This diploma thesis is focused on a franchising business establishment, particularly on the business establishment of a fictive coffe house which could become a well known franchising concept on the Czech market. Primary advantage of franchise compared to the other types of business cooperation is mainly risk reduction for the investor (franchisee) who obtains proven model of the business. For the franchisor the franchise means an easier way how to widen his business without taking higher lability. The main reason for choosing this topic is an attractivity of this field. Correct preparation and serving of a cup of coffee is not an easy process and there is not a big number of coffee houses and restaurants which can offer such a coffee. By offering high-quality and delicious coffee at user acceptable price our company would like to achieve its objectives and be a successful. The primary objective of this diploma thesis is to analyse and define strategy, mission, vision, marketing activities, product strategy and price policy of a new coffee house. The diploma thesis "How to establish a company" focuses on pre-school education. Specifically, on the establishment of a fictitious company day care at a planned business center comprised of office buildings and an ambulance station. Primary purpose of this study is to summarize the activities and difficulties that are associated with the foundation of a successful project. The study describes business strategy, competitive advantage, business vision and mission and legislative issues relating to operation of a company day care. The thesis is divided into two parts, theoretical and practical. In the first, theoretical part, we focus on description of terms associated with starting a business, entrepreneurs, and legal environment in the Czech Republic. The second, practical and analytical, part consists of detailed project of establishment of a company day care and also outlines problems arising in this area. These are hidden discrimination against women and the lack of capacity in public nurseries. The final financial plan evaluates feasibility of the project and assesses whether the investment is the right choice.

The aim of my work was to explore the possibility that the mammalian JAK2 signalling pathway influences the structure and function of chromatin. I have demonstrated that JAK2 is present in the nucleus of both human haematopoietic cell lines and primary cells. My results suggest that JAK2 functions as a histone tyrosine kinase and phosphorylates histone H3 at tyrosine-41 (H3Y41). This novel histone modification, the first described tyrosine phosphorylation on any of the non-variant histones, regulates the binding of heterochromatin protein 1-alpha (HP1α) at a new binding site on chromatin. HP1α uses its chromo-shadow domain to bind the H3Y41 region. Phosphorylation of H3Y41 by JAK2 reduces its affinity for chromatin. This reciprocal relationship was given a functional context by demonstrating its relationship to the expression of a key haematopoietic oncogene Imo2. Genome-wide studies demonstrate that H3Y41ph is present at the 5’ end of genes and is highly correlated with active transcription. This is the first comprehensive genome wide mapping of a histone phosphorylation mark and potentially highlights a role for this novel modification in the regulation of transcription. H3Y41ph was also present at specific cis-regulatory elements on JAK2-STAT5 target genes and genome-wide mapping of STAT5 binding confirmed that STAT5 binding and H3Y41ph was coincident at a significant number of sites within the human genome. This interesting observation suggests that canonical JAK2-STAT5 signalling is not confined to the cytoplasma but also occurs at chromatin. These findings extend the existing paradigm of JAK-STAT signalling and provide a platform for a better understanding of this critical signalling pathway, which is important in both normal development and oncogenesis.

In accordance with its main goal, the main thesis shows which published data and indicators contemporary financial markets use to assess the state of major economies and forecast their short-term future development. Using Bloomberg as the primary source, the thesis provides a detailed analysis of the indicators and surveys sought by finance professionals when assessing the performance of the economy in the United States, the euro-zone (with emphasis on Germany) and, in the context of the impact on the global economy and markets, in China and Japan. A preliminary hypothesis about the similarity or closeness between the theoretical view and the practical approach of financial markets has been refuted. Instead, after close scrutiny and detailed analysis, we have established that financial markets put great emphasis on forward-looking indicators and monetary policy. This emphasis was confirmed by a survey of local investors. Using expert opinion and analysis, the thesis charts an overview of select economic indicators and their over- (payrolls, consumer sentiment) or undervaluation (regional Fed activity indexes, CFNAI).

INTRODUÇÃO: A desregulação da via de sinalização JAK-STAT é uma característica marcante das neoplasias mieloproliferativas (NMPs), doenças clonais da célula tronco hematopoética, dentre as quais encontra-se a mielofibrose (MF). Diversos inibidores de JAK foram desenvolvidos para o tratamento da MF e encontram-se em diferentes fases de desenvolvimento clínico. Devido ao seu desenvolvimento recente, pouco se sabe a respeito do papel de transportadores de membrana na farmacocinética desses compostos. Essas proteínas realizam o influxo e efluxo celular de substratos endógenos e xenobióticos, e alterações na expressão desses transportadores podem influenciar a resposta a esses fármacos. OBJETIVO: Avaliar o efeito de um inibidor comercial da via JAK-STAT na expressão gênica dos transportadores de membrana ABCB1, ABCG2, SLC22A1 e SLCO1A2 em células HepG2, Caco-2 e HEL92.1.7. MÉTODOS: Linhagens de carcinoma hepatocelular (HepG2), adenocarcinoma colorretal (Caco-2) e eritroleucemia humana homozigotas para JAK2V617F (HEL92.1.7) foram cultivadas e tratadas o inibidor comercial da via JAK-STAT JAK Inhibitor I. Para determinar a concentração ideal para o tratamento com o inibidor, as células foram tratadas com diversas concentrações do inibidor de JAK por 24 horas e foram feitos testes de viabilidade celular e fragmentação do DNA. Com as condições de tratamento padronizadas, foi extraído o RNA total das células e sintetizado o cDNA, para análise das expressões de RNAm dos genes ABCB1, ABCG2, SLC22A1 e SLCO1A2 por PCR em tempo real. Foi também avaliada a expressão dos transportadores de efluxo ABCB1 e ABCG2 por citometria de fluxo, utilizando anticorpos primários direcionados a essas proteínas. RESULTADOS: Nas células HepG2, foi observado um aumento da expressão de RNAm de ABCB1 nas células tratadas com 4,00 µM do inibidor de JAK, quando comparado com o controle (células incubadas apenas com o veículo) (P=0,041). Não foi observada alteração da expressão de RNAm de ABCG2 e SLC22A1 com o tratamento com o inibidor de JAK nessa linhagem (P>0,05); a expressão de RNAm de SLCO1A2 não foi detectada nessa linhagem. Nas células Caco-2, a expressão de ABCB1, ABCG2, SLC22A1 e SLCO1A2 não se alterou com o tratamento com o inibidor de JAK nas concentrações utilizadas (0,25 µM a 1,00 µM) por 24 horas (P>0,05). Para as células HEL92.1.7, não foi observada diferença na expressão de RNAm de ABCB1, ABCG2 e SLC22A1 com o tratamento com 1,00 µM do inibidor de JAK por 24 horas em comparação ao controle (P>0,05); nessa linhagem, a expressão de RNAm de SLCO1A2 não foi detectada. A expressão proteica dos transportadores ABCB1 e ABCG2 não sofreu alteração com o tratamento com o inibidor de JAK nas condições utilizadas nas três linhagens celulares estudadas (P>0,05). CONCLUSÕES: Apenas as células HepG2 apresentaram um aumento da expressão de RNAm do transportador de efluxo ABCB1 em concentrações elevadas do inibidor de JAK, sugerindo que os inibidores de JAK podem modular a expressão do gene desse transportador no fígado. O tratamento com o inibidor da via JAK-STAT não foi associado com alterações na expressão proteica de ABCB1 e ABCG2 em todas as células estudadas.BACKGROUND: JAK-STAT pathway signaling disregulation is a hallmark of myeloproliferative neoplasms (MPN), hematopoietic stem cell clonal diseases, among which is myelofibrosis (MF). Several JAK inhibitors have been developed for MF treatment and are found in different stages of clinical development. Because the recent development of these compounds, the role of drug transporters in their pharmacokinetics is poorly understood. These proteins perform celular influx and effux of endogenous substrates and xenobiotics, and changes in the expression of these drugs transporters may affect the response to these drugs. AIM: To evaluate the effect of a JAK-STAT pathway commercial inhibitor in gene expression of drug transporters ABCB1, ABCG2, SLC22A1 and SLCO1A2 in HepG2, Caco-2 and HEL92.1.7 cells. METHODS: Hepatocellular carcinoma cell line HepG2, colorectal adenocarcinoma cell line Caco-2 and human erythroleukemia homozygous JAK2V617F cell line HEL92.1.7 were grown and treated with the JAK-STAT pathway inhibitor JAK Inhibitor I. In order to determine the optimal concentration for treatment with the inhibitor, cells were treated with several concentrations of JAK inhibitor by 24 hours, and cell viability and DNA fragmentation tests were performed. Once the treatment conditions were standardized, total RNA were obtained from the cells, and cDNA was synthesized in order to evaluate the mRNA expression of ABCB1, ABCG2, SLC22A1 and SLCO1A2 genes, performed by real time PCR. We also evaluate the expression of drug efflux transporters ABCB1 and ABCG2 by flow cytometry, using primary antibodies directed to these proteins. RESULTS: In HepG2 cells, it was observed an increase in ABCB1 mRNA expression in cells treated with 4,00 µM of JAK inhibitor, when compared with controls (cells exposed only to the vehicle) (P=0.041). There was no change in ABCB2 and SLC22A1 mRNA expression with the treatment with JAK inhibitor in this cell line (P>0.05); SLCO1A2 mRNA was not detected in this cell line. In Caco-2 cells, ABCB1, ABCG2, SLC22A1 and SLCO1A2 mRNA expression did not change with treatment with the JAK inhibitor at the concentrations used (0.25 µM to 1.00 µM) by 24 hours (P>0.05). In HEL92.1.7 cells, it was not observed differences in ABCB1, ABCG2 and SLC22A1 mRNA expression with the treatment with 1 µM of JAK inhibitor by 24 hours when compared with controls (P>0.05); in this cell line, SLCO1A2 mRNA was not detected. Protein expression of ABCB1 and ABCG2 drug transporters has not changed with treatment with the JAK inhibitor under the conditions used in the three cell lines studied. CONCLUSIONS: Only HepG2 cells presented an increase in mRNA expression of drug efflux transporter ABCB1 in presence of high levels of JAK inhibitor, suggesting that JAK inhibitors could modulate this transporter gene expression in liver. Treatment with JAK-STAT pathway inhibitor was not associated with changes in ABCB1 and ABCG2 protein expression in all cell lines studied.

Syfte: Studien syftar till att undersöka och analysera JAK Medlemsbanks syn på det ekonomiska systemet i allmänhet och räntan i synnerhet, samt undersöka om det går att låna pengar utan ränta. Vi belyser också att det finns dolda räntekostnader i det vi konsumerar.Metod: Metoden som använts är en kvalitativ metod och data har främst samlats in genom tre ostrukturerade intervjuer med anställda och engagerade personer inom JAK Medlemsbank. Material till teoriavsnittet har samlats in från databaser, artiklar och böcker. Resultat & slutsats: Studien visar att JAK Medlemsbank är vad den utger sig för att vara, alltså en räntefri bank. Dock är detta beroende av att ränta definieras som en arbetsfri inkomst dvs. en inkomst som inte täcker några verkliga kostnader. Studien visar att ett lån i JAK Medlemsbank ger en relativt hög månadskostnad vilket också gör att det krävs en hög månadsinkomst för att låna ett större belopp. Förslag till fortsattforskning: Det räntefria alternativet är ett relativt outforskat område vilket ger stora möjligheter till fortsatt forskning. Ett av dessa outforskade områden är räntans andel av priset på en vara. För att utforska detta område anser vi att det krävs mer tid då många företag verkat motvilliga att lämna ut sådan information. However, this depends on that the interest rate is defined as a work-free income i.e. an income that does not cover real costs. Suggestions for future research: The interest-free option is a relatively unexplored area which provides significant opportunities for further research. Abstract
Myeloproliferative neoplasms (MPNs) are diseases of hematopoietic stem cell origin and are characterized by uncontrolled growth of cells of the myeloid compartment. The Philadelphia chromosome negative classical MPNs, including polycythemia vera, essential thrombocythemia, and myelofibrosis, are diseases of dysregulated JAK2 signaling. In fact, the majority of MPN patients have activating mutations in JAK2 (e.g JAK2-V617F), a tyrosine kinase that contributes to the growth and survival of myeloid cells. While MPNs were first described over sixty years ago, a significant need remains to develop therapeutic strategies for them. Inhibitors of JAK2 are currently being developed, and one inhibitor, ruxolitinib, was recently approved for certain MPN patients. Ruxolitinib has made profound impacts on improving splenomegaly and constitutional symptoms in MPN patients, but it and other JAK2 inhibitors have not significantly reduced the JAK2 mutant allele burden, and thus such inhibitors have not induced remission in these patients. The current consensus in the MPN field supports JAK inhibition for the treatment of patients, but a further understanding of MPNs and JAK2 signaling, as well as improved JAK2 inhibitors, may be necessary for treating MPN patients.
The work described in this dissertation has uncovered novel requirements for JAK2-V617F-driven signaling and transformation. We demonstrate that JAK2-V617F co-localizes with lipid rafts, cholesterol-rich microdomains within the plasma membrane that function to serve as platforms for signaling complex formation. Signaling complex formation is a necessary component for dysregulated signaling induced by JAK2-V617F. We provide evidence that cholesterol altering-lipid raft disrupting agents attenuate JAK2-V617F-driven signaling. We also show that cholesterol-lowering statins are effective at downregulating JAK2 signaling and inducing apoptosis in JAK2-V617F-driven cell lines. Importantly, we show that statins, inhibitors of the mevalonate pathway, inhibit the growth of primary MPN cells, while the same statin doses have no effect on healthy controls. Impressively, we demonstrate that statins cooperate with multiple JAK inhibitors, including ruxolitinib, to inhibit cell growth and induce apoptosis of JAK2-V617F-driven cells.
This report establishes statin-mediated inhibition of the mevalonate pathway as a potential approach to improve MPN therapeutics. We propose future studies with statins and JAK2 inhibitors in the treatment of MPNs.

The JAK/STAT pathway is a highly evolutionarily conserved signal transduction pathway, whose activation can lead to a broad range of cellular outcomes. The pathway is used repeatedly during multiple developmental stages and in adult tissue, and therefore tight regulation is required to enable accurate responses in a context specific manner. Internalisation and endocytic trafficking of signalling components provides a mechanism whereby spatial compartmentalisation can enable distinct signalling outputs. Within this study I have investigated the role of endocytosis in the regulation of the Drosophila melanogaster JAK/STAT pathway, and demonstrated that internalisation and endocytic trafficking differentially regulates target genes. Although the JAK/STAT pathway is transcriptionally competent and can regulate the expression of particular targets when the activated receptor is at the cell surface, receptor endocytosis and localisation to distinct endosomes is required for the expression of other targets. This appears to be context-dependent, as high levels of ligand stimulation overcomes endocytic regulation. STAT92E, the Drosophila JAK/STAT transcription factor, is a target of endocytic regulation. Although it is efficiently activated and undergoes nuclear translocation when endocytosis is perturbed, it is not capable of regulating a subset of target genes and therefore further STAT92E interacting partners and/or post translational modification must be required to fine-tune its transcriptional competency during endocytic trafficking. Utilising mass spectrometry I identified a novel STAT92E phosphorylation site, at threonine 702. Mutation of this threonine to prevent its phosphorylation, resulted in inhibition of STAT92E signalling and nuclear translocation, and also prevented phosphorylation of a highly conserved tyrosine residue at position 704, which is crucial for ligand activated JAK/STAT signalling outputs. Therefore, this study has enhanced our understanding of mechanisms that can modulate JAK/STAT activity. I have revealed an important role for endocytosis in fine- tuning Drosophila JAK/STAT signalling outputs and also identified a novel phosphorylation site which is crucial in the activity of STAT92E.

Each chapter focuses on one of these elements and defines British theatre in realation to the Czech scene. By comparing selected productions of the same British play in Prague and London the author draws attention to the possible difficulties that might arise while staging British drama in a different cultural context.

Les cavéoles sont des invaginations en forme de coupelle à la membrane plasmique. Ces organelles multifonctionnelles jouent entre autres, un rôle clé dans la mécano-protection et la signalisation cellulaire. En effet, les cavéoles ont la faculté de s’aplanir en réponse à l’augmentation de la tension membranaire, afin de protéger la cellule des contraintes mécaniques. Les cavéoles jouant un rôle clé dans la signalisation cellulaire, nous avions émis l’hypothèse que le cycle mécano-dépendent de désassemblage/réassemblage des cavéoles constitue un interrupteur mécanique de certaines voies de signalisation. Ce projet consiste à élucider le mécanisme moléculaire responsable du contrôle de la voie de signalisation JAK-STAT par la mécanique des cavéoles. Dans ces travaux, nous avons pu démontré que la cavéoline-1 (Cav1), un constitutant essentiel des cavéoles est libérée et devient hautement mobile au niveau de la membrane plasmique. Considérant les propriétés de signalisation de Cav1, Nous avons testé l’effet du désassemblage des cavéoles sur la signalisation cellulaire. Un criblage à haut débit, nous a permis identifié la voie de signalisation JAK- STAT stimulée par l’IFN-α comme voie modèle pour cette étude. En effet, la transduction du signal JAK-STAT induit par l’IFN-α est modulée par la mécanique des cavéoles. Afin de disséquer le mécanisme moléculaire responsable du contrôle de la signalisation JAK-STAT par la mécanique des cavéoles, nous avons déterminé le rôle de Cav1 dans ce contrôle. Nous avons observé que Cav1 est un régulateur négatif de la phosphorylation de STAT3 dépendante de la kinase JAK1. De plus, nous avons démontré que Cav1 interagit avec JAK1 en fonction de la tension membranaire. Nous avons également démontré que cette interaction Cav1-JAK1 fait intervenir le « scaffolding domain » de Cav1 (CSD), et que celui-ci est responsable de l’abolition de l’activité kinase de JAK1. Par conséquent, l’interaction de Cav1 avec JAK1 empêche l’activation de STAT3 par la kinase JAK1. These multifunctional organelles play a key role in cell mechanoprotection and cell signaling. Indeed our laboratory reported that caveolae have the ability to flatten out upon membrane tension increase, protecting cells from mechanical strains. Since caveolae play a key role in cell signaling we hypothesized that the mechano-dependent cycle of caveolae disassembly/reassembly may constitute a mechanical switch for signaling pathways. In this project, we elucidated the molecular mechanism underlying the control of JAK-STAT signaling by caveolae mechanics. We showed that caveolin-1 (Cav1), an essential caveolar component is released and become highly mobile at the plasma membrane under mechanical stress. Considering that caveolae are important signaling hubs at the plasma membrane, we addressed the effects of the mechanical release of Cav1 on cell signaling. Using high throughput screening, we identified the JAK-STAT signaling pathway as a candidate. To further dissect the molecular mechanism underlying the control of JAK-STAT signaling by caveolae mechanics, we addressed the role of Cav1 in the control of JAK-STAT signaling stimulated by IFN-α. We found that Cav1 was a specific negative regulator of the JAK1 dependent STAT3 phosphorylation. Furthermore, the level of Cav1 interaction with JAK1 depended on mechanical stress. We could show that Cav1-JAK1 interaction was mediated by the caveolin scaffolding domain (CSD), abolishing JAK1 kinase activity, hence, interfering with STAT3 activation upon IFN-α stimulation. Altogether our results show that caveolae are mechanosignaling organelles that disassemble under mechanical stress, releasing non-caveolar Cav1, which binds to the JAK1 kinase and inhibits its catalytic activity, preventing thereby JAK-STAT signal transduction

Ligand-activated signal transduction is a process critical to cell survival and function as it serves as a means of communication between the cells and their environment. Endocytosis is generally thought to down-regulate incoming signals by reducing the surface availability of receptors. However, increasing evidence in many systems suggests a notion which is referred to as the „signalling endosome" hypothesis - that endocytosis can also actively contribute to signalling apart from clearance of activated receptors and thereby attenuation of signalling. The functional aspect of signalling endosomes has been well-characterized in several pathways including RTK and TGF-β signalling. There are, however, various other signalling pathways where the active mechanism of endocytotic regulation is yet to be understood.
In this study, we probe this aspect in the cytokine signalling system, where the receptors are known to internalize but the significance of such internalization and precise mechanism is unclear. My thesis aims to elucidate the function and molecular details of internalization of cytokine receptor using interleukin-4 receptor (IL-4R) signalling as a model. IL-4 and IL-13 ligands can induce assembly of three distinct complexes: IL4 induced IL-4Rα – IL-2Rγ (type I), IL-4 induced IL-4Rα – IL-13Rα1 (type II) or the IL-13 induced IL-13Rα1-IL-4Rα (type II). The formation of any of these complexes triggers signalling through the JAK/STAT pathway. However, models of how the oligomerization of the transmembrane receptors and activation takes place are very diverse and lack a clear molecular and biophysical understanding of the underlying receptor dynamics.
Previous results of the lab had shown that the affinities between subunits are low, precluding complex formation at the plasma membrane at physiological concentrations. In addition, IL-4R subunits localize in to endosomal structures adjacent to the plasma membrane. It had already been shown that the shared IL-4R subunit IL-2Rγ is internalized by a specific, actin dependent, Rac1/Pak1 regulated endocytosis route in the IL-2 context. We could show that pharmacological suppression of this endocytosis pathway also prevented IL-4 induced JAK/STAT signalling, placing endocytosis upstream of signalling.
Here I show using immuno-EM techniques that these endosomal structures are multivesicular bodies. Importantly, I could show that receptor subunits are highly enriched in the limiting membrane of these endosomes relative to the adjacent plasma membrane. Using quantitative loading assays I could furthermore demonstrate that this enrichment is achieved by constitutive internalization of receptors from the cell surface into cortical endosomes. The trafficking kinetics of the receptor subunits is independent of ligand occupancy. Pharmacological inhibition shows that receptors and ligand traffic via the previously identified Rac1/Pak1 pathway. Finally, Vav2 was identified as a candidate Guanine Exchange Factor (GEF) that may regulate Rac1 activity and thereby control the actin polymerization cascade driving IL-4R endocytosis. Immunoprecipitations showed that Vav2 interacts both with the cytoplasmic tail region of the receptors and the receptor associated 2 kinase JAK3. Vav2 may thus couple the receptor/JAK complexes to the Rac1/Pak1 mediated endocytosis route.
Taken together, our results suggests that stable „signalling endosomes‟ adjacent to the plasma membrane act as enrichment centres, where ligand and receptor concentrations are locally increased by constitutive trafficking. The confined environment of the endosome then compensates for the weak affinities between the ligand and receptor and facilitates ligand-mediated receptor dimerization. Importantly, overexpression of both type II IL-4R subunits renders signal transduction resistant to endocytosis inhibition, strongly suggesting that the critical factor effecting signalling is sufficient concentration, which the endosomes facilitate achieving. The endosomes are thus dispensable as signalling scaffolds when the receptors are in sufficient concentration, where activated receptors could interact with downstream pathway components.
Endocytosis thus provides a crucial means for the signalling process to overcome the thermodynamic hurdles for receptor oligomerization. In conclusion, our data propose a novel, purely thermodynamic role of endosomes in regulating cytokine receptor signalling not seen in any other signalling pathway.

Syftet med uppsatsen har varit att få djupare förståelse för hur en räntefri bank arbetar och finansieras. Men även för att se vad skillnaden i kostnad blir med ett lån från den räntefria systemet JAK Medlemsbank i jämförelse med ett vanligt banklån. Studien utgick från en kvalitativ ansats. Vi använde oss av intervjuer som informationshämtande metod för att få djupare förståelse och kunskap om både JAK Medlemsbank och dess medlemmar. Den kvalitativa ansatsen gav oss också möjligheten att ta del av medlemmarnas uppfattning av ett räntefritt system och syn på räntan. Resultatet visade att stora lån blir opraktiska på JAK Medlemsbank då de månatliga utbetalningarna blir nästan dubbelt så höga, vilket många inte troligen klarar av att betala. Resultatet visade även att JAK Medlemsbank inte enbart finansieras av medlemsavgiften utan även av den ränta som de får från statsobligationer. Vilket tyckas vara rätt så paradoxalt, då syftet med JAK Medlemsbank är att motverka räntan.The purpose of this essay has been to get a deeper understanding of how a interest free bank works and gets financed. We also wanted to see the difference in the cost for a interest free loan compared to a traditional bank loan. The study was based on a qualitative approach. We used interviews as information retrieval method to get a deeper understanding and knowledge of both the JAK Members Bank and its members. The qualitative approach also gave us the opportunity to take part of the bank members perception of an interest-free system and their view on interest rates. The results showed that large loans becomes impractical at JAK Members Bank when the monthly payments almost was twice as high, which many are not likely able to pay. The results also showed that JAK Members Bank is not solely funded by membership fees, but also on the interest they receive from government bonds. Which could seem quite paradoxical, since the purpose of JAK Members Bank is to counteract the interest.

Na základě prostudovaných materiálů týkajících se zvolených rámců, metodik a standardů a legislativní norem jsem zvolila kritéria pro výběr vhodné kombinace pro implementaci. Struktura a obsah práce jednotlivých kapitol je navržena tak, aby čtenář postupně získával informace o IT governance, o obsahu a zaměření norem, standardů, metodik a legislativních rámců a o výběru rámce, metodik či standardů pro konkrétní oblast řízení informační bezpečnosti a uvedení případové studie z praxe. Závěrem práce je zjištění, že v současnosti neexistuje žádný dostatečně komplexní návod pro efektivní řízení podnikové informatiky. Naopak je vhodné využít správnou kombinaci těchto rámců, metodik a norem.

My final thesis "How informatics, science and technology affect our society" deals with an often ignored topic of the impact technology on our society. The thesis analyzes all possible positive and negative aspects of technological progress. The threats are described as social dependence on the machines and as well as the gradual moving out people from jobs. The existing system of capitalism below this technology cannot resist pressure and the pressure is a threat of his crash. The question remains to what kind of system should our civilization has to survive. Here come to the word famous futurologists, economists, sociologists, psychologists and architects. These groups of people are trying to sketch a rough draft feature of the future system. The threat of collapse of civilization can be averted. Unfortunately, each of them has their own vision and mostly of them work individually on their projects of new world. My task was to unify visions and to create a realistic vision of the whole society. The second part of my work describes the vision of these famous people and tries to find their mutual consistency. All of the visions have some deficiencies, or they are not well thought out in all aspects, so my other goal was to try to eliminate these defects or minimize them. The task for this thesis is to unite the ideas of contemporary visionaries and convince conservatives that it is not only possible to create a new social system, but rather also necessary.

The goal of this thesis is to clarify the status of books in the current Czech culture using scientific methods. Since there are many possible ways to achieve this widely defined goal, it was necessary to choose certain methods (and therefore an overall angle of the examination). Two specific methods were selected - a questionnaire for members of Generation Y and media analysis of culture sections of the Czech daily newspapers, Mladá fronta DNES and Právo during the months of June and December of 2017. Theoretical background, description of the methods and explanation of all the key terms are part of the theoretical section. The objective of the questionnaire is to map reading preferences and habits of young readers. The questions are focused on qualitative and also quantitative aspects of book reading. The overarching objective of the media analysis is to provide an accurate image the media paint about books. This will be achieved by thorough examination of all the articles that mention them. Just like with the questionnaire, qualitative and quantitative aspects will be tracked.

The subject of this thesis entitled What does an invisible city look like is the perception of a city and the way it is represented. The work is based on urban semiotics that sees urban environment as a particular system of communication. The methods of sensory urbanism and rhythm analysis are employed as well. The author asks whether the cities could be represented by other than visual symbols as it is usually the case for the products of tourism industry (guidebooks, souvenirs, etc.). Usually, the symbol of a particular city is formed by the image of its skyline. In Prague, this is the skyline of the Prague Castle, in Paris, it is the Eiffel tower, in New York, it is the Statue of Liberty, etc. But every city has definitely many other qualities. What are they? To find the answer, the author asks for help the visually impaired people. These people can tell easily what are the non-visual qualities of the urban space as they experience them necessarily in their everyday life and use them for better orientation. The author interviewed the visually impaired people and she also observed the way they read the city in real. The practical part of the thesis focuses on the Prague city centre.

This thesis focuses on the controversy in Czech society over the displacement of Germans. The dispute is one of the most significant public debates related to the Czech past held since the fall of the communist regime. The work briefly summarizes the historical development of the controversy over the displacement and introduces the political context of the discussion held already in the democratic regime. The main part of the text consists of analysis of opinions on the issues of displacement in Czech newspapers in the period of one year directly before the Czech Republic entered the European Union Concerning methodology, the work is inspired by discourse analysis. However, the author is not that much focusing on the use of language, which is common within this methodological tradition, but rather on what is being expressed through the language, i. e. the contents of the discourse itself. In doing so, he refers to the work of the Czech-British anthropologist Ladislav Holý. The author concludes that two main approaches may be found within the debate over the displacement, and calls the first of them the "official" discourse and the other the "critical" discourse. The main topic of this diploma thesis is comparison of two different methods of research. The first method is diary and the second method is online community. The subject of research in which these methods are compared with each other is the drinking regime of university students, more precisely the situation in which different drinks are consumed by university students. A total of twenty university students were involved in the research that is of a qualitative nature, with ten students in one research method. For one week, the students recorded not only all the drinks they drank per week, but also the circumstances of consuming these beverages, such as time of consumption, place, reasons for consumption etc. The diploma thesis outlines the differences between these research methods in terms of research results, frequency and quantity of information obtained, preparation and implementation of the methods, analysis of acquired data, as well as the perception of these methods by the respondents themselves. The resulting data analysis presents the benefits and limits of each of the selected methods in the situational research of the drinking regime and also suggests possible implications for further research.

In chapter four, I focus on the types of childcare after divorce and compare its advantages and disadvantages. After that I examine how the media, general public, and professionals view shared custody. The theoretical part ends with a chapter on specific situations in which court can decide for shared custody. That is followed by qualitative research on how shared custody is evaluated by professionals, such as teachers, carers, pedagogical assistants, social workers, workers of advisory centre and psychologists.

The Purpose of this writing is to address the issue of paedobaptism from the perspective of dogmatics. It contains an historical and confessional overview. It articulates the open issues and demonstarates, that so far no fundamentally balanced dogmatic reflection has been developed. Consequently, individual dogmatic views and positions have largely suffered from offering just partial perspectives. It seems critical to avoid such drawback while setting sound methodological background for such type of research. In the following sections an absence of a robust general methodology in the area of sacramentology is observed, an attempt to articulate key attributes of such metodology is presented, taking into account approaches taken so far. Such approaches are structurally analysed through metanarrative, research discipline, discours, argumentative logic. Finally, a practicable approach to deal with abundant available relevant theological material is proposed. Such an approach, if applied, might offer more holistic perspective on paedobaptism. This Master thesis deals with teaching of history of the Roma people in history lessons. The incorporation of Roma history to the curriculum is supported in the national Framework Program for Elementary Education, primarily through the cross-curricular theme of "Multicultural education". The theoretical part of this thesis consists of a brief summary of Roma history to the extent of what should be part of Elementary Education; history textbooks analysis from the perspective of Roma history; and list of other supportive educational materials. The practical part of this thesis presents a survey that was carried out within a small scale sample of secondary schools. Through the survey the current level of the 9th graders' knowledge about Roma history had been detected and subsequently a demonstrative lesson was delivered and evaluated.

The defining question is whether videogame journalists treat videogames as cultural or technical products thorough their work. The role and position of videogame journalism, games as art and aesthetics aspects of videogames are first studied in the theoretical section. These topics are further examined in practice using content analysis of chosen reviews which aims to investigate the means through which reviewers describe videogames in previously theoretically defined categories.

The aim of the interviews was to find out how the project "Come play hockey" was build and how works, including the recruitment event "Week of hockey", and for another to find out the possibilities of accepting a recruitment type of project in a volleyball environment in the Czech Republic. Zvolila jsem jiné výchozí materiály, brala jsem ohled hlavně na rozdílnou jazykovou situaci u nás.Zároveň jsem chtěla ukázat současný vývoj jazykové propedeutiky ve světě i posun ve vnímání této oblasti v České republice a první zaznamenané oficiální úspěchy v plánech Ministerstva školství mládeže a tělovýchovy.

Tento citát jsem zvolila záměrně, protože si myslím, že vystihuje problematiku, které se ve své diplomové práci věnuji. Kdykoliv otevřete jakoukoliv publikaci věnovanou problematice židovského národanebo vyhledáváte ve slovníku heslo "holocaust", narazíte na statistiku, která obecně zní: přibližně 6 miliónů Židů v Evropě bylo v období 2. světové války pronásledováno a nakonec systematicky vyvražděno nacistickým terorem v rozmezí let 1933 - 1945. Tyto informace jsou samozřejmě pravdivé, ale mám obavu, že jsou až příliš obecné na to, aby si jednotlivec, který je ve výchovně - vzdělávacím procesu uvědomil fakt, co to holocaust vlastně byl. Diplomová práce "Jak se žije dospělým dětem alkoholiků" se zabývá problematikou dospělých, kteří mají zkušenost dětství prožitého v rodině s rodičem-alkoholikem. Jedná se o kvalitativní studii, jejímž cílem je dopovědět na následující otázky: Jak vlastně vypadá dětství, dospívání a následně dospělý život lidí, kteří mají za sebou zkušenost dítěte alkoholika? Jak vypadá takové rodinné prostředí? Existují nějaká témata společná těm, kteří vyrůstali v takovém prostředí? Teoretická část obsahuje dosavadní poznatky studií, které se tématem dospělých dětí alkoholiků zabývají. Popisuje charakteristiky jak rodinného prostředí, tak způsoby uvažování a prožívání dospělých dětí alkoholiků. Metodologická část obsahuje definici výzkumného problému, metody získávání vzorku (samovýběr), metodu získávání dat (nestrukturované interview) a způsob jejich zpracování (narativní analýza, metody zachycení vzorců). Analytická část obsahuje samotné zpracování dat pěti respondentů. Jednotlivé příběhy nahlíženy jako určité životní konfigurace. Následně je uvedeno 28 témat, která jsou v různé míře respondentům výzkumu společná.
