Abstract:
Internalisation of appearance ideals moderates the relationship between exposure to media images and body dissatisfaction. To date, the role of thin- and muscular-ideal internalisation in the context of social media remains under explored, particularly for boys. As such, we aimed to explore how social media use (Instagram and Snapchat) was related to body dissatisfaction, and whether thin- and muscular-ideal internalisation would moderate this relationship in a sample of 1153 adolescent boys and girls (55.42% males; Mage = 13.71, SD = 1.14). As hypothesised, social media use, and thin- and muscular ideal internalisation were positively correlated with body dissatisfaction in both genders. In moderation analyses, thin-ideal internalisation emerged as the only variable that had a significant effect on body dissatisfaction in both genders. Additionally, the influence of social media use on body dissatisfaction was moderated by muscular-ideal internalisation in boys, whereby for boys with high muscular-ideal internalisation, greater social media use was associated with greater body dissatisfaction. The two-way (muscular x thin-ideal internalisation) and three-way interaction (social media use x thin-ideal internalisation x muscular-ideal internalisation) effects on body dissatisfaction were non-significant. These findings emphasise the importance of considering the sociocultural environment (i.e., new media influences) as frameworks for understanding body dissatisfaction and suggest targeting of internalisation of appearance ideals in body dissatisfaction prevention programs.

Abstract:
Purpose – The purpose of this paper is to review the key analytical principles of internalisation theory as a general theory of the multinational enterprise (MNE). It illustrates the vitality, relevance and flexibility of the approach in explaining the continued evolution of the MNE. As a grounded social science theory, it provides, in combination with history and economic geography, satisfying and novel explanations of the key phenomena of the modern globalising economy. Design/methodology/approach – This paper examines the origins and principles of internalisation theory as the foundation theory of the MNE. It considers internalisation theory in the context of current and mainstream theories and concepts in the field of international business. Findings – Internalisation theory is equally valid for the MNEs of yesteryear as it is for those today. The theory continues to have strong explanatory power for MNE activity. Current research areas, such as multiple embeddedness, fine-slicing of the value chain, etc., and other theories, such as dynamic capabilities and the resource-based view, either are subsets of internalisation and thus explained by the theory, or contain weakness and/or inconsistencies not found in internalisation theory. Originality/value – This paper coherently synthesises internalisation theory, its origins and evolution. It shows how commonly held and current concepts and theories are related to internalisation theory or have weaknesses, thus making internalisation theory a superior theory to explain the MNE, and identifies potential applications of the theory to novel research areas in the field of international business.

Purpose The purpose of this paper is to extend the purview of internalisation theory in understanding the global allocation of resources in different institutional contexts. Design/methodology/approach Theoretical extensions of internalisation theory are proposed in response to a comment. Findings The traditional focus of internalisation theory can be extended beyond the normal market exchange context, first, by expanding the range of transactions; second, by including non-market actors; and third, by expanding the relevant welfare criteria. Originality/value Comparative institutional analysis is enhanced by expanding the consideration of the range of intermediate goods that are traded, the potential providers examined and extending the criteria of assessment beyond efficiency to include equity, sustainability and legitimacy. These extensions add value to the internalisation theory.

Purpose Although the European Foundation for Quality Management (EFQM) Excellence Model has been widely adopted throughout Europe, a thorough examination of the factors that contribute to the internalisation of the model (i.e. a substantive adoption) has been neglected in the literature. The purpose of this paper is to present a model that analyses the drivers of the real internalisation of the EFQM excellence model, with a focus on the role of motives for adoption, and appraisal and compensation systems. Design/methodology/approach An empirical study was carried out based on a sample of Spanish organisations that had been awarded EFQM recognition. Structural equation models, cluster analysis and ANOVA were used to examine the research questions. Findings Internal motives concerning the creation of a participative style are the main driver of internalisation. Moreover, having an appraisal system-oriented towards the development of employees helps the substantive adoption of the EFQM model. These findings reinforce the importance of the soft elements of the EFQM model. Originality/value This study enhances evidence about the motives for adoption and their influence on the internalisation of the EFQM model. It analyses internalisation in a novel context, EFQM recognised organisations, and contributes to the debate about the efficacy of the EFQM model to performance improvement, by unveiling the factors that could foster the internalisation of the model within the organisational routines.

Abstract:
The chemokine receptors CCR5 and CXCR4 are major co-receptors/receptors for the CD4-dependent and CD4-independent entry of human and simian immunodeficiency viruses. The chemokines that bind and activate these receptors can inhibit the entry of viruses that use the respective co-receptor molecules. Chemokine-induced co-receptor internalisation is a significant component of the mechanism through which chemokines inhibit virus entry. CXCR4 internalisation is induced by the CXCR4 ligand stromal cell derived factor-1 (SDF-1), phorbol esters and, in T cells, cellular activation. Here we show that CXCR4 endocytosis can be mediated through either one of two distinct internalisation signals. A COOH-terminal serine rich domain is required for ligand- but not phorbol ester- induced CXCR4 internalisation. However, a Ser/IleLeu motif, similar to that required for the endocytosis of CD4 and the T cell receptor/CD3 complex, is required for phorbol ester-induced, but not ligand-induced, CXCR4 endocytosis. By contrast, CCR5 internalisation is induced by the beta-chemokine RANTES but not by phorbol esters. CCR5 lacks the Ser/IleLeu sequence required for phorbol ester-induced uptake of CXCR4. Together these results indicate that distinct mechanisms can regulate CXCR4 and CCR5 endocytosis and trafficking.

Desmosomes are intercellular adhesive junctions providing architectural integrity to tissues that are subject to mechanical stress. During wound healing downregulation of desmosomal adhesion is necessary to facilitate tissue remodeling. Ultrastructural observations suggest that downregulation may occur by internalisation of whole desmosomes but the mechanism of downregulation is not understood. Protein kinase C alpha (PKCα) has been shown to colocalise with desmosomes at the wound edge and to modulate their adhesion. This thesis investigates the internalisation of desmosomes with particular reference to the roles of the cytoskeleton and PKC.

Abstract:
ELISAs were used to investigate the effects of a range of opioid ligands on the internalisation of the T7-tagged μ-opioid receptor (MOR) expressed in HEK 293 cells. The different opioid ligands were seen to vary in their abilities to induce MOR internalisation. The effect of the MOR agonists, DAMGO and morphine on MOR phosphorylation mutants (MOR-T180A and MOR-S363A) expressed in HEK 293 cells was also examined by ELISA. The ability of DAMGO to induce internalisation was significantly inhibited in the MOR-T180A in comparison to the wild type MOR. An N-terminal superecliptic pHluorin-tagged μ-opioid receptor (pHMOR) was generated to study MOR trafficking. Superecliptic pHluorin is a pH-sensitive variant of GFP which emits fluorescence at physiological extracellular pH (7.4) but not in acidic environments such as that occurring in intracellular organelles. In HEK 293 cells expressing pHMOR, fluorescence from pHluorin was only observed at the plasma membrane and this was abolished by lowering the extracellular pH to 6. Raising the intracellular pH with NH4Cl (50 mM) revealed fluorescence in intracellular compartments. The pHMOR construct was shown to be functional as indicated by the robust activation of GIRK channels as well as the rapid decrease in the level of cell surface pHMOR fluorescence following treatment with DAMGO. Total internal reflection fluorescence (TIRF) microscopy demonstrated that the surface expressed pHMORs were very dynamic and displayed characteristics which were not detected by confocal microscopy experiments such as receptor clustering following treatment with DAMGO or morphine. The agonist-activated receptors were also seen to colocalise with DsRed-tagged clathrin. The TIRF microscopy technique together with the pHluorin tagged MOR presents a very useful and promising approach to investigate dynamic changes in cell surface MOR expression in live cells.

Abstract:
Adventurous Training (AT) within Army Phase One organisations is used to assist in the development of British Army recruit core values . This study measured the internalisation of British Army recruit core values during the AT week at two separate Phase One training organisations. A pre-test, post-test design was used to evaluate recruit (n = 302) motivational internalisation of core values during a structured 5-day training week, where recruits undertook a mixture of rock climbing, caving, canoeing, kayaking, and hill walking activities, and were required to complete tasks in unfamiliar and challenging environmental conditions. Reflecting the influence of the training, Bonferroni corrected, pair-samples, ttests conducted on the Relative Autonomy Index were significant for the motivational internalisation of All core values and four of the six independent core values (Selfless Commitment, Courage, Loyalty and Respect for Others). Further examination at external, introjected and integrated regulations additionally revealed significant results for all core values with the AT week appearing to have the most robust effect on introjected regulation. A second hypothesis was concerned with the effects of the leadership of AT instructors in developing recruit core values and asked specifically whether high levels of transformational leadership behaviours were associated with an enhanced internalisation of core values. Fifty nine instructors took part in the study and four transformational leadership behaviours were hypothesised to be associated with greater gains in the internalisation of all core values. Analyses revealed mixed results regarding individual transformational leadership behaviours; however, individual consideration was found to be the most significant behaviour. The implications for training developments are discussed.

Le récepteur TSH est un récepteur couplé aux protéines G présentant des spécificités par rapport aux récepteurs homologues de la lutropine (LH) et folliculostimuline (FSH): (i) son volumineux domaine extracellulaire clivé en deux sous-unités (ii) son activité constitutive (iii) l'existence d'auto-anticorps stimulants dirigés contre le récepteur et impliqués dans l'hyperthyroïdisme. Le trafic intracellulaire du TSHR n'était pas documenté. Des lignées permanentes de cellules L exprimant ce récepteur ainsi que des anticorps monoclonaux dirigés contre le récepteur permettent d'étudier sa distribution et son endocytose en microscopie confocale et électronique. Le TSHR est initialement présent sur la membrane plasmique au sens strict et les puits recouverts de clathrine, au niveau desquels il est internalisé. L'internalisation constitutive représente 10% des récepteurs présents à la surface. L'hormone n'augmente que d'un facteur 3 cette endocytose. La majorité du récepteur internalisé est recyclé via des vésicules lisses alors que l'hormone est dégradée dans les lysosomes. Le recyclage est inhibé par la monensine et emprunte une voie indépendante de la cavéoline-1. Les études de microscopie conduites sur les thyrocytes humains en culture primaire montrent une polarisation baso-latérale du récepteur et une voie d'endocytose similaire. Le récepteur LH est lui internalisé massivement puis dégradé dans les lysosomes. Les essais de colocalisation avec le récepteur transferrine démontrent que les récepteurs homologues LH et TSH suivent un trafic intracellulaire différent dans le même système d'expression cellulaire. L'utilisation de récepteurs chimères des TSHR et LHR prouvent que les domaines transmembranaires et intracellulaires contiennent les informations responsables de l'orientation vers la voie de recyclage versus celle de dégradation. The TSH receptor is a G protein coupled receptor, with specific characteristics from the two high homologous lutropin (LH) and folliculostimulin (FSH) receptors (i) its large extracellular domain which is cleaved in two subunits (ii) its constitutive activity towards the cAMP transduction pathway and (iii) the existence of stimulating anti-receptor autoantibody implicated in hyperthyroïdism. Seant information is available on the intracellular trafficking of this receptor. Stahly transfected L cells expressing TSH receptor and anti-receptor antibodies were used to study by confocal and electron microscopies its cellular distribution and endocytosis. The TSH receptor was initially localized on the plasmalemma proper and in clathrin-coated pits. Lt was internalized through clathrin-coated vesicles. Constitutive endocytosis represented 10% of cell surface receptor molecules. Endocytosis was increased only 3 fold by the hormone. The majority of internalized receptor recycled to cell surface via smooth vesicles whereas hormone was degraded in lysosomes. This recycling was inhibited by administration of monensin and occurred via a caveolin-1 independent pathway. Microscopic studies repeated in primary cultures of human thyroid cells showed a baso-lateral distribution and a very similar endocytosis pathway. The LH receptor is endocytosed in high proportion and degraded in lysosomes. Colocalization studies with transferrin receptor confirmed that highly homologous LH and TSH receptors exhibit, when expressed in the same cells, very different cellular trafficking properties. The use of LHITSH receptor chimeras showed that transmembrane and intracellular domains contain information orienting the protein toward recycling or degradative pathways. The extracellular domain seems to play a role in the extent of internalization. These observations should now allow the determination of the molecular signals involved in these processes

Dans le cadre du developpement de vaccins sous-unites, l'immunogenicite de peptides synthetiques contenant un epitope t cytotoxique (np6) et un epitope t auxiliaire (t) issus de proteines du virus de la rougeole a ete etudiee. Le peptide chimere tt-np6 a pour interet d'induire une reponse t cytotoxique en absence d'adjuvants et de composes hydrophobes. Le mecanisme de reconnaissance d'un antigene par les lymphocytes t ainsi que les nouvelles approches vaccinales capables d'induire une reponse immune cellulaire sont decrits succinctement. Pour stimuler in vivo des lymphocytes t cytotoxiques (ctl), l'epitope t cytotoxique doit etre internalise dans le cytosol. C'est pourquoi nous etudions l'internalisation cellulaire et l'affinite pour la membrane plasmique des peptides tt-np6, t-np6 et tt. Les peptides marques par l'oregon green 514 ont ete localises par microspectrofluorimetrie confocale au niveau de la membrane cellulaire. Le peptide le plus immunogene tt-np6 se differencie des peptides tt et t-np6 par une internalisation plus profonde dans le cytosol et une accumulation de type vesiculaire. L'internalisation est directement reliee a la presence du peptide a la membrane plasmique. Des etudes physicochimiques ont ensuite montre que le peptide tt-np6 a une plus grande affinite pour les lipides, qu'il est plus helicogene et plus stable a la proteolyse que t-np6 et tt. L'etude approfondie de la structure de tt-np6 a mis en evidence une propension a s'autoassocier, ce qui favoriserait la resistance a la proteolyse, et elle a permis d'identifier une arginine qui serait responsable d'une perte d'helicite et dont la mutation ameliorerait l'activite biologique du peptide. Enfin, le peptide tt est etudie du point de vue de son affinite pour des alleles humains et de son helicogenicite afin de permettre son application a d'autres epitopes. Ce travail souligne l'importance des analyses physicochimiques dans l'elaboration de vaccins sous-unites capables de stimuler des ctls.

AbstractChapter 1 gave us a clear starting point: responsible companies create integrated value based on financial value (FV), social value (SV), and environmental value (EV). But what does that mean and imply? This chapter outlines what value creation is, and how the various types of value creation can be prioritised. For the alignment of all types of value, the prospect of internalisation is crucial. Internalisation means that the burdens of (social and environmental) externalities are increasingly shifted back from society to the companies and consumers who cause them. If companies’ FV depends on the exploitation of an external impact (i.e., FV at the expense of SV or EV), that FV will be affected if and when internalisation occurs.As responsible companies aim to create value based on FV, SV, and EV, they should have a clear picture of their current value creation profile and of their capabilities to create integrated value. Based on their purpose and area(s) of value destruction, companies can accordingly adjust their strategy and business model. In the case of serious value destruction, they should be able to outline a credible transition pathway. Companies can invest in their capabilities to adapt to sustainable business models. In that way, they increase their transition preparedness.

AbstractWith the introduction of the Construction Products Directive EU305:2011, architects are more than ever required to select products with a high proportion of renewable raw materials. Only then will it be possible to internalise, hence to compensate, for environmental damage caused by technologies that do not conserve resources and are energy-intensive. Using a novel bio-based wood-plastic composite (WPC) as an example, this chapter shows that internalisation can be successful if conventional fossil plastics are “greened” by adding renewable biomass. The results show that this is only the case if the bio-content is not too high but exceeds a minimum value. The degree of sustainability depends on the assessment of the extent of damage to the environment and society. The optimal level of internalisation can then become allocatively efficient, meaning that the costs of avoiding plastics do not increase more than the damage costs decrease. The findings demonstrate that environmental protection can also be economically meaningful and potentially contributes to increasing social welfare in society. A paradigm shift towards restorative economy in construction should take this principle into account.

Environmental charges to rail service operators are still at an early stage of implementation in Europe. Current schemes are dissimilar and most of them have a very limited scope or do not provide effective incentives to abate environmental impacts. This is due to several practical difficulties in implementing internalisation pricing schemes in the railway sector. The first difficulty arises from the lack of internalisation in other transport modes and the small competitive margin of railways. Increasing the overall level of rail charges could affect their survival. A second difficulty is due to the uncertainties in the valuation of external costs and in the establishment of the optimal level of charges. On top of that, the imperfect competition in the rail market and its operational and financial rigidities imply that pricing decisions could be unfair and produce undesired demand effects. These difficulties should not prevent, however, advancing towards the application to rail of adequate charging methods for its environmental costs. This paper addresses the problem of setting environmentally differentiated rail charges through an analytical approach. A generalised formulation is developed that sets the level of charges as a function of the degree of internalisation in other modes. Then, the optimal trade-off in the level of differentiation is assessed to extract general guidelines. Abstract All human activities generate negative externalities, in particular the use of radioactive material for electricity production and radioisotope applications. Both activities produce radioactive waste, which can, therefore, be considered as being specific externalities. The purpose of the paper is to investigate these externalities and to identify appropriate internalisation instruments. Analogue cases in environmental management are discussed. In general the nuclear externalities are not internalised in the management costs charged by Radioactive Waste agencies (RAWA). The paper explores the possibility of having an internalisation of all costs as requested by the strict application of the Polluter Pays Principle. In the case of electricity production a comparison is made between the externalities attributed to nuclear waste and those in relation with CO2-emissions from the combustion of fossil fuel. A brief overview is given on the evaluation approach in ExternE (“Externalities of Energy”). The evaluations are the basis for the design of a carbon tax applicable to fossil fuels for reducing CO2-emissions. A similar tax could be charged on radioactive waste management. Beyond the internalisation objective, the tax proceeds could finance the technological R&D for improving the conditions of storage and disposal, and provide compensations to local residents in the vicinity of nuclear waste management facilities. The management of spent radioisotope equipment in medicine, research, or industry is shown to have similar features to the management of packages, spent electrical appliances, and the disposal of batteries. In general the price of management of the spent material is not included in the purchase price. In case of spent radioisotope equipment, the externality mainly represents the risk of this material becoming a hazard for the public health. It is recommended to internalise the full costs of management to eliminate this risk. Moreover spent material should be registered and RAWA should maintain detailed inventories on their national territories. In order to induce the free return of spent material to the RAWA, deposit refund systems could be set in place as in the package or battery market. A surcharge is paid by purchase, which is refunded to the buyers when they return the product for recycling or proper disposal. The paper concludes by describing lessons and possible implications of the previously discussed environmental tax or surcharge systems on the way the Polluter-Pays Principle is applied in radioactive waste management.

As widely argued [HG97; Sat96], transitive roles play an important role in the adequate representation of aggregated objects: they allow these objects to be described by referring to their parts without specifying a level of decomposition. In [HG97], the Description Logic (DL) ALCHR+ is presented, which extends ALC with transitive roles and a role hierarchy. It is argued in [Sat98] that ALCHR+ is well-suited to the representation of aggregated objects in applications that require various part-whole relations to be distinguished, some of which are transitive. However, ALCHR+ allows neither the description of parts by means of the whole to which they belong, or vice versa. To overcome this limitation, we present the DL SHI which allows the use of, for example, has part as well as is part of. To achieve this, ALCHR+ was extended with inverse roles. It could be argued that, instead of defining yet another DL, one could make use of the results presented in [DL96] and use ALC extended with role expressions which include transitive closure and inverse operators. The reason for not proceeding like this is the fact that transitive roles can be implemented more efficiently than the transitive closure of roles (see [HG97]), although they lead to the same complexity class (ExpTime-hard) when added, together with role hierarchies, to ALC. Furthermore, it is still an open question whether the transitive closure of roles together with inverse roles necessitates the use of the cut rule [DM98], and this rule leads to an algorithm with very bad behaviour. We will present an algorithm for SHI without such a rule. Furthermore, we enrich the language with functional restrictions and, finally, with qualifying number restrictions. We give sound and complete decision proceduresfor the resulting logics that are derived from the initial algorithm for SHI. The structure of this report is as follows: In Section 2, we introduce the DL SI and present a tableaux algorithm for satisfiability (and subsumption) of SI-concepts—in another report [HST98] we prove that this algorithm can be refined to run in polynomial space. In Section 3 we add role hierarchies to SI and show how the algorithm can be modified to handle this extension appropriately. Please note that this logic, namely SHI, allows for the internalisation of general concept inclusion axioms, one of the most general form of terminological axioms. In Section 4 we augment SHI with functional restrictions and, using the so-called pairwise-blocking technique, the algorithm can be adapted to this extension as well. Finally, in Section 5, we show that standard techniques for handling qualifying number restrictions [HB91;BBH96] together with the techniques described in previous sections can be used to decide satisfiability and subsumption for SHIQ, namely ALC extended with transitive and inverse roles, role hierarchies, and qualifying number restrictions. Although Section 5 heavily depends on the previous sections, we have made it self-contained, i.e. it contains all necessary definitions and proofs from scratch, for a better readability. Building on the previous sections, Section 6 presents an algorithm that decides the satisfiability of SHIQ-ABoxes.
