Abstract:
Kettin is a large actin-binding protein with immunoglobulin-like (Ig) repeats, which is associated with the thin filaments in arthropod muscles. Here, we report identification and functional characterization of kettin in the nematode Caenorhabditis elegans. We found that one of the monoclonal antibodies that were raised against C. elegans muscle proteins specifically reacts with kettin (Ce-kettin). We determined the entire cDNA sequence of Ce-kettin that encodes a protein of 472 kDa with 31 Ig repeats. Arthropod kettins are splice variants of much larger connectin/titin-related proteins. However, the gene for Ce-kettin is independent of other connectin/titin-related genes. Ce-kettin localizes to the thin filaments near the dense bodies in both striated and nonstriated muscles. The C-terminal four Ig repeats and the adjacent non-Ig region synergistically bind to actin filaments in vitro. RNA interference of Ce-kettin caused weak disorganization of the actin filaments in body wall muscle. This phenotype was suppressed by inhibiting muscle contraction by a myosin mutation, but it was enhanced by tetramisole-induced hypercontraction. Furthermore, Ce-kettin was involved in organizing the cytoplasmic portion of the dense bodies in cooperation with α-actinin. These results suggest that kettin is an important regulator of myofibrillar organization and provides mechanical stability to the myofibrils during contraction.

Abstract:
Kettin is a giant muscle protein originally identified in insect flight muscle Z-discs. Here, we determined the entire nucleotide sequence of Drosophila melanogaster kettin, deduced the amino acid sequence of its protein product (540 kD) along with that of the Caenorhabditis elegans counterpart, and found that the overall primary structure of Kettin has been highly conserved in evolution. The main body of Drosophila Kettin consists of 35 immunoglobulin C2 domains separated by spacers. The central two thirds of spacers are constant in length and share in common two conserved motifs, putative actin binding sites. Neither fibronectin type III nor kinase domains were found. Kettin is present at the Z-disc in several muscle types. Genetic analysis showed that kettin is essential for the formation and maintenance of normal sarcomere structure of muscles and muscle tendons. Accordingly, embryos lacking kettin activity cannot hatch nor can adult flies heterozygous for the kettin mutation fly.

Abstract:
Kettin is a high molecular mass protein of insect muscle that in the sarcomeres binds to actin and α-actinin. To investigate kettin's functional role, we combined immunolabeling experiments with mechanical and biochemical studies on indirect flight muscle (IFM) myofibrils of Drosophila melanogaster. Micrographs of stretched IFM sarcomeres labeled with kettin antibodies revealed staining of the Z-disc periphery. After extraction of the kettin-associated actin, the A-band edges were also stained. In contrast, the staining pattern of projectin, another IFM–I-band protein, was not altered by actin removal. Force measurements were performed on single IFM myofibrils to establish the passive length-tension relationship and record passive stiffness. Stiffness decreased within seconds during gelsolin incubation and to a similar degree upon kettin digestion with μ-calpain. Immunoblotting demonstrated the presence of kettin isoforms in normal Drosophila IFM myofibrils and in myofibrils from an actin-null mutant. Dotblot analysis revealed binding of COOH-terminal kettin domains to myosin. We conclude that kettin is attached not only to actin but also to the end of the thick filament. Kettin along with projectin may constitute the elastic filament system of insect IFM and determine the muscle's high stiffness necessary for stretch activation. Possibly, the two proteins modulate myofibrillar stiffness by expressing different size isoforms.

with the economic development, the three-kettle condensation process is more and more widely applied. This paper introduces three-kettle condensation and its features and advantages. Comparing the changes of steam consumption and solvent residue in rubber before and after the modification of the cis-1,4-polybutadiene rubber condensation system, the obtained result shows that compared with the two-kettle condensation, the three-kettle condensation system can steam by 25%, and also can reduce 50% of the solvent residual liquid in rubber. The results after the process optimization on three-kettle condensation system show that the bottom temperature of No.1 kettle is 85 °C, and the top pressure is 48 kPa; the bottom temperature of No. 2 kettle is 102 °C, and the top pressure is 90 kPa; and the top pressure of No. 3 kettle is 10 kPa. In addition, here points out the differences in optimization, and gives suggestions to the optimization.

Abstract:
In 2018 KTH alumni Frida Bylund and Linnea Kåwe investigated, on behalf of C3 Scandinavian lifestyle, a new way of boiling water on an induction cooktop. The idea was to develop an induction kettle that shuts itself off when the water reaches 100°C. This product would fulfil the user desire to keep their kitchen counter minimalistic and clean from clutter and home appliance products. This thesis is a continuation of Bylund and Kåwe’s work and carried out together with Svensk konstruktionstjänst AB. The mechanism of the product was to be evaluated and developed, resulting in a refined solution. A repetitive prototype was to be built to present and ensure the functionality of the mechanism. Furthermore, a design concept was to be realised as renderings and a physical model, which included the mechanism. The project was divided into four phases: Frame of reference, Development of the mechanism, Evaluation of final prototype, Industrial design Proposal. The first phase consisted of testing and evaluating Bylund and Kåwe’s mechanism. Furthermore, a literature study was conducted to examine key components and materials for the product. The second phase consisted of generating concepts for the mechanism using the methods; Crazy eight´s, Brainstorming/Braindrawing, Structural and Shape variation and Negative brainstorming. The concepts were evaluated with the Weight criteria matrix to find the most suitable solution. The third phase consisted of building a robust and repetitive prototype which could be tested with users as well as evaluated against the requirements specification. The user tests were carried out with the Think aloud method and Semi structured interviews. Finally, a design proposal for the product was designed and presented with renderings and a physical model in the fourth phase. A solution for the mechanism was found and the functional prototype was successful. It could boil water 20 times in a row over the course of three days without failure - half a liter of water boiled in 4 minutes and 45 seconds. However, the prototype was only able to manage about 1000W which resulted in a long boiling time. The power drop was partly because the jug was convex in the bottom which created an unwanted distance to the induction cooktop, hence decreasing the transferred power. The second reason was most likely that the ferrous material, which established contact with the induction cooktop, had lost some of its magnetic properties during manufacturing of the prototype. If a better jug and ferrous material for the induction cooktop could be manufactured, the prototype would perform better. An induction kettle could be more sustainable than an electric kettle since it does not consist of electrical components. An eco-analysis was conducted to compare the design suggestion of the induction kettle with an electric kettle. The simplified life-cycle assessment showed that the induction kettle had better end of life potential than an electric kettle, however the materials selected for the induction kettle proved to be on the critical materials list which made the carbon footprint of the induction kettle higher than expected. This could be lowered if the induction kettle does not require virgin materials for its components. The purpose and the goal for the project was reached, a solution for the mechanisms was found and evaluated - the mechanism proved to be robust enough to further develop the product. Areas of future work were found; The housing and other plastic components has to be investigated further and the tolerances of the mechanism has to be customised for the solution.
KTH alumnerna Frida Bylund and Linnea Kåwe undersökte ett alternativt sätt att koka vatten på induktionshällar. Projektet utfördes år 2018 på företaget C3 Scandinavian Lifestyles vägnar. Idén bestod i att utveckla en mekanisk vattenkokare som stänger av sig själv när vattnet kokat upp till 100°C. Produkten skulle även uppfylla Skandinavers drömmar om ett minimalistiskt kök fritt från stökiga köksbänkar. Detta Masterexamensarbete är en fortsättning på Bylund och Kåwes arbete och utfördes tillsammans med Svensk Konstruktionstjänst AB mot C3 Scandinavian Lifestyle. Målet var att evaluera och utveckla mekanismen i produkten för att hitta en förfinad robust lösning. En funktionsprototyp skulle tas fram - den skulle visa på mekanismens duglighet och repeterbara funktionalitet. Ett designförslag skulle realiseras i form av renderingar och en fysisk uteseendemodell som inkluderade mekanismen. Projektet delades in i fyra faser: Frame of reference, Development of the mechanism, Evaluation of final prototype, Industrial design Proposal. Den första fasen bestod av att testa och evaluera Bylund och Kåwes mekanismförslag, samt att utföra en litteraturstudie vilken undersökte nyckelkomponenter och material för produkten. I fas två utfördes konceptgenerering och evaluering för konceptet. Metoder som: Crazy Eight´s, Brainstorming/Braindrawing, Structural and Shape variation, Negative brainstorming och Kriterieviktsmetoden användes. Den tredje fasen bestod av att konstruera en robust funktionsprototyp som repeterbart skulle kunna koka vatten. Användartester utfördes med prototypen och viktiga insikter samlades in med hjälp av Think Aloud metoden och Semi strukturerade intervjuer. Prototypen evaluerades även mot en framtagen kravspecifikation. Slutligen konstruerades ett designförslag av den mekaniska vattenkokaren i form av renderingar och en fysisk uteseendemodell, detta utfördes i projektets fjärde fas. En väl fungerande mekanism-lösning banade vägen för en fungerande prototyp. Prototypen kokade vatten 20 gånger i rad, med testerna löpande under tre dagar, utan problem. En 0,5 liter vatten kokades på 4 minuter och 45 sekunder. Koktiden var längre än önskvärt vilket berodde på att kannan var konvex på insidan, vilket resulterade i ett oönskat avstånd till induktionshällen uppstod. Detta resulterade i en förlorad effekt. Prototypen mätte upp till ca 1000W vid användning. En orsak till att effekten var låg i prototypen var då det ferromagnetiska materialet, som förser kontakten med induktionshällen, hade delvis hade förlorat sin magnetiska förmåga i och med värme som uppstått vid tillverkandet av komponenten. Resultatet hade varit annorlunda om kannan varit planare i botten och den ferromagnetiska komponenten hade varit tillräckligt magnetisk. Från ett hållbarhetsperspektiv skulle en mekanisk vattenkokare kunna vara mer hållbar än en elektrisk vattenkokare eftersom den inte in består av några elektriska komponenter. En livscykelanalys var utförd för att jämföra designförslaget för den mekaniska vattenkokaren med en elektrisk vattenkokare. En förenklad livscykelanalys visade att en mekanisk vattenkokare hade en bättre end-of-life potential i jämförelse med en elektrisk vattenkokare, däremot visade det sig att de valda materialen för den mekaniska vattenkokaren var listade som kritiska material vilket gjorde att koldioxidavtrycket för den mekaniska vattenkokaren var högre än förväntan. Detta kan sänkas om den mekaniska vattenkokaren inte kräver rena obehandlade material för dess komponenter. Syftet och målet med projektet uppnåddes, en lösning för en fungerande mekanism hittades och evaluerades - mekanismen visade sig att vara tillräckligt robust för vidare utveckling av produkten. Områden för vidare utveckling var; undersökande av funktionen med plastkomponenter och toleranser för mekanismen.

The study aimed at an investigation of the vegetation of kettle-holes in the agricultural landscape of the Ziemia Chelminska region in northern central Poland. Most of these kettle-holes have been influenced strongly by human activities. 120 community types were described with regard to their species composition, structure, differentiation, synanthropization, ecological conditions and dynamics, as well as their affinities to similar plant communities from other regions in Europe. The significance of the kettle-holes as a natural resource is briefly discussed.

Abstract:
Abstract This paper explores situational factors and attitudes affecting sustainable electric kettle usage. We conducted a two-week long field study with five residents in Denmark who self-reported their electric kettle usage data, purpose of usage, and situations of usage, over the study period. A follow-up interview was also conducted after the field study to explore reasons behind the observed behaviors. Results show, the amount of excess water heated significantly varied by usage purpose as well as the time sensitivity of the usage purpose. There was no observable effect of the minimum fill level marker on the amount of excess water heated. Participants’ considerations for purchasing a specific kettle and their usage behaviors were not influenced by sustainability-related concerns. We identified four important factors that influenced the amount of excess water heated and participants’ decision to retain or dispose it. They included, (1) uncertainty in the need for hot water in the near future, (2) flexibility in the needed volume for a usage purpose, (3) forgetting to use heated water, and (4) concerns about limescale build up.

In modern combustion engines timing chains are used to actuate camshafts, balance shafts, diesel injection pumps, oil pumps and auxiliaries. Optimizing the engine design with respect to operation reliability, efficiency, noise and acoustics requires a profound knowledge of the vibrational behavior of all chain drive components. The amount of expensive experiments can be significantly reduced by analyzing and studying such systems using computer simulation. At the Institute of Applied Mechanics at the Technical University of Munich, Germany, a multi body simulation program has been developed to compute the dynamical behavior of complex timing chain drives. It is used by several members of the German FVV (Forschungsvereinigung fuer Verbrennungskraftmaschinen), e.g. Porsche, Audi, BMW, DaimlerChrysler and others. This paper describes how the components and the effects of timing chain drives are modelled within the simulation tool. the underlying models are to be explained but also a comparison between simulation and experiment will be presented for a DaimlerChrysler 8-cylinder diesel engine (OM628) which drives the Mercedes-Benz S-Class (S 400 CDI).

Context: Non-ketotic hyperglycemia is a rare cause of chorea. Clinical findings are usually unilateral and potentially reversible after treatment for hyperglycemia. Hyperglycemia leads to asymmetric multifocal petechial hemorrhages of the basal ganglia, leading to a dysfunction of neuronal networks that connect the basal ganglia and the motor cortical areas, mainly affecting the subthalamic nucleus and contralateral striatum, which is highlighted by typical hyperdense lesions of the basal ganglia in computed tomography (CT) of the brain. This study aimed to report a case of a patient with choreiform movements due to a rare etiology of hyperglycemia nonketotic in a Hospital Public of Joinville, SC. The study was carried out through the collection and analysis of a patient’s medical record. Case report: Female patient, 54 years old, who presented for 6 days choreiform movements in the face, left upper limb and, discreetly, in the left lower limb. Snake tongue sign and milkmaid’s grip positive, without dysarthria. In the laboratory exam, glucose of 600 mg / dL; without further changes. Cranial tomography showed hyperdensity in the putamen region on the right. The treatment was started to obtain better glycemic control and Risperidone 3 mg / day. Conclusions: It is concluded, then, that non-ketotic hyperglycemia is an uncommon, but reversible cause of chorea, and may manifest itself due to an uncontrolled non-ketotic diabetes mellitus. Its pathogenic mechanism remains to be clarified. In addition, clinical, epidemiological, imaging and laboratory findings, together, corroborate for early diagnosis and proper management.

Introduction: Type 2 diabetes mellitus is one of the diseases that is most associated with chorea, and although it is a rare complication, it is necessary tobe aware of it so that the correct diagnosis and early treatment can be made. Case report: Male patient, 78 years old, diabetic and uncontrolled hypertension. Began uncontrolled glycemic 7 days before admission. Evolves with imbalance when walking and with involuntary movements in the left upper limb. At admission, dextro 682 mg/dl and at neurological exam the presence of asymmetric choreoathetotic movements, more evident in the left dimidium. Investigation with neuroimaging, brain tomography showed the presence of hyperdensity in bilateral basal nuclei, confirmed by brain MRI. Other differential diagnoses were ruled out, such as hyperthyroidism, liver failure and rheumatic fever. The most likely diagnosis was generalized choreoathetosis secondary to a non-ketotic hyperglycemic disorder. During hospitalization, adequate glycemic control was performed and clonazepam was introduced as an aid, with significant improvement of movements and absence of the same at discharge. Conclusions: Cases of non-ketotic hyperglycemia are associated with the onset of chorea, and although it is a rare complication, it is not uncommon given the high incidence of diabetes mellitus in the Brazilian population, and clinical improvement may take weeks to months, even after adequate glycemic control.

Minden szuverén állam a belső jogában önállóan határozhatja meg az állampolgári jogokat és kötelezettségeket, éppen ezért nincs egységes álláspont arra nézve, hogy jogi értelembe mely jogok és kötelezettségek alkotják az állampolgárságot. Ugyan vannak nemzetközi és európai ügyek (lsd. Nottebohm-ügy, Rottmann-ügy, Tjebbes-ügy, Micheletti-ügy), melyek bár kiemelnek bizonyos elvi tételeket, mely tételeket és joggyakorlatot az államoknak figyelembe kellene venniük, azonban ez a jogkövető magatartás nem mindig érvényesül. Az1960-as évek végétől Csehszlovákiában több törvény is rendelkezett a szlovák, a cseh és a szövetségi állam állampolgárságának megszerzéséről és elvesztéséről. A magyar kedvezményes honosítás ellenreakciójaként értelmezhető a szlovák kormánynak a szlovák állampolgárságról szóló törvény alkotmányt sértő, szigorú jellegű módosítása, amely ahhoz vezetett, hogy 2022. decemberéig több mint 4300 ember veszítette el szlovák állampolgárságát. A szlovák Parlament tavaly fogadta el az állampolgársági törvény ismételt módosítását, mely a megfosztott szlovákiai magyarok többségének helyzetén mit sem javított. Felmerül tehát a kérdés, hogy az Európai Unióhoz csatlakozott Szlovákia állampolgársági törvény módosításai megfelelnek-e az európai uniós és a nemzetközi jogi normáknak? Vagy hogy megvalósul-e Szlovákiában az egyenlő elbánás elve az állampolgárság vonatkozásában? Meglátásom szerint ezek mind-mind olyan érdekfeszítő és releváns kérdések, amelyek nagymértékben meghatározzák és rányomják bélyegét a mai társadalmunkra.

The Adaptation Resources for Agriculture Workbook was jointly developed by USDA Climate Hubs and NRCS to support producers, service providers, and educators to manage climate change. The workbook helps producers consider both short-term adaptive management actions (<5 yrs) and long-range strategic plans (5 to 20 yrs, subject to farm type). This workbook pro-motes adaptation through multiple resources including a “menu” of adaptation strategies/approaches and example tactics for cropping and forages, confined livestock, grazing, orchards and small fruit and vegetable production systems. Recent efforts by USDA Climate Hub NRCS Liaisons work to increase the number of examples, and have been documented as Case Studies. These Case Studies are of producers utilizing the 5-step process in the workbook to document their management choices to ameliorate climate change impacts to their operations.

Odonates are significant components of most wetland habitats and important indicators of their health. At Cape Cod National Seashore (CACO), we compiled odonate records dating back to the 1980s and, based partly on that data, identified 41 wetland sites for sampling, representing six freshwater habitats (kettle pond, inter-dune pond, dune slack, riparian marsh, vernal pool, and bog). We surveyed these sites for adult odonates during the 2016?2018 field seasons. Ten sites were surveyed all three years (total 19-20 surveys/site); all ten had at least some historical data. The remaining 31 sites were surveyed for one field season, a total of 6-8 times each. We conducted 391 surveys, recording 53,435 individuals and 74 species (45 dragonflies and 29 damselflies); not all individuals were identified to species. Abundance and species richness varied significantly between habitats. For all individuals recorded, abundance was greatest at vernal pools and kettle ponds. Riparian sites had the lowest abundance. Species richness was highest at kettle ponds, including several species of conservation concern, two listed as Threatened by the state of Massachusetts. Riparian marshes and dune slacks had relatively low richness. Among the 10 sites surveyed three years, we found significant annual variation in abundance and species richness. There was significant and generally greater between-site variation in abundance within a year than between years at sites. Community analysis found pond depth, habitat type, and presence of predaceous fish were significant factors explaining between-site variation in community composition. Habitats also differed significantly in community composition. Multidimensional scaling showed sites tend to cluster together by habitat type. Vernal ponds have the highest average community similarity to all other habitats (53.5%), with dune slack (52.9%), bog (52.0%) and inter-dune (51.5%) close behind. In contrast, riparian sites (46.3%) and kettle ponds (39.5%) are least similar to other habitats. Overall, 86 species of odonates have been recorded at CACO, a rich and diverse assemblage reflecting the variety and quality of freshwater habitats present. Although these habitats are relatively well-protected, stressors include climate change, nutrient inflow from adjacent development, road runoff, and trampling of emergent vegetation. A plan for monitoring is beyond the scope of this project. Ideally, it would be best to use the insight into odonate variation obtained from these surveys to develop a monitoring program designed to meet standards of statistical confidence and power currently employed in NPS monitoring programs.

Along transects and within boxplots, several ponds have experienced significant changes in macrophyte species composition since 1995, while others have remained relatively unchanged. In 2020 new occurrences of Phragmites australis (common reed) were documented in Duck (a single plant, which was immediately removed), Gull, and Higgins ponds. Three state-listed rare species that occur in CACO kettle ponds were not found in 2020. These included Sabatia kennedyanna (Plymouth rose gentian) (found in 2016 at Great-T), Drosera filiformis (threadleaf sundew) (found in 2016 at Great-W) and Sagittaria teres (quill-leaved arrowhead), which was once abundant but has now been absent for decades. Reductions in the cover of N. odorata (white waterlily) along transects was observed in Great-T, Ryder, Higgins, and Snow ponds, although this trend was apparent in boxplots only in Herring Pond. E. acicularis (dwarf harigrass) also diminished in cover in several ponds, including along Duck, Dyer, Gull, Long, Round-E, and Slough ponds. J. militaris (bayonette rush) appears to have undergone a variety of short-term changes including reductions along transects in Great-T, Higgins, Long, and Snow ponds. Boxplots only exhibited a reduction in Higgins Pond, however. L. dortmanna (Cardinal flower) is another taxon common to most CACO ponds. Its abundance along the Gull Pond transects decreased, contrasted by an increase in Long Pond transects, and in Great-W and Round-E boxplots. The proliferation of Utricularia spp. in Great-T Pond is one of the most conspicuous changes that has occurred over the last decade. The plant community shifts reported here are likely the result of both short-term (e.g., trampling, geese grazing, extreme drought or high water, etc.) and long-term (e.g., climate warming, diminishing acidity, increasing alkalinity). That said, many of the statistically significant increases or decreases in cover were restricted to just one or two transects, and sometimes the changes in individual taxa contrasted each other among the different transects. One-dimensional line-intercept transects are likely detecting changes that are not real on any meaningful spatial scale. On the other hand, the number of boxplot sites should be increased to obtain much better spatial resolution. Accordingly, we propose increasing the number of box plot sites in each pond from five to ten for all future monitoring.

Ziel des im Jahr 2018 abgeschlossenen Projektes ist es, eine „nachhaltigere Chemie“ in der textilen Lieferkette zu unterstützen und dabei den Blick von der „reaktiven“ Compliance Position auf eine „proaktive“ Beyond Compliance Perspektive zu erweitern. Strategisch stützt sich dieser Ansatz auf folgende Überlegung: Wer morgen noch „Compliant“ sein will, muss heute bereits „Beyond Compliance“ agieren. Infant formula is a breastmilk substitute fed to babies when mums are unable or do not want to breastfeed. In the UK, almost three quarters of babies will have consumed infant formula by six weeks of age, and almost all will have by six months (McAndrew et al., 2012). Formula fed babies are at greater risk of gastrointestinal infections than breastfed babies because breastfeeding is protective against infections as it helps babies’ immune systems develop, and because bottles of formula are at risk of bacterial contamination. Bacterial contamination is thought to occur in two ways; first, powdered infant formula (PIF) is not sterile and can contain harmful bacteria, including Salmonella and Cronobacter if not prepared properly (Crawley, Westland & Sibson, 2022), and second, bottles and teats are vulnerable to contamination during preparation (Redmond et al., 2009; Cho et al., 2019). It is estimated that in the UK over 3,000 babies end up in hospital each year, and a further 10,000 are reviewed by GPs, due to gastrointestinal infections which may be attributed to formula feeding (Renfrew et al., 2012). NHS (2019) guidance states that PIF must be mixed with water at a temperature of 70o Celsius (oC) or greater, to kill any bacteria which may be present in the PIF. The use of boiled water from a kettle cooled to at least 70oC is recommended, which is then mixed with the PIF before allowing it to cool further before feeding. This should be repeated every time a bottle is needed to ensure the formula is safe. Bacteria can survive and multiply in formula, even if it is stored in a fridge. NHS guidance also contains steps to minimise contamination of baby feeding equipment, including washing hands, disinfecting preparation surfaces, and washing and sterilising all baby feeding equipment. However, research shows many parents do not carry out all these steps, and a third of parents do not feel confident about preparing PIF (Brown, Jones and Evans, 2020). Furthermore, there has been an increase in UK parents using formula preparation machines and their efficacy has not yet been sufficiently investigated.
