Full textStaphylococcus aureus is a major human pathogen that triggers a variety of immune responses, including the sequestration of essential metals (Mn2+, Zn2+), through the degranulation of calprotectin. During my PhD, I characterized a Zn2+-responsive regulatory RNA (sRNA), named ZinS. In fact, I showed that ZinS is expressed during Zn2+ deficiency and is repressed by the transcriptional factor (TF) Zur in the presence of Zn2+. Using the MAPS technique (MS2-affinity purification coupled with RNA sequencing), differential proteomics, and several in vitro techniques, I found that ZinS targets and regulates various mRNAs encoding proteins which use Zn2+ as co-factor, or which are involved in various metabolic pathways and in virulence. In particular, some mRNAs encode enzymes that require Zn2+ as a co-factor, suggesting that ZinS redirects intracellular Zn2+ to essential enzymes. Another mRNA target encodes GpmA, a glycolytic enzyme essential during calprotectin exposure. The results showed that gpmA regulation also includes RsaC and MntR. RsaC is an sRNA expressed during Mn2+ deficiency and repressed by MntR in the presence of Mn2+. Thus, ZinS and RsaC interconnect Zn2+ and Mn2+ homeostasis, helping S. aureus to survive under metal deficiency conditions (also called, nutritional immunity). Other interconnections were observed through the study of MntR and Zur regulons. Hence, the sRNA/FT regulators are one of the strategies used by S. aureus to adapt to metal stress

Full textIn Africa, malnutrition and malaria remain major causes of childhood mortality. To better understand the interaction between these two affections, we have analysed data collected during an intermittent preventive treatment (IPT) of malaria trial conducted on preschool children. Our findings suggest that advertising on malaria and the basic medical services offered during the trial may have improved the height status of the children. We also investigated the impact of malnutrition on susceptibility to malaria. Our results show that, on one hand, wasting was associated with a decreased risk of having a subsequent clinical malaria attack and on the other hand, that specific antibodies (IgG) levels were significantly lower in stunted children compared to well-nourished ones. Altogether, these findings highlight the importance of integrating children nutritional status when defining malaria control interventions, particularly in the development of a malaria vaccine

Full textA growing demand for tools to build and decrypt genetic networks that control cellular processes is felt in biology. We argue that the use of the declarative approach is relevant and applicable to answer questions from biologists about these networks, which are in general partially known. The main idea is to model knowledge about both the structure and the dynamic of a network by a set of constraints representing all the solutions, to check its consistency, to repair a possible inconsistency by an automatic constraint removal, and to infer properties on the structure and dynamic of the network. To demonstrate the feasibility of the approach, we formalize the discrete networks of R. Thomas and relevant biological properties, offer a tool based on constraint logic programming in cooperation with a SAT solver, and validate it on significant biological applications
