Nordick, Wendy Gale. "Insight into insight : a study on understanding schizophrenia." Thesis, University of Canterbury. School of Social Work and Human Services, 2009. http://hdl.handle.net/10092/2531.

Full text
Abstract:
The lack of insight into their illness continues to be a significant barrier to treatment for people with schizophrenia. However, some people with the illness do develop insight which then results in stability and recovery from their condition. Having met the criteria of good insight, nineteen participants participated in semi-structured interviews exploring the concept of insight and recovery in the context of their personal experiences. Research design used grounded theory methodology layered with a further narrative analysis. The literature review and theoretical perspectives of care provide the context for the research, thus illuminating the known and determining what mysteries in insight development remain. Study findings revealed a Paradox of Insight within a 3-stage process of insight development: The Period of Chaos, the Dynamic Period, and the Period of Wisdom. An overarching Theory of Dangerousness explains the turning point from illness to recovery. This turning point occurs with a Trinity of Crisis. Within this theory, Four Constructions of Dangerousness explain how the meaning making of dangerousness affects recovery. Additionally, Four Types of Insight support this recovery. The importance, meaning, and relevance of study findings for recovery are the focus of the discussion. Six inter-rator focus groups affirmed the findings. Policy and practice implications are discussed, along with recommendations for further research and the acknowledgement of study limitations.

Said, Emanuel. "Insight into action : how firms use customer insight." Thesis, Cranfield University, 2014. http://dspace.lib.cranfield.ac.uk/handle/1826/9274.

Abstract:
Customer insight is fundamental for market oriented organizations to understand their markets. However, the use of customer insight in marketing decisions is poorly understood, partly due to the difficulties in obtaining research access within organizations. But in part because under the perspective of market-based learning (MBL) theory, knowledge is a fixed asset so while there has been interest in insight acquisition, there has been less interest in the processes of insight use. This doctoral research focuses on managers’ use of customer insight within the organisation. It applies the case research method within two organisations using multiple sources of data, including interviews with multiple individuals and real-time experience tracking over a period of time. A framework of the process of insight use is developed from a review of literature and then explored and expanded upon through case study analysis. The emergent framework provides a more granular understanding of the multiple stages of the customer insight use cycle within an organization. It identifies that the insight use process is a perpetual feedback loop learning mechanism and involves several stages identified as: acquiring, filtering, transforming, sharing, analysing/interpreting, actioning and storing. The study finds that some phases are more likely to involve an individual manager while others are more likely to involve managers working collectively. For instance, the stages of acquisition and transforming tend to be individual while the stages relating to interpretation and actioning of insight tend to be collective. Managers may also opt to store insight as their next step for potential actioning at a later stage after any of the process stages. In addition to identifying the stages of insight use, this study identifies the pivotal role of organizational memory in the insight use process. Enablers and blockers of insight use are identified including that managers may respond to perceived information overload by (consciously or unconsciously) blocking information.A key contribution of this thesis is that it incorporates the first use in an organizational behaviour context, of the real-time experience tracking (RET) method. This pioneering use of RET demonstrates that this method may address some of the limitations that plague traditional participant observation techniques in organizational settings, such as active or moderate participation. It demonstrates that RET can be used to track the insight use process of individuals in organizations over time, helping to understand their individual and collective insight use processes. This multi-informant, multi-method study of customer insight use thus provides a deeper understanding of the processes of customer insight use than most previous MBL studies, which have typically employed single-informant, cross-sectional survey approaches. Practitioner implications include that new individual competencies in information use may be needed and that organisations may need to foster a new code of etiquette for information sharing and feedback between peers in organizations operating in today’s information rich environment.

Moreton, Cecily. "Executive coaching for conscious leadership : insight into insight." Thesis, View thesis, 2006. http://handle.uws.edu.au:8081/1959.7/46648.

Full text
Abstract:
This thesis is an investigation of the practice of executive coaching for conscious leadership. The researcher, an executive coach with many years of practice as educator, chaplain, counselling psychologist, Buddhist meditator and company director, draws on case studies to present a holistic understanding of the coaching relationship. She examines how the coaching relationship enables executive managers not only to achieve their management goals, but also to gain insight, assemble meaning, become conscious and present, and grow as leaders. The key questions for the thesis are: What does the coach do and how is it manifested through the coaching discourse? What are the key processes and variables in the coaching relationship that promote conscious leadership? In order to explore these questions, the researcher draws on literature from human relations praxis, ethics, psychology (including counselling, psychotherapy, positive psychology, educational and organisational psychology), education, theology, Buddhist practice and spirituality, leadership and leadership development, and executive coaching. The research methodology is qualitative and quantitative, drawing on data based on an intensive coaching program designed and delivered by the researcher to managers in two organisations – a retail company and a utility company. A series of group sessions reinforced the individual coaching process. Over 300 hours of one-on-one executive coaching sessions were recorded, with 24 executive managers (12 in each company), who each participated in twelve (or equivalent) fortnightly sessions over a 6-month period. In addition, a test of emotional intelligence was given to each of the managers at the beginning and end of the coaching program, giving a quantitative measure of the changes. The results of the coaching sessions are analysed in terms of the two companies and their participants, exploring the organisational context and outlining the benefits of coaching and the coaching relationship with each of the participants individually and in groups. Then two detailed individual case studies are presented, one client from each of the organisations in the study. The managers in the study learned new management skills and adopted new leadership behaviours, reflecting their growth in emotional intelligence and awareness. They reflected on gaining deep awareness and insight, signifying their move to conscious leadership, and reported practical benefits of coaching in their workplace practices, e.g. improved communication skills and the ability to give and receive genuine feedback. A quantitative analysis of the emotional intelligence pre- and post-test scores confirmed these positive outcomes. Many of the managers reported that the most significant aspect of the coaching was the quality of the relationship between them and the coach, which enabled conscious leadership to develop. Their relationship was deeply influenced by the personal qualities of the coach such as warmth, presence, qualifications, credibility, trustworthiness, openness, commitment, insight, and spiritual awareness. This made the difference between coaching for management goals alone and the development of conscious leadership. The concepts of executive coaching and of conscious leadership are holistic phenomena. The clients experience a profound sense of feeling known, respected, accepted, cared for and valued. In essence, they experience love. What has emerged from the research is that consciousness is the ‘being’ of leadership. The attributes of conscious leadership indicate an integration of the whole person that manifests in the workplace, in their homes with their families, and in their everyday interactions and responses to the world around them. While instrumental management goals may be achieved with qualified coaches, conscious leadership involves psychological integration via in-depth relationship and psychological interventions, these need to be experienced with a qualified practitioner with demonstrable advanced levels of personal integration and consciousness. The findings call for a more rofessional and holistic approach to executive coaching for leadership.

The purpose of this research is to gain more understanding of the scientific language. Black symbols and representations of the natural world are used to communicate complex thoughts in various dimensions. Through the process of purification I communicated with confusion and the privilege of being ignorant became my inspiration. My new self is like the alchemists; investigating in new dimensions where symbols and imagery exist to comprehend the always-surrounding unknown. With soda-lime and flat glass I created solid glass symbols. Through the microscope I looked at organic specimens and self-made glass samples and recorded what I saw. Every detail is an indispensable part of an important whole and perceiving is the key to entering new universes. My encounter with science has given me the tools to develop a greater awareness to investigate why and how I am making art. I am convinced that artists and scientists are similar in seeing the universe in unique and specific ways. It is only the representations of those visions that are phrased in different languages.

Abstract:
Using external information to gain insights is becoming increasingly important in today’s knowledge based economies. However, from both research and practice it is somewhat unclear how organizations manage these activities. Therefore the purpose of this paper is to explore how four different organizations manage their insight creation process from external information. To do this, a literature review about insight creation is done to create an analytical framework. Based on this framework, the insight creation process is analyzed within the organizations to find characteristics of each process. Next, a cross case analysis is conducted that reveal the main differences between the organizations. The results indicate two main styles of managing the insight creation process that can be differentiated with regards to formality. As such, this paper complements previous research within the area of competitive intelligence and knowledge management in the sense that it increases the understanding of the insight creation process.

Abstract:
As the competition is getting tougher it becomes important for product development companies to increase their efforts of collecting customer insight (CI). This is essential to ensure that products are developed in coherence with the customer’s needs in order to decrease the risk of developing products that does not add value to the customer. However, despite customer insight being crucial for product development, companies appear to struggle with implementing a suitable way to collect customer insight. The purpose of this master thesis has been to compile and analyse existing methods and tools used in the CI process in order to provide recommendations on what to consider when designing such a process. Further, the study aims to design and apply a CI process, targeting the wind turbine blade inspection (WTBI) industry, both in order to test the recommendations as well as deliver customer insight requested by the case company FLIR Systems. The methodology of this master thesis was partly the case study at FLIR, consisting of interviews with the employees and an extensive literature review. This has been denoted as stage one. Based on the recommendations derived from stage one, a CI process was designed and applied targeting the WTBI industry, denoted staged two. The result was a set of identified barriers related to the company’s CI process. Further, recommendations of what to consider when designing the CI process was derived, aiming to help the company overcome the barriers. It was proposed that the company needed to clearly state the scope for their CI projects. Also, it was recommended to adopt the customer value chain framework in order to gain a more structured approach when assessing which customers to target. Further, what have been denoted as the design guide was provided to the company in order to encourage an active evaluation of which tools to include in their CI process. An additional outcome from the test was that the use of a social network channel, such as LinkedIn, seems to reduce several of the barriers. In terms of customer insight for the WTBI industry, the study managed to derive valuable information for the case company which was perceived as useful for further research

To reduce the extensive environmental impact of the textile industry, new methods for textile recycling are being developed. Today, cotton-based fabric is recycled by Renewcell at their facility in Kristinehamn. The recycled product, Circulose®, is a dissolving pulp that can be used to spin new viscose fibers, which in turn can be used to make new clothes. The present work is a feasibility study on the possibility of optimizing Renewcell's recycling process. The work includes an optimization of the pulp process, which results in a more efficient process with a reduced consumption of process chemicals.

Walsh, Kenneth Ronald 1966. "Generating insight for reengineering." Diss., The University of Arizona, 1996. http://hdl.handle.net/10150/290606.

Abstract:
As organizations face stiff competition and changing environments their structure and systems can become less effective to a point at which they require radical change. It is becoming clear that change is often inevitable. However, radical change is difficult. The popular term "reengineering" is used to describe a process by which organizations undergo the challenging radical process change that is critical to remaining competitive. Although the processes of reengineering appears to have more potential than other change methods that merely tweak a poor system or apply outdated principles, it can often fail. A critical step in the reengineering process is the design of the "to-be" model, however no research has been done on how this should be achieved. It appears that improving this step can have a dramatic impact on the success of the reengineering effort as a whole. This study developed a method for creating "to-be" models and tested it with an organization undergoing reengineering. The method used the latest computer support including both group support systems and animated simulation. The design of the "to-be" models was framed as a group problem solving process and therefore the focus theory of group productivity was used to guide the design of the process, including selection of computer support tools. Because so little is known about the process of creating "to-be" models, this study used an exploratory action science approach. What is known about organizational change is that it takes place in a politically charged arena where individuals have strong vested interests in its outcomes. This environment is difficult to create in the laboratory and therefore the research was conducted using a real organization undergoing reengineering. Results suggest that the combined use of collaborative technology and process animation gives organization members a better understanding of current processes and problems associated with them and helps to generate significant ideas for process improvement. Because groups viewing a process animation have sometimes been observed to focus on incremental improvement at the expense of radical change, special attention must be given to facilitation methods and idea generation techniques that are designed to elicit radical change ideas.

Henok, Niyat. "Systemic insight : the interplay between interactivity, incubation and transfer in insight problem solving." Thesis, Kingston University, 2018. http://eprints.kingston.ac.uk/42276/.

Full text
Abstract:
Classical perspectives on problem solving are embedded in computational models of insight problem solving, such as the information-processing model (e.g., Newell & Simon, 1972). Problem-solving activity is viewed as a product of information-processing in which people see or hear problem information, think about the solution, then produce the solution: see or hear, think, then act. More recently, Ohlsson (2011) suggested people solve problems by mentally restructuring the problem information. Hence, insight comes about as a consequence of restructuring (Weisberg, 2014). As such, the origin of insight is commonly understood as a mental experience. However, the traditional frameworks explaining the insight experience commonly overlook the influence of reasoners' immediate environment. Systemic cognition frameworks such as the Extended Mind Thesis (Clark & Chalmers, 1998), Distributed Cognition (Hollan, Hutchins, & Kirsh, 2000) and the Systemic Thinking Model (Vallée-Tourangeau, Abadie, & Vallée-Tourangeau, 2015; Vallée-Tourangeau & Vallée-Tourangeau, 2017) assume information-processing is augmented when spread across mental and physical resources. When presented with a physical representation of a task, making changes to that physical representation, even arbitrary ones, may offer cues to new strategies, enabling better planning and efficiency in progressing towards a goal. Accordingly, the opportunity to interact and coordinate with the immediate environment enhances insight performance. This thesis sought to explore insight performance from a systemic cognition perspective. The research program investigated how the level of interactivity influenced solution rate in the Cheap Necklace Problem (de Bono, 1967; Silveira, 1971). Across four experiments, participants attempted to solve the problem either in a low interactivity condition, using only pen-and-paper and relying heavily on mental restructuring, or in a high interactivity condition, with a physical model of the problem with constituent elements they could manipulate while attempting to find a solution. The results across the experiments confirmed that increasing the level of interactivity resulted in enhanced insight performance. Incubation and transfer are often upheld as key determinants for insight performance. Thus, in addition to exploring the impact of interactivity, the experiments investigated how interactivity may interact with incubation and transfer to promote insight. To measure incubation effects, participants in the first two experiments reattempted the same problem after a two-week break. There was evidence of an incubation effect as performance substantially improved on the subsequent attempt. To explore transfer, a new Cheap Necklace Problem variant was introduced, which participants in the final two experiments attempted following the original version of the problem. Transfer was evident as participants were able to successfully transfer their solution to solve the new variant. Moreover, overall performance improved on the subsequent problem. Across the four experiments, the level of interactivity offered on the second problem attempt was important: When the problem presentation changed (low interactivity to high interactivity or high interactivity to low interactivity) performance only improved when working in a highly interactive task environment second. Thus, insight through interactivity fosters stronger performance on both the initial and subsequent task. This thesis further explored how interactivity prompts insight in a dynamic agent-environment by recording and analysing participants' actions. One important finding from these behavioural analyses was the fact that those who spent the largest proportion of their time reconfiguring the task environment, thus making the most of the malleability of the artefacts available, were also most likely to reach insight.

Lack of insight is usually reported in Bipolar Disorder (TB), especially when patients have seen the manic pole of the disorder. This lack of verbal awareness of deficits, signs and symptoms of the disorder may be accompanied by an implicit insight, which is the indirect demonstration of some level of knowledge about the disease or a disability with partial or total loss of verbal recognition. Two studies were conducted to verify the relationships between explicit and implicit forms of insight into TB. For this, a theoretical model known as The Cognitive Awareness Model (CAM) was used, which allows to investigate dissociations in the patterns of implicit and explicit consciousness. In the first study, participants were assessed for a number of clinical variables, including an explicit insight measure, as well as an implicit insight measure with depression and mania-related stimuli. In the second study, participants were also tested for clinical variables, including an explicit insight measure, as well as an implicit insight measure with stimuli linked to healthy and ill status. The results of both studies pointed to differences in measures of explicit insight between the manic and euthymic groups and demonstrated differences in mean reaction time for implied self-associations for condition. From these results, it can be suggested that patients in mania explicitly self-evaluate in a similar way to patients in euthymia, which can lead to impairment in adherence to treatment, whereas bipolar patients presented an implicit insight for symptoms and for the patient s condition evaluated.

The present study aimed expand the comprehension about the processes of insight in the Asperger?s Disorder (AD). Insight can be understood as the knowledge one has about his diagnosis and the meaning constructed from it. The development of insight is taken not as a personal mental function, but as a intersubjective phenomenon, forged in social relations. The study has its basis on the theoretical framework of Historical?cultural psychology with focus in the genetical development perspective. As a central feature of this perspective are the processes of meaning construction, considering the social-cultural insertion of the individual and his experiences. Thereby, the study advocates that the understanding of ?what it is like to be an Asperger? has to go beyond the socially shared idea of cognitive disruption as only features of this condition. The effort of comprehending meanings and experiences of these individuals becomes imperative. Therefore, the insight phenomena emerges as a complex construct, demanding a deep and broad research. In order to achieve the study goals, activities to investigate the Asperger experience were proposed in three steps. First, it was produced a narrative surrounding the central question that guides the study: ?what is it like to have asperger?s??. A mediation strategy was used as facilitating tool to the narrative production. It involved an alien character to whom the participants should talk to, explaining the asperger experience, since they came from a planet where there was no such thing. After the initial production, an semi-structured interview was used aiming to investigate aspects regarding the formal knowledge they had of the diagnosis. In sequence, another activity was proposed intending to investigate their ability to identify AD characteristics in others and, at the same time, facilitating the access and production os meaning to their own experiences towards the diagnosis. Three individuals with ages between 15 and 20 years old participated of the study, selected by convenience. They all had a previous diagnosis of AD and had knowledge of it since at least 5 years. That was considered enough time to the construction of meanings about the diagnosis. Also participated in the study their respective parents, who took part in the same investigation steps as their subjects and responded as if they were those. Results showed that they produced narratives about their AD diagnosis and that in their discourse there were many elements notedly present in the media and scientific material. As of the meanings surrounding the diagnosis, is was noted that media elements were also present, but specially approximations were seen between their and their parents perceptions. Therefore, it was noted that the discourse constitution about their own diagnosis for the AD individuals had many influences of the social discourses, notedly of closest people. The results sugest that a characteristic os the processes of insight in AD can be described as a disruption in the transition from a monologic to a dialogic way of thinking. It I believed that the results here exposed can contribute to the comprehension of the singularity of the existence and subjective experience of these individuals.

From nanoparticles system over colloids and polymers to biological systems like membranes and finally living cells, a broad range of materials was considered from a theoretical standpoint. In this dissertation five chemistry-related problem are addressed by means of theoretical and computational methods. The main results can be outlined as follows. (1) A systematic study of the effect of the concentration, chain length, and charge of surfactants on fullerene aggregation is presented. The long-discussed problem of the location of C60 in micelles was addressed and fullerenes were found in the hydrophobic region of the micelles. (2) The interactions between graphene sheet of increasing size and phospholipid membrane are quantitatively investigated. (3) A model was proposed to study structure, stability, and dynamics of MoS2, a material well-known for its tribological properties. The telescopic movement of nested nanotubes and the sliding of MoS2 layers is simulated. (4) A mathematical model to gain understaning of the coupled diffusion-swelling process in poly(lactic-co-glycolic acid), PLGA, was proposed. (5) A soft matter cell model is developed to explore the interaction of living cell with artificial surfaces. The effect of the surface properties on the adhesion dynamics of cells are discussed.

Abstract:
Most patients with schizophrenia exhibit full or partial non-adherence to pharmacological treatment. Only about one-third reliably take antipsychotic medication as prescribed. Poor adherence (i. E. , both complete and partial non adherence) has been found to be associated with serious negative outcomes and as such, interventions aimed at improving and maintaining adherence are of great interest to clinicians, researchers, and policy makers. Objective: To assess the efficacy of a psychotherapy based on motivational enhancement and cognitive therapies designed to improve patient's adherence to treatment and motivation to change. Method: 54 patients diagnosed with schizophrenia about to be discharged following inpatient treatment were included in a six month repeated measures study. Patients were randomly assigned to either the experimental or control therapies and were blind to group assignment. All patients received long acting injectable antipsychotic medications and were rated as compliant when the injection was confirmed and non compliant if the injection was refused or the appointment was missed. Results show that compared to the control psychotherapy the cognitive and motivational therapy improved motivation for change, insight and attitudes toward treatment

Abstract:
Many psychotic patients fail to admit they are mentally ill. The current study evaluated the associations between insight, specific symptoms, and neurocognitive impairments. Thirty-three acute inpatients with a schizophrenia, schizoaffective disorder, or psychotic disorder NOS diagnosis were rated on the SAIE, Birchwood's IS, and the BPRS. Neurocognitive assessments of attention and frontal lobe functioning were also conducted. Stepwise multiple regression analyses found composites representing delusions, disorganization, and anxiety/depression, as well as CPT-IP shapes hit rate, served as significant predictors of total insight or the specific insight dimensions. At least for acute patients, symptoms tended to have stronger relationships with and were more regularly predictive of insight than neurocognitive measures, though the attentional task associated with right hemisphere functioning, contributed significantly.

Abstract:
The need to form inter-organisational collaborative working arrangements is now common across community, public and private sectors. Working collaboratively however, is extremely complex and failures abound. Much research has recently been directed at understanding the nature of inter-organisational collaboration. Insight gained through such research provides the basis for informing, pragmatically, those trying to manage collaborative activities in practice. To date, attempts at making the insight on collaboration available and accessible to practice appear limited in scope and success. Many of those who embark on collaborative working arrangements also seem unaware of the need to consider explicitly the management of their collaborative processes. the high level of complexity, coupled with poor awareness of the need to consider the management of collaboration render the task of making insight available to practice difficult. This is the challenge addressed by the research upon which this thesis is based. The aim of research was to generate process theory on the transfer of insight on collaboration to practice. The work was undertaken in Participatory Action Research and Action Research capacities with individuals pragmatically concerned with collaboration in practice. Ten Design Principles for Transferring Insight to Practice were developed. Conceptualisations of who should be targeted, how they should be targeted and what the substance of the insight should be were also developed. These developments address relevant issues pertaining the Transfer of Insight on Collaboration to Practice.

Abstract:
Duchenne Muscular Dystrophy (DMD) is a genetic disorder characterized by progressive degeneration of muscle fibers and dystrophic changes on muscle biopsy¹. DMD accounts for approximately 50% of all dystrophinopathies, with around 21,000 male babies born with the disease each year², ³, ⁴, ⁵. It is also the most lethal X-linked recessive disorder as phenotypic traits are not immediately present at birth¹¹, ³. Patients usually do not live past their 20's without medical intervention to treat associated respiratory and cardiac dysfunctions¹¹, ³. For these reasons DMD remains one of the greatest threats, amongst a range of pediatric pathologies, to the normalcy of child development and parental care. Although treatment options have shown to mitigate the progression of DMD, most are controversial and costly - the estimated annual treatment cost of DMD per patient is $50,953⁵⁸. In light of this, disease awareness and public health education are critical components for acquiring funds needed for research towards a cure¹². My hope is that through this integrated overview of DMD, the medical layman will better understand the depths of this lethal disease, and how it can be detrimental to both the affected child and his caretaker.

Insight has been interpreted in behavior analysis as the sudden resolution of a problem when two or more different repertoires, previously learned separately, interconnect in a new situation without direct training, producing original sequences of behavior. A small body of research has shown some variables which appear to contribute to the production of this phenomenon, but methodological problems, in particular the use of arbitrary objects in the problem-situation and the delay between response and reinforcer, limit its relevance. Under the light of these considerations, the objective of this research was to investigate the phenomenon of spontaneous interconnection of repertoires (or insight ) with variations in the procedures employed in the area. These were: (a) the object that should be reached by the subjects in the problem-situation was established as a reinforcing stimulus before the phases of testing and training; (b) the delay between the target response (from training and testing) and consumption of the primary reinforcer (water) was reduced by the construction of a drinking magazine that could be moved toward the subject. Two Wistar rats were used as experimental subjects. The equipment and materials were a circular chamber of 69 cm in diameter and 50 cm high, four acrylic cubes coated with black cardboard, a nylon string that drove the movement of the cube along the camera, and a buzzer. The procedure was divided in six phases: (a) training to the drinking magazine, (b) pre-test of insight, (c) training to push the cube, (d) intermediate test of insight, (e) training to climb on the cube and rise, (f) final test of insight. In the test of insight situation, the subject should reach the drinking magazine positioned at the top of the chamber by pushing an acrylic cube in its direction and climbing on the cube to drink. Water were used as reinforcement in a continuous schedule of reinforcement. The results showed that none of the subjects solved the problem in a sudden, direct and continuous way, criteria used to qualify the performance as insight . Nevertheless, the procedure used in this experiment has heuristic value as it represents an attempt to produce the phenomenon without the use of arbitrary objects (which significantly reduces the number of training sessions), and rendered the concept of functional generalization unnecessary. Finally, this research points out obstacles that the area faces in the study of insight , such as the difficulty in replicating Epstein s research and the problems generated by the use of arbitrary objects in the problem-situation, not to mention the limitations imposed by the behavioral interpretation of the phenomenon

Abstract:
Dissertation presented to obtain the PhD degree in Biochemistry
This dissertation portrays recent development on the knowledge of the stability determinants and of functional characteristics of multicopper oxidases (MCO). Multicopper oxidases are a family of enzymes that includes laccases (benzenediol oxygen oxidoreductase; EC 1.10.3.2), ascorbate oxidase (L-ascorbate oxygen oxidoreductase, EC 1.10.3.3) and ceruloplasmin (Fe2+ oxygen oxidoreductase, EC 1.16.3.1). MCO are characterized by having four copper ions that are classified into three distinct types of copper sites, namely type 1 (T1), type 2 (T2) and type 3 (T3). The classical T1 copper site comprises two histidine residues and a cysteine residue arranged in a distorted trigonal geometry around the copper ion with bonding distances approx. 2.0 Å (1 Å=0.1 nm); a weaker fourth methionine ligand completes the tetrahedral geometry. The copper–cysteine linkage is characterized by an intense S(π)→Cu(dx2−y2) CT (charge transfer) absorption band at approximately 600 nm, and a narrow parallel hyperfine splitting A\\ = (43–90)×10−4 cm−1 in the electron paramagnetic resonance (EPR) spectrum. The function of the T1 copper site is to shuttle electrons from substrates to the trinuclear copper centre where molecular oxygen is reduced to two molecules of water during the complete four-electron catalytic cycle. The trinuclear center contains a T2 copper coordinated by two histidine residues and one water molecule, lacks strong absorption bands and exhibits a large parallel hyperfine splitting in the EPR spectrum (A\\ = (150–201)×10−4 cm−1). The T2 copper site is in close proximity to two T3 copper ions, which are each coordinated by three histidine residues and typically coupled, for example, through a dioxygen molecule. The T3 or coupled binuclear copper site is characterized by an intense absorption band at 330 nm originating from the bridging ligand and by the absence of an EPR signal due to the antiferromagnetically coupling of the copper ions.(...)

Abstract:
The emphasis of undergraduate mathematics content is centered around abstract reasoning and proof, whereas students' pre-college mathematical experiences typically give them limited exposure to these concepts. Not surprisingly, many students struggle to make the transition to undergraduate mathematics in their first course on mathematical proof, known as a bridge course. In the process of this study, eight students of varied backgrounds were interviewed before during and after their bridge course at BYU. By combining the proof scheme frameworks of Harel and Sowder (1998) and Ko and Knuth (2009), I analyzed and categorized students’ initial proof schemes, observed their development throughout the semester, and their proof schemes upon completing the bridge course. It was found that the proof schemes used by the students improved only in avoiding empirical proofs after the initial interviews. Several instances of ritual proof schemes used to generate adequate proofs were found, calling into question the goals of the bridge course. Additionally, it was found that students’ proof understanding, production, and appreciation may not necessarily coincide with one another, calling into question this hypothesis from Harel and Sowder (1998).

Abstract:
This content analysis assesses the insight facilitating capacity of some very common inquiry-based writing activities (featured in today's mainstream first-year college composition texts). It accomplishes that assessment by using three language-based insight facilitating methods--one centered on metaphor, another on opposition, and the other on paradox--as evaluative lenses. The position of this study is that these methods--advanced by widely published scholars in the fields of science, psychology and business as effective insight facilitators--can shed light on development opportunities (where insight facilitation is concerned) in the design and protocol of the writing activities selected for analysis. The outcome is ultimately a comparison of sorts drawn between key insight facilitators at work in the proven methods and comparable features capable of eliciting insight in the writing activities. While the analysis aims to show just how effectively insight facilitation is prompted in the selected writing activities, it also--through its evaluative lens--suggests ways the activities could more effectively do so.

Falconieri, Antonella. "Insight into non-canonical pVHL functions." Doctoral thesis, Università degli studi di Padova, 2019. http://hdl.handle.net/11577/3422841.

Full text
Abstract:
One of the most frequent key factors driving tumorigenesis is the functional impairment of tumor suppressor proteins. Mutations of the von Hippel-Lindau tumor suppressor protein (pVHL) are causative of the von-Hippel Lindau syndrome, a hereditary predisposition to develop cancers. The protein exists in two main isoforms: the full-length pVHL30 and a shorter pVHL19 lacking the N-terminal portion. Both these proteins exert their function as oncosuppressor by regulating the hypoxia-inducible factor 1-alpha (HIF-1a) stability. Besides its well-characterized role in HIF-1α regulation, pVHL is proposed as a multifunctional adaptor protein involved in multiple different cellular processes. These additional functions are collectively referred as non-canonical pVHL functions. Aim of this thesis is the further characterization of these HIF-independent functions. A multidisciplinary strategy combining experimental and bioinformatics approaches was used to address three different biological questions: the relationship between pVHL and cell-cycle regulation, the role of pVHL30 in testis-tissue and the existence of a functional link between pVHL30 and the androgen receptor (AR). My work identified novel protein-protein interactions that further link pVHL to cell-cycle control. In particular, I identified a new direct interaction between pVHL and the Cip/Kip protein family (i.e. p21,p27,p57), mediated by the pVHL30 β-domain and a COOD-like motif located into the linear CDKN1s N-terminal domain. In parallel, my efforts in dissecting pVHL also demonstrated pVHL30 to bind the human Mouse double minute 2 homolog (MDM2), an E3-ubiquitin ligase involved in the degradation of the Tumor protein p53 (p53). This interaction was found to be pVHL30-specific and mediated by its N-terminal tail, with the shorter pVHL19 lacking this binding property. The molecular dissection distinguished the pVHL30 B-domain and the N-terminal tail as the regions interacting with a long disordered portion located at the MDM2 C-terminus. On note, the association was found to be mediated by two specific intrinsically disordered portions confirming the pVHL ability to recognize and bind linear motifs. Then using a Y2H genome-wide screening approach I investigated the unknown pVHL30 interactome in testis-tissue. This analysis identified 260 positive clones further classified into 2 main groups addressing 61 human proteins and 118 non coding sequences, respectively. Among the first group I found 6 already known pVHL30 interactors (e.g. Elongin C) that confirmed the approach reliability, whereas the remaining 55 novel interactors were never associated to pVHL30 so far. These testis-specific interactors include proteins involved in relevant cellular pathways such as cell-cycle regulation, DNA damage repair, apoptosis and cytoskeleton regulation. On the other hand, a preliminary protein-protein interacting network generated using these data showed limited internal connections at the pathway level, suggesting a broader involvement of pVHL30 in other pathways not yet identified. Finally considering the relevance of AR in testis-tissue, I investigated the functional relationship between pVHL30 and this protein. Multiple cellular biology techniques were employed to identify a physical interaction between pVHL30 and AR wild-type (AR24Q) or poly-Q expanded (AR65Q). The expansion of the AR poly-Q stretch is linked to protein dysfunction that leads to pathological conditions. My findings demonstrated pVHL30 to inhibit the AR transcriptional activity as well as influencing AR protein stability by accelerating its turnover. In summary, findings reported in this thesis identify 4 novel general interactors that point to pVHL30 involvement in cell-cycle regulation, while 55 new interactors were found to be testis-specific. Moreover, important pieces of evidence shedding light on the functional relationship between pVHL and AR are also discussed. Collectively taken, all of these evidence suggest pVHL30 to have isoform-specific functions as well as putative testis-specific and sex-linked functions.

Our studies 4 and 5 assessed the quality of clinical and cognitive insight and its impact on impulsivity and symptomatology. In study 5 specifically we linked the metacognition dimension to specific mechanisms and confirmed the presence of good insight level in borderline patients as well as a high level of metacognition. Our study 3 examined the difference between impulsivity and aggression in the field of personality disorders. Finally, our 6th study examines the dynamics of positive urgency in relation to insight. Our following studies explore the dynamics of impulsivity, highlighting the important role of negative but also positive urgency. This work made it possible to complete theoretical knowledge, to highlight the importance of positive emotions in the impulsive processes of TPBs, to confirm the presence of good quality insight and to explore new avenues for therapies for this disorder

Abstract:
Debates of contemporary psychiatry point that problems on definition has led to clinical inaccuracies. One of the main situations described in the literature concerns to the expansion of delusion toward a refusal of treatment. Such situations are denominated from a wide semantic spectrum that ranges from the lack of insight, lack of awareness of mental illness, lack of critique and even simple non-adherence to treatment. To contribute to this disagreement as from the psychoanalysis we suggest that, like other unconscious phenomena linked, the delusion can be reset from the subject\'s ability to criticize it, which depends on reorganization of perceptual aspects (insight) of the delusion itself. Because critical terminology is present in the philosophy and history of psychopathology, we discuss changes in delusion ranging from Kant to Pinel, from Freud to Lacan\'s doctoral thesis. Critique is an activity of judgment that can be linked to perceptual interpretation and reordering (insight), but can also be the result of the activity of critical instances of the subject. In order to differentiate these two types of criticism that we find, critique of delusion and critique in delusion, we suggest considering it as a form of subjective implication, linked to synthesis of representations, intentionality of judgment and responsibility of conduct

Abstract:
This thesis investigates the relationship between analysis and insight in Prasarigika Madhyamika Buddhism. More specifically it asks whether analysis is a necessary and/or a sufficient condition for the generation of insight. The thesis is divided into six chapters which include an introduction, an appendix which outlines Prasangika and Svatantrika views regarding the syllogism (svatantra) and a conclusion. The remaining chapters seek to demonstrate that analysis for the Prasarigika and the Tibetan Geluk-ba school is a necessary, but not a sufficient condition for the arising of insight. Chapter one is an investigation of certain Western psychological theories which deal with the effects on the mind of cogitating upon contradictions. Though the psychological theories are only suggestive, this chapter will lend support to the claim a) of the Prasaiigika, that analysis generates an altered state of consciousness; and b) that analysis may be a necessary and sufficient condition for the generation of insight. Chapter two seeks to construct a logico-psychological model of how insight is generated. In this model it is argued that insight is a conceptual and intuitive experience: i.e. non-inferential, and that all thought comes via the intuition. This model argues that analysis is a necessary, but not a sufficient condition for the generation of insight. In chapter three an investigation of the writings of Dzong-ka-ba (Tsong-Kha-pa) is undertaken in order to ascertain how the Tibetan Geluk-ba (dGe lugs pa) school regard the relationship between analysis and insight. The model of chapter two will be compared with the Tibetan Geluk-ba accounts to gauge its explanatory power and correspondence with the Geluk-ba views. Chapter four is an investigation of the writings of certain Western scholars. This investigation seeks to ascertain how these scholars may have regarded the relationship between analysis and insight. The chapter then compares these views with the model developed in chapter two.

Abstract:
Bibliography: leaves 151-164.
Insight has enjoyed the reputation of an elusive phenomenon in psychology and insight problems are very difficult to solve. Only very specific hints concerning their solution have been found to significantly increase the number of problem solvers who are able to solve insight problems. The result of this has been to suggest that insight does not exist, that it is a mysterious phenomenon, or that it is an aspect of problem solving which we have so far failed to understand. Insight in problem solving is investigated from the perspective that the phenomenon needs explanation and it is argued that, while insight has been operationally defined and a clear set of key empirical findings have been established, the conceptual explanation of insight has been largely ignored. It is suggested that a conceptual account of insight is needed so that this aspect of cognitive processing can be incorporated into the main body of cognitive research on problem solving. The current tension in cognitive science and cognitive psychology is examined and it is argued that writing a conceptual account of insight in neural network theoretical terms will not only advance our understanding of insight, but will also reflect on the debate in cognitive theory. This is a result of its status as an aspect of problem solving and as a phenomenon which symbolic theory has so far failed to offer a clear explanation for. A conceptual account of insight in neural network terms is advanced which offers a comprehensive account of the key empirical findings on insight. It is suggested that insight can be understood as the recognition of a pattern to insight problems. Predictions derived from the theory suggest that overcoming the effects of past learning, employing conceptual transfer, and fostering expertise at insight problem solving will significantly facilitate insightful problem solution.

Abstract:
Failure to acknowledge their mental illness occurs in approximately half of all psychotic patients. Interest has been recently been refocused on insight (i.e., awareness of mental illness), and its associations with treatment compliance and better prognosis. Researchers have called into question the traditional factor structure of insight, instead viewing and defining it as a multidimensional and continuous construct. While factor analytic research has suggested that insight is an independent feature of psychotic disorders rather than a secondary manifestation of psychotic symptoms, several factor analytic studies have identified only one higher-order factor. Furthermore, a significant amount of the research literature has assessed insight or analyzed its relationships using only a single insight score. The current study evaluated the structural model of insight and assessed the associations between the different proposed dimensions of insight and psychotic symptoms. One hundred and six participants recruited from both inpatient and outpatient settings with a diagnosis of schizophrenia, schizoaffective disorder, psychotic disorder NOS, or bipolar disorder with psychotic features were rated on David's Schedule for Assessing Insight-Expanded Version (SAI-E), Birchwood's Insight Scale (IS), and the Brief Psychiatric Rating Scale (BPRS) or the Positive and Negative Syndrome Scale (PANSS). Structural equation modeling (SEM) was utilized to provide stringent, confirmatory statistical tests of the hypothetical factor models while accounting for measurement error. Principal findings from the current study were that the three factor model of insight was supported and that the insight factors were meaningfully correlated to the two symptom factors. Moreover, the three factor insight model provided significantly better fit than a single factor model of insight.

This PHD is part of the work for the NASA InSight mission, which will see a seismometer launched for Mars in 2018. One of the main objectives of the mission is to determine the state of the planet (whether it is liquid or solid) by measuring the amplitude of the main amplitude of the Phobos tide with the SEIS seismometer. As such, this PHD aims at modelising all sources of signal, like the Phobos tide itself but also intrumental noises, in order to estimate the true signal that would be seen on site. Other sources of noises will be studied, such as wind noise and others. A second goal of ours is to develop a code to calculate the tidal forces seen by binary systems, notably asteroids without particular shapes, in order to deduce the tidal displacements and stresses inside them

After tens of thousands years, dogs are ecologically adapted to human societies. This species has a diversity of roles in human societies and their cognition and behavior are of interest in experimental research. Common cognitive processes between humans and dogs are the objective on most problem solving researchs with this participants. However, problem solving research did not observed the behavior of this species on the box displacement task. The objective of this work was to observe the performance of two dogs at two diferente experiments with problem situations. On the first one a white board and a box were available. The problem solving occurred if the dog pushed the box toward the white board, stop pusing below it, climbed at the box and snout poked the board. On the second one, food was suspended and a box was available at the experimental environment. The readily problem solving comprehend pushing the box toward the food, stop pushing below it, climbing at the box and grabbing the food. Each dog was subject to a problem situation pre-test. After, they were trained on two prerequisite operant classes. To push a box toward a monochromatic stimuli and to climb on the box. For one dog, Experiment 1, this second operant class was to climb the box and snout poke a white board, for the other one, Experiment 2, was to climb the box and grab the food. Both dogs solved the problem situations in a sudden and directedness behavior, insight like performance. CPB, Experiment 1, solved the problem situation by pushing the box toward the white board, did so on the first attempt in the recombination test with the box on the training position, on this attempt, the dog pushed the box toward the white board, stop pushing below it, straightened the box using his paw and climbed the box and poked the white board, the whole operant recombination lasted 30 seconds. CAL, Experiment 2, resolved the problem situation by pushing the box toward the food, climbed on it and grabbed the food, the whole operant recombination lasted 4 seconds

Abstract:
In this study, theoretical and computational approaches have been utilized to investigate the mechanisms of natural and artificial metalloproteases. The active sites of most natural metalloproteases contain a tetrahedral zinc center, coordinated by three amino acid residues combinated from His(N), Cys(S), Glu(O), and Asp(O) with a water molecule as the fourth ligand. However, the roles played by the ligands environment in the catalytic functions of enzyme are not clear. In this study, the effects of different ligand combinations (NS2, N2S, N2O, N3, S3, NO2 and NSO) in the mechanism were investigated energy barriers were compared. The machanism and energetics of the substrate bound artificial metalloproteases Ni(II)cyclen (cyclen: 1,4,7,10-tetraazacyclododecane) and Cd(II)cyclen have been investigated. In addition, the mechanism of hydrolysis of Phe-Phe peptide bond catalyzed by another artificial metalloprotease [Pd(H2O)4]2+ has also been studied.

Abstract:
The first part of this thesis is an introduction to the different energy conversion and desalination methods that will be invoked in this work. In a second part, we show that the ionic conductance of a nanopore array is sub-additive with the number of pores. Individal contributions of each pore to the global conductance tend to a null value, if the network is big enough. We note that this phenomenon only involves length ratios, and that working at a nanometric scale does not have any influence. Then, in a third part, we measure the permeability of a pore array at a macroscopic scale. There too, the effect of the array does not depend on the scale of the system. Permeability evolves inversely to conductance: permeability is enhanced by the presence of neighboring pores, but in a smaller proportion than the ionic conductance falls under the same cause. The fourth part uses the results of the two preceding ones, to determine a scaling law for the electric power produced by streaming current and diffusio-osmosis, two methods of osmotic energy conversion. We show that entrance effects have a negative impact on such conversion, more efforts are needed to understand them better and circumvent them. The fifth and last part of this thesis is a numerical work on a new desalination device. It relies on osmosis through a gas phase which is trapped within a hydrophobic nanotube. Its main interest is to use nanotubes bigger than the pores of currently used materials, thus less prone to fouling. We use molecular dynamics methods to study the permeability and selectivity of this device

Abstract:
The purpose of this study is to develop and test the validity of an algorithm allowing the classification of the decedents as were dying of, died of and died with BC, using administrative data, for the study of service delivery to the terminally ill.
Validation was carried out through a chart review of a sample of BC decedents extracted from a tumor registry. The algorithm was then applied to the decedents of a cohort of women with BC. The three groups were compared for their distributions of age and place of death.
The validation showed an excellent crude agreement (0.96). Of the 3334 deaths, 2293 were classified as were dying of, 142 as died of, and 949 as died with. The comparison of age showed that women who died with were older than women who were dying of. These two groups differed in their distributions of place of death.

Abstract:
The report is an attempt to incorporate consumer insight in the female sportswear segment to figureout how it can be used in the clothing industry. The consumer insight methods are performed on anexperimental basis to get a perception of how it can be done and figure out how it differs fromtraditional market research. The research aims to clarify the important parameters on which buyingdecisions are made. The parameters will be based on price, function, fit, brand, design etc. Thereport is made in cooperation with the company Röhnisch sportswear. The consumer insight concept is a part of a consumer driven or consumer oriented businessapproach. The concept strives to find unconscious needs, the driving forces behind them and furtherput them in a context to solve a problem for the consumer. The aim is to satisfy consumers and atthe same time develop a competitive and successful business model. While traditional marketingresearch usually describe what is, a consumer insight approach aim to understand why.The methods used in this research are participative observations, in-depth interviews, gymobservations and market survey. The qualitative and quantitative methods have been compared anddiscussed. The relevance and the importance of the methods used are highlighted and prioritizinglists are estimated. The result shows a wide variety of shopping behaviours. Varied shoppingbehaviour can be seen depending on products. Essential parameters for an exercising garment are fitand design. The importance of other parameters is also discussed in the report. On one hand aselective, focused and deterministic approach is seen and on the other hand a more passive andopen attitude is observed. Papadopoulos, Stavros. "Novel graph analytics for enhancing data insight." Thesis, Imperial College London, 2015. http://hdl.handle.net/10044/1/42992.

Full text
Abstract:
Graph analytics is a fast growing and significant field in the visualization and data mining community, which is applied on numerous high-impact applications such as, network security, finance, and health care, providing users with adequate knowledge across various patterns within a given system. Although a series of methods have been developed in the past years for the analysis of unstructured collections of multi-dimensional points, graph analytics has only recently been explored. Despite the significant progress that has been achieved recently, there are still many open issues in the area, concerning not only the performance of the graph mining algorithms, but also producing effective graph visualizations in order to enhance human perception. The current thesis deals with the investigation of novel methods for graph analytics, in order to enhance data insight. Towards this direction, the current thesis proposes two methods so as to perform graph mining and visualization. Based on previous works related to graph mining, the current thesis suggests a set of novel graph features that are particularly efficient in identifying the behavioral patterns of the nodes on the graph. The specific features proposed, are able to capture the interaction of the neighborhoods with other nodes on the graph. Moreover, unlike previous approaches, the graph features introduced herein, include information from multiple node neighborhood sizes, thus capture long-range correlations between the nodes, and are able to depict the behavioral aspects of each node with high accuracy. Experimental evaluation on multiple datasets, shows that the use of the proposed graph features for the graph mining procedure, provides better results than the use of other state-of-the-art graph features. Thereafter, the focus is laid on the improvement of graph visualization methods towards enhanced human insight. In order to achieve this, the current thesis uses non-linear deformations so as to reduce visual clutter. Non-linear deformations have been previously used to magnify significant/cluttered regions in data or images for reducing clutter and enhancing the perception of patterns. Extending previous approaches, this work introduces a hierarchical approach for non-linear deformation that aims to reduce visual clutter by magnifying significant regions, and leading to enhanced visualizations of one/two/three-dimensional datasets. In this context, an energy function is utilized, which aims to determine the optimal deformation for every local region in the data, taking the information from multiple single-layer significance maps into consideration. The problem is subsequently transformed into an optimization problem for the minimization of the energy function under specific spatial constraints. Extended experimental evaluation provides evidence that the proposed hierarchical approach for the generation of the significance map surpasses current methods, and manages to effectively identify significant regions and deliver better results. The thesis is concluded with a discussion outlining the major achievements of the current work, as well as some possible drawbacks and other open issues of the proposed approaches that could be addressed in future works.
