Inhibition of protein synthesis in perfused rat liver deprived of either methionine or tryptophan results from a defect in peptide-chain initiation. Similarly, the decreased rate of protein synthesis in liver from rats deprived of food for 24 h and in skeletal muscle after 2 days of diabetes results from a defect in initiation. In the present study, the tissue content of tRNA(iMet) and its level of aminoacylation were measured in these conditions to determine whether methionyl-tRNA(iMet) formation is a mechanism involved in the regulation of initiation. The extent of aminoacylation of tRNA(iMet) in livers perfused with supplemented medium or medium deficient in either methionine or tryptophan was 64 +/- 2, 61 +/- 3, and 66 +/- 2% of the total accepting activity, respectively. The total tissue content of tRNA(iMet), expressed as a percentage of total RNA, was 1.7 +/- 0.1, 1.6 +/- 0.1, and 1.6 +/- 0.1 for the three conditions, respectively. In livers from starved rats, the extent of aminoacylation of tRNA(iMet) was 80 +/- 7% and the total tissue content of tRNA(iMet) was 1.9 +/- 0.1% compared with control values of 82 +/- 6 and 2.0 +/- 0.1%, respectively. In skeletal muscle from diabetic rats, the extent of aminoacylation of tRNA(iMet) was 79 +/- 4% and the total tissue content of tRNA(iMet) was 2.0 +/- 0.3% compared with values of 79 +/- 5 and 2.0 +/- 0.2% for control animals.(ABSTRACT TRUNCATED AT 250 WORDS)

Temperature measurements of InterMET Inc. aluminum-coated iMET-XQ sensors were tested in an outdoor setting under a variety of solar radiation and wind speed conditions. Twelve unshielded sensors were mounted side-by-side on the tower of a South Alabama Mesonet weather station next to a reference sensor on the tower. The iMET-XQ temperatures were most precise and accurate in solar radiation values that were close to zero, regardless of wind speed. Under overcast conditions, wind speeds of 2 m s−1 were sufficient to obtain precise and accurate temperature measurements. During the day-time, aspiration of wind speeds higher than or equal to 3 m s−1 is sufficient. An iMET-XQ was placed in a radiation shield next to the tower reference sensor to test the need for a radiation shield. A second iMET-XQ was placed unshielded on the tower. The iMET-XQ sensors with aluminum coating do not need to be shielded, but they do need to be aspirated. It is recommended that, when taking temperature measurements using unmanned aerial vehicles (UAV) with iMET-XQ sensors, the UAV either fly at 3 m s−1, be embedded in winds of those speeds, or to use the propeller wash of the UAV to aspirate the sensors.

Abstract:
Dilution of HbS with non-sickling hemoglobin or hemoglobin with increased oxygen affinity is clinically beneficial in sickle cell disease. Aldehydes, including 5-HMF, tucaresol or GBT440, modify the N-terminus of HbS by reversible covalent imine formation generating modified forms of HbS that resist polymerization under low oxygen concentrations. In contrast to reversible imine formation by aldehydes, we hypothesize that stable modification of HbS will result from N-terminal retention of the initiator methionine (iMet) and subsequent N-terminal acetylation of the iMet (acetyl-iMet). MetAP2 is the methionine aminopeptidase able to cleave iMet from Val1 on α-globin and βS-globin as the unfolded N-terminal peptides emerge from the ribosome. Enzyme kinetic studies with pure MetAP2 and N-terminal octapeptides showed that βS-globin peptide is a 5-fold better substrate than α-globin peptide. Lentiviral shRNA knock-down of MetAP2 in differentiating erythroid HUDEP cells in vitro confirmed that α-globin is more extensively modified than βS-globin, consistent with the enzyme kinetic data. Selective MetAP2 inhibitors used to treat cultured human erythroid cells (HUDEP and PBMC derived CD34+) and Townes SCD mice in vivo confirmed that both α-globin and βS-globin domains of HbS are extensively modified by N-terminal iMet and acetyl-iMet. N-terminal retention of iMet and subsequent acetylation creates a mixture of modified HbS tetramers with combined modifications on both globins. Cation exchange chromatography separated nine different modified HbS variants from unmodified HbS as identified by LCMS. Purified samples of HbS modified by N-terminal iMet and acetyl-iMet had increased oxygen affinity as measured by decreased P50. Modified HbS containing the acetyl-iMet-βS-globin were found to have delayed polymerization under complete hypoxia (sodium metabisulfite triggered hypoxia in 1.8 M phosphate). Two modified HbS variants were further purified for X-ray crystallography studies (βS-globin / iMet-α-globin and acetyl-iMet-βS-globin / iMet-α-globin). Oxyhemoglobin structures of both modified HbS variants were in the R2-state previously described in structures of aldehyde modified HbS. This R2-state stabilizes the oxygenated R-state of HbS from conversion to the deoxygenated T-state that initiates HbS polymerization in sickle RBC. Treatment by selective irreversible covalent or reversible MetAP2 inhibitors resulted in high levels of HbS modification (>75%) in cultured erythroid cells (HUDEP and CD34+ cells). Dose dependent modification of HbS was observed in Townes sickle cell mouse blood RBC in vivo with total modification of HbS approaching 50%. In whole blood ex vivo studies, modification of HbS decreased RBC sickling under hypoxia (4% O2) and significantly increased the affinity of RBC for oxygen (decreased P50). Blood samples from MetAP2 inhibitor treated mice were analyzed for single-cell O2 saturation of the RBC and for the fractional flow velocity drop in whole blood rheology under decreasing partial oxygen pressures. In blood from vehicle treated sickle mice, a low-saturation peak of deoxy-HbS was observed in 7.8% O2, in contrast to blood from MetAP2 inhibitor-treated mice where the low-saturation peak was only observed in 6.4% O2. Similarly, in an assay of O2 dependent blood flow rheology, the half-maximum fractional velocity drop occurred at 5% O2 in control blood decreasing to 2% O2 in MetAP2 inhibitor treated blood. Our studies show that MetAP2 inhibition results in retention of iMet on βS-globin and α-globin and allows further acetylation of the retained iMet to create a mixture of N-terminal modified HbS tetramers. These modified HbS variants resist polymerization and RBC sickling under conditions of low O2 by delaying HbS polymerization and increasing O2 affinity. Meetings are very important in any organisation and the Australian public service is no exception. Unfortunately, meetings are costly and time consuming, and often are ineffective and inefficient. Participants are regularly left with feelings of dissatisfaction after the meeting. Electronic meeting systems (EMS) were first developed in the United States in the 1980s to make meetings more effective and efficient. They are now more user-friendly, internet-connected and support multimedia. However, EMS have not been adopted to the extent that could be expected. This study draws from an extensive literature review supplemented by three case studies of Australian companies that provide EMS products and services (Global Learning Pty Ltd, Grouputer Pty Ltd and Zing Technologies Pty Ltd). The study provides answers to six questions: • What are electronic meeting systems (EMS)? • What is the evidence that using an EMS does improve meeting productivity (efficiency and effectiveness) and satisfaction? • Are there other benefits from using these systems? • How does the use of an EMS bring about these improvements in meetings and group collaboration? • What factors need to be managed in order for the organisation to obtain the most benefit from these systems? • Could EMS be used to improve meetings and business processes in Australian (including state/territory) government organisations? The study concluded that EMS could be used to great benefit to improve meetings and their outcomes for an enormous range of tasks that are carried out in all levels in the Australian public sector. EMS have successfully supported meeting sizes from two people to 700, and an even larger number of participants is possible.

The semiarid rainfall regime is northeastern Brazil is highly variable. Climate processes associated with rainfall are complex and their effects may represent extreme situations of drought or floods, which can have adverse effects on society and the environment. The regional economy has a significant agricultural component, which is strongly influenced by weather conditions. Maximum precipitation analysis is traditionally performed using the intensity-duration-frequency (IDF) probabilistic approach. Results from such analysis are typically used in engineering projects involving hydraulic structures such as drainage network systems and road structures. On the other hand, precipitation data analysis may require the adoption of some kind of event identification criteria. The minimum inter-event duration (IMEE) is one of the most used criteria. This study aims to analyze the effect of the IMEE on the obtained rain event properties. For this purpose, a nine-year precipitation time series (2002- 2011) was used. This data was obtained from an automatic raingauge station, installed in an environmentally protected area, Ecological Serid? Station. The results showed that adopted IMEE values has an important effect on the number of events, duration, event height, mean rainfall rate and mean inter-event duration. Furthermore, a higher occurrence of extreme events was observed for small IMEE values. Most events showed average rainfall intensity higher than 2 mm.h-1 regardless of IMEE. The storm coefficient of advance was, in most cases, within the first quartile of the event, regardless of the IMEE value. Time series analysis using partial time series made it possible to adjust the IDF equations to local characteristics

MBA Professional Report
In 2005, IMET research investigated the problems and benefits GSBPP International Military Students (IMS) experience, and how problems could be alleviated. This proposal focuses on implementing the suggestions made by the IMS on Academic Support Facilities and Quality of Life issues. The objective of this MBA Project is to evaluate the prior proposals, identify new problem areas IMS face while at NPS, and follow up on implementation of those recommendations. The research includes the conduct of a survey among the current international students at GSBPP. The proceedings of this initiative that contributed to both developing an implementation strategy and facilitating the gathering of critical information IMS need to achieve a comprehensive understanding were analyzed. Some issues explored include opportunities for streamlining the library, such as more computer facilities, more access to home-country resources for both news and research, opportunities for improving the quality of life such as the non-saluting issue, parking, and finally, the opportunity toward achieving better communication between IMS and the faculty at GSBPP, with the establishment of a new faculty briefing/workshop. The ultimate goal of this study is to ensure maximization of the learning experience of the international students at GSBPP or NPS as a whole.

Abstract:
Integrating anaerobic digestion into electrochemical reactors is an advanced technology for biomethane recovery. Imposing low electric potential between electrodes, supplies CO2, electrons, and hydronium ions from anodic oxidation of organic and/or inorganic compounds. Then, autotrophic methanogens on the cathode produce methane from CO2 and H+ by electron uptake from the cathode. However, in mixed microbial environments, acetogens produce acetate as well. These reactions can take place via two different mechanisms, DIET (direct interspecies electron transfer) or IMET (indirect mediated electron transfer). This work investigates CO2 conversion to acetate and methane in an electrochemical biofilm reactor comparing the efficiency of CO2 reduction via DIET and IMET mechanisms at hydrogen evolving potentials from -0.3 to -0.7 vs SHE. The other goal is to prove the importance of mass balance in CO2 reduction at applied voltages. Simulations are done in AQUASIM version 2.1. Simulation results depicted that higher H+ concentration at -0.7 V vs SHE can reduce more CO2 in DIET with less current generation compared to IMET. This shows DIET the more efficient mechanism. Methane production is dominant in IMET model, however higher current is needed for CO2 fixation in this mechanism. Also, biomass concentration, acetate and methane production, substrate concentration, biofilm thickness, biomass distribution in biofilm, and current density over time in both mechanisms are investigated at variant voltages and substrate concentrations. Simulations showed that at high CO2 levels in both mechanisms CO2 conversion cannot reach maximum if the voltage is not high enough to supply H+.

Abstract:
3D semantic scene completion and 2D semantic segmentation are two tightly correlated tasks that are both essential for indoor scene understanding, because they predict the same semantic classes, using positively correlated high-level features. Current methods use 2D features extracted from early-fused RGB-D images for 2D segmentation to improve 3D scene completion. We argue that this sequential scheme does not ensure these two tasks fully benefit each other, and present an Iterative Mutual Enhancement Network (IMENet) to solve them jointly, which interactively refines the two tasks at the late prediction stage. Specifically, two refinement modules are developed under a unified framework for the two tasks. The first is a 2D Deformable Context Pyramid (DCP) module, which receives the projection from the current 3D predictions to refine the 2D predictions. In turn, a 3D Deformable Depth Attention (DDA) module is proposed to leverage the reprojected results from 2D predictions to update the coarse 3D predictions. This iterative fusion happens to the stable high-level features of both tasks at a late stage. Extensive experiments on NYU and NYUCAD datasets verify the effectiveness of the proposed iterative late fusion scheme, and our approach outperforms the state of the art on both 3D semantic scene completion and 2D semantic segmentation.
