Abstract:
Over the last decade there has been a substantial research effort directed toward understanding recidivism and risk prediction among adult sex offenders. In contrast, the juvenile sex offender field has received much less empirical attention. Consequently, there are very few risk assessment instruments designed for use with young persons who have committed sexual offences. Available measures do not appear to take into account the higher rates of non-sexual recidivism typical of this population. This thesis aimed to identify risk factors for sexual and non-sexual recidivism among juvenile sex offenders (JSOs) and explore their utility in the context of a risk assessment instrument. In Study One, a meta-analysis was conducted on JSO research to examine risk factors for sexual recidivism. The meta-analysis involved 17 studies and 22 predictor constructs. Seven variables emerged as reliable predictors of sexual recidivism: an index sexual offence involving a stranger victim, a history of physical abuse in the offender's background, the presence of sexual deviance, a non-contact index sexual offence, sexually offending against a greater number of victims, a history of sexual offending, and a history of non-sexual offending. In Study Two, a meta-analysis was performed on JSO research to identify risk factors for non-sexual recidivism. The meta-analysis involved 14 studies and 19 predictor constructs. Six variables emerged as reliable predictors of non-sexual recidivism: a history of non-sexual offending, a history of sexual abuse (negative relationship), the presence of an antisocial orientation, an index sexual offence involving a child victim (negative relationship), a history of physical abuse in the offender's background, and the use of threats/force in the index sexual offence. Taken together, the two meta-analyses were able to identify risk factors specific to sexual recidivism (e.g., sexual deviance) and non-sexual recidivism (e.g., antisocial orientation) as well as risk factors common to both outcomes (e.g., a history of non-sexual offending). These studies therefore made an original and significant contribution to the understanding of recidivism among JSOs. Based upon the meta-analyses, Study Three involved the construction of a risk assessment checklist that could be easily scored from file information. The Juvenile Risk Assessment Checklist (J-RAC) contained 11 items based upon the most consistent operationalisations of the predictors that emerged from the meta-analyses. The J-RAC is unique in the JSO risk assessment field as it contains two scales designed to provide separate estimates of the risk of youth engaging in further sexual and non-sexual recidivism, respectively. The J-RAC was scored based on file information from 82 youth who had been found guilty of sexual offences and referred to a treatment service in Queensland, Australia. The majority of the items possessed good inter-rater reliabilities and the two scales both possessed high inter-rater reliabilities. An existing JSO risk assessment instrument, the Juvenile Sex Offender Assessment Protocol Version 2 (J-SOAP-II; Prentky & Righthand, 2003), was also administered and the J-RAC possessed significant correlations with the majority of the J-SOAP-II scales. The J-RAC was also found to significantly discriminate between juveniles in detention and juveniles in the community, with those in detention found to be at higher risk on both scales. Thus, the reliability, convergent validity, and discriminant validity of the J-RAC was established. Consistent with current adult sex offender recidivism theory, the findings demonstrated the distinct roles of sexual deviance and antisocial orientation in the prediction of sexual and non-sexual recidivism of JSOs, respectively. The current thesis goes some way towards advancing a theory of juvenile sex offender recidivism by drawing upon the variables that emerged as reliable risk factors in the two meta-analyses. While the J-RAC could not yet be considered an actuarial instrument capable of assigning low, medium and high risk levels, the results of the research conducted in this thesis are encouraging. The utility of the J-RAC may be further enhanced through the aid of prospective research that incorporates substantial numbers of juvenile recidivists.

Abstract:
One of the notable features of JM Coetzee's work is the portrayal in his novels, particularly in his 1999 Booker Prize-winning novel, Disgrace, of worlds that often seem bleak and bereft of redemption. In this dissertation I will argue that there is a resonance in Coetzee's writing that goes beyond the political and historical and that adds another dimension to his work. The focus of my dissertation will be an attempt to account for this other 'dimension' within the broader socio-political-historical nexus that informs much of his work. I will argue that this other dimension represents an ongoing conccrn with the questions of redemption and salvation, particularly as they are manifested in his latcr works, Disgrace and Elizabeth Costello. These concerns, however, are not just a consequence of Coetzee's evolving thought. As I will show, they are present in his earlier texts as well. To this end, I will consider the ways in which the themes of redemption and salvation, in the specifically religious and Christian sense of these words, also resonate in In the Heart of the Country (1977) and Life and Times of Michael K (1983).

Abstract:
With the upcoming release of the fifth generation of cellular networks, 5G, it is expected to be a lot more connected devices. As a consequence, the importance of the capacity of the networks is increased, especially the radio access network (RAN), which will be changed a lot from previous generations. The best method to ensure a RAN has enough capacity for its targeted area is a simulation, and therefore, there is a demand for such a simulator. The development of a simulator for RAN started last year in another thesis with the possibility to model nodes, links, and set loads. In this thesis, the functionality is extended with the addition of mobility and user equipment (UE) to create load to more accurately simulate the network traffic. Roads are added as an entity to allow mobility, and cells are modeled to determine coverage. Most requested functionality was implemented, with the main difficulties being integrating the new code into the existing code base. The simulator was then evaluated, both the validity of the model and the performance. To improve the simulator for its purpose, even more, functionalities such as geography elements like buildings or mountains blocking the signal should be considered. Another improvement would be to make links and cells less reliable. The simulator is modeling the perfect day scenario at the moment which does not always reflect upon reality.

Abstract:
This essay explores how in the Harry Potter series, J. K. Rowling's magical heroes function asparadigms whose roles reflect on issues of race, prejudice and racism. Those issues include goodand evil, socialism and aristocracy, purity and impurity, freedom and indebtedness. This essayfocuses on showing how those themes are reflected and confronted in the dipole between HarryPotter and Draco Malfoy. Additionally, the Critical Race Theory, a theory that examines howculture uses and assorts power and race in society, is implemented to show how race andprejudice are reflected in the magical world. By further analyzing Harry and Draco's upbringingand social milieus in relation to the theme of good vs. evil, the development of these characters ispresented in response to their contrasting surroundings. The paper concludes that thesecharacters evolve in the final novels and make conscious choices to achieve the common causeof defying evil, despite their opposing backgrounds.

Abstract:
This study examines intersectionality and identity in rapper Jermaine Lamarr Cole’s studio album 4 Your Eyez Only. Through discourse analysis, the essay studies the representation of aspects of identity in the album, with a focus on gender, race, class and sexuality. The study also examines prosthetic memory in the album, with the intention of finding correlating narratives that create a general concensus of how society is viewed from marginalised groups in the US. The findings conclude that gender, race and class are expressed as the main buildingblocks for a characters identity in the album. It is telling that sexuality remains untouched in this regard. The reader is exclusively exposed to the normative heterosexual perspective, completely ignoring any notion of varied sexual orientations. Furthermore the findings conclude that J. Cole showcases a generally negative view on the govornment, as it is percieved as one of many instances keeping marginalised groups at the bottom of a societal foodchain

Abstract:
Ce travail a concerné l'étude des modifications post-traductionnelles des protéines Ras, et des effets de la déhydroépiandroséerone (DHEA) sur l'isoprénylation des protéines Ras. Le travail a été réalisé sur les cellules RTAC (RAS transformed adrenocortical) issues de la corticosurrénale de rat nouveau-né et transformées par l'oncogène HA-RAS#E#J. La protéine Ras doit impérativement se loger a la face interne de la membrane plasmique pour remplir sa fonction biologique. Cette translocation n'est possible qu'après isoprénylation du résidu cystéinyle situé près de l'extrémité carboxyle terminale de la protéine. Jusqu'à présent, il avait été admis que toutes les protéines Ras étaient farnesylées. Apres avoir démontré que les cellules RTAC surproduisent les protéines KI-RAS, nous avons montré que dans ces mêmes cellules les protéines KI-RAS sont géranylgéranylées en plus d'être farnésylees (Osman et coll. , biochem. Biophys. Res. Commun. , 231, 789-792, 1997). Ce résultat original est d'une importance fondamentale pour le développement de nouveaux inhibiteurs dirigés contre la géranylgéranyl transférase I qui pourraient être des anticancéreux potentiels et constitueraient une nouvelle voie thérapeutique pour le traitement des tumeurs dépendantes de l'oncogene KI-RAS. Dans la seconde partie de ce travail, nous avons montré que la DHEA, un stéroïde corticosurrénalien décrit pour avoir une action préventive contre la carcinogenèse chez le rat et la souris mais aussi chez l'homme, inhibe de manière réversible et dépendante de la dose la prolifération des cellules RTAC avec une CI#5#0 DE 50 M. Nous avons démontré que la DHEA n'induit pas l'apoptose dans les cellules RTAC, mais inhibe l'isoprénylation des protéines cellulaires et en particulier des protéines RAS. Le site d'inhibition se situe en aval de l'HMG-COA réductase, et vraisemblablement au niveau de la géranylgéranyl transférase I. Les résultats de ce travail ont été soumis à Journal of Biological Chemistry.

Abstract:
From the first generation, 1G, to the fourth generation, 4G, the development and technological advancements in telecommunications network systems have been remarkable. Faster and better connections have opened up for new markets, ideas and possibilities, to that extent that there now is a demand that surpasses the supply. Despite all these advancements made in the mobile communications field most of the concept of how the technology works and its infrastructure has remained the same. This however, is about to change with the introduction of the fifth generation (5G) mobile communication. With the introduction of 5G much of the technology introduced will be different from that of previous generations. This change extends to include the entire infrastructure of the mobile communications system. With these major changes, many of the tools available today for telecommunications network evaluation do not really suffice to include the 5G network standard. For this reason, there is a need to develop a new kind of tool that will be able to include the changes brought by this new network standard. In this thesis a simulation framework adapted for the next generation telecommunication standard 5G is set to be developed. This framework should include many of the characteristics that set 5G aside from previous generations.

Abstract:
This thesis is concerned with the evaluation, in-situ, of the elastic strain energy release rate of cracks. This can define the criteria for crack propagation, and it is usually necessary to obtain this via calculation from the geometry and applied load. A new method is proposed, based on the conjoint use of digital image correlation to measure full-field displacements and finite element to extract the strain energy release rate of surface cracks. It has been extended to 3-D datasets with the use of digital volume correlation and tomographic imaging. A finite element model with imported full-field displacements measured by DIC/DVC acting as boundary conditions is solved and the J-integral is calculated. For linear elastic materials, modal contributions can be separated via the interaction integral. The method has been benchmarked using synthetic datasets to assess its sensitivity to noise and experimental uncertainties. It is very robust to experimental noise and can be used without knowledge of the specimen geometry and applied loads. The application of the method in 2-D is demonstrated in an analysis of experimental data for a mode I fatigue crack, introduced to an aluminium alloy compact tension specimen. Analysis of mixed-mode cracks in 2-D is shown on a PMMA sample with the Arcan geometry. In 3-D, static loading of a fatigue crack in nodular graphite cast iron is studied and the results from the method are compared with those obtained via a field-fitting approach. Diffraction analysis of polycrystalline materials can determine the full tensor of the elastic strains within them. Maps of elastic strains can thus be obtained typically using synchrotron X-rays or neutrons. A method is presented to calculate the elastic strain energy release rate of a crack from 2-D diffraction strain maps. The diffraction data is processed via a finite element approach to obtain the parameters required to calculate the $J$-integral. A validation is presented using a synthetic dataset from a finite element model. Its experimental application is demonstrated in an analysis of synchrotron X-ray diffraction strain maps of a propagating fatigue crack in a bainitic steel, before and after an overload. Finally, a complex case study of stable fracture propagation in polygranular isotropic nuclear graphite is presented. Synchrotron X-ray tomography and strain mapping by diffraction were combined with DVC and image analysis to extract the full-field displacements and elastic crystal strains. The displacement fields have been analysed using the developed methods to extract the critical strain energy release rate for crack propagation. Non-linear properties described the effect of microcracking on the elastic modulus in the fracture process zone. The analysis was verified by comparison of the predicted and measured elastic strain fields.

This research deals with a central question in philosophy: the problem of foundation. Although the concept of philosophical foundation has undergone profound transformations throughout history, it including the loss of their metaphysical, ontological and theological references, the underlying question to the theme remains the same: the search for rational grounds legitimating of thought and action. After the "Copernican turn" in philosophy undertaken by Kant and the pragmatic-linguistic turn in contemporary philosophy the question of foundation can to express as way follows: what is the transcendental element that is the practical attitude of human? In order to answer this question we will concentrate on the analysis of the thought of two great philosophers of contemporary: Theodor Adorno and J?rgen Habermas. In the meantime, we are dedicated to also examine the concept of rationality of each of these authors, as well as its normative statement of proposals of ethics. We wonder what is the model of reason more apt to examine the normative nature of issues and gnostic of our time, marked on the one hand, the technical prestige and technological sciences, which causes an instrumental view prevails human, on the other hand, often by perniciosa- influenced the mass media, which together with the deficit in the educational and cultural level prevents the full realization of the concept of majority (M?ndigkeit) as telos of individual and collective life as the promised Enlightenment. Our thesis is that dialectical reason developed by Adorno presents itself as a more comprehensive and effective proposal to equate these and other issues of our historic time.
Esta pesquisa versa sobre uma quest?o central na filosofia: o problema da fundamenta??o. Ainda que o conceito de fundamenta??o filos?fica tenha sofrido profundas metamorfoses ao longo da hist?ria, incluindo a perda de suas refer?ncias metaf?sicas, ontol?gicas e teol?gicas, a querela subjacente ao tema continua a mesma: a busca das bases racionais legitimadoras do pensamento e da a??o. Tendo em vista a ?virada copernicana? na filosofia empreendida por Kant e a reviravolta pragm?tico-lingu?stica na filosofia contempor?nea, a quest?o da fundamenta??o deixa-se expressar nos seguintes termos: qual ? o elemento transcendental que reside na atitude pr?tica do homem? Com o objetivo de responder a essa pergunta nos deteremos na an?lise do pensamento de dois grandes fil?sofos da contemporaneidade: Theodor Adorno e J?rgen Habermas. Abstract:
The developmental processes of lymphopoiesis generate mature B lymphocytes from hematopoietic stem cells through increasingly restricted intermediates. Networks of transcription factors regulate these cell fate choices and are composed of both ubiquitously expressed and B lineage-specific factors. E-protein transcription factors are encoded by the three genes E2A, E2-2 (SEF2-1), and HEB. The E2A gene is required for B cell development and encodes the alternatively spliced proteins E12 and E47. During B lymphocyte development, the cells have to pass several checkpoints verifying the functionality of their antigen receptors. Early in the development, the expression of a pre-B cell receptor (pre-BCR) with membrane-bound immunoglobulin (Ig) heavy chain protein associated with surrogate light chain (SLC) proteins is a critical checkpoint that monitors for functional Ig heavy chain rearrangement. Signaling from the pre-BCR induces survival and a limited clonal expansion. Here it is shown that pre-BCR signaling rapidly down-regulates the SLCs l5 and VpreB and also the co-receptor CD19. Ca2+ signaling and E2A were shown to be essential for this regulation. E2A mutated in its binding site for the Ca2+ sensor protein calmodulin (CaM), and thus with CaM-resistant DNA binding, makes l5, VpreB and CD19 expression resistant to the inhibition following pre-BCR stimulation. Thus, Ca2+ down-regulates SLC and CD19 gene expression upon pre-BCR stimulation through inhibition of E2A by Ca2+/CaM. A general negative feedback regulation of the pre-BCR proteins as well as many co-receptors and proteins in signal pathways from the receptor was also shown. After the ordered recombination of Ig heavy chain gene segments, also Ig light chain gene segments are recombined together to create antibody diversity. The recombinations are orchestrated by the recombination activating gene (RAG) enzymes, other enzymes that cleave/mutate/assemble DNA of the Ig loci, and the transcription factor Pax5. A key feature of the immune system is the concept that one lymphocyte has only one antigen specificity that can be selected for or against. This requires that only one of the alleles of genes for Ig chains is made functional. The mechanism of this allelic exclusion has however been an enigma. Here pre-BCR signaling was shown to down-regulate several components of the recombination machinery including RAG1 and RAG2 through CaM inhibition of E2A. Furthermore, E2A, Pax5 and the RAGs were shown to be in a complex bound to key sequences on the IgH gene before pre-BCR stimulation and instead bound to CaM after this stimulation. Thus, the recombination complex is directly released through CaM inhibition of E2A. Upon encountering antigens, B cells must adapt to produce a highly specific and potent antibody response. Somatic hypermutation (SH), which introduces point mutations in the variable regions of Ig genes, can increase the affinity for antigen, and antibody effector functions can be altered by class switch recombination (CSR), which changes the expressed constant region exons. Activation-induced cytidine deaminase (AID) is the mutagenic antibody diversification enzyme that is essential for both SH and CSR. The AID enzyme has to be tightly controlled as it is a powerful mutagen. BCR signaling, which signals that good antibody affinity has been reached, was shown to inhibit AID gene expression through CaM inhibition of E2A.  SH increases the antigen binding strength by many orders of magnitude. Each round of SH leads to one or a few mutations, followed by selection for increased affinity. Thus, BCR signaling has to enable selection for successive improvements in antibodies (Ab) over an extremely broad range of affinities. Here the BCR is shown to be subject to general negative feedback regulation of the receptor proteins as well as many co-receptors and proteins in signal pathways from the receptor. Thus, the BCR can down-regulate itself to enable sensitive detection of successive improvements in antigen affinity. Furthermore, the feedback inhibition of the BCR signalosome and most of its protein, and most other gene regulations by BCR stimulation, is through inhibition of E2A by Ca2+/CaM. Differentiation to Ab-secreting plasmablasts and plasma cells is antigen-driven. The interaction of antigen with the membrane-bound Ab of the BCR is critical in determining which clones enter the plasma cell response. Genome-wide analysis showed that differentiation of B cells to Ab-secreting cell is induced by BCR stimulation through very fast regulatory events, and induction of IRF-4 and down-regulation of Pax5, Bcl-6, MITF, Ets-1, Fli-1 and Spi-B gene expressions were identified as immediate early events. Ca2+ signaling through CaM inhibition of E2A was essential for these rapid down-regulations of immediate early genes after BCR stimulation in initiation of plasma cell differentiation.

Apres une analyse des caracteres specifiques de ce type d'experience, les sequences d'impulsions pour la spectroscopie a deux dimensions in vivo ont ete adaptees pour optimiser le rapport signal/bruit aussi bien a l'acquisition que lors du traitement des donnees. L'importance de la connaissance des temps de relaxation t#2 a conduit a mesurer ces temps a l'aide de methode de type cpmg dans le champ tres inhomogene des bobines de surface. Parallelement, une etude des fluctuations du signal dues au mouvement macroscopique a conduit a chercher et experimenter des methodes de traitement du signal compensant en partie ces fluctuations, permettant ainsi d'ameliorer le rapport signal/bruit, de reduire le bruit en t#1 des experiences a deux dimensions et de mesurer des temps de relaxation t#2 avec plus de precision. Ces recherches methodologiques ont ete appliquees au cas de tumeurs intracerebrales induites chez le rat. Cette etude s'est etendue a un travail in vivo et ex vivo d'identification et de quantification des metabolites du tissu tumoral (gliome c6) par rapport au tissu cerebral sain chez le rat

La poliorcétique romaine connut apogée et suprématie sous le Haut Empire, dans un monde antique urbanisé où les guerres ne cessèrent jamais. Elle est bien davantage que le simple art de faire le siège d’une ville ou d’une place forte. Elle comprend tous les modes d’action, violents ou non. La guerre de siège et la défense en font également partie. Pragmatique, le général romain cherchait la conquête au moindre coût et n’utilisait la violence qu’en dernier lieu. L’assaut dans la foulée, très pratiqué, était d’une efficacité redoutable. Il évitait les désagréments d’un siège incertain, techniquement complexe, demandant une logistique considérable, dévoreur d’hommes, de temps et d’argent. Pour toutes ces raisons, le siège en règle était finalement assez peu fréquent. Une fois entrepris, il pouvait requérir, mais sans doute moins souvent qu’on ne le pense, des travaux lourds et complexes, exigeant de l’armée une division du travail poussée. Au combat, la complémentarité et l’efficacité du couple légion/auxiliaires n’a pas été assez soulignée, de même que l’importance du combat interarmes, incluant l’utilisation systématique des appuis, artillerie, archers et parfois frondeurs. L’assaut final semble avoir été assez rare et les dénouements alternatifs fréquents. Le traitement des vaincus dépendait de leur opiniâtreté. Sans pitié pour ceux qui leur résistaient, les Romains étaient assez pragmatiques pour les autres, en fonction des buts recherchés. Enfin, la prise d’une ville, était génératrice de richesses. Elle renforçait la légitimité impériale et le butin, matériel et humain, était un complément non négligeable pour le chef et le soldat
Roman siege warfare achieved its upmost efficiency and supremacy under the Early Empire in an antique world in which wars were permanent. Siege warfare is far more than the simple art of besieging cities or fortresses. It includes violent and non-violent courses of action. Siege wars and defense are part of it. Pragmatic, the roman general was eager to capture cities at low cost and use of violence came last. Storming assault was frequently employed and very effective. It avoided dangers of an uncertain siege, characterized by technical complexity, an important logistics, potentially high losses, time consuming and requiring huge financial resources. For all those reasons, methodical siege was not a very usual course of action. It sometimes meant, less frequently than expected, heavy siege works, requiring a demanding division of labor. During the battle, the complementarity and efficiency offered by the couple legion/auxiliaries hasn’t been stressed enough. Such is the importance of combined warfare, which systematically included the use of fire support, artillery, archeries and sometimes slingers. Regarding the capture, final assaults appear to have been rare. Alternative conclusions were most of the time the rule. Pitiless with resistant forces, Romans were rather pragmatic with others, depending on the final desired end state on the ground. When overrun, the city was generally a wealth provider. It concurred to strengthen the imperial legitimacy and, bounty, material and humans as well, was an important financial contribution for the general and the soldier

Ce travail a pour objectif de mettre en place des techniques de spectroscopie rmn du proton localisee, et de developper des techniques a deux dimensions frequentielles (2d), en vue d'etudier un modele de gliome intracerebral chez le rat. Une technique de localisation mono-voxel (press) a ete implementee afin de mesurer les temps de relaxation longitudinaux et transversaux des principaux metabolites visibles par rmn dans le cerveau de rat. L'implementation d'une sequence d'imagerie spectroscopique multi-tranches multi-echos a ensuite permis d'obtenir la distribution des principaux metabolites presents dans le cerveau de rat tumoral avec une resolution spatiale digitale d'environ 1l et dans un temps d'experience d'un peu plus d'une heure. Une seconde technique d'imagerie spectroscopique a permis d'obtenir des cartes du ph extracellulaire sur le meme modele animal. Ces cartes ont ete correlees avec la distribution de plusieurs metabolites detectes par rmn. Une sequence 2d j-resolu localisee a ete developpee en vue d'augmenter le nombre de metabolites identifiables par rapport a une technique 1d. Parallelement, une methode a ete proposee pour obtenir les cartes 2d sans suppression du signal de l'eau. Ces methodes ont permis d'etudier le signal des lipides mobiles presents dans les tumeurs intracerebrales chez le rat. Enfin, une sequence 2d j-resolu, spatialement resolue, utilisant les techniques d'apodisation a l'acquisition et multi-echos, a ete developpee. Des cartes 4d j-resolu ont ete obtenues sur le cerveau de rat tumoral, avec une resolution spatiale nominale de 12 l et dans un temps d'experience d'une heure environ.

ENGLSIH ABSTRACT: This study is a political biography of D.F. Malan (1874–1959), the first of the apartheid-era Prime Ministers, and covers the years 1874 to 1954, when Malan retired from politics. It endeavours to provide a warts-and-all account of D.F. Malan which challenges prevalent myths and stereotypes surrounding his public persona and his political orientation. While the overwhelming focus is on Malan’s political career, special attention is paid to his personal life in order to paint a multi-faceted picture of his character. The biography is written in the form of a seamless narrative and employs a literary style of writing. It is based on archival research which utilised Malan’s private collection, as well as the private collections of his Nationalist contemporaries. Malan takes the centre stage at all times, as the biography focuses on his perceptions and experiences. Malan’s views regarding Afrikaner nationalism, which was his foremost political priority, are described, and are related to his views of British imperialism as well as other ideologies such as communism and totalitarianism. This study demonstrates that there is a notable link between Malan’s perceptions of race relations and his concerns about the poor white problem. It reveals that Malan’s racial policy was, to some extent, fluid, as were his views on South Africa’s constitutional position. Debates about South Africa’s links to Britain and the nature of the envisioned republic preoccupied Afrikaner nationalists throughout the first half of the twentieth century – and served as an outlet for regional and generational tensions within the movement. Malan’s clashes with nationalists such as Tielman Roos, J.B.M. Hertzog and J.G. Strijdom are highlighted as an indication of the internecine power struggles within the National Party (NP). By emphasising these complexities, this study seeks to contribute to a nuanced understanding of the South African past.
AFRIKAANSE OPSOMMING: Hierdie studie is politieke biografie van D.F. Malan (1874–1959), die eerste van die apartheid-era Eerste Ministers, en dek die jare 1874 tot 1954, toe Malan uit die politiek getree het. Dit poog om onversuikerde beeld van Malan te skets wat heersende mites en stereotipes aangaande sy openbare beeld en sy benadering tot die politiek uitdaag. Die fokus is hoofsaaklik op Malan se politieke loopbaan, maar besondere aandag word aan sy private lewe geskenk om sodoende veelsydige portret van sy karakter te skilder. Die biografie is in die vorm van naatlose narratief geskryf en maak van literêre skryfstyl gebruik. Dit is gebaseer op argivale navorsing, waartydens daar van D.F. Malan se privaat versameling gebruik gemaak is, sowel as die privaat versamelings van sy tydgenote. Malan is ten alle tye die sentrale figuur en die biografie fokus op sy persepsies en ervarings. Enos John Nganani Mabuza's life and work is used as a case study to highlight the conceptual and methodological theories and challenges in academic biographical writing regarding history, memory and legacies. This thesis answers the question: Who was Mabuza and what is his place or relevance in South Africa's history? We over-simplify and stereotype people because it is convenient. We judge others but there is no saint without a past or a sinner without a future. Mabuza reconciled with the main liberation movements and moved from a maligned position on the political fringes to a respectable one within the emerging black economic elite. History, like reference systems and values is not calcified or static. It is prone to interpretation, adaptation, modification, invention, manipulation, decomposition and re-composition. Bourdieu's habitus-field analysis, theory or logic of practice, notions of capital (symbolic, political, social, cultural and economic) is used together with Latour's actor-network theory as the basis of analysis of the social contracts and trust bonds that Mabuza was able to create and which enabled him to navigate South Africa's socio-political and economic milieus during apartheid and the transition in the early 1990s. As people or actors, we believe in the mantra, nothing ventured, nothing gained. We exercise agency and take risks every day. We make choices and those choices have consequences. Mabuza's choices in the fields of education, politics and business had implications for how he is perceived or has been written into history. His choices put notions of identity, citizenship, power, legitimacy, ambition, elite accommodation, class, personal and professional networks, popular struggles, agency and structure under the spotlight. Mabuza's involvement in Bantustan politics, for instance, is contextualised in terms of a historical overview of the unpopular role played by traditional authorities in South Africa before and during colonialism and apartheid. His later foray into the world of business, however, was facilitated in part, by the personal and professional contacts that he made whilst he was in politics and the opportunities which opened up during the country's political transition. Mabuza adapted to changing circumstances and demonstrated a level of versatility which other Bantustan functionaries did not or could not exercise.

Advances in medical technology have altered the need for certain types of surgery to be performed in traditional inpatient hospital settings. Less invasive surgical procedures allow a growing number of medical treatments to take place on an outpatient basis. Hospitals face growing competition from ambulatory surgery centers (ASCs). The competitive threats posed by ASCs are important, given that inpatient surgery has been the cornerstone of hospital services for over a century. Additional research is needed to understand how surgical volume shifts between and within acute care general hospitals (ACGHs) and ASCs. This study investigates how medical technology within the hospital industry is changing medical services delivery. The main purposes of this study are to (1) test Clayton M. Christensen’s theory of disruptive innovation in health care, and (2) examine the effects of disruptive innovation on appendectomy, cholecystectomy, and bariatric surgery (ACBS) utilization. Disruptive innovation theory contends that advanced technology combined with innovative business models—located outside of traditional product markets or delivery systems—will produce simplified, quality products and services at lower costs with broader accessibility. Consequently, new markets will emerge, and conventional industry leaders will experience a loss of market share to “non-traditional” new entrants into the marketplace. The underlying assumption of this work is that ASCs (innovative business models) have adopted laparoscopy (innovative technology) and their unification has initiated disruptive innovation within the hospital industry. The disruptive effects have spawned shifts in surgical volumes from open to laparoscopic procedures, from inpatient to ambulatory settings, and from hospitals to ASCs. The research hypothesizes that: (1) there will be larger increases in the percentage of laparoscopic ACBS performed than open ACBS procedures; (2) ambulatory ACBS will experience larger percent increases than inpatient ACBS procedures; and (3) ASCs will experience larger percent increases than ACGHs. The study tracks the utilization of open, laparoscopic, inpatient and ambulatory ACBS. The research questions that guide the inquiry are: 1. How has ACBS utilization changed over this time? 2. Do ACGHs and ASCs differ in the utilization of ACBS? 3. How do states differ in the utilization of ACBS? 4. Do study findings support disruptive innovation theory in the hospital industry? The quantitative study employs a panel design using hospital discharge data from 2004 and 2009. The unit of analysis is the facility. The sampling frame is comprised of ACGHs and ASCs in Florida and Wisconsin. The study employs exploratory and confirmatory data analysis. This work finds that disruptive innovation theory is an effective model for assessing the hospital industry. The model provides a useful framework for analyzing the interplay between ACGHs and ASCs. While study findings did not support the stated hypotheses, the impact of government interventions into the competitive marketplace supports the claims of disruptive innovation theory. Regulations that intervened in the hospital industry facilitated interactions between ASCs and ACGHs, reducing the number of ASCs performing ACBS and altering the trajectory of ACBS volume by shifting surgeries from ASCs to ACGHs.

Restenosis or neointimal hyperplasia is still a clinical limitation of percutaneous coronary angioplasty (PTCA) for treatment of arterial obstruction. Abnormal proliferation and migration of vascular smooth muscle cells (VSMCs) have been demonstrated to be involved in the development of restenosis. The purpose of the present study was to investigate whether Ugonin J, a bioactive compound isolated from Helminthostachys zeylanica (L) Hook. (Helminthostachyaceae), has potential effects to prevent neointimal formation, and to explore its molecular mechanisms. Our current findings demonstrated that at 12, 24 and 36 uM Ugonin J markedly inhibited cell growth and migration of VSMCs (A7r5) by using MTT assay, wound scratch assay and transwell assay. Cytometric analysis suggested that Ugonin J was not siginificantly regulating cell cycle at 12 and 24 uM, but arrested VSMCs in the G1 phase at 36 uM. Zymography assay showed that Ugonin J was able to suppress the activities of matrix metalloproteinase (MMP)-2 and MMP-9 in a concentration-dependent manner. Western blot results demonstrated that treatment with Ugonin J decreased the expression levels of p-AKT, p-ERK1/2, p-FAK, ERP57, PGK1, PI3K and MMP-2 and MMP-9, whereas the total protein levels of AKT, ERK1/2 and FAK weren’t siginificantly changed. The pharmacological effects of Ugonin J in preventing balloon injury-induced neointimal hyperplasia and migration were evident in the rat model of carotid artery balloon angioplasty. Our data demonstrated that Ugonin J can be a potential regulator to inhibit cell migration and neointimal formation.

The recombination-activating gene products, RAG1 and RAG2, initiate V(D)J recombination during lymphocyte development by cleaving DNA adjacent to conserved recombination signal sequences (RSSs). The reaction involves DNA binding, synapsis, and cleavage at two RSSs located on the same DNA molecule and results in the assembly of antigen receptor genes. Since their discovery full-length, RAG1 and RAG2 have been difficult to purify, and core derivatives are shown to be most active when purified from adherent 293-T cells. However, the protein yield from adherent 293-T cells is limited. Here we develop a human suspension cell purification and change the expression vector to boost RAG production 6-fold. We use these purified RAG proteins to investigate V(D)J recombination on a mechanistic single molecule level. As a result, we are able to measure the binding statistics (dwell times and binding energies) of the initial RAG binding events with or without its co-factor high mobility group box protein 1 (HMGB1), and to characterize synapse formation at the single-molecule level yielding insights into the distribution of dwell times in the paired complex and the propensity for cleavage upon forming the synapse. We then go on to investigate HMGB1 further by measuring it compact single DNA molecules. We observed concentration dependent DNA compaction, differential DNA compaction depending on the divalent cation type, and found that at a particular HMGB1 concentration the percentage of DNA compacted is conserved across DNA lengths. Lastly, we investigate another HMGB protein called TFAM, which is essential for packaging the mitochondrial genome. We present crystal structures of TFAM bound to the heavy strand promoter 1 (HSP1) and to nonspecific DNA. We show TFAM dimerization is dispensable for DNA bending and transcriptional activation, but is required for mtDNA compaction. We propose that TFAM dimerization enhances mtDNA compaction by promoting looping of mtDNA.

For years Black women’s subjectivity in the use of their bodies and movements has been overshadowed or completely erased by dominant hegemonic systems that created its own narrative of Black women, their bodies, and their movement. This thesis works to acknowledge and analyze the dialogic relationship among the narratives of Black women, Black women’s performances of their “theories of the flesh” through dance as well as their everyday activities, and the race, gender, and class conditions that inform said “theories of the flesh.” During football season, everyone in the African-American community of Jackson, Mississippi is looking at and talking about the dance company, the Prancing JSettes. There are audience members who critique their movements and costumes and there are those who view the group as a vital part of the community. Either way every audience member is captivated by the J-Settes because their cultural history is depicted by the women’s performance. How does this work? How is the Prancing J-Sette image constructed and by whom, and why and how does it persist? These are the questions I ask to examine the gender, class, and racial relations that are inscribed upon the movements of Black women in the African Diaspora. For a group whose African ancestors viewed dance as very spiritual, with such activities as the ring shout, it is interesting to note the ambivalence that surrounds the public dancing body in Jackson, Mississippi. While some Jacksonians view the female body in the public sphere with a Protestant Christian lens, they also enjoy the Africana aesthetics and aggressive energy of the J-Settes’ performances. Also, while the J-Settes buck their society’s hegemonic system of propriety, they also comply with some of these standards in their performance. I examine this ambivalence through the discourses of critical race theory, Black feminism, the social significance of African Diaspora dance conventions and HBCUs, and the classed, racial, and gendered power relations in the African Diaspora. I argue that the stories about the Prancing J-Settes can be expanded to present a genealogy and present state of contradictory values and issues of visibility affecting all Black women.

Between 1985 and 1997 a hip hop culture emerged in Halifax, Nova Scotia through rap music artists' live performances at various venues, and by releasing original music on commercial and non-commercial cassette tapes and compact discs. This thesis examines the evolution of this grassroots musical culture through the lenses of Halifax's geography, innovative musical and technological trends, ever-present racial politics, and a strong "do-it-yourself" ethic. This thesis argues that hip hop in Halifax during these years can be divided into two eras distinguished by dynamic racial and stylistic changes. While the 1980s saw a predominantly Black hip hop community take root around Uniacke Square and Gottingen Street, the 1990s saw a geographic and demographic shift as the rap music scene expanded and competed with the mainstream music scene of the city. In doing so, the integrated downtown hip hop community produced a significant amount of work, overcoming institutional opposition

In eutherian mammals, the size of the testes and number of spermatozoa produced and stored in the excurrent ducts vary widely between species, with the hydromyine rodents of Australia exhibiting a greater range of interspecific variation than any other closely related group of species. This study compared the efficiency of germ cell production and sperm storage capacity in the extra-testicular ducts of two arid zone species, the plains rat, Pseudomys australis, and the spinifex hopping mouse, Notomys alexis, that have vast differences in testes size and number of stored spermatozoa. Results are discussed.

“The Transgressive Stage: The Culture of Public Entertainment in Late Victorian Toronto,” argues that public entertainment was one of the most important sites for the negotiation of identities in late Victorian Toronto. From the vantage point of the twenty-first century, where theatre is strictly highbrow, it is difficult to appreciate the centrality of public entertainment to everyday life in the nineteenth century. Simply put, the Victorian imagination was populated by melodrama and minstrelsy, Shakespeare and circuses. Studying the responses to these entertainments, greatly expands our understanding of Victorian culture. The central argument of this dissertation is that public entertainment spilled over the threshold of the playhouse and circus tent to influence the wider world. In so doing, it radically altered the urban streetscape, interacted with political ideology, promoted trends in consumption, as well as exposed audiences to new intellectual currents about art and beauty. Specifically, this study examines the moral panic surrounding indecent theatrical advertisements; the use by political playwrights of tropes from public entertainment as a vehicle for political satire; the role of the stage in providing an outlet for Toronto’s racial curiosity; the centrality of commercial amusements in defining the boundaries of gender; and, finally, the importance of the theatre—particularly through the Aesthetic Movement—in attempts to control the city’s working class. When Torontonians took in a play, they were also exposing themselves to one of the most significant transnational forces of the nineteenth century. British and American shows, which made up the bulk of what was on offer in the city, brought with them British and American perspectives. The latest plays from London and New York made their way to the city within months, and sometimes weeks, of their first production. These entertainments introduced audiences to the latest thoughts, fashion, slang and trends. They also confronted playgoers with issues that might, on the surface seem foreign and irrelevant. Nevertheless, they quickly adapted to the environment north of the border. Public entertainment in Toronto came to embody a hybridized culture with a promiscuous co-mingling of high and low and of British and American influences.

Objects of Memory is a master's dissertation in Multimedia Art with a theoreticalpractical nature, which we analyze the concept of memory associated with the postcard’s object. Starting from the aggregation of dichotomous concepts such as memory / myth and Gestalt / narrative unit, we explore relationships based in a construction of the visuality having as resource the static image and the moving image, which intersection is made from the photography with the video. For an interpretation of the postcard and its artistic faculties as an object, we evoke the Mail Art approaching the use of postcards by the Fluxus Group and by artists such as Robert Filliou and Ray Johnson, who in the late 1950s used postcards as artistic objects to communicate with other artists around the world. Gathering the works of Alain Resnais, Chris Marker, Marcel Broodthaers, Orhan Pamuk and Óscar Muñoz, we observed possible relations with the concept of memory within the scope of the project, specifically the work methodology inserted in narrative and visual discourse. For the Objects of Memory final project, we have been developed three videos - Eraser River, Vanishing and I Remember - each one representing a postcard to be projected through a device. From the intersection of images we explored a working methodology that allowed to create visual events with the purpose of transforming theperception of the postcard as a whole. For this, we connected static images (photography) with images in motion (video) through the process of cut, collage and assembly to ensure a logical and perceptive speech in the body’s postcard. Although independent, each one of these videos tries to express the concept of object of memory preserving, in this way, a relation with images of the past which appear analogous to the transformation of a still image into a moving image

Geref. Kerk beskou word. Die meeste van sy standpunte word vandag as waarheid aanvaar en hy het bygedra tot die intellektuele debat oor die Bybel as Woord van God wat vandag nog aangaan. Du Plessis se herkoms, opleiding en verdienste as predikant en saakgelastigde van die Kerk word kortliks bespreek. Hy was die talentvolle seun en enigste kind van 'n Afrikaanse predikant en 'n Engelse vrou, die dogter van 'n sendeling van die LMS. Hy word professor aan die Kweekskool en met kennis opgedoen tydens verdere studie oorsee, het hy gewys op dele van die Bybel wat nie aan goddelike inspirasie toegeskryf kan word nie. In die studie van bydraes van die drie geleerdes tot ons verstaan van die Bybel, is ook aandag geskenk aan 'n ander aspek van die Bybel as problematiese teks: Die nadruk wat skrywers en redaktors van die Bybelse gebeure, geplaas het op mans as vaders en seuns ten koste van vroue, moeders en dogters. Die gevolg was dat meeste van die sogenaamde Kerkvaders genoeg in die Bybel kon vind om hulle te identifiseer met die neerhalende beeld wat in Die Openbaring (14:3-4) geskep word van vroue, opvallend in stryd met die inhoud van Genesis 1:26-31.
The controversy started by Johannes Du Plessis over the question whether everything in the Bible was inspired by God, caused a crisis in the Dutch Reformed Church, which is listed as a beacon in its history. Most of the points he raised are accepted as truth today, and started an intellectual debate that is still going on. Information relating to Du Plessis’ origins, training, and achievements as a minister and church official is briefly noted. He was the talented son, and only child of an Afrikaner father and minister of religion, and an English mother, the daughter of an LMS missionary. He became a professor at the Seminary in Stellenbosch, after his studies overseas had caused him to question parts of the Old Testament text that could not be regarded as divinely inspired. He also made it quite clear that believing without trying to understand was not possible for him as a theologian and teacher. He proclaimed his findings in Het Zoeklicht, a monthly magazine intended to serve as a searchlight and an organ to reveal the “darkness” that was still prevalent in both religion and politics. Aware of the value of Higher Criticism of the Old Testament, he regarded it his duty to make known what he had found in reading widely and in critical research on the origins, form and content of the books of the Bible. Articles to this effect published first in De Kerkbode and then in Het Zoeklicht, caused an outcry from conservative readers who were opposed to any changes in the teaching of the church. They were especially enraged by his view that the confessions drawn up by Reformers 500 years earlier, were open for review. Complaints that Du Plessis was on a dangerous path, marked the beginning of what was regarded as a crisis in the Church, and in the end led to Du Plessis being relieved of his post by a special Synod convened in 1930. The views of the conservative majority, however, prevailed. As a result, a large number of leading ministers and theologians, who openly supported the Nationalist government that came into power in 1948, provided assurance that the concept of separation of racial groups was in accordance with Scripture. After nearly forty years of an uncritical approach to the Bible, the belief in in-errancy was again questioned. Among the few theologians who dared to point out errors and inconsistencies in various texts, were Ferdinand Deist and Willem Vorster. Both trained as ministers, they chose to continue their studies and openly expressed themselves against the abuse of Biblical texts to support the ideology of apartheid. Trained at the Seminary in Stellenbosch, Deist turned out to be a noted scholar who combined his urge for honest critical study with a pietism expressed in a flood of academic and popular publications ― which were not always consistent with his own statement that The Word of God could not be connected to Scripture with an is-equal-to symbol. Vorster, who received his education at the University of Pretoria, became equally well known as a scholar and gained international recognition for his application of semantics in the study of the Gospels. He also applied his knowledge of languages in what came to be known as The New Quest for the historical Jesus who, as a Jew and a Galilean, distinguished himself during a brief period as a teacher and eschatological prophet. In the study of the contributions of these three outstanding men to our understanding of the Bible, another major aspect of the Bible as problematic text is addressed: The emphasis the authors and redactors of the Scriptures placed on the roles of men, fathers and sons, patently to the exclusion of women, mothers and daughters. As a result many of the so-called Church Fathers found in the Bible sufficient material to look down upon women ― the verdict expressed in Revelations 14:3-4 being one of numerous texts pointing to women as objects of derision in glaring contradiction to the contents of Genesis 1:26-31.
