Abstract:
Background and Objectives: Microtubules are an attractive target for cancer chemotherapy. Previously, we reported that Ivalin exhibited excellent anti-migration and anti-invasion activities in human breast cancer cells. Here, we examined the microtubule inhibition effect of Ivalin in human hepatocellular carcinoma SMMC-7721 cells. Materials and Methods: We used the 3-(4,5-dimethylthiazol)-2,5-diphenyltetrazolium bromide (MTT) assay to evaluate the cell proliferation effect of Ivalin and flow cytometry analysis to detect the apoptotic and cell cycle arrest effects of Ivalin. Immunofluorescence staining was used to measure the effect of Ivalin on the cytoskeleton network, and Western blotting was used to detect the expression levels of Bax, Bcl-2, Cdc2, phosphor-Cdc2, Cdc25A, Cyclin B1, and tubulin. Results: Ivalin induced cell cycle G2/M arrest and subsequent triggered apoptosis in human hepatocellular carcinoma SMMC-7721 cells. Furthermore, microtubules were shown to be involved in Ivalin-meditated apoptosis. In this connection, Ivalin treatment suppressed cellular microtubule network formation by regulating microtubule depolymerization. Moreover, Western blotting revealed Cdc25A and Cyclin B1 were upregulated in Ivalin-meditated cell cycle arrest. Subsequently, the induction of Bax (a proapoptotic protein) and reduction of Bcl-2 (an anti-apoptotic protein) expression were observed in Ivalin-treated SMMC-7721 cells. Conclusion: Ivalin induced microtubule depolymerization, then blocked cells in mitotic phase, and eventually resulted in apoptosis in SMMC-7721 cells. Collectively, these data indicate that Ivalin, acting as a novel inhibitor of microtubules, could be considered as a promising lead in anticancer drug development.

Abstract:
Ivalin, a natural compound isolated from Carpesium divaricatum, showed excellent microtubule depolymerization activities among human hepatocellular carcinoma in our previous work. Here, we investigated its functions on mitochondria-mediated apoptosis in hepatocellular carcinoma SMMC-7721 cells. DAPI (4′,6-diamidino-2-phenylindole) staining, annexin V-fluorexcein isothiocyanate (FITC) apoptosis detection, and western blotting were applied to explore the apoptotic effect of Ivalin. Next, the induction effect of Ivalin on the mitochondrial pathway was also confirmed via a series of phenomena including the damage of mitochondria membrane potential, mitochondria cytochrome c escape, cleaved caspase-3 induction, and the reactive oxygen species generation. In this connection, we understood that Ivalin induced apoptosis through the mitochondrial pathway and the overload of reactive oxygen species. Furthermore, we found that the activation of nuclear factor-κB (NF-κB) and subsequent p53 induction were associated with the apoptotic effect of Ivalin. These data confirmed that Ivalin might be a promising pro-apoptotic compound that can be utilized as a potential drug for clinical treatment.

The inhibition of ATP synthesis, proton uptake, and electron transport (basal, phosphorylating, and uncoupled) from water to methylviologen by ivalin (a naturally occurring sesquiterpene lactone in Zaluzania triloba and Iva microcephala) indicates that it acts as electron transport inhibitor. Since photosystem I and electron transport from DPC to QA were not affected, while the electron flow of uncoupled photosystem II from H2O to DAD and from water to silicomolybdate was inhibited, we concluded that the site of inhibition of ivalin is located at the oxygen evolution level. Abstract:
Vermeersiekte or “vomiting disease” is an economically important disease of ruminants following ingestion of Geigeria (G.) species in South Africa. Sheep are more susceptible, and poisoning is characterized by stiffness, regurgitation, bloat, paresis, and paralysis. Various sesquiterpene lactones have been implicated as the cause of poisoning. The in vitro cytotoxicity of two sesquiterpene lactones, namely, ivalin (purified from Geigeria aspera) and parthenolide (a commercially available sesquiterpene lactone), were compared using mouse skeletal myoblast (C2C12) and rat embryonic cardiac myocyte (H9c2) cell lines, representing the oesophageal, skeletal and cardiac muscles, which are affected in sheep. For 24, 48, and 72 h, both cell lines were exposed. A colorimetric viability assay, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), was used to assess cytotoxicity. A concentration-dependent cytotoxic response was observed in both cell lines, however, the C2C12 cells were more sensitive, with the half-maximal effective concentrations (EC50s) ranging between 2.7 and 3.3 µM. In addition, the effect that ivalin and parthenolide has on desmin, an important cytoskeletal intermediate filament in myocytes, was evaluated using the C2C12 myoblasts. Disorganization and aggregation of desmin were caused by both sesquiterpene lactones, which could clarify some of the ultrastructural lesions described in vermeersiekte.

The flower heads of Inula britannica L. of Bulgarian origin afforded sesquiterpene lactones (gaillardin, britannin, 11,13-dihydroinuchinenolide B, ivalin, pulchellin C), triterpenoids (3- O-palmitates of 16β-hydroxylupeol, 16β-hydroxy-β-amyrin, and faradiol) and flavonoids (quercetin, luteolin, luteolin-7- O-glucoside). All compounds are known and they were identified by spectral methods. The observed differences in the chemical content of the chloroform and methanol extracts were also reflected in their free radical scavenging activity, evaluated by DPPH and ABTS assays. Intraspecific variability of I. britannica is discussed.

Abstract:
Marine aquaculture development is recently impeded by parasitic leech Zeylanicobdellaarugamensis (Hirudinea, Piscicolidae) in Sabah, Malaysia. The parasitic leech infests a variety of cultured fishes in aquaculture facilities. In this study, we evaluated the antiparasitic activity of the chromatographic fractions of the medicinal plant Nephrolepis biserrata methanol extract against Z.arugamensis and highlighted the potential metabolites responsible for the antiparasitic properties through liquid chromatography (LC)–quadrupole time-of-flight (QTOF)–mass spectrometry (MS) analysis. Out of seven fractions obtained through flash column chromatography techniques, three fractions demonstrated antiparasitic properties. Significant parasitic mortality was indicated by fraction 3 at a concentration of 2.50 mg/mL, all the leeches were killed in a time limit of 1.92 ± 0.59 min. followed by fraction 4 (14 mg/mL) in 34.57 ± 3.39 and fraction 5 (15.3 mg/mL) in 36.82 ± 4.53 min. LC-QTOF-MS analysis indicated the presence of secondary metabolites including phytosphingosine (6), pyrethrosin (1), haplophytine (9), ivalin (2), warburganal (3), isodomedin (4) and pheophorbide a (16), representing sphingoid, alkaloid, terpenoid, phenolic and flavonoid groups. Thus, our study indicated that the chromatographic fractions of N. biserrata demonstrated significant antiparasitic activity against the marine parasitic leeches due to the presence of potent antiparasitic bioactive compounds.

Abstract:
Fungi from the genus Candida are very important human and animal pathogens. Many strains can produce biofilms, which inhibit the activity of antifungal drugs and increase the tolerance or resistance to them as well. Clinically, this process leads to persistent infections and increased mortality. Today, many Candida species are resistant to drugs, including C. auris, which is a multiresistant pathogen. Natural compounds may potentially be used to combat multiresistant and biofilm-forming strains. The aim of this review was to present plant-derived preparations and compounds that inhibit Candida biofilm formation by at least 50%. A total of 29 essential oils and 16 plant extracts demonstrate activity against Candida biofilms, with the following families predominating: Lamiaceae, Myrtaceae, Asteraceae, Fabaceae, and Apiacae. Lavandula dentata (0.045–0.07 mg/L), Satureja macrosiphon (0.06–8 mg/L), and Ziziphora tenuior (2.5 mg/L) have the best antifungal activity. High efficacy has also been observed with Artemisia judaica, Lawsonia inermis, and Thymus vulgaris. Moreover, 69 plant compounds demonstrate activity against Candida biofilms. Activity in concentrations below 16 mg/L was observed with phenolic compounds (thymol, pterostilbene, and eugenol), sesquiterpene derivatives (warburganal, polygodial, and ivalin), chalconoid (lichochalcone A), steroidal saponin (dioscin), flavonoid (baicalein), alkaloids (waltheriones), macrocyclic bisbibenzyl (riccardin D), and cannabinoid (cannabidiol). The above compounds act on biofilm formation and/or mature biofilms. In summary, plant preparations and compounds exhibit anti-biofilm activity against Candida. Given this, they may be a promising alternative to antifungal drugs.

Vermeersiekte or ‘vomiting disease’ is an economically important disease of ruminants following ingestion of Geigeria species in South Africa. Sheep are more susceptible and poisoning is characterised by stiffness, regurgitation, bloat, paresis and paralysis. Geigeria aspera was collected in the Vrede district (27° 25′ 48″ S; 29° 9′ 36″ E), Free State Province. The plant material was dried, milled and the toxic principles, known as sesquiterpene lactones, were extracted and isolated following chromatographic procedures. Even though geigerin and ivalin were previously isolated, an unknown sesquiterpene lactone, isogeigerin acetate, was also purified. Mouse myoblast (C2C12) and rat embryonic cardiac myocyte (H9c2) cell lines were exposed to different concentrations of geigerin, ivalin, isogeigerin acetate and a commercially available sesquiterpene lactone, parthenolide. An in vitro colorimetric assay 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was used to assess cytotoxicity. The median effective concentrations (EC50) indicated that ivalin and parthenolide were significantly (p<0.05) more toxic than geigerin and isogeigerin acetate. A concentration-dependent cytotoxic response was observed in both cell lines, however, C2C12 cells were more sensitive. A high performance liquid chromatography (HPLC) analysis was used to evaluate the in vitro metabolism of parthenolide, following the addition of a mouse liver microsomal fraction. Results revealed that parthenolide, incubated with the microsomal fraction, undergoes enzymatic transformation to form a metabolite.

Resumo: Estudo do coreógrafo e professor Ivaldo Bertazzo, que desde os anos 70 desenvolve trabalhos com "cidadãos-dançantes", centrado na sua pesquisa de movimento. A partir de 2000, Bertazzo também se dedica a projetos de arte-educação com adolescentes da periferia do Rio de janeiro e São Paulo. O estudo tem três metas principais: traçar um panorama da trajetória de Ivaldo Bertazzo, como coreógrafo e diretor de espetáculos e de uma escola de dança; entender as bases da sua pesquisa de movimento e fazer a descrição analítica de seu trabalho específico com o corpo (Reeducação do Movimento), necessária para o entendimento da criação coreográfica; e investigar de que modo o conceito da construção da identidade pelo movimento, apontado por Bertazzo, ocorre no seu trabalho. Princípios coreográficos e técnica corporal foram abordados aqui no viés da "identidade e autonomia do movimento", entendida como forma de pensar o corpo, sua articulação e desenho no espaço, numa técnica híbrida, influenciada, por diversas fontes (incluindo a dança indiana, estudos de coordenação matara por Piret e Béziers e das cadeias musculares por Denys-Struyf). A obra de Bertazzo põe em xeque não só nossas idéias sobre o corpo, mas também nossa idéia de Brasil. Com isso em mente, durante a análise foi levada em conta a situação política do país e sua influência na evolução da dança nesse período, examinando as marcas que deixa no desenvolvimento dos criadores, obras, escolas e movimentos da nossa cultura. Abstract: This is a study of the Brazilian choregrapher and teacher IvaIdo Bertazzo, who has developed an original work with "citizen-dancers" since the 1970s, based on his own research on movement techniques. From 2000 on, Bertazzo has also developed a series of art-education projects with teenagers from poor neighbourhoods in Rio de Janeiro and São Paulo. The thesis has three main aims: to draw a panorama of Bertazzo's career as a choreographer, artistic director and head of a dance school; to explain the bases for his research on movement, and to write an analytical descrition of his work on the body (Movement Reeducation), which is necessary in order to understand his choreographic creations; and to investigate the many ways in which his concept of "identity built by movement" pIays a role in this oeuvre. Choreographic principIes and body technique have been studied in the light of his ideas on "identity and autonomy of movement", taken as a way to think about the body, its shape and its forms of articulation in space, and leading to a hybrid technique, that draws on many different sources (including Indian dance, studies of coordination by Piret and Béziers, and the study ofmuscle chains by Denys-Struyf). Bertazzo's work challenges not only our ideas on the body, but aIso our ideas about Brazil. With that in mind, the research brings to attention many facets of the polítical situation and its influence upon Brazilian dance over these decades. It also examines the marks this has left on the work ofmany authors, schools and movements in Brazilian culture. The thesis collects photographs and video footage of many works performed over the Jast thirty years - an image bank which may be of great assistance for a better understanding of this period in Brazilian dance history

This thesis aims to plan and design an urban monument in Seoul, Republic of Korea, that can reflect the political movement of the city at the urban area which has been historically associated with political representations of the government and civic organizations. The advent of digital technologies allows a massive amount of social and political ideas of citizens to be collected efficiently through online networks, and enables this gathered data to be linked to physical environments through digital interactive interfaces. In this system, when users input data, interfaces respond to it by generating physical components interactively; therefore, individuals can create urban environments with own intentions. However, at the same time, the technological movement also raises a demand for a comprehensive master plan which can properly control digital systems in physical space. The goal of planning and design in this project is to create an urban master plan that can integrate the political symbolism of the site and the new technological system.

I den här uppsatsen undersöks Herr Ivan, en av de tre Eufemiavisorna, med syfte att utröna vilka lexikala skillnader det finns i arvorden mellan de tre svenska och två danska handskrifterna av romanen som finns bevarade från 1400-talet, kallade A–C resp. E–F. Det excerperade materialet består av de arvord som enligt Erik Noreens kritiska utgåva avviker i E–F i förhållande till A–C, med morfologiska och ortofonologiska skillnader borträknade. Vissa gränsfall diskuteras dock. Undersökningen tar upp ett axplock av de utbyten som bedömts som mest relevanta i fråga om mängd, ändringsfrekvens och sammanfall med det nutida ordförrådet i båda språken. Orden i materialet delas upp i ordklasser, och både antalet belägg och lexem har räknats. Materialet visar ingen stor skillnad mellan språken, men det finns dock tendenser till att de danska handskrifternas ordförråd ligger närmare det nutida än vad de svenska gör.

The basis of this dissertation confronts through different criteria Czech photographer Ivan Pinkava and Israeli Adi Nes. The works of both artists deal with stage photography based on direct references to well-known works in art history, biblical stories or other great stories from European culture. Part one will discuss their work within the brother context of postmodern art. Part two will talk about the formal aspects of their photographs, e.g. the character of lighting, structure of the picture. Finally the last chapter will explore the meaning and the subject-matter of their attitudes, specifically translating their photographic language in the relationship to painting, photography and cinematography.

Professor M. Zhytaryuk’s review is about a book scientific novelty – a monograph by Professor M. Tymoshyk «Ukrainian journalism in the diaspora: Great Britain. Monograph. K.: Our culture and science, 2020. 500 p. – il., Them. pok., resume English, German, Polish.». Well-known scientist and journalism critic, Professor M. S. Tymoshyk, wrote a thorough work, which, in terms of content, is a combination of a monograph, a textbook and a scientific essay. This book can be useful for both students and practicing journalists or anyone interested in the history of the Ukrainian diaspora, Ukrainian journalism and Ukrainian culture. The author dedicated his work to Stepan Yarmus from Winnipeg, Canada – archpriest, journalist, editor, professor. As the epigraph to the book were taken the words of Ivan Bagryany: «Our press, born under the sword of Damocles of repatriation», not only survived and survived to this day, but also showed a brilliant ability to grow and develop. It was shown that beggars that had come to the West without money at heart can and know how to act so organized. It was also an example of how a modern «enbolshevist» and «denationalized» by the occupier man person is capable of a combined mass action».

Abstract:
The article investigates functional techniques of extralinguistic expression in multimedia texts; the effectiveness of figurative expressions as a reaction to modern events in Ukraine and their influence on the formation of public opinion is shown. Publications of journalists, broadcasts of media resonators, experts, public figures, politicians, readers are analyzed. The language of the media plays a key role in shaping the worldview of the young political elite in the first place. The essence of each statement is a focused thought that reacts to events in the world or in one’s own country. The most popular platform for mass information and social interaction is, first of all, network journalism, which is characterized by mobility and unlimited time and space. Authors have complete freedom to express their views in direct language, including their own word formation. Phonetic, lexical, phraseological and stylistic means of speech create expression of the text. A figurative word, a good aphorism or proverb, a paraphrased expression, etc. enhance the effectiveness of a multimedia text. This is especially important for headlines that simultaneously inform and influence the views of millions of readers. Given the wide range of issues raised by the Internet as a medium, research in this area is interdisciplinary. The science of information, combining language and social communication, is at the forefront of global interactions. The Internet is an effective source of knowledge and a forum for free thought. Nonlinear texts (hypertexts) – «branching texts or texts that perform actions on request», multimedia texts change the principles of information collection, storage and dissemination, involving billions of readers in the discussion of global issues. Mastering the word is not an easy task if the author of the publication is not well-read, is not deep in the topic, does not know the psychology of the audience for which he writes. Therefore, the study of media broadcasting is an important component of the professional training of future journalists. The functions of the language of the media require the authors to make the right statements and convincing arguments in the text. Journalism education is not only knowledge of imperative and dispositive norms, but also apodictic ones. In practice, this means that there are rules in media creativity that are based on logical necessity. Apodicticity is the first sign of impressive language on the platform of print or electronic media. Social expression is a combination of creative abilities and linguistic competencies that a journalist realizes in his activity. Creative self-expression is realized in a set of many important factors in the media: the choice of topic, convincing arguments, logical presentation of ideas and deep philological education. Linguistic art, in contrast to painting, music, sculpture, accumulates all visual, auditory, tactile and empathic sensations in a universal sign – the word. The choice of the word for the reproduction of sensory and semantic meanings, its competent use in the appropriate context distinguishes the journalist-intellectual from other participants in forums, round tables, analytical or entertainment programs. Expressive speech in the media is a product of the intellect (ability to think) of all those who write on socio-political or economic topics. In the same plane with him – intelligence (awareness, prudence), the first sign of which (according to Ivan Ogienko) is a good knowledge of the language. Intellectual language is an important means of organizing a journalistic text. It, on the one hand, logically conveys the author’s thoughts, and on the other – encourages the reader to reflect and comprehend what is read. The richness of language is accumulated through continuous self-education and interesting communication. Studies of social expression as an important factor influencing the formation of public consciousness should open up new facets of rational and emotional media broadcasting; to trace physical and psychological reactions to communicative mimicry in the media. Speech mimicry as one of the methods of disguise is increasingly becoming a dangerous factor in manipulating the media. Mimicry is an unprincipled adaptation to the surrounding social conditions; one of the most famous examples of an animal characterized by mimicry (change of protective color and shape) is a chameleon. In a figurative sense, chameleons are called adaptive journalists. Observations show that mimicry in politics is to some extent a kind of game that, like every game, is always conditional and artificial.
