Abstract:
Background:Janus kinase 1 (JAK1) inhibitors are efficacious in rheumatoid arthritis (RA). Despite having similar efficacy, in vitro studies have shown differences in JAK selectivity profiles for the small-molecule JAK inhibitors (JAKi) baricitinib (BARI), tofacitinib (TOFA), and upadacitinib (UPA).1For example, BARI and UPA are JAK1/JAK2 selective, while TOFA is JAK1/JAK3 selective, but each JAKi has some activity against other JAKs. As JAKs form signaling pairs, differences in selectivity could lead to distinct pharmacologic profiles that may impact clinical efficacy and safety.Objectives:As a first step to understand the basis of potential differences at therapeutic doses, we compared the selectivity and potency of filgotinib (FIL) and its major metabolite (MET) to those of BARI, TOFA, and UPA in cytokine-stimulated peripheral blood mononuclear cells (PBMCs) and whole blood (WB).Methods:PBMCs and WB from healthy donors were incubated in vitro with 8 doses of each JAKi, and levels of signal transducer and activator of transcription phosphorylation (pSTAT) were measured following cytokine stimulation. Half maximal inhibitory concentration (IC50) values were calculated in phenotypically sorted leukocyte populations by flow cytometry. Therapeutic dose relevance of the in vitro analyses was assessed using calculated mean concentration-time profiles from JAKi population pharmacokinetic data in RA subjects. For each JAKi, the time above IC50and average daily pSTAT inhibition were calculated for each cytokine/STAT pair in B cells, CD4+ T cells, CD8+ T cells, monocytes, and/or NK cells.Results:Cellular assays in PBMCs and WB showed dose-dependent inhibition of cytokine-induced pSTATs with all JAKi (correlation between the protein-adjusted IC50values from PBMCs and IC50values from WB, r2=0.98). Among the most potently inhibited pathways were JAK1/TYK2-dependent cytokine, interferon alpha (IFNα), and the JAK1/2-dependent cytokine, interleukin (IL)-6. FIL and MET had weaker potencies against JAK2/TYK2 (G-CSF/pSTAT3), JAK1/2 (IFNƴ/pSTAT1), and JAK2/2 (granulocyte-macrophage colony-stimulating factor [GM-CSF])-dependent pathways compared to JAK1/TYK2 (IFNα/pSTAT5). FIL and MET showed the greatest selectivity vs the JAK2/2 pathway (GM-CSF/pSTAT3) in monocytes.The mean concentration-time profiles and time above IC50over 24 hr for each cytokine/STAT pathway showed that JAK1/2 (IL-6/pSTAT1) and JAK1/TYK2 (IFNα/pSTAT1) pathways were strongly modulated with all tested JAKi. FIL (200 mg) showed similar activity in average target coverage and time above IC50to the approved low doses of TOFA (5 mg) and UPA (15 mg); conversely, FIL had reduced mean average inhibition and time above IC50levels against JAK1/2 (IFNƴ/pSTAT1), JAK1/3-dependent cytokines (IL-2, -4, and -15), JAK2/TYK2 (G-CSF/pSTAT3), and JAK2/2 (GM-CSF/pSTAT5)-dependent pathways compared to TOFA and UPA, and in certain cases to BARI (2 mg).Conclusion:Different JAKi modulate distinct cytokine pathways to varying degrees, and no agent potently and continuously inhibited an individual cytokine signaling pathway throughout the dosing interval. FIL (200 mg) showed a similar inhibition profile to TOFA, BARI, and UPA against the JAK1/TYK2- (IFNα/pSTAT1) or JAK1/2-dependent (IL-6/pSTAT1) responses, consistent with the role of these pathways in clinical efficacy.2However, FIL displayed a differentiated pharmacologic profile from the other JAKi, showing biologically reduced activity on the JAK1/2 (IFNγ)-, JAK1/3 (IL-2, -4 and -15)-, JAK2/TYK2 (G-CSF)-, and JAK2/2 (GM-CSF)-dependent pathways, which play important roles in hematopoiesis and immune function. These data suggest that FIL (200 mg) may have less impact on a subset of homeostatic immune functions signaling via JAK2 and JAK3 than those observed at the clinically approved doses of TOFA (5 mg and 10 mg), UPA (15 mg), and BARI (4 mg).References:[1]McInnes IB, et al. Abstract:
Jake hard red spring wheat (Triticum aestivum L.) was developed using a modified bulk breeding method at the University of Alberta, Edmonton, AB. Jake is an awned, hollow-stemmed line with high yield potential, medium tall plants, and medium maturity. During the three years (2015–2017) of evaluation in the Parkland Wheat Cooperative test, Jake yielded 6% higher than the mean of all of the checks, and matured 0.7 and 1.7 d later than Parata and Splendor but 2.9 d earlier than Glenn. Jake was 91.2 cm tall, shorter than AC Splendor (95.8 cm), but similar in height to Glenn (91.8 cm) and Parata (92 cm). The lodging score of Jake (2.2) was lower than Parata (3.1) and AC Splendor (3.1), but similar to Glenn. The test weight of Jake (80.8) was higher than AC Splendor (78.3), similar to Parata (80.5), but lower than Glenn (82.5). The grain weight of Jake (35.6 g) was similar to Parata (35.6 g), but lower than Glenn (36.7 g) and AC Splendor (37.4 g), while the NIR Protein of Jake (15.9%) was higher than Glenn (15.5%) and similar to the other checks. Jake was moderately resistant to resistant to leaf, stem, and stripe rusts, and moderately resistant to common bunt during the 3 yr of testing. The reaction of Jake to Fusarium head blight was variable and ranged from moderately susceptible to moderately resistant, with DON values similar to Carberry and Glenn. Three years of end-use quality evaluation has indicated that Jake is acceptable for the CWRS class.

With his thought-provoking and socially relevant music, American composer Jake Runestad has quickly become one of the most performed choral composers of the 21st century. Although music and social justice have been tied together for centuries, there is a new movement bringing social justice to American choral music in a noticeably increased manor, and Jake Runestad is a leading composer in this movement. In this paper, I provide a detailed analysis into the social justice themes employed by Runestad, interviews with him and several well-respected American choral directors programming and commissioning his music, as well as compositional devices employed within his compositions. The purpose of this study is to show Jake Runestad's place as an American choral composer by offering a historical overview of the social justice themes in American music and Western choral music separately. I will then narrow the scope to Jake Runestad, who since 2013 has been using his choral music to bring awareness to human inequalities within the United States today.

My paintings are a mediated autobiography focusing on fragmented, cropped or collaged elements derived from photographic sources. Banal scenes based on my experience growing up in rural Washington state are charged with multivalent interpretations via close ups, veiling and intentional obfuscation. Through these means, altered color and expressionistic paint handling, they subtly point outside of their beginnings as the photographic and personal and into a dialogue with historical and social sites such as punk music, abstract expressionism and existential literature.

Abstract:
This dissertation explores the development of commercial entertainments and film exhibition in the urban South around the turn of the last century through the growth and decline of Jake Wells Enterprises. A former professional baseball player, Wells invested in a wide variety of public amusements, with the core of his early business centered on establishing and organizing a string of vaudeville, popularly priced, and legitimate theaters throughout the largest cities in the region, a network he later transitioned to showing exclusively motion pictures. A thorough analysis of period newspapers, trade journals, and some business records covering Wells’ career provides much-needed evidence for film and cultural historians wishing to understand the genesis and evolution of public amusements in the region, and its negotiation of traditional social and cultural institutions. In the 1890s, Wells played and managed several professional baseball teams in the South. The sport educated players and spectators alike to both the values and creed of New South progress, and to rising tensions confronting the intersection of modern and traditional forms of culture. Using his experiences and contacts gained in baseball, Wells helped foster a culture of entrepreneurship and innovation required for the progress of media industries in the region, establishing social networks of knowledge and improving distribution flows of entertainment. The dissertation explores how race and the genteel emerged as regional characteristics most influential to the success of this conversion in many urban areas. Protestants and evangelical culture served as the bulkhead supporting opposition to new amusements. Wells’ expansion plans and violations of Sabbath day laws evoked a “spatial” battle between commercialism and religion where political, social, and cultural power drawn from place and identity were challenged and reconfigured. Another chapter explores the exhibition and reception of early Civil War films in the region. Wells and other exhibitors were influential in their production and circulation nationwide, and positioned cinema as an alternative shrine to commemorate the Lost Cause in many communities. The last chapter shows how Wells failed to meet local demands and consumer desires in competition with the rise of national chain theaters and Hollywood’s vertical integration.

Each of the artists engages, in various ways, with issues relating to biotechnology and the body. The convergence of technology and the body arouses both utopian desires and dystopian nightmares; many of these desires and fears are prefigured in ancient myth and legend. U za­mjenu za takve usluge, posebne interesne skupine nude političku potporu izravno ili neizravno utječući na stajališta opće javnosti ili na oba načina. Ko­risti za svakog pojedinca male lobističke grupe od posebnog interesa mogu biti ogromne, dok se troškovi takvih političkih „transakcija” obično raspore­đuju kroz veće poreze (ili dug) na cijelu populaciju poreznih obveznika. Po­sljedično, troškovi za prosječnog poreznog obveznika čine se malima i sto­ga će se vjerojatno tolerirati. Kao rezultat toga, stvara se mali trošak za vla­du da zadovolji zahtjeve interesnih skupina, dok (političke) koristi mogu biti znatne. Na taj način interesne skupine mogu u konačnici utjecati na djelova­nje vlade i njezin udio u gospodarstvu. Međutim, nisu sve interesne skupine jednako učinkovite u ostvarivanju svojih ciljeva. Glavni cilj ovog rada je istra­žiti teorijske postavke i empirijske pokušaje procjene odnosa između veličine javnog sektora u gospodarstvu i interesnih skupina.

Abstract:
Single Point Incremental Metal Forming(SPIMF) is a process that allows architectural panels to be incrementally formed from sheet metal into doubly-curved complex shapes using a robotic arm and a stylus-like end effector. SPIMF leverages industrial robots’ precision and strength by gradually pushing the end effect or into vertically supported sheet metal.This work was inspired by Anmar Kalo andMichael Jake Newsum’s Incremental SheetMetal Forming and CITA’s Stressed Skins project. SPIMF examined how different materials, tools, and tool path generation methods impact the finished quality of completed pieces along with applications for the formed metal parts. Once an understanding of howSPIMF worked, focus was put on how to accurately program the robotic arm to produce repetitive geometries that correspond with a digital model. By creating a feedback loop that studies how the sheet metal deforms and how accurately the robot performs the forming task, new and more accurate geometry can be used to program the arm.

Abstract:
Státy se musí rozhodnout, jak budou řešit schodek veřejných rozpočtů. Tato otázka je klíčovou pro všechny státy, které se potýkají s rostoucí inflací a dalšími krizovými jevy. Jak situaci řešit a zároveň nenarušit rámec veřejné podpory stanovené v evropských smlouvách se řeší z politického a ekonomického hlediska. Je otázkou, zda to může být i daňové právo, které vnáší do veřejné podpory určité odchylky a jak se s touto situací vyrovnávají nejen státy jako celek, ale i jednotlivé články územní samosprávy.

Przedmiotem artykułu jest próba odczytania pojęcia patriotyzmu jako określonej konstrukcji semantycznej i interpretacyjno-narracyjnej, osadzonej w historycznych i kulturowych uwarunkowaniach XIX w., oraz wskazanie jego cech konstytutywnych. Punktem odniesienia w tych rozważaniach jest broszura zatytułowana: O miłości Ojczyzny – przykład niezwykle interesującej publicystyki społecznej, wydanej po raz pierwszy w 1899 r. przez Jadwigę z Działyńskich Zamoyską (1831-1923) – założycielkę Szkoły Domowej Pracy dla Kobiet w Kórniku (1882), żonę gen. Władysława Zamoyskiego (1803-1868), głównego przedstawiciela polistopadowej emigracji. W społecznym programie narodowego odrodzenia sformułowanym przez Zamoyską, patriotyzm rozumiany jest głównie jako przywiązanie do rodzinnego kraju i gotowość całkowitego poświęcenia się dla jego dobra, a także jako systematyczne odwoływanie się do kluczowych wartości, takich jak wiara katolicka, przeszłość i narodowe tradycje oraz praca organiczna u podstaw.

Słownik języka polskiego bohatera definiuje jako osobę „odznaczającą się niezwykłymi czynami”, „męstwem”, co czyni ją wyjątkową, ale też podziwianą i wartą pamięci potomnych. Informacje o bohaterach i ich osiągnięciach są więc upowszechniane oraz przekazywane z pokolenia na pokolenie. Treści na ten temat obecne były także na łamach polskiej prasy dla kobiet. Ukazywano wątki związane z historią oraz postaciami, które zapisały się na jej kartach w różnych dziedzinach życia publicznego, m.in.: w polityce, nauce, sztuce, literaturze, sporcie. Celem badań była rekonstrukcja obrazu bohaterki historycznej w polskiej prasie dla kobiet w latach 1989-1992. Aby zrealizować cel badawczy, sformułowano następujące pytania badawcze: 1) jaki jest udział pisarstwa o bohaterach, a jaki o bohaterkach na łamach polskiej prasy dla kobiet w latach 1989-1992?; 2) czym różnił się obraz bohatera od obrazu bohaterki?; 3) w jakiej dziedzinie życia dominowały „bohaterki”? Przedmiotem badań były artykuły opublikowane w „Kobiecie i Życiu”.

Abstract:
ABSTRACT Jake field, discovered in July, 2006, contains 10 oil-producing and 12 condensate gas-producing zones. The wells have high flow capacities, producing from long-perforation interval of 3,911 ft (from 4,531 to 8,442 ft). Production mechanisms include gas injection in downdip wells and traditional gas lift in updip, zonal production wells since the start-up of field in July, 2010. Following pressure depletion of oil and condensate-gas zones and water breakthrough, traditional gas-lift wells became inefficient and dead. Based on nodal analysis of entire pay zones, successful innovations in gas lift have been made since March, 2013. This paper highlights them in the following aspects: Extend end of tubing to the bottom of perforations for commingled production of oil and condensate gas zones, in order to utilize condensate gas producing from the lower zones for in-situ gas lift.Produce well stream from the casing annulus while injecting natural gas into the tubing.High-pressure nitrogen generated in-situ was used to kick off the dead wells, instead of installation of gas lift valves for unloading. After unloading process, the gas from compressors was injected down the tubing and back up the casing annulus.For previous high water-cut producers, prior to continuous gas lift, approximately 3.6 MMcf of nitrogen can be injected and soaked a couple of days for anti-water-coning.Two additional 10-in. flow lines were constructed to minimize the back pressure of surface facilities on wellhead. As a consequence, innovative gas-lift brought dead wells back on production, yielding average sustained liquid rate of 7,500 bbl/d per well. Also, the production decline curves flattened out than before.

„Przyjaciółka” będąca w okresie PRL najpopularniejszym czasopismem kobiecym edukowała kobiety w takich sferach jak rodzina, małżeństwo, gospodarstwo domowe czy rolne. Udzielała porad zdrowotnych, wychowawczych, kulinarnych czy prawnych. Pełniła także funkcję emancypacyjną wśród kobiet wiejskich, do których była kierowana. Stosując metodę analizy zawartości prasy, opracowanie ukazuje w jaki sposób tygodnik kreował proces emancypacji przedstawicielek płci pięknej. Celem opracowania jest próba ukazania propagowanych przez „Przyjaciółkę” sposobów osiągania autonomii przez kobiety wiejskie. Ponadto w oparciu o wybrane numery periodyku z lat 1956-1975 zaprezentowano formy „wyzwolenia” Polek poprzez pracę, edukację, podział obowiązków domowych i relacje w rodzinie.
