Abstract:
This thesis addresses the issues faced by the urban poor in their efforts to gain housing in developing countries. It argues that participation in housing is a matter of governance, in which the Public, Private and Popular sectors interact, and Intermediary Agents (IA) are necessary to act as catalysts in this interaction. In Indonesia there are two major problems related to housing the urban poor. The first problem concerns the quantity of housing. The second problem concerns the quality of housing, infrastructure and the environment. The government usually addresses these problems through a project-based, sectoral approach that lacks community engagement and accountability. Intermediary Agents have assisted many of these housing projects. However, little research has been undertaken on their role in these housing projects, as well as in the informal housing process. This thesis examines the role of IAs in the housing process of four different communities: one community in a government-sponsored project and three communities in informal settlements. The findings showed that the IAs, in engaging with the four different communities, played different roles and have different values. The involvement of IAs in the housing process elucidates a fundamental issue. On one hand, the IAs offer a mechanism to assist communities in providing and improving access to housing, infrastructure and services. However, there are limitations to their role: they have not the capacity to improve security of land tenure for the residents, which is essential for sustainable housing development. Where there has been some recognition of residents' rights to occupy the land, there have been substantial gains in community engagement in investing and improving their living environment. The thesis demonstrates the importance of community participation and acknowledges stakeholder engagement and community empowerment are crucial, roles that are often dependent on the involvement of IAs. It also extended the research on movement and development IAs demonstrating that both functions are complementary and important in housing delivery and that there are five roles that these IAs could play depending on the housing situation. The thesis offers new approaches to housing delivery and policy in Indonesia by involving urban poor communities, as the beneficiaries of housing for the poor programs. The qualitative method of the research has not been able to provide a generalised conclusion, but the lessons learned are important not only for providing housing for the urban poor in Indonesia but also possibly to addressing housing problems in other Asian countries. This thesis addresses four areas of intervention in transposing skills for a more democratic approach in housing the poor. Within these areas the research proposes new models of engagement for relevant stakeholders in public, popular and private sectors involving Intermediary Agents. These models are underpinned by a set of basic principles to guide IA engagement. Lastly the thesis proposes three models for IAs involvements in the housing process.

Abstract:
The 10,000 travel agencies in Germany and 4,000 travel agencies in France shape the tourist landscape and play an important role for the tourism industry in both countries. We analyse the development of the number of travel agencies in France and Germany in order to identify factors that may explain the heterogeneity of the two systems. In this context, France can be seen as an exception: in comparison to many other countries, the number of French travel agencies has not decreased during the last decade. Thus, the French system challenges the relationship (often considered a causal one) between the rise of e-tourism and the decrease of travel agencies. By means of quantitative and qualitative methods we compare legal, spatial, technological and cultural factors in the two countries. We show that differences in the legislation affect the travel agency structure. Furthermore, French travel agencies are more likely to have a receptive character which means that spatial factors of the two countries can be different. Our study also indicates that e-tourism leads to a growth of specialized travel agencies. Furthermore, cultural differences between travel agency customers are smaller than expected. We present examples which demonstrate that the tourism press conveys a misleading impression regarding this topic and reinforces stereotypes

Abstract:
Thesis (MTech (Biomedical Technology))--Cape Peninsula University of Technology, 2015.
The increase in antibiotic resistant bacterial and fungal infections and the prevalence of oxidative stress-related conditions including cancers, cardiovascular diseases and diabetes has led to a consensus among pharmaceutical companies, clinicians and researchers that novel antimicrobial and antioxidant approaches are needed. These should be ideally efficacious, non toxic, easily accessible and affordable. There has been an increased interest in the identification of medicinal plants that possess both these bioactivities in an intrinsically related manner, allowing the simultaneous prevention of these ailments. Two South African herbal teas, rooibos and honeybush have been associated with a long history of medicinal use, hence their consideration for the current study. Numerous studies have been performed to evaluate the antioxidant activities of these South African herbal teas, however limited information about their antimicrobial activity currently exists.

Kirke, Alexis. "Application of intermediate multi-agent systems to integrated algorithmic composition and expressive performance of music." Thesis, University of Plymouth, 2011. http://hdl.handle.net/10026.1/895.

Full text
Abstract:
We investigate the properties of a new Multi-Agent Systems (MAS) for computer-aided composition called IPCS (pronounced “ipp-siss”) the Intermediate Performance Composition System which generates expressive performance as part of its compositional process, and produces emergent melodic structures by a novel multi-agent process. IPCS consists of a small-medium size (2 to 16) collection of agents in which each agent can perform monophonic tunes and learn monophonic tunes from other agents. Each agent has an affective state (an “artificial emotional state”) which affects how it performs the music to other agents; e.g. a “happy” agent will perform “happier” music. The agent performance not only involves compositional changes to the music, but also adds smaller changes based on expressive music performance algorithms for humanization. Every agent is initialized with a tune containing the same single note, and over the interaction period longer tunes are built through agent interaction. Agents will only learn tunes performed to them by other agents if the affective content of the tune is similar to their current affective state; learned tunes are concatenated to the end of their current tune. Each agent in the society learns its own growing tune during the interaction process. Agents develop “opinions” of other agents that perform to them, depending on how much the performing agent can help their tunes grow. These opinions affect who they interact with in the future. IPCS is not a mapping from multi-agent interaction onto musical features, but actually utilizes music for the agents to communicate emotions. In spite of the lack of explicit melodic intelligence in IPCS, the system is shown to generate non-trivial melody pitch sequences as a result of emotional communication between agents. The melodies also have a hierarchical structure based on the emergent social structure of the multi-agent system and the hierarchical structure is a result of the emerging agent social interaction structure. The interactive humanizations produce micro-timing and loudness deviations in the melody which are shown to express its hierarchical generative structure without the need for structural analysis software frequently used in computer music humanization.

Gordon, Gregory Ernest Robert. "A chemo-enzymatic process for the production of beta-thymidine, a key intermediate in antiretrovirol manufacture." Thesis, Nelson Mandela Metropolitan University, 2010. http://hdl.handle.net/10948/d1016217.

Full text
Abstract:
The socio-economic impact of HIV/AIDS on South Africa has resulted in lower gross domestic product, loss of skills in key sectors such as education, and increased health-care costs in providing access to treatment. Currently active pharmaceutical ingredients (API’s) such as stavudine (d4T) and azidothymidine (AZT) are imported from India and China, while formulation is conducted locally. A strategy was initiated between CSIR Biosciences and LIFElab under the auspices of Arvir Technologies to investigate the feasibility of local antiretroviral manufacture (d4T and AZT) or the manufacture of a key intermediate such as β- thymidine (dT). Several advantages associated with successful implementation of this strategy include ensuring a local supply of API’s, thus reducing reliance on procurement from foreign sources and reducing the effect of foreign exchange rate fluctuations on providing cost effective access to treatment. A local supply source would also reduce the imports and thus aid the balance of payments deficit, and in addition to this, provide stimulus in the local pharmaceutical manufacturing industry (which has been in decline for several decades), resulting in increased skills and employment opportunities. This thesis describes the development of a superior chemo-enzymatic process for the production of β-thymidine (72 percent yield, prior to isolation), a key intermediate in the preparation of anti-retrovirals. Alternative processes based purely on chemical or bioprocess transformations to prepare either 5-methyluridine (5-MU) or dT suffer from several disadvantages: lengthy transformations due to protection/deprotection strategies, low selectivties and product yields (30 percent in the chemical process) and isolation of the product from dilute process streams requiring the use of large uneconomical reactors (bioprocesss). This contributes significantly to the cost of d4T and AZT manufacture. Our novel chemoenzymatic process comprises of a biocatalytic reaction for the production of 5-MU, with subsequent chemical transformation into dT (3 steps) negating and circumventing the limitations of the chemical or bioprocess routes. During the course of this project development, the β-thymidine selling price declined from 175 $/kg (2005) to 100 $/kg (2008). However, the process described in this work is still competitive based on the current β- thymidine selling price of 100 $/kg. The process economics show that with further optimization and increasing the isolated dT yield from 70 percent to 90 percent, the variable cost decreases from 136 $/kg to 110 $/kg. The increase in isolated yield is highly probable, based on solubility data of β-thymidine. The decrease in β-thymidine selling price and technological improvement in dT manufacture should translate into lower API manufacture costs and more cost effective access to treatment. Our novel biocatalytic process producing 5-MU uses a coupled enzyme system employing PNP, Purine Nucleoside Phosphorylase and PyNP, Pyrimidine Nucleoside Phosphorylase. The overall transglycosylation reaction may be decoupled into the phosphorolysis reaction (PNP) and synthesis reaction (PyNP). During the phosphorolysis reaction, guanosine is converted into guanine and ribose-1-phosphate (R-1-P) in the presence of PNP enzyme. The reaction intermediate R-1-P is then coupled to thymine in the presence of PyNP enzyme during the synthesis reaction, producing 5-MU. The process was scaled up from lab-scale to bench-scale (10 - 20 L) and demonstrated to be robust and reproducible. This is evident from the average guanosine conversion (94.7 percent ± 2.03) and 5-MU yield (88.2 percent ± 6.21) and mole balance (104 percent ± 7.61) which were obtained at bench-scale (3 replicates, 10 L). The reaction was carried out at reactor productivities of between 7 – 11 g.L-1.h-1. The integration of the biocatalytic process and chemical processes was successfully carried out, showing that 5-MU produced using our novel biocatalytic process behaved similarly to commercially available 5- MU (ex. Dayang Chemicals, China). A PCT patent application (Ref. No. P44422PC01) on this chemo-enzymatic process has been filed and currently public private partnerships are being explored through Arvir Technologies to evaluate and validate this technology at one ton scale.

Abstract:
Ten years ago, the only contenders for the role of setting the fashion media agenda were mainstream fashion media magazines such as Vogue. With the evolution of blogging and the popularity of blog sites associated with magazine websites, a new contender was born: the fashion-themed blog. Now mainstream fashion media outlets and fashion bloggers sit side-by-side in the front row at designer fashion shows covering fashion news and trends. This study focused on ten elite fashion-themed blogs (fashion blogs) reporting on women’s fashion and ten mainstream fashion media websites during the summer 2016 fashion season looking retrospectively at the fashion forecast from October 2015 New York Fashion Week. The study examined how media outlets respond to fashion blogs and whether the blogs were used as sources by mainstream fashion media or vice a versa. This study has filled a research gap by conducting the first analysis examining fashion blogs through the lens of agenda setting. This research uncovered a media landscape in transition with fashion bloggers and mainstream fashion media communicating in a new and different way. Communicating fashion news is no longer a one-way street controlled by mainstream fashion media magazines. Instead it is a two-way street where each outlet uses the other and neither plays the role of lead agenda setter. This new synergy suggests the acceptance of fashion blogs as credible sources for fashion news and potential agenda setters for the fashion community. The relationship between bloggers and mainstream fashion media also creates excitement regarding trends because fashion bloggers have the ability to instantly comment and generate buzz on a trend when the mainstream fashion media are reporting on other issues or waiting for their magazines to be published. Out of the five trends studied, denim was the leading trend with 44 percent of the posts or articles discussing denim. With every trend, bloggers were present in the five earliest published posts or articles and continued to post on the trends throughout the study period. Style.com dominated the mainstream fashion media landscape, with 25 published articles. For fashion bloggers, Gal Meets Glam and Atlantic Pacific tied with 19 blog posts each. Over 57.6 percent of the total blog posts and blog articles linked to fashion designer websites where readers could purchase the trend being discussed. Fashion bloggers did not quote sources in their posts; 74 blog posts and articles recognized no sources at all. Understanding the new relationship between fashion bloggers and fashion magazine websites will help guide future reporters of fashion and fashion trends.

Abstract:

Abstract:
The purpose of this study was to measure the perceptions of teachers regarding their most recent evaluation experience and to determine whether teachers perceive any significant relationships between attributes of teacher evaluation and its quality and impact on teacher growth. The study also considered whether there are significant differences between elementary and secondary teachers, as well as between classroom and nonclassroom teachers, in their perceptions of evaluation. The suburban school district in this study serves approximately 22,000 students. The subjects included 402 elementary and secondary teachers who were randomly selected from 1,081 permanent teachers. The Teacher Evaluation Profile (TEP) instrument was used to measure teachers' perceptions of their most recent evaluation experience. The 44 items on the questionnaire were examined based on the following five categories of evaluation: (a) teacher attributes; (b) evaluator attributes; (c) evaluation procedures; (d) evaluation feedback; and (e) evaluation context. A total of 284 respondents, or 71 percent of the teachers surveyed, returned a completed questionnaire. Data were reported in terms of frequency distributions, means, and standard deviations. Data analysis consisted of correlational analyses and an analysis of variance. The results of this study suggested that teachers judge the quality of their evaluation based on the attributes of the person who evaluates them and the feedback they receive. The quality of evaluation appears to be determined by the following attributes of effective feedback: the merit of the ideas and suggestions contained in the feedback, the depth of information provided, the specificity of information provided, and the amount of information received. Teachers appreciate an evaluator who gives useful suggestions for improvement, has a persuasive rationale for suggestions, and is a credible source of feedback. None of the attributes on the TEP had a significant relationship to the overall impact of evaluation on teacher growth. The results indicated that significant differences exist between elementary and secondary teachers, as well as between classroom and nonclassroom teachers, in their perceptions of evaluation. Recommendations were made for establishing a teacher evaluation system that is supportive of professional growth.

Abstract:
Scholars of intermedia agenda-setting have examined how news organizations can affect one another's content, but research is lacking on the influence of foreign news programs on fledging media outlets, such as Rwanda Television (RTV). A quantitative content analysis conducted between October and December 2016 indicates that media outlets in core countries dominated RTV's international news edition, which depended mostly on foreign programs, mainly from Euronews and Al Jazeera English. The 2016 U.S. election was the predominant topic. More than half the stories had a negative tone. In addition, qualitative interviews with the RTV editorial team revealed that the main factors influencing story selection were proximity, prominence, impact, cultural values, and relevance. Cross-lagged correlation coefficients indicated that both RTV and two Rwandan newspapers—igihe.com and Imvaho Nshya—focused on the same news topics but with slightly different sources. In Rwanda, the defining factors for this intermedia agenda-setting are not media type, ownership, or technology, as other studies have found, but institutional barriers, language, and the globalization of news. Faced with limited finances and a shortage of trained journalists, fledging media organizations in Rwanda will continue to depend on big media for their daily international news. Strategies are suggested for helping to break the cycle of foreign media domination and news homogenization in Rwanda

Naser, Md Abu. "INTERMEDIA AGENDA-SETTING RELATIONSHIPS BETWEEN POLITICAL BLOGS AND LEGACY NEWS MEDIA: A STUDY OF THE `OBAMA-IS-A-MUSLIM' RUMOR." OpenSIUC, 2014. https://opensiuc.lib.siu.edu/dissertations/843.

Full text
Abstract:
A number of incidents such as the "Obama-is-a-Muslim" rumor during the 2008 presidential election suggest that influential political blogs are playing a major role in setting the agenda of the legacy media. The power of the political blogosphere is forcing media practitioners, communication scholars and political theorists to re-conceptualize how issues arrive on the political and public agenda. This new phenomenon challenges the fundamental assumptions of agenda-setting theory to the extent that the 42-year old model, including the notion of a shared public agenda, should be reevaluated. By situating news agenda research within a larger intellectual context of agenda setting, this dissertation explored the intermedia agenda-setting relationship between political blogs and legacy news media, such as newspapers, network TV, and cable TV channels. Analyzing the contents count data of political blogs and legacy media and Internet Search Volume Index (SVI) data, this dissertation explored how coverage of a certain issue by top-ranking and influential political blogs may predict the coverage of the same issue by legacy news media. Employing a combination of quantitative and qualitative research methods, the results of the study revealed that political blogs significantly predicted the coverage of the legacy media on the "Obama-is-a-Muslim" rumor over a period of five days. The results also showed that the coverage of the rumor by both the political blogs and legacy media significantly predicted the level of Internet searches related to the rumor on all five days. The study also found positive and statistically significant correlations between the coverage by the political blogs and legacy media on the Obama-is-a-Muslim rumor and the volume of Internet searches on that rumor. The correlation was high on day 1, and decreased on each subsequent day. The study also found bi-directional agenda-setting relationships between the coverage of the political blogs and legacy media on the `Obama-is-a-Muslim' rumor. However, there was hardly any bi-directional relationship between the coverage of political blogs of the rumor and the volume of Internet searches related to the rumor. Although, the study found bi-directional relationship between the coverage of the legacy media on the rumor and the volume of Internet searched related to the rumor, the relationship faded away quickly. However, the study findings suggest that the perceived power of political blogs should be seen as trivial and ephemeral while that of the legacy news media should be seen as substantial and indelible in setting the public agenda. These findings may broaden our understanding of intermedia agenda-setting.

Abstract:

This study intends to examine the impact of a well known debate page in one of Sweden’s biggest newspapers, Dagens Nyheter. We have done this by analysing Ekot, Aktuellt, Göteborgs Posten and Västerbottens-Kuriren to see to what extent they “take” news from the debate page.

The method used was content analysis and to describe some of our thoughts about how we placed the hits in different categories we chose eight of our hits to exemplify.

The material from the newspapers, radio and television was examined to discover the potential impact from the debate page.

To carry out this study we have mainly used theories about how media get influenced by each other’s news and how they do follow-ups and refer to the original media. Other theories of importance for our study are for example those about what kinds of events that become news and how media influences people’s thoughts about what’s important.

One of the most important conclusions we were able to draw after carrying out our study was that the debate page in Dagens Nyheter has quite a small impact on what our chosen media reported on. The biggest impact was found in the newspapers, whereas radio and television very seldom referred to the debate page.

To sum up, one can absolutely say that the debate page in Dagens Nyheter is powerful and do have impact on other media, but an article published on the page is not automatically discussed and debated in other media during the day.

Abstract:
Karlheinz Stockhausen’s Kathinkas Gesang als Luzifers Requiem presents mental, physical, and musical challenges that go beyond the usual expectations of an instrumentalist, extending and redefining the traditional idea of virtuosity. Using firsthand performance experience, score and recording study, and flutist interviews, this document explores the effects of some of these heightened demands and argues that the particular performance situation presented by Kathinkas Gesang brings up critical questions about the performer’s role, the nature of performance and of the musical work, and the existence of an authoritatively “authentic” interpretation. Employing an expanded definition of virtuosity that includes interpretation and encompasses both choices and actions, the document discusses the extensions of virtuosity into two main areas: first, memory; and second, staging and movement, covering both practical suggestions and larger implications. Finally, it examines how the performer’s negotiation of these challenges relates to questions about authenticity and agency. Performance is defined here as a creative and collaborative act, not attempting to duplicate previous performances or recordings, but rather to give the best realization of the piece possible in the given circumstances, according to the individual’s interpretation of the score’s directions. There is no single “authentic” interpretation, but rather a rich multiplicity of possibilities, and the performer’s creative agency and personal authenticity are necessary for the full realization of the work.

Abstract:
The thesis here presented is focused on the molecular study of three human proteins related to conformational diseases, or Amyloidoses. The mechanisms of native folding and aggregation in a cross-β sheet structure have been described using several spectroscopic techniques. These two pathways are competing by the presence of intermediate states of the reaction. An oligomeric intermediate is the precursor of the β-aggregates and is considered the main responsible of the cellular toxicity in many amyloid diseases. Therefore, is necessary to perform a deeper study of the oligomeric state to delve into the etiology of these human diseases. On one hand, the expression and extraction of a single-chain variable fragment (scFv-h3D6), a derivative of an antibody specific for Aβ-oligomers, was performed. Here is the first description of a conformational mechanism by which a scFv-h3D6 withdraws Aβ1–42 oligomers from the amyloid pathway and, consistently, it prevents Aβ-oligomer toxicity in the SH-SY5Y human neuroblastoma cell line. Moreover, we modeled the 3D-structure of scFv-h3D6, obtained by sequence homology. This model has allowed to a rational redesign of the molecule in order to improve its stability, increase its recombinant production, and decrease the effective dose already demonstrated in a mice model of Alzheimer's disease. For another hand, we studied the AL-12 protein, an Ig variable domain (VL) obtained from a patient diagnosed with light chain Amyloidoses. This amyloidogenic domain shows seven non-conservative mutations from his germinal protein κI O18/O8. The thermodynamic study of AL-12 and four restoratives mutants, allowed us to determine the effect of somatic mutations on the VL stability. Thereby, the restoration of a residue toward the germinal sequence implies a destabilization of the domain and increases the aggregation tendency. Here we demonstrate that a single somatic mutation couldn’t be the cause of the amyloidogenic potential of AL-12, rather it is due to several factors. The destabilization of the native folding promotes a conformational reorganization more susceptible to β-aggregation. Finally, we work with PSD95-PDZ3 domain, a neuronal post-synaptic protein and related to human disease such as Alzheimer or Autism. PDZ3 follows a reversible aggregation pathway, a feature which is relevant to determining the conformational changes that promote it. In addition, the design of six variants of PDZ3 has helped determine the regions involved in the folding and stability of the global PDZ3 domain and its role in the aggregation process. This work demonstrates the importance of the states in the intermediate conformational space of proteins, their relationship with conformational diseases, and the application of knowledge in the development of effective therapies.

Abstract:
The biological control of the weedy complex Lantana camara (L.) (Verbenaceae) has been ongoing in South Africa for over 40 years. Despite this, the weed is still not under sufficient control and continues to invade new territories in the country. The biological control programme needs to be bolstered with releases of new and potentially damaging biological control agents. A promising biological control agent endemic to Central America, Falconia intermedia (Distant) (Hemiptera: Miridae), was imported into quarantine from Jamaica in 1994. This agent was released on sites in KwaZulu-Natal and Limpopo provinces of South Africa in 1999. Even though it initially established and damaged L. camara, populations died out at most of the release sites. As varietal difference and adverse climate have been cited as the reason for non-establishment and ineffective control in L. camara biocontrol programmes worldwide, this study attempts to investigate the role that these two factors play in this weed herbivore relationship. Laboratory no-choice trials were conducted to determine the varietal performance of F intermedia, among five Eastern Cape varieties of the weed from East London, Whitney Farm, Heather Glen Farm, Port Alfred and Lyndhurst Farm, and a variety from the Plant Protection Research Institute (PPRI), Pretoria. However, there were differences in performance as the adult mirids performed better on white-pink varieties from Whitney Farm and Heather Glen Farm. To test varietal preference in field conditions, field releases of F intermedia were also made at East London, Whitney Farm, Heather Glen Farm, Port Alfred and Lyndhurst Farm. Post release evaluations were conducted monthly for two years (2002 and 2003). The insect established at East London and Whitney Farm, both of which have white-pink varieties. Insect populations quickly died out at the Lyndhurst Farm and Port Alfred sites, which have dark pink varieties. It is suggested that field conditions may have resulted in poor plant quality and led indirectly to varietal preference, and to non-establishment at these two sites. With the onset of cooler weather, populations disappeared at Heather Glen Farm. This suggested that F. intermedia was suitable for release in more subtropical areas within South Africa where climatic conditions would be suitable throughout the year. The mirid performed well at Whitney Farm, resulting in significant reduction in plant growth parameters such as height and percentage cover, and increasing the cover of other flora growing beneath L. camara plants. Finally, ways to improve the efficacy of this agent are considered in an effort towards better control of L. camara in South Africa.

Abstract:
Information and communication technology (ICT) radically affects the tourism industry by overturning its production and distribution vertical value chain. Direct exchange between tourist and suppliers is the beginning of a new era. ICT has also given the opportunity to many new players to introduce themselves into tourism market. From now on, travel agencies and tour operators are not the only ones able to connect supply with demand. Internet has also contributed in changing the tourist’s demands and activities. Tourism is no longer limited to a mass product. The tourism intermediaries must evolve in a less favourable environment. The question is about their place and value creation in the industry. A literature review allows us to understand that the tourism intermediary is present to overcome market’s imperfections and propose value through the following functions: informational, expertise, assurance, economic, innovation and networking. If this literature review shows that the intermediation functions are value creators, it does not necessarily claims that all functions are always carriers of value creation. Therefore, we analyse the intermediaries’ value propositions by trying to find the functions they perceive as being important to cope with tourism evolution. Consequently, we will complete our theoretical analysis with an empirical observation to identify the functions on which the intermediaries rely the most to face the evolution of the tourist market. By means of statistical analysis of our semi-structured interviews, we show in particular that: the expertise and assurance are the most important value propositions of tourism intermediaries; tour operators take less financial risks except for owning a competition-free offer. Furthermore, specialisation and destination management are revealed as strategic choices

Abstract:
A highly variable invasive shrub, Lantana camara L. (Verbenaceae), has been notoriously difficult to control thus far despite a well established biological control programme in South Africa. A promising leaf-feeding biological control agent, Falconia intermedia (Distant) (Hemiptera: Miridae), released to control this invasive plant eventually crashed at three out of five sites in the Eastern Cape Province. In the Mpumalanga Province, after initially colonising and building up high numbers on the L. camara stands the agent populations crashed. Several reasons for these population crashes have been suggested, but induced plant defences have not been investigated. Although plants face the challenge of herbivory by various organisms while remaining immobile, some plants may possess the ability to induce physical and/or chemical defensive responses following feeding and thus prevent further plant tissue damage and loss. Laboratory trials were conducted to determine the existence, nature and effect of physical and chemical feeding-induced responses of L. camara on the performance of the leaf-feeding biological control agent, F. intermedia. Lantana camara plants used in the study were obtained from five localities in the Eastern Cape Province, South Africa, while the insect culture was established from field populations. Plants from all varieties on which F. intermedia was released significantly increased the toughness of their leaves compared to control treatment plants. In addition, plants from three localities: Lyndhurst Farm, East London and Port Alfred, significantly increased trichome density after prolonged feeding by F. intermedia. On the three varieties showing increases in these two factors (i.e. leaf toughness and trichome density), oviposition, survival and feeding damage by the mirid agent was significantly lower on previously damaged plants. A significant negative correlation between trichome density and population numbers was found (R²= 0.52, p < 0.0003), suggesting that an increase in trichome density strongly contributes to a reduction in F. intermedia's growth. The growth and reproduction of the resistant plants was not significantly impacted by F. intermedia feeding. The defensive responses were found to be plant systemic and rapidly induced as they were elicited and expressed throughout the plant in both damaged and undamaged leaves within five weeks after insect release. Leaf toughness and trichome density were not significantly increased after feeding on plants from Whitney Farm and Heather Glen Farm. On the contrary, mirid individuals performed significantly better on plants from Whitney Farm and Heather Glen Farm than on plants of other varieties, indicating their susceptibility and suitability to the agent and the lack of induced resistance against the agent. Plants from all localities besides East London showed some level of tolerance and overcompensated for feeding damage by increasing plant growth and reproductive factors on plants fed upon. This was however only significant in two variables of the more susceptible localities, Whitney Farm and Heather Glen Farm. This increase in plant fitness did however indicate an induced defence response by these plants to feeding, a response designed to lessen the effects of agent feeding. Headspace volatile analysis was used to investigate any volatile chemical responses by L. camara due to F. intermedia feeding at two of the five localities chosen: East London and Whitney Farm. There was no significant difference in headspace volatiles emitted by leaves of plants from the East London insect infested and control treatment plants. On the Whitney Farm damaged plants however there was a 2.5 fold increase in the emission intensity of one of the three main compounds, later identified as Beta-caryophyllene. Three major chemical constituents which were found to be common to leaf volatiles of the two varieties were identified through gas chromatography-mass spectrometry (GC-MS) from the damaged and undamaged leaves of these two varieties. The methods used in collecting leaf volatiles were shown to be significant in the strength of chromatogram peaks. Using general authentication methods and purified standards, one of these was identified as the sesquiterpene, Beta-caryophyllene (C₁₅H₂₄). This compound is one of the major constituents found in isolations of L. camara varieties worldwide. This is the first such work done on a variety of L. camara in South Africa, and hopefully the beginning of more in-depth studies of the volatile organic chemicals from the numerous naturalised varieties of L. camara. It is suggested that the sum of these responses may play a role bigger than is currently understood in this plant-insect relationship. It is also argued that feeding induced plant defences may play an important role in attempts to control alien plants using insect agents.

Abstract:
Lantana camara L. (Verbenaceae) remains a highly invasive and ecologically damaging weed in South Africa, despite some 50 years of biological control efforts. Lack of success has been ascribed to varietal differences, climate and predation of agents but these have not been tested. In this study, the effects of ant predation were tested on populations of three biological control agents for L. camara. Colonies of two species, Crematogaster sp. 1 and 2 were investigated. Crematogaster sp. 1 colonies were offered no choice between immature stages of the agents Hypena laceratalis Walker (Lepidoptera: Noctuidae), Falconia intermedia Distant (Hemiptera: Miridae) or Teleonemia scrupulosa Stål (Hemiptera: Tingidae) on lantana shoots. Density-dependent predation on F. intermedia and T. scrupulosa nymphs on lantana shoots was tested using Crematogaster sp. 2 colonies. In choice experiments Crematogaster sp. 2 colonies were offered F. intermedia or T. scrupulosa nymphs on potted lantana plants. Preliminary food trials confirmed that colonies foraged for protein, thereby validating results of no-choice experiments. Crematogaster sp.1 foragers removed 50% of F. intermedia nymphs, followed by 45% of H. laceratalis larvae and only 9% of T. scrupulosa nymphs. Foragers recruited most actively to H. laceratalis larvae and significantly more H. laceratalis biomass was removed than either F. intermedia or T. scrupulosa. A trade-off existed in prey size selection because larger larvae provided considerably more biomass but required forager cooperation and a longer time to subdue than did smaller prey. This increases both forager energy expense and mortality risk by other predators. This study showed that all Crematogaster sp. 1 colonies removed small (≤10mm) H. laceratalis larvae more frequently than larvae larger than 10mm. Thus, of these biological control agents, predators probably prefer small H. laceratalis larvae. Significantly more F. intermedia than T. scrupulosa nymphs were removed by Crematogaster sp. 1, while Crematogaster sp. 2 colonies removed comparable numbers of both agent species. Falconia intermedia nymphs' fast movement triggered a predatory response by these ant species. In contrast, the relatively immobile behaviour of T. scrupulosa nymphs was identified as a highly effective predator avoidance strategy. Since T. scrupulosa nymphs are unable to escape predators by moving, they appear to depend on the presence of alternative prey attracting predator attention. At high agent and/or forager density, T. scrupulosa nymphs attempted escape, but foragers identified them as prey once they moved and caught them. Predation on F. intermedia was also density dependent in that at high nymph and/or forager densities, escape routes were congested and nymphs were more easily caught. Survival of F. intermedia and T. scrupulosa nymphs in particular was low on ant-accessed shrubs in choice experiments and high on ant-excluded shrubs. It is likely that ants significantly depress F. intermedia populations in the field since besides predation, ant foragers probably interrupt F. intermedia feeding and ovipositioning. The combination of parasitism and predation on early instar larvae may explain why H. laceratalis occurs across lantana's range in South Africa but populations remain low. It is unlikely that T. scrupulosa nymphs are habitually preyed on by ant species unless they attract attention by being mobile. Although biological control of L. camara is influenced by climate and physiological defence mechanisms, this study has shown that predation by two ant species severely impacts leaf-feeding agents for L. camara. Thus, it is recommended that future selection of additional agents to control lantana should exclude leaf-feeding .

Kumar, Lalit. "DEVELOPMENT OF NOVEL CHEMICAL TOOLS FOR PROTEASOME BIOLOGY & A NEW APPROACH TO 1-AZASPIROCYCLIC RING SYSTEM." UKnowledge, 2012. http://uknowledge.uky.edu/chemistry_etds/14.

Abstract:
The proteasome, a multiprotease complex, is clinically validated as an anticancer target by the FDA approval of bortezomib and carfilzomib for the treatment of multiple myeloma. The emergence of resistance to proteasome inhibitors however remains a major clinical challenge. Recently, distinct types of proteasomes termed ‘intermediate proteasomes’, which contain unconventional mixtures of catalytic subunits, have been implicated with drug resistance of tumor cells. In elucidating the role of intermediate proteasomes in drug resistance, a crucial step is to unequivocally determine the subunit composition of intermediate proteasomes in cells. With this in mind, the goal of the studies reported in this dissertation is to develop novel chemical tools which can facilitate the investigation of intermediate proteasomes via two complementary approaches: a FRET-based approach and a bifunctional cross-linking approach. Chapter 2 describes the structure-based design, synthesis, and characterization of a peptide epoxyketone-based fluorescent probe, named as LKS01-B650, which selectively targets the immunoproteasome subunit β5i/LMP7. In addition to its utility in determining the identity of intermediate proteasomes as FRET-based probe, this imaging agent may also serve as a valuable tool in visualizing the immunoproteasome in living cells. Chapter 3 describes the design and synthesis of various epoxyketone-based bifunctional agents. The ability of these bifunctional agents to cross-link different catalytic subunits within a proteasome complex is shown by mobility shift assays. These bifunctional agents may provide important information in determining the subunit composition of proteasomes. Chapter 4 describes a systematic study of the relationship between the proteasome inhibitor structure and the inhibitory activity against critical subunits of the proteasome. Given the reported role of β5i/LMP7 in autoimmune diseases, this study may provide useful insights in developing therapeutic agents for autoimmune diseases as well as other diseases. Chapter 5 describes a separate study which is not related to proteasome biology. A concise approach to synthesize 1-azaspirocyclic ring systems is developed by utilizing a novel semi-pinacol/Beckmann rearrangement. Additionally, an environmentally benign, microwave-assisted, and solvent-free self-condensation of carbonyl compounds is reported.

Abstract:
Political blogs have emerged as a new journalistic format that has gained influence on the political discourse in the United States. Previous research has shown that this influence stems mainly from attention given to blogs by traditional news media. Based on the concepts of intermedia agenda setting and agenda building, this study explored the source interaction between 10 elite traditional news media and 10 political filter blogs during the 2006 Congressional Midterm Elections. An analysis of 2587 sources used in the election context found that traditional news media frequently cited blogs in their election coverage, but that the source attributions to the blogs were vague. Blogs, on the other hand, heavily cited traditional news media, but the analysis revealed that conservative blogs cited elite traditional news media less than did liberal blogs. Conservative blogs relied more on conservative media outlets in their election coverage. A case study of the dominant election topic, the Mark Foley scandal, showed that the daily interaction between the two media formats was driven by the use of breaking news elements as well as controversial opinions. The findings of this study show that the blog agenda is strongly influenced by traditional news media sources and that blogs at the same time have become part of the routine newsgathering process of traditional news media journalists. However, the findings also raise questions about changes in the standard journalistic research and attribution procedures as both media formats often rely on each other as sources rather than on original reporting.

Abstract:
The aim of this thesis is to contribute to the understanding of ancient Egyptian kinship in the First Intermediate Period and Middle Kingdom (ca 2150–1650 BCE) by exploring how forms of relatedness were displayed in the monumental record. Kinship and marriage are contextually driven sociocultural phenomena that should be approached from the actors' perspective; such an approach can achieve some insight into emic notions of kinship, because monuments were integral to society and contributed to perpetuating and sustaining its fabric. The introduction (chapter 1) presents the theoretical background on which the thesis is based, namely the notion of kinship as process, where relationships can be constructed and reconstructed throughout one’s life. In addition, it provides a working definition of 'kin group', an analytical category that is taken as the primary unit of social analysis that can encompass several ways of being related. Chapter 2 offers a discussion of kinship terminology in the First Intermediate Period and Middle Kingdom. The focus is less on basic kinship terms than on the little understood terminology for kin groups and how these were presented in the written record. Chapter 3 treats stelae, which constitute the core corpus of material for the thesis. Stelae present a variety of images of kin groups and, moreover, they should be considered within the larger units of which they were part. Many of these stelae are unprovenanced but have been attributed to Abydos. At this site, memorial chapels have been identified archaeologically, and some stelae have been found in association with them. Thus, the site offers a materialisation of constellations of relationships. Possible reconstructions of such chapels – one from Saqqara and two from Abydos – are presented in chapter 4, and the impact they may have had on the social memory of visitors is assessed. Display, presence, and performance were some of the ways in which the social role of those groups was communicated. Chapter 5 is concerned with how change and time may be represented in apparently static objects. On the basis of the model of the developmental cycle of domestic groups first introduced by Meyer Fortes, the dynamism of the social fabric is explored through three case studies of groups at different stages of their developmental cycle. The strategies of survival can be seen pervasively in the monumental record, allowing for a glimpse into time and change in kin groups. The conclusion (chapter 6) offers a holistic approach to the material presented in the thesis, emphasising the ways in which the different theoretical approaches proposed intertwine with the material.

Abstract:
Years of high stakes testing and managerial directives to improve student test scores created a trend of teachers’ declining sense of efficacy and agency. Researchers have yet to examine the perceptions of teachers following requirements to improve student engagement and school climate in an effort to improve academic performance following the authorization of local and national educational accountability reforms. The purpose of this study is to examine how teachers perceive their efficacy and agency in response to the addition of nonacademic measures and the requirement of documented input from teachers and other stakeholders into educational policy planning procedures. Veteran K-12 teachers’ responses to survey and interview questions were coded, analyzed, and organized into themes to generate an educational theory. Grounded Theory Methodologies (GTM), Culturally Responsive Methodologies (CRM) and Critical Pedagogy (CP) informed data collection methods and theoretical foundations for this study. The creation of a safe dialogical space between the practitioner researcher and participants developed a relationship for both to engage as co-researchers. Abstract:
Lantana camara (lantana), a thicket-forming shrub, a number of different varieties of which were introduced into South Africa as ornamental plants but which has become a serious invasive weed. Conventional control measures for lantana are expensive and ineffective and it has therefore been targeted for biological control since 1961. To date, eleven biological control agent species have become established on lantana in South Africa. However, most agents persist at low densities and only occasionally impact plant populations. Three species regularly cause significant damage, but only reach sufficiently high numbers by midsummer after populations crash during the winter. Overall, the impact of the biological control programme on the weed is negligible and this has been ascribed to the poor selection of agents for release, the accumulation of native parasitoids, differences in insect preference for different varieties of the weed and variable climatic conditions over the weed’s range. This study suggests that the importance of varietal preferences has been over-estimated. A predictive bioclimatic modelling technique showed that most of the agents established in South Africa have a wide climatic tolerance and that the redistribution and importation of new climatypes of these agents will not improve the level of control. Additional agents are required to improve the biocontrol in the temperate conditions, and also to increase damage in the sub-tropical areas where most of the agents are established and where the weed retains its leaves year round. New candidate agents that possess biological attributes that favour a high intrinsic rate of increase, a high impact per individual and that improve the synchrony between the weed and the agent in climatic conditions that promote the seasonal leaflessness of plants should receive prior consideration. A survey in Jamaica indicated that additional biological control agents are available in the region of origin but that care should be taken to prioritise the most effective agents. The various selection systems currently available in weed biocontrol produce contradictory results in the priority assigned to candidate agents and a new selection system is proposed. The biology and host range of two new candidate natural enemies, the leaf-galling weevil, Coelocephalapion camarae and the leaf-sucking mirid, Falconia intermedia were investigated for the biocontrol of lantana. The studies indicated that these have considerable biocontrol potential, in that the weevil has a wide climatic tolerance and has the potential to survive the host leaflessness typical of temperate conditions, while the mirid has a high intrinsic rate of increase, and the potential for several generations a year. Both agents caused a high level of damage to the leaves, with the weevil galling the vascular tissue in the leaf-petiole and the mirid causing chlorotic speckling of the leaves. During laboratory trials both agents accepted indigenous species in the genus Lippia. However, under multiple choice conditions these agents showed a significant and strong oviposition preference for lantana. A risk assessment and post release field trials indicated that F. intermedia is likely to attack some Lippia species in the presence of lantana, but the levels of damage are predicted to be relatively low. A possible low incidence of damage to indigenous species was considered a justifiable ‘trade-off’ for the potentially marked impact on L. camara. Preference and performance studies on the two candidate agents suggested that most of the South African lantana varieties are suitable host plants. The mirid preferred certain varieties in multiple choice experiments, but this is unlikely to affect its impact under field conditions. Permission for release was accordingly sought for both species. Finally, the challenges facing the biological control programme and the potential for improving the control of L. camara in South Africa are considered.

Abstract:
Since researchers Maxwell McCombs and Donald Shaw published the seminal work of Chapel Hill on agenda-setting theory (AS) at the beginning of the 1970s, its academics have developed their investigations using two methodological tools: a first phase of content analysis, used to study the news coverage and, therefore, the prominence or salience of certain topics or attributes in the agenda; and a second study of statistical nature, based on the verification of the level of correlation between the issues, their senders or the global of the different agendas. Content analysis has generally focused on the relative or comparative importance of some topics over others. It is assumed that some media or social actors are busier than others in certain topics, which has led to a general study of the relative volume of information, to the detriment of the detailed content analysis. Such differences (or similarities) have been traditionally evaluated from the comparison of the frequencies of their main contents, which, by means of Spearman correlations between the senders, have fostered an analysis on the hierarchical order, disregarding the relative or absolute and, therefore, any other information. In view of this need, and to resolve these procedures when the interest lies in the relative importance of the parts of a whole, the thesis deals with the analysis of Compositional Data (CoDa), founded by Aitchinson in 1982, along with the graphic representation through biplots, promoted by Gabriel in 1971, for the study and mapping of the AS theory. The interest in CoDa lies in the proportion of each part or component, since the absolute quantities are irrelevant and only expose the sample size. A very similar problem in all aspects that is established in the study of the AS, where the absolute data are irrelevant and mostly point to the content, the intent of the content, the popularity of the issuer, etc. In this line, only the proportions of each content category or the relative size of one type of content over another are 3/3 truly informative. According to these premises, the present doctoral thesis is structured in five articles that distinguish the originality of the study in presenting the CoDa-biplot as a tool for mapping and studying the AS theory. Using the period of the 2015 General Elections in Spain, the thesis is presented as a thematic unit that addresses, throughout five publications, the mapping and compositional study of the theory of AS of opinion on politics in the Spanish media system (media agenda), as well as political communications on Twitter of the main Spanish parties (political agenda). Likewise, the aggregate study of both agendas (the media and politics) allows us to approach the analysis and representation of the intermediate AS

Abstract:
The purpose of this thesis is to clarify what a football agent does in Sweden and describe how legitimacy is created in an industry that is associated with crime and a lack of transparency. Qualitative interviews have been used to answer the two main issues that follows: What do football agents do in Sweden and what services do they offer football players and football clubs? And how do football agents construct legitimacy in an industry with a bad reputation? The respondents were picked from three different professions: three footballers, three football agents and three sports directors. The result shows that football agents in Sweden mainly worked with contracts negotiation, professional contact creation, scouting and giving career advice. Some agents even offered their players the opportunity for extra training sessions, match analysis and rehabilitation. The result also shows that some agents in Sweden uses work methods that could be considered unethical, for example giving bribes to players. When it came to how agents constructed legitimacy, it was mainly about creating a trust between themselves and other actors in Swedish football. This was done mostly through gaining external approval from other actors in football. In addition, it was about following the regulations that exist and staying away from anything that can be associated with the shadier side of the football agent industry.

Abstract:
The purpose of this bachelor thesis was to examine how traditional news media vis-à- vis alternative news media in Sweden portrayed the Quran burnings and the riots which followed during august in the summer of 2020. With the basis in framing theory and intermedia agenda setting we examined these research questions: How do the news papers frame the Quran burnings? What discourses are reproduced? How do the traditional news papers and the alternative news papers framing of the Quran burnings differ from each other?  The qualitative methods used in this bachelor thesis were Robert Entmans framing theory combined with Rens Vliegenthart & Stefaan Walgraves intermedia agenda setting theory. Also, we used different theories regarding populism, clash of civilizations and freedom of speech in order to find various themes in the reporting. For instance, the study applies Mikhail Bakhtin’s theory of the carnivalesque to examine how the alternative news paper Samnytt framed the Quran burnings.  Through examining news articles, we discovered several preexisting frames and some additional ones. The results showed that the traditional news papers framed the Quran burnings in a laconic way as an intolerant act against Muslims, while the alternative news papers framed the Quran burnings as a clash of civilizations and simultaneously as an act of freedom of speech. In relation to the alternative news papers overly dramatic and sensationalistic discourse, the traditional news papers discourse was more moderate. In this way the populist discourse in the alternative news media and their focus on the “man on the street” is an example of how the alternative news media defines itself as an opposition to the hegemonic “elite” news media institutions. In the alternative news papers, we also found one specific frame concerning anti-Semitism in Malmö, which was not given the same amount of attention by the traditional news papers.  The result of this study shows differences in framing, in line with previous research regarding alternative news media and populism, that could be an example of how the alternative news media is born and defines itself as an alternative to a narrative they deem wrong or lacking in traditional news media. This is primarily an effect of the fact that the traditional news papers lacked diversity in their interviewees since they mostly relied on authority sources, or in one case their own opinion piece writer, to tell the story. The results of this study highlights processes and effects that lies at the core of several phenomenon in this sphere, therefore it should be used to further the research on framing, alternative news media and populism.

Abstract:
In the laboratory, investigations on neurofilament biology and on their interactions with other cytoskeleton components showed tubulin-binding sites (TBS) on different intermediate filaments. Among the synthesized peptide corresponding to these sequences, the NFLTBS.40-63 peptide derived from the neurofilament light subunit (NFL), revealed in vitro and in vivo targeting properties and anti-tumour effect on glioblastoma cells, the most frequent and aggressive brain tumour, for which current treatments show very limited effects. The therapeutic value of this peptide is reinforced by its lower uptake and the lack of a major cytotoxic effect in healthy cells (astrocytes, neurons). Works in this thesis describe the uptake mechanisms of the NFL-TBS.40-63 peptide in glioblastoma cells, to better understand its selective internalization. Its specificaction on glioblastoma cells has been completed by the analysis of the peptide effects on the mitochondrial organization and function. Finally, we show that the peptide targets neural stem cells, which could be at the origin of glioblastoma, and serve as a therapeutic tool inseveral brain diseases, such as traumatic injuries and neurodegenerative diseases. Altogether these works indicate that this NFL-TBS.40-63 peptide is a promising tool for therapeutic strategies of glioblastoma. It can also target neural stem cells in order to develop new treatments for various brain disorders

Abstract:
The diploma thesis deals with intermedia relations and agenda-setting in the case of the purchase of one-crown bonds of the company Agrofert by its owner Andrej Babiš. The news story took place in the first half of 2017 and its research was ended on the date of filing of a motion to remove Andrej Babiš from the position of Minister of Finance in the then government of Bohuslav Sobotka. The subject of quantitative analysis of media content is the determination of the influence in intermedia relations, the development of the story coverage over time and the degree of use of links to social networks in news articles. The case study then uses the grounded theory method to identify the variables that represented the media inputs, political context, and political outputs that occurred during the development of the story, and also to look for relationships between the phenomena observed. The work is based on the theory of agenda-setting and a model of contingency of the mass media's political agenda-setting power. The thesis is divided into four parts. The first chapter contains a summary of theoretical concepts and results of empirical studies related to the media agenda building and agenda- setting and the relationship between the media and political agenda. Awad, Nathalie C. "Knowledge transfer intervention theory : a model grounded in the strategies used by intermediate agents in the context of education." Thèse, 2014. http://hdl.handle.net/1866/11595.

Full text
Abstract:
The present thesis was based on Grounded Theory (Strauss & Corbin, 1998) principles in order to address the lack of documented strategies implemented by “intermediate agents” to promote school practitioners’ use of research-based evidence. The term “intermediate agent” refers to the individuals who are positioned at the interface between scientific knowledge producers and users, and who encourage and support users’ uptake of research-based evidence in their practice. The study is part of the Quebec Ministry of Education, Recreation and Sports’ project aimed at improving success rates of secondary school students in underprivileged areas. Those solicited to participate in the study were intermediate agents from different levels of the educational system who had obtained the mandate to transfer research-based evidence to practitioners in schools targeted by the project. Snowball sampling (Biernacki & Waldorf, 1981; Patton, 1990) was used to identify individuals who were recognized by their peers for the quality of the support they offered to school practitioners in applying research. Sixteen semi-structured interviews were conducted. Analysis of the data yielded an intervention model coined Knowledge Transfer Intervention Theory, composed of 32 strategies of influence that were categorized into 6 intervention components: Relational, Cognitive, Political, Facilitative, Evaluative, as well as Continuous Support and Follow-up. The findings suggest that strategies embedded in the Relational, Cognitive, and Political components are interdependent and aim to establish a favourable climate, allowing agents to exercise a greater influence towards potential users’ ownership of the knowledge utilization process. Moreover, the Continuous Support and Follow-up component is key to sustaining changes in school practitioners’ use of research. Theoretical implications linked to the model, along with underlying explanations of the mechanisms involved in the different components are compared with the literature in knowledge transfer in the sectors of both health and education. Reports from related disciplines (notably in psychology) are also exposed. Finally, recommendations for practice are proposed.

Other theoretical concepts like gatekeeping, news values or media routine are also briefly presented. So are profiles of selected media outlets are also presented. Next part of the thesis introduces the reader to methodology upon which the research is based. The research itself is divided into two parts. The first one is a quantitative content analysis of ČTK's foreign news agenda during the specified period. The same analysis was also carried out for selected Czech daily newspapers. The research focused on several variables - what media sources are being used, what topics do the articles cover or who are the actors mentioned in the articles. The second part of the research focused on whether and on what scale do daily papers use texts provided by ČTK and if they're being used without adjustments. Main findings of the research with commentary are presented in the conclusion section of this thesis.

Nowadays, social movement activists tend to manage their own online media during the movement period. Activists could not only use online media as a platform to communicate the public, but also could use it to influence mass media’s coverage. Past studies on agenda-setting showed that media could set each other’s agenda in the context of election. However, there are few studies focused on the context of social movement. Therefore, this study focused on the case of Shihlin Urban Renewal to examine the flow of agenda between mass media and online media, whether online media owned by SMO could influence mass media’s coverage, or mass media still has its agenda-setting ability on others. The current study raised 5 research questions to examine the event theme, frame, source, standpoint used by mass media and online media, and the flow of agenda between these two media. Content analysis was adopted to analyze print press (CHINA TIMES and Liberty Times) and online media (Facebook fan pages managed by the pros and cons during this movement), and the sampling time began from January 28th in 2012 to March 28th in 2013. The result showed that the theme of law and politics were the most emphasized event themes of mass media during the movement, while online media stressed on the themes of protest and attribution of the whole event. Second, both media spent lots of space discussing treatment recommendation of urban renewal and this movement. Third, mass media tended to quote different sources in a single coverage, but online media preferred to quote sources that supported their action. Fourth, the mass media coverage showed its sympathy toward the Wang family while online media holded distinct standpoint. Last, the finding showed that during social movement, mass media could set online media’s news themes, but online media could decide mass media’s frame of the agenda. These results suggest that under the context of social movement, mass media and online media both has their influence on each other.

The asymmetric Reformatsky reaction is a versatile and straight forward approach to obtaining chiral alcohols ( -hydroxy esters), a motif found in biologically active compounds and synthetic intermediates. Although a variety of asymmetric Reformatsky reactions have been developed. In our study the compound 3 with  -hydroxycarboxylic acid was reported as an important intermediate of triazole antifungal agent. We found the Reformatsky reaction with acetophenone (Ketone) was carried out under the metal-halogen exchange reaction of the α-Bromo oxazolidinones with diethyl zinc and catalyst loading of Ni(acac)2. And we obtained that the best diastereoselectivity up to 97%, which provide a practical method to synthesize the new triazole antifungal agents.

Because of the growth of new-type intermediate traders and the increase of independent visitors, the travel agency is being divided gradually. In recent years, tourism industry is being upgraded by big data’s applications which are all the rage. Big data analysis which is different from traditional databases can predict customers’ demands and offer product experiences. By doing so, it can offer a new opportunity for conventional travel agencies. This research bases on Resource-Based Theory (RBT) and uses big data applications as intervening variables. By observing these cases and gathering the second-handed information by interviewing the managers of three different kinds of travel agencies, this research studies what kinds of core capabilities which travel agencies should possess. Besides, this research also studies the competitive relationship between traditional ravel agencies and new-type intermediate traders. By reference reviews and cases interviews, this research sorts out conventional travel agencies’ key factors of success in developing e-commerce area. The top issue for travel agencies is to know their own advantages of sources. Moreover, travel agencies should further enhance and raise their four new core capabilities which include tourism professions, social network’s asset managements, strategies of brand data and integrations between upstream and downstream sources by using their own advantages. After possessing these core capabilities, travel agencies should manage social network’s asset by analyzing brand data. Furthermore, they should also integrate corporations’ sources and improve performance systems. Ultimately, they should take online to offline(O2O) and Co-opetition strategies to build a new industry’s ecosystem. This method is the main key point of enhancing organizations’ performance and reintermediation for traditional travel agencies.

Teamwork has been regarded as essential basic skills of the 21st century citizens. Collaborative problem solving ability has begun to attach importance to international , which PISA (Programme for International Student Assessment) to implement the collaborative problem solving computerized assessment in 2015, the object is for 15 years of age students. This study aimed at understanding on collaborative problem solving performance situation in 3rd & 4th graders of elementary school, therefore develop suitable computerized assessment for them. In this study, based on PISA 2015 collaborative problem solving framework design topics, in order to reduce real dialogue emotions and personality traits error to get a more accurate student ability, test method using one on one human-computer interaction options dialogue computerized test, two units were designed "classes uniform design" and "two three-legged". The results of this study are as follows: 1. Develop suitable computerized assessment for student in 3rd & 4th graders of elementary school and can effectively measure the collaboration problem solving. 2. We can find most CPS skills common answer correct rate has more than 0.5。 3. The analysis showed most CPS skills are in high correlation. 4. Most CPS assessment analysis showed little difference A fourth-grade performance is better than B fourth- grade performance。We can not rule out the possibility of the gap between urban and rural schools and culture deprivation. And the reading understanding indirectly affect the ability of collaborative problem solving.

The main purpose of this study is based on PISA 2015 cooperative problem- solving assessment framework, aims to establish interactive and cooperative problem-solving online assessment questions for one on two, human- agent machine, and find out what collaborative skills that elementary school students are lack of. The results of the research can be provided as a reference in teaching, which are as bellow. 1. This study successfully builds the online cooperative problem-solving assessment for elementary school students. 2. The results have shown that among the three core competencies in cooperation performance for students, students performed better in “Establishing and maintaining shared understanding” and “Establishing and maintaining team organization” , but has a relatively poor performance in “Taking appropriate action to solve the problem.” The result indicates that students should strengthen the abilities to set up a plan and act it out when they encounter problems. 3. The results have shown that in the course of the four problem-solving performance, students performed best in “Represent and articulate”; the second is “Explore and understand”; the third is “Plan and execute”, and the worst is the part of “Monitor and reflect”. Therefore, it suggests that the training of “Monitor and reflect” needs to be strengthened. 4. The results have shown that the CPS’s ability of elementary school students in the city is much more than“better” than that of the rural areas, but there are significant gap and differences. This research confirms that there is indeed the existence of the Urban-Rural differences.

Since social media platforms have been widely applied in election campaigns as candidate-controlled media tools, candidates expect their social media outlets being influential when employing strategic actions to affect the public perception and the mass media reports in not only the topics, but also the attributes of reports. For which we know the effect as the intermedia agenda setting. This study examined whether candidates’ social media influence the news’s coverage in framing two leading candidates in both news frames and affective tones during the 2014 Taipei mayoral election, giving evidence to support both first-level and second-level agenda setting theory. With content analysis of two leading campaign issues during the election, MG149 account controversy and wiretap case, results showed strong correlation between the candidates’ Facebook posts and four major newspaper coverage in the quantity of news frame, but results for in affective tones were not confirmed. Intermedia agenda setting effect is proved occurring in both Ko’s and Lien’s campaign, since the quantity of four categories of frames (issue, image, strategy, and ideology frame) in two campaigns members’ Facebook post are aligned with those in newspaper. However, the results for affective attributes differ in each team. Ko’s team members determined a large part of the media frame at both positive and negative frame, showing that second-level intermedia agenda setting effects occurred in his campaign, while Lien’s didn’t have significant correlation with news coverage in affective dimension, only his team member apparently transferred a powerful negative frame to mass media in wiretap case.
