Abstract:
The paper examines the literary technique of Ihor Kostetskyi and the innovative forms and content of his prose from the late 1940s. It focuses on the author’s attempts to update the genre nature of a short story. The analysis covers the principles of creating a figurative system, the structure of a prose narrative, the use of cinematic constructions, the ‘stream of consciousness’ technique, the situation of a lacuna, ‘text within a text’, etc. The research involved the works included in the collection “Stories about the Winners”, as well as the short stories “You Own the Whole World”, “Before the Upcoming Day”, “Divine Lies”, and samples of their critical reception (in particular, the studies by Victor Petrov, Volodymyr Derzhavyn, Yurii Sherekh, and others). Kostetskyi’s collection of short prose “The Story of the Winners” is regarded as a kind of laboratory in which the writer explored the possibilities of language and literary techniques, experimenting with the concepts of genre and image. It is argued that the writer sought to create a technical picture of the world that has nothing to do with the physical world, and the images he created are signs and symbols that need to be deciphered. In the short story “You Own the Whole World”, which is perceived as a “telegraphic mini-version of a psychological novel” or a “mini-novel with a mini-prologue”, one can find the ‘stream of consciousness’ technique and the situation of a gap, that is, a gap between the story of traumatic event and reality when the reader does not receive an unambiguous answer to the question of where the event occurs and whether it occurs at all. The compositional technique of ‘text within a text’, tested by Kostetskyi in the short story “Before the Upcoming Day”, performs the function of a hidden ‘message’ and allows inserting ‘internal’ actions and dialogues into ‘external’ ones. In general, Kostetskyi’s works appeal to the subconscious, enhanced associativity and imagination, and the play factor is important in them as one of the elements of the author’s poetics.

Abstract:
Our laboratory studies a morphogen molecule called Hedgehog (Hh) using Drosophila melanogaster as an animal model. The Hh signaling pathway is evolutionarily conserved from invertebrates to vertebrates, and plays regulatory roles in various aspects of animal development and tissue homeostasis, such as stem cell renewal, tissue repair, and organ regeneration. Hh is a dually lipidated molecule modified by cholesterol at its C-terminus and palmitic acid at its N-terminus, and therefore tightly binds the plasma membrane. Although the hydrophobic nature of the molecule, Hh exerts its function over a long-range of distance. One particular way cells adopted to communicate over long distances is through a new mechanism based on direct cell-cell contacts via long actin-based filopodia extensions, called cytonemes. This new modality for information transfer is at the core of my present project. In this work, we studied this Hh transport mechanism in a polarized epithelial tissue, called the wing imaginal disc, a larval precursor tissue from which the adult wings develop. Cytonemes have been extensively studied in this tissue with the use of the overexpression of a fluorescently tagged protein called Interference of hedgehog (Ihog). The expression of Ihog protein in the wing disc is necessary and sufficient to stabilize these long plasma membrane extensions, otherwise they would be too fragile and easy to be disrupted by conventional fixatives. Here we present Ihog, Hh and Disp as important players in cytoneme growth. In absence of different Ihog domains, we found a significant reduction of cytoneme length and numbers. In addition, Ihog/Boi loss of function was able to reduce the number and length of wild type cytonemes, marked with mCD8GFP. Further, we saw that in loss of function and gain of function genotype for Hh, cytoneme length was reduced and increased respectively, without any change at the cytoneme numbers. With this work, we suggest that Hh has a novel, non-canonical function in the cytoneme growth. To understand the role of cytonemes in Hh secretion we also analyzed discs entirely mutant for Dispatched (Disp). In disp mutants, the Hh gradient is strongly restrained, with most of Hh targets expressed only in anterior cells juxtaposing the A/P border. This phenotype could not be rescued by Ihog overexpression, despite the fact that Hh was very abundant on cytonemes in the disp mutant. Additionally, in the absence of Disp, we observed a reduction of cytoneme length, which suggests a role for Disp in the formation of these filaments, which is likely independent from its function in Hh secretion. In conclusion, although the aforementioned proteins contribute to the structure of cytonemes, we could not measure any direct correlation between the expression of Hh target genes and the simultaneous manipulation of cytoneme length in any mutant condition checked. Finally, we wanted to correlate the time of cytoneme initiation with the establishment of the Hh gradient. In order to do that, we have introduced in the laboratory an alternative system to study cytoneme formation: the abdominal histoblast model, which allowed us to directly analyze the formation and dynamics of membrane extensions in live animals. In particular, we looked at two separated dorsal histoblast nests of the pupal stage (distanced by 30 microns at the onset of pupariation) where the posterior group of cells produces Hh and the anterior cells show expression of various Hh targets. Approximately 15 hours after the onset of the metamorphosis, the abdominal histoblast cells begin to divide and migrate while simultaneously inducing apoptosis in the surrounding larval cells. Our results indicate that the establishment of the Hh gradient occurs before the juxtaposition of the two nests. Nevertheless, we could only observe cytoneme formation after the juxtaposition of the anterior and posterior histoblast cells and not when the two nests were still separated

Abstract:
Abstract Heterotopic ossification is a pathological condition in which bone forms outside the skeletal system. It can also occur in skin, which is the case in some genetic disorders. In multiple miliary osteoma cutis (MMOC), tiny bone fragments develop in the dermis and nearby subcutaneous tissue of the face and upper chest region during middle age. The etiology of the disease is poorly understood. The origin of the osteoma-forming cells is not known and also unknown are the signaling factors that direct the skin cells towards an osteogenic lineage. The purpose of this study was to investigate MMOC and the pathogenesis of ectopic bone formation by combining patient study and cell biology methods. The results from an extensive review of the literature and five new cases revealed MMOC as a distinct disease entity, where heterotopic bone formation is intramembranous. No correlation was found between MMOC and acne scars, hormonal disturbances or GNAS gene mutations. In cell culture studies mouse and human dermal fibroblasts and mouse dermal papilla (DP) cells were found to differentiate into osteoblast-like matrix mineralizing cells. The bone morphogenetic protein 4 (BMP-4) homodimer and BMP-2/7 heterodimer had significant effects on the osteogenic differentiation of the above mentioned cells. Interestingly, the BMPs enhanced the differentiation of mouse cells but reduced it in human cells. In mouse DP cells and human fibroblasts BMP-2/7 was more potent than BMP-4. The skin area affected by osteomas in patients was compared to their unaffected skin and also to the corresponding skin areas in controls with regard to osteogenic differentiation and gene expression studies. MMOC patients’ skin differs from controls both in osteoma and unaffected skin areas, which suggests MMOC is not only a local but also a systemic skin disease. The results confirm the previous findings that gene expression in skin is different in different parts of the body, which could explain why the osteomas develop in certain skin areas. The results of this study provide new information about MMOC and heterotopic ossification in skin and could be useful when developing treatments for MMOC. This study also presents new information about BMPs and their different effects in mouse and human cells, which may stimulate discussion about the generalization of mouse studies in humans and the clinical use of BMPs
Tiivistelmä Virhesijaintinen luutuminen on patologinen tila, jossa luuta muodostuu luisen tukirangan ulkopuolelle. Laajasta kirjallisuuteen perehtymisestä ja viidestä omasta potilaasta saadut tulokset osoittivat ihon lukuisten jyvämäisten osteoomien olevan oma erillinen tautinsa, jossa virhesijaintinen luutuminen tapahtuu suoran luutumisen mekanismilla.

The purpose of the study is to investigate how guidance counsellors in a Swedish municipally looks at the educational cooperation with guidance as well as the actions that can promote an equivalent way of working. To get answers to the above, we conducted thorough interviews with five guidance counsellors who work at primary schools, each of them against their respective schools principal. The background to the investigation is seen by examining the Swedish comprehensive school guidance and counseling, which reports an invalid and non equal educational cooperation. To analyze the collected data we have chosen the Action Theory. This is done using understood automatic and conscious actions, through taking a hold of and changing, as well as actual experiences, reflective observations, abstract thinking and active experimentation. The result shows that the guidance counsellors see the educational cooperation as substandard. That this is seen dependent in both educators as managerial approach, and that this work also may differ from school to school, which thus affects its equivalence.

Abstract:

Abstract:

Abstract:
Most of the engineering courses focus more on theory and very little on hands-on, project-based learning in the classroom. Integration of real-world engineering problems and applications in lower division engineering courses will produce engineering students, who will be technically sound and be able to execute and manage real-world projects, when they will do senior design projects in their final year of engineering study. To overcome the engineering design challenges we have developed iHOP (Ingenieŕia Hands on Project) and integrate it with our lower division engineering courses. iHOP has been developed to emphasis the design component at the University of Texas at Brownsville (UTB) Engineering Physics curriculum and the project is now an integral part of Introduction to Engineering class. The iHOP project is one that is challenging, fun, requires teamwork, associated with the engineering material being studied, low cost, and doable in a limited amount of time. The experience from iHOP project motivates our freshman students to choose a better senior design project in senior year of their college career. The objectives of the iHOP projects are — to have students develop teamwork skills, and to teach students basic engineering design concepts in a complementary format to the traditional lecture. Various techniques related to team selection, encouraging teamwork, incorporation of engineering topics, keeping costs down, project results presentations, and gathering feedback from students will also be presented in this paper. Integrating iHOP Project with Introduction to Engineering class helped us to improve our retention effort in the engineering department.

Rudyk, Myroslava. BLOGGING PLATFORMS AS ARENAS FOR THE MEDIA ACTIVITIES OF CITIZEN JOURNALISTS. Ivan Franko National University of Lviv, March 2024. http://dx.doi.org/10.30970/vjo.2024.54-55.12164.

Full text
Abstract:
The article is dedicated to the investigation of blogging platforms as information and communication arenas during wartime. It analyzes the media activities of citizen journalists as a crucial component of the overall information landscape in Ukraine, outlining the peculiarities of their work, the specifics of information dissemination, and the prospects for the development of citizen journalism as a whole. The advantages and disadvantages of citizen journalism are highlighted. Journalism is one of the most dynamic spheres of our existence, and the information space in Ukraine has undergone significant changes since the full-scale war. The development of technological tools always transforms journalism, elevating it to new levels of possibilities. The use of modern blogging platforms, widely accessible not only to professional journalists but also to activists, has contributed to powerful changes in the information sector. Citizen journalism during the Russo-Ukrainian war has taken on entirely different dimensions than before, prompting new reflections on the role of citizen journalists. This expands the scope of the researched issue, from ethical considerations and adherence to journalistic standards to understanding the safety of information activities for both journalists and humanity as a whole. Not every blogger can be called a citizen journalist. The crucial characteristic of citizen journalism, as opposed to mere blogging, is self-awareness, active social action, dedication to an idea, drawing attention to a problem, lobbying for public interests, and serving the function of public control. In the article were examined the activities of citizen journalists, who meet professional standards and exemplify citizen journalism. These include Ihor Lachenkov, Serhiy Sternenko, and Kostyantyn Liberov. Their blogging platforms and social networks were analysed, their content characterised, and posting frequency assessed. The activities of citizen journalists become most in-demand when media representatives cannot capture an event promptly at a particular moment. Citizen journalists find it easier to disseminate information through blogging platforms and social media, especially considering the tremendous trust these platforms enjoy. Survey results illustrate the colossal trust in these platforms. When asked, «What mass media tools did citizens use to get news in 2022?» the responses were as follows: 1. Social networks – 74%, 2. News websites – 42%, 3. Television – 36%, Radio – 11%, Print media – 3%. We observe a trend in the Ukrainian information space where some bloggers position themselves as citizen journalists, even though they previously did not have such a pronounced civic position. This trend became particularly evident at the beginning of Russia’s full-scale invasion of Ukraine when influencers started using their blogs to inform their audience about the events in Ukraine, mobilizing them for active volunteer actions and resisting enemy challenges. Keywords: citizen journalism, information dissemination, blogging platforms, war.
