"Direktiva 2003/49/EZ o zajedničkom sustavu oporezivanja isplate kamata i autorskih naknada između povezanih trgovačkih društava različitih država članica - analiza Direktive i hrvatskog prijevoda nekih termina iz Direktive." Abstract:
Službeni prijevod direktiva EU-a važan je, prvo, zbog njihove implementacije u pravne sustave država članica, a zatim i zbog mogućnosti izravne primjene direktiva pod određenim uvjetima.

Abstract:
Abstract Three novel splice site mutations and two novel missense mutations were identified by molecular analysis of pyruvate kinase (PK) deficiency associated with hereditary nonspherocytic hemolytic anemia. A Nepalese PK variant, PK Kowloon, was found to have a homozygous transversion at the 5′-splice site of the seventh intervening sequence (IVS) of the L-type PK gene (Ivs7[+1]gt → tt). Using a reverse transcription polymerase chain reaction (RT-PCR) assay, we showed that the R-type PK mRNA in the proband's reticulocytes included the seventh IVS between the seventh and eighth exon, introducing a stop codon 3 nucleotides downstream of the mutated site. Consequently, the translational product may lack 44% of the R-PK polypeptide. A transition at the last nucleotide of exon 9 (1269GCG → GCA) was found in a Japanese PK variant, PK ‘Kamata.’ The mutation did not alter the amino acid sequence, but caused skipping of the ninth exonic sequence in the R-PK transcripts. As a result, the affected R-type PK lost 51 amino acid residues (373Met-423Ala del). A transversion at the splice acceptor site of the third IVS (Ivs 3[-2]ag → tg) was identified in PK ‘Aomori.’ The mutation resulted in aberrant splicing at a cryptic splice site within exon 4, causing deletion of two codons in the aberrant R-PK transcript (95 Gly-96 Pro → del). Both PK ‘Kamata’ and PK ‘Aomori’ had a missense mutation on the other allele, 1044AAG → AAT (348Lys → Asn) and 1075CGC → TGC (359Arg → Cys), respectively. Although both 348Lys and 359Arg were located in the sixth loop of A domain (β/α)8 barrel, which has been shown to contain the substrate and cation binding sites, the degree of anemia was much more severe in PK ‘Kamata’ than PK ‘Aomori,’ possibly because the 51 amino acid deletion of PK ‘Kamata’ but the 2 amino-acid deletion of PK ‘Aomori’ may abolish PK catalytic activity.

Abstract:
Islam is the entire system of life. Through the Qur'an, the Almighty Allah gave instructions to humanity on how to live in this transitory world. As not a single segment of human life is left out of Allah's Code, so the issue of work, sustenance and trade is not left out either. It is in nature, but also in man's need, to earn and save in order to feed himself and his family. Certainly, one thing should be kept in mind, and that is how he acquires that income and wealth. There are many who reach for various illegal things, such as fraud, various speculations and engaging in easy ways of earning, which is especially visible in the modern age, and that is in terms of usury or interest, as the work itself is called. The main goal of the work is actually to point out the perniciousness and harmfulness and the bad consequences of that society or individuals who are encouraged to engage in interest business. First, that Allah, the Exalted, has strictly and categorically forbidden it, and secondly, that man brings misfortune and misery upon himself if he does the opposite of what he was ordered to do. Therefore, if we want good for ourselves, our family, society and humanity -let us leave what Almighty Allah has ordered us to leave, seek halal earnings, let us be among those who will enjoin good and forbid evil, let us be good and sincere believers because that is the only way we will deserve the Lord's mercy and forgiveness.

Full text
Abstract:
We previously reported that in mesenteric arteries from streptozotocin (STZ)-induced diabetic rats that 1) endothelium-derived hyperpolarizing factor (EDHF)-type relaxation is impaired, possibly due to a reduced action of cAMP via increased phosphodiesterase 3 (PDE3) activity (Matsumoto T, Kobayashi T, and Kamata K. Am J Physiol Heart Circ Physiol 285: H283–H291, 2003) and that 2) PKA activity is decreased (Matsumoto T, Wakabayashi K, Kobayashi T, and Kamata K. Am J Physiol Heart Circ Physiol 287: H1064–H1071, 2004). Here we investigated whether chronic treatment with cilostazol, a PDE3 inhibitor, improves EDHF-type relaxation in mesenteric arteries isolated from STZ rats. We found that in such arteries 1) cilostazol treatment (2 wk) improved ACh-, A-23187-, and cyclopiazonic acid-induced EDHF-type relaxations; 2) the ACh-induced cAMP accumulation was transient and sustained in arteries from cilostazol-treated STZ rats; 3) the EDHF-type relaxation was significantly decreased by a PKA inhibitor in the cilostazol-treated group, but not in the cilostazol-untreated group; 4) cilostazol treatment improved both the relaxations induced by cAMP analogs and the PKA activity level; and 5) PKA catalytic subunit (Cat-α) protein was significantly decreased, but the regulatory subunit RII-β was increased (and the latter effect was significantly decreased by cilostazol treatment). These results strongly suggest that cilostazol improves EDHF-type relaxations in STZ rats via an increase in cAMP and PKA signaling.

Abstract:
Current energy issues have prompted demand for the innovations in constructing a clean and sustainable energy platforms. One of examples for energy carrier to actualize a zero carbon society is hydrogen, which is a promising carbon-free fuel with an excellent weight energy density. Hydrogen production through alkaline water electrolysis using renewable energy resources (light and wind)-derived electricity is an attractive ecofriendly and cost-effective technology because cheap nonprecious electrocatalysts can be employed in alkaline media. However, oxygen evolution reaction (OER) at the anode side in the water electrolysis is basically slow and prevents the widespread applications of water electrolysis. Thus, highly efficient and cheap electrocatalysts for OER are highly desirable. Iron (Fe)-based compounds have recently received much attention as OER electrocatalysts because Fe is a quite earth-abundant metallic element that is a very cheap and non-toxic. Therefore, Fe is a promising candidate as electrocatalyst for OER. Due to the insufficient OER activity on simple Fe oxides, previous studies have attempted to enhance the activity by incorporating Fe and other metallic elements.[1,2] Recently, we established a novel comprehensive structural descriptor for the OER on Fe-based simple and multimetal oxides;[3] i.e., shorter minimum Fe–O bond length in the crystalline structures led to higher OER activity regardless of elemental composition, Fe–O coordination number, and crystal category. Thus, this descriptor enabled us to quickly develop excellent OER catalyst, Ba0.65Ca0.35Fe12O19, comprising extremely short Fe–O bond lengths.[4] Furthermore, we found that post-spinel CaFe2O4 catalyzes OER via "multi-iron-site mechanism" involving a reaction intermediate with O–O direct formation on multiple Fe atoms, and CaFe2O4 consequently displayed outstanding OER activity despite the long Fe–O bond length.[5] However, we speculate that OER activity of Fe-based oxides cannot be further enhanced based on crystal structures, and it necessitates to employ another type of compounds. In this study, we report the OER performances of Fe-based phosphates in alkaline media. We selected three kinds of Fe-based phosphates with distinct crystal structures: trigonal FePO4, trigonal Fe3O3(PO4), and monoclinic Fe4(P2O7)3. The electrochemical measurements and post-characterizations identified the Fe-based phosphates as efficient precatalysts that can be electrochemically converted into highly active Fe-based compounds. Notably, the OER performances of the electrochemically converted Fe-based compounds overcame those of the above-mentioned most active Fe-based multimetal oxides and any previously reported crystalline OER electrocatalysts. Therefore, the Fe-based phosphates are promising candidates as OER precatalysts, which can be in situ converted into excellent OER electrocatalysts, and have further advantages of low price, environmental friendliness, and easy production.[6] The part of this paper is based on results obtained from a project, JPNP14021, commissioned by the New Energy and Industrial Technology Development Organization (NEDO).

An exploratory investigation is reported to test the utility of Kamata, Tenenbaum, and Hanin’s (2002) probabilistic model in determining individual affect-related performance zones (IAPZs) in a simulated car-racing task. Three males completed five separate time-trials of a simulated racing task by which self-reported affective states (i.e., arousal and pleasure) and physiological measures of arousal (i.e., heart rate and skin conductance) were integrated with performance and measured throughout each trial. Results revealed each performer maintained unique IAPZs for each of the perceived and physiological measures in terms of the probability and range of achieving each zone. The practical applications of this approach are discussed.

An Electromagnetic shower in rock salt is generated by a computer code of Geant4.5. A modified Greisen parameterization in longitudinal distribution of the shower and a modified Nishimura, Kamata, and Greisen function in lateral distribution of the shower (structure function) are fitted to a space distribution of the excess electrons. Abstract:
A recently introduced probabilistic methodology (Kamata, Tenenbaum, & Hanin, 2002) was implemented in the current study to ascertain the idiosyncratic Individual Affect-related Performance Zones (IAPZs) of four intercollegiate tennis players. The current study advances upon previous empirical works by its use of multiple performance levels, use of athletes’ introspective affective intensity, and recording multiple data points duringcompetition. Results present within- and between-player comparisons, and highlight the dynamic nature of competitive athletic events. A brief discussion regarding the implications of this methodology and the pursuant results for sport psychology consultants is also proffered. Being idiosyncratic in nature, the observations from this study are not intended to generalize across samples, but rather to introduce how knowledge of the systematic and dynamic linkage between an individual’s affect and his or her performance can be uncovered and possibly used with individual athletes to facilitate more consistently optimal performances.

Abstract:
On the occasion of the 25th anniversary of his death in 2019, Netflix released two limited series focused on Mexican presidential candidate Luis Donaldo Colosio and his assassination. 1994 (directed by Diego Enrique Osorno) and Crime Diaries: The Candidate (Historia de un crimen: Colosio, directed by Hiromi Kamata and Natalia Beristáin) are, respectively, a documentary series and a drama, both delving into the political conjuncture surrounding both Colosio’s candidacy and the investigations subsequent to his assassination. Since his dramatic death, the image of Colosio has maintained its associations with heroism and his unrealized presidency has come to embody the “México que no fue,” a radically innovated Mexican state that never was and, possibly, will never be. Both series rely on the exploitation of original footage and underlying political takes on the events. This analysis will critically compare the two series from three main thematic perspectives: the Mexican political context at the time of the assassination; Colosio’s characterization and private life; and the procedural aspects of the investigations.

A new hybrid experiment (YAC-II+Tibet-III+MD) located at Yangbajing has started to improve the capability of explicit measurements of the cosmic-ray components (P, He, CNO, Fe, etc) at the knee energy region since 2014. Considering the different features of air shower development for different primary cosmic-ray nuclei, using a full Monte Carlo simulation, we modified the lateral distribution functions for proton-induced, helium-induced and iron-induced air showers, respectively. The results show that the air shower size of different nuclei obtained by the modified Nishimura-Kamata-Greisen function is consistent with the true shower size within 5% systematic errors. Furthermore, we studied the Extensive Air Shower age parameter determination from the lateral distribution functions of charged particles near the air shower core with the Tibet hybrid experiment. The derived age parameter can be well used to estimate the particle type of the incident primary cosmic rays.

Abstract:
Abstract In this work, we show the results of a comparative analysis of different reconstruction methods of shower cores produced by Cosmic Rays (CR’s) in a layer of Resistive Plate Chambers (RPC’s) that is under consideration for MATHUSLA (Massive Timing Hodoscope for Ultra-Stable neutraL pArticles). The analysis was performed with Monte Carlo simulations. Extensive air showers were simulated with CORSIKA using the GEISHA and QGSJET-II-04 hadronic interaction models for primary protons and iron nuclei in the energy range from 10 TeV to 100 PeV and zenith angles: θ ≤ 20°. The detector response was simulated with a simplified Python-based program. The location of the shower core was found using the center of mass of the deposited charge distribution, the maximum of the charge distribution, a fit with an exponential function to the projected distributions on the X and Y horizontal directions of the 2D charge distributions, and fits with an exponential and a Nishimura-Kamata-Greisen (NKG) lateral functions to the 3D charge distributions of the RPC. We found that the fits of the X and Y projections with an exponential function and the fits with a NKG function to the 3D signal distributions have a better performance than the other methods.

Abstract:
Present energy issues have increasingly stimulated the demand for the development of clean and sustainable energy platforms which can achieve net zero carbon emissions. Renewable energy resources are quite promising candidates for the construction of sustainable decarbonized society. One of examples of harnessing renewable energy-derived electricity is the electrolysis of earth-abundant water into hydrogen, which is a prominent carbon-free chemical fuel with a high weight energy density. Compared with polymer electrolyte water electrolysis in acidic environment, electrochemical alkaline water splitting can employ inexpensive nonprecious metals as electrocatalysts, which can make this technology a large-scale and cost-effective process. However, the anodic oxygen evolution reaction (OER) in the water splitting has a large overpotential, and it is a great bottleneck for widespread use. Thus, highly active and inexpensive electrocatalysts for OER are desired to overcome this concern. Iron (Fe) is a very earth-abundant element that is a quite cheap and non-toxic metal, and thus it is a potential candidate as electrocatalyst for OER. Previous studies have reported several multimetal oxides with Fe and other metallic elements, such as perovskite and brownmillerite types, which have attracted great attentions due to their facile synthesis and easily tunable OER activities by element substitution and doping. However, most of these studies focused on the choice of elements. Contrarily, the effects of crystal structures on OER activity have not yet been sufficiently interpreted, and it can be a crucial aspect to understand the OER electrocatalysis on Fe-based oxides and for the rational design of outstanding OER catalysts. In this study, we aim to comprehensively understand the effects of crystal structures on electrocatalytic OER activities on Fe-based oxides. To obtain versatile insights into OER electrocatalysis, we collected OER performances of Fe-based simple and bimetal oxides from our experiments and literature, and their structural characteristics were obtained from databases for inorganic compounds. Thus, we found that OER efficiencies on the Fe-based oxides were correlated with Fe–O bond lengths in their crystals; i.e., a shorter Fe–O bond length led to a higher OER activity.[1] We then exploited databases to mine potential candidates based on the structure–activity relationship and selected unreported Fe-based bimetal and trimetal oxides with various structures, which can dramatically accelerate our catalyst research. We synthesized these Fe-based multimetal oxides and evaluated their OER activity in alkaline media; remarkably, the OER efficiency on the unreported Fe-based multimetal oxides also exhibited excellent correlation with Fe–O bond lengths, as plotted in Figure 1. It is notable that the trend in the Fe–O bond length and OER activity is applicable to a wide variety of Fe-based simple, bimetal, and trimetal oxides regardless of chemical compositions, crystal categories, and Fe valence states. Furthermore, machine learning analysis proved that Fe–O bond length is the most dominant structural descriptor for OER efficiency compared with other structural parameters.[2] However, it is required to overcome the abovementioned catalytic trend because theoretical Fe–O bond length in Fe-based oxides is limited, and superior OER catalysts cannot be achieved according to this design guideline. Hence, we attempted to switch the OER electrocatalytic mechanism on a Fe-based oxide by the arrangement of crystal structure because different mechanisms should be involved in different descriptors; i.e., the mechanism switch could surpass the abovementioned catalytic trend. Thus, we found post-spinel CaFe2O4 that comprises a large amount of edge-shared FeO6 connectivity in its crystal. DFT calculations unveiled that the structural characteristics induced an unprecedented OER mechanism, multi-iron-site mechanism, where OER process goes through a reaction intermediate with an O–O bridge on multiple Fe sites as depicted in upper side in Figure 1. Consequently, CaFe2O4 exhibited prominent OER activity despite long Fe–O bond length in its crystal.[3] These findings provide a novel insight into the OER electrocatalysis on Fe-based oxides and contribute to the innovation of electrochemical hydrogen production. Abstract:
The Koch Dynasty occupies an important place in the history of Assam. There were many great warriors of history who were associated with this dynasty. Viśva Siṁha was the first king of the Koch Dynasty. Viśva Siṁha had eighteen sons and Chilārāi was the third son. After Viśva Siṁha, the Koch Dynasty was ruled by King Naranārāyaṇa. Chilārāi was appointed as the commander-in-chief during the reign of Naranārāyaṇa. Being a commander-in-chief, Chilārāi brought various states under the control of the Kamata-Koch power. Chilārāi was very popular for his military exploits during the reign of Naranārāyaṇa. The name of Chilārāi during his birth was Śukladhvaja. But later on, the name chilārāi was given to him due to his traits. It was said that Śukladhvaja got the name chilārāi because of the swift mobility of his army and sudden and surprising attack. There was another tradition regarding the name chilārāi. History said that when Śukladhvaja crosses the Bhoroli River in one jump, it appears like a hawk is pouncing on its prey. Thus he got the name chilārāi. In the history, Chilārāi was always remembered for his prowess. But people hardly know about the connection and contribution of Chilārāi in the field of literature. Thus the present seminar paper tries to bring out the connection of Chilārāi with the contemporary literary world of Assam. Significance of p27 as a predicting marker for medroxyprogesterone acetate therapy against endometrial endometrioid adenocarcinoma. Int J Gynecol Cancer 2006;16(Suppl. 1): 452–457.We reported that p27 induced by medroxyprogesterone acetate (MPA) may be involved in the progestin-induced growth suppression of human endometrial adenocarcinoma cells. This study aimed at investigating whether p27 expression could be a predicting marker to evaluate the effectiveness of MPA therapy. The clinical responses of 15 patients with endometrial carcinoma treated with MPA were examined. p27 expression was evaluated by immunohistochemical staining. Percentage of positive nuclear staining was expressed as a strongly positive (SP) labeling index (LI). Before MPA treatment, SP LIs in the effective and noneffective groups were 22.6 ± 14.3% and 9.1 ± 9.2%. At 1–6 weeks in the MPA treatment, SP LIs increased in both groups and were significantly higher than those before the therapy. At 7–12 weeks, SP LIs in both groups decreased to the level of pretherapy. At 13–18 weeks, SP LIs in the effective group were 14.9 ± 5.7%, whereas in the noneffective group, 1.1 ± 2.0%. The former was significantly higher than the latter. p27 expression could predict the effectiveness of MPA treatment for endometrial carcinoma at an early stage of the 4-month period in MPA therapy and could be a useful predicting marker for MPA.

Abstract:
Using superior mesenteric artery rings isolated from age-matched controls and streptozotocin (STZ)-induced diabetic rats, we recently demonstrated that EDHF-type relaxation is impaired in STZ-induced diabetic rats, possibly due to a reduced action of cAMP via increased phosphodiesterase (PDE) activity (Matsumoto T, Kobayashi T, and Kamata K. Am J Physiol Heart Circ Physiol 285: H283–H291, 2003). Here, we investigated the activity and expression of cAMP-dependent protein kinase (PKA), an enzyme that is produced by a pleiotropic and plays key roles in the transduction of many external signals through the cAMP second messenger pathway and in cAMP-mediated vasorelaxation. The relaxation induced by cilostamide, a selective PDE3 inhibitor, was significantly weaker in superior mesenteric artery rings from STZ-induced diabetic rats than in those from age-matched controls. The relaxation responses to 8-bromo-cAMP (8Br-cAMP) and N6, O2-dibutyryl-adenosine-cAMP (db-cAMP), a cell-permeant cAMP analog, were also impaired in the STZ diabetic group. PKA activity in the db-cAMP-treated mesenteric artery was significantly lower in the STZ diabetic group. The expression levels of the mRNA and protein for PKA catalytic subunit Cat-α were significantly decreased in the STZ diabetic group, but those for PKA regulatory subunit isoform RII-β were increased. We conclude that the abnormal vascular relaxation responsiveness seen in STZ-induced diabetic rats may be attributable not only to increased PDE activity but also to decreased PKA activity. Possibly, the decreased PKA activity may result from an inbalance between PKA catalytic and regulatory subunit expressions.

Abstract:
The enzyme GK (glucokinase), which phosphorylates glucose to form glucose 6-phosphate, serves as the glucose sensor of insulin-producing β-cells. GK has thermodynamic, kinetic, regulatory and molecular genetic characteristics that are ideal for its glucose sensor function and allow it to control glycolytic flux of the β-cells as indicated by control-, elasticity- and response-coefficients close to or larger than 1.0. GK operates in tandem with the K+ and Ca2+ channels of the β-cell membrane, resulting in a threshold for glucose-stimulated insulin release of approx. 5 mM, which is the set point of glucose homoeostasis for most laboratory animals and humans. Point mutations of GK cause ‘glucokinase disease’ in humans, which includes hypo- and hyper-glycaemia syndromes resulting from activating or inactivating mutations respectively. GK is allosterically activated by pharmacological agents (called GK activators), which lower blood glucose in normal animals and animal models of T2DM. On the basis of crystallographic studies that identified a ligand-free ‘super-open’ and a liganded closed structure of GK [Grimsby, Sarabu, Corbett and others (2003) Science 301, 370–373; Kamata, Mitsuya, Nishimura, Eiki and Nagata (2004) Structure 12, 429–438], on thermostability studies using glucose or mannoheptulose as ligands and studies showing that mannoheptulose alone or combined with GK activators induces expression of GK in pancreatic islets and partially preserves insulin secretory competency, a new hypothesis was developed that GK may function as a metabolic switch per se without involvement of enhanced glucose metabolism. Current research has the goal to find molecular targets of this putative ‘GK-switch’. The case of GK research illustrates how basic science may culminate in therapeutic advances of human medicine.

Abstract:
We previously reported that in mesenteric arteries from aged Otsuka Long-Evans Tokushima fatty (OLETF) rats (a type 2 diabetes model) endothelium-derived hyperpolarizing factor (EDHF)-type relaxation is impaired while endothelium-derived contracting factor (EDCF)-mediated contraction is enhanced (Matsumoto T, Kakami M, Noguchi E, Kobayashi T, Kamata K. Am J Physiol Heart Circ Physiol 293: H1480–H1490, 2007). Here we investigated whether acute and/or chronic treatment with metformin might improve this imbalance between the effects of the above endothelium-derived factors in mesenteric arteries isolated from OLETF rats. In acute studies on OLETF mesenteric arteries, ACh-induced relaxation was impaired and the relaxation became weaker at high ACh concentrations. Both metformin and 5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside [AICAR, an AMP-activated protein kinase (AMPK) activator that is also activated by metformin] 1) diminished the tendency for the relaxation to reverse at high ACh concentrations and 2) suppressed both ACh-induced EDCF-mediated contraction and ACh-stimulated production of prostanoids (thromboxane A2 and PGE2). In studies on OLETF arteries from chronically treated animals, metformin treatment (300 mg·kg−1·day−1 for 4 wk) 1) improved ACh-induced nitric oxide- or EDHF-mediated relaxation and cyclooxygenase (COX)-mediated contraction, 2) reduced EDCF-mediated contraction, 3) suppressed production of prostanoids, and 4) reduced superoxide generation. Metformin did not alter the protein expressions of endothelial nitric oxide synthase (eNOS), phospho-eNOS (Ser1177), or COX-1, but it increased COX-2 protein. These results suggest that metformin improves endothelial functions in OLETF mesenteric arteries by suppressing vasoconstrictor prostanoids and by reducing oxidative stress. Our data suggest that within the timescale studied here, metformin improves endothelial function through this direct mechanism, rather than by improving metabolic abnormalities.

Abstract:
Ayurveda considers Jaundice (Kamala) as a disorder of Raktavahasrotas. Vitiated Pitta is the main causa-tive factor in the pathogenesis of Kamala. In Ayurveda, Kamala is a next step of Pandu (anaemia). Ac-cording to Acharya Charaka, Kamala is mainly caused due to frequently consumption of Ushna (hot), Tik-shna (spicy) food i.e. Pittaprakopak Ahara (increasing level of Pitta). Due to these factors vitiation of Pitta (bile), Rakta (blood) and Mamsa (muscles) Dhatu will occur. Kamala is clinically correlated with jaundice due to its similarity of symptoms like yellowish of skin (Twak pitata), sclera (Netra), urine (Mutra), stool (Purisha) etc. In Kamala Vyadhi all Acharyas in Ayurveda texts has explained about Virechana Karma. Modern science has its own limitations in treating Kamala Vyadhi (jaundice) but Ayurvedic literature clearly explained pathology and treatment of Kamala Vyadhi which shows the specificity of Ayurveda. Ayurvedic line of management i.e., Virechana therapy and Shamana Chikitsa are successful in Kamala by removing toxic waste from the body and by correction of Agni (digestive fire).

Abstract:

Abstract:
Abstract Gastric cancer (GC) in the fifth-leading types of cancer and the third-leading cause of death from cancer. Characterization of tumor microenvironment (TME) in GC is important not only to comprehensively understand tumor biology in GC but also to develop a novel cancer therapeutics targeting TME. In this study, we conducted multi-omics analyses of single-cell RNA sequencing (scRNA-seq), bulk RNA sequencing (bulk RNA-seq), and whole exome sequencing (WES) using biopsied tumor samples through endoscopy from 23 GC patients: 3 treatment-naïve patients, 7 patients in treatment with chemotherapy (Chemo), 5 patients in combination treatment with chemotherapy and immune checkpoint blockade (Chemo+CPI), 3 patients in combination treatment with chemotherapy and Her2-antibody targeted therapy (Chemo+Her2i), and 5 patients in third-line or later treatment. We initially classified the GC samples into 4 molecular subtypes (CIN, GS, MSI, and EBV) using datasets of WES, bulk RNA-seq and scRNA-seq, showing that our samples were mainly composed of CIN (n=7) and GS (n=16) types. In scRNA-seq, we revealed that drug treatments profoundly modulated not only proportions of TME components but also their molecular characteristics. For instance, Chemo+CPI treated patients showed higher proportion of CD8+ cytotoxic T cells with activation and/or exhaustion markers. Combination treatments with Chemo+Her2i, in addition, resulted in higher Trail and Estrogen signaling pathways in epithelial cells of TME components. These findings suggest that interventions with certain cancer therapeutic drugs could substantially convert transcriptome landscape and reconstitute GC TME components. High-resolution of GC TME analyses should provide a clue for developing a next generation of cancer therapeutic drugs. Abstract:
INTRODUCTION: Kamala is both raktapradoshaja vyadhi and pittaja nanatmaja. According to Charakacharya, kamala is a more advanced level of panduroga. While Vagbhatacharya described kamala as a distinct ailment, Sushrutacharya considered it a separate disease that might be caused by a further panduroga problem. In con-temporary medicine, kamala is associated with jaundice. Vyadhi Aacharyas have described Virechana Karma in Kamala. The limitations of modern science in treating Kamala Vyadhi (jaundice) are evident in the Ayurvedic literature, which elucidates the etiology and therapy of the condition and highlights the peculiarity of Ayurveda. The majority of individuals have grown accustomed to spicy fast food over time. Humanity has been separated from nature by this. The intake of alcohol is likewise rising daily in the modern day. Hepatocellular jaundice is referred to in Ayurvedic writings as Kamala. "Kamali tu virechanama" has also been cited by ancient Acharyas as the Chikitsa Sutra for Kamala. AIMS AND OBJECTIVES To find out the effect of Ayurvedic medicine in the management of Kamala (Jaundice) managed with Shaman chikitsa. MATERIAL AND METHODES A 15yr. Old girl patient came with the complaint of Anorexia, Urine -Pale yellow, Ab-dominal pain after taking of food, Face yellow, and yellowish coloration of eyes and nails. The patient has taken medicine, but no relief was found, so she came to us for Ayurveda treatment. RESULT AND DISCUSSION The outcomes were noteworthy Regarding the clinical and analytical criteria. According to statistics, there is improvement in the yellowing of the urine and eyes, weakness, appetite loss, and serum bilirubin levels. 15-year-old girl came for Ayurvedic treatment for Kamala.
