Due to high levels of parasite resistance to different commercial active ingredients, new control alternatives are being studied, including the phytotherapy. These measures aim to search for helper methods in the control of parasitic diseases. However, there are many products available in the market and they have no scientific proof of its efficacy or possible side effects. The objective of this study was to evaluate the hematological and hepatic effects following administration of Neem pie (Azadirachta indica) in sheep. Three strengths of Neem cake was added to the mineral salt (1, 2 and 4 %) and administered during 126 days to 32 Lacaune sheep breed, divided into four groups: three groups for different treatments and a control were tested, which received mineral salt. Blood samples were taken every 21 days to perform the complete blood count, serum total plasma protein and fibrinogen and liver biochemical evaluation. In those same times, fecal samples were collected for quantification of eggs per gram of feces (EPG). Statistical differences between times and groups for several variables were observed, but without being related to the administration of Neem pie. The results of complete blood count, measurement of total plasma protein, fibrinogen and hepatic clinical biochemistry indicated that administration of Neem pie at concentrations of 1, 2 and 4%, does not interfere in hematological values , or on the integrity and liver function Lacaune sheep breed
Devido aos altos índices de resistência dos parasitas aos diferentes princípios ativos comerciais, novas alternativas de controle vêm sendo estudadas, entre elas a fitoterapia. Essas medidas visam a busca de métodos auxiliares no controle das parasitoses, entretanto, muitos produtos estão disponíveis no mercado e não têm comprovação científica de sua eficácia ou de possíveis efeitos colaterais. O objetivo deste estudo foi avaliar os efeitos hematológicos e hepáticos após a administração de torta de Neem (Azadirachta indica) em ovinos. Foram testadas três dosagens da torta de Neem adicionada ao sal mineral (1, 2 e 4%), administradas por 126 dias para 32 ovinos da raça Lacaune, divididos em quatro grupos sendo três grupos para os diferentes tratamentos e um controle, o qual recebeu somente sal mineral. Amostras de sangue foram colhidas a cada 21 dias para realização do hemograma completo, dosagem de proteína plasmática total e fibrinogênio e avaliação da bioquímica clínica hepática. Nestas mesmas ocasiões, amostras de fezes foram coletadas para a quantificação de ovos por grama de fezes (OPG). Foram observadas diferenças estatísticas entre momentos e grupos para diversas variáveis, porém sem estarem relacionadas à administração de torta de Neem. Os resultados obtidos de hemograma completo, dosagem de proteína plasmática total, fibrinogênio e de bioquímica clínica hepática indicaram que a administração de torta de Neem nas concentrações de 1, 2 e 4%, não interfere nos valores hematológicos, nem sobre a integridade e função hepática de ovinos da raça Lacaune

Full textPruning is a common practice in tree fruit crops. However, its effects on the yield are variable and hardly predictable. In this thesis, we propose to study the effects of pruning on vegetative growth and reproduction for the mango tree (Mangifera indica L.), at the axis and at the tree scales, in order to better understand how fruit yield built-up following pruning. Pruning was characterized by two factors: pruning intensity, defined at the tree scale as the amount of fresh biomass removed per unit volume of canopy, and pruning severity, defined at the axis scale as the depth of pruning along the axis. At the axis scale, pruning intensity and severity had an effect on the occurrence, intensity and dynamics of vegetative growth. We also highlighted a major role of the diameter of the pruned axis on the occurrence and intensity of vegetative growth. The vegetative growth which results from pruning led to the emergence of new populations of axes whose proportions depend on pruning intensity. Flowering of these different populations of axes differ in terms of occurrence, intensity and dynamics and was affected negatively by pruning intensity and severity. At the tree scale, yields tended to be lower on moderately and intensively pruned trees as a result of a decrease of flowering rate and/or average weight of fruits. Lightly pruned trees had similar yields compared to unpruned trees. The effects of pruning on the vegetative growth, at the structural and temporal levels, were integrated in a functional-structural model of fruit yield and quality build-up. The model has shown that the number of axes and the leaf area removed at the pruning time were widely compensated by the vegetative growth stimulation that followed. The integration of the effects of pruning on reproduction will allow developing crop management techniques integrating pruning techniques in order to respond to agronomic issues related to mango tree as asynchronisms and irregular bearing

Piriformospora indica is a mutualistic endophyte with a broad host range and application potential in agriculture. Mutualistic microbes are able to produce effector proteins to interfere plant physiology for faciliting symbiosis with plants. In consideration of P. indica’s broad host-range, studies of P. indica effector proteins can establish a model for endophyte symbiosis with plants. This study focuses on functional characterization of effector proteins. According to lab’s previous studies, we already screen out two P. indica’s effector genes, PIIN_09643 and PIIN_11103 by bioinformatics approaches. We established overexpression lines of Chinese cabbage hairy root system and transgenic Arabidopsis thaliana. We also do agroinfiltrantion for transient overespression in Chinese cabbage leaves to confirm suppression of salicylic acid signal transduction. To investigate effectors function, we establish cDNA library to undertake yeast two-hybrid screening experiment. We found B. rapa’s Arabinogalactan protein 4 (AGP4) would interact with PIIN_11103. Arabinogalactan proteins are highy glycosylated glycoproteins mostly localized on plasma membrane, involved in cell proliferation, programmed cell death, pollen-tube development and somatic embryogenesis. This study found that BrAGP4 localized on plasma membrane and chloroplast envelope, and confirmed interaction of BrAGP4 and PIIN_11103 by BiFC. To characterize BrAGP4, we treated Chinese cabbage with AGPs inhibitor, found the inhibitory effects on root hair development but up-regulateing of auxin-relaed gene expressions. We also found that AGPs play a critical role in symbiosis process of P. indica and Chinese cabbage.

Piriformospora indica, an endophytic root-colonizing fungus, interacts with various plants species. From our previous researches, Chinese cabbage root colonization by P. indica results in an increase in plant growth, root propagation and adaptation to abiotic stress. Although P. indica has been reported to colonize and cause various beneficial function on plants, little is known about its early colonizeal mechanism. Previously, we screened several putative effector genes of P. indica by the bioinformatics screening system established from previous researchers. Further to characterize the function of P. indica effector proteins, four candidate genes PIIN_04020, PIIN_04363, PIIN_07152 and PIIN_08445 were chosen to establish Chinese cabbage transgenic hairy-root system、transgenic Arabidopsis thaliana for further investigation. Yeast two hybrid screening were used to screen the target interacting proteins against the effector proteins. B. rapa’s copper chaperone (CCH) against PIIN_04363 and glutathione S-transferase U24 (GSTU24) against PIIN_07152 were found. In this study, we found that PIIN_04363 gene expression was upregulated in the early colonized stages. Transgenic hairy-root system and transgenic Arabidopsis thaliana showed that PIIN_04363 suppress the expression level of SA pathway marker gene, PR-1 and cause a promotion of colonization efficiency. PIIN_07152 enhances the expression level of JA pathway marker genes, LOX2 and VSP2. Redox status relative genes DHAR and MDAR are also upregulated. From the observation of 7days-old transgenic Arabidopsis thaliana, the growth rate of PIIN_07152 OE was retard. It suggests that it is caused by the up-regulation of JA pathway. On the other hand, 7days-old transgenic Arabidopsis thaliana of PIIN_08445 OE show the higher growth rate in seedling stage, suggesting the effects of the up-regulation of auxin-related genes.

Piriformospora indica (P. indica) is a new root endophytic fungus belonging to the Hymenomycetes of the Basidiomycota. P. indica interacts with the roots of various mono- and dicotyledonous plants, showing the positive effect on biomass production. In addition to this growth promoting effect, P. indica also has the potential to induce resistance to fungal disease and to increase the antioxidative activities of plants. After the colonization of P. indica, plant becomes more tolerance to salt and drought. In this study, we used Chinese cabbage (Brassica campestris subsp. chinensis L.) for colonization with P. indica. After colonization for 7 days, six genes expression level of glutathione-S-transferase parC (BcGSTU), aux resistant 1 (BcAUX1) , efflux carrier, (BcPIN3), V-type proton ATPase (BcV-ATPase), expansin A17 (BcEXPA17),and root hair defective 4 (BcRHD4) were up-regulated and we have constructed above genes in pCAMBIA1300-35S-nos vector and tend to investigate gene function of the overexpressing cabbages. In glutathione assay, we found out the concentration of glutathione increased especially in leaves when Chinese cabbage colonized with P. indica. According to this result, we studied the tolerance Chinese cabbage colonized with P. indica under different stress. The results show that Chinese cabbage colonized with P. indica reduced the concentration of hydrogen peroxidase under NaCl stress indicated P. indica could enhance the salt tolerance of Chinese cabbage. We overexpressed these six gene in Arabidopsis and found out these gene could up-regulate the glutathione content and promote the growth of laterial roots, root length, and biomass. 35S::BcRHD4 could promote root hiars and 35S::BcGSTU could against the salt stress and drought sress. We figured out that glutathione play a role in plant growth and tolerance by P. indica colonized with Chinese cabbage.
