The objective of this work was to evaluate the performance of the Crotalaria juncea, under applications of different doses of cattle manure and of earthworm humus. An experiment was carried out in the period of August to October of 2007 in the greenhouse of the Plant Science Department of the Universidade Federal Rural do Semi-Árido (UFERSA), Mossoró-RN. A 2 x 5 factorial scheme with thee replications of two plants per plot in a completely randomized design was used. The treatments consisted of the combination of two sources of organic matter ( cattle manure and earthworm humus ) with five doses of these sources ( 0, 15, 45, 60 t ha-1 ) the doses of organics fertilizers were incorporated to the total soil volume and, soon after application of the treatments it was sowed five seeds per pot with Crotalaria juncea, leaving later two plants per pot. The evaluated traits were: plant height and stalk diameter, number of leaves per plant, shoot fresh and dry mass. Significant interaction between sources of organic matter and their doses was observed on plant height, stalk diameter and number leaves per plant. The highest plant height ( 173.50 cm) and stalk diameter (4,83 cm) were obtained with the dose application of 45 t ha-1 of cattle manure, being this value 24% higher as compared to that value in the same dose of earthworm humus. These was not significant the interaction between sources of organic matter and their doses on shoot fresh and dry mass. The Crotalaria juncea responded very well the input of the organic matter sources. The dose of organic matter to be recommended for fresh production of Crotalaria juncea is of 40 t ha-1 . the production of more vigorous plants was provided by the cattle manure
O objetivo deste trabalho foi avaliar a performance da Crotalaria juncea sob a aplicação de diferentes doses de esterco bovino e húmus de minhoca. Um experimento foi conduzido no período de agosto a outubro de 2007 na casa de vegetação do Departamento de Ciências Vegetais da Universidade Federal Rural do Semi-Árido (UFERSA), Mossoró-RN. O delineamento experimental utilizado foi em blocos completos casualizados em esquema fatorial 2 x 5, com três repetições com duas por parcela. Os tratamentos consistiram da combinação de duas fontes de matéria orgânica (esterco bovino e húmus de minhoca) com cinco doses destas fontes ( 0, 15, 30, 45, 60 t ha-¹). As doses dos fertilizantes orgânicos foram incorporados ao volume total do solo, e, logo após a aplicação dos tratamentos, semeou-se cinco sementes de Crotalaria juncea por vaso, posteriormente deixando-se das plantas por vaso. As características avaliadas foram: altura de plantas e diâmetro do caule, número de folhas por planta, massa verde e seca da parte aérea. Interação significativa entre as fontes de matéria orgânica e suas doses foi observada na altura de planta, diâmetro de caule e número de folhas por planta. A maior altura de planta (173,50 cm ) e o maior diâmetro de caule (4,83 cm) foram obtidos com aplicação da dose de 45 t ha-¹ de esterco bovino. Para o número de folhas por planta o maior valor foi observado na dose de 30 t ha-¹ de esterco bovino, sendo este valor 24 % superior ao obtido com o húmus de minhoca na mesma dose, não houve interação significativa entre fontes de matéria orgânica e suas doses na massa fresca e seca da parte aérea. A Crotalaria juncea respondeu muito bem ao aporte das fontes de matéria orgânica.a dose de matéria orgânica recomendada para a produção da Crotalaria juncea é de 40 t ha-¹. Em poucas áreas da economia o Brasil pode desempenhar papel tão destacado quanto no uso sustentável e inteligente do potencial de sua biodiversidade. As plantas medicinais sempre foram objeto de estudo, buscando-se novas fontes para obtenção de princípios ativos, responsáveis por sua ação farmacológica ou terapêutica. Jungia sellowii é uma planta endêmica brasileira que tem suas raízes utilizadas para distúrbios digestivos, porém permanece pouco explorada e conhecida cientificamente. Com o intuito de promover a investigação fitoquímica da espécie para avaliação da atividade biológica in vitro dos componentes majoritários, o extrato bruto das raizes foi submetido a sucessivos procedimentos cromatográficos. Dos quais foi possível purificar e caracterizar 10 substâncias, sendo dois esteroides; o ß-sitosterol e o estigmasterol; um derivado do ácido cafeico, o cafeato de etila; sete terpenóides dos quais cinco são derivados da mesma via metabólica sendo eles a perezona, o a e ß- piptizóis, a diperezona, a curcuhidroquinona-O-ß-Glicopiranosideo; dois derivados do linalol e ácido jungico. Todas as substancias estão sendo descritas pela primeira vez para a espécie e a curcuhidroquinona-O-ß-Glicopiranosideo uma substancia nova, além do derivado trixano ter sido isolado pela primeira vez de fonte natural. A avaliação da atividade leihmanicida in vitro em promastigotas de L. infantum, L. donovani e L. amazonenses da fração Butanol mostrou na dose de 500µg/mL uma porcentagem de inibição de 73%, 60% e 72% respectivamente. Contudo a substancia majoritária curcuhidroquinona-O-ß-Glicopiranosideo apresentou atividade somente contra L. amazonenses sugerindo um efeito sinérgico de outras substancias presentes na fração responsáveis pela atividade. Já a perezona apresentou potente atividade leishmanicida contra amastigotas de L. amazonenses Com uma IC50 de 6,28 ±0,14 µM. Os resultados obtidos ressaltam a importância do estudo fitoquímico e o potencial biológico de Jungia sellowii.
Abstract : In a few areas of the economy Brazil can play a role as prominent and sustainable as the use of the potential of its biodiversity. Medicinal plants have always been the focus of many studies, as sources for active principles responsible for their pharmacological or therapeutic action. Jungia sellowii is a Brazilian endemic plant and has its roots are used for digestive disorders, but remains poorly explored and unknown scientifically. In order to promote the phytochemical research of the specie and to evaluate the biological activity in vitro of it s major components, the crude extract of the roots was subjected to successive chromatographic procedures. Of which was possible to purify and characterize 10 substances, two steroids ß-sitosterol and stigmasterol; a derivative of caffeic acid, ethyl caffeate; seven terpenoids five derived from the same metabolic pathway: perezone, a and ß- piptizols, diperezone and curcuhydroquinone-O-ß-glucopyranoside; two derivatives of linalol, and jungic acid. All substances were described for the first times in the species, curcuhydroquinone-O-ß-glucopyranoside a new substance, and the trixane derivative was isolated for the first time from natural source. The evaluation of the in vitro activity leishimanicid against promastigotes of L. infantum, L. donovani and L. amazonenses of the butanol fraction showed inhibition percentage of 73%, 60% and 72%, respectively. Curcuhydroquinone-O-ß-glucopyranoside the majority substance was active only against L. Amazonensis, suggesting a synergistic effect of other substances present in the fraction responsible for the activity. And perezone has show potent leishmanicid activity against amastigotes of L. amazonenses with an IC50 of 6,28 ±0,14 µM. The results highlight the importance of phytochemical study and the biological potential of J. sellowii.

O desenvolvimento de procedimentos sensíveis para rápida obtenção de informações precisas sobre o potencial fisiológico de lotes de sementes é essencial para o desenvolvimento de programas de controle de qualidade pelas empresas produtoras das mesmas. Nesta pesquisa, foi avaliada a eficiência de diferentes testes de vigor na determinação do potencial fisiológico de sementes de crotalária e, o desempenho das mesmas sob condições de estresse hídrico. O estudo foi conduzido em duas etapas, sendo a primeira relativa à seleção de procedimentos adequados para a condução dos testes de condutividade elétrica, envelhecimento acelerado, tetrazólio e de um sistema automatizado de análise de imagem de plântulas (Seed Vigor Imaging System – SVIS). Na segunda etapa, as sementes foram submetidas a diferentes situações de disponibilidade hídrica, utilizando-se soluções aquosas de polietileno glicol (PEG 6000). Para tal, foram utilizados cinco lotes de sementes. Os testes de condutividade elétrica, envelhecimento acelerado com solução salina saturada e o uso do software SVIS em plântulas com três dias de idade podem detectar eficientemente diferenças de vigor entre lotes de sementes de crotalária. O desempenho fisiológico de sementes dessa espécie é fortemente afetado pelo estresse hídrico

Resumo: A presença de metais pesados no ambiente é atualmente, um dos principais problemas de contaminação ambiental, uma vez que, os metais liberados no ambiente contaminam o solo e entram na cadeia alimentar através das plantas, causando efeitos tóxicos a curto e a longo prazo aos animais e seres humanos. No caso do metal pesado Níquel (Ni), foi constatado que a sua presença nas plantas pode diminuir o crescimento, reduzir a taxa de fotossíntese e provocar alterações, tanto nas atividades enzimáticas quanto metabólicas.Pouca informação está disponível na literatura, com referência a resposta antioxidante das plantas à expressão a esse metal. Neste sentido, os objetivos deste trabalho foram realizar diferentes ensaios para avaliar o efeito fitotóxico do Ni em plântulas de C. juncea. Neste estudo foram analisados parâmetros bioquímicos relativos à atividade das enzimas antioxidantes, Catalase (CAT), Superóxido Dismutase (SOD) e Glutationa Redutase (GR). Constatou-se que, não houve alteração da atividade GR e CAT nas raízes. Entretanto, atividades destas enzimas apresentaram aumento significativo na parte aérea. O aumento na atividade da GR, na parte aérea, pode ser explicado pelo fato de ser o ciclo Halliwell-Asada o principal mecanismo que age na desintoxicação de Espécies Ativas de Oxigênio. Quanto à atividade da SOD, quando comparados ambos tecidos, foi constatado que na parte aérea foi pouco alterada, entretanto, a atividade da SOD foi estimulada nas raízes na presença do Ni. Para aumentar a precisão dos resultados, quantificou-se pela técnica de Fluorescência de Raios X, a concentração de NiCl2 e o seu efeito na absorção de nutrientes nas plântulas de crotalaria. Nesta análise, observou-se o acúmulo do metal nas raízes e baixa translocação para a parte aérea. Também foi analisada a concentração de Malonaldeído (MDA) na parte aérea de plântulas de C. juncea, sendo constatada, a ocorrência de peroxidação lipídica na presença do Ni. Finalmente, determinou-se o efeito do Ni sobre as concentrações de aminoácidos solúveis em raízes e na parte aérea, como estratégia desta planta para superar o efeito do Ni , não tendo sido observadas variações significativas nas concentrações dos mesmos com os diferentes tratamentos com Ni. Abstract: The contamination of the environment with heavy metals has become a world-wide problem. Heavy metals can have direct effects on human health and decreases agricultural yields. In the presence of Nickel (Ni), plant growth and the net photosynthetic rate have been shown to be significantly reduced, even at low concentrations. Moreover, Ni can also inhibit the activity of several groups of enzymes and metabolism. The effects of the Ni on the activies of the antioxidant enzymes and metabolism were investigated in C.juncea seedlings. Enzymatic assays for Catalase (CAT) Glutathione Reductase (GR) and Superoxide Dismutase (SOD) were carried out. Response patterns for CAT and GR were similar with low values in the roots and increased activity in the shoots. A significant increase in CAT and GR activity in the shoots suggests that these enzymes play a role in the detoxification process, of reactive oxygen species (ROS) produced by the induced Ni stress, possibly by subquesent dismutase. On the other hand, the activity of SOD was stimulated in the roots, and decreased in the shoots. To gain insight into the response of C. juncea seedlings to the oxidative damage induced by excess Ni, were conducted studies of Malondialdehyde (MDA). It was evident that Ni induced an increase an lipid peroxidation in the shoots during the studied period. The Ni concentration in shoots and roots was determined for C. juncea and demonstrated an increase in the concentration of Ni in the roots, however the shoots did not present an accumulation. Further experiments were carried out on total free amino acids. It was observed that Ni treatment did not cause any change in the accumulation of total free amino acids in the roots. The results suggest that in C. juncea seedlings did not exhibit any major variation following Ni treatment. Key words: Antioxidant enzymes, Amino acids, C. juncea, Nickel, Environmental Pollution

Full textThe bicycle use has been diffused among the population in general, the people started to see it as a way of transport. It has been used both for transportation and for professional cyclists who use it as tool to reach their goals or to win bicycle races. The evolution, both on materials and cycling/bike riding techniques in the second half of last century has changed. In the other hand, fatigue cases which follow some disasters got some relevancy. The first goal was to find some explanation for what happened and how to avoid the problem – the crack, or in some cases the disaster. The cyclism followed the evolution, both on materials, on the road bikes and off-road bikes. Actually many riders know that some samples of bike frames have failed during use or even worse the crack appeared and the frame broke down suddenly. The discussion concerning the bicycle frame resistance doesn’t look finished. The cyclists demands are different. The loads encountered during cycling can be very different comparing on and off-road bikes – Road and Mountain Bike. The start point for the present work involved a sample of a Mountain Bike frame which had failed. The work development was to propose a kind of load which could test the fatigue strength of the material analyzed. The load proposed should be easy to simulate in finite element analysis – FEA, and it must reproduce a normal use condition. The the load were defined, the structure were simulated using a specific software – FEA. Finite element analysis. After the test the responses were collected. The primary goals were to find the location of the main tensile and its measurement in the frame. It coincided exactly with the crack in the real bike frame. Using the normal procedure to predict the fatigue life in the welded joint, the result reached 40% of the fatigue resistance for that joint, considering the load applied. The analysis compared the project resistance to the load to check its capacity to absorb loads igual or above the one which were used. Some riders that are more strength, those who can apply more than the load proposed or even those who are more radical in their way of riding could use the combination of these situations making the load easly amplified by dynamics effects, bumps and high speed for example. This hard condition of use is complex to be reproduced because there isn’t any information concerning strain, speed, angles and the montion that the rider could do when riding a mountain bike.
