Abstract:
ABSTRACT Genetic structures surrounding the carbapenem-hydrolyzing oxacillinase gene bla OXA-58 were characterized in a series of OXA-58-positive Acinetobacter baumannii strains isolated from different countries. We showed that in most of the cases, acquisitions of the bla OXA-58-containing overall structure, including insertion sequence elements, may be likely the results of recombination events. In type strain A. baumannii MAD, the genetic structure surrounding the bla OXA-58 gene was bracketed by two 27-bp repeated sequences. The isolation of a clonally related OXA-58-negative A. baumannii isolate that possessed the same plasmid backbone as A. baumannii MAD but lacked this genetic structure indicated that the mechanism of acquisition could be reversible. Parts of the structure identified in A. baumannii MAD were conserved in other bla OXA-58-positive isolates from various European countries. Primer extension experiments showed that bla OXA-58 expression was related to promoter sequences brought by different insertion sequence elements, such as an ISAba3-like element, ISAba1, ISAba2, and IS18. This work identified novel structures at the origin of acquisition and expression of a carbapenem-hydrolyzing β-lactamase identified in non-clonally related A. baumannii isolates.

Abstract:
Abstract This article critically re-examines the textual relationship between the Enarratio in prophetam Isaiam attributed to Basil of Caesarea and Basil’s (genuine) homily In ebriosos. It studies how Basil makes use of Isaiah 5:11–12 in his In ebriosos, and shows its degree of resemblance to the Enarratio in Isaiam 5:11–12. Moreover, it refutes the arguments in support of Basil’s authorship of the Enarratio as well as its anteriority and influence on the In ebriosos. The study argues instead that the anonymous author of the Enarratio made use of a certain part of Basil’s In ebriosos in his attempt to fabricate the beginning of his explanation on Isaiah 5:11–12. Finally, the article examines the process of transformation of the homily under discussion into a commentary.

Abstract:
Acinetobacter baumannii is an important cause of nosocomial infections worldwide. The elucidation of the carbapenem resistance mechanisms of hospital strains is necessary for the effective treatment and prevention of resistance gene transmission. The main mechanism of carbapenem resistance in A. baumannii is carbapenemases, whose expressions are affected by the presence of insertion sequences (ISs) upstream of blaCHDL genes. In this study, 61 imipenem-nonsusceptible A. baumannii isolates were characterized using phenotypic (drug-susceptibility profile using CarbaAcineto NP) and molecular methods. Pulsed field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST) methods were utilized for the genotyping. The majority of isolates (59/61) carried one of the following acquired blaCHDL genes: blaOXA-24-like (39/59), ISAba1-blaOXA-23-like (14/59) or ISAba3-blaOXA-58-like (6/59). Whole genome sequence analysis of 15 selected isolates identified the following intrinsic blaOXA-66 (OXA-51-like; n = 15) and acquired class D β-lactamases (CHDLs): ISAba1-blaOXA-23 (OXA-23-like; n = 7), ISAba3-blaOXA-58-ISAba3 (OXA-58-like; n = 2) and blaOXA-72 (OXA-24-like; n = 6). The isolates were classified into 21 pulsotypes using PFGE, and the representative 15 isolates were found to belong to sequence type ST2 of the Pasteur MLST scheme from the global IC2 clone. The Oxford MLST scheme revealed the diversity among these studied isolates, and identified five sequence types (ST195, ST208, ST208/ST1806, ST348 and ST425). CHDL-type carbapenemases and insertion elements upstream of the blaCHDL genes were found to be widespread among Polish A. baumannii clinical isolates, and this contributed to their carbapenem resistance.

Abstract:
This article is a part of a Doctoral Thesis titled “Isaan Heritage Tree: From the Belief and the Aesthetic to the Creation of Visual Arts,” with the objectives of studying the belief and the aesthetic that exist in the Isaan Heritage Tree using the qualitative method. The study’s target groups can be divided into three following groups: the experts, the practitioners, and the related people. The area of study is in the Isaan region. The region is divided into upper Isaan, mid-Isaan, and lower Isaan. The research tools consist of surveys, non-participatory observation, and structured and non-structured interviews. The data collected from documents and field data was analyzed using Aesthetic Theory and Symbolic Interactionism Theory and presented using descriptive analysis. The study results show that Isaan has a long history and development both in geography, the administration, society, the culture, and the migration of people who came to settle in the area from Luang Phrabang, Vientiane, and Champasak. This had caused the people and nation’s coming together and led to social management, which consists of regulations, religion, and belief. The beliefs of the Isaan people are connected to forest spirits, household spirits, or tree spirits. Five following characteristics of the Heritage Trees were also found: 1) The traditional beliefs related to the Heritage Trees of Isaan. 2) The new belief. 3) The beliefs that are connected to the locations. 4) The beliefs in the tree spirits whose identity and gender cannot be identified. 5) Auspicious and inauspicious beliefs. There are three aspects for the aesthetics: Aesthetic elements are the feeling of amazement due to the gigantic size filled with astonishment, mystery, and the fear of power. The interesting aspects of Art elements are the unity and relationship to the seasons, such as the Fall season, Rainy Season, and the blooming of flowers that contribute to the changes in the aesthetics changes. The visual art elements consist of six following components; bodies and shapes, lines, colors, textures, light and shadows, and area. It was found that the gigantic size and height cause amazements to the viewers. The physical lines of the Isaan Heritage trees were the lines along the trunks, the lines on the branches, and the lines that go along the leaves and flowers. There are different colors of the trunk, the leaves, and the flowers. The texture was rough, harsh, and the cracks follow the same directions as the trunk. There are botanical differences in the light and shadows of the heritage trees. As for the area, there are differences between the area of the heritage trees and the surrounding areas, as well as the differences within the Isaan Heritage Trees area. Anthropogenic activities along coastal areas have contributed to the unwarranted discharge of toxic metals into mangrove swamps, posing risks to marine deposits and ecological environments. In this research, we studied the Isaka–Bundu tidal swamp area in the Niger Delta, which is an impacted mangrove creek located along the Bonny river, exposed to pollution pressures. The ecological risks (Er) of toxic metals in the sediments and water of the Isaka–Bundu tidal mangrove swamp followed a decreasing order (Cu > Zn > Cd > Cu > Pb > As), according to our results, while the potential ecological risk index (PERI) of the toxic metals in the sediments and water of the Isaka–Bundu tidal mangrove swamp can be said to have a very high ecological risk (PERI ≥ 600). The sediment pollution load index (PLI) was higher than 1 in all three analyzed stations, suggesting extremely toxic pollution. The enrichment evaluation shows that the studied stations have a moderate potential ecological risk of Cd, with the enrichment value for Pb showing low potential ecological risk. Our study shows that the Isaka–Bundu tidal mangrove swamp has a significant level of toxic metal pollution, which is evidence of the illegal activities performed in the Niger Delta.

Abstract:
The South African Police Services (SAPS) has a constitutional responsibility to prevent, combat, and investigate crime that would naturally occur within communities. It therefore stands to reason that, if the police are to fullfil this responsibility, community members should be involved. This paper examines the phenomenon of lack of community involvement in the fight against stock theft in Dr. Pixley ka Isaka Seme Municipality, Mpumalanga Province. To achieve this objective, it was necessary to first determine the extent to which community in the Dr. Pixley ka Isaka Seme Municipality is involved in the fight against of stock theft. A qualitative research approach was adopted for this study with the use of semi-structured in-depth interviews to collect data. The sample consisted of 22 participants selected through purposive sampling, who included farmers, herdsmen, Community Police Forum members, and community leaders. The study found that the community in the Dr. Pixley Ka Isaka Seme Municipality is not involved in the fight against stock theft and that this could mainly be attributed to the poor relationship between the South African Police Service and the community. It was also found that there are no regular meetings taking place to ensure that there is a common understanding regarding various stakeholders’ responsibilities in an attempt to combat stock theft within the area of study. There are no awareness campaigns to discuss the stock theft trends in Dr. Pixley Ka Isaka Seme Municipality area between the police and the community.

In order to illustrate the contribution of the Great Isaiah Scroll from Qumran (1QIsaa) to textual criticism, this paper presents six viable readings for Isaiah 1–39, i.e. variants that most likely represent the original Hebrew text. In assessing the cases of Isaiah 3 : 24 ; 9 : 16 [English 9 : 17] ; 18 : 7 ; 19 : 18 ; 21 : 8 ; 23 : 10, the author takes into account the textual character of the manuscript, the scribal habits of the copyist, the work of scholars and commentaries on the Book of Isaiah, recent translations of Isaiah, as well as the context and the overall sense of the passage. In all six instances — with the possible exception of Isa 19 : 18 — the reading in 1QIsaa is found to be compelling. These readings and similar ones should therefore be included as part of the main text of Isaiah (not the apparatus) in future editions of the Hebrew Bible, and part of the main text of Isaiah in future translations (not the footnotes).

ABSTRACT The bla OXA-51-like gene with an upstream ISAba1 (ISAba1-bla OXA-51-like gene) was originally found on the chromosomes of carbapenem-resistant or -susceptible Acinetobacter baumannii isolates. However, a plasmid-borne ISAba1-bla OXA-51-like gene has recently been identified in Acinetobacter genomic species 13TU and several A. baumannii isolates in Taiwan, and all of the isolates are carbapenem resistant. This study aimed to characterize the plasmids bearing the ISAba1-bla OXA-51-like gene and their significance in A. baumannii. Among the 117 ISAba1-bla OXA-51-like-harboring isolates collected from 10 hospitals in Taiwan, 58 isolates (49.6%) from 24 clones had the genes located on plasmids that likely originated from a common progenitor. Among the 58 isolates, four had additional copy of the ISAba1-bla OXA-51-like gene on their chromosomes. Based on the analysis of these four isolates, the plasmid-located ISAba1-bla OXA-51-like gene appeared to be acquired via one-ended transposition (Tn6080). The isolates with a plasmid bearing the ISAba1-bla OXA-51-like gene had higher rates of resistance to imipenem (98% versus 46.6%; P < 0.001) and meropenem (98% versus 69%; P = 0.019) than those with the genes chromosomally encoded, which is most likely due to increased gene dosage provided by the higher copy number of associated plasmids. Transformation with a recombinant plasmid harboring only the ISAba1-bla OXA-51-like gene was enough to confer a high level of carbapenem resistance to A. baumannii, eliminating the possible contribution of other factors on the original plasmids. This study demonstrated that the carbapenem resistance-associated plasmids carrying the ISAba1-bla OXA-51-like gene are widespread in A. baumannii strains in Taiwan.

Multidrug-resistant Klebsiella pneumoniae (MDR-KP) is a major public health problem that is globally associated with disease outbreaks and high mortality rates. As the world seeks solutions to such pathogens, global and regional surveillance is required. The aim of the present study was to examine the antimicrobial susceptibility pattern and clonal relatedness of Klebsiella pneumoniae isolates collected for a period of three years through pulse field gel electrophoresis (PFGE). Isolate IDs, antimicrobial assays, ESBL-production, and minimum inhibitory concentrations (MICs) were examined with the Vitek 2 Compact Automated System. IDs were confirmed by 16S rRNA gene sequencing, with the resulting sequences being deposited in NCBI databases. DNA was extracted and resistance genes were detected by PCR amplification with appropriate primers. Isolates were extensive (31%) and multidrug-resistant (65%). Pulsotype clusters grouped the isolates into 22 band profiles that showed no specific pattern with phenotypes. Of the isolates, 98% were ESBL-KP, 69% were carbapenem-resistant Enterobacteriaceae (CRE) strains, and 72.5% comprised the carriage of two MBLs (SIM and IMP). Integrons (ISAba1, ISAba2, and IS18) were detected in 69% of the MDR-KP. Additionally, OXA-23 was detected in 67% of the isolates. This study therefore demonstrates clonal diversity among clinical K. pneumoniae, confirming that this bacterium has access to an enormous pool of genes that confer high resistance-developing potential.

Moreover, the mission of the servant in Isaiah 61 is inherently holistic, as had been established early on in the book. This involves the covenant expectation to fulfill a range of righteous actions indicative of the good news of salvation. To argue that Isaiah 61 serves as a paradigm for Christian mission is, on one level, self-evident from Jesus' own reference to the text in Luke 4. However, this essay has a larger hermeneutical point. A Christian reading of Isaiah 61 in its own context complements what we learn from the New Testament's employment of the text. Isaiah 61 reveals a call placed upon all Christians, not just Jesus. This standard for mission is not only achievable by the Spirit of God (Isa 61:1) but is also to be emulated by all servants of the Servant as the progression of the book implies. As Christians pursue holistic, and distinctively Christian mission, the fundamental idea must be recalled—as servants of the Servant Jesus Christ, righteous action, in terms outlined in Isaiah 61 and its context, is a covenant obligation.

This study aimed to investigate the composition, abundance, distribution, and diversity of the phytoplankton community of the Isaka-Bundu waterway in Rivers state, which is a polluted tidal mangrove wetland. Phytoplankton was collected bimonthly from July to December 2021 at high tide from four stations according to APHA methods. The species diversity was calculated using standard indices. The total composition of 334 individual phytoplankton was identified in the 3 stations and control. The total composition of 220 species from 5 Phylum was recorded. This observed decrease in the species diversity and richness could also be attributed to changes in environmental variables due to pollution resulting from industrial effluent discharge into the river which has adversely affected the aquatic biota. Based on these activities, there is an urgent need to carry out a regular study on the phytoplankton community that supports its fisheries in this aquatic environment. The results of this study indicate the characteristic species and compositions of phytoplankton in the Isaka-Bundu waterway in Rivers state. The species abundance and distribution are a confirmation while the composition, and diversity gave an insight into the adverse effect of these cumulative activities. The result showed that the Isaka-Bundu waterway had been extremely polluted.

Recent studies on both the redactional and literary unity of Isaiah have emphasized the pivotal role of Isaiah 36–39 as a transitional bridge between the two major parts of the book of Isaiah. More specifically, a growing number of commentators have understood that Hezekiah is cast as an ideal role model and remains exemplary even in Isaiah 39. However, this interpretation of Hezekiah as an idealized king is an oversimplification and does not recognize the complexity of the character of Hezekiah portrayed in these chapters. The purpose of this article is to propose a synchronic reading of how Isaiah's prediction at the end of the royal narratives in the book of Isaiah plays a key role in providing a coherent evaluation of Hezekiah as a blemished monarch who used to be a success but has become a failure.

Researchers looked at the characteristics of Isaiah 45 and the concept of monotheism to get the implications of the concept of monotheism in Isaiah 45 for alternatives to Indonesian Christians. This type of research is qualitative descriptive research with the use of hermeneutic methods and socio-historical approaches. Researchers conduct a literature study of sources related to the research topic. The results explain that the implications of the concept of monotheism in Isaiah 45 show that Yahweh, who is God, is not exclusive in relationships with mankind. Based on the implications of the concept of monotheism in Isaiah 45, Christians can choose transformative inclusive attitudes in establishing relations with other religious people.

This article argues that it contains a prophetic call by Isaiah, speaking with God’s authority, that Jerusalem should welcome the Moabite refugees who have fled there for safety. Isaiah tells Jerusalem that by welcoming these refugees they are participating in the coming of a Kingdom and a Davidic King who will rule with justice, righteousness, love, and faithfulness.
