Abstract:

Thermodynamic and kinetic properties of O2 and CO binding to haemoglobin (Hb) Kempsey [Asp-G1(99) beta----Asn] were investigated and the activation parameters for the two ligands were determined. At every temperature the O2-binding isotherms display a weak co-operativity, n ranging between 1.1 and 1.2, and dissociation kinetics show a single-exponential behaviour. O2-binding kinetics were studied at 25 degrees C by temperature jump and are characterized at each saturation (from Y = 0.31 to Y = 1.0) by two processes, a fast bimolecular one and a slow monomolecular one (tau -1 = 20 s-1), which contributes to approx. 30% of the whole relaxation amplitude at every Y. CO-binding kinetics to Hb Kempsey were followed at several temperatures by flash photolysis and stopped flow. The process is biphasic, as reported elsewhere [Bunn, Wohl, Bradley, Cooley & Gibson (1974) J. Biol. Chem. 249, 7402-7409], and the relative contributions of the two bimolecular rates to the whole process are only slightly affected by temperature. On taking account for the fraction of dimers at every protein concentration, the slow phase corresponds to approx. 50% of the ligand binding to tetramers. Correlation of these results with previous spectroscopic data leads to the hypothesis that the biphasic time course of CO binding may be attributed to alpha/beta heterogeneity of the R-state of tetrameric Hb Kempsey.

Abstract:

Through the use of mitochondrial DNA (ATP8 gene), the prediction of intermediate genetic structuring was investigated in two species of estuarine glassfish (Ambassis marianus and Ambassis jacksoniensis) (Perciformes : Ambassidae) to determine the possibility of a generalised ‘estuarine’ genetic structure. Individuals were collected from estuaries in eastern Australia between Tin Can Bay (Queensland) in the north and Kempsey (New South Wales) in the south. Analysis of the haplotype frequencies found in this region suggested panmictic populations with star-like phylogenies with extremely high levels of genetic diversity, but with no correlation between geographic distance and genetic distance. Non-significant FST and ΦST suggested extensive dispersal among estuaries. However, Tajima’s D and Fu’s FS values suggest ‘mutation–genetic drift equilibrium’ has not been reached, and that population expansions occurring 262 000 (A. marianus) and 300 000 (A. jacksoniensis) years ago may obscure any phylogeographic structuring or isolation by distance. The finding of panmixia was contrary to the prediction of genetic structuring intermediate between that of marine fish (shallowly structured) and freshwater fish (highly structured), suggesting high dispersal capabilities in these species.

Abstract:

On March 30, 1984, I was given the privilege of delivering the introductory speech at the first American Medical Association (AMA) meeting exclusively for child abuse and neglect. Although one might wonder why 22 years lapsed between the Journal of the American Medical Association (JAMA) publication of the milestone article by Henry Kempe1 and the first meeting, the fact remains that the AMA is now paying attention to the needs of abused children and their families. My talk at this conference came just 3 weeks after the death of Henry Kempe, some 20 years almost to the date when he asked if I would like to be involved with his study of the battered child syndrome.

Dalam mencapai misi dan matlamat, setiap kempen berbeza dari segi pendekatannya bergantung kepada objektif dan khalayak sasaran. Matlamat utama kempen kesedaran awam bertujuan untuk memberi kesedaran kepada masyarakat mengenai sesuatu dasar, perkhidmatan, program atau mengenai sesuatu isu. Oleh itu, kerajaan bertanggungjawab merancang dan melaksanakan kempen Hari Kebangsaan dengan teliti bagi mencapai tujuan yang direncanakan agar dapat dihayati dengan sepenuh hati oleh golongan sasar. Dalam usaha mendidik dan meningkatkan kesedaran masyarakat terhadap kepentingan bulan Kebangsaan, setiap tahun kerajaan akan menganjurkan kempen Hari Kebangsaan bagi menyuntik semangat patriotisme di kalangan masyarakat. Oleh itu, wacana tentang kemerdekaan yang diketengahkan oleh pihak kerajaan melalui kempen bulan Kebangsaan saban tahun harus dirancang dengan teliti agar kewibawaan, konsep dan motif yang direncanakan dalam sesebuah kempen dapat dihayati dengan sepenuh hati oleh golongan sasar. Penggunaan media sosial dalam kempen kesihatan di Malaysia kebelakangan ini dilihat kian dipentingkan. Senario positif ini memberikan gambaran bahawa pelbagai pihak mulai melihat kelebihan yang dimiliki oleh medium ini dalam membantu menangani isu-isu yang menjadi risiko dan ancaman kepada kesihatan awam. Walau bagaimanapun, kajian-kajian yang dijalankan bagi menilai keberkesanan medium ini dalam kempen kesihatan awam umumnya dan Kempen Bahaya Penyalahgunaan Dadah khususnya dilihat amatlah terhad. Tujuan kajian ini dijalankan adalah untuk menilai tahap keberkesanan media sosial dan strategi keunikannya dalam Kempen Bahaya Penyalahgunaan Dadah di Malaysia. Reka bentuk kajian keratan rentas kaedah campuran digunakan bagi kajian ini iaitu kaedah kuantitatif (survei) dan kaedah kualitatif (temu bual mendalam). Sejumlah 400 orang responden direkrut bagi perolehan data survei dan seramai 12 orang informan direkrut bagi perolehan data temu bual mendalam. Dapatan utama survei menunjukkan bahawa keberkesanan strategi keunikan media sosial adalah di tahap yang tinggi. Manakala, dapatan utama temu bual mendalam pula menunjukkan bahawa adanya keberkesanan media sosial AADK dari segi (1) keupayaannya membantu pihak AADK dalam konteks kebolehgunaannya yang mudah dan murah, (2) keupayaannya membantu mempengaruhi masyarakat, (3) perolehan sambutan yang menggalakkan daripada masyarakat, (4) perolehan capaian yang tinggi, serta (5) strategi keunikan berjaya menarik perhatian masyarakat. Secara keseluruhannya, penggunaan media sosial dalam Kempen Bahaya Penyalahgunaan Dadah di negara ini mempunyai keberkesanan. Penggunaan medium ini wajar diteruskan serta diperhebatkan lagi dengan strategi yang lebih kreatif dan inovatif sekaligus meningkatkan keberkesanan sedia ada. Setiap tahun, perayaan Hari Kebangsaan disambut meriah dengan pelbagai kempen dan program anjuran kerajaan bagi menyuntik semangat patriotisme dan nasionalisme di kalangan masyarakat. Sambutan perayaan Hari Kebangsaan ini dianjurkan dengan harapan ia dapat membina jiwa kenegaraan yang tinggi dalam kalangan masyarakat yang majmuk. Pengisian dalam kempen Hari Kebangsaan adalah sepenuhnya menjadi tanggungjawab kerajaan untuk mengatur pelan strategi yang paling sesuai dengan keadaan masyarakat Malaysia yang berbilang bangsa dan agama. Di sini, pengaruh realiti sosial takrifan autoriti dijangka memainkan peranan dalam mengawal situasi untuk mengekalkan sesuatu penguasaan yang telah diperolehi. Oleh itu, wacana tentang kemerdekaan yang diketengahkan oleh pihak kerajaan melalui kempen bulan Kebangsaan saban tahun harus dirancang dengan teliti agar kewibawaan, konsep dan motif yang direncanakan dalam sesebuah kempen dapat dihayati dengan sepenuh hati oleh golongan sasar. Dalam masa yang sama kebijaksanaan kerajaan dalam merangka kandungan kempen perlu agar integriti kerajaan dalam memimpin negara diperolehi dan dipandang tinggi di mata dunia. Kaedah perbincangan kumpulan fokus dan temubual mendalam telah dijalankan untuk mengenalpasti adakah wujudnya unsur ‘authority-defined’ dalam pemilihan isi kandungan kempen bulan Kebangsaan. Dapatan yang digarap daripada perbincangan kumpulan fokus dan temu bual mendalam dikaitkan dengan konsep realiti sosial takrifan autoriti sebagai pemangkin kepada pembentukan terhadap penguasaan dalam kalangan rakyat negara ini. Kajian ini diharap dapat memberikan pemahaman yang lebih jelas mengapa konsep takrifan autoriti (authority-defined) itu lebih penting untuk dipraktiskan dalam pengurusan sesebuah pemerintahan berbanding amalan takrifan harian (everyday-defined).

Penggunaan papan tanda sebagai medium penyampaian maklumat sudah tidak asing lagi di negara ini. Kempen-kempen kesihatan di negara ini turut tidak ketinggalan menggunakan platform ini sebagai salah satu agen penyebarluasan informasi kepada masyarakat. Tujuan kajian rintis ini dijalankan adalah untuk menilai impak penggunaan papan tanda sebagai medium promosi Kempen Penyalahgunaan Dadah. Reka bentuk kajian keratan rentas kuantitatif digunakan bagi kajian ini. Sejumlah 50 orang responden yang terdiri daripada belia berusia 18 hingga 29 tahun telah direkrut sebagai sampel kajian dari kawasan negeri Pulau Pinang. Hasil kajian rintis ini menunjukkan bahawa penggunaan papan tanda sebagai medium promosi bahaya penyalahgunaan dadah mempunyai keberkesanan kepada golongan belia. Penambahbaikan dari segi kreativiti persembahan kandungan dalam papan tanda boleh terus dijalankan bagi meningkatkan impak medium ini kepada masyarakat.

Abstract:

Abstract Background: Mechanisms of hereditary erythrocytosis have been elucidated recently. These include high oxygen affinity (HOA) hemoglobin (Hb) variants, bisphosphoglycerate mutase (BPGM) deficiency, abnormalities in the erythropoietin receptor (EPOR) and oxygen-sensing pathway (OSP) proteins prolyl hydroxylase domain-2 (PHD2), hypoxia-inducible factor-2 alpha subunit (HIF2A), and von Hippel-Lindau (VHL). We present our experience with these disorders. Methods: Evaluation of erythrocytosis patients' blood samples for HOA Hb variants by protein or DNA sequencing methods has been performed in our laboratory for over 30 years. Testing has included alkaline/acid electrophoresis, isoelectric focusing, capillary electrophoresis, high performance liquid chromatography, mass spectrometry and Sanger sequencing. BPGM deficiency was determined by enzyme activity assay and/or genetic testing. Since 2012 we have evaluated the EPOR, EGLN1 (PHD2), EPAS1(HIF2A), and VHL genes. Reflexive evaluations, which include testing for p50 and pertinent Sanger sequencing of HBB, HBA1, HBA2, EPOR (exon 8), EGLN1 (exons 1-5), EPAS1 (exon 12) and VHL, have been performed on a subset of cases. Genetic results were correlated with clinical and phenotypic data. Results: Retrospective database review identified 62 confirmed HOA Hb variants (48β, 14α) with clinical erythrocytosis (Table 1). Multiple Hb variants were silent on at least one protein method but showed decreased p50 values. One homozygous BPGM mutation case was identified. Of the 394 cases evaluated for EPOR and OSP abnormalities, 39 cases (10%) contained genetic alterations. Of these, 11 were known pathogenic mutations and 28 were novel alterations including 4 pathogenic, 12 likely pathogenic and 12 variants of unknown significance (VUS). Eighteen EGLN1, 10 EPAS1, and 7 EPOR alterations were detected, all heterozygous. Four VHL mutation cases were identified: 3 homozygous/compound heterozygous known mutations and one novel heterozygous VUS (p.R200Q). All EPOR mutations resulted in a premature stop codon. Most of the EPAS1 alterations were located near proline 531, although two predicted splice site disturbances were found. The EGLN1 alterations were novel, variable, and affect the enzyme's catalytic domain. All 4 VHL cases involved the R200 amino acid position in at least one allele. Serum Epo levels varied: decreased in EPOR (5/5); normal in PHD2 (11/11) and most HIF2A (6/7) and VHL (3/4) cases; increased in 1 HIF2A and 1 VHL case. p50 was normal in EPOR and OSP cases (31/31 tested). Conclusion: A spectrum of abnormalities was identified. Salient points include: 1) Diagnostically, a high index of suspicion for HOA Hb variants is required as they can give false negative results by routine protein detection methods. p50 and Epo levels were useful for triage. 2) Phenotypically, serum Epo levels were low in EPOR and normal in PHD2 and HIF2A cases. Unexpectedly, the two classic Chuvash polycythemia VHL cases were associated with normal Epo levels. 3) Mechanistically, the alterations were truncating in EPOR, variable and scattered in EGLN1 and clustered near proline 531 in EPAS1. VHL mutations were rare and consistently affected the R200 amino acid position in at least one allele. Novel alterations were frequent, especially in EGLN1. 4) Clinically, many patients were asymptomatic but a subset demonstrated recurrent headaches and one had chest pain. Keberadaan mesofauna tanah bergantung pada sumber energi seperti bahan organik dan biomassa hidup yang terkait dengan aliran siklus karbon di dalam tanah. Penelitian ini bertujuan untuk mengidentifikasi kelimpahan dan keragaman mesofauna tanah pada tiga tipe penggunaan lahan di Bogor, Jawa Barat. Pengambilan sampel tanah dilakukan pada 5 titik lokasi sampel pada tipe penggunaan lahan hutan, kebun, dan tegalan. Ekstraksi mesofauna tanah pada masing-masing tipe penggunaan lahan menggunakan metode berlese tulgren, kempson extractor , dan pitfall traps. Kelimpahan mesofauna tanah terbesar terdapat pada tipe penggunaan lahan tegalan yang diekstraksi menggunakan metode kempson sampel seresah dengan densitas 835 individu/m2. Indeks diversitas tertinggi meso fauna tanah terdapat pada tipe penggunaan lahan hutan.

Abstract:

We report the discovery of carbonates in the Planetary Nebulae NGC 6302 and NGC 6537 (Kemper et al. 2002). In the ISO LWS spectra far-infrared features have been identified with calcite and dolomite by comparison with laboratory spectra of these minerals. This is the first time that carbonates have been identified outside the solar system. In a follow-up study (Kemper et al., in prep.) a detailed analysis of the mineral composition of the dust in NGC 6302 is presented.

Abstract:

The 4-color theorem was proved by showing that a minimum counterexample cannot exist. Birkhoff demonstrated that a minimum counterexample must be internally 6-connected. We show that a minimum counterexample must also satisfy a coloring property that we call Kempe-locking. The novel idea explored in this article is that the connectivity and coloring properties are incompatible. We describe a methodology for analyzing whether an arbitrary planar triangulation is Kempe-locked. We provide a heuristic argument that a fundamental Kempe-locking configuration must be of low order and then perform a systematic search through isomorphism classes for such configurations. All Kempe-locked triangulations that we discovered have two features in common: (1) they are Kempe-locked with respect to only a single edge, say x y , and (2) they have a Birkhoff diamond with endpoints x and y as a subgraph. On the strength of our investigations, we formulate a plausible conjecture that the Birkhoff diamond is the only fundamental Kempe-locking configuration. If true, this would establish that the connectivity and coloring properties of a minimum counterexample are indeed incompatible. It would also imply the appealing conclusion that the Birkhoff diamond configuration alone is responsible for the 4-colorability of planar triangulations.

Declining state provision, low levels of financial capability (FSA/DTI, 2002), and, it must be added, regulation on savings and investments have impacted severely on the ability of some sections of the UK population to purchase life assurance and savings products. In addition, according to a report submitted to the Treasury, ‘there is a wide consensus that the UK population is not saving enough for retirement. Savings levels are 20 per cent or more below what they should be. The problem appears to be particularly acute amongst the less affluent, where insufficient levels of saving are likely to have a more serious impact’ (Sandler, 2002: 2). This is due in no small part to current banking and insurance practices that have excluded many of the least affluent customers from essential financial services (Kempson and Whyley, 1998; Kempson et al., 2000; NCC, 1997), that afford them the protection of:[bull ] Financial security for a rainy day;[bull ] Greater comfort during retirement and old age;[bull ] Access to greater independence and opportunity throughout their lives; (HM Treasury, 2001b)Ideally for less affluent consumers, product design should incorporate simplicity and transparency, preferably offer automatic savings of small amounts weekly or fortnightly and be flexible enough for payments to be suspended during times of financial hardship without incurring penalties (Kempson and Whyley, 1998: 50–51).

Dr Ir Johan Vanpaemel beschrijft in een artikel, getiteld 'De Industrialisatie van de Kempen' het ontstaan van metallurgische en chemische industrie tussen Lommel en Balen. De auteur laat zien, hoe in de tweede helft van de 19e eeuw talrijke sterk vervuilende industrieën werden overgebracht naar de dunbevolkte delen van de Noorderkempen. Ook daar rezen echter grote bezwaren tegen de enorme pollutie, waardoor de regen niet als water maar als zwavelzuur uit de hemel kwam vallen! Door ingrijpen van de nationale overheid waren de bedrijven gedwongen nieuwe technische procedé's te gaan toepassen, waardoor zij enorme hoeveelheden zwavelzuur overhielden. Dit product bleek evenwel uitermate geschikt als grondstof voor de vervaardiging van kunstmeststoffen. Ten gevolge van de grote agrarische crisis ontstond sedert 1880 een grote vraag juist naar deze producten. Dientengevolge ontwikkelden zich naast de zinkfabrieken in de Kempen chemische bedrijven die de derivaten van de zinkproductie verwerkten.

ABSTRACT The Concept of Kempel in Keprakan and Dhodhogan in Yogyakarta-style Menak Wooden Puppet Show. KKeprakan and dhodhogan are two of the accompaniments in Menak wooden puppet show in Yogyakarta. The term keprakan is derived from the essential word ‘keprak’ which refers to the principle of form, material, variety, and technique of creating sounds from the iron plates pounded by using cempala made from iron or wood. The term dhodhogan is taken from the root ‘dhog’ which refers to the sound produced by iron or wood that is pounded on kothak. The term kempel has meaning as ‘whole’ and ‘blend into one’. Further, in the context of keprakan, its meanings is that the sound created is harmonically integrated with the movement of the puppet and the motif of kendhangan. This study aims to disclose the pattern of keprakan and dhodhogan to produce a sense of kempel in supporting the aesthetic expression of puppet characters and the scenes’ ambiences. The author collected the data through direct participation, in-depth interviews, and observations of recordings of Menak puppet show in which Sukarno as the puppeteer. The data analysis was conducted to draw a conclusion as a result of an investigation of the relationship among the pattern of both keprakan and dhodhogan; the movement diversity of puppet characters; and the motif of kendhangan. According to the results, it can be stated that the design of keprakan and dhodhogan in Menak wooden puppet show in Yogyakarta consists of two styles – the one is free style and the other is bound style. The sense of kempel lies in the accuracy of keprakan and dhodhogan diverse sound combinations in relation to the movement varieties of puppet figures incorporated with the motif of kendhangan.ABSTRAK Keprakan dan dhodhogan adalah salah satu bagian dari iringan pergelaran wayang golek Menak gaya Yogyakarta. Istilah keprakan dari kata dasar keprak menunjuk pada persoalan bentuk, bahan, ragam, serta teknik menghasilkan bunyi dari lempengan besi yang dipukul menggunakan cempala berbahan besi maupun kayu. Istilah dhodhogan diambil dari kata dasar dhog yang menunjuk pada bunyi yang dihasilkan cempala besi atau kayu yang dipukul pada kothak. Kosa kata kempel mempunyai makna utuh dan melebur menjadi satu, dalam konteks keprakan maknanya bahwa bunyi yang dihasilkan menyatu dengan gerak wayang dan motif kendhangan. Penelitian ini bertujuan untuk mengungkap pola keprakan dan dhodhogan untuk menghasilkan rasa kempel dalam mendukung ekspresi estetis tokoh wayang maupun suasana adegan. Data diperoleh melalui partisipasi terlibat, wawancara mendalam, serta pengamatan terhadap rekaman pergelaran wayang golek Menak dengan dalang Ki Sukarno. Analisis dilakukan untuk mendapatkan simpulan berdasarkan telaah terhadap relasi antara pola keprakan dan dhodhogan dengan ragam gerak tokoh wayang serta motif atau pola kendhangan. Berdasarkan hasil penelitian dapat dijelaskan bahwa pola keprakan dan dhodhogan dalam pergelaran wayang golek Menak gaya Yogyakarta terdiri dari dua ragam, yaitu ragam bebas dan ragam berpola. Rasa kempel terletak pada ketepatan dalam memadukan ragam bunyi keprakan dan dhodhogan sesuai dengan ragam gerak tokoh wayang yang menyatu dengan motif kendhangan.

In collecting data the techniques used were through observation, interviews, and triangulation with data analysis using idea theorizing or conceptualization of ideas. qard on sharia microfinance carried out by the KHAS Micro Waqf Bank, plays a role in the increase in the number of sales of operating income, operating profit, and economic conditions. Most of the implementation of the qard contract in BWM KHAS Kempek according to the conclusion of the researcher is fully in accordance with the corridors that are theorized in Islamic law.
