Full textThis study has as its aim to show the importance of the awareness of the intermediary managership, through environmental education, to actuate as multiplying agent in the implantation of a environmental management system(SGA) at a company of metropolitan trains. As a method, the option is the study of occurrence, with documental analisis, watching the work sections and questionnaire application. The survey was realized at São Paulos Company of Metropolitan Trains(CPTM), in the period of time between March 2004 and November 2005, during preparatory stage for the implantation of the SGA. The CPTM is a company of São Paulos States government, and it is responsible for the operation of the collective transportation above railroads in the metropolitan region of the city of São Paulo. The surveyed were 36 multiplying agents(intermediary managership), divided in 18 maintenance supervisors, 02 of equipment and 16 general heads of stations. They receive training about concepts of environmental awareness, that covers the environmental legislation that is in force in the country ant the most important environmental aspects and impacts generated at CPTM. After the training, they impart the acquired knowledges to their teams, in which they perform the role of leadership. Nine months later, it is applied a questionnaire for the intermediary managership to answer, that covers, among other aspects, the attitudes and actions to be taken in face of environmental issues. Among the most significative results, these stand out: (1) the improvement in the rational use of water in 25%, for the use of electrical energy in 22,2% and for the use of paper in 19,4%; (2) 86,1% of the multiplying agents notified, after imparting the training about the environment, between average and a lot of change in the attitude of their teams facing environmental issues. The results show the efficacy of using the intermediary managership as multiplying agent in the implantation of a environmental managership system at CPTM.

Full textThe 10,000 travel agencies in Germany and 4,000 travel agencies in France shape the tourist landscape and play an important role for the tourism industry in both countries. We analyse the development of the number of travel agencies in France and Germany in order to identify factors that may explain the heterogeneity of the two systems. In this context, France can be seen as an exception: in comparison to many other countries, the number of French travel agencies has not decreased during the last decade. Thus, the French system challenges the relationship (often considered a causal one) between the rise of e-tourism and the decrease of travel agencies. By means of quantitative and qualitative methods we compare legal, spatial, technological and cultural factors in the two countries. We show that differences in the legislation affect the travel agency structure. Furthermore, French travel agencies are more likely to have a receptive character which means that spatial factors of the two countries can be different. Our study also indicates that e-tourism leads to a growth of specialized travel agencies. Furthermore, cultural differences between travel agency customers are smaller than expected. We present examples which demonstrate that the tourism press conveys a misleading impression regarding this topic and reinforces stereotypes
Die 10 000 Reisebüros in Deutschland und 4 000 Reisebüros in Frankreich formen die touristische Landschaft und spielen eine wichtige Rolle im Vertrieb touristischer Dienstleistungen. Wir analysieren die Reisebürodynamik in Frankreich und Deutschland und identifizieren Faktoren, welche die heterogene Entwicklung der beiden Systeme erklären können. Frankreich eignet sich dabei als Vergleichsland, da die Anzahl französischer Reisebüros im Vergleich zu vielen anderen Ländern in der letzten Dekade nicht abgenommen hat. Das gibt Anlass, den häufig als kausal beschriebenen Zusammenhang zwischen der Zunahme des e-Tourismus und der Abnahme der Anzahl Reisebüros zu hinterfragen. Mit Hilfe von quantitativen und qualitativen Methoden vergleichen wir politisch-juristische, räumliche, technische und kulturelle Einflussfaktoren auf die Reisebürostruktur der beiden Länder. Wir zeigen auf, dass vor allem Unterschiede in der Gesetzeslage die Reisebürostruktur beeinflussen. Auβerdem nehmen französische Reisebüros in Teilen eine rezeptive Rolle ein, so dass die räumlichen Einflussfaktoren Unterschiede aufweisen. Unsere Studie weist darauf hin, dass der e-Tourismus eine Profilschärfung der Reisebüros in Richtung Spezialist bewirkt. Die kulturellen Unterschiede zwischen den Reisebürokunden der beiden. Länder sind kleiner als erwartet. Wir zeigen anhand von Beispielen, dass hier die touristische Fachpresse einen falschen Eindruck erweckt und Stereitypen zwischen den beiden Ländern verstärkt

Full textThe increase in antibiotic resistant bacterial and fungal infections and the prevalence of oxidative stress-related conditions including cancers, cardiovascular diseases and diabetes has led to a consensus among pharmaceutical companies, clinicians and researchers that novel antimicrobial and antioxidant approaches are needed. These should be ideally efficacious, non toxic, easily accessible and affordable. There has been an increased interest in the identification of medicinal plants that possess both these bioactivities in an intrinsically related manner, allowing the simultaneous prevention of these ailments. Two South African herbal teas, rooibos and honeybush have been associated with a long history of medicinal use, hence their consideration for the current study. Numerous studies have been performed to evaluate the antioxidant activities of these South African herbal teas, however limited information about their antimicrobial activity currently exists.

Full textThis study intends to examine the impact of a well known debate page in one of Sweden’s biggest newspapers, Dagens Nyheter. We have done this by analysing Ekot, Aktuellt, Göteborgs Posten and Västerbottens-Kuriren to see to what extent they “take” news from the debate page.
The method used was content analysis and to describe some of our thoughts about how we placed the hits in different categories we chose eight of our hits to exemplify.
The material from the newspapers, radio and television was examined to discover the potential impact from the debate page.
To carry out this study we have mainly used theories about how media get influenced by each other’s news and how they do follow-ups and refer to the original media. Other theories of importance for our study are for example those about what kinds of events that become news and how media influences people’s thoughts about what’s important.
One of the most important conclusions we were able to draw after carrying out our study was that the debate page in Dagens Nyheter has quite a small impact on what our chosen media reported on. The biggest impact was found in the newspapers, whereas radio and television very seldom referred to the debate page.
To sum up, one can absolutely say that the debate page in Dagens Nyheter is powerful and do have impact on other media, but an article published on the page is not automatically discussed and debated in other media during the day.

Full textThe thesis here presented is focused on the molecular study of three human proteins related to conformational diseases, or Amyloidoses. The mechanisms of native folding and aggregation in a cross-β sheet structure have been described using several spectroscopic techniques. These two pathways are competing by the presence of intermediate states of the reaction. An oligomeric intermediate is the precursor of the β-aggregates and is considered the main responsible of the cellular toxicity in many amyloid diseases. Therefore, is necessary to perform a deeper study of the oligomeric state to delve into the etiology of these human diseases. On one hand, the expression and extraction of a single-chain variable fragment (scFv-h3D6), a derivative of an antibody specific for Aβ-oligomers, was performed. Here is the first description of a conformational mechanism by which a scFv-h3D6 withdraws Aβ1–42 oligomers from the amyloid pathway and, consistently, it prevents Aβ-oligomer toxicity in the SH-SY5Y human neuroblastoma cell line. Moreover, we modeled the 3D-structure of scFv-h3D6, obtained by sequence homology. This model has allowed to a rational redesign of the molecule in order to improve its stability, increase its recombinant production, and decrease the effective dose already demonstrated in a mice model of Alzheimer's disease. For another hand, we studied the AL-12 protein, an Ig variable domain (VL) obtained from a patient diagnosed with light chain Amyloidoses. This amyloidogenic domain shows seven non-conservative mutations from his germinal protein κI O18/O8. The thermodynamic study of AL-12 and four restoratives mutants, allowed us to determine the effect of somatic mutations on the VL stability. Thereby, the restoration of a residue toward the germinal sequence implies a destabilization of the domain and increases the aggregation tendency. Here we demonstrate that a single somatic mutation couldn’t be the cause of the amyloidogenic potential of AL-12, rather it is due to several factors. The destabilization of the native folding promotes a conformational reorganization more susceptible to β-aggregation. Finally, we work with PSD95-PDZ3 domain, a neuronal post-synaptic protein and related to human disease such as Alzheimer or Autism. PDZ3 follows a reversible aggregation pathway, a feature which is relevant to determining the conformational changes that promote it. In addition, the design of six variants of PDZ3 has helped determine the regions involved in the folding and stability of the global PDZ3 domain and its role in the aggregation process. This work demonstrates the importance of the states in the intermediate conformational space of proteins, their relationship with conformational diseases, and the application of knowledge in the development of effective therapies.

Full textInformation and communication technology (ICT) radically affects the tourism industry by overturning its production and distribution vertical value chain. Direct exchange between tourist and suppliers is the beginning of a new era. ICT has also given the opportunity to many new players to introduce themselves into tourism market. From now on, travel agencies and tour operators are not the only ones able to connect supply with demand. Internet has also contributed in changing the tourist’s demands and activities. Tourism is no longer limited to a mass product. The tourism intermediaries must evolve in a less favourable environment. The question is about their place and value creation in the industry. A literature review allows us to understand that the tourism intermediary is present to overcome market’s imperfections and propose value through the following functions: informational, expertise, assurance, economic, innovation and networking. If this literature review shows that the intermediation functions are value creators, it does not necessarily claims that all functions are always carriers of value creation. Therefore, we analyse the intermediaries’ value propositions by trying to find the functions they perceive as being important to cope with tourism evolution. Consequently, we will complete our theoretical analysis with an empirical observation to identify the functions on which the intermediaries rely the most to face the evolution of the tourist market. By means of statistical analysis of our semi-structured interviews, we show in particular that: the expertise and assurance are the most important value propositions of tourism intermediaries; tour operators take less financial risks except for owning a competition-free offer. Furthermore, specialisation and destination management are revealed as strategic choices

Full textDesde que a inicios de la década de los años setenta los investigadores Maxwell McCombs y Donald Shaw publicaran el trabajo seminal de Chapel Hill sobre la teoría de la agenda-setting (AS), sus académicos han abordado, fundamentalmente, las investigaciones mediante dos herramientas metodológicas: una primera fase de análisis de contenido, empleada para estudiar la cobertura noticiosa y, por ende, la prominencia o saliencia de determinados temas o atributos en la agenda; y un segundo estudio de naturaleza estadística, basado en la verificación del nivel de correlación existente entre los temas, sus emisores o el global de las distintas agendas. El análisis de los contenidos generalmente se ha enfocado en la importancia relativa o comparativa de algunos temas sobre otros. Se supone que algunos medios o actores sociales están más ocupados que otros en determinados temas, lo que ha llevado a un estudio general del volumen relativo de información, en detrimento del análisis detallado del contenido. Tales diferencias (o similitudes) se han evaluado tradicionalmente a partir de la comparación de las frecuencias de sus contenidos principales, que, por medio de las correlaciones de Spearman entre los emisores, han fomentado un análisis sobre el ordenamiento jerárquico, despreciando las diferencias relativas o 2/3 absolutas y, por lo tanto, cualquier otra información. A la vista de esta necesidad, y para resolver estos procedimientos cuando el interés reside en la importancia relativa de las partes de un todo, la tesis aborda el análisis de Datos Composicionales (CoDa), fundamentado por Aitchinson en 1982, junto con la representación gráfica a través de biplots, impulsada por Gabriel en 1971, para el estudio y mapeo de la teoría de la AS. El interés en CoDa reside en la proporción de cada parte o componente, ya que las cantidades absolutas son irrelevantes y sólo exponen el tamaño de la muestra. Un problema muy similar en todos los aspectos al que se establece en el estudio de la AS, donde los datos absolutos son irrelevantes y en su mayoría apuntan sobre la temática del contenido, la intencionalidad del mismo, la popularidad del emisor, etc. En esta línea, sólo las proporciones de cada categoría de contenido o el tamaño relativo de un tipo de contenido sobre otro, son verdaderamente informativos. De acuerdo con estas premisas, la presente tesis doctoral se estructura en cinco artículos que distinguen la originalidad del estudio en presentar el CoDa-biplot como herramienta para el mapeo y estudio composicional de la teoría de la AS. Sirviéndose del período de las Elecciones Generales en España de 2015, la tesis se presenta como una unidad temática que aborda, a lo largo de cinco publicaciones, el mapeo y estudio composicional de la teoría de la AS de la opinión sobre política en el sistema mediático español (agenda de los medios), así como de las comunicaciones de carácter político en Twitter de los principales partidos españoles (agenda política). Asimismo, el estudio agregado de ambas agendas (la de los medios y la política) nos permite abordar el análisis y representación de la intermedia AS

Full textThe purpose of this thesis is to clarify what a football agent does in Sweden and describe how legitimacy is created in an industry that is associated with crime and a lack of transparency. Qualitative interviews have been used to answer the two main issues that follows: What do football agents do in Sweden and what services do they offer football players and football clubs? And how do football agents construct legitimacy in an industry with a bad reputation? The respondents were picked from three different professions: three footballers, three football agents and three sports directors. The result shows that football agents in Sweden mainly worked with contracts negotiation, professional contact creation, scouting and giving career advice. Some agents even offered their players the opportunity for extra training sessions, match analysis and rehabilitation. The result also shows that some agents in Sweden uses work methods that could be considered unethical, for example giving bribes to players. When it came to how agents constructed legitimacy, it was mainly about creating a trust between themselves and other actors in Swedish football. This was done mostly through gaining external approval from other actors in football. In addition, it was about following the regulations that exist and staying away from anything that can be associated with the shadier side of the football agent industry.

Full textIn the laboratory, investigations on neurofilament biology and on their interactions with other cytoskeleton components showed tubulin-binding sites (TBS) on different intermediate filaments. Among the synthesized peptide corresponding to these sequences, the NFLTBS.40-63 peptide derived from the neurofilament light subunit (NFL), revealed in vitro and in vivo targeting properties and anti-tumour effect on glioblastoma cells, the most frequent and aggressive brain tumour, for which current treatments show very limited effects. The therapeutic value of this peptide is reinforced by its lower uptake and the lack of a major cytotoxic effect in healthy cells (astrocytes, neurons). Works in this thesis describe the uptake mechanisms of the NFL-TBS.40-63 peptide in glioblastoma cells, to better understand its selective internalization. Its specificaction on glioblastoma cells has been completed by the analysis of the peptide effects on the mitochondrial organization and function. Finally, we show that the peptide targets neural stem cells, which could be at the origin of glioblastoma, and serve as a therapeutic tool inseveral brain diseases, such as traumatic injuries and neurodegenerative diseases. Altogether these works indicate that this NFL-TBS.40-63 peptide is a promising tool for therapeutic strategies of glioblastoma. It can also target neural stem cells in order to develop new treatments for various brain disorders

Full textThe present thesis was based on Grounded Theory (Strauss & Corbin, 1998) principles in order to address the lack of documented strategies implemented by “intermediate agents” to promote school practitioners’ use of research-based evidence. The term “intermediate agent” refers to the individuals who are positioned at the interface between scientific knowledge producers and users, and who encourage and support users’ uptake of research-based evidence in their practice. The study is part of the Quebec Ministry of Education, Recreation and Sports’ project aimed at improving success rates of secondary school students in underprivileged areas. Those solicited to participate in the study were intermediate agents from different levels of the educational system who had obtained the mandate to transfer research-based evidence to practitioners in schools targeted by the project. Snowball sampling (Biernacki & Waldorf, 1981; Patton, 1990) was used to identify individuals who were recognized by their peers for the quality of the support they offered to school practitioners in applying research. Sixteen semi-structured interviews were conducted. Analysis of the data yielded an intervention model coined Knowledge Transfer Intervention Theory, composed of 32 strategies of influence that were categorized into 6 intervention components: Relational, Cognitive, Political, Facilitative, Evaluative, as well as Continuous Support and Follow-up. The findings suggest that strategies embedded in the Relational, Cognitive, and Political components are interdependent and aim to establish a favourable climate, allowing agents to exercise a greater influence towards potential users’ ownership of the knowledge utilization process. Moreover, the Continuous Support and Follow-up component is key to sustaining changes in school practitioners’ use of research. Theoretical implications linked to the model, along with underlying explanations of the mechanisms involved in the different components are compared with the literature in knowledge transfer in the sectors of both health and education. Reports from related disciplines (notably in psychology) are also exposed. Finally, recommendations for practice are proposed.

Nowadays, social movement activists tend to manage their own online media during the movement period. Activists could not only use online media as a platform to communicate the public, but also could use it to influence mass media’s coverage. Past studies on agenda-setting showed that media could set each other’s agenda in the context of election. However, there are few studies focused on the context of social movement. Therefore, this study focused on the case of Shihlin Urban Renewal to examine the flow of agenda between mass media and online media, whether online media owned by SMO could influence mass media’s coverage, or mass media still has its agenda-setting ability on others. The current study raised 5 research questions to examine the event theme, frame, source, standpoint used by mass media and online media, and the flow of agenda between these two media. Content analysis was adopted to analyze print press (CHINA TIMES and Liberty Times) and online media (Facebook fan pages managed by the pros and cons during this movement), and the sampling time began from January 28th in 2012 to March 28th in 2013. The result showed that the theme of law and politics were the most emphasized event themes of mass media during the movement, while online media stressed on the themes of protest and attribution of the whole event. Second, both media spent lots of space discussing treatment recommendation of urban renewal and this movement. Third, mass media tended to quote different sources in a single coverage, but online media preferred to quote sources that supported their action. Fourth, the mass media coverage showed its sympathy toward the Wang family while online media holded distinct standpoint. Last, the finding showed that during social movement, mass media could set online media’s news themes, but online media could decide mass media’s frame of the agenda. These results suggest that under the context of social movement, mass media and online media both has their influence on each other.

The asymmetric Reformatsky reaction is a versatile and straight forward approach to obtaining chiral alcohols ( -hydroxy esters), a motif found in biologically active compounds and synthetic intermediates. Although a variety of asymmetric Reformatsky reactions have been developed. In our study the compound 3 with -hydroxycarboxylic acid was reported as an important intermediate of triazole antifungal agent. We found the Reformatsky reaction with acetophenone (Ketone) was carried out under the metal-halogen exchange reaction of the α-Bromo oxazolidinones with diethyl zinc and catalyst loading of Ni(acac)2. And we obtained that the best diastereoselectivity up to 97%, which provide a practical method to synthesize the new triazole antifungal agents.

Because of the growth of new-type intermediate traders and the increase of independent visitors, the travel agency is being divided gradually. In recent years, tourism industry is being upgraded by big data’s applications which are all the rage. Big data analysis which is different from traditional databases can predict customers’ demands and offer product experiences. By doing so, it can offer a new opportunity for conventional travel agencies. This research bases on Resource-Based Theory (RBT) and uses big data applications as intervening variables. By observing these cases and gathering the second-handed information by interviewing the managers of three different kinds of travel agencies, this research studies what kinds of core capabilities which travel agencies should possess. Besides, this research also studies the competitive relationship between traditional ravel agencies and new-type intermediate traders. By reference reviews and cases interviews, this research sorts out conventional travel agencies’ key factors of success in developing e-commerce area. The top issue for travel agencies is to know their own advantages of sources. Moreover, travel agencies should further enhance and raise their four new core capabilities which include tourism professions, social network’s asset managements, strategies of brand data and integrations between upstream and downstream sources by using their own advantages. After possessing these core capabilities, travel agencies should manage social network’s asset by analyzing brand data. Furthermore, they should also integrate corporations’ sources and improve performance systems. Ultimately, they should take online to offline(O2O) and Co-opetition strategies to build a new industry’s ecosystem. This method is the main key point of enhancing organizations’ performance and reintermediation for traditional travel agencies.

Teamwork has been regarded as essential basic skills of the 21st century citizens. Collaborative problem solving ability has begun to attach importance to international , which PISA (Programme for International Student Assessment) to implement the collaborative problem solving computerized assessment in 2015, the object is for 15 years of age students. This study aimed at understanding on collaborative problem solving performance situation in 3rd & 4th graders of elementary school, therefore develop suitable computerized assessment for them. In this study, based on PISA 2015 collaborative problem solving framework design topics, in order to reduce real dialogue emotions and personality traits error to get a more accurate student ability, test method using one on one human-computer interaction options dialogue computerized test, two units were designed "classes uniform design" and "two three-legged". The results of this study are as follows: 1. Develop suitable computerized assessment for student in 3rd & 4th graders of elementary school and can effectively measure the collaboration problem solving. 2. We can find most CPS skills common answer correct rate has more than 0.5。 3. The analysis showed most CPS skills are in high correlation. 4. Most CPS assessment analysis showed little difference A fourth-grade performance is better than B fourth- grade performance。We can not rule out the possibility of the gap between urban and rural schools and culture deprivation. And the reading understanding indirectly affect the ability of collaborative problem solving.

The main purpose of this study is based on PISA 2015 cooperative problem- solving assessment framework, aims to establish interactive and cooperative problem-solving online assessment questions for one on two, human- agent machine, and find out what collaborative skills that elementary school students are lack of. The results of the research can be provided as a reference in teaching, which are as bellow. 1. This study successfully builds the online cooperative problem-solving assessment for elementary school students. 2. The results have shown that among the three core competencies in cooperation performance for students, students performed better in “Establishing and maintaining shared understanding” and “Establishing and maintaining team organization” , but has a relatively poor performance in “Taking appropriate action to solve the problem.” The result indicates that students should strengthen the abilities to set up a plan and act it out when they encounter problems. 3. The results have shown that in the course of the four problem-solving performance, students performed best in “Represent and articulate”; the second is “Explore and understand”; the third is “Plan and execute”, and the worst is the part of “Monitor and reflect”. Therefore, it suggests that the training of “Monitor and reflect” needs to be strengthened. 4. The results have shown that the CPS’s ability of elementary school students in the city is much more than“better” than that of the rural areas, but there are significant gap and differences. This research confirms that there is indeed the existence of the Urban-Rural differences.

Since social media platforms have been widely applied in election campaigns as candidate-controlled media tools, candidates expect their social media outlets being influential when employing strategic actions to affect the public perception and the mass media reports in not only the topics, but also the attributes of reports. For which we know the effect as the intermedia agenda setting. This study examined whether candidates’ social media influence the news’s coverage in framing two leading candidates in both news frames and affective tones during the 2014 Taipei mayoral election, giving evidence to support both first-level and second-level agenda setting theory. With content analysis of two leading campaign issues during the election, MG149 account controversy and wiretap case, results showed strong correlation between the candidates’ Facebook posts and four major newspaper coverage in the quantity of news frame, but results for in affective tones were not confirmed. Intermedia agenda setting effect is proved occurring in both Ko’s and Lien’s campaign, since the quantity of four categories of frames (issue, image, strategy, and ideology frame) in two campaigns members’ Facebook post are aligned with those in newspaper. However, the results for affective attributes differ in each team. Ko’s team members determined a large part of the media frame at both positive and negative frame, showing that second-level intermedia agenda setting effects occurred in his campaign, while Lien’s didn’t have significant correlation with news coverage in affective dimension, only his team member apparently transferred a powerful negative frame to mass media in wiretap case.
