We have isolated and sequenced genes from Saccharomyces cerevisiae (SRP54SC) and Schizosaccharomyces pombe (SRP54sp) encoding proteins homologous to both the 54-kD protein subunit (SRP54mam) of the mammalian signal recognition particle (SRP) and the product of a gene of unknown function in Escherichia coli, ffh (Römisch, K., J. Webb, J. Herz, S. Prehn, R. Frank, M. Vingron, and B. Dobberstein. 1989. Nature (Lond.). 340:478-482; Bernstein H. D., M. A. Poritz, K. Strub, P. J. Hoben, S. Brenner, P. Walter. 1989. Nature (Lond.). 340:482-486). To accomplish this we took advantage of short stretches of conserved sequence between ffh and SRP54mam and used the polymerase chain reaction (PCR) to amplify fragments of the homologous yeast genes. The DNA sequences predict proteins for SRP54sc and SRP54sp that are 47% and 52% identical to SRP54mam, respectively. Like SRP54mam and ffh, both predicted yeast proteins contain a GTP binding consensus sequence in their NH2-terminal half (G-domain), and methionine-rich sequences in their COOH-terminal half (M-domain). In contrast to SRP54mam and ffh the yeast proteins contain additional Met-rich sequences inserted at the COOH-terminal portion of the M-domain. SRP54sp contains a 480-nucleotide intron located 78 nucleotides from the 5' end of the open reading frame. Although the function of the yeast homologues is unknown, gene disruption experiments in S. cerevisiae show that the gene is essential for growth. The identification of SRP54sc and SRP54sp provides the first evidence for SRP related proteins in yeast.

The synchrotron Laue technique has been applied to high-resolution structure refinement of the ribotoxin, restrictocin [Yang & Moffat (1996). Structure, 4, 837–852]. By employing carefully designed data-collection strategies and the data-reduction algorithms incorporated in the software system LaueView [Ren & Moffat (1995a). J. Appl. Cryst. 28, 461–481; Ren & Moffat (1995b). J. Appl. Cryst. 28, 482–493], a set of high-resolution Laue data with a completeness and accuracy comparable to excellent monochromatic data was obtained. Through detailed comparison with the monochromatic data and electron-density maps derived from the Laue data, optimum data-collection and reduction strategies were identified and the application of Laue diffraction techniques to conventional crystallographic refinement was demonstrated.

Surgical defects often have adverse effects on patient perception of esthetics and self-esteem. Rehabilitation of such surgical defects poses a challenge to the clinician. Presented here is a clinical report of an interim prosthetic rehabilitation of a patient who underwent partial rhinectomy for basal cell carcinoma. Nasal resection included part of the nasal septum, the entire cartilage, and the ala. An interim nasal prosthesis was fabricated for this patient 3 weeks after surgery, to provide early rehabilitation. This prosthesis provided a sociopsychological benefit to the patient, and the prosthesis was well tolerated. The spectacle glasses retained the prosthetic nose. (Eur J Dent 2010;4:482-486)

The generation of sensory and autonomic neurons from the neural crest requires the functions of two classes of basic helix-loop-helix (bHLH) transcription factors, the Neurogenins (NGNs) and MASH-1, respectively (Fode, C., Gradwohl, G., Morin, X., Dierich, A., LeMeur, M., Goridis, C. and Guillemot, F. (1998) Neuron 20, 483–494; Guillemot, F., Lo, L.-C., Johnson, J. E., Auerbach, A., Anderson, D. J. and Joyner, A. L. (1993) Cell 75, 463–476; Ma, Q., Chen, Z. F., Barrantes, I. B., de la Pompa, J. L. and Anderson, D. J. (1998 Neuron 20, 469–482). We have cloned two chick NGNs and found that they are expressed in a subset of neural crest cells early in their migration. Ectopic expression of the NGNs in vivo biases migrating neural crest cells to localize in the sensory ganglia, and induces the expression of sensory neuron-appropriate markers in non-sensory crest derivatives. Surprisingly, the NGNs can also induce the expression of multiple pan-neuronal and sensory-specific markers in the dermomyotome, a mesodermal derivative. Taken together, these data suggest that a subset of neural crest cells may already be specified for a sensory neuron fate early in migration, as a consequence of NGN expression.

The historical evolution of the heptanese during the classical and hellenistic periods has been little studied until now. The main aim of this thesis is consequently, thanks to a geopolitical approach, to show the richness and the complexity of the international life of the four principal ionian islands forming this archipelago, namely, corcyra (corfu), leucas, cephallenia, and zacynthos. C., le roi du Pont Mithridate VI Eupator, en tant qu'instigateur des vêpres éphésiennes qui coûtèrent la vie à des milliers de Romains, déclencha un conflit avec Rome qui dura un quart de siècle. Au cours de ces guerres dites « mithridatiques », les armées républicaines remportèrent de nombreuses victoires au terme desquelles les généraux romains devaient traiter avec les vaincus et décider du sort qui leur serait réservé. Ce mémoire s'intéresse plus particulièrement à cette relation entre les vainqueurs et les vaincus en cherchant à déterminer si les généraux romains firent preuve de clementia envers les peuples battus. En plus des sources littéraires, un corpus, composé de six textes épigraphiques, est sollicité. Ce travail consiste en une édition commentée des Grenouilles, comédie présentée par Aristophane en 405 avant notre ère, et ayant obtenu une seconde représentation l’année suivante. Aristophane y présente la descente aux Enfers de Dionysos en quête du poète Euripide, et la dispute entre ce dernier et Eschyle pour la suprématie tragique. Notre édition comporte une introduction générale qui resitue les deux représentations dans leur cadre – historique, idéologique et littéraire – et montre comment le contexte immédiat, notamment la représentation des Bacchantes d’Euripide et la mort de celui-ci et de Sophocle, a influencé la structure de la pièce et ses thématiques ; ainsi que la nature et les causes des modifications apportées au texte entre la première et la seconde représentation. Le cœur de l’introduction analyse l’intrigue de la comédie de manière plus ou moins linéaire, montrant comment Aristophane utilise dans la première partie les motifs traditionnels de la catabase et du voyage initiatique, et comment, dans la deuxième partie, il détourne et critique les théories sophistiques du langage pour mettre face à face Euripide et Eschyle et en dégager une réflexion sur la fonction du théâtre dans la cité. This work consists of an edition and commentary of the Frogs of Aristophanes, a comedy performed in 405 BC and a second time the following year. In it, Aristophanes shows the descent to the Underworld of Dionysos as he searches for the poet Euripides, and the latter’s contest with Aeschylus over tragic supremacy.. This edition contains a general introduction replacing both performances of the play in their historical, ideological and literary frame, and showing how immediate context – mainly the performance of Euripides’ Bacchae and his and Sophocles’ death – influenced the play’s structure and themes. It also shows the nature and causes of the revisions that Aristophanes made between the first and the second performance. The central part of the introduction analyses the plot in a mostly linear way, showing how Aristophanes uses the traditional narratives of catabasis and initiatic journey in the first part of the play, and how, in the second part, he appropriates and criticises the sophists’ theories of language in order to oppose Aeschylus and Euripides and consider the role of theatre in the city. One chapter is dedicated to hypotheses concerning the original stage performance, and another to the transmission of the text between Antiquity and modern times. The line-by-line commentary preoccupies itself with matters of textual criticism, vocabulary, grammar, metric, performance and intertextuality. The text is completed by a critical apparatus that reports the readings of four of the oldest mediaeval manuscripts (and some others when the need arises) and an original French translation

Cette thèse explore la notion de corruption dans le Corpus hippocratique et dans les tragédies d’Euripide. L’objectif est non seulement de relire et de réinterpréter les termes qui expriment la corruption, mais surtout de les replacer dans leur contexte historique et social. Une analyse sémantique préliminaire explique la signification de deux termes grecs fondamentaux qui expriment la corruption, φθείρω et σήπω, et montre l’importance de ces termes dans la période classique. La notion de corruption dans le Corpus hippocratique est étudiée à travers l’examen de diverses maladies liées à la tête, au thorax, à l’utérus et aux deux fluides essentiels du corps humain, le sang et la semence. Cette analyse démontre ainsi que la corruption dans le Corpus hippocratique est étroitement liée à l’évolution des maladies et concerne principalement les dysfonctionnements du corps. En revanche, la notion de corruption chez Euripide, telle que révélée par l’étude des tragédies sélectionnées, dépasse la sphère corporelle et est employée métaphoriquement pour exprimer l’intensité dramatique des situations où les héros vivent une détérioration physique ou mentale extraordinaire. Malgré la différence flagrante entre les deux œuvres, une évaluation comparative des deux œuvres met en exergue une similitude intrigante entre les deux auteurs concernant le rôle particulier des thèmes liés aux femmes ou à la mort imminente dans le contexte de la corruption. Dans son ensemble, cette étude met en évidence une complexité et une richesse sémantique associées à la corruption, largement inconnues du lecteur moderne
This thesis explores the meaning of corruption in the Hippocratic Corpus and in the tragedies of Euripides. The aim is not only to reread and reinterpret the terms that express corruption, but above all to place them in their historical and social context. A preliminary semantic analysis describes the meaning of two basic Greek terms that express corruption, φθείρω and σήπω and shows the importance of these terms in the classical period. The notion of corruption in the Hippocratic Corpus is studied through the examination of a range of diseases related to the head, thorax, uterus and the two essential fluids of the human body, blood and semen. This analysis shows that corruption in the Hippocratic Corpus is closely related to the evolution of disease and primarily concerns malfunctions of the body. In contrast, the notion of corruption in Euripides, as revealed by the analysis of a selection of tragedies, goes beyond the corporeal sphere and is used metaphorically to express the dramatic intensity in situations when heroes experience extraordinary physical or mental deterioration. Despite this clear distinction, a comparative evaluation of the two works illustrates an intriguing similarity between the two authors regarding the particular role of themes related to women or imminent death in the context of corruption. Overall, this study reveals a complexity and a semantic richness associated with corruption that is largely unknown to the modern reader

Aesthetic appreciation, like any ordinary phenomenon of human life, triggers affective and cognitive processes that can provide the subject with sensations of hedonic pleasure and cognitive self-reward (Leder H, Belke B, Oeberst A, Augustin D. Br J Psychol 95(4):489–508, 2004). Although humans share several neuropsychological processes, the experience of aesthetic appreciation is undeniably idiosyncratic, and sometimes it is not that simple to find beauty where we were supposed to find it, and more often the same object can elicit different reactions amongst observers.

AbstractThis qualitative study on steep social mobility is based on interviews with descendants of guest-worker immigrants from Turkey. They currently work in highly prestigious positions in the field of professional business services in the Netherlands. The majority of studies on social capital and career mobility is based on quantitative methods which makes it difficult to gain insight into its mechanisms. In line with Lin (Annu Rev Sociol 25:467–487, 1999) and Smith (Am J Sociol 111:1–57, 2005) the current article makes the distinction between the access to and the activation of social capital. By explaining how the professional characteristics of the social climbers triggered their network connections to become ‘donors’ (Portes A, Annu Rev Sociol 24:1–24, 1998), we attempt to shed light on the mechanisms of social capital activation. For this claim we apply the concepts of ‘likeability’ and ‘reliability’. Furthermore, we categorize two types of donors that have emerged from our data: ‘ambassadors’ and ‘coaches’. By comprehending how the descendants of migrants activated their social capital, this article gives insight into the mechanisms of their steep social mobility, and it deepens our understanding of the relation between social capital and career mobility.

ABSTRACT Geothermal formations are generally fractured using water-based fracturing fluid, which has several issues, such as high-water consumption and environmental issues. In this study, a waterless stimulation method was used on granite samples. Studying the optimum injection time of LN2 in HDR formations for efficient fracturing will be beneficial for the future of EGS. Effects of varying LN2 exposure time on heated granite samples and their mechanical and physical properties were studied. Eleven core samples were heated to different temperatures, up to 482°F, and treated with LN2 for different times of 10,20, and 30 minutes. CT scan, porosity, permeability, and ultrasonic velocities tests were conducted both before and after LN2 treatment. Later, unconfined compressive strength and X-Ray Diffraction tests were conducted on the treated samples. The results indicated that the physical and mechanical properties of the core samples were altered after implementing LN2 treatment. The permeability enhancement was more significant than the porosity improvement. The ultrasonic velocities of the samples decreased after the cryogenic treatment owing to the creations of the new fractures inside the samples. Increasing the treatment temperature and treatment time results in an increase in the permeability of samples, which will improve the heat extraction in geothermal formations. INTRODUCTION As an abundant and sustainable energy source, geothermal energy, particularly stored in hot dry rocks (HDR), can play a critical role in reducing the fossil fuel consumption and the associated environmental dangers such as carbon dioxide emissions, water pollution, and acid rain (Lu, 2018; Y. Shi et al., 2018; Tester et al., 2006). The amount of heat present in HDR is large, approximately 1.4 × 1025 J in the United States and over 2.52×1025 J in China, and the temperature is higher than in the other geothermal systems, 300°F – 1200°F (Song et al., 2018). The most suitable rock type for HDR is granite or other crystalline basement rock with temperatures from 300°F to 950°F at depths from 3 to 4 miles (Potter et al., 1974). The method of producing energy from hot dry rocks is known as Enhanced Geothermal System (EGS). Even though there are startling numbers regarding the potential of EGS from numerous studies around the world, tapping energy from HDR is still in progression (Breede et al., 2013).

Powerful IR radiation tunable in the wavelength range of 1.44-4.05 μm can be generated by an OPO of potassium niobate (KNbO3) pumped by the fundamental of a pulsed Nd:YAG laser 1. The OPO consists of a 10-mm-long AR- coated crystal (critical type-l phase-matching, θ=41°, ϕ=0°) placed in a 12-mm-long flat-flat mirror cavity resonant for the signal wave The energy density at threshold increases from 0.485 J/cm2 at 1.9 μm to more than 0.7 J/cm below 1 48 μm. To avoid damage of the coating of the KNbO3 crystal the pump energy densities should be less than 1 J/cm2. Due to the rather large walk-off of 3.5° the threshold decreases with increasing pump beam diameter (from 0.6 J/cm3 at 2.35 mm to 0.485 J/cm2 at 4,85 mm). Hence, enlarging the pump beam diameter front 3.05 mm to 4.85 mm. the OPO energy and efficiency increases at constant pump energy density (0.975 J/cm2) from 6.4 mJ to 30 mJ and from 9 % to 16.7 %, respectively.

In a collaborative program, J-R results were compared from multi-specimen and single-specimen procedures for toughness testing with single-edge notched tensile loaded SEN(T) specimens. Grade 483 MPa (X70) grade pipeline steel was used to prepare surface notched axially loaded specimens. Test procedures followed the multi-specimen method and a single-specimen recommended practice recently developed at the CANMET Materials Technology Laboratory (recently name-changed to CanmetMATERIALS) in a program jointly funded by PERD, PRCI, and DOT. The multi-specimen method adopted side grooves with the objective of comparison with results from the single–specimen method. The specimen geometry was B × B for both test procedures. The target ao/W was about 0.5. The multi-specimen testing was performed at BMT Fleet Technology. The single-specimen testing was performed at the CANMET laboratory, formerly located in Ottawa. The paper describes experimental and analysis details, and compares results from the two techniques, using J expressions developed at CANMET. The results showed similar J-resistance curves at small crack extensions for both techniques, and higher J-resistance values at larger crack extensions for the multi-specimen method. The results are discussed in terms of initial crack length and the analysis methods adopted for the two techniques. Validity criteria according to ASTM E1820 were applied and the results are presented.

Abstract Recent destructive analysis of six ASTM A350 LF2 flanges has revealed vastly different low temperature (-50°F) Charpy impact toughness from 4 J (3 ft-lbs) to greater than 298 J (220 ft-lbs). These relatively low strength flanges, minimum 248 MPa (36 ksi) yield and 483-655 MPa (70-95 ksi) tensile strength, had nominally the same yield and UTS despite the difference in toughness. Detailed chemical and microstructural analysis was undertaken to elucidate the cause of the toughness range. The majority of the flanges had aluminum additions and a fine grain size with the toughness differences mostly explained by the cooling rate after normalizing with the still air cool showing the lowest toughness and the fastest air cooled sample the highest. For flanges of this strength level a quench and temper operation is not required to obtain good low temperature toughness but forced air cooling after normalizing is a minimum cooling rate to ensure good toughness and overall strength.

Serotonin (5-HT) stimulates prostacyclin (PGI2) production by bovine aortic smooth muscle cells in culture via 5-HT2 receptors (1). These cells express a synthetic phenotype (2), whereas the majority of the smooth muscle cells in the media from adult arteries are in a contractile state. We have now shown that 5-HT (1-10 μM) also stimulates PGI2 production by a preparation of contractile smooth muscle cells : explants from bovine aortic media cultured for short periods. This effect is independent from 5-HT2 receptors : it is only partially inhibited (±30%) by ketan-serin (a selective and potent 5-HT2 antagonist) and is perfectly mimicked by a 5-HT1 agonist, 5-carboxamidotryptamine. 5-HT2 receptors seem to be linked to a phospholipase C (3), with subsequent accumulation of inositol tr isphosphate , Ins(1,4,5)P3, and diacylglycerol, an activator of protein kinase C. We have observed a stimulatory effect of phorbol 12-myristate, 13-acetate (a selective activator of kinase C) on PGI2 production by the bovine aortic smooth muscle cells (synthetic state), whereas it was totally ineffective on media explants preparation (contractile state). Furthermore, in the smooth muscle cells in culture, the 5-HT effect can be inhibited by (ethyl-isopropyl)amiloride, a potent and selective inhibitor of the Na+/H+ antiporter. In conclusion it appears that the regulation mechanisms of PGI2 production in arterial smooth muscle cells are strongly dependent an the phenotypic state of these cells. The control of PGI2 release via 5-HT2 receptors seems to involve a cytoplasmic alkalinization, via the activation of protein kinase C. Abstract:
X-ray lasers have been available for almost ten years with the majority still being pumped by high energy (≈ 1 kJ) modest duration (≈ 500 psec) optical lasers. The recent availability of ultrashort pulse (≈ 100 fsec) optical lasers with modest energies (≈ 1 J), has renewed the interest in x-ray laser schemes that could be pumped by these relatively inexpensive, compact lasers. In one x-ray laser scheme, the ultrashort pulse, high intensity laser ionizes the target gas via the electric field of the laser with subsequent recombination and lasing between the first excited state and the ground state. In contrast with collisional ionization, this ionization mechanism may result in nearly complete emptying of a given ionization stage and has the potential of leaving relatively cold free electrons. There is potential for lasing down to wavelengths of order 100Å, e.g. Li-like Ne at 98Å, using lasers with energies of order 1 J.1 Lasing at significantly shorter wavelengths, e.g. Li-like Al at 52Å or H-like B at 48Å, appears to be very difficult because of excessive heating associated with Raman instabilities.2 There have been recent experimental results showing possible evidence of lasing in H-like Li at 135Å.3,4 We discuss theoretical efforts on this approach to lasing and modeling efforts to understand the recent data. A second approach to x-ray lasing using ultrashort pulse lasers is based on innershell photoionization which has the potential of reaching short wavelengths, e.g., 15Å in Ne. The indirect ionization process of this scheme, where incoherent x rays emitted from a nearby plasma ionize the innershell electron, results in higher energy requirements. For example, lasing in Ne is calculated to require a pump laser with approximately 10 J in a 100 fsec pulse. The energy requirements are significantly reduced by considering longer wavelengths, e.g., 45Å in C, but a rapid rise time (≈ 50 fsec) for the x rays is still required to minimize collisional ionization. For both approaches the use of modest energy ultrashort pulse lasers allow for much higher repetition rates than available with conventional x-ray lasers pumped by high energy laser drivers. We discuss new applications appropriate for such high-repetition-rate x-ray lasers.

The increased automation of the car manufacturing process, in which much of the assembly has been delegated from man to machine, has done much to relieve workers of the burden of heavy lifting. However, despite ergonomic improvements in the workplace, many jobs still require workers to perform repetitive tasks [1]. In the automotive industry, Work-related Musculoskeletal Disorders (WMSD) are one of the most common occupational problems due to repetitive working tasks. Workers that perform manual work are often prone to awkward postures, repetitive movements, forceful exertions, and overextensions, which are some of the main factors for the arising of WMSD [2]. Besides, these work-related factors, also the personal factors contribute to the occurrence of this kind of injury, making WMSD a complex condition that involves contributions from many factors [3,4]. The current study aims to compare different observational methods commonly used to assess the WMSD risk in repetitive tasks. To accomplish this goal a case of study in assembly workstation of an automotive company in Portugal was applied. It was selected methods that are widely used by ergonomists and are validated for implementation in the industry [5,6]. Therefore, the following methods were applied to an assembly workstation: (i) Rapid Upper-Limb Assessment (RULA), (ii) Occupational Repetitive Actions (OCRA), (ii) Key Indicator Method – Manual Handling Operations (KIM-MHO), and (iv) Revised Strain Index (RSI) This multi-method approach was very important, as it allowed for a more comprehensive assessment, which will support the proposals for improvement The results show that workstation present a considerable WMSD risk in 3 of 4 methods applied. These results suggest that a change to the workstation is necessary. A possible solution would be to implement a Human-robot collaboration solution, in order to reduce the physical demands associated with repetitive movements [7] to which workers are subjected.
