Abstract:
We reported previously that Ro-318220 blocked expression of mitogen-activated protein kinase phosphatase-1 (MKP-1) induced by tumor necrosis factor-α (TNF-α) and subsequently caused apopotosis in mesangial cells (Y.-L. Guo, B. Kang, and J. R. Williamson. J. Biol. Chem. 273: 10362–10366, 1998). These data support our hypothesis that a TNF-α-inducible phosphatase may be responsible for preventing sustained activation of c-Jun NH2-terminal protein kinase (JNK) and consequent cell death in these cells (Y.-L. Guo, K. Baysal, B. Kang, L.-J. Yang, and J. R. Williamson. J. Biol. Chem. 273: 4027–4034, 1998). In this study, we investigated the involvement of protein kinase C (PKC) in regulation of MKP-1 expression in mesangial cells together with effects on viability. Although originally characterized as a PKC inhibitor, Ro-318220 inhibited TNF-α-induced MKP-1 expression through a mechanism other than blocking the PKC pathway. Furthermore, inhibition of the PKC pathway neither significantly affected TNF-α-induced MKP-1 expression nor made cells susceptible to toxic effect of TNF-α. Thus PKC activation is not essential for cells to achieve the resistance to TNF-α cytotoxicity displayed by normal mesangial cells. However, activation of PKC by phorbol 12-myristate 13-acetate (PMA) dramatically increased cellular resistance to the apoptotic effect of TNF-α. Coincidentally, PMA stimulated MKP-1 expression and suppressed JNK activation. Therefore, PMA-induced MKP-1 expression may contribute to the protective effect of PMA. These results provide a mechanistic explanation for previous documentation that PKC activation can rescue some cells from apopotosis.

1. We have purified membrane-associated Ins(1,4,5)P3/Ins(1,3,4,5)P4 5-phosphatases from bovine testis and human erythrocytes by chromatography on several media, including a novel 2,3-bisphosphoglycerate affinity column. 2. The enzymes have apparent molecular masses of 42 kDa (testis) and 70 kDa (erythrocyte), as determined by SDS/PAGE, and affinities for Ins(1,4,5)P3 of 14 microM and 22 microM respectively. 3. The two enzymes hydrolyse both Ins(1,4,5)P3 and Ins(1,3,4,5)P4 and are therefore type I Ins(1,4,5)P3 5-phosphatases [nomenclature of Hansen, Johanson, Williamson and Williamson (1987) J. Biol. Chem. 262, 17319-17326]. 4. On chromatofocusing, the partially purified testicular enzyme migrates as two peaks of activity, with pI values of about 5.8 and 5.5. The erythrocyte enzyme exhibits only the latter peak. 5. The testis 5-phosphatase is labile at 37 degrees C, but its activity can be maintained in the presence of 50 mM phorbol dibutyrate (PdBu). After PdBu treatment, a third form of the enzyme, with pI about 6.2, appears on chromatofocusing, but without change in its Km or Vmax. 6. Consideration of the properties of these enzymes and of the 5-phosphatases from other tissues suggests that type I Ins(1,4,5)P3 5-phosphatases are of two well-defined subtypes. We propose that these be termed type Ia [typified by the testis enzyme: approximately 40 kDa, higher affinity for Ins(1,4,5)P3] and Type Ib [typified by the erythrocyte enzyme: approximately 70 kDa, lower affinity for Ins(1,4,5)P3].

The article presents a laser dazzle generating three overlapping laser beams of different colours and an analysis of the possibility of dazzling people with this radiation. The analysis took into account the eye’s photopic sensitivity and the additivity of the three radiation beams. Such analysis, to the knowledge of the authors, is presented for the first time. The possibility of dazzling for four different dazzle levels and three levels of ambient luminance was presented. It has been shown that in some conditions it is possible to damage the eye without causing the assumed dazzle effect. Targeted deletion of mXinα results in cardiac hypertrophy and cardiomyopathy with conduction defects (Gustafson-Wagner, E., Sinn, H. W., Chen, Y.-L., Wang, D.-Z., Reiter, R. S., Lin, J. L.-C., Yang, B., Williamson, R. A., Chen, J. N., Lin, C.-I., and Lin, J. J.-C. (2007) Am. J. Physiol. 293, H2680-H2692). To understand the underlying mechanisms leading to such cardiac defects, the functional domains of mXinα and its interacting proteins were investigated. Interaction studies using co-immunoprecipitation, pull-down, and yeast two-hybrid assays revealed that mXinα directly interacts with β-catenin. The β-catenin-binding site on mXinα was mapped to amino acids 535-636, which overlaps with the known actin-binding domains composed of the Xin repeats. The overlapping nature of these domains provides insight into the molecular mechanism for mXinα localization and function. Purified recombinant glutathione S-transferase- or His-tagged mXinα proteins are capable of binding and bundling actin filaments, as determined by co-sedimentation and electron microscopic studies. The binding to actin was saturated at an approximate stoichiometry of nine actin monomers to one mXinα. A stronger interaction was observed between mXinα C-terminal deletion and actin as compared with the interaction between full-length mXinα and actin. Furthermore, force expression of green fluorescent protein fused to an mXinα C-terminal deletion in cultured cells showed greater stress fiber localization compared with force-expressed GFP-mXinα. These results suggest a model whereby the C terminus of mXinα may prevent the full-length molecule from binding to actin, until the β-catenin-binding domain is occupied by β-catenin. The binding of mXinα to β-catenin at the adherens junction would then facilitate actin binding. In support of this model, we found that the actin binding and bundling activity of mXinα was enhanced in the presence of β-catenin.

À partir des mises en scène de L’Orestie d’Eschyle, d’Œdipe roi de Sophocle et des Bacchantes d’Euripide diffusées dans les institutions nationales en Allemagne, en France et au Royaume-Uni entre 1973 et 2010, la thèse procède à une archéologie du commun, en explorant, d’une part, le concept de commun, et en particulier ses enjeux politiques, à travers une analyse des mises en scène contemporaines du chœur tragique et en étudiant, d’autre part, ces mises en scène à travers l’expression du commun. Ce travail propose donc de mettre au jour la construction et la circulation du discours sur le commun dans et entre ces trois pays. L’analyse des spectacles, d’abord, expose les éléments qui font ou entendent faire du chœur une incarnation du commun et met en perspective ces choix avec la réception de la tragédie grecque. Le discours sur le commun qui se construit ainsi au théâtre est ensuite confronté aux discours philosophiques et anthropologiques du moment mais aussi aux événements économiques, politiques et sociaux afin de faire apparaître les échos, les analogies, les ruptures et les discontinuités. Ainsi, entre 1973 et 1980, la mise en scène du chœur des Bacchantes a donné du commun une représentation utopiste où la communauté est fondée par le rituel. Dès 1980, à partir des Orestie de Peter Stein et Peter Hall qui tiennent lieu de modèles, le chœur devient un collectif où ce que les individus ont en commun est précisément leur singularité. Dans la continuité, jusqu’en 1999, les mises en scène d’Œdipe roi racontent la naissance de l’individu moderne à laquelle le chœur sert de cadre archaïque. Enfin, et malgré les tentatives dans des mises en scène de L’Orestie, au tournant du millénaire, pour refonder la communauté à partir d’une mémoire commune, les tragédies grecques montées dans les années 2000 présentent un désespoir de communautés – au double sens objectif et subjectif de l’expression. Analysing productions of Aeschylus’ The Oresteia, Sophocles’ Oedipus the King and Euripides’ The Bacchai in national theatres in France, Germany and the United-Kingdom between 1973 and 2010, this thesis proposes an archaeology of the common (in the sense of « what we have in common ») both exploring the political implications of the concept – thrown into sharp relief by the various ways ancient choruses were staged – and studying the productions themselves through the type of community that they make manifest. This work intends to highlight the construction and the circulation of contemporary discourses about the common within, and between, these three countries. Performance analyses first focus on the elements that make, or intend to make, the chorus into an incarnation of the common and put these choices into perspective through the reception of Greek tragedy. The discourse about the common thus built in theatres, is then confronted with philosophical and anthropological discourses, as well as with economic, political and sociological events in order to call attention to echoes, analogies, disruptions and discontinuities. Thus, between 1973 and 1980, performances of choruses in The Bacchai were built upon rituals, putting forward a utopian conception of the common. From 1980 onward, as Peter Stein’s and Peter Hall’s Oresteia became established models, the chorus morphed into a collective in which individuals had their singularity in common. Following this, until 1999, the performances of Oedipus the King hailed the birth of the modern individual, for whom the chorus acts as archaic backdrop. Lastly, and despite attempts in performances of The Oresteia at the turn of the millennium to rebuild a community out of common memory, Greek tragedies staged in the 2000s show the despair of, and about, communities. This archaeology of the common, reflecting the globalisation of European societies, is therefore indirectly an archaeology of the individual

Abstract:
Ever since the speculations of the Victorians about the inexorable progress of Man from the savagery of foraging to agriculture and civilization, Europe has been one of the main theatres of debate about transitions from foraging to farming (Chapter 1). The dominant model in the twentieth century, first developed explicitly by Gordon Childe in The Dawn of European Civilization (1925) and The Danube in Prehistory (1929), has been that of ex oriente lux, ‘light from the Near East’. According to this theory, farming began in Europe because it was introduced by Neolithic farmers from South-West Asia, who brought with them domesticated plants and animals together with a new technology that included pottery and polished stone tools. They colonized a land thinly occupied by Mesolithic foragers except at the coastal margins. In southern Europe, the first farmers would have ‘taken to their boats and paddled or sailed on the alluring waters of the Mediterranean to the next landfall—and the next’ (Childe, 1957: 16). In temperate Europe, expansion was facilitated by ‘slash-and-burn’ (swidden) agriculture practised by the first farmers: they arrived at a particular location, cleared the forest, burnt the cut timber, and planted their crops, and then moved on after a few years. The first suite of 14C dates from European Neolithic sites obtained in the 1960s astonished archaeologists, because the (uncalibrated) dates of c.6000 bc from Greek Neolithic settlements such as Nea Nikomedeia and Knossos (Fig. 9.1) were 3,000 years older than Childe’s suggested date for the beginning of the European Neolithic: c.3000 BC. He established the latter by an elaborate process of cross-dating European prehistoric sites with historically dated cultures in the eastern Mediterranean, in turn dated by links to Pharaonic Egypt. At the same time, the 14C data appeared to confirm Childe’s ex oriente lux theory, because there was a clear trend of increasingly younger dates with distance from South-West Asia (J. G. D. Clark, 1965; Fig. 1.7). The dates of c .6000 BC in south-east Europe were in the same time-frame as dates for PPNB Neolithic settlements in South-West Asia, dates in central Europe and the Mediterranean were of the order of 4500 BC, and dates from Early Neolithic sites on the Atlantic margins of Europe were nearer 3000 BC.

Purpose/Background: Adverse Childhood Experiences (ACEs) have an impact on health throughout the lifespan (Filletti et al., 1999; Hughes et al., 2017). These experiences range from physical and mental abuse, substance abuse in the home, parental separation or loss, financial instability, acute illness or injury, witnessing violence in the home or community, and incarceration of family members (Hughes et al., 2017). Understanding and screening for ACEs in children with urinary incontinence can help practitioners identify psychological stress as a potentially modifiable risk factor. Methods: A 5-month chart review was performed identifying English speaking patients ages 6-11 years presenting to the outpatient urology office for an initial visit with a primary diagnosis of urinary incontinence. Charts were reviewed for documentation of individual or family risk factors for ACEs exposure, community risk factors for ACEs exposures, and records where no related documentation was included. Results: For the thirty-nine patients identified, no community risk factors were noted in the charts. Seventy-nine percent of patients had one or more individual or family risk factors documented.
