Full textThe goal of my dissertation is to characterize the contribution of industrial food animal production to between-farm transmission of zoonotic influenza A viruses and transmission of these viruses from industrial food animals to humans. The intention of this research is to improve the capacity of public health policies in the United States to prevent the emergence of pandemic influenza viruses.
Preventing and controlling outbreaks within animal populations and avoiding human infection with zoonotic influenza A viruses can reduce the risk of emergence of pandemic influenza viruses in human populations. Industrial food animal production, which dominates the market in the United States and much of the developed world – and increasingly, the developing world as well – has long been considered biosecure. However, emerging research indicates that these industrial systems are vulnerable to disease incursions and suggests that they may play a central role in driving the emergence of zoonotic diseases. The implications of these industrial systems for human influenza risk, particularly the emergence of novel zoonotic influenza A viruses, remains largely unaddressed in the current literature and in health policy strategies in the United States.
Chapter 1 of this dissertation outlines my research goals and provides background on my central research themes and topics. Chapter 2 documents the limits of biosecurity within industrial systems, highlighting risks to food animal workers. Chapter 3 details a cross-sectional serology study of a cohort of industrial poultry workers and community members (n=99) in the Delmarva Peninsula, a tri-state area of intense poultry production in the Mid-Atlantic region of the United States. No evidence of infection with avian influenza viruses is observed in this population.
Chapter 4 contains a quantitative modeling study to estimates risk of between-farm transmission of avian influenza viruses among industrial poultry farms. This study concluded that company affiliation was a significant source of exposure risk from vehicular transmission. Chapter 5 is a policy analysis of the limitations of current pandemic preparedness policy in the United States to adequately incorporate primary prevention. The central results of this dissertation, their significance to public health and opportunities for further research are highlighted in Chapter 6.

Full textOf all the food produced, about one-third is discarded, of which up to 50% is estimated to be avoidable food waste, avoidable food waste is unnecessary food waste, food that could have been eaten if it had been managed better. For example, moldy vegetables. Households account for the largest part of the unnecessary food waste and single person households waste the most food on a per capita basis. This study has examined the waste of fresh fruits and vegetables, as it is the most frequently thrown food. Wasting food is to waste resources which harmfully affects the environment and nature. There is a lot of research on how much food is thrown away and how it affects the environment in a negative way, but there is a lack of research on what is driving the waste. This study, therefore, with research through design, examined the underlying factors of wasting food and how design can be used as an aid to reduce the avoidable waste. Empirical data were gathered from people living in single households. This has been done through internet-based survey, observation and interview and probes. What emerged was that all people throw away fresh fruit and vegetables between one to five times a week even though almost everyone expressed that they do not throw much. Fruits and vegetables are usually thrown away because they are forgotten, usually in the refrigerator boxes caused of the lack of visibility. This study focused on solutions on how to prevent fruit and vegetable from being wasted, by exposing the fresh fruit and vegetables when stored and supporting produce while storing. The purpose of this study was to investigate how research through design can contribute to reducing wastage for individuals in single-person households, and to develop a preventive invention. The conclusion is that a preventive solution should support the fresh fruit and vegetables and at the same time make them visible when stored so that it is not forgotten and thus thrown away. This study shows an example of how such a product could look like.

Full textMantle cell lymphoma (MCL) is an aggressive B-cell malignancy with bad prognosis, which predominates in males with advanced age. However, studies of the endocannabinoid system and how it affects tumour behaviour provides the basis for designing innovative therapeutic strategies that could open new opportunities for treatment of patient with MCL. It has earlier been shown that the cannabinoid receptor ligand (R)-(+)-methanandamide (R-MA) induce cell death in MCL by accumulation of ceramide. Ceramide has a pro-apoptotic effect on the cell but could be metabolized by the enzymes glucosylceramide synthase (GCS) and sphingosine kinase 1 (SphK1) to molecules with pro-proliferative effect. Therefore, treatments with R-MA on Jeko-1 MCL cell line were performed in this study to determine interference in the proliferative behaviour as well as in the gene expression of the enzymes GCS and SphK1. In addition, treatments with chemotherapeutic substances, such as doxorubicin or cytarabine (Ara-C), and combinations of R-MA and chemotherapeutic substance, were performed for the same reason. Results showed that the proliferation behaviour of Jeko cells remained unaffected when treated with R-MA, in contrast to the decreased proliferative effects shown when treated with cytostatics or combinations of R-MA and cytostatics. Furthermore, a tendency for up-regulation of GCS and SphK1 expression was recognized when cells were treated with cytostatics or combination of cytostatics and R-MA, in contrast to cells treated with R-MA alone. Although, R-MA alone had a tendency for a small down-regulation of GCS expression, it contributed to a potential elevation of GCS expression when combined with Ara-C or doxorubicin. It is believed that the effect from upregulated levels of the metabolizing enzymes GCS and SphK1 is balanced by, earlier observed, up-regulations of the ceramide synthesis enzymes.

Full textFat in milk and cream is present as tiny droplets, which are each enveloped in a thin membrane, called the milk fat globule membrane (MFGM). The MFGM can easily be damaged by factors such as pumping the milk and applying other forms of agitation. MFGM damage is believed to reduce processing efficiency and compromise the quality of manufactured products. A comprehensive review of the literature showed that our understanding of changes occurring in the MFGM post secretion of the fat globule by the mammary secretory cell is still rudimentary. Furthermore, it was found that a fundamental understanding of MFGM damage in raw milk is lacking. Hence, this study sought to develop analytical techniques for studying the MFGM. Fluorescent probes were identified that associated with the MFGM (bovine, ovine, human) in one of two ways: either by embedding in the phospholipid bilayer (lipophilic probe) or by binding to carbohydrate moieties of glycosylated chains in the glycocalyx (lectin probes). The use of these probes, in combination with either conventional fluorescence microscopy or confocal laser scanning microscopy, allowed 2-D images and 3-D images of fat globules to be made. Application of water-soluble lipophilic probes and the lectin wheat germ agglutinin (WGA) directly to milk allowed the staining of the MFGM in its native environment. Variable distribution patterns of the probes in the MFGM were observed, which suggests that the MFGM of fat globules in harvested milk is structurally and chemically heterogeneous both within and among globules from the same species and between species, and even among fat globules within the milk of an individual animal. Furthermore, the binding behaviour of WGA to the MFGM of native fat globules (in bovine milk) and washed fat globules (in model systems) following heat treatment implicated β-lactoglobulin, α-lactalbumin, immunoglobulin M and/or the glycosylated proteins Periodic acid Schiff 6/7 in the disappearance of fat globule aggregation upon elevated heat treatment of milk. The results of the current study showed that the use of membrane-specific fluorescent probes, particularly in combination with confocal laser scanning microscopy, has significant potential for providing real time structural and chemical information about the MFGM in matrices such as harvested milk and milk products. In addition to the fluorescence microscopy techniques, development of other techniques was also conducted. Flow cytometry was shown to have significant potential for the quantitative determination of various properties of fat globules and their membranes. Although no suitable sample preparation technique could be developed in this study, atomic force microscopy is believed to have significant potential for studying structural and physical properties of the MFGM. Selective harvesting of individual fat globules was shown to be possible by using a micromanipulator. In future work, this technique is expected to be used in combination with fluorescence microscopy, or atomic force microscopy. The present study has shown that the development and application of novel analytical techniques has advanced, and in the future will further advance, understanding of the MFGM.
