Inhibition of protein synthesis in perfused rat liver deprived of either methionine or tryptophan results from a defect in peptide-chain initiation. Similarly, the decreased rate of protein synthesis in liver from rats deprived of food for 24 h and in skeletal muscle after 2 days of diabetes results from a defect in initiation. In the present study, the tissue content of tRNA(iMet) and its level of aminoacylation were measured in these conditions to determine whether methionyl-tRNA(iMet) formation is a mechanism involved in the regulation of initiation. The extent of aminoacylation of tRNA(iMet) in livers perfused with supplemented medium or medium deficient in either methionine or tryptophan was 64 +/- 2, 61 +/- 3, and 66 +/- 2% of the total accepting activity, respectively. The total tissue content of tRNA(iMet), expressed as a percentage of total RNA, was 1.7 +/- 0.1, 1.6 +/- 0.1, and 1.6 +/- 0.1 for the three conditions, respectively. In livers from starved rats, the extent of aminoacylation of tRNA(iMet) was 80 +/- 7% and the total tissue content of tRNA(iMet) was 1.9 +/- 0.1% compared with control values of 82 +/- 6 and 2.0 +/- 0.1%, respectively. In skeletal muscle from diabetic rats, the extent of aminoacylation of tRNA(iMet) was 79 +/- 4% and the total tissue content of tRNA(iMet) was 2.0 +/- 0.3% compared with values of 79 +/- 5 and 2.0 +/- 0.2% for control animals.(ABSTRACT TRUNCATED AT 250 WORDS)

Temperature measurements of InterMET Inc. aluminum-coated iMET-XQ sensors were tested in an outdoor setting under a variety of solar radiation and wind speed conditions. Twelve unshielded sensors were mounted side-by-side on the tower of a South Alabama Mesonet weather station next to a reference sensor on the tower. The iMET-XQ temperatures were most precise and accurate in solar radiation values that were close to zero, regardless of wind speed. Under overcast conditions, wind speeds of 2 m s−1 were sufficient to obtain precise and accurate temperature measurements. During the day-time, aspiration of wind speeds higher than or equal to 3 m s−1 is sufficient. An iMET-XQ was placed in a radiation shield next to the tower reference sensor to test the need for a radiation shield. A second iMET-XQ was placed unshielded on the tower. The iMET-XQ sensors with aluminum coating do not need to be shielded, but they do need to be aspirated. It is recommended that, when taking temperature measurements using unmanned aerial vehicles (UAV) with iMET-XQ sensors, the UAV either fly at 3 m s−1, be embedded in winds of those speeds, or to use the propeller wash of the UAV to aspirate the sensors.

Abstract:
Dilution of HbS with non-sickling hemoglobin or hemoglobin with increased oxygen affinity is clinically beneficial in sickle cell disease. Aldehydes, including 5-HMF, tucaresol or GBT440, modify the N-terminus of HbS by reversible covalent imine formation generating modified forms of HbS that resist polymerization under low oxygen concentrations. In contrast to reversible imine formation by aldehydes, we hypothesize that stable modification of HbS will result from N-terminal retention of the initiator methionine (iMet) and subsequent N-terminal acetylation of the iMet (acetyl-iMet). MetAP2 is the methionine aminopeptidase able to cleave iMet from Val1 on α-globin and βS-globin as the unfolded N-terminal peptides emerge from the ribosome. Enzyme kinetic studies with pure MetAP2 and N-terminal octapeptides showed that βS-globin peptide is a 5-fold better substrate than α-globin peptide. Lentiviral shRNA knock-down of MetAP2 in differentiating erythroid HUDEP cells in vitro confirmed that α-globin is more extensively modified than βS-globin, consistent with the enzyme kinetic data. Selective MetAP2 inhibitors used to treat cultured human erythroid cells (HUDEP and PBMC derived CD34+) and Townes SCD mice in vivo confirmed that both α-globin and βS-globin domains of HbS are extensively modified by N-terminal iMet and acetyl-iMet. N-terminal retention of iMet and subsequent acetylation creates a mixture of modified HbS tetramers with combined modifications on both globins. Cation exchange chromatography separated nine different modified HbS variants from unmodified HbS as identified by LCMS. Purified samples of HbS modified by N-terminal iMet and acetyl-iMet had increased oxygen affinity as measured by decreased P50. Modified HbS containing the acetyl-iMet-βS-globin were found to have delayed polymerization under complete hypoxia (sodium metabisulfite triggered hypoxia in 1.8 M phosphate). Two modified HbS variants were further purified for X-ray crystallography studies (βS-globin / iMet-α-globin and acetyl-iMet-βS-globin / iMet-α-globin). Oxyhemoglobin structures of both modified HbS variants were in the R2-state previously described in structures of aldehyde modified HbS. This R2-state stabilizes the oxygenated R-state of HbS from conversion to the deoxygenated T-state that initiates HbS polymerization in sickle RBC. Treatment by selective irreversible covalent or reversible MetAP2 inhibitors resulted in high levels of HbS modification (>75%) in cultured erythroid cells (HUDEP and CD34+ cells). Dose dependent modification of HbS was observed in Townes sickle cell mouse blood RBC in vivo with total modification of HbS approaching 50%. In whole blood ex vivo studies, modification of HbS decreased RBC sickling under hypoxia (4% O2) and significantly increased the affinity of RBC for oxygen (decreased P50). Blood samples from MetAP2 inhibitor treated mice were analyzed for single-cell O2 saturation of the RBC and for the fractional flow velocity drop in whole blood rheology under decreasing partial oxygen pressures. In blood from vehicle treated sickle mice, a low-saturation peak of deoxy-HbS was observed in 7.8% O2, in contrast to blood from MetAP2 inhibitor-treated mice where the low-saturation peak was only observed in 6.4% O2. Similarly, in an assay of O2 dependent blood flow rheology, the half-maximum fractional velocity drop occurred at 5% O2 in control blood decreasing to 2% O2 in MetAP2 inhibitor treated blood. Our studies show that MetAP2 inhibition results in retention of iMet on βS-globin and α-globin and allows further acetylation of the retained iMet to create a mixture of N-terminal modified HbS tetramers. These modified HbS variants resist polymerization and RBC sickling under conditions of low O2 by delaying HbS polymerization and increasing O2 affinity. The accuracy and precision of iMET-XQ (InterMET Inc., Grand Rapids, MI, USA) temperature measurements in ten different locations on an off-the shelf rotary-wing unmanned aerial vehicle (rw-UAV) were assessed, as a function of atmospheric conditions. The rw-UAV hovered near an instrumented South Alabama Mesonet tower. The mean ± standard deviation of all the temperature differences between the tower and the ten iMET-XQ sensors for all experiments are −0.23 °C ±0.24 °C. Both the UAV and the environment influence the accuracy and precision of the iMET-XQ temperature measurements. Heat generated by the electronic components within the UAV body has a significant influence on the iMET-XQ temperature measurements, regardless of solar radiation conditions, and is highly dependent on wind direction. Electronic components within the UAV body heat up and can cause sensors downwind from the UAV body to record temperatures that are too warm, even if the sensors are aspirated by propeller wash. iMET-XQ sensors placed on rotor arms not near UAV body heat sources, and properly aspirated by propeller wash, perform well. Measurements from iMET-XQ sensors suspended below the UAV are also accurate. When using an off-the-shelf UAV for atmospheric temperature sensing, the electronic components inside the body of the UAV must be properly located. It is recommended that multiple sensors are placed on the UAV. Sensor redundancy will mitigate data loss in case of malfunction during flight and the identification of poorly performing sensors.

How do U.S. International Military Education and Training programs affect the recipient states` behavior in militarized interstate disputes? While the relationship between U.S. military aid in the form of arms and equipment transfer and MID involvement has been studied extensively in international relations literature the effects of U.S. IMET programs on the same phenomena has been largely ignored. This study intends to fill some of this gap. This paper proposes that American educated and trained foreign military personnel return home with a better understanding about the role of the military as an instrument of national power, civil-military relations, and the cost of war. These military personnel advise their political masters against the use of military force during international disputes leading to a decreased probability of both MID initiation and escalation. To test this argument the analysis employs a merged dataset from the Correlates of War Projects and the most prominent U.S. IMET and coups data. Using logistic regression analysis this study finds that more U.S. IMET support a country receives the less likely it initiates MIDs. The analysis also finds that countries that receive U.S. IMET support are less likely to escalate ongoing MIDs to higher levels of hostility.

Abstract The annual mean heat budget of the upper ocean beneath the stratocumulus/stratus cloud deck in the southeast Pacific is estimated using Simple Ocean Data Assimilation (SODA) and an eddy-resolving Hybrid Coordinate Ocean Model (HYCOM). Both are compared with estimates based on Woods Hole Oceanographic Institution (WHOI) Improved Meteorological (IMET) buoy observations at 20°S, 85°W. Net surface heat fluxes are positive (warming) over most of the area under the stratus cloud deck. Upper-ocean processes responsible for balancing the surface heat flux are examined by estimating each term in the heat equation. In contrast to surface heat fluxes, geostrophic transport in the upper 50 m causes net cooling in most of the stratus cloud deck region. Ekman transport provides net warming north of the IMET site and net cooling south of the IMET site. Although cold-core eddies are often generated near the coast in the eddy-resolving model, they do not significantly impact the heat budget in the open ocean in the southeast Pacific.

We compare coincident, balloon-borne, in situ measurements of temperature and pressure by two radiosondes (Vaisala RS92, Intermet iMet-1-RSB) and measurements of relative humidity (RH) by Vaisala RS92 sondes and frost point hygrometers. Data from a total of 28 balloon flights with mixed payloads are analyzed in 1-km altitude bins to quantify measurement biases between sensors and how they vary with altitude. The disparities between sensors determined here are compared to measurement uncertainties quoted by the two radiosonde manufacturers. Our comparisons expose several flight profiles with anomalously large measurement differences. Excluding these anomalous profiles, 33 % of RS92-iMet median temperature differences exceed the uncertainty limits calculated from manufacturer-quoted uncertainties. A statistically significant, altitude-independent bias of about 0.5 ± 0.2 °C is revealed for the RS92-iMet temperature differences. Similarly, 23 % of RS92-iMet median pressure differences exceed the quoted uncertainty limits, with 83 % of these excessive differences above 16 km altitude. The pressure differences are altitude dependent, increasing from −0.6 ± 0.9 hPa at the surface to 0.7 ± 0.1 hPa above 15 km. Temperature and pressure differences between redundant RS92 sondes on the same balloon exceed manufacturer-quoted reproducibility limits 20 % and 2 % of the time, respectively, with most of the excessive differences belonging to anomalous difference profiles. Relative humidity measurements by RS92 sondes are compared to other RS92 sondes and to RH values calculated using frost point hygrometer measurements and coincident radiosonde temperature measurements. For some flights the RH differences are anomalously large, but in general are within the ±5 % RH measurement uncertainty limits quoted for the RS92. The quantitative effects of RS92 and iMet pressure and temperature differences on frost point-based water vapor mixing ratios and RH values, respectively, are also presented.

Nucleotide substitutions in positions 54 and 60 to cytidines and guanosines (C54, G54, C60, and G60) did not prevent cell growth. In contrast, the double mutation U/rT54C60 blocked cell growth, and changing the A-U base pair 1-72 to a G-C base pair was deleterious to the cell, although these tRNAs were synthesized and accepted methionine in vitro. From our data, we suggest that an A-U base pair in position 1-72 is important for tRNA(iMet) function, that the hypothetical requirement for adenosines at positions 54 and 60 is invalid, and that a U/rT at position 54 is an antideterminant distinguishing an elongator from an initiator tRNA in the initiation of translation.

Each comparison (T, P, RH) exposes several profiles of anomalously large measurement differences. Measurement difference statistics, calculated with and without the anomalous profiles, are compared to uncertainties quoted by the radiosonde manufacturers. Excluding seven anomalous profiles, T differences between 19 pairs of RS92 and iMet sondes exceed their measurement uncertainty limits (2 σ) 31% of the time and reveal a statistically significant, altitude-independent bias of 0.5 ± 0.2 °C. Similarly, RS92-iMet P differences in 22 non-anomalous profiles exceed their uncertainty limits 23% of the time, with a disproportionate 83% of the excessive P differences at altitudes >16 km. The RS92-iMet pressure differences increase smoothly from −0.6 hPa near the surface to 0.8 hPa above 25 km. Temperature and P differences between all 14 pairs of RS92 sondes exceed manufacturer-quoted, reproducibility limits (σ) 28% and 11% of the time, respectively. About 95% of the excessive T differences are eliminated when 5 anomalous RS92-RS92 profiles are excluded. Only 5% of RH measurement differences between 14 pairs of RS92 sondes exceed the manufacturer's measurement reproducibility limit (σ). RH measurements by RS92 sondes are also compared to RH values calculated from frost point hygrometer measurements and coincident T measurements by the radiosondes. The influences of RS92-iMet Tand P differences on RH values and water vapor mixing ratios calculated from frost point hygrometer measurements are examined.

Seeing the current conditions, if Indonesia buys military equipment from Russia, such as the Su-35, it will be subject to sanctions, so this research can conclude that the coercive diplomacy carried out by the United States against Indonesia is not effective.

Abstract Sickle cell disease (SCD) is associated with hemolysis, vascular inflammation, and organ damage. Affected patients experience chronic painful vaso-occlusive events requiring hospitalization. Hypoxia-induced polymerization of sickle hemoglobin S (HbS) contributes to sickling of red blood cells (RBCs) and disease pathophysiology. Dilution of HbS with nonsickling hemoglobin or hemoglobin with increased oxygen affinity, such as fetal hemoglobin or HbS bound to aromatic aldehydes, is clinically beneficial in decreasing polymerization. We investigated a novel alternate approach to modify HbS and decrease polymerization by inhibiting methionine aminopeptidase 2 (MetAP2), which cleaves the initiator methionine (iMet) from Val1 of α-globin and βS-globin. Kinetic studies with MetAP2 show that βS-globin is a fivefold better substrate than α-globin. Knockdown of MetAP2 in human umbilical cord blood–derived erythroid progenitor 2 cells shows more extensive modification of α-globin than β-globin, consistent with kinetic data. Treatment of human erythroid cells in vitro or Townes SCD mice in vivo with selective MetAP2 inhibitors extensively modifies both globins with N-terminal iMet and acetylated iMet. HbS modification by MetAP2 inhibition increases oxygen affinity, as measured by decreased oxygen tension at which hemoglobin is 50% saturated. Acetyl-iMet modification on βS-globin delays HbS polymerization under hypoxia. MetAP2 inhibitor–treated Townes mice reach 50% total HbS modification, significantly increasing the affinity of RBCs for oxygen, increasing whole blood single-cell RBC oxygen saturation, and decreasing fractional flow velocity losses in blood rheology under decreased oxygen pressures. Crystal structures of modified HbS variants show stabilization of the nonpolymerizing high O2–affinity R2 state, explaining modified HbS antisickling activity. Further study of MetAP2 inhibition as a potential therapeutic target for SCD is warranted.

Inhomogeneous magneto-electric effect (IMEE) driven by exchange forces is studied. A phenomenological theory of the effect is proposed. The theory predicts the existence of the radial electric polarization in the systems with the magnetic structures of vortex type. Polarization does not depend on the sign of the winding number and appears even in a magnetic “hedgehog”. A microscopic theory of the exchange IMEE in a medium with the vortex magnetization distribution is developed. Electric polarization is caused by the exchange interaction of free charge carriers with the localized ones. On the basis of the microscopic theory, the electrical polarization is estimated for vortices in magnetic semiconductors and metals.

To understand how phosphorylation of eukaryotic translation initiation factor (eIF)-2 alpha in Saccharomyces cerevisiae stimulates GCN4 mRNA translation while at the same time inhibiting general translation initiation, we examined the effects of altering the gene dosage of initiator tRNA(Met), eIF-2, and the guanine nucleotide exchange factor for eIF-2, eIF-2B. Overexpression of all three subunits of eIF-2 or all five subunits of eIF-2B suppressed the effects of eIF-2 alpha hyperphosphorylation on both GCN4-specific and general translation initiation. Consistent with eIF-2 functioning in translation as part of a ternary complex composed of eIF-2, GTP, and Met-tRNA(iMet), reduced gene dosage of initiator tRNA(Met) mimicked phosphorylation of eIF-2 alpha and stimulated GCN4 translation. In addition, overexpression of a combination of eIF-2 and tRNA(iMet) suppressed the growth-inhibitory effects of eIF-2 hyperphosphorylation more effectively than an increase in the level of either component of the ternary complex alone. These results provide in vivo evidence that phosphorylation of eIF-2 alpha reduces the activities of both eIF-2 and eIF-2B and that the eIF-2.GTP. Met-tRNA(iMet) ternary complex is the principal component limiting translation in cells when eIF-2 alpha is phosphorylated on serine 51. Analysis of eIF-2 alpha phosphorylation in the eIF-2-overexpressing strain also provides in vivo evidence that phosphorylated eIF-2 acts as a competitive inhibitor of eIF-2B rather than forming an excessively stable inactive complex. Finally, our results demonstrate that the concentration of eIF-2-GTP. Met-tRNA(iMet) ternary complexes is the cardinal parameter determining the site of reinitiation on GCN4 mRNA and support the idea that reinitiation at GCN4 is inversely related to the concentration of ternary complexes in the cell.

However, there are challenges associated with the process and Professor Woeichyn Chu, who is a distinguished professor based within the Department of Biomedical Engineering at the National Yang Ming Chiao Tung University in Taiwan, is developing a medical device to help surgeons overcome these challenges, benefiting patients and clinicians. The device is called iMET (intra-medullary endo-transilluminating) and came about after Woeichyn Chu spoke with his brother, Dr William Chu, General Orthopedic Department, Cheng Hsin General Hospital, Taiwan, about some of the challenges relating to intramedullary nailing which led the brothers to begin working together to develop a medical device to help target distal locking holes in intramedullary nailing procedures. The iMET device that the brothers are developing is designed to improve target aiming accuracy and reduce dose absorption concerns. In the process, it will save significant time, effort and money. As well as benefiting patients and clinicians, iMET also reduces X-ray fluoroscopy time, which has health benefits for all involved. Woeichyn Chu and his brother are also developing a device called iKypho, which applies negative pressure during bone cement injection in percutaneous vertebroplasty surgeries. iKypho reduces bone cment leakage risks when using low viscosity cement and reducing the costs associated with high viscosity bone cement.

Therefore, it is urgent to evaluate the risk of electric power grid investment in China under new socio-economic development situation, which can help the investors manage risk and reduce risk loss. In this paper, a hybrid novel multi-criteria decision making (MCDM) method combining the latest group MCDM method, namely, Bayesian best–worst method (BBWM) and improved matter-element extension model (IMEEM) is proposed for risk evaluation of electric power grid investment in China under new socio-economic development situation. The BBWM is used for the weights’ determination of electric power grid investment risk criteria, and the IMEEM is employed to rank risk grade of electric power grid investment. The risk evaluation index system of electric power grid investment is built, including economic, social, environmental, technical and marketable risks. The risk of electric power grid investment under new socio-economic development situation in Inner Mongolia Autonomous Region of China is empirically evaluated by using the proposed MCDM method, and the results indicate that it belongs to “Medium” grade, but closer to “High” grade. The main contributions of this paper include: (1) it proposes a hybrid novel MCDM method combining the BBWM and IMEEM for risk evaluation of electric power grid investment; and (2) it provides a new view for risk evaluation of electric power grid investment including economic, social, environmental, technical and marketable risks. The proposed hybrid novel MCDM method for the risk evaluation of electric power grid investment is effective and practical.

The business risk of ERC in China is evaluated by using the proposed MCDM method, and the result shows the current business risk belongs to ‘High’ grade and closer to ‘Very High’ grade more. The proposed MCDM method for business risk evaluation of ERC is effective and practical, which can provide references for risk management and sustainable development of ERC.

The offsets vary during the ascent both in absolute and percent pressure differences. To investigate this problem further, a total of 731 radiosonde/ozonesonde launches from the Southern Hemisphere subtropics to northern mid-latitudes are considered, with launches between 2005 and 2013 from both longer term and campaign-based intensive stations. Five series of radiosondes from two manufacturers (International Met Systems: iMet, iMet-P, iMet-S, and Vaisala: RS80-15N and RS92-SGP) are analyzed to determine the magnitude of the pressure offset. Additionally, electrochemical concentration cell (ECC) ozonesondes from three manufacturers (Science Pump Corporation; SPC and ENSCI/Droplet Measurement Technologies; DMT) are analyzed to quantify the effects these offsets have on the calculation of ECC ozone (O3) mixing ratio profiles (O3MR) from the ozonesonde-measured partial pressure. Approximately half of all offsets are > ±0.6 hPa in the free troposphere, with nearly a third > ±1.0 hPa at 26 km, where the 1.0 hPa error represents ~ 5% of the total atmospheric pressure. Pressure offsets have negligible effects on O3MR below 20 km (96% of launches lie within ±5% O3MR error at 20 km). Ozone mixing ratio errors above 10 hPa (~ 30 km), can approach greater than ±10% (> 25% of launches that reach 30 km exceed this threshold). These errors cause disagreement between the integrated ozonesonde-only column O3 from the GPS and radiosonde pressure profile by an average of +6.5 DU. Comparisons of total column O3 between the GPS and radiosonde pressure profiles yield average differences of +1.1 DU when the O3 is integrated to burst with addition of the McPeters and Labow (2012) above-burst O3 column climatology. Total column differences are reduced to an average of −0.5 DU when the O3 profile is integrated to 10 hPa with subsequent addition of the O3 climatology above 10 hPa. The RS92 radiosondes are superior in performance compared to other radiosondes, with average 26 km errors of −0.12 hPa or +0.61% O3MR error. iMet-P radiosondes had average 26 km errors of −1.95 hPa or +8.75 % O3MR error. Based on our analysis, we suggest that ozonesondes always be coupled with a GPS-enabled radiosonde and that pressure-dependent variables, such as O3MR, be recalculated/reprocessed using the GPS-measured altitude, especially when 26 km pressure offsets exceed ±1.0 hPa/±5%.

Abstract The accuracies of the meteorological sensors (air temperature, relative humidity, barometric pressure, near-surface temperature, longwave and shortwave radiation, and wind speed and direction) that compose the Improved Meteorological (IMET) system used on buoys at long-term ocean time series sites known as ocean reference stations (ORS) are analyzed to determine their absolute error characteristics. The predicted errors are compared to in situ measurement discrepancies and other observations (direct flux shipboard sensors) to confirm the predictions. The meteorological errors are then propagated through bulk flux formulas and the Coupled Ocean–Atmosphere Response Experiment (COARE) algorithm to give predicted errors for the heat flux components, the freshwater flux, and the momentum flux. Absolute errors are presented for three frequency bands [instantaneous (1-min sampling), diurnal, and annual]. The absolute uncertainty in the annually averaged net heat flux is found to be 8 W m−2 for conditions similar to the current ORS deployments in the subtropics.

The offsets vary during the ascent both in absolute and percent pressure differences. To investigate this, a total of 501 radiosonde/ozonesonde launches from the Southern Hemisphere subtropics to northern mid-latitudes are considered, with launches between 2006–2013 from both historical and campaign-based intensive stations. Three types of electrochemical concentration cell (ECC) ozonesonde manufacturers (Science Pump Corporation; SPC and ENSCI/Droplet Measurement Technologies; DMT) and five series of radiosondes from two manufacturers (International Met Systems: iMet, iMet-P, iMet-S, and Vaisala: RS80 and RS92) are analyzed to determine the magnitude of the pressure offset and the effects these offsets have on the calculation of ECC ozone (O3) mixing ratio profiles (O3MR) from the ozonesonde-measured partial pressure. Approximately half of all offsets are > ±0.7 hPa in the free troposphere, with nearly a quarter > ±1.0 hPa at 26 km, where the 1.0 hPa error represents ~5% of the total atmospheric pressure. Pressure offsets have negligible effects on O3MR below 20 km (98% of launches lie within ±5% O3MR error at 20 km). Ozone mixing ratio errors in the 7–15 hPa layer (29–32 km), a region critical for detection of long-term O3 trends, can approach greater than ±10% (>25% of launches that reach 30 km exceed this threshold). Comparisons of total column O3 yield average differences of +1.6 DU (−1.1 to +4.9 DU 10th to 90th percentiles) when the O3 is integrated to burst with addition of the McPeters and Labow (2012) above-burst O3 column climatology. Total column differences are reduced to an average of +0.1 DU (−1.1 to +2.2 DU) when the O3 profile is integrated to 10 hPa with subsequent addition of the O3 climatology above 10 hPa. The RS92 radiosondes are clearly distinguishable in performance from other radiosondes, with average 26 km errors of +0.32 hPa (−0.09 to +0.54 hPa 10th to 90th percentiles) or −1.31% (−2.19 to +0.37%) O3MR error. Conversely, iMet-P radiosondes had average 26 km errors of −1.49 hPa (−2.33 to −0.82 hPa) or +6.71% (+3.61 to +11.0%) O3MR error. Based on our analysis, we suggest that ozonesondes always be coupled with a GPS-enabled radiosonde and that pressure-dependent variables, such as O3MR, be recalculated/reprocessed using the GPS-measured altitude, particularly when 26 km pressure offsets exceed ±1.0 hPa/±5%.

Digital games are a viable alternative to accomplish crowdsourcing tasks that would traditionally require paid online labor. This study compares the quality of crowdsourcing with games and paid crowdsourcing for simple and complex annotation tasks in a controlled exper-iment. While no difference in quality was found for the simple task, paid contributors’ response quality was sub-stantially lower than players’ quality for the complex task (92% vs. 78% average accuracy). Results suggest that crowdsourcing with games provides similar and potentially even higher response quality relative to paid crowdsourcing.

Mental disorders are among those illnesses, which in particular cause suffering to the individual and society.MethodTo study burden of disease for mental in comparison to somatic disorders, 2099 patients from 40 general practitioners filled in (a) the Burvill scale which measures acute and chronic illnesses in ten different body systems and (b) the IMET scale which measures impairment in ten different areas of life.ResultsPatients were suffering on average from acute and/or chronic illness in 3.5 (SD: 2.0) body systems and 56.6% of patients complained about acute and/or chronic mental disorders. The most significant negative impact on the IMET total score have acute and chronic mental disorders, followed by chronic neurological and musculoskeletal and acute respiratory and gastrointestinal disorders, while cardiovascular, metabolic, urogenital, haematological and ear/eye disorders have no greater impact. Acute as well as chronic mental disorders cause impairment across all areas of life and most burden of disease (functional burden of disease 1.69), followed by musculoskeletal disorders (1.62).ConclusionMental disorders are among the most frequent health problems with high negative impact across all areas of life. When combining frequency and impairment mental disorders cause most burden of disease in comparison to other illnesses. This should be reflected in the organization of medical care including family medicine.

It is shown that steel 10KH3A with a super-equilibrium nitrogen content (up to 0,2) electroslag remelting under pressure DESHP-0,1 (FSUE «VIAM») provides a high level of properties (ultimate strength 1670 MPa) while maintaining high mechanical characteristics (elongation 10%, constriction 50%).

Retroviruses and long terminal repeat-containing retroelements use host-encoded tRNAs as primers for the synthesis of minus strong-stop DNA, the first intermediate in reverse transcription of the retroelement RNA. Usually, one or more specific tRNAs, including the primer, are selected and packaged within the virion. The reverse transcriptase (RT) interacts with the primer tRNA and initiates DNA synthesis. The structural and sequence features of primer tRNAs important for these specific interactions are poorly understood. We have developed a genetic assay in which mutants of tRNA(iMet), the primer for the Ty1 retrotransposon of Saccharomyces cerevisiae, can be tested for the ability to serve as primers in the reverse transcription process. This system allows any tRNA mutant to be tested, regardless of its ability to function in the initiation of protein synthesis. We find that mutations in the T psi C loop and the acceptor stem regions of the tRNA(iMet) affect transposition most severely. Conversely, mutations in the anticodon region have only minimal effects on transposition. Further study of the acceptor stem and other mutants demonstrates that complementarity to the element primer binding site is a necessary but not sufficient requirement for effective tRNA priming. Finally, we have used interspecies hybrid initiator tRNA molecules to implicate nucleotides in the D arm as additional recognition determinants. Ty3 and Ty1, two very distantly related retrotransposons, require similar molecular determinants in this primer tRNA for transposition.
