manipulation among multiple machines, some of the motion variables measured by sensors may only be available through the communication layer, which requires to formulate the optimal state estimator subject to lossy network. In contrast to standard dynamic systems, the kinematic model used in the KKF relies on sensory data not only for the output but also for the process input. This paper studies how the packet dropout occurring from the input sensor as well as the output sensor affects the performance of the KKF. When the output sensory data are delivered through the lossy network, it has been shown that the mean error covariance of the KKF is bounded for any non-zero packet arrival rate. On the other hand, if the input sensory data are subject to lossy network, the Bernoulli dropout model results in an unbounded mean error covariance. More practical strategy is to adopt the previous input estimate in case the current packet is dropped. For each case of packet dropout models, the stochastic characteristics of the mean error covariance are analyzed and compared. Simulation results are presented to illustrate the analytical results and to compare the performance of the time varying (optimal) filter gain with that of the static (sub-optimal) filter gain.

Alterations of the kallikrein-kinin system (KKS) consistent with activation and increased consumption have been reported in septic patients and it has been suggested that this activation could contribute to the development of septic shock. As similar alterations have been found in other critically ill patients, many of them prone to shock, we wonder if activation of the KKS could be a consequence rather than a cause of the hemodynamic instability. To answer this question we compared 12 patients with septic shock (8 fatal) with 10 cases of cardiogenic shock secondary to myocardial infarction (8 fatal) as a model of non septic shock. In adition 25 episodes of uncomplicated sepsis and 10 intra-intensive care unit controls were studied. A functional measure of factor XII, high molecular weight kininogen (HMWK) (coagulative methods), prekallikrein (PK), Cl-inhibitor (Cl-INH), α2-macroglobulin (α2-M)- Antithrombin III (AT-III) and kallikrein inhibitor activity (KIA) (chromogenic methods) was performedRESULTS: Patients with septic shock, specially in fatal cases, showed a highly significant decrease in activities of factor XII (P<0.001), PK (P<0.0001), HMWK (P<0.005), α2-M (P<0.001), AT-III (P<0.0001) and KIA (P<0.005). Cl-INH activity was increased in uncomplicated sepsis (P<0.001) but came back to normal or was slightly decreased in septic shock. Components and inhibitors of the KKS were within normal limits in all patients with cardiogenic shock.Our findings support the idea of a contribution of the KKS to the development of septic shock but this system neither seems to play a role in the pathogenesis of cardiogenic shock nor to be altered as a consequence of it.

Acute pancreatitis (AP) was induced in juvenil pigs by injection of Na-taurocholate into the pancreatic duct. Eight animals remained untreated (group A), while 7 pigs (group B) received 60000 KIU/kg aprotinin (Tras-ylol, Bayer AG Leverkusen, West Germany) intravenously during 30 min before the induction of AP, thereafter 10000 KIU/kg/h during a 6 h observation period. Seven pigs (group C) received 60000 KIU/kg aprotinin during 30 min starting 3 h after the induction of AP, thereafter 10000 KIU/kg/h. The total infusion volume was the same in all 3 groups (5 ml/kg/h). Using an enzym-linked immunosorbent assay for aprotinin, maximum plasma concentration of aprotinin in group B was found 30 min after start of the infusion (2.8(1.9-5.4)umol/l). The aprotinin concentration thereafter remained elevated for the rest of the observation period (1.4(0.4-1.9) umol/1 after 6 h). The aprotinin concentration gradually increased in the peritoneal exudate during ongoing aprotinin infusion. After 1 h the aprotinin conc. in plasma and the exudate were within the same range. Similar results were observed in group C experiments, where the aprotinin conc. in plasma and peritoneal exudate both were approximately 2 umol/1 1 h after start of the infusion. Assayed by the chromogenic peptide substrate S-2302 (Kabi Vitrum AB, Stockholm, Sweden), markedly elevated values of plasma kallikrein inhibition were detected both in plasma and peritoneal exudate after start of aprotinin infusion in group B and C.In group A, 5 out of 8 animals died in a circulatory collapse, while the animals in group B and C remained hemodynamic stable and all survived the observation.Aprotinin concentrations of 1-3 umol/1 in plasma and peritoneal exudate improves the outcome during experimental acute pancreatitis in pigs.
