Abstract:
ABSTRACT A variety of serovars of the food-borne pathogen Vibrio parahaemolyticus normally cause infection. Since 1996, the O3:K6 strains of this pathogen have caused pandemics in many Asian countries, including Taiwan. For a better understanding of these pandemic strains, the recently isolated clinical O3:K6 strains from India, Japan, Korea, and Taiwan were examined in terms of pulsed-field gel electrophoresis (PFGE) typing and other biological characteristics. After PFGE and cluster analysis, all the O3:K6 strains were grouped into two unrelated groups. The recently isolated O3:K6 strains were all in one group, consisting of eight closely related patterns, with I1(81%) and I5(13%) being the most frequent patterns. Pattern I1 was the major one for strains from Japan, Korea, and Taiwan. All recently isolated O3:K6 strains carried the thermostable direct hemolysin (tdh) gene. No significant difference was observed between recently isolated O3:K6 strains and either non-O3:K6 reference strains or old O3:K6 strains isolated before 1996 with respect to antibiotic susceptibility, the level of thermostable direct hemolysin, and the susceptibility to environmental stresses. Results in this study confirmed that the recently isolated O3:K6 strains of V. parahaemolyticus are genetically close to each other, while the other biological traits examined were usually strain dependent, and no unique trait was found in the recently isolated O3:K6 strains.

ABSTRACT Historically, Vibrio parahaemolyticus infections have been characterized by sporadic cases caused by multiple, diverse serotypes. However, since 1996, V. parahaemolyticus serotype O3:K6 strains have been associated with several large-scale outbreaks of illness, suggesting the emergence of a “new” group of organisms with enhanced virulence. We have applied three different molecular subtyping techniques to identify an appropriate method for differentiating O3:K6 isolates from other serotypes. Pulsed-field gel electrophoresis (PFGE) following NotI digestion differentiated seven closely related subtypes among O3:K6 and related strains, which were distinct from PFGE patterns for non-O3:K6 isolates. Ribotyping and tdh sequencing were less discriminatory than PFGE, but further confirmed close genetic relationships among recent O3:K6 isolates. In vitro adherence and cytotoxicity studies with human epithelial cells showed that O3:K6 isolates exhibited statistically higher levels of adherence and cytotoxicity to host cells than non-O3:K6 isolates. Epithelial cell cytotoxicity patterns were determined with a lactate dehydrogenase release assay. At 3 h postinfection, high relative cytotoxicities (>50% maximum lactate dehydrogenase activity) were found among a greater proportion of recently isolated O3:K6 and closely related strains (75%) than among the non-O3:K6 isolates (23%). A statistically significant relationship between adherence and cytotoxicity suggests that the pathogenic potential of some isolates may be associated with increased adherence to epithelial cells. Our findings suggest that enhanced adherence and cytotoxicity may contribute to the apparent unique pathogenic potential of V. parahaemolyticus O3:K6 strains.

Abstract:
Barrier-to-autointegration factor (BAF) is a highly conserved protein in metazoans that has multiple functions during the cell cycle. We found that BAF is SUMOylated at K6, and that this modification is essential for its nuclear localization and function, including nuclear integrity maintenance and DNA replication. K6-linked SUMOylation of BAF promotes binding and interaction with lamin A/C to regulate nuclear integrity. K6-linked SUMOylation of BAF also supports BAF binding to DNA and proliferating cell nuclear antigen and regulates DNA replication. SENP1 and SENP2 catalyze the de-SUMOylation of BAF at K6. Disrupting the SUMOylation and de-SUMOylation cycle of BAF at K6 not only disturbs nuclear integrity, but also induces DNA replication failure. Taken together, our findings demonstrate that SUMOylation at K6 is an important regulatory mechanism that governs the nuclear functions of BAF in mammalian cells.

Vibrio parahaemolyticus O3:K6 strains responsible for the increase in the number of cases of diarrhea in Calcutta, India, beginning in February 1996 and those isolated from Southeast Asian travelers beginning in 1995 were shown to belong to a unique clone characterized by possession of the tdh gene but not thetrh gene and by unique arbitrarily primed PCR (AP-PCR) profiles (J. Okuda, M. Ishibashi, E. Hayakawa, T. Nishino, Y. Takeda, A. K. Mukhopadhyay, S. Garg, S. K. Bhattacharya, G. B. Nair, and M. Nishibuchi, J. Clin. Microbiol. 35:3150–3155, 1997). Evidence supporting a hypothesis that this clone emerged only recently and is spreading to many countries was obtained in this study. Of 227 strains isolated in a hospital in Bangladesh between 1977 and 1998, only 22 strains isolated between 1996 and 1998 belonged to the new O3:K6 clone (defined by the serovar, the tdh andtrh typing, and AP-PCR profiles). The O3:K6 strains isolated from clinical sources in Taiwan, Laos, Japan, Thailand, Korea, and the United States between 1997 and 1998 were also shown to belong to the new O3:K6 clone. The clonality of the new O3:K6 strains was also confirmed by analysis of the toxRS sequence, which has been shown to be useful for phylogenetic analysis of the members of the genus Vibrio. The toxRS sequences of the representative strains of the new O3:K6 clone differed from those of the O3:K6 strains isolated before 1995 at least at 7 base positions within a 1,346-bp region. A new PCR method targeted to 2 of the base positions unique to the new O3:K6 clone was developed. This PCR method could clearly differentiate all 172 strains belonging to the new O3:K6 clone from other O3:K6 strains isolated earlier. One hundred sixty-six strains belonging to 28 serovars other than O3:K6 were also examined by the new PCR method. The tdh-positive andtrh-lacking strains that belonged to the O4:K68 and O1:K untypeable serovars and were isolated in three countries and from international travelers beginning in 1997 gave positive results. The AP-PCR profiles of these strains were nearly identical to those of the new O3:K6 clone, and their toxRS sequences were 100% identical to that of the new O3:K6 clone. The results suggest that these strains may have diverged from the new O3:K6 clone by alteration of the O:K antigens. In conclusion, this study presents strong evidence for the first pandemicity in the history of V. parahaemolyticus and reports a novel toxRS-targeted PCR method that will be useful in epidemiological investigation of the cases associated with the current pandemic spread.

Abstract Keratin intermediate filament is one major cytoskeleton architecture in epithelial cells, providing necessary mechanical support to maintain cell shape and tissue integrity. In a previous study, we discovered the existence of a novel K6-K14 keratin fusion variant (K6-K14/V7) in oral squamous cell carcinoma (OSCC) cells which can promote cancer stemness and aggressiveness. However, the underlying mechanism remains to be elucidated. Here, we report that overexpression of K6-K14/V7 in OSCC cells can influence the mechanical properties of the cells, leading to cell protrusion and enhanced extracellular vesicle (EV) secretion. Consequently, the expression levels of inflammatory cytokines (e.g. IL-6, TGF-[[Unsupported Character - Symbol Font β]]) in the culture medium can be also increased. Normal oral fibroblast cells co-cultured with K6-K14/V7-expressing OSCC cells can be converted into a pro-inflammatory type of cancer-associated fibroblasts (iCAFs). Those findings suggest the ability of K6-K14/V7-expressing OSCC cells in remodeling tumor microenvironment. In this study, we will discuss how K6-K14/V7-expressing OSCC cells educate iCAFs to promote cancer cell migration and invasion via intercellular communication by EVs. The key signaling cascades and the associated regulatory networks will be also discussed. Our study, therefore, uncovers the pro-tumorigenic functionality of K6-K14/V7 by creating an inflammatory tumor microenvironment. Mental distress is highly prevalent in university students, and autistic traits can hinder academic education. The substantial lifestyle changes experienced by new university students can induce mood and anxiety dysfunctions and subsequent suicide-related behaviors. The aims of this study were to evaluate the detectability of suicidal ideation using the Kessler Psychological Distress (K6) questionnaire, a short and easily administered measure, and to elucidate the relationship between K6 scores and developmental disorders following university admission in medical students. The mental states of 341 new students at University School of Medicine were prospectively evaluated using the short questionnaire including the K6 at admission and 6 months later. Participants were simultaneously assessed on the five core features of autism spectrum disorder (ASD), four features of attention-deficit hyperactivity disorder (ADHD), and presence of suicidal ideation on this short questionnaire. K6 scores were significantly higher 6 months after admission than at admission ( p = .028). Students with suicidal ideation had significantly higher K6 scores than those without ( p < .001), and K6 scores at admission differentiated students with suicidal ideation both at admission (area under the curve [AUC] = 0.947) and 6 months after admission (AUC = 0.913). K6 scores were positively correlated with scores of ASD- and ADHD-related items (ρ = .451 and .440, respectively) and with autism-spectrum quotient scores ( p = .041, ρ = .501). In conclusion, the K6 questionnaire may be useful in efficiently detecting university students with suicidal ideation or autistic traits.

Abstract:
Emerging evidence suggests that probiotics are beneficial in non-alcoholic fatty liver disease (NAFLD). This study aimed to explore the effects of two Lactobacillus plantarum strains, ATG-K2 and ATG-K6 (isolated from Korean fermented cabbage), in a rat model of high fat/high fructose (HF/HF) diet-induced NAFLD. Rats with NAFLD were randomized into four groups (HF/HF diet control, (HC); HF/HF diet with silymarin, (PC); HF/HF diet with ATG-K2, (K2); and HF/HF diet with ATG-K6, (K6)) with healthy rats on a normal diet serving as the negative control. After treatment, histopathological and biochemical analyses of the blood and liver tissue were conducted. In addition, fecal microbiota was analyzed using the MiSeq platform. Compared with HC rats, K2 and K6 rats experienced significantly lower body weight gain, displayed decreased hepatic lipid accumulation, had lower serum levels of aspartate aminotransferase and alanine aminotransferase, and showed increased antioxidant enzyme activities. Moreover, de novo lipogenesis-related genes were downregulated following K2 and K6 administration. The fecal microbiota of K2 and K6 rats contained a higher proportion of Bacteriodetes and a lower proportion of Fimicutes than that of HC rats. Taken together, our results suggest that L. plantarum strains ATG-K2 and ATG-K6 are potential therapeutic agents for NAFLD.

The genomes of the O3:K6 strains of Vibrio parahaemolyticus which abruptly emerged in Calcutta, India, in February 1996 and which demonstrated an unusual potential to spread and an enhanced propensity to cause infections were examined by different molecular techniques to determine clonality. No restriction fragment length polymorphism (RFLP) in the gene encoding the thermostable direct hemolysin was observed among the O3:K6 isolates of V. parahaemolyticus. Clonal diversity among the O3:K6 strains became evident by examining the RFLPs of the rrn operons and by the use of pulsed-field gel electrophoresis. Five ribotypes were distinguished among the O3:K6 strains examined, with ribotype R4 constituting the major type. Strains of O3:K6 isolated between June and August 1996 showed different pulsotypes compared to the pulsotypes of strains isolated before and after this period, indicating genetic reassortment among these strains, but those isolated between August 1996 and March 1998 showed identical or nearly similar pulsotypes. It is clear that there is a certain degree of genomic reassortment among the O3:K6 clones but that these strains are predominantly one clone.

Abstract:
The investigation of structure-property behavior of extruded cast films prepared from blends of thermotropic liquid crystalline copolyesters with polyethylene terephthalate (PET). Data were obtained which showed not only the temperature dependence of the moduli and stress-strain behavior but also the orientation effects that must be prevalent in order to explain the differences between the moduli measured parallel and perpendicular to the extrusion direction. Only at high liquid crystal polymer (LCP) compositions is the modulus particularly increased. The modulus enhancement with lower LCP content and utilization of process variables are discussed. Specifically, the extruder gear pump speed did not enhance Young's modulus at the same LCP content as extensively as did the effect of extruder screw speed. Also a study to synthesize and characterize new segmented copolymers that could produce unusual film properties are discussed. The approach involved the synthesis of high Tg (220 C) isotropic poly (aryl ether sulfone) oligomers of varying segment molecular weights. The thermal and mechanical studies of the copolymers have been carried out to probe the potential of these copolymers for signs of liquid crystalline character and to note their ability to thermally crystallize as well as to crystallize by solvent or strain inducement. Along these lines, thermal analysis, polarizing hot-stage microscopy, wide angle x-ray scattering and mechanical testing were utilized in this investigation

L'analyse du comportement de hanseniaspora guilliermondii k6 en culture continue a ete approfondie afin de mieux comprendre l'apparition spontanee d'oscillations aux niveau de certains parametres (biomasse, concentrations en co2, o2 et metabolites produits). Ce phenomene est probablement du a une inhibition par un des metabolites formes. Enfin, trois esterases de la souche k6 ont ete isolees (dont deux purifiees) et caracterisees.

Abstract:
Understanding how atoms interact is a fundamental aspect of chemistry, biology and materials science. There have been great advances in the knowledge of covalent and ionic bonding over the past twenty years but one of the major challenges for chemistry is to develop full understanding of weak interatomic/intermolecular forces. This thesis describes fundamental studies that develop the basic understanding of weak interactions between heavier polarisable elements. The chosen methodology is to constrain heavy atoms using a rigid naphthalene backbone. When substituents larger than hydrogen, are positioned at close proximity at the peri-positions of a naphthalene molecule they experience steric strain; the extent of which is dictated by intramolecular interactions. These interactions can be repulsive due to steric hindrance or attractive due to weak or strong bonding. In efforts to understand the factors which influence distortion in sterically crowded naphthalenes and study possible weak intramolecular interactions between peri-atoms, investigations focussed on previously unknown mixed 1,8-disubstituted naphthalene systems. Mixed phosphorus-chalcogenide species were initially studied; three mixed phosphine compounds of the type Nap[ER][PPh2] were prepared along with their chalcogenides and a series of metal complexes. The study of interactions between heavy atoms was progressed by investigations into a series of mixed chalcogenide compounds of the type Nap[EPh][E’Ph] (E = S, Se, Te). Subsequent reaction of the chalcogenide systems with the di-halogens, dibromine and diiodine, afforded a mixture of charge transfer and insertion adducts displaying an array of different geometries around the chalcogen atom. From molecular structural studies, a collection of intramolecular peri-interactions were found, extending from no interaction due to repulsive effects, weak attractive 3c-4e type interactions and one example containing a strong covalent peri-bond. Further weak intramolecular interactions observed include CH-π and E•••E’ type interactions plus π-π stacking between adjacent phenyl rings. It was discovered that the bulk of the peri-atoms is influential on the distance between them, but this is not the only factor determining the naphthalene geometry. Inter- and intramolecular interactions can also have an impact and furthermore the number, size and electronic properties of substituents attached to the peri-atoms can determine molecular distortion.

Abstract As one of the most culturally diverse countries in the world, Australia has a high proportion of minority communities. However, its ageing population, particularly within these ethnic minorities, faces a range of barriers or difficulties in gaining access to and using health and aged care services. This study aims to identify the acculturation factors that affect the health status of Iranian-born elderly immigrants to Australia and their utilisation of health and community aged care services. The results of this study will be of value to Iranian elders, their families, and Australian aged health care service providers. The findings could also contribute towards enriched multicultural policy and improved social fairness, access to services, and equity for the aged from different ethnic backgrounds. 302 Iranian migrants aged 65 years who had lived in the Sydney Metropolitan area for at least six months were surveyed via a written questionnaire, face-to-face interviews, and telephone interviews. The results were analysed using SPSS and then compared to the findings from a 1999 survey of NSW elderly. The results indicate that Iranian migrants suffer higher levels of psychological distress and are more limited in their physical functioning than the general population of older Australians. They are in greater need of assistance with activities of daily living, have a lower sense of wellbeing, and are far less likely to utilise aged care services. Iranian migrant who do not speak English at home experience these disadvantages to an even greater extent English language proficiency was the only acculturation factor found to affect whether Iranian elderly utilised health and community aged care services, while ability to engage in activities of daily living (ADL) was the only health variable associated with their utilisation of community supportive aged care services. This variable did not predict the use of community aged care services in the broader sample of NSW respondents. Abstract As one of the most culturally diverse countries in the world, Australia has a high proportion of minority communities. However, its ageing population, particularly within these ethnic minorities, faces a range of barriers or difficulties in gaining access to and using health and aged care services. This study aims to identify the acculturation factors that affect the health status of Iranian-born elderly immigrants to Australia and their utilisation of health and community aged care services. The results of this study will be of value to Iranian elders, their families, and Australian aged health care service providers. The findings could also contribute towards enriched multicultural policy and improved social fairness, access to services, and equity for the aged from different ethnic backgrounds. 302 Iranian migrants aged 65 years who had lived in the Sydney Metropolitan area for at least six months were surveyed via a written questionnaire, face-to-face interviews, and telephone interviews. The results were analysed using SPSS and then compared to the findings from a 1999 survey of NSW elderly. The results indicate that Iranian migrants suffer higher levels of psychological distress and are more limited in their physical functioning than the general population of older Australians. They are in greater need of assistance with activities of daily living, have a lower sense of wellbeing, and are far less likely to utilise aged care services. Iranian migrant who do not speak English at home experience these disadvantages to an even greater extent English language proficiency was the only acculturation factor found to affect whether Iranian elderly utilised health and community aged care services, while ability to engage in activities of daily living (ADL) was the only health variable associated with their utilisation of community supportive aged care services. This variable did not predict the use of community aged care services in the broader sample of NSW respondents. Since limited proficiency in English placed elderly Iranian migrants at greater health risk and impeded their access to necessary assistance, the findings suggest that they would clearly benefit from English classes and from access to health and community care services and information regarding these services in the Farsi language.

Abstract:
Novel ceramic membrane cells of BaCe₀.₅Zr₀.₃Y₀.₁₆Zn₀.₀₄O[subscript(3-δ)] (BCZYZ), a proton-conducting oxide, have been developed for electrocatalytic ammonia synthesis. Unlike the industrial Haber-Bosch process, in this work an attempt to synthesise ammonia at atmospheric pressure has been made. The membrane cell fabricated by tape casting and solution impregnation comprises of a 200 μm-thick BCZYZ electrolyte and impregnated electrode composites. Electrocatalysts for anode and cathode were investigated. For the anode, the co-impregnation of Ni and CeO₂ provided excellent electrode performance including high catalytic activity, sintering stability and compatibility with the BCZYZ electrolyte. The best composition was the mixture of 25 wt% NiO and 10 wt% CeO₂. A symmetrical cell prepared with this electrode composition revealed low polarisation resistances of 1.0 and 0.45 Ωcm² in humidified 5% H₂/Ar at 400 and 500 °C, respectively. For the cathode, 25 wt% of impregnated Fe oxide provided a satisfactory performance in non-humidified N₂ atmosphere. Significant amounts of ammonia were produced from the single cell with Ni-CeO₂ anode and Fe oxide cathode at 400-500 °C under atmospheric pressure. Ammonia formation rate was enhanced by Pd catalyst addition and electrochemical performance was improved by Ru addition. The highest ammonia formation rate of 4 x 10⁻⁹ mols⁻¹cm⁻² was attained using the cell with a Pd-modified Fe cathode at 450 °C. The formation reaction of ammonia typically consumed around 1-2.5 % of total applied current while most of the applied current was employed in H⁺ reduction. The total current efficiency of around 90-100 % could be obtained from the membrane cells.

This paper describes and evaluates the use of SIMD floating point instructions for scientific calculations. The performance of these instructions is compared with ordinary floating point code. Implementation concerns, the effects of loop unroll as well as matrix size variations are analyzed. Execution speeds are compared using matrix multiplication. The intrinsic incompatibility of the SIMD floating point implementations used by different manufacturers requires the use of two different instruction sets: 3DNOW! on the AMD K6 processor and the Streaming-SIMD Extensions (SSE) on the Intel Pentium III processor.

The first advanced boiling water reactor (ABWR)—Kashiwazaki-Kariwa Nuclear Power Station Unit 6 (K6)—was built in Japan by Tokyo Electric Power Corporation (TEPCO) in 1996. Subsequently, Kashiwazaki-Kariwa Nuclear Power Station Unit 7 (K7) was constructed (completed in 1997). The K6 and K7 have been operating safely for more than 6 years. Presently, four additional ABWRs are under construction (two in Japan and two in Taiwan), with an additional nine ABWRs in Japan that are currently in the planning stage. One of the ABWRs now under construction is Hamaoka Unit 5 (H5), owned by Chubu Electric Power Co., Inc. The H5 has an estimated completion date of January 2005. The latest technologies have been applied to H5, which will result in improved economics, ease of maintenance, increased reliability, and a reduction in radiation exposure. Below, you will find the latest major technologies applied to H5 in terms of the history of development and verification of manufacturing stages: (1) Sealless Fine-Motion Control Rod Drive (S-FMCRD): S-FMCRD provides fine-motion control by the electric motor for normal operation and hydraulic pressure for scram insertion. This redundancy of power supply is similar to the conventional FMCRD. Additionally, S-FMCRD experiences excellent performance due to its magnetic coupling. As a result, S-FMCRD provides improved operation, ease of maintenance, increased reliability, and reduced occupational radiation exposure. (2) Multi-drive configuration of Reactor Internal Pump (RIP) Adjustable Speed Drive (ASD): Under a conventional configuration, one ASD drives one RIP. Under a multi-drive configuration, one ASD drives two or three RIPs. As a result, four ASDs would drive ten RIPs resulting in improved economics, ease of maintenance, and a smaller footprint. (3) Debris Filter Integrated Fuel Support (DFFS): In order to eliminate the risk of debris causing a fuel leak, debris filters have been developed that will be attached at the core-support orifice. DFFS enhances reliability and improves economics.
